Roepstorff A. and Andreassen J. 1982. Course of heavy primary infections and earlier immunologically mediated rejection of secondary infections of Hymenolepis diminuta in mice. International Journal for Parasitology12: 23–28. The worms of heavy (50–100 worms) primary Hymenolepis diminuta infections in inbred C57-mice were 1–2 mm long when growth ceased about day 4. Thereafter the mean length decreased by shrinkage and/or ‘decollation’, the worms moved backwards in the small intestine and were rejected from day 6 to day 10. Heavy secondary infections given 14 days after a heavy primary infection were severely stunted (0.2–0.3 mm) but normally situated in the intestine on day 2 and nearly all were rejected by day 4. Even when the time between the primary and secondary infections was increased to 21 or 42 days, therecovery, position and length of the secondary worms were significantly different from primary infections. These results show that an immunologically mediated memory was involved, and that functional antigens can be released from the scolex and/or the neck alone.  相似文献

Hopkins C. A. and Barr I. F. 1982. The source of antigen in an adult tapeworm. International Journal for Parasitology12: 327–333. Although a primary infection of Hymenolepis diminuta is not rejected for 9–15 days by a mouse, it has been shown that a primary infection terminated chemically after only 3 days induces as good protection against challenge. This demonstrated that a scolex and 1–2 mm of neck tissue (all that is formed by day 3 post infection) are an adequate source of ‘protective’ antigen. Irradiated (350 Gy) cysticercoids which survive but show little growth immunize as effectively as normal cysticercoids which indicates actively growing neck tissue is not essential and hence the scolex alone is a sufficient source of ‘protective’ antigens. In the rat irradiated cysticercoids were found to establish, double their length over 3–6 days and then slowly shrink, but 14% of the worms were still present 49 days p.i. Although a primary infection of normal worms in a rat markedly depresses growth of a secondary infection administered 7 days after the chemical expulsion of the primary, irradiated scoleces induced no measurable protection. These results are discussed in relation to the source of antigen and the fundamental difference in the protective response of mice (an abnormal host) and rats (a normal host) to the tapeworm H. diminuta in the small intestine.  相似文献

Changes in the ultrastructure of the tegument and subtegumental cells of 4-day-old Hymenolepis diminuta were studied in vitro in 50% fresh normal rat serum over a 5-h period and compared with heat-inactivated serum and saline controls. First, membrane-bound vesicles accumulate above the microthrix-border. After 30–40 min large vacuoles, which may contain membranous elements, appear in the tegument at a time when the surface of the young strobila is virtually denuded of the microthrix-border. With prolonged incubations there are subtegumental secretory inclusions with dark, enveloping cytoplasm in the tegument and finally the apical plasma membrane, together with the majority of the matrix, is lost. The disrupted portion of the worm is abruptly demarcated from the comparatively intact scolex/anterior neck region by a constriction. Even after 5 h incubation there is no evidence of loss of tegumental matrix components from regions anterior to the constriction but the neck region shows a significant denudation of the microthrix layer and the tegument contains numerous inclusions. The scolex tegument only showed little evidence of loss of membrane from the surface. Possible mechanisms for the avoidance of complement-mediated lysis in the anterior region are discussed.  相似文献

The activities of 5-nucleotidase (E.C. 3.1.3.5), adenosine deaminase (E.C. 3.5.4.4), adenosine kinase (E.C. 2.7.1.20), AMP deaminase (E.C. 3.5.4.6) and adenylate kinase (E.C. 2.7.4.3) were demonstrated in homogenates of Hymenolepis diminuta. The Km values for adenosine and AMP of the above enzymes were determined. Growth and development of Hymenolepis nana in mice maintained at different environmental temperatures. International Journal for Parasitology16: 13–17. On days 3 and 4 post infection (p.i.) the number of Hymenolepis nana cysticercoids in the villi of male mice kept at 5°C did not differ from those in controls (21°C), but fewer larvae were observed in hosts at 35°C. However, on day 10 and 14 p.i. in cysticercoid and egg-induced infections respectively, the incidence of infection was higher and significantly more worms per host were found in mice at 5°C than in those kept at 21 or 35°C. Also, worms from mice maintained at 5°C were significantly heavier and became patent 1 day earlier than those from 21°C, which, in turn, were significantly heavier than those grown in hosts at 35°C.  相似文献

