In the present study, the aphrodisiac activity of Butea frondonsa Koen. ex Roxb (Papillionaceae) bark extract was investigated. The extract (400 mg/kg body wt./day) was administered orally by gavage for 28 days. Mount latency (ML), intromission latency (IL), ejaculation latency (EL), mounting frequency (MF), intromission frequency (IF), ejaculation frequency (EF) and post-ejaculatory interval (PEI) were the parameters observed before and during the sexual behavior study at day 0, 7, 10, 14, 21, and 28. The extract reduced significantly ML, IL, EL and PEI (p < 0.05). The extract also increased significantly MF, IF and EF (p < 0.05). These effects were observed in sexually active and inactive male rats.  相似文献

In this study, the aphrodisiac activities of Crocus sativus stigma aqueous extract and its constituents, safranal and crocin, were evaluated in male rats. The aqueous extract (80, 160 and 320 mg/kg body wt.), crocin (100, 200 and 400 mg/kg body wt.), safranal (0.1, 0.2 and 0.4 ml/kg), sildenafil (60 mg/kg body wt., as a positive control) and saline were administered intraperitoneally to male rats. Mounting frequency (MF), intromission frequency (IF), erection frequency (EF), mount latency (ML), intromission latency (IL) and ejaculation latency (EL) were the factors evaluated during the sexual behavior study. Crocin, at all doses, and the extract, especially at doses 160 and 320 mg/kg body wt., increased MF, IF and EF behaviors and reduced EL, IL and ML parameters. Safranal did not show aphrodisiac effects. The present study reveals an aphrodisiac activity of saffron aqueous extract and its constituent crocin.  相似文献

将50只同期怀孕的大鼠分为5组,在怀孕第7-18d,每天分别给两组大鼠腹腔注射1.00和20.00mg/kg体重2,2’,4,4’-四氯联苯(PCB47);分别给另两组注射0.25和1.00mg/kg体重3,3’,4,4’-四氯联苯(PCB77);对照组注射0.10mL芝麻油。幼鼠出生后第70-91d、97-101d测试F1代雌鼠的接近时间(AL)、爬跨后返回时间(MRL)、插入后返回时间(IRL)、射精后不应期(PER)、脊柱前凸系数(LQ)和雄鼠的爬跨频率(MF)、爬跨等待时间(ML)、插入频率(IF)、插入等待时间(IL)、射精等待时间(EL)、射精后不应期(PEI)、插入率等性行为。结果显示,妊娠期腹腔注射PCB47和PCB77显著降低F1代雌鼠的LQ(对照组为84.4%;PCB47组分别为76.0%、67.8%;PCB77组分别为64.4%、53.3%;P〉0.05),但对雌鼠的AL、MRL、IRL、PER和雄鼠的MF、ML、IF、IL、EL、PEI和插入率无显著影响(P〉0.05)。  相似文献

The stem barks and the leaves from Bridelia ferruginea (BF, Euphorbiaceae), a medicinal plant, are used in traditional medicine for the treatment of several ailments including male impotency. The present study was aimed at investigating the effects of the dried stem bark of BF on the sexual behaviour of normal and castrated sexually inexperienced male Wistar rats. Animals were orally administered with 100 mg/kg of either the aqueous or the ethanol extracts of BF whilst the neutral control group received in the same way 10 ml/kg of distilled water. The positive control group was treated with a subcutaneous injection of testosterone propionate (20 mg/kg/day/3days) prior to the experiment. The sexual behaviour of all rats was monitored on days 0, 1 and 7 by measuring frequencies of penile erection, mount, intromission and ejaculation. In a separate group of normal sexually inexperienced rats, the pro-sexual effects induced by a single dose of the aqueous extract (100 mg/kg) were measured after pre-treatment with either haloperidol (10 mg/kg), atropine (10 mg/kg) or L-omega-nitro-arginine methyl ester (Lω-NAME, 10 mg/kg). Results obtained showed a significant influence of the duration of treatment on the frequencies of penile erection, mount and intromission of both normal and castrated rats. An increase in all sexual performance parameters was observed when compared to respective controls. The intromission frequency of normal animals was significantly increased (P < 0.05–0.01) on day 1 of treatment and the effect was more expressed in rats receiving the aqueous extract. In castrated animals, a statistical increase was noticed on day 7 for rats treated with testosterone. The sexual effects induced by the aqueous extract of BF were completely abolished after pre-treatment of rats with atropine or haloperidol while pre-administration of Lω-NAME did not produce any significant effects. Flavonoids and sterols revealed in the aqueous and ethanolic extracts from the barks of BF may account for the enhancement of sexual activity in naive rats which could be expressed through dopaminergic and/or cholinergic receptor(s). Results of this work also give value to the traditional use of the plant for the improvement of male sexual behaviour.  相似文献

