The response of faba bean seedlings to the combined effects of increased atmospheric CO2 concentrations ([CO2]) and solar UV-B irradiance was studied using open-top chambers transparent to UV-B radiation. The purpose of the study was to determine whether effects of increased [CO2] on growth and physiology are modified by the present solar UV-B fluence rate in the Netherlands. Seedlings were exposed to 350 or 700 micromoles mol-1 CO2. At both [CO2], solar UV-B irradiance was either present or reduced using polyester foil opaque to UV-B radiation. To obtain information on the time dependence of increased [CO2] and UV-B radiation effects, three harvests were performed during the experiment. CO2 enrichment resulted in increased biomass production at all harvests. At the final harvest, UV-B radiation did not affect biomass production but a significant decrease was observed after 14 d of treatment. A reduction of the UV-B fluence increased shoot length at both [CO2] throughout the experiment. UV-B radiation slightly altered biomass allocation. Plants grown at reduced levels of UV-B radiation invested less biomass in flowers and more in stem material compared to plants grown at ambient UV-B levels. CO2 enrichment resulted in a stimulation of net photosynthesis after 26 and 38 d of treatment. UV-B reduction did not alter this response. After 26 d of treatment, photosynthetic acclimation to CO2 enrichment was observed, which was probably the result of accumulation of carbohydrates in the leaves. After 38 d, photosynthetic acclimation was no longer present. The UV absorbance of methanolic leaf extracts was increased by CO2 enrichment only. Both CO2 enrichment and solar UV-B reduced the transmittance of radiation through intact attached leaves. Interaction between [CO2] and UV-B radiation was limited to UV-A transmittance of leaves. Under prevalent experimental conditions, UV-B radiation did not affect the measured physiological parameters. Most open-top chambers used for climate change research are constructed of materials which do not transmit UV-B radiation. Our results indicate that part of the 'chamber effects' on plant height often described in the literature might be explained by the absence of solar UV-B radiation in these chambers.

Predicted increase in ultraviolet-B (UV-B: 280–320 mn) radiation may have adverse impacts on growth and yield of rice ( Oryza sativa L.), as has been found in studies hitherto. However, most of the studies were conducted in growth chambers or greenhouses where the plants are generally more sensitive to UV-B than in the field, presumably because of the distorted balance between UV-B and ultraviolet-A as well as PAR. This study was conducted to address the effects of enhanced UV-B on growth and yield of rice under a realistic spectral balance in the field. Three cultivars, "Koshihikari",'IR 45'and'IR 74'were pot-grown and irradiated with enhanced UV-B for most of the growing season in the field at Tsukuba, Japan (36°01'N, 140°07'E). The UV-B enhancement simulated ca 38% depletion of stratospheric ozone at Tsukuba. The results showed no UV-B effects on plant height, numbers of tillers and panicles, dry weight of the plant parts or the grain yield for any of the 3 cultivars. Natural abundance of 13C in the flag leaves was not altered by the UV-B enhancement either. While UV-absorbing compounds showed no response to the UV-B enhancement, chlorophyll contents decreased with enhanced UV-B. However, the decrease of chlorophyll was limited to an early growth stage with no effect later. We thus found no extraordinary impact of the nearly doubled UV-B radiation on rice in the field, and it would appear that a reliable prediction of the effects of UV-B will require experiments carried out over a number of years under various climatic and solar UV-B regimes.

