The hexaploid wheat (Triticum aestivum) adult plant resistance gene, Lr34/Yr18/Sr57/Pm38/Ltn1, provides broad‐spectrum resistance to wheat leaf rust (Lr34), stripe rust (Yr18), stem rust (Sr57) and powdery mildew (Pm38) pathogens, and has remained effective in wheat crops for many decades. The partial resistance provided by this gene is only apparent in adult plants and not effective in field‐grown seedlings. Lr34 also causes leaf tip necrosis (Ltn1) in mature adult plant leaves when grown under field conditions. This D genome‐encoded bread wheat gene was transferred to tetraploid durum wheat (T. turgidum) cultivar Stewart by transformation. Transgenic durum lines were produced with elevated gene expression levels when compared with the endogenous hexaploid gene. Unlike nontransgenic hexaploid and durum control lines, these transgenic plants showed robust seedling resistance to pathogens causing wheat leaf rust, stripe rust and powdery mildew disease. The effectiveness of seedling resistance against each pathogen correlated with the level of transgene expression. No evidence of accelerated leaf necrosis or up‐regulation of senescence gene markers was apparent in these seedlings, suggesting senescence is not required for Lr34 resistance, although leaf tip necrosis occurred in mature plant flag leaves. Several abiotic stress‐response genes were up‐regulated in these seedlings in the absence of rust infection as previously observed in adult plant flag leaves of hexaploid wheat. Increasing day length significantly increased Lr34 seedling resistance. These data demonstrate that expression of a highly durable, broad‐spectrum adult plant resistance gene can be modified to provide seedling resistance in durum wheat.  相似文献

The rusts of wheat are important fungal plant pathogens that can be disseminated thousands of kilometers across continents and oceans by wind. Rusts are obligate parasites that interact with resistance genes in wheat in a gene-for-gene manner. New races of rust develop by mutation and selection for virulence against rust resistance genes in wheat. In recent years, new races of wheat leaf rust, wheat stripe rust, and wheat stem rust have been introduced into wheat production areas in different continents. These introductions have complicated efforts to develop wheat cultivars with durable rust resistance and have reduced the number of effective rust-resistance genes that are available for use. The migration patterns of wheat rusts are characterized by identifying their virulence against important rust resistance genes in wheat and by the use of molecular markers.  相似文献

Abstract

Leaf rust of wheat (Triticum aestivum L.), incited by Puccinia recondita ex Desm. f. sp. tritici Eriks, is one of the most important wheat diseases in Egypt. Methyl jasmonate (MJ) is a potential plant elicitor which induces a wide range of chemical and anatomical defense reactions in conifers and might be used to increase systemic resistance against biotic damage. In the greenhouse, different concentrations of MJ (10, 20 and 30 mM) were applied as seed soaking plus foliar spray or only as foliar spray to control leaf rust and induction of secondary compound production in leaves of wheat plants. Foliar spray was applied after 30 and 50 days of sowing. Results indicated that all concentrations and treatments reduced the severity of rust disease caused by P. recondita f. sp. tritici in wheat leaves during 45 days of inoculations. Disease incidence was decreased significantly in MJ-treated plants as seed soaking plus foliar spray with 20 and 30 mM when compared to 10 mM MJ or control plants. The study revealed that, with increasing concentrations of MJ, the secondary metabolites were greatly increased. Endogenous levels of both free and conjugated putrescine, spermidine and spermine increased in response to the elicitor. Activities of polyamine biosynthetic enzymes of ornithine decarboxylase (ODC) and polyamine oxidase (PAO) displayed up to threefold increases relative to untreated control. Moreover, significant increases in activities of plant defense-related protein, enzymes as peroxidase and chitinase as well as free and conjugated phenols contents were recorded in treated plants compared with untreated and infected plants. Furthermore, MJ treatment increased the chlorophyll-a, chlorophyll-b and carotenoids pigments contents, the higher increase was obtained with combined treatment between seeds soaking plus foliar spray at 20 and 30 mM of MJ. Under field conditions, three concentrations of MJ, i.e. 10, 20 and 30 mM as combined treatment between seeds soaking plus foliar spray or only as foliar spray were applied to study their effect against rust disease. Foliar spray was applied after 30 and 80 days of sowing. Results showed that the high reduction in disease severity was obtained with combined treatments between seeds soaking plus foliar spray with MJ at 20 and 30 mM compared with other treatments and control. At the same time, all treatments increased the growth and grain yield of wheat plants. It could be suggested that combination treatment between seeds soaking plus foliar spray with methyl jasmonate might be used commercially for controlling rust disease of wheat plants under field conditions.  相似文献