We studied the effects of Eurycoma longifolia Jack, commonly known as Tongkat Ali in Malaysia, on the initiation of sexual performance and the weights of sexual accessories in inexperienced castrated male rats. The doses of 200, 400 and 800 mg/kg body weight, which were extracted from E. longifolia Jack, were orally administered to the rats twice daily for 10 days prior to the tests and continued throughout the test period. Testosterone was used as a positive control after injecting 15 mg/kg daily subcutaneously for 32 days. Results showed that E. longifolia Jack produced a dose-dependent increase in sexual performance of the treated animals, but the E. longifolia Jack groups showed lower sexual performance in mounting, intromission and ejaculation than the testosterone group. Further results also showed that E. longifolia Jack promoted the growth of both ventral prostate and seminal vesicles as compared with the control, but the growth of sexual accessories at 800 mg/kg of butanol, methanol, water and chloroform fractions of E. longifolia Jack was less than that of testosterone treated group. The present study therefore gives further evidence of the folkuse of E. longifolia as an aphrodisiac.  相似文献

This study examined the effects of rhizomes of Alpinia calcarata Roscoe (Zingiberaceae) on male sexual competence and fertility, using a hot water extract (HWE) and rats. Different doses of HWE (150, 250 and 500 mg/kg) were orally administrated to male rats and their sexual behaviour was monitored (for 15 min) 3 h later using receptive females. Fertility was determined in a separate group (with the highest dose) using a noncompetitive copulation test. In the sexual behaviour study, the HWE impaired the number of rats ejaculating and markedly prolonged the latency for ejaculation. Further, the number of rats mounting and intromitting, and the latencies for mounting and intromission were inhibited. Collectively, these observations indicate a strong aphrodisiac action. The other parameters remained unchanged indicating non-impairment in libido, sexual arousability, sexual vigour and sexual performance or penile erectile ability. However, a slight impairment was evident in sexual motivation (with the highest dose) in a partner preference test. In the fertility test, HWE induced profound oligozoospermia but fertility was uninhibited. The highest dose of HWE also elevated the serum testosterone level and the number of spontaneous penile erections rapidly and markedly. Further, the HWE was nontoxic. It is concluded that A. calcarata rhizomes possess a strong and safe oral aphrodisiac activity.  相似文献

The role of the cingulate cortex in regulating male sexual behavior was studied in testosterone propionate-treated castrated male rats. Males with lesions in the anterior part of the cingulate cortex showed lower levels of mount, intromission and ejaculation activities than sham-operated control males and males with lesions in the posterior part of the cingulate cortex or the frontal cortex. In male rats in which lateral connections of the anterior cingulate cortex were bilaterally interrupted by sagittal cuts, the sexual activity was much lower than in the control rats, being comparable to that of the anterior cingulate cortex lesion group, but transection of the anterior connections by a transverse cut made in the anterior part of the anterior cingulate had no effect. These results suggest that the anterior cingulate cortex and its lateral connections are critical in regulating male sexual behavior in male rats.  相似文献

In the present study, the aphrodisiac properties of Microdesmis keayana J. Léonard root extract and major isolated alkaloids were evaluated by observing the sexual behavior of male rats.Aqueous extract (150 mg/kg body weight) and pure alkaloids (3 mg/kg body weight) were administered orally by gavage to male rats. Latent times of observation, intromission and ejaculation, mounting behavior, number of intromissions and mating performances were evaluated and compared to those obtained with untreated rats in the presence of receptive and non-receptive females. The results have shown that aqueous extract and alkaloids of M. keayana stimulate sexual parameters in rats’ sexual behavior.A short-term toxicity study undertaken to establish the therapeutic index of aqueous extract, showed that a high dose of the extract (2 g/kg body weight) caused no mortality or changes in rats’ behavior.  相似文献