Solar ultraviolet-B radiation (UV-B) can have large impacts on the interactions between plants and herbivorous insects. Several studies have documented effects of UV-B-induced changes in plant tissue quality on the feeding performance of insect larvae. In contrast, the effects of UV-B-induced plant responses on the behavior of adult insects have received little attention. We carried out a series of field and glasshouse experiments using the model plant Arabidopsis thaliana L. and the crucifer-specialist insect Plutella xylostella L. (diamondback moth) to investigate the effects of UV-B on natural herbivory and plant–insect interactions. Natural herbivory under field conditions was less severe on plants exposed to ambient UV-B than on plants grown under filters that attenuated the UV-B component of solar radiation. This reduced herbivory could not be accounted for by effects of UV-B on larval feeding preference and performance, as P. xylostella caterpillars did not respond to changes in plant quality induced by UV-B. In contrast, at the adult stage, the insects presented clear behavioral responses: P. xylostella moths deposited significantly more eggs on plants grown under attenuated UV-B levels than on plants exposed to ambient UV-B. The deterring effect of UV-B exposure on insect oviposition was absent in jar1-1, a mutant with impaired jasmonic acid (JA) sensitivity, but it was conserved in mutants with altered ethylene signaling. The jar1-1 mutant also presented reduced levels of UV-absorbing phenolic compounds than the other genotypes that we tested. Our results suggest that variations in UV-B exposure under natural conditions can have significant effects on insect herbivory by altering plant traits that female adults use as sources of information during the process of host selection for oviposition. These effects of natural UV-B on plant quality appear to be mediated by activation of signaling circuits in which the defense-related hormone JA plays a functional role.

The effects of elevated UV-B radiation on growth, symbiotic function and concentration of metabolites were assessed in purely symbiotic and NO3-fed nodulated plants of Lupinus luteus and Vicia atropurpurea grown outdoors either on tables under supplemental UV-B radiation or in chambers covered with different types of plexi-glass to attenuate solar ultraviolet radiation. Moderately and highly elevated UV-B exposures simulating 15% and 25% ozone depletion as well as sub- ambient UV-B did not alter organ growth, plant total dry matter and N content per plant in both L. luteus and V. atropurpurea. In contrast, elevated UV-B increased (P <0.05) flavonoid and anthocyanin concentrations in roots and leaves of L. luteus, but not of V. atropurpurea. Feeding nodulated plants of L. luteus under elevated UV-B radiation with 2 mM NO3 increased (P <0.05) nodule, leaf and total dry matter, and whole plant N content. With V. atropurpurea, NO3 reduced (P <0.05) nodule activity, root %N and concentrations of flavonoids, anthocyanins in roots and leaves and soluble sugars in roots, in contrast to an observed increase (P <0.05) in nodule dry matter per plant. Similarly, supplying 2 mM NO3 to L. luteus plants exposed to sub-ambient UV-B radiation significantly reduced individual organ growth, plant total biomass, nodule dry matter, nodule %N, and whole plant N content, as well as root concentrations of flavonoids, anthocyanins, soluble sugars, and starch of L. luteus, but not V. atropurpurea plants. These results show no adverse effect of elevated UV-B radiation on growth and symbiotic function of L. luteus and V. atropurpurea plants. However, NO3 supply promoted growth in L. luteus plants exposed to the highly elevated UV-B radiation.

Plant response to the combination of two or more abiotic stresses is different than its response to the same stresses singly. The response of maize (Zea mays L.) photosynthesis, growth, and development processes were examined under sunlit plant growth chambers at three levels of each day/night temperatures (24/16°C, 30/22°C, and 36/28°C) and UV-B radiation levels (0, 5, and 10 kJ m?2 d?1) and their interaction from 4 d after emergence to 43 d. An increase in plant height, leaf area, node number, and dry mass was observed as temperature increased. However, UV-B radiation negatively affected these processes by reducing the rates of stem elongation, leaf area expansion, and biomass accumulation. UV-B radiation affected leaf photosynthesis mostly at early stage of growth and tended to be temperature-dependent. For instance, UV-B radiation caused 3–15% decrease of photosynthetic rate (P N) on the uppermost, fully expanded leaves at 24/16°C and 36/28°C, but stimulated P N about 5–18% at 30/22°C temperature. Moreover, the observed UV-B protection mechanisms, such as accumulation of phenolics and waxes, exhibited a significant interaction among the treatments where these compounds were relatively less responsive (phenolics) or more responsive (waxes) to UV-B radiation at higher temperature treatments or vice versa. Plants exposed to UV-B radiation produced more leaf waxes except at 24/16°C treatment. The detrimental effect of UV-B radiation was greater on plant growth compared to the photosynthetic processes. Results suggest that maize growth and development, especially stem elongation, is highly sensitive to current and projected UV-B radiation levels, and temperature plays an important role in the magnitude and direction of the UV-B mediated responses.

