藓类提取物对植物病原真菌的抑菌性研究   总被引:3,自引:0,他引:3  
以立枯丝核菌(Rhizoctonia)、尖孢镰孢菌(Fusarium oxysporum)、苹果轮纹病菌(Macrophoma kawatsukai)、梨黑星病菌(Fusicladium pirina)、草莓灰霉菌(Botrytis cinerea)等七种植物病原真菌为供试菌,通过生长速率法测定了24种藓类的乙醇提取物对病原真菌的抑制作用.结果表明,在提取物浓度为1.2mg干样/mL,大镰刀藓提取物对立枯丝核菌的抑制作用最强,抑菌率为67.8%.藓类提取物不仅对植物病原真菌有抑菌活性,而且还有多种藓类提取物对病原真菌菌丝有促进生长活性,如锐尖匍灯藓对立枯丝核菌菌丝生长的促进率为95.1%.选出大镰刀藓、扁灰藓、紫萼藓、山地水灰藓、疣小金发藓、橙色净口藓、锐尖匍灯藓和羊角藓对立枯丝核菌和灰霉菌的抑菌性或菌丝生长促进性进行了进一步研究,藓类提取物对病原菌的抑菌性或菌丝生长促进性在一定范围内随浓度的增加而增强,并且抑菌性随时间的延长呈下降趋势.  相似文献

植物病原真菌侵入寄主组织并在寄主组织中生长是其致病的基本条件(Deising et al., 2000; Hartmann et al.,1996; Tucker & Talbot, 2001; Buller,1931).但是,关于病原真菌是靠机械力还是酶解作用,亦或是二者结合穿透寄主的争论,已经持续了多年(Latunde-Dada,2001;Mendgen et al.,1996).许多植物病原菌能形成称为附着胞的特异侵染结构,可以产生很大的机械压力,推动侵染菌丝侵入寄主组织.顶端生长是菌丝生长的特点之一,通过顶端生长,可以侵入固体基质获取营养.真菌细胞内的渗透压是维持顶端生长、推动病原真菌侵入寄主的动力.早在1931年, Buller(1931)在一个简单的实验中已经观察到真菌菌丝细胞内巨大膨压的产生并转化为压力的现象:一种鬼伞Coprinus sterquilinus的菌柄直径仅有6mm,却可以撑起200g的重物,压强至少可达0.07 MPa (0.7 bar).许多蘑菇种类可以顶起障碍物及顶开沥青路面或石块等,都显示了菌丝的膨压可以使菌丝细胞产生很大的机械强度.最近关于稻瘟病菌Magnaporthe grisea的研究结果表明,病菌可以产生足够的机械力直接穿透水稻的表皮细胞(De Jong et al.,1997).本文对真菌穿透寄主的机械力的研究进展进行简述.  相似文献

以河南郑州和江苏徐州采集到的杜仲为材料,采用纯培养方法对杜仲内生真菌进行分离。将分离纯化得到的90株真菌通过形态学鉴定并进行多样性分析,90株内生真菌分别属于13个属,其中茎点霉属(Phoma)和链格孢属(Alternaria)为优势菌群,分别占总菌株数的18%和16%;其次为黑孢属(Nigrospora)和枝孢属(Cladosporium),均占总菌株数的10%。用平板对峙法对分离得到的杜仲内生真菌进行抗植物病原真菌实验,发现有18个菌株对至少一种病原真菌具有明显的拮抗作用。通过比色法进行内生真菌产IAA(吲哚乙酸)定性实验,结果显示,有37株真菌具有产IAA能力,通过分光光度法对这37株真菌进行产IAA定量实验,发现有8株有较好的产IAA活性。  相似文献

在马铃薯葡萄糖琼脂 ( PDA)上测定了酸溶性壳聚糖 ( S- )和 2种水溶性壳聚糖 ( S- 和 S- )对 15种植物病原真菌的作用 ,结果表明 ,三种壳聚糖对供试的 15种植物病原真菌均有一定程度的抑制作用 ,抑制强度因壳聚糖的理化性质以及不同的病原真菌而有较大差异。水溶性壳聚糖 S- 可明显抑制水稻恶苗病菌分生孢子的萌发。壳聚糖的抑菌作用机制与其增加菌丝细胞膜的透性有关。  相似文献