Complement-mediated lysis in vitro of newly excysted tapeworms: Hymenolepis diminuta, Hymenolepis microstoma, Hymenolepis nana and Hymenolepis citelii. International Journal for Parasitology16: 157–161. Newly excysted worms of Hymenolepis diminuta were lysed in 50% normal serum from all 13 animal species tested, including man. Since H. diminuta was neither lysed in complement inactivated serum—by heat or adding EDTA, LPS or CVF—nor in C5-deficient mouse serum, it is concluded that the lysis was associated with the complement cascade. It is shown that H. diminuta can activate the complement system via both the classical and alternative pathway. Furthermore, it is indicated that the lysis is independent of serum antibodies. Hymenolepis nana and H. citelli were also lysed in all normal sera tested, eight and six respectively, while newly excysted worms of H. microstoma were lysed in normal sera from 10 mammals and birds, but not in sera from its hosts, the mouse, rat and golden hamster. This indicates that the complement system of these three species differs from that of the other species tested in such a way that H. microstoma is able to avoid lysis in these sera.  相似文献

The effects of the density of Hymenolepis diminuta and the effects of thermal acclimation on the water balance of Tenebrio molitor were examined. Also, the subsequent infectivity of the cysticercoids for rats were investigated. T. molitor beetles were fed known numbers of H. diminuta eggs and then were kept at 15° or 25°C for 14 days. After 14 days, beetles were desiccated and water loss was determined. Parasite density did not significantly affect transpiratory water loss in T. molitor kept at 15° or 25°C following 24 or 48 hr of desiccation. However, after 72 hr of desiccation, beetles maintained at 15°C evidently could not regulate water efficiently since there was a significant increase in the transpiratory water loss as parasite density increased. Beetles acclimated at 15°C produced fewer cysticercoids than did beetles maintained at 25°C. Also, fewer adult worms were recovered from rats intubated with cysticercoids from heavily infected, 15°C-acclimated beetles. Apparently, heavily infected beetles acclimated to 15°C do not produce viable cysticercoids.  相似文献

An adult hymenolepidid tapeworm was recovered from a 52-year-old Tibetan woman during a routine epidemiological survey for human taeniasis/cysticercosis in Sichuan, China. Phylogenetic analyses based on sequences of nuclear 28S ribosomal DNA and mitochondrial cytochrome c oxidase subunit 1 showed that the human isolate is distinct from Hymenolepis diminuta and Hymenolepis nana, the common parasites causing human hymenolepiasis. Proglottids of the human isolate were unfortunately unsuitable for morphological identification. However, the resultant phylogeny demonstrated the human isolate to be a sister species to Hymenolepis hibernia from Apodemus mice in Eurasia. The present data clearly indicate that hymenolepidid tapeworms causing human infections are not restricted to only H. diminuta and H. nana.  相似文献

Mice were infected orally with 1,6, or 30 cysticercoids of Hymenolepis diminuta. These were allowed to develop for different periods of time before elimination with anthelminthic, thus exposing the hosts to antigens from the prestrobilate, early strobilate, or fully strobilate worms. Other groups of mice were immunized by intraperitoneal (ip) implantation of a live strobilate worm or by ip implantation of live worms from cysticercoids excysted in vitro. Strong protection against challenge with a surgically transplanted strobilate worm was achieved by prior infection with 6 or 30 worms eliminated as early as Day 3 of infection. By this time these worms would not have strobilated. Conversely, a single worm, strobilating extensively over 16 days, stimulated only weak protection. Parenteral implantation of excysted worms protected mice but parenteral implantation of a strobilate worm had no effect. It is suggested that (i) the tapeworm protective antigens are primarily related to the scolex and/or the germinative region; (ii) the number of worms and the duration of antigenic stimulation in an immunizing infection determine the magnitude of a protective secondary response.  相似文献

