Summary Mitotic index (MI) was determined in the cap, epidermis, cortex and stele at successive intervals along the apical mm of lateral roots of various specific lengths. This parameter was found to decrease basally along roots of each length examined and to decline as the lateral root elongated. Moreover, MI was different in the various tissues investigated. These results have been discussed with respect to changes in the other parameters of cell proliferation and to root growth by elongation. It is suggested on the basis of the data obtained, and other results in the literature, that epidermal initial cells could be differentiated from those of the cortex and stele before the quiescent centre appeared in these roots, but the initial cells of the latter two tissues were indistinguishable until after the quiescent centre began to form.  相似文献

Nine accessions ofVicia narbonensis, considered to be the wild progenitor of faba bean (Vicia faba), were investigated to ascertain the nature and extent of intraspecific karyotypic polymorphism. The chromosome complements resolved into four distinct types (A, B, C, D), and the meiotic data of F1 hybrids (A × B, B × C, A × C) revealed that alteration in chromosome morphology is the result of segmental interchanges. The interchange complexes indicate that the parents differ from each other by 1 to 2 interchanges. It is also evident that karyotype B, and not A as previously reported, is the normal karyotype of the species, and A and C are single homozygotes for unequal interchange. The comparative karyomorphology of the parents and the hybrids, and of two interchange heterozygotes of four chromosomes each in F1 hybrids of A × C shows that the chromosomes involved in the single interchange homozygotes (A, C) are not common and the breaks in both interchanges occurred in short and long arms of the involved chromosomes. Identification of the interchanged chromosomes in the complements and the frequency of ring and chain quadrivalents in the heterozygotes enabled location of the breakpoints. The present results provide probably the first example indicating that interchange homozygosity (A) is not only firmly established but also has enabled the species to spread further by adapting to a wide range of habitats. — The genetic relationships between A and D are very different. All seven chromosome pairs in D could be distinguished from A, and for that matter, B and C as well. From the meiotic pairing properties it is also amply clear that genome D is well differentiated from A and possibly B, and C, and deserves special status.  相似文献

To reveal the behavior of silver stainable material localized mainly in the nucleoli and nucleolar organizing regions (NORs), the somatic cells ofVicia faba were investigated by silver staining throughout the mitotic cell cycle. Nucleoli of interphase and early prophase nuclei were darkly stained. From late prophase to anaphase the secondary constrictions were discriminated as silver stained NORs and many silver grains appeared throughout the cytoplasm. At late prophase the NOR condensed at the same rate as the chromosome arm. Small spherical bodies and two new nucleoli appeared in telophase nuclei and at the same time the cytoplasmic grains disappeared. On the basis of the above observations on the silver stainable material during each mitotic phase, the behavior of silver stainable material is interpreted.  相似文献

Age of explant and six different media were evaluated with the objective of regenerating higher numbers of interspecific hybrids between sexual and apomictic Brachiaria. Immature embryos of 7–8, 9–10 and 11–12 days after pollination (DAP), from artificial hybridization between Brachiaria ruziziensis (R) as female parent, and B. brizantha (B) or B. decumbens (D) as male parent, were cultured in modified MS media (M4) – supplemented with different combinations of growth regulators and vitamins. Embryos cultured 9–12 DAP showed high percentage (85–100%) of germination for all the crosses examined. Germination and survival rates varied according to accessions within crosses. Six different media (all modified MS with different growth regulators and vitamins) were tested with the objective of inducing multiple shoots from 7 to 10 DAP embryos, from crosses between R × B. The media M1, supplemented with Kinetin (13.94 μM) and NAA (5.37 μM), and media M3, supplemented with BA (4.44 μM and IAA 2.85 μM), regenerated adventitious shoots and calli about 30–40 days after inoculation. The highest multiplication rate observed was 2.85 shoots per explant in media M1, 60–70 days after culturing. Two other media, M6, supplemented with 2,4-D (13.57 μM) and M2, with 2,4-D (9.05 μM) and BA (8.87 μM) exclusively induced the formation of calli. The described protocols proved to be efficient in regenerating healthy seedlings from immature embryos of interspecific hybrids in Brachiaria. A rapid regeneration protocol for proembryos of Phaseolus angustissimus as young as 1 day after pollination (DAP) involving pod culture for 1 week followed by embryo culture for 2 weeks and embryo germination for 1 or 2 weeks is provided. Optimization of the media was conducted with pods collected 3 DAP. The best pod culture medium was composed of basal medium [(Phillips and Collins 1979) salts with (Geerts et al. 2001) vitamins], 1000 mg l−1 glutamine, 1000 mg l−1 casein hydrolysate, 3% sucrose and 0.5% agar. Embryo culture medium consisted of basal medium with 500 mg l−1 glutamine, 250 mg l−1 casein hydrolysate, 1.9 μM ABA, 3% sucrose and 0.5% bacto-agar. Embryos developed into plantlets on germination medium containing basal medium with 0.25 μM BA, 3% sucrose and 0.7% bacto-agar. Fertile, normal plants were recovered from direct embryogenesis and from micrografted embryo-derived shoots. Embryos obtained from pods collected 3 DAP regenerated plantlets at a rate of 29.3%, while embryos from pods collected 2 DAP and 1 DAP regenerated at rates of 20.2 and 4%, respectively. A second accession of P. angustissimusregenerated at a rate of 26.2%. Using this 5-week protocol for P. vulgaris resulted in a plantlet regeneration rate of 12.5%.  相似文献

