The effect of gamma irradiation on potato microtuber production in vitro   总被引:3,自引:0,他引:3  
The effects of low doses of gamma irradiation and potato (Solanum tuberosum L.) cultivar on the production of microtubers in vitro were investigated. Nodal segments from virus free explants of three potato cultivars (cv.) were placed on tuberization inducing medium and irradiated with 4 doses of gamma radiation (2.5, 5, 10, 15 Gy). Cv. Diamant produced the highest number of microtubers followed by Draga and Spunta. Irradiation of the explants with 2.5 Gy of gamma radiation led to a significant increase in the number of microtubers (38% increase over the control). Average weight of microtubers was not significantly influenced by low doses of gamma irradiation. Draga microtubers were the largest followed by Diamant and Spunta. Microtubers resembled mature tubers in shape (Spunta was oval and Draga and Diamant were spherical). Size of microtubers was crucial for sprouting in vivo. It is suggested that only microtubers larger than 5 mm in diameter (250 mg) be used to produce minitubers in vivo. Since 2.5 Gy is a low irradiation dose, it can be used to enhance tuberization in vitro without fear of genetic changes in the used cultivars. Aphidophagous ladybird beetles (Coleoptera: Coccinellidae) are attracted to and feed heavily on aphids, but many species will also feed opportunistically on other prey that they encounter. In potatoes (Solanum tuberosum L.) in Washington State, USA, coccinellids feed on both green peach aphids (“GPA,” Myzus persicae Sulzer) and eggs of the Colorado potato beetle (“CPB,” Leptinotarsa decemlineata Say). The guild of aphidophagous ladybirds includes two native species, Hippodamia convergens Guérin-Méneville and Coccinella transversoguttata Brown. Recently, an introduced species, Coccinella septempunctata L., has invaded and apparently displaced its native congener. A second exotic, Harmonia axyridis Pallas, has colonized the area and is becoming more abundant. We compared larval development of each species on a monotypic diet of GPA, a monotypic diet of CPB eggs, or a mixed diet of both GPA and CPB eggs. Our goal was to answer two questions: (1) do larvae of the four ladybird species benefit from including CPB eggs in their diet and (2) do the four ladybird species differ in their ability to utilize CPB eggs as prey? No larva of any species completed development on a pure diet of CPB eggs, and survivorship was highest for all species when they fed on a pure diet of GPA. One native species, H. convergens, and one exotic species, H. axyridis, exhibited significantly lower survivorship on a mixed diet of both CPB eggs and GPA, compared to a pure GPA diet; H. axyridis also took longer to develop from egg to adult when both prey were provided. Survivorship of the two Coccinella spp. was not altered by the inclusion of CPB eggs with GPA, although CPB eggs lengthened the development time of C. transversoguttata. Adult size was not consistently affected by diet for any of the coccinellids. Overall, no ladybird species benefited from the inclusion of potato beetle eggs in its diet. The two Coccinella species responded similarly to the inclusion of CPB eggs, and so we would not expect any difference in the success of coccinellid larval development in potato fields following the replacement of C. transversoguttata by C. septempunctata. Hippodamia convergens and H. axyridis, the two species whose survivorship was depressed by combining CPB egg and aphid prey, were also the two species that consumed the greatest number of CPB eggs during successful larval development. A comparison of total egg consumption by each species cohort suggested that displacement of the other species by H. axyridis would not alter CPB biological control, because the higher per capita feeding rate by H. axyridis larvae compensated for individuals’ greater mortality risk on a diet including CPB eggs.  相似文献

Summary The effects of 0,5 and 10 Gy doses of gamma irradiation on the enhancement of embryogenesis and plant regeneration efficiency of three barley (Hordeum vulgare L.) genotypes, Igri, Arabi Abiad and AECS 76, were evaluated. Embryo yields at 5 and 10 Gy doses were significantly higher than those of the control (OGy). This effect was genotype-dependent. The most responsive genotype was Igri, with 592.8 embryos 32 anthers exposed to 10 Gy. However, despite a high embryo induction rate, the green plant regeneration rate was low. Arbi Abiad had a higher ability to generate green plants produced from, with 28. 13 plantlets obtained from 32 anthers at 10 Gy; irradiation had no significant effect on regeneration of Igri and AECS 76 genotypes. In general, the 10 Gy dose produced a much higher embryo yield than the 5 Gy dose. The root-tip chromosome number and the fertility of 298 regenerating green plants of cv. Igri revealed that 64% of the tested plants were spontaneously doubled haploids (DHs) and fertile.  相似文献

