Grazing of protozoa and its effect on populations of aquatic bacteria 总被引:23,自引:0,他引:23
Predation by bacterivorous protists in aquatic habitats can influence the morphological structure, taxonomic composition and physiological status of bacterial communities. The protistan grazing can result in bacterial responses at the community and the species level. At the community level, grazing-induced morphological shifts have been observed, which were directed towards either larger or smaller bacterial sizes or in both directions. Morphological changes have been accompanied by changes in taxonomic community structure and bacterial activity. Responses at the species level vary from species to species. Some taxa have shown a pronounced morphological plasticity and demonstrated complete or partial shifts in size distribution to larger growth forms (filaments, microcolonies). However, other taxa with weak plasticity have shown no ability to reduce grazing mortality through changes in size. The impact of protistan grazing on bacterial communities is based on the complex interplay of several parameters. These include grazing selectivity (by size and other features), differences in sensitivity of bacterial species to grazing, differences in responses of single bacterial populations to grazing (size and physiology), as well as the direct and indirect influence of grazing on bacterial growth conditions (substrate supply) and bacterial competition (elimination of competitors). 相似文献

Islet amyloid contributes to the loss of beta-cell mass in type 2 diabetes. To examine the roles of glucose and time on amyloid formation, we developed a rapid in vitro model using isolated islets from human islet amyloid polypeptide (hIAPP) transgenic mice. Islets from hIAPP transgenic and non-transgenic mice were cultured for up to 7 days with either 5.5, 11.1, 16.7 or 33.3mmol/l glucose. At various time-points throughout the culture period, islets were harvested for determination of amyloid and beta-cell areas, and for measures of cell viability, insulin content, and secretion. Following culture of hIAPP transgenic islets in 16.7 or 33.3mmol/l glucose, amyloid formation was significantly increased compared to 5.5 or 11.1mmol/l glucose culture. Amyloid was detected as early as day 2 and increased in a time-dependent manner so that by day 7, a decrease in the proportion of beta-cell area in hIAPP transgenic islets was evident. When compared to non-transgenic islets after 7-day culture in 16.7mmol/l glucose, hIAPP transgenic islets were 24% less viable, had decreased beta-cell area and insulin content, but displayed no change in insulin secretion. Thus, we have developed a rapid in vitro model of light microscopy-visible islet amyloid formation that is both glucose- and time-dependent. Formation of amyloid in this model is associated with reduced cell viability and beta-cell loss but adequate functional adaptation. It thus enables studies investigating the mechanism(s) underlying the amyloid-associated loss of beta-cell mass in type 2 diabetes. 相似文献

【目的】发掘有效生防菌株以控制枣缩果病,并探究生防菌株的拮抗特性。【方法】采用梯度稀释法和平板对峙法筛选出2株对枣缩果病菌细极链格孢(Alternaria tenuissima)具有显著拮抗作用的菌株STO-12和STO-45,通过PCR扩增得到其16S rRNA基因序列,进行同源比对分析和分子系统树的构建,并结合形态学观察和生理生化实验对菌株进行鉴定。采用皿内对峙、显微观察及蛋白质抑菌试验等测定了菌株STO-45的拮抗效应。【结果】两拮抗菌株STO-12和STO-45均鉴定为枯草芽孢杆菌,但菌株STO-12能产生橘红色色素,且两菌株在大小、生理生化特征及系统发育关系上具有生理差异。这两株拮抗细菌的发酵液、发酵上清液及发酵滤液均对枣缩果病菌具有显著抑菌活性。STO-45发酵滤液在0.8%的低体积浓度下即达到抑制中浓度,可使病原菌A. tenuissima MY5的菌丝及芽管部分畸形膨大,发酵液中的蛋白类物质可能是其拮抗物质之一。【结论】拮抗菌株STO-12和STO-45对枣缩果病的生物防治均具有潜在的应用价值,其抑菌机制及生物制剂的开发利用值得进一步研究。 相似文献