Studies of ultrastructure and ATPase localization of the mature embryo sac in Vicia faba L. show that the egg cell has no cell wall at thechalazal end, it has a chalazally located nucleus and a large micropylar vacuole. There are many nuclear pores in the nuclear membrane. The cytoplasm is restricted around the nucleus. Dictyosome and mitochondria are few. There are some starch grains and lipid grains in the egg cytoplasm. There are no obvious differences between two synergids. No cell wall is seen at the chalazal end either, but there are some vesicles which project to vacuole of the central cell and fuse with its vacuolar membrane. Plasmodesmata connections occur within the synergid wall where it is adjacent to the central cell. The synergid has a micropylarly located nucleus and a chalazal vacuole, the nucleus is irregularly shaped. The synergid cytoplasm is rich in organelles. The filiform aparatus is of relatively heterogeneous structure. The central cell is occupied by a large vacuole and its cytoplasm is confined to a thin layer along the empryo sac wall, but is rich in various organelles, starch grains and lipid bodies. Nucleolar vacuoles are often present two polar nuclei. The nuclear membranes of two polar nuclei have partly fused. ATPase reactive product was located obviously at the endoplasmic reticulum in cytoplasm of the egg cell and central cell. The embryo sac wall consists of different density of osmiophilic layer. There are some wall ingrowths in chalazal region of the embryo sac. The long-shaped and cuneate cells of chalazal region are peculiar. Special tracks of ATPase reactive products are visible at their intercellular space which may be related to transportation of nutrients.  相似文献

Summary DNA sequences reassociating within a Cot value of 1.8×10–1 and those producing a light satellite in a CsCl density gradient were isolated fromVicia faba DNA and hybridizedin situ on squashes of roots of the same species. Silver grains were seen to be scattered over both the interphase nuclei and the metaphase chromosomes after hybridization with fast renaturing DNA sequences, indicating these are fairly regularly interspersed in theV. faba genome. Clustered labeling occurred after hybridization with satellite DNA sequences, indicating these are clustered in the genome. The localization of satellite DNA in chromosomes appeared to correspond closely to the position of the bright bands detectable after staining with quinacrine mustard. After hybridization with both DNA probes, labeling intensity over the nuclei of meristematic cells was higher than that over the nuclei of differentiating and/or differentiated cells. These results are discussed in relation to the structure of the cell nucleus, the mechanism of quinacrine banding and to previous data suggesting underrepresentation of nuclear repeated DNA sequences in differentiatingV. faba root cells.  相似文献

本文对34种基因型的小麦未成熟胚在离体培养中的反应进行了比较。结果表明,94%的供试基因型愈伤组织诱导率都可达到80%以上,若排除供体植株环境条件的不同和接种过程中的人为因素可能造成的影响,不同基因型的愈伤组织诱导率看来没有根本的差异。愈伤组织分化率因基因型的不同变动在0—60%之间,平均为32.7%。虽然同一基因型的盾片愈伤组织分化率在不同年份中有所不同,但是愈伤组织是否具有再生能力?看来是个稳定的遗传性状。因此小麦未成熟胚对愈伤组织诱导的反应和愈伤组织的再生能力可能具有不同的遗传基础。本文的结果还表明,虽然最适于培养的未成熟胚的大小为1毫米左右,伹小至0.3毫米的未成熱胚仍能以几乎100%的频率形成愈伤组织,60%左右的愈伤组织能分化出再生檀株,只是所需的时间比1毫米左右的胚较长。  相似文献