Predation upon lady beetle (Coleoptera: Coccinellidae) eggs in the field is most often instances of egg cannibalism by larvae or adults while the majority of the remaining predation events upon coccinellid eggs is done by other species of Coccinellidae. Thus the recent introduction and establishment of Harmonia axyridis in the US could negatively affect native species of Coccinellidae via egg predation. However, little is known regarding the suitability of interspecific coccinellid eggs as a food source for larval development. In this study, it was found that native first or third instar Coleomegilla maculata and Olla v-nigrum larvae were incapable of surviving to the adult stage when provided solely exotic H. axyridis eggs. In stark contrast, H. axyridis larvae survived equally well when cannibalizing eggs or eating eggs of either native species. When C. maculata and O. v-nigrum were grouped as ‘native’ and compared with the exotic H. axyridis, more native eggs were attacked than exotic eggs and a higher percentage of eggs was attacked by H. axyridis larvae. Native and exotic larvae attacked a similar percentage of native eggs but native larvae attacked significantly fewer exotic eggs than did exotic larvae. These data suggest that H. axyridis may prey upon the eggs of these native species, when encountered in the field, compared with the likelihood of the native species preying upon H. axyridis eggs. Therefore, eggs of the native species C. maculata and O. v-nigrum will continue to be subjected to cannibalism and also to possible predation by other native species and the exotic H. axyridis.  相似文献

Summary The bz2-mu1 allele contains a 1.4 kb Mu element insertion in the open reading frame of the bronze-2 locus. This insertion suppresses gene activity. In an active Mutator line, however, the bz2-mu1 allele shows high somatic instability resulting in numerous purple spots of full gene activity against a beige background in the aleurone tissue of the kernel; restoration of gene activity results from excision of the Mu element. In contrast, in plants with an inactive Mutator system, uniformly bronze kernels are found, and the Mu element at bz2-mu1 is stabilized. Accompanying a loss of somatic instability, this Mu element, as well as the Mu elements elsewhere in the genome, have an increased level of DNA modification. Spontaneous reactivation of somatic instability in inactive Mutator lines rarely occurs; however, reactivation can be induced with gamma irradiation. Reactivated plants regain both the spotted kernel phenotype indicative of element excision from the bz2-mu1 reporter allele and diagnostic restriction sites within the Mu elements indicative of a hypomethylated state. The reactivated plants transmit these characters to their progeny. These data support the hypothesis that genomic shock can elicit cryptic transposable element activities in maize. Possible mechanisms for inactivation and reactivation of the Mutator transposable element system are also discussed.  相似文献

Summary The role of lignin as a physical defence against Dendroctonus micans was investigated in laboratory feeding experiments. The effect of lignin is dose-dependent, reducing larval survival, growth rate, and weight, as well as affecting gallery construction. Adults lay fewer eggs in lignified bark and also tend to construct abnormal galleries. The distribution of lignin in trees suggests a role in defence against bark beetles that feed in the thicker bark on the lower bole.  相似文献

