In this experiment we (i) tested the hypothesis that, besides decreasing leaf C fixation, lime induced iron (Fe) deficiency increases root C fixation via PEP carboxylase and (ii) assessed the Fe-induced modifications in the elemental composition of plant tissues. Sugar beet plants were grown in nutrient solutions with Fe (45 M Fe-EDTA; +Fe control) or in a similar nutrient solution without Fe (–Fe) and in presence of CaCO3 (1.0 gL–1), either labelled with 13C (20 at. %) or unlabelled. After 7 and 17 days from treatment imposition, plants were harvested and single organs analysed for total O, C, H, macro and micronutrients. 13C abundance was also assessed in control, unlabelled and labelled –Fe plants. Iron deficiency caused significant growth reductions; chlorophyll and net photosynthesis decreased markedly in Fe-deficient plants when compared to the controls, whereas leaf transpiration rates and stomatal conductance were not affected by Fe deficiency. Iron deficient plants had leaf biomass with lower C (2 to 4%) and higher O (3 to 5%) concentrations than +Fe plants. The 13C was higher (less negative) in +Fe than in –Fe unlabelled plants. Iron deficient plants grown in the nutrient solution enriched with labelled CaCO3 absorbed a relatively small amount of labelled C, which was mainly recovered in the fine roots and accounted for less than 2% of total C gain in the 10 d treatment period. Evidences suggest that iron deficient sugar beets grown in the presence of CaCO3 do not markedly shift their C fixation from leaf RuBP to root PEPC.  相似文献

Maize plants of CPB2 and CPB8 hybrids were kept under water deficit for 22 d. In the CPB8 hybrid, leaf rolling initiated at the 9th d of water deficit period, while in CPB2 hybrid it was at the 15th d. Both hybrids showed leaf rolling initiation at the same leaf water potential, ΨW of -0.480±0.095 MPa. At leaf rolling initiation, the leaf osmotic potential, ΨS was -0.730±0.085 MPa in CPB8 and 0.630±0.110 MPa in CPB2. The leaf temperature and stomatal conductance were higher in CPB8 than in CPB2. Values of leaf ΨW, ribulose-1,5-bisphosphate carboxylase activity, chlorophyll content, and specific leaf area were similar in both hybrids. Phosphoenolpyruvate carboxylase activity and protein content were lower in the CPB2 hybrid than in CPB8. In both hybrids leaf rolling initiation was associated with: (1) higher leaf temperature, with leaf rolling effect related to leaf temperature reduction, and (2) lower leaf ΨS, related to osmotic adjustment as an additional component of drought-tolerance strategy. The initial (in vivo) and total (activity present after preincubation with CO2 and Mg2+) activities of ribulose bisphosphate carboxylase were both assayed in extracts of leaves of soybean (Glycine max) plants which had been grown under 4 different irradiance levels. The total carboxylase activity per unit leaf area decreased with decreased irradiance during growth but was not different on a dry weight basis. The initial activity as a percentage of the total activity was unchanged (approximately 95%) except in leaves of plants grown at the lowest irradiance (74%). When the plants grown at the lowest irradiance were exposed to high irradiance, the initial activity was increased to 93% of the total. Light saturated rates of photosynthesis per unit leaf area were lower and saturated at lower irradiance for plants grown at lower irradiances. Initial carboxylase activity was correlated closely (r2=0.84) with leaf photosynthesis rate on a dry weight basis.  相似文献

