The interaction of dietary protein type and fat level on the body fat-reducing activity of conjugated linoleic acid (CLA) was studied in male rats fed diets containing casein (CAS) or soy protein (SOY) as a protein source with low fat (LF, 6.0% soybean oil) or high fat (HF, 13.0% soybean oil) combinations for 4 weeks. CLA was added at the 1.0% level to all diets. The weight of perirenal adipose tissue tended to be lower in the SOY groups than in the corresponding CAS groups, and the difference between the LF diets was significant. The weight of epididymal adipose tissue showed a similar but insignificant trend. The weight of brown adipose tissue was heaviest on the SOY-HF diet and lowest on two CAS diets, the SOY-LF diet being intermediate. The concentration of serum leptin was lowest on the SOY-LF diet and was significantly lower than that of the corresponding CAS group, but this difference disappeared when the dietary fat level increased. The serum cholesterol-lowering activity of SOY in relation to CAS was reproduced even when CLA was given. Thus the body fat-reducing activity of CLA was most marked when rats were fed the SOY-LF diet. Although the CAS-HF diet increased body fat deposition, the magnitude of the reduction by lowering dietary fat level was more marked than in the case of SOY. These results indicate a complicated interaction of dietary manipulations with the body fat-reducing effect of CLA, but the combination of CLA with the SOY-LF diet appears to be an appropriate approach.  相似文献

The effects of dietary oligosaccharides on the hepatotoxic action of D-galactosamine (GalN) were investigated in this study. Male Wistar rats fed with 20% casein diets containing 10% oligosaccharide or D-galactose (Gal) for 2 weeks were injected with GalN (1,900 mg/kg of body weight), and the plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities and the hepatic glycogen concentration were examined 20 hours after the injection. The plasma AST and ALT activities in experiment 1 for the Gal+neomycin (NEO) group were significantly lower than those for the control (C), NEO, raffinose (RAF)+NEO and galacto-oligosaccharide (GALO)+NEO groups. In experiment 2, these activities were significantly lower in the Gal, Gal+NEO and RAF groups than in the RAF+NEO group when the groups were treated with GalN. On the other hand, in respect of the hepatic glycogen concentration in experiment 1, that of the Gal+NEO group was higher than that of the C, NEO, RAF+NEO or GALO+NEO groups. In experiment 2, this parameter was significantly higher in the Gal, Gal+NEO and RAF groups than in the RAF+NEO group after the GalN treatment. As a result, it is suggested that the GalN-hepatitis-suppressive effects of indigestible oligosaccharides such as RAF or GALO is mediated by the action of intestinal bacteria.  相似文献

The effects of polychlorinated biphenyls (PCB) and dietary protein level on the liver and serum lipid metabolism of rats were studied. Rats were fed an experimental diet containing 7 or 30% casein with or without 0.1 % PCB for 24 days. Dietary PCB increased the level of triglyceride, phospholipid and cholesterol in the liver. The accumulation of triglyceride and cholesterol in liver was markedly increased with a low protein diet. The incorporation of injected 3H2O into liver cholesterol was increased by PCB, but not affected by the dietary level of protein. The incorporation of the tracer into liver fatty acids was not increased by PCB intake. Dietary PCB also raised serum cholesterol and phospholipid, while PCB decreased triglyceride level, especially in rats on low protein diet. In addition, PCB intake clearly raised serum high density lipoprotein and diminished very low density lipoprotein. In the low protein group, PCB markedly repressed the incorporation of 3H2O into serum lipids. The results suggest that the hepatic lipids accumulation by the addition of 0.1 % PCB to a low protein diet might be mainly ascribed to a repression in the transport of triglyceride from liver to blood. Four new polyacetylenic glucosides isolated from the leaf of Jerusalem artichoke (Helianthus tuberosus L.) were characterized as methyl β-D-glucopyranosyl helianthenate C (5), D (6), E (7), and F (8). The absolute stereochemistry of the glucosyloxymethine was also determined to be of R configuration by preparing the relative compounds with Sharpless asymmetric epoxidation as the key step and source of optical activity.  相似文献