The effects of hyperprolactinemia on male copulatory behavior in adult male and female rats were studied. Hyperprolactinemia was induced by the transplantable purely prolactin-secreting tumor 7315b. Male rats were castrated and received testosterone-filled capsules of different sizes which induced normal and subnormal testosterone levels. After sexual training the rats of the experimental groups were inoculated with tumor 7315b. Three weeks after tumor-inoculation high prolactin levels (2000-30000 ng/ml) were found. During this hyperprolactinemia ejaculation latency increased significantly, while the mount frequency and intromission frequency remained unchanged. Only 9 out of 22 rats ejaculated 19 days after inoculation. Moreover, it appeared that the inhibitory effect of the tumor was as strong in the presence of normal (2.33 +/- 0.07 ng/ml) as in the presence of low (0.35 +/- 0.01 ng/ml) testosterone levels. The inhibitory effect of tumor 7315b on copulatory behavior was not influenced by adrenalectomy. In gonadectomized female rats bearing testosterone-filled capsules tumor 7315b induced prolactin levels of about 2000 ng/ml and an almost complete cessation of mounts and intromission patterns 4 weeks after tumor-inoculation. It was concluded that tumor 7315b causes a strong inhibitory effect on male copulatory behavior in male and female rats and that this effect is not influenced by the presence of normal or low testosterone levels or removal of the adrenals, suggesting a direct effect of prolactin on brain functions.  相似文献

The mitogenic agent that disrupts male and female sexual behavior has been isolated from corncob bedding. The disrupting activity resides in an isomeric mixture of linoleic acid derivatives with a tetrahydrofuran ring and two hydroxyl groups (THF-diols) that include 9, (12)-oxy-10, 13-dihydroxtstearic acid and 10, (13)-oxy-9, 12-dihydroxystearic acid. We examined the effects of exposure of male rats to THF-diols in drinking water on several parameters of male sexual behavior. THF-diols disrupt sexual behavior in male rats by reducing mounting and intromission frequencies. The mount, intromission and ejaculatory latencies are enhanced while the ejaculatory responses are diminished. These findings suggest that the THF-diols modulate hypothalamo-pituitary axis to regulate steroid hormone-dependent male sexual behavior.  相似文献

Three groups of inexperienced castrated male rats were treated daily for 15 days with oil, estradiol benzoate (1 μg), or dihydrotestosterone (1 mg), and thereafter injected daily with testosterone (1 mg) for 21 days. Sexual behavior was tested every third day after the start of the pretreatment until day 36. Estradiol benzoate or dihydrotestosterone failed to elicit sexual behavior. Pretreatment with dihydrotestosterone, but not estradiol benzoate, significantly shortened the intervals to initiation of mounting and intromission in response to testosterone. The results suggest that fully developed genitals (penis and/or sexual accessories) facilitate initiation of copulatory behavior in response to testosterone administration.  相似文献

The synthetic steroid methyltrienolone (R 1881) binds specifically with high affinity to intracellular androgen receptors and is not metabolized to androstanediol. Administration of R 1881 (1 mg/day) to castrated male rats facilitated intromission in significantly more animals than did 5α-dihydrotestosterone (DHT) (1 mg/day); however, the percentage of animals ejaculating and the pattern of behavior displayed were equivalent in the two groups. Combined administration of estradiol benzoate (EB) (2 μg/day) plus either R 1881 or DHT further facilitated males' sexual performance to levels previously seen in castrated male rats of the same strain when given testosterone propionate (TP). The results suggest that conversion of DHT to 3α- or 3β-androstanediol neither detracts from nor contributes to its ability to activate sexual behavior in the male rat.  相似文献