A pot experiment was conducted to study the effects of root pruning at the stem elongation stage on the growth and water use efficiency (WUE) of winter wheat (Triticum aestivum). The results showed that stomatal conductance (g) and transpiration (E) of wheat were very sensitive to root pruning. After root pruning, they declined rapidly and but returned to pre-pruning values 15 days after treatment. Under well-watered conditions, there was no significant difference in leaf water potential (ψleaf) between root pruned and control plants after root pruning. Under moderate drought stress, ψleaf of root pruned plants declined significantly compared to the control 3 days after root pruning. After 15 days, ψleaf of root pruned plants was similar to the controls. Under different soil moisture levels, net assimilation rate (A) of root pruned plants was lower than controls 3–7 days after root pruning, but was similar to the controls 15 days after pruning. At anthesis (50 days after root pruning), root pruned plants showed significantly higher A compared with the control. Leaf area per tiller and tiller number of root pruning plants was significant lower than the control at booting stage, which showed that root pruning restrained the growth of plants in the early growing stage, but leaf area per stem, of root pruned plants, was similar to the control at anthesis. Under both soil moisture levels, there was no significant difference in grain yield between root pruned and the control plants in the monoculture. In mixture with the control plants, the root pruned plants was less productive and had a lower relative yield (0.92 and 0.78, respectively) compared with the control (1.13 and 1.19, respectively), which suggested that the pruned plants lost some of its competing ability and showed a lower ability to acquire and use the same resources in the mixture compared with the control plant. Over the whole growing cycle, root pruning reduced water consumption (by 10% under well-watered conditions and 16% under moderate drought stress) of wheat significantly compared to the control (< 0.05), and but there was no significant difference in grain yield between root pruned and control plants. Therefore root pruned wheat had a higher WUE with respect to grain yield compared with the controls. In conclusion, lowering water consumption by root pruning in the early growing stage is an effective way to improve water use efficiency in arid and semi arid areas.  相似文献

Leaf physiological and gas-exchange traits of a summer-green herbaceous perennial, Parasenecio yatabei, growing along a stream were examined in relation to leaf age. In its vegetative phase, the aerial part of this plant consists of only one leaf and provides an ideal system for the study of leaf longevity. Volumetric soil water content (SWC) decreased with increasing distance from the stream, whereas relative light intensity was nearly constant. The light-saturated net CO2 assimilation rate (A sat) and leaf stomatal conductance (gs) were approximately 1.5-fold and 1.4-fold higher, respectively, in the lower slope near the mountain stream than in the upper slope far from the mountain stream. The lifespan of aerial parts of vegetative plants significantly increased with decreasing SWC. The leaf mass-based nitrogen content of the leaves (N mass) was almost constant (ca. 2.2%); however, the maximum carboxylation rate by ribulose-1,5-biphosphate carboxylase/oxygenase (rubisco) (V cmax) and photosynthetic nitrogen use efficiency (PNUE, A sat/N area) decreased more slowly in the upper slope than in the lower slope. The higher leaf photosynthetic activity of P. yatabei plants growing lower on the slope leads to a decrease in V cmax and PNUE in the early growing season, and to a shorter leaf lifespan.  相似文献

Activity of ribulose 1,5-bisphosphate (RuBP) carboxylase in leaf extracts of the constitutive Crassulacean acid metabolism (CAM) plant Kalanchoe pinnata (Lam.) Pers. decreased with increasing leaf age, whereas the activity of phosphoenolpyruvate (PEP) carboxylase increased. Changes in enzyme activities were associated with changes in the amount of enzyme proteins as determined by immunochemical analysis, sucrose density gradient centrifugation, and SDS gel electrophoresis of leaf extracts. Young developing leaves of plants which received high amounts of NO 3 - during growth contained about 30% of the total soluble protein in the form of RuBP carboxylase; this value declined to about 17% in mature leaves. The level of PEP carboxylase in young leaves of plants at high NO 3 - was an estimated 1% of the total soluble protein and increased to approximately 10% in mature leaves, which showed maximum capacity for dark CO2 fixation. The growth of plants at low levels of NO 3 - decreased