Response of 19 cultivars of soybeans to ultraviolet-B irradiance
Nineteen soybean cultivars were grown for four weeks in controlled environmental chambers with artificial daylight supplemented by five UV-B irradiance regimes to determine the range of growth and development responses of seedlings. Data from nine plant characteristics were assessed: leaf area, dry weight of leaves, stems and roots, total plant dry weight, height, ratio of roots to shoots and leaf area to weight and rating of leaves for damage. Significant differences were observed in the responses noted. Stunting, leaf chlorosis and loss of apical dominance were three general symptoms apparent on all cultivars which received UV-B irradiance. Varying degrees of reduced leaf area and dry weight of the plants and altered ratios of weights of leaves per unit area and weight of roots to shoots were also found. It was concluded that different soybean cultivars demonstrate a marked difference in sensitivity to UV-B radiation under the artificial conditions of controlled environmental growth chambers and this may indicate a genetic basis for variability in sensitivity of soybean cultivars to this waveband. However, the sensitivity to UV-B radiation was increased by the lower than normal photon fluence of photosynthetically active radiation (225 μE m−2 s−1).

Interest in the potential consequences of stratospheric ozone depletion has led to numerous studies that have evaluated the effects of ultraviolet-B (UV-B) radiation on plant growth and productivity. However, few studies have been conducted on plants from natural ecosystems. Differences in solar UV-B radiation along latitudinal or elevational gradients may have resulted in plants from diverse habitats developing contrasting sensitivities to UV-B radiation. In this study, seeds were collected along a 3,000-m elevational gradient in Hawaii and then germinated and grown in an unshaded greenhouse with either no UV-B radiation or one of two daily UV-B irradiances, 15.5 or 23.1 kj m2. Seedlings were grown for 12 weeks and harvested to determine whether UV-B radiation altered plant biomass. The responses to UV-B radiation varied among species, but, in general, sensitivity to UV-B radiation was reduced as the elevation of seed collection increased. Of the 33 species tested, UV-B radiation significantly reduced plant height in 14 species and biomass in eight species. Biomass increased in four species grown under UV-B radiation. This study provides clear evidence that natural plant populations exhibit wide variation in UV-B radiation sensitivity and that this variation is related to the natural (ambient) UV-B radiation environment in which these plants grow.

As a result of stratospheric ozone depletion, more solar ultraviolet-B radiation (UV-B, 280–315 nm) is reaching the Earth's surface. Enhanced levels of UV-B may, in turn, alter ecosystem processes such as decomposition. Solar UV-B radiation could affect decomposition both indirectly, by changes in the chemical composition of leaves during growth, or directly by photochemical breakdown of litter and through changes in decomposer communities exposed to sunlight. In this experiment, we studied indirect and direct effects of solar UV-B radiation on decomposition of barley (Hordeum vulgare). We used barley straw and leaf litter grown under reduced UV-B (20% of ambient UV-B) or under near-ambient UV-B (90% of ambient UV-B) in Buenos Aires, Argentina, and decomposed the litter under reduced or near-ambient solar UV-B for 29 months in Tierra del Fuego, Argentina. We found that the UV-B treatment applied during growth decreased the decay rate. On the other hand, there was a marginally significant direct effect of elevated UV-B during the early stages of decomposition, suggesting increased mass loss. The effect of UV-B during growth on decomposition was likely the result of changes in plant litter chemical composition. Near-ambient UV-B received during plant growth decreased the concentrations of nitrogen, soluble carbohydrates, and N/P ratio, and increased the concentrations of phosphorus, cellulose, UV-B-absorbing compounds, and lignin/N ratio. Thus, solar UV-B radiation affects the decomposition of barley litter directly and indirectly, and indirect effects are persistent for the whole decomposition period.