 评估臭氧(O3)污染对农田冬小麦生长和产量的影响是污染生态学和生理生态学研究的重要内容之一。该研究运用开顶式气室(OTC),对冬小麦‘ 嘉403’(Triticum aestivum cv. Jia 403)进行了O3动态暴露的田间原位试验。实验设置过滤空气组(CF)、自然大气组(NF)和两个不同浓度的 O3动态暴露组(DO100和DO150)。结果表明:1) O3浓度增加,一方面可以改变灌浆期冬小麦叶片气体交换参数的日变化规律;另一方面引起表观 光量子产额、光饱和点和光补偿点等光响应参数的显著降低。这表明灌浆期叶片光合能力的下降是气孔因素和非气孔因素共同作用的结果。2) O3暴露可以改变小麦形态特征,如植株变矮、叶片衰老加速、 叶面积变小,并最终导致产量大幅下降。  相似文献

Reliable and accurate estimation of plant disease severity at the field scale is a key factor for predicting yield losses, disease management and food security. A field experiment was designed and conducted during 2017–18 and 2018–19 with 24 wheat cultivars to estimate the stripe rust severity by supervised classification of thermal and visible images using parallelepiped, minimum distance, mahalanobis distance, maximum likelihood, support vector machine and neural network methods of image classification. Results demonstrated the potential of thermal and visible imaging techniques to estimate wheat stripe rust severity with good accuracy. For both visible and thermal images used in this study, support vector machine gave the best estimates of the rust severity, while the parallelepiped method was the worst-performing method. Support vector machine and neural network methods showed d-index, Nash-Sutcliffe efficiency and coefficient of determination values above 85%, with accuracies above 98% and kappa coefficient above 0.97 for both thermal and visible images. Comparison of thermal and visible image classification performance revealed that for all the methods except support vector machine, the estimated rust severity, overall accuracy and kappa coefficient of thermal images were better than visible images. The present study clearly showed that both thermal and visible image analysis can be applied as a rapid non-destructive technique to estimate the wheat rust severity under field conditions. The study also provided a comparative insight into thermal and visible image classification methods that have great potential for sustainable plant disease management in modern agriculture.  相似文献

The Indian bread wheat cultivar HD2009 has maintained its partial resistance to leaf rust and stripe rust in India since its release in 1976. To examine the nature, number and mode of inheritance of its genes for partial leaf rust and stripe rust resistance, this cultivar was crossed with cultivar WL711, which is susceptible to leaf rust and stripe rust. The F1, F2, F3 and F5 generations from this cross were assessed separately for adult plant disease severity under artificial epidemic of race 77-5 of leaf rust and race 46S119 of stripe rust. Segregation for rust reaction in the F2, F3 and F5 generations indicated that resistance to each of these rust diseases is based on 2 genes, each with additive effects. Although the leaf rust resistance of HD2009 is similar in expression to that conferred by the gene Lr34, but unlike the wheats carrying this gene, cultivar HD2009 did not show leaf tip necrosis, a morphological marker believed to be tightly linked to the leaf rust resistance gene Lr34. Thus, the non-hypersensitive resistance of HD2009 was ascribed to genes other than Lr34.  相似文献

Summary The direct and indirect effects of plant genetic diversity on epidemics and the influence of disease on plant competition were investigated using the wheat (Triticum aestivum)/stripe rust (Puccinia striiformis) system. Replacement series consisting of a susceptible and a resistant wheat genotype or two wheat genotypes susceptible to different races of stripe rust were grown in the presence and absence of the pathogen. Stripe rust severity, number of seed heads, seed yield, and seed weight were determined separately for each wheat genotype in the mixtures and the pure stands. The frequency of susceptible genotypes in a mixture explained up to 67% of the variation in disease severity. However, competitive interactions among plant genotypes sometimes appeared to alter susceptibility and obscured the relationship. In pure stands of single genotypes, disease severity explained between 52 and 58% of the variation in seed yield. In mixtures, coefficients of determination were only 10 and 31%, suggesting a strong influence of plant-plant interactions on seed yield. These results suggest that host-parasite coevolutionary models need to account for the strong effect that specific plant genotype combinations may have on disease severity and plant reproduction.Paper No. 9818 of the journal series of the Oregon Agricultural Experiment Station  相似文献

