Summary Mutations at the Adh1 locus in maize were selected from plants infected with barley stripe mosaic virus (BSMV). Pollen from the infected inbred line 1s2p, which is homozygous for Adh1-S (abbreviated S), Adh2-P, c and r was treated with allyl alcohol and applied to silks of a tester stock homozygous for Adh1-F, Adh2-N, C and R. From these pollinations 356 kernels arose on the F1 ears. Of these eight showed no activity of the S allele in scutellar samples while two exhibited low levels. Five of the putative mutant kernels germinated and two of these contained the contamination markers Adh2-P, c and r. The newly arisen mutations were designated S5446 and S5453. S5453 exhibited an abnormally low level of ADH activity in the F1 scutellum. In the F2 generation the mutant reverted at a high frequency with only about 5% of the S5453 alleles expressing low levels. DNA blotting and hybridization analyses showed no alterations in the restriction patterns of S5453 when compared to the progenitor S allele. S5446 which exhibited no ADH activity in the F1 scutellum is unstable in the pollen; reversion frequencies approaching 10-2 were observed in samples from some plants. Restriction digestion patterns of DNA from this mutant revealed the presence of a 3.3 kb insertion at Adh. The insert does not appear to contain sequences homologous to the BSMV genome but rigorous analyses remain to be carried out. It is hypothesized that BSMV infection may mobilize endogenous but dormant transposable elements in maize.  相似文献

The ADH allozyme pattern was tested in seeds of 1553 varieties of the world collection of safflower (Carthamus tinctorius L.) and 36 collections belonging to 14 wild species of the genus Carthamus L. with different chromosome numbers (n=10, 11, 12, 22, and 32). Two genes, Adh 1 and Adh 2, have been identified. The Adh 1 locus controls the allozyme bands found in the faster-moving anodal zone, and the Adh 2 gene controls the cathodal band. A third group of bands which migrates slowly toward the anode and stains weakly is probably interaction products of the two genes. Two codominant alleles Adh 1 S and Adh 1 F , specifying allozymes with different migration rates in the fast-moving anodal zone, were found in cultivated safflower. The frequency of the Adh 1 F allele was very low. A third homologous allele, Adh 1 T , was present only in the polyploid wild species. The Adh 2 was stable, without any variation in migration rate. In addition to the variation in migration rates, there was also variation in activity levels of the products of both the Adh 1 and Adh 2 genes. The contribution of this study to our understanding of the origin of the polyploid species C. lanatus, C. baeticus, and C. turkestanicus is discussed.This research has been financed in part by a grant made to A. Two Drosophila melanogaster strains, each heterozygous for fast and slow alleles at the Adh locus, and each having balanced second chromosomes, were found to differ in the apparent thermostability of the slow allozyme. The two strains were crossed, and F1heterozygotes were separated on the basis of the origin of the slow allele. After electrophoresis, the cellulose acetate strips were treated 1 1/2 min at 35 C. The putatively more sensitive allozyme showed a strikingly greater response to heat. These findings further support the conclusion that electrophoretically cryptic allelic differences exist which are expressed in thermostability differences. Further application of this approach has revealed one similar sensitive slow allozyme and three cases of a relatively resistant fast ADH allozyme in wild-caught flies.This work was supported by NIH Grant GM18967-02-03.  相似文献

In the natural populations +Tüb, +Prov, and +Rov, similar Adh F allele frequencies occur (q F=0.11, 0.18, and 0.08, respectively). However, there is a discrepancy in that the Adh F allele in +Tüb is closely linked to the lethal factor 1(2)Stm, which reduces relative fitness of the F phenotype to zero. In spite of this, polymorphism is maintained also in +Tüb, because the heterozygotes are superior to the homozygous S type (relative fitness=0.88). Under laboratory culture conditions, in +Tüb the relative fitness of the S genotype further decreases to 0.6. After outcrossing the lethal factor, relative fitnesses for S, FS, and F become 0.6, 1, and 0.48, respectively, implying that fitness for S remains the same. Relative values for S, FS, and F in +Prov, not affected by the lethal factor, are calculated by the maximum average fitness method to be 1, 1.2, and 0.2 under the assumption that heterozygous FS are similarly superior to S as in the natural +Tüb population and all allele frequencies found are stable equilibrium values.  相似文献

