Populations of strongyloid nematode infective stages in sheep pastures : comparison between direct pasture sampling and tracer lambs as estimators of larval abundance. International Journal for Parasitology11: 359–367. Over a 2-year period, numbers of infective larvae in samples of pasture herbage, and numbers of worms in previously worm-free “tracer” lambs allowed 4 weeks grazing, were compared as estimators of the abundance of infective larvae on pastures.Transformation of sample estimates of infective larval numbers per 100 g herbage dry matter (DM) and of worm numbers in tracer sheep, according to the expression y = log10 (x+25), was effective in stabilizing variances. Estimates of error variance for each technique did not differ significantly among the genera Haemonchus, Ostertagia or Trichostrongylus and the pooled estimate for the tracer sheep method was 4 times greater than that for pasture sampling. From these results, more tracer sheep than pasture samples would be required to achieve the same level of precision with the two techniques. Using conventional statistical methods, the effects of numbers of pasture samples or tracer sheep on the size of the difference between two means which can be detected as significant and on the width of the confidence interval about a single mean, are illustrated. These can be used as a guide in the choice of sample sizes. Error variances for Nematodirus spp. were significantly less than for the other genera by pasture sampling, and greater by the tracer sheep technique. Possible reasons for this are discussed, but it is concluded that pasture sampling is likely to be much the more precise method for estimating Nematodirus spp. infective larval availability.Changes with time in infective larval abundance, for Haemonchus, Trichostrongylus and Nematodirus spp. which were present in moderate to low numbers, followed similar trends by both techniques. However, for Ostertagia spp. larvae, which were much more abundant, peak levels were defined more sharply and occurred earlier by pasture sampling than by the tracer method. It is suggested that worm counts from tracer sheep, especially those grazing for 4 weeks rather than shorter periods, may systematically underestimate the infective larval population on pasture at high levels of abundance owing to density-dependent worm loss.  相似文献

An observational study was performed to investigate the effects of yarding and ruminoreticulum bypass on the efficacy of oxfendazole against resistant Trichostrongylus spp. helminths. Ruminoreticulum bypass was observed in 9-month old Perendale lambs using video-taped fluoroscopy and the efficacy of oxfendazole was estimated using the faecal egg count reduction test. Yarding and withholding feed for 24 h before drenching with oxfendazole significantly enhanced the faecal egg count reduction for resistant Trichostrongylus spp. parasites (p < 0.05), but ruminoreticulum bypass of the drench in the yarded animals had no negative effect on anthelmintic efficacy (p > 0.05). The effect of ruminoreticulum bypass on the efficacy of oxfendazole may depend on the resistant parasite species present. The positive effects of yarding sheep for 24 h before drenching, on the efficacy of oxfendazole against resistant Trichostrongylus spp., probably outweigh any negative effects of ruminoreticulum bypass. However further studies are required, with different resistant parasite genera present, before firm recommendations can be made.  相似文献

Gastrointestinal (GI) parasitism represents a severe health problem in small ruminant production systems world-wide. The objective of the present study was three-fold: (i) to assess the prevalence of GI parasitism in dairy breeds of sheep and goats in selected areas of Greece; (ii) to determine the species of existing gastrointestinal parasites; and (iii) to investigate the effect of climatic factors on the seasonal variation of parasite population dynamics in dairy sheep and goats reared in two different geographical areas of Greece. The study was conducted in four flocks of either sheep or goats that were equally allocated in northern and central Greece. In each flock, faecal samples were randomly collected from 30 animals at monthly intervals and were used for nematode egg counts and coprocultures. Two animals from each flock were slaughtered monthly and their GI tract were examined for adult worm population and identification. The study revealed that the parasitic burdens in sheep were significantly (P<0.05) higher than those in goats. Faecal egg counts for both sheep and goats were significantly (P<0.01) affected by the month of the year and there was also a significant interaction between month of the year and area of study (P<0.01). Teladorsagia, Haemonchus, Trichostrongylus and Chabertia were the most prevalent nematode genera in both sheep and goats. Such information could be useful for the development of strategic treatments when sheep and goats are reared under systems similar to those described in this study.  相似文献

