甘蔗渣纤维素降解菌的筛选及鉴定   总被引:1,自引:0,他引:1  
通过利用多种选择性培养基,从自然发酵不同阶段的甘蔗渣中分离到多种纤维素分解菌,经过初筛和复筛,获得了降解纤维素的功能菌株c1g3-3及其最适功能培养基蛋白胨纤维素培养基(PCS),并通过形态、生理生化和分子综合鉴定得出c1g3-3鉴定为木糖氧化无色杆菌(Achrom obacter xylosoxidans)。  相似文献

甘蔗渣是制糖工业的主要副产物。筛选甘蔗渣纤维素降解菌株对甘蔗渣乙醇产业具有重要的意义。以甘蔗渣为原料,通过分离和纯化得到14株菌株,对其进行纤维素刚果红平板染色实验和滤纸崩解实验,最终获得3株可以生产纤维素酶的菌株02-2-2、21-1-2和40-1-1。酶活性测定结果表明,菌株40-1-1的酶活力在培养3 d后达到最高,为27.26 U/mg。通过形态学和分子生物学鉴定,菌株02-2-2为枝顶孢属(Acremonium sp.),菌株21-1-2和40-1-1为光滑短梗霉属(Acrophialophora sp.)。研究筛选的菌株将为开展甘蔗渣纤维素降解利用提供参考。  相似文献

以新华滤纸为唯一碳源,从垃圾堆肥中筛选能够分解纤维素的菌株共39株,采用刚果红鉴别培养基进行识别,获取透明圈较大的菌株10株,在此基础上,进行液体培养,测定酶活,得到1株酶活较高的曲霉B-6(Aspergillus sp)。将B-6与绿色木霉(Trichoderma sp)AS3.3711进行了参比试验,比对筛选工作进行评定,经过固体,液体发酵对比试验,发现B-6与AS3.371有相近的产酶性能,B-6在固,液发酵中酶活分别达到39.2IU,14.9IU,而S3.3711则分别为16.6IU与15.7IU,且B-6较AS3.3711有更强的液化CMC的能力,B-6在24h内即能使3%CMC完全液化,而S3.3711则需96h。  相似文献

目的初步筛选牛瘤胃中纤维素降解菌。方法分别采用基本培养基(牛肉膏蛋白胨培养基、马丁培养基),利用好氧、兼性和厌氧3种不同的培养方法进行初选,初步分离牛瘤胃中的细菌与真菌,再通过复选培养基(加入微晶纤维素),筛选降解纤维素的菌种。结果筛选分离出降解纤维素的1株厌氧细菌和1株厌氧真菌。结论此实验方法简单易行,能够有效地从牛瘤胃中筛选出生长良好的纤维素降解菌。  相似文献

目的 针对大庆地区土地盐碱化较严重,盐碱面积较大等现状,从大庆特征性盐碱地区土壤样品中分离出能够降解纤维素的菌株,为降解植物废料以及缓解土壤盐碱化改善土壤环境提供功能菌株。方法 通过刚果红染色法初步筛得到具有纤维素降解能力菌株,进一步用比色法测定其纤维素降解率,同时测定菌株耐盐、耐碱、产酸性能,选择性能优良的3株菌作为纤维素降解菌复合菌系构建菌株,通过耐盐性、耐碱性和纤维素降解率实验测定复合菌系菌株最佳组合,进一步通过上述实验确定复合菌系菌株最佳混合比例。结果 得到由DX-5和DX-9按照2∶3进行组合复合菌系纤维素降解率最高,达到87.96%,且具有较高的耐盐以及耐碱能力。结论 通过实验得到纤维素降解菌的复合菌系,具有较高纤维素降解率以及改善土壤盐碱性能力。  相似文献

从发酵床垫料中初步分离出43株纤维素降解菌。采用刚果红鉴别培养基及滤纸条培养基初筛,得到5株透明圈较大且使滤纸条产生崩解的菌株,通过进一步液体发酵,测定其CMC酶活、FPA酶活和天然纤维素酶活,获得2株具有较高纤维素降解活性菌株,并分别命名为F7和F21。经16S rRNA基因序列分子生物学鉴定和系统发育分析表明,这2株纤维素降解菌分别归属为枯草芽孢杆菌(Bacillus subtilis)和链霉菌(Streptomyces sp.)。  相似文献

三株高效秸秆纤维素降解真菌的筛选及其降解效果   总被引:25,自引:0,他引:25  
【目的】利用多种筛选方法,获得高效秸秆纤维素降解真菌,并研究其秸秆纤维素的降解能力。【方法】采用滤纸片孔洞法、滤纸条降解法、羧甲基纤维素钠(CMC-Na)水解圈测定法、秸秆失重法、纤维素分解率测定法、胞外酶活测定法等常规秸秆纤维素降解菌的筛选方法。【结果】筛选到3株具有较强纤维素降解能力的真菌菌株,经初步鉴定菌株98MJ为草酸青霉(Penicillium oxalicum)、菌株W3为木霉(Trichoderma sp.)、菌株W4为扩张青霉(Penicillium expansum)。菌株W4具有非常强的秸秆纤维素降解能力,10d内对秸秆的降解率可达56.3%,对纤维素、半纤维素和木质素的分解率分别为59.06%、78.75%和33.79%。菌株W4的胞外纤维素酶活力在14.25-49.75U/mL之间。【结论】筛选获得3株高效秸秆纤维素降解真菌菌株,其中菌株W4的纤维素酶活高于已报道的菌株,是一株十分具有研究开发潜力的纤维素酶生产菌株。  相似文献