Rust fungi of the order Pucciniales are destructive pathogens of wheat worldwide. Leaf rust caused by the obligate, biotrophic basidiomycete fungus Puccinia triticina (Pt) is an economically important disease capable of causing up to 50 % yield losses. Historically, resistant wheat cultivars have been used to control leaf rust, but genetic resistance is ephemeral and breaks down with the emergence of new virulent Pt races. There is a need to develop alternative measures for control of leaf rust in wheat. Development of transgenic wheat expressing an antifungal defensin offers a promising approach to complement the endogenous resistance genes within the wheat germplasm for durable resistance to Pt. To that end, two different wheat genotypes, Bobwhite and Xin Chun 9 were transformed with a chimeric gene encoding an apoplast-targeted antifungal plant defensin MtDEF4.2 from Medicago truncatula. Transgenic lines from four independent events were further characterized. Homozygous transgenic wheat lines expressing MtDEF4.2 displayed resistance to Pt race MCPSS relative to the non-transgenic controls in growth chamber bioassays. Histopathological analysis suggested the presence of both pre- and posthaustorial resistance to leaf rust in these transgenic lines. MtDEF4.2 did not, however, affect the root colonization of a beneficial arbuscular mycorrhizal fungus Rhizophagus irregularis. This study demonstrates that the expression of apoplast-targeted plant defensin MtDEF4.2 can provide substantial resistance to an economically important leaf rust disease in transgenic wheat without negatively impacting its symbiotic relationship with the beneficial mycorrhizal fungus.  相似文献

Acyl lipids and pigments were analyzed in young plants of garden pea, spring wheat and spinach exposed to < 5 or 65 nl l?1 ozone 12 h per day for 6 days. In one set of experiments, the plants were exposed to 14CO2 for 2 h 3 days prior to ozone exposure. The plants responded differently to the moderately enhanced level of ozone used Spinach was not at all sensitive while in both pea and wheat, leaves of different ages differed in ozone sensitivity. In pea, ozone sensitivity increased with leaf age. In the second and third oldest leaves, the amounts of galactolipids per leaf area and the proportions of 18:3 of the total lipid extract and of phosphatidylglycerol decreased. In the second oldest leaf, ozone also caused a decreased proportion of 18:3 of monogalactosyldiacylglycerol. In the fourth oldest leaf, lipid composition and galactolipid unsaturation was unaffected, but ozone caused decreased leaf expansion resulting in increased acyl lipid content per leaf area. In both the first and second leaves of wheat, ozone fumigation caused a marked decrease in the content of monogalactosyldiacylglycerol and in the first leaf, the contents of phosphatidylcholine and phosphatidylethanolamine increased. The proportion of 18:3 in phosphatidylcholine was larger in ozone-fumigated than in control plants, while the reverse applied for phosphatidylglycerol. In the oldest sampled leaves of pea and wheat, ozone caused an increase in the radioactivity associated with β-carotene, indicating increased turnover. Thus, while spinach was unaffected, in both pea and wheat ozone caused a decrease in the proportion of chloroplast membrane lipids to non-chloroplast membrane lipids in older leaves while younger leaves were less sensitive.  相似文献

It is important to implement detection and assessment of plant diseases based on remotely sensed data for disease monitoring and control. Hyperspectral data of healthy leaves, leaves in incubation period and leaves in diseased period of wheat stripe rust and wheat leaf rust were collected under in-field conditions using a black-paper-based measuring method developed in this study. After data preprocessing, the models to identify the diseases were built using distinguished partial least squares (DPLS) and support vector machine (SVM), and the disease severity inversion models of stripe rust and the disease severity inversion models of leaf rust were built using quantitative partial least squares (QPLS) and support vector regression (SVR). All the models were validated by using leave-one-out cross validation and external validation. The diseases could be discriminated using both distinguished partial least squares and support vector machine with the accuracies of more than 99%. For each wheat rust, disease severity levels were accurately retrieved using both the optimal QPLS models and the optimal SVR models with the coefficients of determination (R2) of more than 0.90 and the root mean square errors (RMSE) of less than 0.15. The results demonstrated that identification and severity evaluation of stripe rust and leaf rust at the leaf level could be implemented based on the hyperspectral data acquired using the developed method. A scientific basis was provided for implementing disease monitoring by using aerial and space remote sensing technologies.  相似文献