The accurate diagnosis of strongylid nematode infections is central to investigating their epidemiology and for parasite control. To overcome major limitations in sensitivity or specificity of traditional methods, including faecal egg count (FEC) and/or larval culture (LC), we evaluated and established a semi-automated, high throughput multiplexed-tandem PCR (MT-PCR) platform for the diagnosis of gastrointestinal strongylid nematode infections in sheep, and established its diagnostic sensitivity (100%) and specificity (87.5%) based on the testing of 100 faecal DNA samples from helminth-free sheep and 30 samples from sheep with infections confirmed by necropsy. Subsequently, the platform was employed to test 219 faecal samples from sheep with naturally acquired infections from various geographical localities within Australia and the results compared with those from conventional LC using 139 of the 219 samples. The results obtained using both MT-PCR and LC correlated significantly for most nematodes examined, but revealed that Oesophagostomum venulosum and Chabertia ovina (parasites of the large intestine) were significantly under-represented in the LC results. The results showed that Trichostrongylus spp. (87%), Teladorsagia circumcincta (80%) and Haemonchus contortus (67%) had the highest prevalences, followed by O. venulosum (51%) and C. ovina (12%). The molecular-diagnostic platform established can be used for species- or genus-specific diagnosis of patent nematode infections within 24 h (compared with 7–10 days for LC), and is a sensitive and cost effective tool for routine application in research and service laboratories.  相似文献

The sequence of appearance of Eimeria and helminths, in 20 West African Dwarf kids from birth and the pattern of oocyst and strongyle worm egg output for 1.5 years are described. Eimeria oocysts appeared early about 20 days after birth and oocyst output in some kids reached 2.7 million oocysts per gram (opg) of faeces about 39 days after birth and showed a group mean oocyst output of 443,540 opg in the second month but this declined further to 23,840 opg after 5.5 months. Eimeria arloingi (20.50%), Eimeria ninakohlyakimovae (17.02%), Eimeria alijevi (15.07%), Eimeria caprina (12.65%), Eimeria jolchijevi (11.42%), Eimeria apsheronica (8.70%), Eimeria pallida (5.31%), Eimeria caprovina (3.29%), Eimeria hirci (3.20%) and Eimeria christenseni (2.84%) were seen in a descending order of prevalence. Strongyle worm ova were seen 53 days after birth and peaked soon after the fall in Eimeria oocyst output but thereafter fluctuated. The eggs of cestode, Moniezia spp. appeared later but was transient. Both oocyst and worm egg output declined and were almost absent when the kids were about 1 year old. Faecal larval cultures were made and the L3s identified with the dominant ones being Haemonchus spp. and Trichostrongylus spp. Sixty percent of the kids in this study died when they were 7 months old and a total of 70% of the kids had died before they were 1 year old.  相似文献

The nodular worms (Oesophagostomum spp) are important parasites of food animals and primates. The archaic line that parasitizes pigs appears to be less pathogenic than the parallel evolutionary lines in ruminants and primates. Morphological types of O. dentatum in pigs and an apparent new host record for O. venulosum, which normally affects sheep and wild ruminants, in cattle suggests aggressive recruitment within the genus. The same Oesophagostomum spp occur in humans and in non-human primates but it is not clear which is the natural host.  相似文献

A rapid and accurate assay for evaluating antibabesial drugs on a large scale is required for the discovery of novel chemotherapeutic agents against Babesia parasites. In the current study, we evaluated the usefulness of a fluorescence-based assay for determining the efficacies of antibabesial compounds against bovine and equine hemoparasites in in vitro cultures. Three different hematocrits (HCTs; 2.5%, 5%, and 10%) were used without daily replacement of the medium. The results of a high-throughput screening assay revealed that the best HCT was 2.5% for bovine Babesia parasites and 5% for equine Babesia and Theileria parasites. The IC50 values of diminazene aceturate obtained by fluorescence and microscopy did not differ significantly. Likewise, the IC50 values of luteolin, pyronaridine tetraphosphate, nimbolide, gedunin, and enoxacin did not differ between the two methods. In conclusion, our fluorescence-based assay uses low HCT and does not require daily replacement of culture medium, making it highly suitable for in vitro large-scale drug screening against Babesia and Theileria parasites that infect cattle and horses.  相似文献