目的 研究栀子豉汤及栀子、淡豆豉对6种人肠道菌的影响。方法 从特定人的粪便中分离培养肠杆菌、肠球菌、双歧杆菌、乳杆菌、产气荚膜梭菌和脆弱拟杆菌。分别将高、中、低浓度的栀子、淡豆豉及栀子豉汤药液加入选择性培养基中,以不加药液的培养基作为空白对照,将实验菌分别于厌氧或有氧条件下培养,计数菌落。结果 不同浓度的栀子对肠杆菌的抑制率均为100.00%,淡豆豉在20%、50%、70%浓度分别使肠杆菌数量下调100.00%、53.83%和22.11%,而栀子豉汤可使肠杆菌数量维持在一定水平(104~106 CFU/g);两味药单独都表现了对肠球菌较强的抑制作用,而低、中浓度栀子豉汤则使肠球菌保持较稳定的数量;单味药及复方对产气荚膜梭菌和双歧杆菌敏感性较弱;栀子对脆弱拟杆菌抑制作用最强,而淡豆豉和栀子豉汤则可使其数量上调约20.00%;淡豆豉、栀子豉汤和高浓度栀子对乳杆菌非常敏感,低、中浓度栀子对乳杆菌数量仅有轻微下调作用。结论 栀子豉汤比单独的栀子和淡豆豉更能维持肠道菌群的相对平衡。 相似文献

The phenomenon of cyanide-resistant respiration was studied in bacteria as well as its relation to the oversynthesis of metabolites. Cyanide-resistant respiration was shown to be common of aerobic and facultative anaerobic bacteria. The resistance of respiration to cyanide was observed when the growth of a microbial culture was inhibited or ceased entirely, provided the culture lacked the oversynthesis of metabolites. When a culture was capable of metabolites oversynthesis under such conditions, cyanide-resistant respiration (if any) declined. Therefore, there is a competition between metabolites oversynthesis and cyanide-resistant respiration, which is typical of many microorganisms. 相似文献

The mechanism of ethanol conversion in a nonequilibrium glow discharge has been studied. It is shown that molecular hydrogen is produced in reactions between ethanol molecules and hydrogen atoms in the initial stage and in reactions involving active H, CH2OH, CH3CHOH, and formaldehyde in the final stage. Comparison with experimental data shows that the kinetic mechanism used in these calculations correctly predicts the concentrations of the main components of the gas mixture. 相似文献

The effect of T-2 toxin on the growth rates of different bacteria was used as a measure of its toxicity. Toxin levels of 10 micrograms/ml did not decrease the growth rate of Selenomonas ruminantium and Anaerovibrio lipolytica, whereas the growth rate of Butyrivibrio fibrisolvens was uninhibited at toxin levels as high as 1 mg/ml. There was, however, a noticeable increase in the growth rate of B. fibrisolvens CE46 and CE51 and S. ruminantium in the presence of low concentrations (10 micrograms/ml) of T-2 toxin, which may indicate the assimilation of the toxin as an energy source by these bacteria. Three tributyrin-hydrolyzing bacterial isolates did not grow at all in the presence of T-2 toxin (10 micrograms/ml). The growth rate of a fourth tributyrin-hydrolyzing bacterial isolate was unaffected. B. fibrisolvens CE51 degraded T-2 toxin to HT-2 toxin (22%), T-2 triol (3%), and neosolaniol (10%), whereas A. lipolytica and S. ruminantium degraded the toxin to HT-2 toxin (22 and 18%, respectively) and T-2 triol (7 and 10%, respectively) only. These results have been explained in terms of the presence of two different toxin-hydrolyzing enzyme systems. Studies with B. fibrisolvens showed the presence of a T-2 toxin-degrading enzyme fraction in a bacterial membrane preparation. This fraction had an approximate molecular weight of 65,000 and showed esterase activity (395.6 mumol of p-nitrophenol formed per min per mg of protein with p-nitrophenylacetate as the substrate. 相似文献