The effects of nasunin, a major anthocyanin in eggplant, and its aglycone, delphinidin, on the serum cholesterol conpentration were determined in rats fed with a cholesterol-enriched diet. The serum total cholesterol and HDL-cholesterol concentrations tended to be decreased and increased, respectively, by feeding nasunin and delphinidin, while the fecal excretion of both cholesterol and bile acids tended to be increased by feeding with the anthocyanins. There was no difference between the nasunin and delphinidin activity to decrease serum total cholesterol, nor to increase serum HDL-cholesterol or the fecal excretion of cholesterol and bile acids. These results suggest that the slightly lower serum total cholesterol concentration in rats fed with nasunin and delphinidin may in part be due to inhibition of the intestinal absorption of both cholesterol and bile acids by these anthocyanins, and that the delphinidin moiety of nasunin mainly contributes to this activity.  相似文献

研究了成年雌性黑麂(Muntiacus crinifrons)的肝、小肠和大肠的组织学结构及Ghrelin的分布。采用H.E染色法观察组织学结构,免疫组化PV-9000两步法并结合DAB显色技术确定Ghrelin的分布。结果表明,黑麂的肝组织分为被膜、肝小叶、肝中央静脉、门管区等结构。被膜为浆膜结构,肝小叶不明显。肝细胞以中央静脉为中心,呈放射状排列。肝血窦的形状不规则。肠黏膜上皮为单层柱状上皮,小肠、盲肠和结肠的黏膜肌层很薄,管壁皱襞与肠绒毛等形态在消化道各部也存在差异。免疫组化结果显示肝细胞中有Ghrelin阳性细胞的表达;在肠道,免疫阳性细胞在十二指肠、空肠、回肠、盲肠和直肠的黏膜、黏膜下层和肌层均有分布,尤其在肠绒毛上皮和黏膜下层分布较多。黑麂肝、小肠和大肠结构与哺乳动物基本相似,但无十二指肠腺;Ghrelin阳性细胞在肝、小肠和大肠均有分布,这表明Ghrelin可能对消化有一定的调节作用。  相似文献

Introduction

Early detection, assessment of disease progression, and application of an appropriate therapeutic intervention are all important for the care of patients with type 2 diabetes. Currently, however, there is no simple test for early detection of type 2 diabetes. Established diagnostic tests for the disease including oral glucose tolerance, fasting blood glucose, and hemoglobin A1c are relatively late markers where the disease has already progressed. Since blood is in direct contact with many tissues, we hypothesized that pathological tissue changes are likely to be reflected in proteomic profiles of plasma.

Methods

Mice were reared either on regular chow or a high-fat diet at weaning and several physiological responses (i.e., weight, fasting plasma glucose and insulin, and glucose tolerance) were monitored at regular time intervals. Plasma was collected at regular intervals for proteomic analysis by two-dimensional gel electrophoresis and subsequent mass spectrometry.

Results

Onset of hyperinsulinemia with corresponding glucose intolerance was observed in 2 weeks and fasting blood glucose levels rose significantly after 4 weeks on the high-fat diet. Many proteins were found to exist in multiple forms (isoforms). Levels of some isoforms including plasma retinol binding protein, transthyretin, Apolipoprotein A1, and kininogen showed significant changes as early as 4 weeks which coincided with the very early development of glucose intolerance.

Conclusions

These results show that a proteomic approach to study the development of type 2 diabetes may uncover unknown early post-translationally modified diagnostic and/or therapeutic protein targets.  相似文献

Our aim was to compare and combine 3 nutritional strategies to slow down the age-related loss of muscle mass in healthy old rats: 1) increase protein intake, which is likely to stimulate muscle protein anabolism; 2) use leucine rich, rapidly digested whey proteins as protein source (whey proteins are recognized as the most effective proteins to stimulate muscle protein anabolism). 3) Supplement animals with a mixture of chamomile extract, vitamin E, vitamin D (reducing inflammation and oxidative stress is also effective to improve muscle anabolism). Such comparisons and combinations were never tested before. Nutritional groups were: casein 12% protein, whey 12% protein, whey 18% protein and each of these groups were supplemented or not with polyphenols/antioxidants. During 6 months, we followed changes of weight, food intake, inflammation (plasma fibrinogen and alpha-2-macroglobulin) and body composition (DXA). After 6 months, we measured muscle mass, in vivo and ex-vivo fed and post-absorptive muscle protein synthesis, ex-vivo muscle proteolysis, and oxidative stress parameters (liver and muscle glutathione, SOD and total antioxidant activities, muscle carbonyls and TBARS). We showed that although micronutrient supplementation reduced inflammation and oxidative stress, the only factor that significantly reduced the loss of lean body mass was the increase in whey protein intake, with no detectable effect on muscle protein synthesis, and a tendency to reduce muscle proteolysis. We conclude that in healthy rats, increasing protein intake is an effective way to delay sarcopenia.  相似文献