过硬单胚培养十一项特殊的技术,运用此技术可以从果蝇单个原肠胚(同一基因型)中提取细胞进行体外培养,科观察到各种细胞的分化过程,从而能研究导致发生异常或致死突变的机制。通过反复摸索实验,我们在本实验室的条件下,建立了黑腹果蝇(D. melanogaster)单胚培养技术,并对野生型(Canton special, CS.)果蝇的单胚细胞进行体外培养,观察到了各种细胞,如肌肉细胞、神经细胞、脂肪细胞、以及成虫盘等的分化及其发生规律性,为今后研究突变奠定了基础。实验过程中,我们采用了载玻片法和培养皿法。两种方法各有特点及利弊,本文一并进行讨论。Abstract:We can culture the cells of single gastrula (the gene type of cells is same) in vitro,we now have cstablished the technique of single embryo culture in Drosophila.We have seen the differentiated state of myocytes, neurocytes,hemocyte cells,epithelical cells and imagical disk by this technique.  相似文献

以EMS的处理并结合组织培养技术成功地获得胚致死突变的纯合再生植株。该植株生长发育正常,除种子无发芽能力外,纯合突变体的一切性状均与亲本品种表现一致。观察到胚败育的各种表现:(1)仅具有一个球形胚。(2)完全没有胚器的分化。(3)仅具胚根的分化而无胚芽的分化。(4)胚芽分化不完全。(5)胚芽与胚根之间没有输导组织相连接或者输导组织发育不完全等等。(纯合突变体×正常品种)杂种当代的种子(F1)发芽正常,而由F1及R1植株上所产生的种子(F2及R2)约有3/4具发芽能力,而1/4无发芽能力。统计分析的结果表明胚致死突变受隐性单基因控制。据我们所知,获得胚致死突变纯合体的成熟植株,本研究乃是首例报告,至少在水稻上是如此。在以利用无融合生殖之固定杂种优势的“一系法”杂交水稻生产的设想中,胚致死突变可作为胚乳的提供者而加以利用。  相似文献

Summary Two dimensional gel electrophoresis of total cell protein extracts from not expanded, and primary leaves, petioles, and roots ofVicia faba resulted in four actin isoforms at 43 kDa with pI values from 5.9 to 6.05. In contrast to root extracts, in all leaf extracts an additional immunoreactive polypeptide with a molecular mass of 51 kDa and pI 5.75 was detected. This polypeptide was present in high amounts in protein extracts of purified chloroplasts, whereas no actin isoform at 43 kDa could be demonstrated. Compared to the tissue extracts, two actin isoforms at 43 kDa with pI values of 5.9 and 6.0 were enriched, when purified plasma membranes and the membranous fraction of vacuoles were analysed. In contrast, the soluble protein fraction of the plasma membrane preparation contained only two isoactins with pI values of 5.95 and 6.05 and a molecular mass of 43 kDa. Summary The arrangement of chromosome arms in metaphases and anaphases has been studied inVicia faba root meristem cells. During metaphase, the long chromosome arms are aligned parallel to the spindle axis. As a consequence, at the onset of anaphase, one chromatid can move straight ahead to the spindle pole whereas the other has to invert its orientation. Specially in narrow cells it has been observed frequently that some chromatids move in a reverse orientation to the pole, i.e., they move telomere-first instead of centromere-first. This behaviour results in a chromatid which protrudes beyond the main group of late anaphase or telophase chromatids. It is dicussed that the most likely explanation for the phenomenon is that in narrow cells chromatid behaviour is influenced by steric hindrance by the tightly packed surrounding chromatids and microtubules. When there is insufficient room, some chromatids are unable to make the required U-turn. Under such conditions the kinetochore of a non-inverted chromatid pulls the chromatid in a reverse orientation to the pole. An alternative explanation, i.e., protruding chromatids being the result of a neocentric activity at the telomere end of a reverse-directed chromatid or the lateral associations of spindle microtubules, failed to find support by electron microscopical studies.  相似文献