The inter-relationship between the amino acid serine and lipid metabolism in the rat tapeworm Hymenolepis diminuta has been studied under in vitro conditions. The label from U-14C-serine, U-14C-glucose and 1-14C-oleic acid was rapidly incorporated into worm tissue phospho- and glycolipids, the latter illustrating the synthesis of cerebrosides by H. diminuta. Activity from U-14C-serine was recovered in phosphatidylserine, phosphatidylethanolamine, cerebrosides and several unidentified lipid-like compounds. The majority of the label recovered in phosphatidylethanolamine was associated with the ethanolamine moiety; in the cerebrosides with the sphingosine moiety. The sugar moiety of the cerebrosides was galactose.Pulse label studies showed a serine flux phenomenon, and a rapid rate of turnover of some of the unidentified compounds.Exogenous ethanolamine had no detectable effect upon absorption and conversion of serine to tissue phosphatidylethanolamine. Incubation of H. diminuta homogenates with phosphatidyl U-14C-serine resulted in the recovery of considerable activity in phosphatidylethanolamine. The results show that the major pathway of phosphatidylethanolamine synthesis is by decarboxylation of phosphatidylserine.  相似文献

Ultrastructural studies, including stereological analyses of micrographs, have been made of five-worm primary infections of Hymenolepis diminuta from C57 mice to determine whether the immune destrobilation/rejection process was accompanied by significant changes in the fine structure of the scolex tegument. Destrobilation/rejection of worms occurred from Days 9-12 after infection. For the first 5 days after infection, the scolex tegument showed no detectable differences in ultrastructure compared with that of "control" worms from either Wistar rats or immunosuppressed C57 mice. By Day 6, large lipid deposits were observed in the tegument and associated musculature of worms from untreated C57 mice. Further, worms recovered from Days 6-8 after infection also showed increased activity of the Golgi apparatus, GER, and mitochondria of the tegument, resulting in increased numbers of discoidal secretory bodies. Concomitant with destrobilation/rejection from Day 9 was a drop in the number of secretory bodies, an increase in autophagic activity throughout the tegument, and a blistering of the tegument surface plasma membrane. The possible functional significance of the results is discussed in relation to host immunity.  相似文献

Rau M. E. 1979. The frequency distribution of Hymenolepis diminuta cysticercoids in natural, sympatric populations of Tenebrio molitor and T. obscurus. International Journal for Parasitology9: 85–87. Natural, sympatric populations of Tenebrio molitor and T. obscurus were examined for cysticercoids of Hymemlepis diminuta. The distribution of cysticeroids in both species and both sexes conformed to the negative binomial. Cysticeroids were more prevalent and the mean intensity of infection was higher in T. obscurus than in T. molitor. No differences in the intensity of infection were detected between the sexes. Larvae of both beetle species were always very lightly infected. The significance of these factors in the transmission of the infection to the rat definitive host is discussed.  相似文献

Hymenolepis diminuta possesses a tegumental ribonuclease (RNase) which hydrolyzes rat liver and degraded yeast RNA. Polyacrylamide gel electrophoresis and gel chromatography of rat liver RNA after incubation with intact worms demonstrated significant hydrolysis of the high molecular weight RNA fractions (28 S and 18 S), with the appearance of fractions of intermediate molecular weight (i.e., between 18 S and 4 S), as well as ethanol-soluble fractions. Hydrolysis of degraded yeast RNA (with a molecular weight of approximately 25,000) yielded a single ethanol-precipitable hydrolysis product, as well as ethanol-soluble hydrolysis products.  相似文献

Background

The aim of the present paper was to describe the enzymatic antioxidant system in Hymenolepis diminuta collected from rats exposed to chronic cestode invasion.

Methodology

We dissected different tissues of H. diminuta (immature proglottids, genital primordia, hermaphroditic proglottids, early uterus, and gravid uterus) and studied activity of: superoxide dismutases (SOD1 and SOD2), catalase (CAT), glutathione peroxidases (non-Se-dependent GSHPx and Se-dependent GSHPx), glutathione-S-transferase (GST) and glutathione reductase (GSHR), and oxidative stress markers ?? reduced glutathione (GSH), and the lipid peroxidation level (TBARS).