ATP-sensitive potassium (K+ATP) channels regulate cell excitability and are expressed in steroid-responsive brain regions involved in sexual behavior, such as the preoptic area (POA) and medial basal hypothalamus (MBH). We hypothesized that K+ATP channels serve as a mechanism by which testosterone can control the electrical activity of neurons and consequently elicit male sexual responsiveness. RT-PCR analysis indicated that castration induces, while testosterone inhibits, mRNA expression of the K+ATP channel subunit Kir6.2 in both the POA and MBH of adult male rats. Intracerebral infusion of the pharmacological K+ATP channel inhibitor tolbutamide increased the proportion of long-term castrates displaying sexual behavior and restored mount frequency, intromission frequency, and copulatory efficacy to values observed in testes-intact animals. Infusions of tolbutamide, but not vehicle, also decreased latencies to mount and intromit in castrated males. Unilateral tolbutamide infusion directly into the POA significantly reduced mount latency of castrates; however, it did not affect other copulatory measures, suggesting that blockade of K+ATP channels in additional brain regions may be necessary to recover the full range of sexual behavior. These data indicate that blockade of K+ATP channels is sufficient to elicit the male sexual response in the absence of testosterone. Our observations are consistent with the hypothesis that testosterone modulates male sexual behavior by regulating K+ATP channels in the brain. Decreased channel expression or channel blockade may increase the excitability of androgen-target neurons, rendering them more sensitive to the hormonal, chemical, and somatosensory inputs they receive, and potentially increase secretion of neurotransmitters that facilitate sexual behavior.  相似文献

Centrally administered corticotrophin-releasing factor fragment CRF(4-6) (Pro-Pro-Ile) inhibits components of male rat sexual behaviour. Administration of 1 and 2 microg of tripeptide leads to dose-dependent increase in mount, intromission and ejaculation latencies. Alterations of sexual behaviour parameters suggest suppression of both sexual motivation and sexual performance.  相似文献

The seeds of Griffonia simplicifolia Baill. are rich in 5-HTP (5-hydroxytryptophan), a direct precursor of the neurotransmitter serotonin. In the present study we investigated the influence of the plant extract on male sexual behavior. The seed extract was orally administered to Sprague-Dawley male rats at three dose levels (25, 50 and 100 mg/kg) both acutely and subchronically (daily for 9 days). Mating test with receptive female rats was performed 60 min after the acute treatment or the last dose when repetitively administered. Mount, intromission and ejaculation latencies and post-ejaculatory interval were recorded. Food intake and body weight were measured over the 9-day period of treatment. Microdialysis technique was used to detect the extracellular levels of serotonin (5-HT) and its metabolite 5-hydroxyindoleacetic acid (5-HIAA) in rat brain following the acute administration of the extract dosed at 100 mg/kg. The acute treatment significantly increased mount latency (at any dosage), intromission and ejaculation latencies (at 100 mg/kg) and post-ejaculatory interval (at 50 and 100 mg/kg). On the contrary the subchronic treatment failed to exert a significant influence on copulatory behavior. The daily administration of the extract dosed at 50 and 100 mg/kg for 9 days significantly reduced food intake and body weight. Finally in the microdialysis experiments we found a dramatic increase in 5-HT and its metabolite 5-HIAA.  相似文献

According to Ayurvedic literature of Sri Lanka, roasted seeds of Artocarpus heterophyllus Lam. (Family: Moraceae) has aphrodisiac activity. However, some reproductively active young men in rural areas of Sri Lanka claim that consumption of these seeds few hours prior to coitus disrupts sexual function. Because of these two conflicting claims, it was thought useful to scientifically investigate the effects of A. heterophyllus seeds on male sexual function and fertility. This was done using a seed suspension in 1% methylcellulose (SS) in rats. In a sexual behaviour study using receptive female rats, an oral administration of 500 mg/kg dose of SS markedly inhibited libido, sexual arousal, sexual vigour and sexual performance within 2 hr. Further, the treatment induced a mild erectile dysfunction. These antimasculine effects on sexual function was not evident 6 hr post treatment indicating rapid onset and offset of action. Further, these actions on the sexual behaviour was not due to general toxicity, liver toxicity, stress or reduction in blood testosterone level but due to marked sedative activity. In a mating study, SS failed to alter ejaculating competence and fertility. These results suggest that A. heterophyllous seeds do not have aphrodisiac action, at least, in rats.  相似文献