Anthelmintic resistance (AR) against gastrointestinal nematodes (GINs) of sheep and goats is a global concern. To address the problem, this study assessed the status of AR in different government and private sheep and goat farms in Bangladesh. We conducted fecal egg count reduction test (FECRT) and Egg hatch assay (EHA) experiments. For the detection of resistant larvae, pooled fecal samples from treated and non-treated groups were subjected to coproculture. Furthermore, 195 adult Haemonchus parasites were genotyped to ascertain benzimidazole (BZ) resistance allele from seven topographic zones of Bangladesh using allele specific PCR (AS-PCR). In FECRT, the percentage reduction along with 95% confidence intervals indicated that GINs were resistant to albendazole (ABZ), levamisole (LEV) and ivermectin (IVM). Coproculture revealed that Haemonchus spp., Oesophagostomum spp. and Trichostrongylus spp. were resistant to anthelmintics. ABZ resistance was also confirmed by in vitro EHA in all the farms except the private goat farm in Mymensingh. The genotype frequencies were 6% for homozygous resistant (rr), 59% for heterozygous (rS) and 35% for homozygous susceptible (SS) among different topographic zones. The allelic frequency of the mutation conferring resistance (r) ranged from 25% to 47% signifying resistance to BZ in nematodes of sheep/goats. The genotype frequencies (rr, rS and SS) and allelic frequencies (r and S) varied significantly (p˂0.05) in different zones in Bangladesh. Overall, the data suggest an alarming condition created by multiple AR in Bangladesh.  相似文献

Concurrent infections with vector-borne pathogens affected a cattle herd in Switzerland, and one of the pathogens was identified as Babesia bigemina, which had never been observed in this country before. Therefore, a survey of the occurrence of ruminant Babesia spp. and their tick vectors in Switzerland was conducted. A total of 2,017 ticks were collected from sheep, goats, cattle, and wild ruminants (deer, roe deer, and chamois) in southern parts of Switzerland and identified morphologically. The vast majority of the ticks (99.2%) were Ixodes ricinus, but 14 ticks from sheep and goats were identified as Dermacentor marginatus and two ticks from wild ruminants were identified as Hemaphysalis punctata. PCR analyses of 700 ticks revealed the presence of Babesia divergens (n = 6), Babesia sp. genotype EU1 (n = 14), and B. major (n = 2), whose suggested occurrence was confirmed in this study by molecular analysis, and the presence of novel Babesia sp. genotype CH1 (n = 4), which is closely related to B. odocoilei and to Babesia sp. genotype RD61 reported from North America. The identification of B. divergens and B. major in ticks collected from wild ruminants cast doubt on the postulated strict host specificity of these bovine Babesia species. Furthermore, the zoonotic Babesia sp. genotype EU1 was detected in ticks collected from domestic animals but was obtained predominantly from ticks collected from wild ruminants. More than one tick containing DNA of different Babesia spp. were collected from two red deer. Hence, the role of these game animals as reservoir hosts of Babesia spp. seems to be important but requires further investigation.  相似文献

A vaccine containing integral membrane glycoproteins from the intestine of Haemonchus contortus was evaluated in three groups of grazing sheep each containing 13 ewes and their 16 lambs naturally infected with gastrointestinal nematodes. Two groups were vaccinated with either 5 or 50 μg of the antigen per immunisation, while the third, the control group, received adjuvant alone. The sheep were immunised six times at 3 week intervals, partly because the vaccine antigens are hidden and thus no immunological boost would be delivered by subsequent infection and partly because the level of Haemonchus spp. challenge was expected to be high. The vaccinated ewes, first immunised approximately 1 month before lambing, showed a circulating antibody response but no signs of reduced anaemia or Haemonchus spp. egg counts, compared with control ewes. Several ewes with severe haemonchosis in all three groups had to be given precautionary treatment with anthelmintic drugs. In contrast, vaccinating their lambs with either 5 or 50 μg of the antigen per immunisation resulted in 10 fold higher antibody titres. In the case of the lower antigen dose this was associated with significantly less anaemia, 72% reduction in the overall number of Haemonchus spp. eggs produced and significantly fewer worms compared with control lambs. It is hypothesised that the heavily pregnant or lactating ewes did not have sufficient physiological reserves to mount a protective response following vaccination in the tropical weather and high challenge conditions that prevailed. Nevertheless, the vaccine could afford useful protection for lambs against H. contortus.  相似文献