Niemann-Pick disease, type C1 (NPC1) is a fatal, neurodegenerative disorder for which there is no definitive therapy. In NPC1, a pathological cascade including neuroinflammation, oxidative stress and neuronal apoptosis likely contribute to the clinical phenotype. While the genetic cause of NPC1 is known, we sought to gain a further understanding into the pathophysiology by identifying differentially expressed proteins in Npc1 mutant mouse cerebella. Using two-dimensional gel electrophoresis and mass spectrometry, 77 differentially expressed proteins were identified in Npc1 mutant mice cerebella compared to controls. These include proteins involved in glucose metabolism, detoxification/oxidative stress and Alzheimer disease-related proteins. Furthermore, members of the fatty acid binding protein family, including FABP3, FABP5 and FABP7, were found to have altered expression in the Npc1 mutant cerebellum relative to control. Translating our findings from the murine model to patients, we confirm altered expression of glutathione s-transferase α, superoxide dismutase, and FABP3 in cerebrospinal fluid of NPC1 patients relative to pediatric controls. A subset of NPC1 patients on miglustat, a glycosphingolipid synthesis inhibitor, showed significantly decreased levels of FABP3 compared to patients not on miglustat therapy. This study provides an initial report of dysregulated proteins in NPC1 which will assist with further investigation of NPC1 pathology and facilitate implementation of therapeutic trials.  相似文献

The transxylosylation reaction products of β-xylosidase-1, excreted by Penicillium wortmanni IFO 7237 using β-(1→4)-xylobiose as substrate, have been separated by chromatography on activated charcoal into four fractions, designated as P-1, P-2, P-3, and P-4, respectively. They were further purified by preparative paper chromatography. The characterization and structural analysis were done by measurement of the degree of polymerization (DP) and specific rotation followed by methylation analysis. Moreover, the enzymatic structural analysis of transxylosylation products, with high performance liquid chromatography (HPLC), allowed the confirmation of each structure. The first product, P-1, was β-(1→3)-xylobiose and the second, P-2, was β-(1→4)-xylotriose, but, P-3 was O-β-d-xylopyranosyl-(1→3)-O-β-d-xylopyranosyl-(1→4)-d-xylopyranose or isomeric xylo-triose and P-4 was assumed to be O-β-d-xylopyranosyl-(1→4)-[O-β-d-xylopyranosyl-(1→3)]-O-β-d-xylopyranosyl-(1→4)-d-xylopyranose.  相似文献

This study was undertaken to investigate the response of sucrase (SA), alkaline phosphatase (ALP), and leucine aminopeptidase (LAP) activities localized in the brush border membrane of the small intestine to ingested nutrients. In rats previously meal-fed on carbohydrate-free diets, the maximal increase in SA activity after the administration of a 18 % casein and high sucrose diet (HSD) occurred 24 hr after the beginning of the 12-hr period of HSD-feeding, but ALP and LAP activities reached the maximal levels 12 hr after the HSD administration and then rapidly declined. These changes in three enzyme activities were similar to those after HSD-feeding following a 2-day fast, but those were not observed after giving a carbohydrate-free and high fat diet. The increases in these enzyme activities after giving HSD were not observed entirely after giving HSD containing 0.5% concanavalin A, which was undigestible and preferentially bound mitotically active cells of the small intestine. From these findings, it can be suggested that the adaptive response of these enzyme activities to ingested nutrients, as sucrose and casein, was produced on the luminal surface of the small intestine, especially on the immature cells.  相似文献