the content of soluble protein per unit leaf area as well as the extractable activity and the percentage contribution of both RUBP carboxylase and PEP carboxylase to total soluble leaf protein. There was no definite change in the ratio of RuBP carboxylase to PEP carboxylase activity with a varying supply of NO 3 - during growth. It has been suggested (e.g., Planta 144, 143–151, 1978) that a rhythmic pattern of synthesis and degradation of PEP carboxylase protein is involved in the regulation of -carboxylation during a day/night cycle in CAM. No such changes in the quantity of PEP carboxylase protein were observed in the leaves of Kalanchoe pinnata (Lam.) Sour orange (Citrus aurantium L.) grown in low-P (9–12 ppm) and high-P (420 ppm) soil inoculated with or without Glomus intraradices (G.i.), were evaluated for biomass, carbohydrates, ribulose bisphosphate carboxylase (RuBPCase), phosphoenolpyruvate carboxylase (PEPCase) activity, leaf 14CO2 incorporation, and other physiological parameters. Growth of plants in the low-P, noninoculated soil was lowest, with total dry biomass reduced up to half of the low-P, inoculum treatment. Total nonstructural carbohydrates were 40% lower in leaves of plants in the low-P, noninoculated soil, compared with the other treatments. Inoculation of the low-P soil enhanced leaf 14CO2 incorporation by 67%, total chlorophyll content by 28%, and RuBPCase activity by 42%, compared with low-P, noninoculated treatment. Improved P-use efficiency by G.i. in low-P soil was comparable to high-P nutrition in improving leaf 14CO2 incorporation and concentration of major leaf photosynthetic products that include starch and sucrose. Leaf PEPCase activity in the low-P, noninoculated treatment, however, was at least threefold higher than the other treatments, suggesting a possible alteration in organic acid metabolism in sour orange leaves as a result of P deficiency.  相似文献

Phaseolus vulgaris (cv. Hawkesbury Wonder) was grown over a range of NaCl concentrations (0–150 mM), and the effects on growth, ion relations and photosynthetic performance were examined. Dry and fresh weight decreased with increasing external NaCl concentration while the root/shoot ratio increased. The Cl- concentration of leaf tissue increased linearly with increasing external NaCl concentration, as did K+ concentration, although to a lesser degree. Increases in leaf Na+ concentration occurred only at the higher external NaCl concentrations (100 mM). Increases in leaf Cl- were primarily balanced by increases in K+ and Na+. X-ray microanalysis of leaf cells from salinized plants showed that Cl- concentration was high in both the cell vacuole and chloroplast-cytoplasm (250–300 mM in both compartments for the most stressed plants), indicating a lack of effective intracellular ion compartmentation in this species. Salinity had little effect on the total nitrogen and ribulose-1,5-bisphosphate (RuBP) carboxylase (EC 4.1.1.39) content per unit leaf area. Chlorophyll per unit leaf area was reduced considerably by salt stress, however. Stomatal conductance declined substantially with salt stress such that the intercellular CO2 concentration (C i) was reduced by up to 30%. Salinization of plants was found to alter the 13C value of leaves of Phaseolus by up to 5 and this change agreed quantitatively with that predicted by the theory relating carbon-isotope fractionation to the corresponding measured intercellular CO2 concentration. Salt stress also brought about a reduction in photosynthetic CO2 fixation independent of altered diffusional limitations. The initial slope of the photosynthesis versus C i response declined with salinity stress, indicating that the apparent in-vivo activity of RuBP carboxylase was decreased by up to 40% at high leaf Cl- concentrations. Iron nutrition affects cadmium accumulation and toxicity in rice plants   总被引:12,自引:0,他引:12  