Keeping Drosophila cultures at 28 C results in elimination of all minor multiple ADH bands, thought to be due to conformational change. Thus in diploid and triploid adults heterozygous for the Adh F and Adh Salleles, relative staining intensities are found for the three bands which were in conformity with the assumption that both alleles are equally expressed. Among all polymorphic strains derived from natural Central European and Mediterranean populations, the strain +Tüb is unique in that its Adh Fallele is closely linked to a new recessive lethal factor, named 1(2)Stm. All Adh F 1/AdhF 1 pupae are unable to emerge, and die. The lethal effect is obvious 50 hr earlier by retarded eye, bristle, and body wall pigmentation. Although all pupae of the phenotype F die, Adh F allele frequency scarcely seems to be lowered in this natural population.  相似文献

Summary The data of this paper partially define a two-gene, differentially inducible enzyme system in maize. Four major conclusions are drawn. (1) Anaerobic treatment results in the simultaneous expression of two unlinked Adh genes, Adh1 and Adh2. Adh2 is scarcely expressed in the uninduced root and negligibly expressed in the quiescent embryo. (2) The zero-order rate of anaerobic ADH protein induction reflects the zero-order rate of ADH synthesis throughout the induction; all active ADH isozymes display negligible turnover during the induction. (3) The auxin, 2,4-D, overcomes repression by air. The aerobic induction rates are also zero-order with negligible turnover for any isozyme. (4) The ratio of Adh1-Adh2 expression is drastically altered dependent on the mode of induction. The level of this differential regulatory phenomenon is before or at the assembly of ADH polypeptides into enzymologically or antigenically active dimers.  相似文献

Summary The analysis of the individual parts of the Triticum aestivum L. kernel yields a total of 11 peroxidase isozymes: m, n, a, c, d1, d, d2, e, f, g and h (in order from faster to slower migration). Isozymes a, c and d are found in the endosperm (Ed) and seed coats (C), while m, n, d1, d2, e, f, g and h are peculiar to the embryo and scutellum (E + S). The use of the nullitetrasomic and ditellosomic series of Chinese Spring wheat allows peroxidase isozymes to be associated with specific chromosome arms. Isozymes a, c and d (Ed) are associated with chromosome arms 7DS, 4BL and 7AS; whereas isozymes m, d2, e and f are associated with chromosome arms 3DS, 3BL, 3DL and 3DL, respecitvely. Thus, the E + S isozymes are associated with homoeology group 3 and the Ed isozymes with homoeology groups 7 (a and d isozymes) or 4 (c isozymes).  相似文献

Lines homozygous for the Adh S and Adh F alleles were extracted from a French and an African natural population of D. melanogaster. The lines from each geographic region were then crossed and the Mendelian F2 constituted the first generation subjected to selection for an increase in adult survival in the presence of 2% ethanol.The responses to selection were similar in the two strains, in spite of their different ethanol tolerance. In less than 10 generations, life span with ethanol increased from about 7 to more than 12 days, with realized heritabilities of 0.14 and 0.18. This extension of longevity was also observed with other concentrations of ethanol but not with water alone. The increase in life span in presence of alcohol appears to result from unknown metabolic processes, which are not obligatorily related to the capacity of the flies to tolerate starvation, nor to their size, their lipid content of their ethanol tolerance. In the two lines, however, the Adh S allele was quickly eliminated, suggesting a selective advantage for the F allele.  相似文献