It is revealed that administration of exogenous dexamethasone and thyroxin to the 7-8-day old rat pups produces different effect on activity and ratio of membrane and soluble forms of digestive enzymes (lactase, saccharase, maltase, alkaline phosphatase, aminopeptidase M, and glycyl-L-leucine dipeptidase) in jejunum and ileum of animals, whose mothers received different rations during lactation: the standard (control) and low-protein (experiment) ones. Disturbances of the hormonal regulation of the activity and the ratio of membrane and soluble forms of the digestive enzymes in small intestine in early ontogenesis consisted in that the effect of hormones was much less pronounced in the rat pups of the experimental group. An increase of the portion of the soluble form of the enzymes was also revealed. These results indicate a possibility of retardation of maturation of the intestinal digestive enzymes. The most significant difference of these effects in animals of the experimental and control groups was observed for saccharase, maltase, and peptidases.  相似文献

l-Methionine γ-lyase (EC 4.4.1.11) catalyzes α,β-elimination of l-2-amino-3-(N-methylamino)propionic acid and l-2-amino-3-(N-hydroxyethylamino)propionic acid to yield pyruvate, ammonia, and the corresponding amines. These amino acids also undergo the enzymatic β-replacement reaction with thiols to produce the corresponding S-substituted cysteines. Thus, l-methionine γ-lyase cleaves a C-N bond in addition to C-S, C-Se, and C-O bonds at the β position of amino acids by elimination and replacement reactions. A linear relationship between the reactivity, (log(Vmax/Km) and the pKa value of the conjugated acid of the leaving group has been found for Se-methyl-l-selenocysteine, S-methyl-l-cysteine, and O-methyl-l-serine. However, l-2-amino-3-(N-methylamino)propionic acid has shown lower reactivity than that expected from the pKa value of methylammonium ions.  相似文献

The incorporation of 14C of acetate-1-14C into the lipids of the liver and carcass, and the changes in the concentrations of nucleotides and citric acid in the liver were studied in the rats fed individual nutrients; starch, casein or corn oil. And the metabolism of citric acid-1,5-14C was also investigated after the feeding of nutrients. Lipogenesis in the liver and carcass was more markedly stimulated with starch than with casein or corn oil. In the liver of rats fed starch, the concentration of ATP doubled and that of citric acid was one-half of those with casein or corn oil, respectively. And the conversion of citric acid to carbon dioxide and lipids was stimulated with starch.  相似文献

Formaldehyde dehydrogenase from Pseudomonas putida C-83 was found to contain 7 halfcystine residues per subunit monomer, as checked by the method of performic acid oxidation. Approximately 7 sulfhydryl groups per subunit monomer were titrated with 5,5′-dithiobis(2-nitrobenzoic acid) (DTNB) after denaturation with 8 m urea. In the native enzyme, modification of three sulfhydryl groups per subunit with p-chloromercuribenzoate (PCMB) led to the complete loss of enzyme actiyities for both formaldehyde and n-butanol. Hydrogen-peroxide competitively inhibited the enzyme activity for formaldehyde, while it was only slightly inhibitory to the activity for n-butanol. Both formaldehyde and hydrogen-peroxide protected one sulfhydryl group per subunit monomer from modification with PCMB. Moreover, hydrogen-peroxide was hardly reactive to the enzyme which was preincubated with formaldehyde.

From these observations, we conclude that one of three PCMB-reactive sulfhydryl groups is essential for the binding of formaldehyde, and hydrogen-peroxide modifies this sulfhydryl group.  相似文献

A significant decrease of protein content in epithelial, stromal, and muscular-serosal layers of jejunum and ileum, especially in aged animals, is revealed in rats of different age groups (young, mature, aged) after 10-day-long protein deprivation. The responses of peptide hydrolases (aminopeptidase M and glycylleucine dipeptidase) were different. There were, as a rule, no changes of these enzyme activities in small intestine of young rats, except for decreased activity of aminopeptidase M in stromal layer of jejunum and increased activity of both peptide hydrolases in epithelial layer of ileum. In mature animals, increased activities of these enzymes also was observed in the epithelial layer of jejunum and ileum and in the stromal layer of ileum in comparison with the rats receiving a full-value nutrition. In aged rats, a much more pronounced rise of these enzyme activities in the layers of small intestine was revealed after protein starvation in comparison with young and mature rats. Probably, such increase of the peptide hydrolase activities in the layers of small intestine after protein deprivation can be considered as an adaptive-protective reaction of the organism in response to formation of significant amounts of low-molecular peptides as a result of protein catabolism  相似文献