从生产烯唑醇的农药厂废水处理池的活性污泥中驯化、分离得到6株能以烯唑醇为唯一碳源士长的细菌,分别命名为1#、2#、3#、4#、5#、6#。实验结果表明,这六种菌都足好氧菌种,对烯唑醇降解率大于30%的有1#、3#、4#、6#,其中1#为优势菌种。该优势菌降解烯唑醇的最适温度为30~35℃,最适pH值为7.0,投加0.404mg/L的菌株母液量为10%,烯唑醇的最佳初始浓度为30mg/L。该菌在上述最适条件下,150r/min摇床培养6d,埘怖唑醇的降解率可达78.14%;优势菌1#单菌对烯唑醇的降解要比复合菌对其降解的效果好。  相似文献

纤维素高效降解菌YN1的筛选及其降解特性   总被引:3,自引:3,他引:3  
为了获得降解天然纤维素的微生物菌株,并用于农田秸秆腐熟及竹林地稻壳促腐。从牛羊粪堆肥中筛选出一株纤维素降解菌YN1,对其进行了形态和系统发育分析;测定了YN1的液、固体发酵酶活;进行了YN1对纤维素的失重测定;对YN1在纤维素表面上的定殖及其对纤维素的降解进行了电镜观察。经菌株形态分析和ITS基因序列分析,将YN1鉴定为曲霉(Aspergillussp.);在YN1菌株的液体发酵培养第3天,内切酶、外切酶、β-糖苷酶和总纤维素酶活性都达到最大值,分别为95.7、14.6、20.5和26.6U/mL,固体发酵培养第5d各酶活均达到峰值,依次为1192.2、100.6、136.9和210.7U/g;在失重测定中,YN1在7d内可降解41.87%的秸秆,31.59%的稻壳;在扫描电镜下可明显观察到YN1对滤纸、秸秆和稻壳的降解。YN1可有效降解天然纤维素,对农田秸秆腐熟及竹林地稻壳促腐具有较好的应用前景。  相似文献

The interactions between organisms and their environments can shape distributions of spatial genetic variation, resulting in patterns of isolation by environment (IBE) in which genetic and environmental distances are positively correlated, independent of geographic distance. IBE represents one of the most important patterns that results from the ways in which landscape heterogeneity influences gene flow and population connectivity, but it has only recently been examined in studies of ecological and landscape genetics. Nevertheless, the study of IBE presents valuable opportunities to investigate how spatial heterogeneity in ecological processes, agents of selection and environmental variables contributes to genetic divergence in nature. New and increasingly sophisticated studies of IBE in natural systems are poised to make significant contributions to our understanding of the role of ecology in genetic divergence and of modes of differentiation both within and between species. Here, we describe the underlying ecological processes that can generate patterns of IBE, examine its implications for a wide variety of disciplines and outline several areas of future research that can answer pressing questions about the ecological basis of genetic diversity.  相似文献

Reported in this study are the experimental design and results of an immunosensor for the detection of the explosive, 2,4,6-trinitrotoluene (TNT) in seawater using a reversed-displacement format. This reversed-displacement immunosensor methodology has successfully measured TNT in seawater by direct injection, eliminating the need for preconcentration or pretreatment of samples. A microcolumn containing an Affi-Gel resin derivatized with a 2,4,6-trinitrobenzene (TNB) moiety and a fluorophore-labeled anti-TNT antibody composed the immunoassay reactive chamber. Fluorophore-labeled anti-TNT antibody was incubated with the modified Affi-Gel resin until binding equilibrium was reached. Under a constant flow, samples containing TNT were introduced into the flow stream displacing the fluorophore-labeled TNT antibody. Limits of detection were 2.5ng/mL or part-per-billion (ppb) for TNT in saline buffer and 25ppb in seawater with an analysis time of 10 min. Two anti-TNT antibodies with differing binding affinities were compared in the reversed-displacement assay format, and a correlation between affinity and detection limits was observed. Furthermore, we have demonstrated that the reversed-displacement format can be used to screen seawater samples containing TNT, remains effective after dozens of cycles, and provides significant fluorescence response before regeneration is required.  相似文献

An obligately chemolithoautotrophic and aerobic hydrogen-oxidizing bacterium was isolated from seawater of the Shonan Coast, Kanagawa Pref., Japan. The isolate was a Gram-negative, comma-shaped rod cell measuring 0.2 to 0.5 by 1 to 2 m. The cells occurred singly and were motile by a polar flagellum. The deoxyribonucleic acid base composition of the isolate was 44.1 mol% guanine plus cytosine. The optimal temperature for autotrophic growth on H2–O2–CO2 was around 37°C, and no growth was observed at 5° C or 45° C. The optimal pH for growth was around 6.5. NaCl was required for growth with an optimum of 0.5 M. Elemental sulfur, thiosulfate or tetrathionate was utilized, as well as molecular hydrogen, as the sole energy source. No heterotrophic growth was observed on organic media tested. To our knowledge, this is the first report of the isolation of a marine, aerobic hydrogen-oxidizing bacterium, and of an obligately chemolithoautotrophic, mesophilic and aerobic hydrogen-oxidizing bacterium.  相似文献