Results

We demonstrated changes in antioxidant enzyme activities and levels of oxidative stress markers in different tissues of the parasite. The levels of TBARS and GSH indicate that oxidative stress occurred in tissues located proximal to the intestine wall. Activity of SOD1 was high in all parts of H. diminuta, but the GST activity was the highest of all studied antioxidant enzymes. SOD2 activity differed significantly in various parts of H. diminuta. Significant differences were observed for nonSeGSHPx and activity of other GSH-dependent enzymes was generally similar in all the tissues.

Conclusions

Our results show that the enzymatic antioxidant system of H. diminuta, allows the parasite to adapt and live under conditions of chronic oxidative stress. It suggests an oxidative-antioxidative balance during interactions between parasite and host.  相似文献

Engelkirk P. G., Williams J. F. and Signs M. M. 1981. Interactions between Taenia taeniaeformis and host cells in vitro: rapid adherence of peritoneal cells to strobilocerci. International Journal for Parasitology11: 463–474. Strobilocerci of Taenia taeniaeformis, incubated for l h in vitro with various combinations of serum and peritoneal cells from infected or non-infected rats, were examined at the ultrastructural level for evidence of cell adherence and tegumental damage. Maximal adherence and surface alterations occurred when larvae were incubated in the presence of cells and fresh serum. This was true regardless of whether the cells or the serum had been obtained from infected or non-infected donors. No tegumental damage was seen when parasites were incubated with or without cells in the absence of serum. Serum enhancement was either much reduced or abolished by heat treatment (56°C for 1 h). In the presence of EDTA, tegumental lesions still developed, but adherence of cells, especially those from non-infected rats, decreased markedly. The predominant cells interacting with the larval surface were eosinophils; these took up parasite material within phagosomes and appeared to strip microtriches from the tegumental free surface. Mast cells, some of which became degranulated, were also present in the adherent cell masses. The results indicate that potent non-specific effector mechanisms can rapidly damage the tegument of T. taeniaeformis, in vitro, in contrast to the failure of recognition and rejection by host defenses in vivo. Established strobilocerci are therefore not invulnerable but the balance of the host-parasite relationship in vivo must favor their survival.  相似文献

Interleukin (IL)-22, an immune cell-derived cytokine whose receptor expression is restricted to non-immune cells (e.g. epithelial cells), can be anti-inflammatory and pro-inflammatory. Mice infected with the tapeworm Hymenolepis diminuta are protected from dinitrobenzene sulphonic acid (DNBS)-induced colitis. Here we assessed expulsion of H. diminuta, the concomitant immune response and the outcome of DNBS-induced colitis in wild-type (WT) and IL-22 deficient mice (IL-22-/-) ± infection. Interleukin-22-/- mice had a mildly impaired ability to expel the worm and this correlated with reduced or delayed induction of TH2 immunity as measured by splenic and mesenteric lymph node production of IL-4, IL-5 and IL-13 and intestinal Muc-2 mRNA and goblet cell hyperplasia; in contrast, IL-25 increased in the small intestine of IL-22-/- mice 8 and 12 days post-infection compared to WT mice. In vitro experiments revealed that H. diminuta directly evoked epithelial production of IL-25 that was inhibited by recombinant IL-22. Also, IL-10 and markers of regulatory T cells were increased in IL-22-/- mice that displayed less DNBS (3 mg, ir. 72h)-induced colitis. Wild-type mice infected with H. diminuta were protected from colitis, as were infected IL-22-/- mice and the latter to a degree that they were almost indistinguishable from control, non-DNBS treated mice. Finally, treatment with anti-IL-25 antibodies exaggerated DNBS-induced colitis in IL-22-/- mice and blocked the anti-colitic effect of infection with H. diminuta. Thus, IL-22 is identified as an endogenous brake on helminth-elicited TH2 immunity, reducing the efficacy of expulsion of H. diminuta and limiting the effectiveness of the anti-colitic events mobilized following infection with H. diminuta in a non-permissive host.  相似文献