The effect of iron (Fe) nutrition on cadmium (Cd) toxicity and accumulation in rice plants was studied using a hydroponic system. The inhibitory effect of Cd on plant growth and chlorophyll content (SPAD value) was dependent on Fe level and the genotype. Malondialdehyde (MDA) content in leaves and roots was not much affected by an increased Cd stress at 0.171 mg l−1 Fe, but it showed a rapid increase when the plants were exposed to moderate (1.89 mg l−1) and high (16.8 mg l−1) Fe levels. High Fe nutrition caused a marked reduction in Cd content in both leaves and roots. Fe content in plants was lower at high Cd (5.0 μM) stress than at low Cd (<1.0 μM) stress. Cd stress increased both superoxide dismutase (SOD) and peroxidase (POD) activities at low and moderate Fe levels. However, with high Fe level, it increased the POD activity, but reduced the SOD activity. Our results substantiate the hypothesis that cell membrane-bound iron transporter (carrier) involved in high-affinity iron transport systems can also transport Cd, and both these ions may compete for this common carrier. The study further showed that there were significant correlations between MDA and Fe contents in leaves and roots of rice plants. It is suggested that the occurrence of oxidative stress in plants exposed to Cd stress is mediated by Fe nutrition. The present results also show that Cd stress affects the uptake of Cu and Zn.  相似文献

Some elemental levels, morphological and photosynthetic characteristics and cellular metabolites of wheat plants (Triticum aestivum L. cv. Vergina) growing in the field on an ore body (Cu concentration in the soil 3050 ug g?1) were compared to control plants growing in the same environment where the Cu concentration in the soil was 140 μg g1. The concentrations of Cu, K, Pb, Zn, Mg and Fe were higher in the ore plants but Ca was lower. Growth of the ore plants was inhibited, with decreased height (25%), weight (5%), leaf area (7%) and leaf dry weight (5%) compared to the control plants. Leaf protein concentration of the ore plants was 16. 2 mg cm2 leaf area, 63% of that of the control plants. The ore plants were chlorotic and chlorophyll concentration was 3. 8 μg cm?2 leaf area, 6. 4 times lower than that in the control plants. Ribulose 1. 5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4. 1. 1. 39) activity was 13. 4 μmol CO2 fixed (mg dry weight)?1 min?1, 164% of the activity in control plants. Therefore, growth inhibition did not appear to result from inhibition of the dark reactions of photosynthesis. High Rubisco activity appears to be maintained to permit maximal CO2 fixation rates whenever energy is available. Parameters of chlorophyll fluorescence Fm, Fv, I1/2 and Fv/Fm were lower in the ore-grown plants than in control plants; Fm was higher. These values indicate that there is a decrease in the pool size of the electron acceptors on the reducing side of photosystem II (PSII), a destruction of PSII centres and interference in the photochemistry of PSII. The nitrogen content, soluble sugars, starch and lipid content were lower in ore-grown plants. Lower carbohydrate levels appear to result from low photosynthetic activity. The fatty acid composition of lipids was similar in both groups. A lower proportion of polyunsaturated fatty acids was found in the ore-grown plants. Stress caused by high Cu concentration in the soil appears to affect the light reactions of photosynthesis leading to growth inhibition.  相似文献

The specific activity of cytokinin oxidase/dehydrogenase (EC 1.5.99.12) (CKX) was determined in leaves of wild type (wt) and ethylene-insensitive mutant (eti5) of Arabidopsis thaliana (L.) Heynh plants. Comparative studies showed that this mutation has lower basal CKX activity than wt. Application of 4PU-30 (N1-(2-chloro-4-pyridyl)-N2-phenylurea) resulted in decreased CKX activity in both wt and mutant plants. The treatment increased leaf blade thickness and the volume of chlorophyll-containing cells per unit leaf area in wt but these changes were not observed in the eti5 mutant. The reduction in chlorophyll “a” and “b”, as well as in carotenoids content in the treated wt tissues resulting from altered leaf morphology was not detected in eti5 plants.  相似文献