The influence of solar UV-A and UV-B radiation at Beltsville, Maryland, on growth and flavonoid content in four cultivars of Cucumis sativus L. (Ashley, Poinsett, Marketmore, and Salad Bush cucumber) was examined during the summers of 1994 and 1995. Plants were grown from seed in UV exclusion chambers consisting of UV-transmitting Plexiglas, lined with Llumar to exclude UV-A and UV-B, polyester to exclude UV-B, or cellulose acetate to transmit UV-A and UV-B. Despite previously determined differences in sensitivity to supplemental UV-B radiation, all four cultivars responded similarly to UV-B exclusion treatment. After 19–21 days, the four cultivars grown in the absence of solar UV-B (polyester) had an average of 34, 55, and 40% greater biomass of leaves, stems, and roots, respectively, 27% greater stem height, and 35% greater leaf area than those grown under ambient UV-B (cellulose acetate). Plants protected from UV-A radiation as well (Llumar) showed an additional 14 and 22% average increase, respectively, in biomass of leaves and stems, and a 22 and 19% average increase, respectively, in stem elongation and leaf area over those grown under polyester. These findings demonstrate the extreme sensitivity of cucumber not only to present levels of UV-B but also to UV-A and suggest that even small changes in ozone depletion may have important biological consequences for certain plant species.

Field experiments were conducted to determine the effects of exclusion of UV-A/B and UV-B alone on growth parameters, activity of antioxidant enzymes, level of antioxidants and yield, to evaluate the intra-specific variations in sensitivity of these responses in eight soybean (Glycine max) varieties (PK-472, PK-1029, Pusa-24, JS-7105, JS-335, Hardee, NRC-7 and Kalitur). The plants were grown in specially designed UV-exclusion chambers which lined with selective UV filters to exclude either UV-B (<320 nm) or UV-A/B (<400 nm). Plants grown under UV-exclusion filters were compared with those grown under polythene filter which transmitted ambient UV-B and UV-A radiation. The results indicate that the exclusion of solar UV-B and UV-A/B enhanced the vegetative growth (plant height and leaf area), total biomass accumulation and yield (number of seeds and seed weight) of all the varieties as compared with those grown under ambient UV. The activities of SOD, GPX, APX and GR, and levels of ASA were significantly decreased, while α-tocopherol increased after the exclusion of UV-B and UV-A/B in all varieties. These results suggest that the ambient level of UV-B and UV-A radiation evoked some active oxygen species to accumulate, which in turn retarded the growth, development and yield of soybean varieties. On the basis of biomass, UV-B (280–315 nm) sensitivity can be arranged in decreasing order as PK-472 > Hardee > JS-335 > Kalitur > JS 71-05 > Pusa-24 > NRC-7 > PK-1029 and UV-A/B sensitivity can be arranged in decreasing order as PK-472 > Kalitur > JS-335 > Hardee > Pusa-24 > JS 71-05 > NRC-7 > PK-1029. The results indicate var. PK-472 is more sensitive than other varieties and PK-1029 is least sensitive to ambient level of UV radiation. This study in the area of UV-B and UV-A/B stress provides an extensive data that can be used as a predictive basis in crop sciences to further investigate some of the tolerant varieties in field studies.

Solar ultraviolet (UV)-B radiation (280-315 nm) has a wide range of effects on terrestrial ecosystems, yet our understanding of how UV-B influences the complex interactions of plants with pest, pathogen and related microorganisms remains limited. Here, we report the results of a series of experiments in Lactuca sativa which aimed to characterize not only key plant responses to UV radiation in a field environment but also consequential effects for plant interactions with a sap-feeding insect, two model plant pathogens and phylloplane microorganism populations. Three spectrally modifying filters with contrasting UV transmissions were used to filter ambient sunlight, and when compared with our UV-inclusive filter, L. sativa plants grown in a zero UV-B environment showed significantly increased shoot fresh weight, reduced foliar pigment concentrations and suppressed population growth of green peach aphid (Myzus persicae). Plants grown under a filter which allowed partial transmission of UV-A radiation and negligible UV-B transmission showed increased density of leaf surface phylloplane microbes compared with the UV-inclusive treatment. Effects of UV treatment on the severity of two plant pathogens, Bremia lactucae and Botrytis cinerea, were complex as both the UV-inclusive and zero UV-B filters reduced the severity of pathogen persistence. These results are discussed with reference to known spectral responses of plants, insects and microorganisms, and contrasted with established fundamental responses of plants and other organisms to solar UV radiation, with particular emphasis on the need for future integration between different experimental approaches when investigating the effects of solar UV radiation.