Abstract The antagonistic effect of Trichoderma reesei and Trichoderma harzianum was studied towards a range of phytopathogenic fungi; Alternia solani, Botrytis fabae, Cladosporium cucumerinum, Fusarium oxysporum, Fusarium tricintum . Coculture of the phytopathogens and Trichodema under laboratory conditions clearly showed dominance of the Trichoderma species. In all cases, Trichoderma overgrew the phytopathogens and subsequently developed a conidial lawn over the surface. In these studies the lytic action of the pathogen was clearly apparent and the inhibition of growth appears directly related to its ability to hydrolyze the cell walls of the tested microorganisms rather than through the inhibitory action of antibiotics or toxins. Proteinase, mannanase, laminarinase and chitinase activities were determined in the extracellular fluid of Trichoderma . Glucose and laminaribiose were detected after the in vitro hydrolysis of the cell walls of the phytopathogens. The data imply that the nature of T. reesei and T. harzianum antagonism is based on mycoparasitism (lysis) and appears to optimalize with contact between the mycelia.  相似文献

Verticillium dahliae is a soilborne fungus that causes a vascular wilt disease of plants and losses in a broad range of economically important crops worldwide. In this study, we compared the proteomes of highly (Vd1396‐9) and weakly (Vs06‐14) aggressive isolates of V. dahliae to identify protein factors that may contribute to pathogenicity. Twenty‐five protein spots were consistently observed as differential in the proteome profiles of the two isolates. The protein sequences in the spots were identified by LC‐ESI‐MS/MS and MASCOT database searches. Some of the identified sequences shared homology with fungal proteins that have roles in stress response, colonization, melanin biosynthesis, microsclerotia formation, antibiotic resistance, and fungal penetration. These are important functions for infection of the host and survival of the pathogen in soil. One protein found only in the highly aggressive isolate was identified as isochorismatase hydrolase, a potential plant‐defense suppressor. This enzyme may inhibit the production of salicylic acid, which is important for plant defense response signaling. Other sequences corresponding to potential pathogenicity factors were identified in the highly aggressive isolate. This work indicates that, in combination with functional genomics, proteomics‐based analyses can provide additional insights into pathogenesis and potential management strategies for this disease.  相似文献

Methanol extract, obtained from Tagetes patula plant, was assayed against three phytopathogenic fungi: Botrytis cinerea, Fusarium moniliforme and Pythium ultimum. The antifungal activity was tested both in the dark and in the light, using two different lighting systems. The data showed that the extract proved to have a dose-dependent activity on all the fungi with a marked difference between treatments in the light than in the dark. Good growth inhibition was observed in fungi only when these were treated with the highest dose of the extract and irradiated, whereas the same dose gave only a modest inhibition when the experiment was conducted in the dark. At 5 and 10 microg/ml in the dark, growth increased. The results indicated that the presence of a luminous source enhances the antifungal activity, with small differences between UV-A and solar spectrum light. SEM and TEM observations on Pythium ultimum revealed that the Tagetes patula extract induced alterations on cell fungal membranes with a photoactivation mechanism possibly involving the production of free radicals and leading to a premature aging of the mycelium.  相似文献

Summary Aeriobiological studies are essential for understanding the distribution, ecology and deposition patterns of both phytopathogenic and nonpathogenic fungal spores which are carried away from their source. Many spores and conidia are devitalized during aerial transportation as a consequence of being exposed to atmospheric agents. Nonetheless, a sufficient number remain viable, causing infections of various kinds, some of which extremely serious and with an epidemic trend.In order to predict the onset of fungi-induced diseases, it is necessary to be able to determine the inoculum source of the pathogenic agent. As air is the main vector transporting pathogenic fungal spores and conidia, periodical monitoring is required. Thus, having established the critical stages of plant infection, necessary precautionary measures can be undertaken in order to control diseases onset and development.It is therefore necessary to gain a through understanding of spore takeoff and dispersal mechanisms so as to determine how the spores and conidia are transported by air currents onto the plants and how they cause infective impaction. Spores and conidia suspended in the atmosphere can be collected by means of appropriate traps filtering a predetermined amount of air at predetermined time intervals in order to be able to make predictions as to the possibility of plant infection. Volumetric air sampling allows not only to determine the concentration of spores and conidia in a given period of time but also to establish the hours of the day in which they are present in highest concentrations and in which therefore they are more liable to cause infection. This information may be used in estimating the incidence of disease symptoms, the duration of infection and the seriousness of the disease.On the basis of this data, mathematical models for predicting epidemics can be worked out.  相似文献