Effects of an increased lead (Pb) content in soil on growth, photosynthesis (P N) and anatomical parameters of Plantago major L. plants grown under controlled conditions were studied. The total dry weights of plants at 500 and 2000 mg kg–1 Pb in soil were correspondingly 70% and 54% of those of control plants. A reduced leaf area and changed leaf structure caused a decrease in P N in the whole plant. The specific leaf weight (SLW) increased as compared with that of control plants. An increasing Pb content in soil caused a larger number of chloroplasts and larger sizes of protoplasts, a decrease in the chlorophyll a+b contents and a larger number of stomata per unit leaf area based both in adaxial and abaxial epidermis, as compared with control plants. However their conductance was from 40% to 50% lower than that in control plants. It was noted that the dimensions of conducting bundles decreased mainly because of the reduced xylem area. The lower functional activity and the changes at different structural levels of the photosynthetic apparatus caused a decrease in the growth rate of plants at a high Pb content in soil.  相似文献

Morpho-physiological responses to bicarbonate-induced Fe deficiency were investigated in five Vitis vinifera L. Tunisian varieties (Khamri, Blanc3, Arich Dressé, Beldi, and Balta4). One-month-old woody cuttings were cultivated for 85 days on a free calcareous soil irrigated with tap water containing increasing bicarbonate levels (0, 4, 8, 12, and 16 mM NaHCO3). After this screening, a second experiment compared root biochemical responses of two contrasting genotypes (tolerant-sensitive) dealing with bicarbonate-induced iron deprivation (20 μM Fe ± 10 mM HCO3) for 75 days. Using morpho-physiological criteria, grapevine tolerance to HCO3-induced Fe shortage appeared to be genotype-dependent: Balta4 and Beldi varieties showed the highest leaf-chlorosis score (especially at the extreme HCO3 levels), in contrast to Khamri variety. Growth parameters (shoot height, total leaf area, leaf number, and biomass production) as well as juvenile leaf chlorophyll content were also differently affected depending on both genotype and bicarbonate dose. At 16 mM HCO3, Khamri was the less sensitive variety, contrasting with Balta4. On the other hand, chlorophyll content correlated positively with HCl-extractible Fe content of the juvenile leaves, suggesting that the grapevine response to iron deficiency may partly depend on to the plant ability to adequately supply young leaves with this element. Root biochemical responses revealed a relatively higher root acidification capacity in Khamri (tolerant) under Fe-deficiency while no significant changes occurred in Balta4 (sensitive). In addition, Fe(III)-reductase and phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31) activities were strongly stimulated by Fe-deficiency in Khamri, while remaining constant in Balta4. These findings suggest that biochemical parameters may constitute reliable criteria for the selection of tolerant grapevine genotypes to iron chlorosis.  相似文献

Mature green leaves from tobacco (Nicotiana tabacum L.) plants were submitted to contrasting light conditions; half of each leaf was shaded (changed from 60 to 25 mol photons· m-2 ·s-1=LL) and the other half was exposed to higher light (changed from 60 to 360 mol·m-2· s-1=HL) for 24 h. The activity and quantity of ribulose-1,5-bisphosphate carboxylase (RuBPCase) were measured during the first 24 h in each leaf region and the variation was compared with that of small subunit (SSU)-and large subunit (LSU)-mRNA contents determined by a hybridot technique. Each leaf half responded separately to the actual light received. The activity of RuBPCase increased progressively in the HL zones and decreased in the LL zones. The RuBPCase-protein content was not significantly modified during the first 24 h but SSU-mRNA content responded very rapidly to the treatment. Within 2 h a significant difference in SSU mRNA appeared between LL and HL zones: at the end of the photoperiod the content in LL zones was approx. 