In glasshouse experiments with plants in pots, applications of potassium chloride to the soil at 0.5 g/plant, 4 days before inoculation with Puccinia striiformis, decreased the severity of yellow rust on several winter wheat cultivars in comparison with untreated plants. Conversely, yellow rust was encouraged by applications of sodium nitrate. Sodium chloride in solution (8.6 g/l) reduced yellow rust when applied to the soil at the rate of 20 ml/plant but not when it was sprayed on to the leaves. In small-plot field experiments, sodium and potassium chlorides applied to the soil as dry powders in the spring at rates of 376, 1130 or 2260 kg/ha, significantly decreased the severity of yellow rust on most of the winter wheat cvs examined at each rate. The chlorides at these rates did not adversely affect the overall growth or yield in the absence of yellow rust.  相似文献

Abstract

‘Santa Fe’ is a hard red winter cultivar that has had long-lasting resistance to the leaf rust fungus, Puccinia triticina. The objective of this study was to determine the chromosome location of the adult plant leaf rust resistance in Santa Fe wheat. A partial backcross line of ‘Thatcher’ (Tc) wheat with adult plant leaf rust resistance derived from Santa Fe was crossed with Thatcher to develop a Thatcher//Tc*2/Santa Fe F6 recombinant inbred line (RIL) population. The RIL population and parental lines were evaluated for segregation of leaf rust resistance in three field plot tests and in an adult plant greenhouse test. A genetic map of the RIL population was constructed using 90,000 single-nucleotide polymorphism (SNP) markers with the Illumina Infinium iSelect 90K wheat bead array. A significant quantitative trait locus for reduction of leaf rust severity in all four tests was found on chromosome 3BL that segregated as a single adult plant resistance gene. The RILs with the allele from the resistant parent for SNP marker IWB10344 had lower leaf rust severity and a moderately resistant to moderately susceptible response compared to the susceptible RILs and Thatcher. The gene derived from Santa Fe on chromosome 3BL was designated as Lr77. Kompetitive allele-specific polymerase chain reaction assay markers linked to Lr77 on 3BL should be useful for selection of wheat germplasm with this gene.

通过荧光显微镜和扫描电镜分别对条形柄锈菌夏孢子在寄主植物-小麦叶表和非寄主植物-水稻叶表以及小麦穗部和茎秆上的萌发过程进行了观察。结果发现,夏孢子在小麦叶片体表萌发产生芽管后,可依次分化形成气孔下囊、初生菌丝与吸器母细胞;在小麦颖片、稃片及茎秆部位表面,同样可观察到病菌在体外分化形成吸器母细胞;并且在水稻叶片上也观察到病菌侵染结构存在体外分化现象。经荧光染色发现,条形柄锈菌在体外与在小麦组织中形成的侵染结构没有明显的差别。观察结果可为条形柄锈菌侵染结构的离体诱导与调控机理研究提供依据。  相似文献

Rust diseases are a major cause of yield loss in wheat worldwide, and are often controlled through the incorporation of resistance genes using conventional phenotypic selection methods. Slow-rusting resistance genes are expressed quantitatively and are typically small in genetic effect thereby requiring multiple genes to provide adequate protection against pathogens. These effects are valuable and are generally considered to confer durable resistance. Therefore an understanding of the chromosomal locations of such genes and their biological effects are important in order to ensure they are suitably deployed in elite germplasm. Attila is an important wheat grown throughout the world and is used as a slow-rusting donor in international spring wheat breeding programs. This study identified chromosomal regions associated with leaf rust and stripe rust resistances in a cross between Attila and a susceptible parent, Avocet-S, evaluated over 3 years in the field. Genotypic variation for both rusts was large and repeatable with line-mean heritabilities of 94% for leaf rust resistance and 87% for stripe rust. Three loci, including Lr46/Yr29 on chromosome 1BL, were shown to provide resistance to leaf rust whereas six loci with small effects conferred stripe rust resistance, with a seventh locus having an effect only by epistasis. Disease scoring over three different years enabled inferences to be made relating to stripe rust pathogen strains that predominated in different years.  相似文献

Abstract:  A convertible glasshouse was established to study annual transgenic plants under near-field environmental conditions while simultaneously ensuring a high level of biological containment. This system can provide a useful step in the assessment of transgenic plants prior to open-field experiments. Two transgenic wheat lines (cv. Bobwhite) were investigated and compared with their corresponding non-transformed wildtypes with respect to plant performance, expression of the transgenic trait and interactions with antagonists. The first line expressed snowdrop lectin [ Galanthus nivalis agglutinin (GNA)] for enhanced resistance to aphids, and the second one overexpressed the endogenous Lr10 gene to enhance resistance to leaf rust. Interestingly, 1000-kernel weight of Lr10 -transgenic plants was significantly reduced, indicating that the overexpression of the Lr10 gene caused a significant fitness cost for the plant. GNA-transgenic plants expressed the lectin at levels too low to affect the target aphids. A detached leaf bioassay with Lr10 -transgenic plants revealed an increased resistance to leaf rust. No differences in the performance of aphids or cereal leaf beetles on transgenic and non-transformed plants were recorded in the convertible glasshouse and in complementary glasshouse studies. Similarly, infection levels with powdery mildew did not differ between transgenic and non-transformed plants but Bobwhite plants were significantly more infected when compared with conventional Swiss spring wheat cultivars. Overall, the assessment revealed that for the plants investigated here, their genetic background had a stronger impact on the performance of a plant and its interactions with insect herbivores and pathogens than the expression of the transgene.  相似文献