Improvement in the use of existing anthelmintics is a high priority need for the pharmaco-parasitology research field, considering the magnitude and severity of anthelmintic resistance as an important issue in livestock production. In the work described here, monepantel (MNP) was given alone or co-administered with either macrocyclic lactone (ML) or benzimidazole (BZ) anthelmintics to calves naturally infected with ML- and BZ-resistant gastrointestinal (GI) nematodes on two different commercial cattle farms. Both pharmacokinetic (PK) and efficacy assessments were performed. On Farm A, male calves (n = 15 per group) were treated with either MNP orally (2.5 mg/kg), IVM s.c. (0.2 mg/kg), ricobendazole (RBZ) s.c. (3.75 mg/kg) or remained untreated. On Farm B, eight groups (n = 15) of male calves received treatment with either: MNP, abamectin (ABM, oral, 0.2 mg/kg), RBZ (s.c., 3.75 mg/kg), albendazole (ABZ, oral, 5 mg/kg), MNP+ABM, MNP+RBZ, MNP+ABZ (all at the above-mentioned routes and doses) or remained untreated. Seven animals from each treated group (Farm B) were randomly selected to perform the PK study. MNP and its metabolite monepantel sulphone (MNPSO2) were the main analytes recovered in plasma after HPLC analysis. The combined treatments resulted in decreased systemic exposures to MNP parent drug compared with that observed after treatment with MNP alone (P < 0.05). However, the systemic availability of the main MNP metabolite (MNPSO2) was unaffected by co-administration with either ABM, RBZ or ABZ. Efficacies of 98% (Farm A) and 99% (Farm B) demonstrated the high efficacy of MNP given alone (P < 0.05) against GI nematodes resistant to ML and BZ in cattle. While the ML (IVM, ABM) failed to control Haemonchus spp., Cooperia spp. and Ostertagia spp., MNP achieved 99% to 100% efficacy against those nematode species on both commercial farms. However, MNP alone failed to control Oesophagostomum spp. (60% efficacy) on Farm A. The co-administered treatments MNP+ABZ and MNP+RBZ reached a 100% reduction against all GI nematode genera. In conclusion, the oral treatment with MNP should be considered to deal with resistant nematode parasites in cattle. The use of MNP in combination with BZ compounds could be a valid strategy to extend its lifespan for use in cattle as well as to reverse its poor activity against Oesophagostomum spp.  相似文献

Madura cattle, which are native to Indonesia and mainly kept on Madura Island, East Java, are expected to contribute to improving the regional meat self-sufficiency. Eimeria spp. are the most pathogenic protozoans among gastrointestinal parasites in livestock but no molecular surveys of Eimeria spp. in Madura cattle have been conducted to date. In this study, a total of 183 fecal samples were collected from Madura cattle and 60 (32.8%) were positive for parasites of protozoans and nematodes by the sugar floatation method. Among the samples with parasites, Eimeria spp. oocysts were detected in 50 samples (27.3%) with an average OPG value of 1686.1. Eimeria spp. were successfully identified to the species level in 26 samples with Eimeria bovis being the most prevalent, followed by E. zuernii and E. aubrunensis. A total of 21 samples showed mixed infection of more than two species of Eimeria. E. bovis and E. zuernii have been recognized as having high virulency and, thus, these parasites are potential sources of severe coccidiosis and the cause of infections in other cattle. Although additional studies are warranted, these results can be helpful for improving the management and productivity of Madura cattle.  相似文献