25% of the initial value. The increase in the exposed zone, however, was not significant, indicating that there was a dissymmetry of the response to variation in incident white light. The LSU-mRNA contents from the same leaf extracts were totally unaffected by the light treatment. A hydroponics culture experiment was conducted to investigate the effect of iron plaque on Cd uptake by and translocation within rice seedlings grown under controlled growth chamber conditions. Rice seedlings were pre-cultivated for 43 days and then transferred to nutrient solution containing six levels of Fe (0, 10, 30, 50, 80 and 100 mg L−1) for 6 days to induce different amounts of iron plaque on the root surfaces. Seedlings were then exposed to solution containing three levels of Cd (0, 0.1 and 1.0 mg L−1) for 4 days. In order to differentiate the uptake capability of Cd by roots with or without iron plaque, root tips (white root part without iron plaque) and middle root parts (with iron plaque) of pre-cultivated seedlings treated with 0, 30 and 50 mg L−1 Fe were exposed to 109Cd for 24 h. Reddish iron plaque gradually became visible on the surface of rice roots but the visual symptoms of the iron plaque on the roots differed among treatments. In general, the reddish color of the iron plaque became darker with increasing Fe supply, and the iron plaque was more homogeneously distributed all along the roots. The Fe concentrations increased significantly with increasing Fe supply regardless of Cd additions. The Cd concentrations in dithionite–citrate–bicarbonate (DCB)-extracts and in shoots and roots were significantly affected by Cd and Fe supply in the nutrient solution. The Cd concentrations increased significantly with increasing Cd supply in the solution and were undetectable when no Cd was added. The Cd concentrations in DCB-extracts with Fe supplied tended to be higher than that at Fe0 at Cd0.1, and at Cd1.0, DCB-Cd with Fe supplied was significantly lower. Cd concentrations in roots and shoots decreased with increasing Fe supply at both Cd additions. The proportion of Cd in DCB-extracts was significantly lower than in roots or shoots. Compared to the control seedlings without Fe supply, the radioactivity of 109Cd in shoots of seedlings treated with Fe decreased when root tips were exposed to 109Cd and did not change significantly when middle parts of roots were exposed. Our results suggest that root tissue rather than iron plaque on the root surface is a barrier to Cd uptake and translocation within rice plants, and the uptake and translocation of Cd appear to be related to Fe nutritional levels in the plants.  相似文献

The ability of Fe-deficient cucumber plants to use iron complexed to a water-extractable humic substances fraction (WEHS), was investigated. Seven-day-old Fe-deficient plants were transferred to a nutrient solution supplemented daily for 5 days with 0.2 μM Fe as Fe-WEHS (5 μg org. C mL-1), Fe-EDTA, Fe-citrate or FeCl3. These treatments all allowed re-greening of the leaf tissue, and partial recovery of dry matter accumulation, chlorophyll and iron contents. However, the recovery was faster in plants supplied with Fe-WEHS and was already evident 48 h after Fe supply. The addition of 0.2 μM Fe to the nutrient solution caused also a partial recovery of the dry matter and iron accumulation in roots of Fe-deficient cucumber plants, particularly in those supplied with Fe-WEHS. The addition of WEHS alone (5 μg org. C mL-1, 0.04 μM Fe) to the nutrient solution slightly but significantly increased iron and chlorophyll contents in leaves of Fe-deficient plants; in these plants, dry matter accumulation in leaves and roots was comparable or even higher than that measured in plants treated with Fe-citrate or FeCl3. After addition of the different iron sources for 5 days to Fe-deficient roots, morphological modifications (proliferation of lateral roots, increase in the diameter of the sub-apical zones and amplified root-hair formation) and physiological responses (enhanced Fe(III)-chelate reductase and acidification of the nutrient solution) induced by Fe deficiency, were still evident, particularly in plants treated with the humic molecules. The presence of WEHS caused also a further acidification of the nutrient medium by Fe-deficient plants. The Fe-WEHS complex (1 μM Fe) could be reduced by intact cucumber roots, at rates of reduction higher than those