Very few studies have evaluated the effects of UV-B radiation on trees. especially deciduous species. In this study we evaluate the effects of supplemental UV-B radiation on the growth and photosynthetic capacity of sweetgum (Liquidambar styraciflua L.). Sweetgum seedlings were grown for 2 years in the field under either ambient or supplemental UV-B radiation. Artificial UV-B radiation was supplied by fluorescent lamps at a maximum daily supplementation of either 3.1 or 5.0 kJ of biologically effective UV-B radiation. Over the 2-year period, supplemental UV-B radiation had little effect on total plant biomass or photosynthetic capacity. However, subtle changes in leaf physiology, carbon allocation, and growth were observed. Supplemental UV-B radiation reduced photosynthetic capacity only during the first year, while leaf area and biomass were reduced in the second year. Alterations in carbon allocation included an increase in branch number and root to shoot ratio. While these data do not indicate that the productivity of sweetgum would likely be compromised by an increase in solar UV-B radiation, they do suggest that the UV-B portion of the solar spectrum contributes to the regulation of sweetgum growth and development. Therefore the possibility of significant consequences to sweetgum due to possible increases in UV-B radiation cannot be ruled out.

UV-B radiation is absorbed effectively by nucleic acids and other sensitive targets, potentially causing harmful photochemical effects. Protection against UV-B radiation may be afforded by flavonoids and other phenolics, which absorb strongly in the UV region, but little is known about the role played by other compounds, such as terpenes. Grindelia chiloensis, native of Patagonia (Argentina), can accumulate as much as 25% resin (terpenes) in its leaves. The present investigation was carried out to test the effect of solar UV-B radiation on the allocation of photoassimilates to biomass and terpenes. Exposure to UV-B radiation reduced whole plant biomass, plant height and leaf area, and increased leaf thickness and resin accumulation in Grindelia chiloensis. Higher absorbance was found for refined resin in the UV-B waveband from plants grown under solar UV-B radiation than plants without UV-B radiation. These chemical and structural changes could protect the plant from UV radiation, and may help elucidate the importance of epicuticular resins for a species as G. chiloensis native to an environment with maximum daily integrated values of solar UV-B irradiance.

The impact of simulated solar radiation on DNA and the mitigation of DNA-damaging effects by photoreactivation was studied in a cyanobacterium Anabaena variabilis PCC 7937. Cultures were irradiated under 295, 320 and 395 nm cut-off filters as well as seven other filters such as WG 280, WG 295, WG 305, WG 320, WG 335, WG 345 and GG 400. Growth of the test organism was found to be affected mostly under UV-B radiation as compared to PAR and PAR + UV-A radiations. Amplification of 16s rDNA and RAPD profile was significantly affected following exposure of genomic DNA to UV-B radiation. The formation of T<>T CPDs was recorded only in the cultures irradiated with UV-B radiation (i.e., under 295 nm as well as under WG 280, WG 295 and WG 305 nm cut-off filters), but maximum yield was found under 280 nm cut-off filter. Furthermore, the considerable induction of thymine dimers was observed with increasing UV-irradiation times. Fluorometric analysis of DNA unwinding (FADU) assay for UV-induced DNA strand breaks exhibited the maximum loss in the percentage of dsDNA under UV-B radiation followed by UV-A and PAR in comparison to the light control samples. We observed that T<>T CPD repair is light-dependent, since these lesions were more efficiently removed upon exposure to visible light than in the darkness. Blue radiation was found to be the most effective in photoreactivation than any other wavebands of light. Furthermore, the rate of photoreactivation was measured under varying temperatures (10, 20 and 30 °C); the repair rate was found to be the maximum at 20 °C under white fluorescent light. Our results indicate that photoreactivation play an important role in survival of the organism under natural conditions in spite of being exposed to the UV-B component present in the solar drops.