Loblolly pine (Pinus taeda L.) produces an induced defensive hypersensitive response in inner bark colonized by the southern pine beetle, Dendroctonus frontalis Zimm. (Coleoptera: Scolytidae), and its associated fungi. Adult beetles forced to colonize the induced response tissue in a laboratory study constructed galleries that were the same length as adults boring in normal phloem. However, each female laid fewer eggs in the induced tissue. Larval and pupal mortality were also higher in this tissue when compared to surrounding phloem. Beetles colonizing trees in response to pheromone baits constructed less gallery and laid fewer eggs in induced tissue than in surrounding normal phloem. These results suggest that the lesions produced by the induced defense system in conifers may not only contain the growth of fungi inoculated into trees during the attack phase of beetle colonization, but may also affect survival of bark beetle progeny.
Résumé Une réaction hypersensible de défense a été induite dans les couches profondes de l'écorce de P. taeda colonisée par D. frontalis et le champignon qui lui est associé. Au laboratoire, des adultes, contraints de coloniser les tissus où une réaction hypersensible e été induite, ont foré des galeries de même longueur que celles creusées dans du phloème sain. Cependant les femelles ont pondu moins d'oeufs dans les tissus induits; les mortalités larvaires et nymphales étaient aussi plus fortes dans ce tissu que dans le phloème voisin. Les scolytes, ayant colonisé les arbres après attraction par des pièges à phéromones, forent moins de galeries et pondent moins d'oeufs dans le tissu induit que dans le phloème sain voisin. Ces résultats suggèrent que les lésions, provoquées par le système de défense induit des conifères, peuvent non seulement limiter la croissance du champignon inoculé dans l'arbre au cours de la colonisation des scolytes, mais aussi affecter la survie des descendants dans l'écorce.

Summary The interaction between grazing by Gastrophysa viridula and interspecific plant competition was investigated for Rumex obtusifolius growing in the field. During an eight-month growing season non-competing R. obtusifolius grew larger than competing plants although herbivore loads (beetle productivity per unit leaf area) were similar for competing and non-competing plants. However, grazing significantly reduced the growth of R. obtusifolius only in competing plants thus demonstrating a synergism between plant competition and invertebrate herbivory. The relevance of these findings for the distribution of plant species is discussed.  相似文献

This study investigated the effect of azadirachtin A (AZA) on the ultrastructure of the endocrine glands (the prothoracic gland, the corpus allatum, and the corpus cardiacum) responsible for controlling moulting and ecdysis in Lucilia cuprina larvae. All constituent endocrine glands within the ring complex exhibited ultrastructural change after exposure of larvae to AZ A in the diet. The effects on the nucleus were the most noticeable and included: (1) crenulation of nuclear shape, (2) clumping of heterochromatin, and (3) pyknosis. The degenerative changes to the nuclei appeared to precede changes within the cytoplasm. It was concluded that the degeneration of all three endocrine glands within the ring complex would contribute to a generalised disruption of neuroendocrine function.  相似文献

A survey of 100 Scots pine trees (Pinus sylvestris) showed that those windthrown with a declination > 50° from vertical were preferentially colonized by the larger pine shoot beetle, Tomicus piniperda (L.) (Coleoptera: Scolytidae). A factorial experiment was done with logs of 0°, 45° and 90° declination, and different diameters and spacing to test hypotheses, involving olfactory, visual and tactile mechanisms. In the experiment, the attack rates depended strongly on log diameter with a preference being shown for larger diameter logs having a rough bark structure. In contrast to the field survey, experimental declination did not increase attack densities. The spacing between neighbouring attack sites in the bark was regular rather than random, which could be due both to spacing between conspecifics and regularities in bark structure. The contrary data from survey and experiment is explained by release of odours from wind damaged trees, and host odour and bark roughness stimuli are suggested to operate in different steps in the behavioural sequence of host location and colonization.
Résumé L'observation de 100 individus de Pinus sylvestris a montré que les arbres inclinés de plus de 50° par le vent sont colonisés de préférence par T. piniperda L., Scolytidae. Des expériences avec des branches de différents diamètres, plus ou moins espacées et inclinées de 0°, 45° et 90°, ont permis d'examiner les influences de l'olfaction, de la vision et du tact. Les attaques ont montré une préférence marquée pour les diamètres les plus grands et pour la structure la plus rugueuse. Contrairement aux observations dans la nature, l'inclinaison n'a pas augmenté les attaques.L'analyse sur l'écorce de la distance entre les points d'attaque montre une tendance à une distribution régulière plutôt qu'au hasard, tenant compte de la régularité structurale de l'écorce.Les résultats contradictoires entre les expériences et les observations peuvent s'expliquer par l'abondante émission d'odeurs par les arbres endommagés par le vent, paramètre non contrôlé dans les observations. Les stimuli dus à la rugosité et à l'odeur de l'arbre devraient opérer à différentes étapes de la découverte et de la colonisation de l'hôte.