Beef cattle production represents the largest cattle population in Thailand. Their productivity is constrained by tick-borne diseases such as babesiosis and theileriosis. In this study, we determined the prevalence of Babesia bigemina, Babesia bovis and Theileria orientalis using polymerase chain reaction (PCR). The genetic markers that were used for detection of the above parasites were sequenced to determine identities and similarity for Babesia spp. and genetic diversity of T. orientalis. Furthermore the risk factors for the occurrence of the above protozoan parasites in beef cattle from northern and northeastern parts of Thailand were assessed. A total of 329 blood samples were collected from beef cattle in 6 provinces. The study revealed that T. orientalis was the most prevalent (30.1%) parasite in beef cattle followed by B. bigemina (13.1%) and B. bovis (5.5%). Overall, 78.7% of the cattle screened were infected with at least one of the above parasites. Co-infection with Babesia spp. and T. orientalis was 30.1%. B. bigemina and T. orientalis were the most prevalent (15.1%) co-infection although triple infection with the three parasites was observed in 3.0% of the samples. Sequencing analysis revealed that B. bigemina RAP1 gene and B. bovis SBP2 gene were conserved among the parasites from different cattle samples. Phylogenetic analysis showed that the T. orientalis MPSP gene from parasites isolated from cattle in north and northeast Thailand was classified into types 5 and 7 as reported previously. Lack of tick control program was the universal risk factor of the occurrence of Babesia spp. and T. orientalis infection in beef cattle in northern and northeastern Thailand. We therefore recommend training of farmers on appropriate tick control strategies and further research on potential vectors for T. orientalis and elucidate the effect of co-infection with Babesia spp. on the pathogenicity of T. orientalis infection on beef in northern and northeastern Thailand.  相似文献

Since regulation of population size requires the existence of one or more density-dependent processes affecting parasite numbers, the literature was examined for evidence of density-dependence in establishment, reproduction and mortality of trichostrongylids of ruminants. Because of differences between the two environments required for completion of the trichostrongylid life-cycle, this evidence was sought in processes occurring within the host, rather than in dung-pats or on pasture. Evidence was found of at least one density-dependent population process in several economically important species of the genera Haemonchus, Ostertagia and Trichostrongylus, although further observations on populations derived from continuous infection at several rates of larval intake are required to substantiate much of this evidence. It is suggested that populations of trichostrongylids of ruminants are regulated at the level of the suprapopulation by density-dependent constraints on egg production of constituent infrepopulations; rates of larval intake may play a central role in coordinating regulation within each ecosystem.  相似文献

Q fever is a widespread zoonosis that is caused by Coxiella burnetii (C. burnetii), and ruminants are identified as the main sources of human infections. Some human cases have been described, but very limited information was available about Q fever in ruminants on Reunion Island, a tropical island in the Indian Ocean. A cross-sectional study was undertaken from March 2011 to August 2012 to assess the Q fever prevalence and to identify the major risk factors of C. burnetii infection in ruminants. A total of 516 ruminants (245 cattle, 137 sheep and 134 goats) belonging to 71 farms and localized in different ecosystems of the island were randomly selected. Samples of blood, vaginal mucus and milk were concomitantly collected from females, and a questionnaire was submitted to the farmers. Ticks from positively detected farms were also collected. The overall seropositivity was 11.8% in cattle, 1.4% in sheep and 13.4% in goats. C. burnetii DNA was detected by PCR in 0.81%, 4.4% and 20.1% in cow, sheep and goat vaginal swabs, respectively. C. burnetii shedding in milk was observed in 1% of cows, 0% in sheep and 4.7% in goats. None of the ticks were detected to be positive for C. burnetii. C. burnetii infection increased when the farm was exposed to prevailing winds and when there were no specific precautions for a visitor before entering the farm, and they decreased when a proper quarantine was set up for any introduction of a new ruminant and when the animals returned to the farm at night. MLVA genotyping confirmed the role of these risk factors in infection.  相似文献