The distribution of the F420-reactive and F420-nonreactive hydrogenases from the methylotrophic Methanosarcina strain Gö1 indicated a membrane association of the F420-nonreactive enzyme. The membrane-bound F420-nonreactive hydrogenase was purified 42-fold to electrophoretic homogeneity with a yield of 26.7%. The enzyme had a specific activity of 359 mol H2 oxidized · min-1 · mg protein-1. The purification procedure involved dispersion of the membrane fraction with the detergent Chaps followed by anion exchange, hydrophobic and hydroxylapatite chromatography. The aerobically prepared enzyme had to be reactivated anaerobically. Maximal activity was observed at 80°C. The molecular mass as determined by native gel electrophoresis and gel filtration was 77000 and 79000, respectively. SDS gel electrophoresis revealed two polypeptides with molecular masses of 60000 and 40000 indicating a 1:1 stoichiometry. The purified enzyme contained 13.3 mol S2-, 15.1 mol Fe and 0.8 mol Ni/mol enzyme. Flavins were not detected. The amino acid sequence of the N-termini of the subunits showed a higher degree of homology to cubacterial uptake-hydrogenases than to F420-dependent hydrogenases from other methanogenic bacteria. Four Drosophila melanogaster strains, each homozygous for one of the two major ADH aliozymes, Fast and Slow (Adh F1, Adh S , Adh F2 and Adh S2) were used to study the interaction of the Adh locus with ethanol and temperature. The separate and especially the combined effects of these two parameters allow the conclusion that the Adh locus of D. melanogaster intervenes in the adaptation process through the heat shock protein system.  相似文献

Photon yields of oxygen evolution at saturating CO2 were determined for 44 species of vascular plants, representing widely diverse taxa, habitats, life forms and growth conditions. The photonyield values on the basis of absorbed light ( a) were remarkably constant among plants possessing the same pathway of photosynthetic CO2 fixation, provided the plants had not been subjected to environmental stress. The mean a value ±SE for 37 C3 species was 0.106±0.001 O2·photon-1. The five C4 species exhibited lower photon yields and greater variation than the C3 species ( a=0.0692±0.004). The a values for the two Crassulaceanacid-metabolism species were similar to those of C3 species. Leaf chlorophyll content had little influence on a over the range found in normal, healthy leaves. Chlorophyll fluorescence characteristics at 77 K were determined for the same leaves as used for the photon-yield measurements. Considerable variation in fluorescence emission both at 692 nm and at 734 nm, was found 1) among the different species; 2) between the upper and lower surfaces of the same leaves; and 3) between sun and shade leaves of the same species. By contrast, the ratio of variable to maximum fluorescence emission at 692 nm (Fv/FM, 692) remained remarkably constant (The mean value for the C3 species was 0.832±0.004). High-light treatments of shade leaves resulted in a reduction in both a and the Fv/FM, 692 ratio. The extent of the reductions increased with time of exposure to bright light. A linear relationship was obtained when a was plotted against Fv/FM, 692. The results show that determinations of the photon yield of O2 evolution and the Fv/FM, 692 ratio can serve as excellent quantitative measures of photoinhibition of overall photosynthetic energy-conversion system and of photochemistry of photosystem II, respectively. A rapid and reproducible enzymatic rate assay for the quantitative determination of the concentration of active sites is presented for the alleloenzymes AdhS and AdhF from Drosophila melanogaster. Using this procedure the turnover numbers as catalytic-center activities were found to be 12.2 sec–1 for AdhF and 3.4 sec–1 for AdhS with secondary alcohols. This showed a slower dissociation of the coenzyme from the binary enzyme-NADH complex with AdhS and hence a stronger binding of NADH to this alleloenzyme. With ethanol, the catalytic-center activity was 1.4 sec–1 for AdhS and 2.8 sec–1 for AdhF, and hence the single amino acid mutation distinguishing the two alleloenzymes also affected hydride transfer.  相似文献