The purpose of this study was to examine the relationship between plasma norepinephrine concentrations (PNE) and efferent muscle sympathetic nerve activity to noncontracting muscle (MSNA) during graded, rhythmic exercise in humans. In the initial study, six healthy men (ages 20-30 yr) performed 2-min bouts of two-arm cycling exercise at power outputs of 0, 10, 20, 40, 60 (n = 6), and 80 (n = 3) W. Heart rate (HR) was recorded and intraneural measurements of MSNA (right peroneal nerve) were made continuously for 2 min before (control) and during exercise at each work load. At least 2 wk later, subjects performed the same exercise bouts at which time HR was measured and a venous (forearm) blood sample was obtained for the subsequent determination of PNE by high-performance liquid chromatography. During exercise, HR increased progressively from 0 to 80 W. Neither MSNA nor PNE increased above control in response to arm cycling at 0, 10, and 20 W [0-16 +/- 1% (SE) of peak work load], but both variables increased progressively at the 40-, 60-, and 80-W (33 +/- 1 to 67 +/- 2% of peak work load) levels (all P less than 0.05). The effect of amphotericin B on the permeability properties of liposomes prepared by reverse-phase evaporation was examined by using an osmotic method. This study has revealed that the magnitude and type of the alterations in permeability induced by amphotericin B in liposomes made of egg phosphatidylcholine and ergosterol depend not only on the amphotericin B concentration in the external aqueous solution but also on the time elapsed after mixing. Thus, low amphotericin B concentrations (from 0.2 to 1.2 microM) led to, (1) an small increment of the total extent of shrinkage of liposomes suspended in non-electrolytes such as urea or salts like KNO3, (2) an enhancement of urea and salt permeabilities at the same time scale at which volume changes were measured (ms to s), (3) a maximal blocking by tetraethylammonium of amphotericin B-induced urea permeability and (4) an enhancement of glucose permeability but only after liposomes were incubated with amphotericin B for some minutes before mixing. The high amphotericin B concentration regime (beyond 1.2 microM) led to, (1) a decrease of the total extent of shrinkage of liposomes immediately after rapid mixing of liposomes with urea solutions containing amphotericin B and (2) a 50% reduction of the tetraethylammonium blocking of amphotericin B-induced urea permeability. The disposal of reject brine, a highly concentrated waste by-product generated by various industrial processes, represents a major economic and environmental challenge. The common practice in dealing with the large amounts of brine generated is to dispose of it in a pond and allow it to evaporate. The rate of evaporation is therefore a key factor in the effectiveness of the management of these ponds. The addition of various dyes has previously been used as a method to increase the evaporation rate. In this study, a biological approach, using pigmented halophilic bacteria (as opposed to chemical dyes), was assessed. Two bacteria, an Arthrobacter sp. and a Planococcus sp. were selected due to their ability to increase the evaporation of synthetic brine. When using industrial brine, supplementation of the brine with an iron source was required to maintain the pigment production. Under these conditions, the Planococcus sp. CP5-4 produced a carotenoid-like pigment, which resulted in a 20% increase in the evaporation rate of the brine. Thus, the pigment production capability of halophilic bacteria could potentially be exploited as an effective step in the management of industrial reject brines, analogous to the crystallizer ponds used to mine salt from sea water. 相似文献

【背景】梨火疫病是由梨火疫病菌(Erwinia amylovora)引起的细菌性病害,对苹果、梨、山楂等40余属220多种植物危害极大。近年来,与中国毗邻的中亚多国相继暴发梨火疫病。梨火疫病菌成为国内特别是对新疆地区的特高风险有害生物,梨火疫病已被中国农业农村部列入《一类农作物病虫害名录》。【目的】从新疆本土材料以及实验室现有菌株中进行梨火疫病菌拮抗菌株的筛选,为梨火疫病的防控奠定基础。【方法】采用平板对峙法从新疆香梨种植区土壤及实验室现有菌株中分离、筛选具有显著拮抗效果的菌株,通过生理生化特性分析和16SrRNA基因测序等方法对菌株进行初步鉴定,并采用牛津杯扩散法对抑菌效果进行复筛。利用两年生盆栽杜梨苗对拮抗菌进行温室内保护型和治疗型防效测定。【结果】筛选获得11株具有显著拮抗作用的菌株,均为革兰氏阳性菌。其中9株为芽胞杆菌属(Bacillus)、2株为乳杆菌属(Lactobacillus)。平板对峙实验结果表明贝莱斯芽胞杆菌(Bacillus velezensis) JE7、植物乳杆菌(Lactobacillus plantarum) LP1和植物乳杆菌LP2拮抗作用较强,其次为贝莱斯芽胞杆菌JE4。生理特性实验结果表明:JE4、JE7可在NaCl浓度为1%-7%时生长,JE4、JE7在pH 4.0-9.0时生长,LP1可在NaCl浓度为1%-9%时生长,在pH 5.0-9.0时生长良好。温室防效测定结果显示,贝莱斯芽胞杆菌JE4防效最佳,可达到73%以上。【结论】筛选获得一批梨火疫病拮抗菌,其中部分菌株在温室内防效显著并具有较强的盐碱耐受能力。 相似文献