measured for Fe-EDTA at equimolar iron concentration. Plasma membrane vesicles, purified by two-phase partition from root microsomes of Fe-deficient plants, were also able to reduce Fe-WEHS. Results show that Fe-deficient cucumber plants can use iron complexed to water soluble humic substances, at least in part via reduction of complexed Fe(III) by the plasma membrane Fe(III)-chelate reductase of root cells. In addition, the stimulating effect of humic substances on H+ release might be of relevance for the overall response of the plants to iron shortage. In the present work, the response of tobacco (Nicotiana tabaccum L.) wild-type SR1 and transgenic CAT1AS plants (with a basal reduced CAT activity) was evaluated after exposure to the herbicide paraquat (PQ). Superoxide anion (O2.−) formation was inhibited at 3 or 21 h of exposure, but H2O2 production and ion leakage increased significantly, both in SR1 or CAT1AS leaf discs. NADPH oxidase activity was constitutively 57% lower in non-treated transgenic leaves than in SR1 leaves and was greatly reduced both at 3 or 21 h of PQ treatment. Superoxide dismutase (SOD) activity was significantly reduced by PQ after 21 h, showing a decrease from 70% to 55%, whereas catalase (CAT) activity decreased an average of 50% after 3 h of treatment, and of 90% after 21 h, in SR1 and CAT1AS, respectively. Concomitantly, total CAT protein content was shown to be reduced in non-treated CAT1AS plants compared to control SR1 leaf discs at both exposure times. PQ decreased CAT expression in SR1 or CAT1AS plants at 3 and 21 h of treatment. The mechanisms underlying PQ-induced cell death were possibly not related exclusively to ROS formation and oxidative stress in tobacco wild-type or transgenic plants.  相似文献

The activities of several enzymes, including ribulose-1,5-diphosphate (RuDP) carboxylase (EC 4.1.1.39) and phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) were measured as a function of leaf age in Z. mays. Mature leaf tissue had a RuDP-carboxylase activity of 296.7 mol CO2 g-1 fresh weight h-1 and a PEP-carboxylase activity of 660.6 mol CO2 g-1 fresh weight h-1. In young corn leaves the activity of the two enzymes was 11 and 29%, respectively, of the mature leaves. In senescent leaf tissue, RuDP carboxylase activity declined more rapidly than that of any of the other enzymes assayed. On a relative basis the activities of NADP malic enzyme (EC 1.1.1.40), aspartate (EC 2.6.1.1) and alanine aminotransferase (EC 2.6.1.2), and NAD malate dehydrogenase (EC 1.1.1.37) exceeded those of both PEP and RuDP carboxylase in young and senescent leaf tissue. Pulse-chase labeling experiments with mature and senescent leaf tissue show that the predominant C4 acid differs between the two leaf ages. Labeling of alanine in senescent tissue never exceeded 4% of the total 14C remaining during the chase period, while in mature leaf tissue alanine accounted for 20% of the total after 60 s in 12CO2. The influence of increasing concentrations (0.1, 1.0 and 5.0 mg l−1) of fluoranthene (FLT) on growth, endogenous abscisic acid (ABA) level and primary photosynthetic processes in 21-day-old pea plants (Pisum sativum L.) in vitro was investigated. Murashige and Skoog’s (MS) medium, with or without FLT, was enriched with indole-3-acetic acid (IAA; 0.1 mg l−1) or a combination of IAA (0.1 mg l−1) plus N6-benzyladenine (BA; 0.1 mg l−1). The level of endogenous ABA significantly increased with increasing FLT concentrations in the presence of both IAA and IAA plus BA. An increased level of endogenous ABA was observed in plants treated with IAA alone. The growth of shoot, callus and the content of photosynthetic pigments (chlorophyll a and b, carotenoids), in both IAA- and IAA plus BA-treated plants, were significantly stimulated by FLT at its lowest concentration (0.1 mg l−1) assayed in this study. However, FLT at higher concentrations (1.0 and 5.0 mg l−1) significantly inhibited all these parameters. Chlorophyll fluorescence imaging showed that FLT only at the highest concentration (5.0 mg l−1) in the presence of IAA (0.1 mg l−1) significantly increased F0, but decreased FV/FM and ΦII.  相似文献