Striped mutants of Petunia were isolated after -ray treatment of zygotes and from the progeny of a haploid plant. This striped phenotype appeared to be associated with a telocentric translocation in which one arm of chromosome I was transferred to the end of the long arm of chromosome II.The phenotype and the breeding behaviour of the striped plants are explained on the basis of a mechanism similar to the breakage-fusion-bridge cycle described earlier in maize endosperm.The translocation is marked by the dominant allele P1 responsible for the synthesis of petunidin in flowers. Chromatium vinosum DSM 185 was grown in continuous culture at a constant dilution rate of 0.071 h-1 with sulfide as the only electron donor. The organism was subjected to conditions ranging from phosphate limitation (S R-phosphate=2.7 M and S R-sulfide=1.8 mM) to sulfide limitation (S R-phosphate=86 M and S R-sulfide=1.8 mM). At values of S R-phosphate below 7.5 M the culture was washed out, whereas S R-phosphate above this value resulted in steady states. The saturation constant (K ) for growth on phosphate was estimated to be between 2.6 and 4.1 M. The specific phosphorus content of the cells increased from 0.30 to 0.85 mol P mg-1 protein with increasing S R-phosphate. The specific rate of phosphate uptake increased with increasing S R-phosphate, and displayed a non-hyperbolic saturation relationship with respect to the concentration of phosphate in the inflowing medium. Approximation of a hyperbolic saturation function yielded a maximum uptake rate (V max) of 85 nmol P mg-1 protein h-1, and a saturation constant for uptake (K t) of 0.7 M. Summary In this paper, the results of the preceding electrophysiological study of sodium-alanine cotransport in pancreatic acinar cells are compared with kinetic models. Two different types of transport mechanisms are considered. In the simultaneous mechanism the cotransporterC forms a ternary complexNCS with Na+ and the substrateS; coupled transport of Na+ andS involves a conformational transition between statesNCS andNCS with inward- and outward-facing binding sites. In the consecutive (or ping-pong) mechanism, formation of a ternary complex is not required; coupled transport occurs by an alternating sequence of association-dissociation steps and conformational transitions. It is shown that the experimentally observed alanine- and sodium-concentration dependence of transport rates is consistent with the predictions of the simultaneous model, but incompatible with the consecutive mechanism. Assuming that the association-dissociation reactions are not rate-limiting, a number of kinetic parameters of the simultaneous model can be estimated from the experimental results. The equilibrium dissociation constants of Na+ and alanine at the extracellular side are determined to beK N <-64mm andK S <-18mm. Furthermore, the ratioK N /K N S of the dissociation constants of Na+ from the binary (NC) and the ternary complex (NCS) at the extracellular side is estimated to be <-6. This indicates that the binding sequence of Na+ andS to the transporter is not ordered. The current-voltage behavior of the transporter is analyzed in terms of charge translocations associated with the single-reaction steps. The observed voltage-dependence of the half-saturation concentration of sodium is consistent with the assumption that a Na+ ion that migrates from the extracellular medium to the binding site has to traverse part of the transmembrane voltage.  相似文献

The Adh and αGpdh allozyme loci (both located on the second chromosome) showed considerable fluctuations in allele frequencies in a seminatural population of Drosophila melanogaster during 1972–97. Both long-term and short-term fluctuations were observed. The short-term fluctuations occurred within almost all years and comparison of allele frequencies between winters and summers showed significantly higher AdhS (P < 0.001) and αGpdhF (P < 0.01) allele frequencies in summers. Frequencies of these alleles were significantly positively correlated with environmental temperature, suggesting the adaptive significance of these allozyme polymorphisms. Frequency changes of the Odh locus (located on the third chromosome) showed no seasonal pattern and were not correlated with environmental temperature. Almost all short-term and long-term increases in AdhS frequency were accompanied by a corresponding decrease in αGpdhS frequency (r = –0.82, P < 0.001) and vice versa. Further analysis showed that gametic disequilibria between the Adh and αGpdh loci, which frequently occurred, were due to the presence of inversion In(2L)t located on the same chromosome arm and In(2L)t frequencies were positively correlated with environmental temperature. Gametic disequilibria between Adh and Odh and between Odh and αGpdh were hardly observed. Because In(2L)t is exclusively associated with the AdhS/αGpdhF allele combination, the observed correlated response in Adh/αGpdh allele frequencies is (at least partly) explained by hitchhiking effects with In(2L)t. This means that the adaptive value of the allozyme polymorphisms has been overestimated by ignoring In(2L)t polymorphism. Fluctuations in Adh allele frequencies are fully explained by selection on In(2L)t polymorphism, whereas we have shown that αGpdh frequency fluctuations are only partly explained by chromosomal hitchhiking, indicating the presence of selective differences among αGpdh genotypes in relation with temperature and independent of In(2L)t. Frequency fluctuations of αGpdh and In(2L)t are consistent with their latitudinal distributions, assuming that temperature is the main environmental factor varying with latitude that causes directly or indirectly these frequency distributions. However, the results of the tropical greenhouse population show no correlation of Adh (independent of In(2L)t) and Odh allele frequencies with environmental temperature, which may indicate that the latitudinal distribution in allele frequencies for these loci is not the result of selection on the F/S polymorphism in a direct way.  相似文献