Few efforts have been directed at understanding how the rhizosphere microbiology of continuous corn may effect crop yields.
This relationship may explain, in part, the decreases in yield associated with continuous corn as compared to the corn in
rotation with a second crop. This study was conducted to determine the importance of soil-borne microorganisms to yield declines
in long term continuous corn. Continuous corn (Zea mays L.) or rotated corn-soybean (Glycine max L.) field plots, established in 1975, under either fall plowing or no-till tillage treatments were used. Treatments consisted
of methyl bromide applied at 48.8 g m−2 3 days prior to planting in all four combinations. Total plant samples from both the fumigated and non-fumigated areas were
collected 14 days after planting. Rhizosphere bacteria were recovered and tested for their ability to impact plant growth.
Bacterial assessments were made in a test tube bioassay where germinated corn was transported in to agar containing a bacterial
isolate. In the first year of the study a highly significant interaction of fumigation and rotation was indicated. With fumigation
continuous corn yields were similar to that of rotated corn-bean. Rotated corn yields were less affected by fumigation. In
the second year, the effects were similar but less significant. Over 130 bacterial isolates were tested for their effect on
plant growth. Approximately 22% were able to inhibit plant growth. Of these, 72% were from the continuous corn system. Clearly,
the interaction of rotation and yield is at a microbiological level. The suggestion that microorganisms similar to those isolated
are responsible for controlling early plant growth in the continuous corn system is indicated. 相似文献

目的观察黄瓜香等对糖尿病大鼠脂质代谢的影响,并进一步探讨其调脂机制。方法选取糖尿病大鼠灌胃给予黄瓜香提取液120mg/(kg.d),共8周,检测其空腹血脂变化,及其肠道菌群变化。结果(1)经黄瓜香提取物治疗后,治疗组较糖尿病组,甘油三脂、低密度脂蛋白和胆固醇明显降低(P〈0.01);(2)糖尿病组肠道菌群较正常组有明显变化(P〈0.01),尤其双歧杆菌、乳杆菌等明显减少,而治疗后肠道菌群得到调整。结论黄瓜香提取物可能通过扶植肠道菌群生长繁殖,对脂质代谢发挥作用,从而达到其调脂目的。 相似文献

This article describes and particularly explains a new phenomenon of persistent microbicidal effect of water previously exposed to the low-temperature plasma, which cannot be attributed to the acidification only. The direct microbicidal action of plasma is well documented, being mediated by number of reactive particles with a short lifetime. However, we observed the microbicidal effect also in exposed water stored for a month, where it must be mediated by stable particles. In water and in phosphate-buffered saline, the formation of NOx and corresponding acids, H2O2 and O3 was confirmed after exposition to the low-temperature plasma generated in air by DC negative glow corona and positive streamer discharge. The time course of acidification, H2O2 and O3 formation were deremined. Except uncertain traces of HCN, SIFT-MS analysis of exposed liquids reveals no additional reactive compounds. The microbicidal effect persists almost unchanged during 4 weeks of storage, although O3 completely and H2O2 almost disappears. Staphylococcus epidermidis and Escherichia coli were inactivated within 10 min of incubation in exposed liquids, Candida albicans needs at least 1 h. The solutions prepared by artificial mixing of reactive compounds mimic the action of exposed water, but in lesser extent. The acid milieu is the main cause of the microbicidal effect, but the possibility of still unidentified additional compound remains open. 相似文献