Wheat leaf rust, stem rust, stripe rust, and powdery mildew caused by the fungal pathogens Puccinia triticina, P. graminis f. sp. tritici, P. striiformis f. sp. tritici, and Blumeria graminis f. sp. tritici, respectively, are destructive diseases of wheat worldwide. Breeding durable disease resistance cultivars rely largely on continually introgressing new resistance genes, especially the genes with different defense mechanisms, into adapted varieties. Here, we describe a new resistance gene obtained by mutagenesis. The mutant, MNR220 (mutagenesis-derived new resistance), enhances resistance to three rusts and powdery mildew, with the characteristics of delayed disease development at the seedling stage and completed resistance at the adult plant stage. Genetic analysis demonstrated that the resistance in MNR220 is conferred by a single semidominant gene mapped on the short arm of chromosome 2B. Gene expression profiling of several pathogenesis-related genes indicated that MNR220 has an elevated and rapid pathogen-induced response. In addition to its potential use in breeding for resistance to multiple diseases, high-resolution mapping and cloning of the disease resistance locus in MNR220 may lead to a better understanding of the regulation of defense responses in wheat.  相似文献

Plants of the winter wheat cv. Bounty were inoculated with Septoria nodorum and then assayed for ergosterol. A detached leaf technique was used in which leaf segments were incubated on agar containing benzimidazole. Four levels of a conidial suspension inoculum were applied using a measured droplet technique. Ergosterol in plant extracts was measured using HPLC and its identity confirmed by co-injection with pure ergosterol as an internal standard. The extracts were also assayed by silica-gel thin layer chromatography with the sterol being visualised with rhodamine; the presence of ergosterol in the material from the two higher disease level treatments was confirmed by analysing the eluted spots in a spectrophotometer. Additional confirmation of ergosterol was obtained using gas-liquid chromatography comparing a mixture of known plant sterols with a sample from the diseased leaf tissue. The ergosterol assay was found to be very sensitive and offers a high level of reproducibility. It would therefore appear that such assays could be of value in breeding programmes when it is necessary to screen wheat cultivars for their reaction to S. nodorum or other fungal pathogens.  相似文献

Stripe rust and leaf rust, caused by Puccinia striiformis Westend. f. sp. tritici Erikss. and P. triticina, respectively, are devastating fungal diseases of common wheat (Triticum aestivum L.). Chinese wheat cultivar Bainong 64 has maintained acceptable adult-plant resistance (APR) to stripe rust, leaf rust and powdery mildew for more than 10?years. The aim of this study was to identify quantitative trait loci/locus (QTL) for resistance to the two rusts in a population of 179 doubled haploid (DH) lines derived from Bainong 64?×?Jingshuang 16. The DH lines were planted in randomized complete blocks with three replicates at four locations. Stripe rust tests were conducted using a mixture of currently prevalent P. striiformis races, and leaf rust tests were performed with P. triticina race THTT. Leaf rust severities were scored two or three times, whereas maximum disease severities (MDS) were recorded for stripe rust. Using bulked segregant analysis (BSA) and simple sequence repeat (SSR) markers, five independent loci for APR to two rusts were detected. The QTL on chromosomes 1BL and 6BS contributed by Bainong 64 conferred resistance to both diseases. The loci identified on chromosomes 7AS and 4DL had minor effects on stripe rust response, whereas another locus, close to the centromere on chromosome 6BS, had a significant effect only on leaf rust response. The loci located on chromosomes 1BL and 4DL also had significant effects on powdery mildew response. These were located at the same positions as the Yr29/Lr46 and Yr46/Lr67 genes, respectively. The multiple disease resistance locus for APR on chromosome 6BS appears to be new. All three genes and their closely linked molecular markers could be used in breeding wheat cultivars with durable resistance to multiple diseases.  相似文献