Ostertagia spp. obtained at slaughter from the abomasa. of previously infected sheep and transferred via cannulae into the abomasa of previously worm-free sheep produced severe effects. This was shown with populations of mainly adult parasites given to three sheep and also when mixed populations of 4th stage larvae and adults were given to three other sheep. Although more severe effects were produced when both larval and adult worms were transferred, there were clearly effects when isolates composed predominantly of adults were transferred. Sheep of each group had reductions in food intake and elevations in plasma pepsinogen and gastrin within 24 h of transfer of the parasites. Further increase in plasma pepsinogen and gastrin occurred when abomasal pH rose 5–7 days after infection. Plasma pancreatic polypeptide varied but showed a general tendency to be markedly lower after infection. Ostertagia spp. eggs were detected in the faeces of infected sheep 48 h after transfer. The proportions of adults and inhibited larvae recovered from the abomasa were similar to those in the donor sheep, showing that inhibition, or arrested development of larvae, continued after their transfer to worm free sheep.  相似文献

High copper concentration is toxic for living organisms including humans. Biosorption is a bioremediation technique that can remove copper and other pollutants from aqueous medium and soils, consequently cleaning the environment. The aim of this study was, therefore, to investigate the influence of different copper compounds (Cu(II) as CuCl2; Cu(II) as CuSO4; and Cu(I) as CuCl) on copper bioreduction and biosorption using four copper-resistant bacteria isolated from the rhizosphere of two plants (Avena sativa and Plantago lanceolata) in aqueous matrix. Copper resistance profile, bioreduction, and biosorption after 48 h of incubation were evaluated. The isolates displayed high copper resistance. However, isolate A1 did not grow very well in the CuCl2 and isolate T5 was less resistant to copper in aqueous solutions amended with CuCl (Cu(I)). The best copper source for copper bioreduction and biosorption was CuSO4 and the isolates removed as much as ten times more copper than in aqueous solutions amended with the other copper compounds. Moreover, Cu(I) did not succumb to biosorption, although the microbes were resistant to aqueous solutions of CuCl. In summary, Cu(II) from CuSO4 was furthermost susceptible to bioreduction and biosorption for all isolates. This is an indication that copper contamination of the environment from the use of CuSO4 as an agrochemical is amenable to bioremediation.  相似文献

The objective was to evaluate long-acting moxidectin (Cydectin® 2% LA for sheep) against trichostrongylids in sheep. We performed a blocked, parallel, controlled clinical trial. We included 45 ewe-lambs, allocated into treated (n = 30, 1.0 mg moxidectin per kg bw, subcutaneously at base of ear on D 0) or controls (n = 15). Animals had been naturally infected (pre-treatment geometric mean epg counts: 233 and 249, respectively) and throughout the study grazed at the same pasture. Fortnightly, we collected faecal samples and performed established parasitological techniques (epg counts, coprocultures). We used analysis of covariance for post-treatment results, using pre-treatment counts as covariate, treatment as fixed effect, block as random effect. Geometric mean epg counts in treated animals were 0 up to D 112, 1 on D 126, 4 on D 140 and 13 on D 161. Respective figures for controls were 369–820 up to D 140 and 168 on D 161 (p < 0.05). Persistent efficacy (>90%) was 140 d for Teladorsagia, 119 d for Haemonchus and 115 d for Trichostrongylus. This is the first reported clinical trial of efficacy of long-acting moxidectin in sheep in Europe. Cydectin 2% LA was found to be effective against trichostrongylids of sheep. The particularly long persistent efficacy can provide new possibilities in formulating anthelmintic programs for sheep.  相似文献

Little is known of the prevalence of Cryptosporidium and Giardia parasites in sheep and the genotypes that they harbor, although potentially sheep may contribute significantly to contamination of watersheds. In the present study, conducted in Western Australia, a total of 1,647 sheep fecal samples were screened for the presence of Cryptosporidium and Giardia spp. using microscopy, and a subset (n = 500) were screened by PCR and genotyped. Analysis revealed that although both parasites were detected in a high proportion of samples by PCR (44% and 26% for Giardia and Cryptosporidium spp., respectively), with the exception of one Cryptosporidium hominis isolate, the majority of isolates genotyped are not commonly found in humans. These results suggest that the public health risk of sheep-derived Cryptosporidium and Giardia spp. in catchment areas and effluent may be overestimated and warrant further investigation.  相似文献