Androgen deprivation therapy, also known as chemical castration, has been used as an adjunct to psychotherapy for sex offenders. Goserelin and bicalutamide are drugs used for chemical castration. Serotonin (5-hydroxytryptamine, 5-HT) is a key neurotransmitter involved in mood changes, such as depression. We investigated the effects of surgical and chemical castration on depressive symptoms in rats. Surgical castration was performed through a bilateral orchiectomy. Bicalutamide was administrated orally once a day for 84 consecutive days. Goserelin acetate was implanted subcutaneously into the anterior abdominal wall, and this implantation was repeated 3 times at 28-day intervals. Testosterone levels were detected by enzyme-linked immunosorbent assays. Sexual behaviors were analyzed by measuring mount latency, mount frequency, intromission latency, and intromission frequency. The forced swimming test was performed to evaluate rats’ depression status. To detect 5-HT and tryptophan hydroxylase (TPH)-positive cells in the dorsal raphe, immunohistochemistry for 5-HT and TPH and western blotting for 5-HT1A receptors and TPH were performed. Surgical castration and goserelin decreased testosterone levels and suppressed sexual behaviors. However, bicalutamide did not inhibit sexual behaviors, although it reduced testosterone levels to a limited extent. Both surgical and chemical castration induced depression in rats. The expression of 5-HT, TPH, and 5-HT1A receptors in the dorsal raphe was significantly decreased by both surgical castration and chemical castration via bicalutamide and goserelin. The present results showed that surgical and chemical castration for 12 weeks induced a depressive state in rats by inhibiting serotonergic function through 5-HT1A receptors.  相似文献

Two groups of weanling rats were subjected to malnutrition, one with periodic injections of testosterone (males) and the other with estradiol (females). Two other groups (castrated males or castrated females) received normal feedings. In control animals, the relative weights (mg/gm body weight) of testes, seminal vesicles, and ovaries were greater than in malnourished rats. However, relative weights of those organs in hormone-treated, malnourished animals were greater than in those subjected to malnutrition alone and still greater than in controls. Normal sexual cranial dimorphism (SCD) was decreased 16% by male castration, 23% by malnutrition, and 83% by estradiol treatment in malnourished females. On the other hand, normal SCD was increased 20% by female castration and more than 200% by testosterone treatment in malnourished males. All monosexual comparisons corroborated the bisexual range of distances found. Testicular but not ovarian secretions seemed to influence sexual cranial dimorphism. Malnutrition delayed SCD because of a deficiency of testosterone level in stressed males. It is suggested that estradiol in females may counteract sexual cranial development and that its inhibitory effect may be additive to the testosterone deficit evoked by malnutrition.  相似文献

In order to investigate the participation of estrogen during the period of brain sexual differentiation, male rats were treated with clomiphene citrate in the neonatal phase. Fertility and sexual behavior were assessed during adult life. Sexual maturation, body weight, and wet weight of the testes were unchanged. Although the adult male rats treated with clomiphene in the neonatal phase presented a significant reduction in the frequency of mounts, 90% of these rats were able to mate with normal females, which became pregnant. However, these females exhibited a significantly increased number of pre- and post-implantation losses. When these adult male rats were castrated and received estrogen, 60% presented female sexual behavior (receptive behavior and acceptance of mount). Thus, treatment of pups with clomiphene immediately after birth has a long-term effect on the reproductive physiology and sexual behavior of male rats.  相似文献

Daily injections of 100 μg estradiol benzoate activated the whole pattern of sexual behavior in castrated sexually experienced male rats. If compared to rats treated daily with 100 μg testosterone propionate, the estrogen-treated males tended to have longer latencies and more mounts and intromissions prior to ejaculation. Fifty micrograms of estradiol benzoate stimulated the display of mounts and intromissions in prepuberally castrated male rats. No peripheral effects of the estrogen treatment were noted. These results suggest that estrogen has central “androgen-like” effects, but no such effects in the periphery. Estrogen treatment (5, 50, and 200 μg/kg for 3 weeks) of intact sexually experienced male rats resulted in testicular atrophy and loss of body weight, but had no significant effects on the sexual behavior.  相似文献