The properties of the six common phenotypes (F1, S1, I1, F1S1, S1I1, F1I1) of placental alkaline phosphatase have been studied by DEAE-Sephadex chromatography, gel filtration, two-dimensional paper-starch gel and starch block electrophoresis. The rapidly migrating component (component I) had the same molecular weight (118,000) in all phenotypes studied but different negative charge (F1>I1>S1). The fastest of the slow-moving components (component II) had a molecular weight of 130,000 and a higher absolute negative charge, but slightly lower net negative charge than I. The very slow-moving components (II) were usually composed of two major and several minor components. The major components had a higher absolute negative charge but a lower net negative charge than II. All II components had a molecular weight of over 200,000. It is suggested that II is an enzyme bound to a small molecule, while II components are aggregates of enzyme with various substances or with itself.This work was supported in part by U.S. Public Health Service Grant HD02552. Summary Cytogenetic studies were made with particular emphasis on the sex-determining mechanism in Rumex acetosella (6 x = 42) and its hybrids (F 1, F 2, BC 1 and BC 2) with R. hastatulus (synthetic 4 x = 16 = 4 A +4 X = and 4 x = 18 = 4 A + 2 (X Y 1 Y 2) = ). Rumex acetosella was almost strictly dioecious with 5050 male and female. Breeding tests revealed that the males were heterogametic. The longest chromosomes (S), usually two, are the sex chromosomes of this hexaploid species. The S chromosomes are homomorphic in both male and female. The sex chromosome: autosome ratios, and the strong epistatic male effect of the S M chromosome in the polyploid dioecious species and in the hybrids, are evidence of an X/Y Melandrium type sex-determining mechanism controlled by a single pair of homomorphic sex chromosomes. Thus, the sex chromosome formula of the males was S F S M and that of females was S F S F. The present approach is a new method for resolving the sex-determining mechanism in a dioecious species.  相似文献

Two unlinked genes, Adh 1 and Adh 2, control the production of alcohol dehydrogenase (ADH) in seeds of the annual sunflower (Helianthus annuus). Each gene is polymorphic, having F and S alleles. Starch gel electrophoretic zymograms of the four possible double homozygotes have three bands, representing two homodimers and an intermediately migrating intergenic isozyme. Zymograms of double heterozygotes consist of nine bands produced by ten isozymes: six intragenics and four intergenics, two of which are coincident. Results of dissociation-recombination (D-R) experiments are reported which demonstrate the subunit composition of the intergenic isozymes, thus supporting the relationships suggested by genetic studies. Densitometric tracings of the zymogram of a cleared gel and measurements of activities of homodimer isozymes eluted from gels following D-R of an intergenic isozyme showed that the Adh 2 isozymes were more than twice as active as those of Adh 1. Measurements of activities of crude extracts from the four possible double homozygous genotypes indicated that the seeds of the genotype Adh 1 F /Adh 1 F , Adh 2 S /Adh 2 S produced more activity than the other three. This genotype is the most common one found in wild and cultivated stocks. Isozymes eluted following electrophoresis of the same extracts had averages of 19%, 70%, and 11% of total activity contributed by the Adh 1, Adh2, and intergenic isozymes, respectively. A simple but efficient method of isozyme elution from starch gels is described which resulted in nearly full expected recovery (approximately 46%) of the ADH activity in the applied sample.Supported by Graduate School and BioMed grants and by NSF Grant GB35853.  相似文献