Transgenic plants producing insecticidal proteins from Bacillus thuringiensis (Bt) can control some major insect pests and reduce reliance on sprayed insecticides. However, large scale adoption of this technology has raised concerns about potential negative effects, including evolution of pest resistance to Bt toxins, transgene flow from Bt crops to other plants, and harm to non-target beneficial organisms. Furthermore, concern has also been expressed over the effects this technology may have on biodiversity in general. Ecologically relevant risk assessment is therefore required (Risk = Hazard × Exposure). Transgenic plants that produce Bt toxins to kill insect pests could harm beneficial predators. This might occur directly by transmission of toxin via prey, or indirectly by toxin-induced reduction in prey quality (Hazard). To test these hypotheses, we determined the effects of Bt-producing canola on a predatory ground beetle (Pterostichus madidus) fed larvae of diamondback moth (Plutella xylostella) that were either susceptible or resistant to the Bt toxin. Survival, weight gain, and adult reproductive fitness did not differ between beetles fed prey reared on Bt-producing plants and those fed prey from control plants. Furthermore, while Bt-resistant prey was shown to deliver high levels of toxin to the beetle when they were consumed, no significant impact upon the beetle was observed. Subsequent investigation showed that in choice tests (Exposure), starved and partially satiated female beetles avoided Bt-fed susceptible prey, but not Bt-fed resistant prey. However, in the rare cases when starved females initially selected Bt-fed susceptible prey, they rapidly rejected them after beginning to feed. This prey type was shown to provide sufficient nutrition to support reproduction in the bioassay suggesting that Bt-fed susceptible prey is acceptable in the absence of alternative prey, however adults possess a discrimination ability based on prey quality. These results suggest that the direct effects of Bt-producing canola on predator life history was minimal, and that predators’ behavioural preferences may mitigate negative indirect effects of reduced quality of prey caused by consumption of Bt-producing plants. The results presented here therefore suggest that cultivation of Bt canola may lead to conservation of non-target predatory and scavenging organisms beneficial in pest control, such as carabids, and may therefore provide more sustainable agricultural systems than current practices. In addition, minimal impacts on beneficial carabids in agro-ecosystems suggest that Bt canola crops are likely to be compatible with integrated pest management (IPM) systems.  相似文献

Summary Mortality of neonatal pine beauty moth larvae varied from 0% to 100% depending on the duration of food deprivation and temperature. At 20°C and 100% RH all larvae had died by the fourth day of food deprivation, whereas at 10°C and 100% RH larvae survived for twelve days without food. Although larvae were able to survive at 15°C for up to seven days without food, establishment on thier host at this temperature was seriously affected by three days of starvation. No establishment took place after four days of starvation at 15°C. At 10°C establishment was significantly affected by starvation but not to such a marked degree. Establishment at both 15°C and 10°C was enhanced if the larvae were presented with a highly suitable host. The mean relative growth rate of those larvae reaching fifth instar was unaffected by the duration of their prefeeding starvation period. However, overall mean relative growth rates of the larvae were significantly affected by the duration of the starvation period. The implications of these results are discussed in relation to the population dynamics of this insect.  相似文献

Summary A study of the effects of defoliation by insects on the chemistry of lodgepole pine (Pinus contorta), and on the performance of Panolis flammea (Lepidoptera; Noctuidae) larvae, was carried out in a forest in northwest Scotland I year after a severe outbreak of P. flammea had caused extensive defoliation. Larval weight and survival were not significantly different on trees that had experienced different levels of defoliation in 1986. The nitrogen and tannin content of current and previous years' pine needles was not significantly affected by defoliation (although both were slightly greater in the foliage of defoliated trees). Phosphorus content of young pine foliage was lower (but not significantly lower except on one occasion) on heavily defoliated trees. On all sampling occasions, however, the nitrogen: phosphorus ratio was significantly higher on heavily defoliated trees. There were large differences in monoterpene composition of the previous year's shoots associated with defoliation intensity, but these differences had largely disappeared in the new growth. The results are discussed in relation to other studies on the effects of insect damage on plant chemistry and insect performance and in relation to the abundance of P. flammea in Scotland.  相似文献