Three different culture media have been examined for their ability to support growth in culture of embryos of two pea lines near-isogenic except for the r-locus. Embryos showed a greater increase in fresh weight on a medium containing 10% sucrose and a high level of a mixture of amino acids than on either one containing an equivalent amount of glutamine as the sole nitrogen source or one containing both inorganic nitrogen and a low level of glutamine. Small embryos (up to 10 mg fresh weight) showed the greatest relative increase in fresh weight. Decreasing the osmotic pressure of an agar medium by lowering the sucrose content to 2% and reducing the concentration of amino acids induced precocious germination. Shoot growth was more sensitive than root growth to increasing sucrose concentrations and optimum development was obtained when embryos were cultured in liquid culture at a high osmotic pressure followed by growth on an agar medium at low osmotic pressure. Alternatively, precocious germination could be induced by removing the cotyledons. Embryos of all sizes and of both genotypes of pea responded in a similar manner.  相似文献

建立昆明小鼠受孕模型,分离并体外培养胚胎细胞.检测了各培养浓度下的细胞增殖、分化与凋亡.胚胎细胞在0.2mmol/L和5.56mmol/L葡萄糖浓度的KSOM培养基中能正常发育和孵化;而在浓度为15.56mmol/L和25.56mmol/L葡萄糖培养基中胚胎发育和孵化均受到损害(P<0.005),且总细胞数和内细胞团细胞数也明显减少(P<0.01),但其细胞凋亡率与0.2mmol/L和5.56mmol/L葡萄糖浓度下胚胎细胞凋亡率无显著性差异(P>0.05).随着葡萄糖浓度的增高,胚泡总的表面积无明显变化,但胚胎细胞密度呈增加趋势.高血糖对早期胚胎的发育具有毒性作用,提示高糖可能导致妊娠合并糖尿病患者的流产和胎儿畸形率升高.  相似文献

Interploid sexual hybridizations were completed in 2001 and 2002 between seven lemon (Citrus limon(L.) Burm. f.) varieties, Key lime (C. aurantifolia (Cristm.) Swing), Palestine sweet lime (C. imettioides Tan.), Lakeland limequat (C. aurantifolia x Fortunella japonica (Thumb.) Swing.), and Etrog citron (C. medica L.) as diploid progenitors and four allotetraploid somatic hybrids (Key lime + Valencia orange, Hamlin orange + Femminello lemon, Valencia orange + Rough lemon, and Valencia orange+ Femminello lemon) in efforts to generate improved seedless triploid acid fruit hybrids. Efficient recovery of triploid progeny from such crosses requires embryo rescue to avoid embryo abortion due to endosperm failure. Germination of rescued genetically diverse immature embryos was induced on two culture media (EME and Gamborgs B5), with two sucrose concentrations (50 or 70 g l–1). All media contained 0.5 g l–1 malt extract and 4.50 M GA3. Germination of globular, heart and torpedo shaped embryos (defined as small embryos) was significantly (p < 0.05) affected by medium and genotype. Gamborgs medium induced 82.89% germination. Of germinated embryos, 11–65% developed into normal plants with differences among crosses. Cotyledonary embryos (defined as immature embryos with fully developed cotyledons) germinated and developed into normal plants at higher rates than less-developed embryos. In efforts to improve the efficiency of plant recovery, small embryos from Todo el año × HF and Lisbon × HF crosses conducted during 2002 were rescued and cultured on three media (MS, Gamborgs, and RMA) for comparison. The comparison of Hy- and C-bands reveals three types of heterochromatin (Hy+, Hy−, and Hy 0) inVicia faba. By C-banding, the total of constitutive heterochromatin is identified. The Hy-banded heterochromatin is restricted to the M-chromosome: The nucleolus-associated heterochromatin appears as two darkly stained (Hy+)-bands bordering the secondary constriction, while reduced staining (Hy−)-bands are located near the centromere. The heterochromatin of the S-chromosomes is of the (Hy 0)-type, i.e., it does not respond to the Hy-banding procedure. A complete karyogram of the C-banded chromosome complement is presented and discussed. A comparison of C-bands and cold-sensitive segments clearly shows that negatively as well as positively reacting cold-induced segments are partly heterochromatic, partly euchromatic.  相似文献