Indigenous actinomycetes isolated from rhizosphere soils were assessed for in vitro antagonism against Colletotrichum gloeosporioides and Sclerotium rolfsii. A potent antagonist against both plant pathogenic fungi, designated SRA14, was selected and identified as Streptomyces hygroscopicus. The strain SRA14 highly produced extracellular chitinase and β-1,3-glucanase during the exponential and late exponential phases, respectively. Culture filtrates collected from the exponential and stationary phases inhibited the growth of both the fungi tested, indicating that growth suppression was due to extracellular antifungal metabolites present in culture filtrates. The percentage of growth inhibition by the stationary culture filtrate was significantly higher than that of exponential culture filtrate. Morphological changes such as hyphal swelling and abnormal shapes were observed in fungi grown on potato dextrose agar that contained the culture filtrates. However, the antifungal activity of exponential culture filtrates against both the experimental fungi was significantly reduced after boiling or treatment with proteinase K. There was no significant decrease in the percentage of fungal growth inhibition by the stationary culture filtrate that was treated as above. These data indicated that the antifungal potential of the exponential culture filtrate was mainly due to the presence of extracellular chitinase enzyme, whereas the antifungal activity of the stationary culture filtrate involved the action of unknown thermostable antifungal compound(s).  相似文献

The frequency and distribution of microsatellites were analyzed in the 19 mitogenomes of phytopathogenic fungi covering five phyla. Our analysis revealed that in all the mitogenomes studied, the frequency and relative abundance varied, and it was neither influenced by genome size nor by GC content. SSRs were found to be differential distributed in genic and intergenic regions. An average of 5.14 (23.6%) SSRs were present in genic sequences and 21.7 (76.4%) SSRs were located in the intergenic sequences. Relative abundance of SSRs in mitogenomes was the highest in Aspergillus tubigensis, whereas, it was the least in Phaeosphaeria nodurum, the average being 0.45. Trinucleotide repeats were the most abundant motifs in the genic and intergenic regions of the mitogenomes of the phytopathogenic fungi. Among the genes, cox1 harbors the maximum SSRs, whereas cox3 and nad 7 contain the least. Based on the presence of SSRs in a particular gene, genetic relationships among individual organisms were also established.  相似文献

丝状真菌不仅是致病菌,而且在异源表达工业酶、化学制品以及药物活性物质中发挥着越来越重要的作用。随着人类基因组计划的实施和推进,生命科学研究已进入了功能基因组时代,特别是蛋白质组学,在蛋白质水平对丝状真菌细胞生命过程中蛋白质功能和蛋白质之间的相互作用以及特殊条件下的变化机制进行研究,对生命的复杂活动进行深入而又全面的认识也为丝状真菌工业酶制剂和重组药物的开发提供广阔的创新空间。本文综述了蛋白质组学的研究内容和方法,总结了其在丝状真菌致病菌、抗生素产生菌和纤维素酶产生菌中的应用现状。不同层次的功能基因组学分析可以从各个角度掌握生物体的代谢网络和调控机制,本文还对蛋白质组学以及功能基因组学各部分内容的整合运用进行了展望。  相似文献

通过对丝状真菌G蛋白偶联受体(GPCR)的结构、分类以及功能方面进行综述,以期明确丝状真菌与其他模式生物GPCR之间的关系。基于已报道的模式生物及丝状真菌等不同生物中的GPCR,通过SMART保守结构域分析,以及利用Clustal X、MEGA等软件对上述GPCR进行遗传关系分析。明确丝状真菌典型GPCR具有七跨膜结构域,新型GPCR则含有PIPK、RGS等保守结构域,明确不同学者对于GPCR的分类情况,以及新型GPCR所具有的特殊功能,明确模式生物GPCR、丝状真菌GPCR分别各自聚类。丝状真菌中GPCR的数量较模式生物少,不同分类单元中真菌之间GPCR的数量也不尽相同,同时,丝状真菌GPCR除具有典型的七跨膜结构域外,还含有一些其他保守的结构域,上述研究为进一步开展其功能研究提供重要的理论基础。  相似文献