The attraction of Tomicus piniperda (L.) (Coleoptera: Scolytidae) to Scots pine trees of low and higher vigor with and without previous attacks was monitored with sticky traps. Somewhat higher numbers of beetles were caught on low vigor trees than on trees of higher vigor, indicating differences in olfactory stimuly between the two classes of trees.Many more beetles were caught on trees attacked by T. piniperda and on trees with simulated bark beetle galleries (manually drilled holes) than on control trees with no or only a few attacks. The strong attraction of beetles to attacked trees is attributed to the beetles responding to host volatiles released from the galleries.
Zusammenfassung Der Anflug von Tomicus piniperda (L.) (Coleoptera: Scolytidae) an Kiefern in schlechtem oder besserem Gesundheitszustand, mit oder ohne vorherigen Befall, wurde mit Hilfe von Leimfallen untersucht. An Bäumen in schlechtem Zustand wurden etwas mehr Käfer gefangen als an Bäumen in besserem Gesundheitszustand. Das deutet auf Unterschiede in Geruchsreizen zwischen den beiden Klassen des Baumzustands hin.An Kiefern, die von T. piniperda befallen waren oder simulierte Einbohrungen (von Hand gebohrte Löcher) hatten, wurden viel mehr Käfer gefangen als an Kontrollbäumen ohne oder mit sehr geringem Befall. Der starke Anflug von Käfern an befallene Bäume wird gedeutet als Reaktion auf Duftstoffe des Wirts, die aus den Borkenkäfergängen abgegeben werden.

Summary Genetic neighborhood size and area were estimated from pollinator movements over 3 years in a scarab beetle-pollinated clonal herb, Dieffenbachia longispatha (Araceae) at the La Selva Biological Station, Costa Rica. This species was characterized by low densities of reproductive individuals and asynchronous flowering within the population. The pollinator flight distributions were characterized by relatively long mean distances between consecutive visits to inflorescences (83 m) and movements to the nearest neighboring inflorescence in the appropriate phase of flowering. Pollinator movement distributions between consecutive visits to inflorescences were significantly leptokurtic in 2 of the 3 years. I calculated neighborhood sizes incorporating the levels of kurtosis and found minimal estimates of N e to be 227–611 ramets and neighborhood area to be 88 000–180 000 m2. The three beetle species that made 94% of the visits (Cyclocephala gravis, C. amblyopsis, and Erioscelis columbica) varied in their flight distributions and in their contributions to the estimates of neighborhood size. Cyclocephala amblyopsis exhibited the greatest degree of kurtosis in its movement patterns, and neighborhood size based on its movement was large relative to N e calculated from movement distributions of the other two beetle species. Long-distance movements of C. amblyopsis (>300 m) accounted for 68% of the neighborhood size.  相似文献

The effects of ultraviolet-B radiation on loblolly pine   总被引:11,自引:0,他引:11  
Summary Depletion of stratospheric ozone and the resulting increase in ultraviolet-B (UV-B) radiation may negatively impact the productivity of terrestrial ecosystems. This concern has led to a number of studies that report the influence of supplementing UV-B radiation on plant growth and development. However, only two of these field studies have included tree species and both were singleseason experiments. In this study, loblolly pine (Pinus taeda L.) from seven seed sources was grown under natural and supplemental levels of UV-B radiation. Irradiation treatments were continued for three seasons on plants from four of the seven groups and for 1 year only for three groups. The supplemental irradiances simulated those that would be anticipated with stratospheric ozone reductions of 16% and 25% over Beltsville, Md. The effects of UV-B radiation during the 1st year on plant growth varied among the seed sources. The growth of plants from two of the seven seed sources tested showed significant reductions following a single irradiation season and plants from one group tended to be larger under increased UV-B radiation. However, after 3 years of supplemental irradiation, plant biomass was reduced in all four groups by 12–20% at the highest simulated ozone depletion. These results suggest that the effects of UV-B radiation may accumulate in trees and that increased UV-B radiation could significantly reduce the growth of loblolly pine over its lifetime. However, they also point to a need for multiple season research in any analysis of potential consequences of global change on the long-term growth of trees.  相似文献