利用化学全分析、XRD分析、原子力显微镜等方法对内蒙古产黑色电气石进行测试表征,并研究了该电气石对海水pH值的影响,探讨了电气石用量、海水盐度、初始pH值等因素对pH调控的影响.结果表明,电气石能够调节初始pH值为3和10的海水pH值分别至7.9和8.1;电气石对低盐度海水pH调控速率大于高盐度海水,处理120 min后初始pH均为5,盐度为5、10、15、20、35的海水pH分别增加了3.2、3.16、3.06、2.99、2.85;电气石对海水电导率基本无影响.本研究为将电气石应用于调控水产养殖水体pH提供了实验基础.  相似文献

The screening of catalysts, substrates or conditions in the early stages of bioprocess development requires an enormous number of experiments and is a tedious, expensive and time-consuming task. Currently available screening systems can only be operated in batch or fed-batch mode, which can lead to severe misinterpretations of screening results. For example, catalysts that are inhibited by substrates or accumulating products will be excluded from further investigations in the early stages of process development despite the fact that they might be superior to other candidates in a different operational mode. Important and advantageous properties such as turnover stability can also be overshadowed by product inhibition. The aim of this study was to develop a novel screening system that enables continuous feeding of substrates and continuous removal of products. A prototype based on the membrane reactor concept was designed and operated for a model reaction, the hydrolysis of cellulose.  相似文献

内生细菌存在于健康植物体内,一些内生细菌具有促生长、抗病和固氮等生物学功能.本项研究采用化学药剂表面灭菌方法从黑龙江省大豆品种合丰25的根、茎、叶和种子中分离到大量内生细菌,其种群数量在根部最多,为3.4×103CFU/g,在叶部次之,为2.8×103CFU/g,在茎部和种子中最少,为2.9×102 CFU/g和1.4×102CFU/g.从121株内生细菌中筛选到31株对大豆根腐病菌Fusarium oxysporum f. sp.soybean具有较强抑制作用的拮抗内生细菌,其中菌株TF28抑菌谱广,抑菌率高,对不同植物的病原菌F. oxysporum的抑菌率为80.2%~96.7%.经形态、生理生化和16S rRNA鉴定为解淀粉芽孢杆菌(Bacillus amyloliquefaciens).  相似文献

Survival of Aeromonas salmonicida in natural (non-sterile) seawater, as determined from colony counts on marine agar, was found to be influenced by the presence of potentially inhibitory organisms, i.e., Acinetobacter, Aeromonas hydrophila, Chromobacterium, Escherichia coli, Flavobacterium and Pseudomonas, and their metabolites. Yet, samples, thought to be devoid of culturable A. salmonicida, were found to contain cells, which were filterable through 0.22 and 0.45 microns Millipore Millex porosity filters, and were recoverable on a specialised medium for L-forms, i.e. L-F medium.  相似文献

红树林内生细菌及拮抗菌分离筛选结果表明:各品种红树体内均有大量的内生细菌,不同红树品种及部位内生细菌的数量不同,其中以红海榄体内的含量最高,达4.225×104cfu/g(fw),其它依次为木榄、桐花树、秋茄和白骨壤等;不同部位以茎组织体内内生细菌的含量最多,达1.649×104cfu/g(fw),其次为根和叶。获得的内生细菌中约有43.53%的内生细菌菌株对枯萎病菌(Fusarium oxysporum)、炭疽病菌(Colletotrichum sp.)等病原真菌及番茄青枯病菌(Ralstonia solanaceance)具有较强拮抗作用。对番茄生长测定发现,13株菌株中有9株(69.23%)表现为促进生长效果,4株(30.77%)表现为抑制生长作用。经初步鉴定,上述拮抗细菌均为芽孢杆菌(Bacillus sp.)。  相似文献

Summary Growing agricultural crops with direct seawater irrigation has progressed within the past few years from the conceptual to the experimental phase. This has been accomplished by selecting halophytes with inherently high salinity tolerance for use as crop plants rather than by increasing the ability of traditional crop plants to tolerate seawater. Some of the halophytes being investigated for use as crops in seawater irrigation scenarios have high nutritional value as forage or fodder crops. Most of them also have high digestibility. The limiting factor in such use is their high salt content, but this limitation can be moderated. However, since seeds of halophytes do not accumulate salt any more than do those of glycophytes, the greatest promise for seawater-irrigated halophytes probably will be as seed crops. The seeds of many halophytes have high protein and oil contents and compare favorably with traditional oilseed crops. Sustained high yields of seed and biomass already have been obtained from some halophytes irrigated with seawater, and within the next few years seawater agriculture should proceed from the experimental to the operational phase.  相似文献