Lethbridge R. C. 1976. The architecture of the eggshell of Hymenolepis diminuta. International Journal for Parasitology6: 87–90. Scanning and transmission electron microscopy demonstrated that the eggshell of H. diminuta consisted of an inner continuous band from which arose an outer zone of finger-like projections with rounded tips. These tips were revealed as globule-like units lacking a regular spatial distribution but entirely covering the eggshell's surface. The appearance of these structures suggested that the shell constituents may have been deposited in an initially plastic state and possible methods of attaining the final architecture of the hardened eggshell have been discussed on the basis of this hypothesis.  相似文献

To determine geographical patterns of natural parasite infections among wild rodents, a total of 46 wild rodents from 3 different localities in northern Gangwon-do (Province), Korea were examined for intestinal parasite infections. Along with nematodes such as hookworms and Syphacia spp., Plagiorchis muris (2 specimens) (Trematoda) were collected from striped field mice, Apodemus agrarius. In a Korean wood mouse, Apodemus peninsulae, the overall nematode infections were similar to A. agrarius, but an adult worm of Echinostoma hortense (Trematoda) was collected. In addition, 2 species of cestodes, i.e., Hymenolepis nana and Hymenolepis diminuta, were collected from A. agrarius. Through this survey, A. agrarius and A. peninsule were confirmed as the natural definite hosts for zoonotic intestinal helminths, i.e., P. muris, E. hortense, H. nana, and H. diminuta, in northern Gangwon-do, Korea. Considering increased leisure activities around these areas, seasonal and further comprehensive surveys on wild rodents seem to be needed to prevent zoonotic parasite infections.  相似文献

Damage and early destruction of Taenia taeniaeformis larvae in resistant hosts, and anomalous development in susceptible hosts: a light microscopic and ultrastructural study. International Journal for Parasitology15: 377–384. Taenia taeniaeformis larvae in resistant C57 mice have been studied from 5th to 15th day post-infection (L5–L15) both at the light and electron microscopic level. L5 stages were already damaged and total destruction occurred by approx. 15 days post-infection. In stage L5, unlike fertile larvae from C3H mice, the perilarval amorphous layer (PAL) was generally absent, and the host's cells were in close contact with the parasite surface. At this stage eosinophils were already present together with neutrophils and macrophages. Larvae were seen increasing in volume between stages L6 and L8, but remained constant from stages L9 to L14, while both the tegumental distal cytoplasm (TDC) and the subtegumental cellular layer (SCL) gradually decreased. In stages L10–L14 only a narrow TDC separated the larval cavity from host cells. After the larval tegument had been reduced in thickness the eosinophil lytic enzyme release onto the parasite surface contributed to produce a ‘hole’ in the TDC where host cells penetrated and gradually filled the larval cavity of L15, destroying the parasitic residues. Therefore anomalous small larvae (L50 and more) from C3H mice (susceptible host) have been studied: in these the scolex anlagen was absent or greatly reduced; the TDC was very narrow and the SCL greatly damaged. Outside the larva the ‘host tissue’ appeared as an unidentifiable amorphous material. These larvae cannot be considered ‘dead’ but are defined as sterile.  相似文献

The present study sought to assess the potential of the cestode Hymenolepis diminuta as a bioindicator for lead accumulation in two industrial areas of the city of Riyadh, Saudi Arabia. Rats (Meriones libycus) were collected from two sites (industrial area II and Salbukh) in Riyadh. In the industrial area II, the mean levels of lead concentrations were found to be 1.96, 1.92, 1.4 and 30.72 μg/g in the rats’ liver, kidney and intestine, and in H. diminuta, respectively. In Salbukh, meanwhile, the lead concentrations were 1.63, 1.52, 1.20 and 21.31 μg/g in the rats’ liver, kidney, and intestine, and in H. diminuta, respectively. In addition, in industrial area II, compared with the liver, kidney and intestine of their host, the bioconcentration factors of lead were found to be, respectively, 15.6, 16 and 21.9 times higher in H. diminuta, and were 7.5, 8, and 10.2 times higher in the same organs compared to H. diminuta in Salbukh. The present study, therefore, proved that H. diminuta could be used as a bioindicator for heavy metal contamination in the industrial areas of the city of Riyadh.  相似文献