Osmoderma eremita is a threatened scarab beetle living in the hollows of old deciduous trees and is regarded as an umbrella species of the beetle fauna associated with this habitat. Several methods like pitfall trapping and wood mould sampling have been used to monitor the occurrence of O. eremita, but these methods cannot be applied for trees with certain characteristics. Recently, (R)-(+)--decalactone was identified as a male-produced sex pheromone of the species. Here, we show that -decalactone can be detected in hollow trees by air sampling and that the presence of the compound is strongly correlated with the occurrence of living male beetles in the same trees. Air was sampled from tree cavities and extracts analysed using gas chromatography–and mass spectrometry. There was a 89% match between the detection of -decalactone in extracts and the occurrence of male O. eremita±2 days from the sampling event. In the absence of males, samples never contained -decalactone, and the presence of this compound in a tree cavity appears to be a good predictor of O. eremita occupancy. Air sampling can be a useful complement to other methods when trying to detect as many trees housing this beetle as possible, which is crucial when estimating populations sizes and developing conservation strategies for this species.  相似文献

Limonin and ten structurally modified limonins were evaluated as antifeedants against 4th instar larvae of the Colorado potato beetle, Leptinotarsa decemlineata Say, in no-choice leaf dics assays. The epoxide and furan groups were shown to be essential structural requirements for high antifeedant activity.

Aucune activité dissuasive n'ayant été observée avec le tétrahydrolimonine ou le déoxylimonine, l'anneau furane et le groupe époxy sont donc nécessaires pour obtenir cette activité. Ni la réduction de la fonction 7-kéto, ni la rupture de l'anneau A de la limonine n'ont eu d'effêt sur cette activité. La réduction des fonctions 16-carbonyl et 7-kéto de la limonine ont diminué l'activité, mettant en évidence une action possible de l'anneau D-lactone dans l'activité phagodissuasive. Tandis que la déépoxydation de la limonine entraîne une perte d'activité totale, une restauration partielle de cette activité a pu être obtenu par une réduction ultérieure de la fonction 7-kéto.

In contrast to the northern hemisphere where species of Chironomidae are usually the dominant benthic invertebrates in the coldest upper reaches of glacial streams, mayflies (Deleatidium spp.: Leptophlebiidae) predominate in equivalent conditions in New Zealand. We examined the life histories and annual production of Deleatidium spp. at two sites on the Matukituki River (South Island, New Zealand) and at three sites in its glacier-fed tributary, Rob Roy Stream. Mean annual water temperature at the five sites ranged from 2.1 to 7.0°C. Monthly sampling showed that mayfly populations were poorly synchronised at all sites but were probably univoltine. The large Deleatidium cornutum was the dominant mayfly species found at the upper sites (Sites 1 and 2) on Rob Roy Stream, whereas above the confluence with Matukituki River (Site 3) it co-existed with a complex of smaller species we refer to as D.angustum”. Deleatidiumangustum” also dominated at the Matukituki sites. Deleatidium production calculated for the five sites, assuming an 11-month nymphal life, ranged from 0.48 g dry weight/m2/year (Site 1) to 3.07 g dry weight/m2/year (Site 3). The values for D. cornutum at Sites 2 and 3 are high for a species of Deleatidium and reflect its large size. This species appears to be strongly adapted for growth at low temperatures. Climate change scenarios for New Zealand predict the gradual and ultimate loss of small South Island glaciers and a consequent warming of streams as runoff from rainfall and snow melt becomes more dominant in spring. As a result, suitable habitats will be lost for cold-water specialists such as D. cornutum, and they are likely to suffer reductions in their distributional range and local extinction. In contrast, species such as those in the D.angustum” complex may extend their ranges into streams formerly dominated by glacial meltwater. Onychoteuthid squids are among the most common cephalopods found in New Zealand waters, and comprise a major portion of the regional diets of teuthophagous marine mammals. Although several recent publications have addressed aspects of various species’ biology and reproduction, the systematics of the group remains poorly understood. Herein the ontogenetic and adult morphologies of regionally occurring known species of the genera Moroteuthis, Onychoteuthis, and Notonykia are redescribed, and Notonykia nesisi sp. nov. is described for the first time. Ontogenetic and sexually dimorphic variation in characters and character states associated with body proportions, and beak, radula, tentacular hook, palatine tooth and gladius morphologies are also described and compared between local onychoteuthid taxa for the first time.  相似文献
