The immunomodulatory effect of lithium chloride (LiCl) administered i.p. to adult mice (150 mg/kg/day) on cellular immunity in vivo was investigated. A short (6-day) treatment with LiCl of either spleen cell donors in semiallogeneic or xenogeneic GVH reaction, or recipients in semiallogeneic HVG reaction, significantly diminished the early sings of local cell-mediated immunity. A short-term LiCl treatment of donor mice also abrogated systemic GVH reaction in newborn F1 recipients, while a treatment prolonged for 20 days did not alter the high GVH response of spleen cells typical for untreated donors. Thus, a striking time-dependent efficacy of LiCl on the reduction of GVH reaction was found.  相似文献

In order to study pregnancy-induced changes in cell-mediated immunity to Listeria monocytogenes, acquired resistance and T cell functions in pregnant mice were compared with those in nonpregnant mice after immunization with viable listerial cells. Impaired generation of acquired resistance was evident in pregnant mice from the impaired elimination of bacteria and poor survival after secondary challenge. Delayed footpad reactivity to listerial antigen was also lower in the pregnant mice. When immune spleen cells were examined for their ability to produce macrophage activating factor in vitro, culture supernatants from pregnant-mouse spleen cells with listerial antigen showed far less ability to render macrophages cytostatic for P815 mastocytoma cells. To elucidate further the impairment of listeria-immune T cell generation in pregnant mice, a local transfer experiment was carried out. When a given number of immune spleen cells was transferred locally into the footpads of naive mice, both delayed footpad reaction and local protection were much lower in the pregnant mice. This local transferability of the reactions was abrogated after treatment of cells with anti-Thy 1 antibody plus complement. These findings indicate that pregnancy impairs the generation of specific T cells capable of contributing to acquired resistance to L. monocytogenes. Possible mechanisms for this impairment and the relationship to macrophage functions are discussed.  相似文献

Progesterone concentration in jugular blood of bred beef heifers was determined on days 9 and 16 in two trials. Human chorionic gonadotrophin (HCG) was administered to some of the heifers in each trial in an attempt to improve pregnancy percentage.In Trial 1, 183 heifers were divided into a control group and three groups of animals which were subjected to a 9-day estrous synchronization treatment prior to breeding. The treatment consisted of an ear implant containing 17 α acetoxy-11-beta-methyl 17 nor preg 4-ene 3, 20 dione (norgestomet) left in place for 9 days and an injection of 5 mg estradiol valerate (EV) and 3 mg of norgestomet given at the time of implantation. The heifers in one group received .25 mg estradiol-17β at time of implant removal; heifers in the 2nd group received 1500 IU of HCG in 5% beeswax and 95% sesame oil at breeding time, while heifers in the 3rd group received a placebo injection containing 5% beeswax and 95% sesame oil at breeding time. No differences in serum levels of progesterone were observed (P>0.5) between treatments or between pregnant and non-pregnant heifers on day 9 or 16 (P>.05). Pregnancy percentage in heifers receiving HCG was similar to that noted in the control heifers or the placebo injected heifers while injection of estradiol 17β decreased the proportion of heifers which became pregnant.In trial 2, 58 heifers which had been bred 1 or 2 times without becoming pregnant were divided into a control group and a group in which heifers received 1000 IU of HCG 96 hr. after observed estrus. In heifers receiving HCG, serum levels of progesterone were higher (P<.01), on day 9 and 16 post estrus than in controls but no difference in serum progesterone was noted (P>.05) between pregnant and non-pregnant heifers on day 9 or 16. The proportion pregnant did not differ (P>.05) between heifers receiving HCG and control heifers.  相似文献

Cervical extensibility increased from Day 16 to term in the pregnant rat. Following ovariectomy on Day 16 of pregnancy the cervix became as inextensible by Day 20 as that of non-pregnant animals. Fetal growth was maintained in rats ovariectomized on Day 16 if given oestradiol benzoate plus progesterone but cervical extensibility only increased to a small extent. Relaxin given to these animals further increased cervical extensibility, suggesting a role for this hormone.  相似文献

We have evaluated the effect of purified human α-fetoprotein (AFP) on several in vitro correlates of cell-mediated immunity. AFP (100 μg/ml) had no effect on antigen-induced migration inhibitory factor (MIF) production or on the ability of T cells to bind sheep erythrocytes. In contrast, AFP had a twofold effect on mitogen-and antigen-induced lymphocyte proliferation. In a dose-dependent fashion (1–100 μg/ml), AFP was mitogenic to lymphocyte cultures and also suppressed tritiated thymidine incorporation by PHA- and SK-SD-stimulated cultures. Albumin had somewhat similar effects on lymphocyte proliferation but only at 100 μg/ml. The reason for the latter was not clear since the albumin preparation was free of any detectable AFP. Our studies suggest that human AFP may not have any biologically significant immunosuppressive function.  相似文献

The effect of increasing age on various tests of cell-mediated immunity was investigated in BALB/c mice both in vitro and in vivo with four different assay systems. The following results were obtained. 1) In contact sensitivity to DNFB, old mice (age 60 to 80 weeks) showed no differences in sensitization when compared to young adult mice (age 8 to 12 weeks). (In contrast, old NZB/W mice showed impaired contact sensitization when compared with young NZB/W MICE.)2) Unlike the reaction in contact sensitivity, cells from old BALB/c mice were defective in eliciting a graft-vs-host reaction. This was true also when a partially purified population of T cells was transferred. 3) In the mixed lymphocyte reaction, cells from old mice were as efficient or better than cells from young adult BALB/c mice in responding to or stimulating allogeneic cells. 4) Responses to PHA and Con A (Both T cell mitogens) were greatly reduced when old cells were cultured as compared with cells from young adult mice. Thus, we have found that within the same batch of mice, increasing age was associated with increased capabilities in some measures of cell-mediated immunologic function and decreased capabilities in other measures of the same.  相似文献

The lymphocyte transformation test with phytohemagglutinin as mitogen estimated according to the incorporation of 2-(14)C-thymidine in DNA was used as an indicator of cell-mediated reactivity in 53 healthy subjects. Three age groups were examined: up to 20 years (21 subjects), 21-40 years (10 subjects) and over 70 years (22 subjects). The responsiveness of lymphocytes decreased significantly with age. Female CBA mice were mated to fertile CBA males, to vasectomized CBA males, to fertile C57BL males or to vasectomized C57BL males. After allogeneic or syngeneic mating the extent of thymic involution on the 10th day of pregnancy and pseudopregnancy was similar. Lumbar lymph node weight was not affected by pseudopregnancy but increased similarly in allogeneic and syngeneic pregnancies. Serum progesterone levels on the 10th day of pseudopregnancy were similar to those of non-pregnant females, and significantly lower than those of pregnant females. On the 4th to 7th days progesterone levels in pseudopregnant animals were equal to those in pregnant animals. Progesterone levels and thymic involution were similar in syngeneically and allogeneically pregnant females. Progesterone levels were negatively correlated with thymus weight but reached significance only when the mating was allogeneic. It is suggested that there is an interaction between progesterone concentrations and the degree of thymic involution during pregnancy.  相似文献

Women mount more vigorous antibody- and cell-mediated immune responses following either infection or vaccination than men. The incidence of most autoimmune diseases is also higher in women than in men; however, during pregnancy many autoimmune diseases go into remission, only to flare again in the early post-partum period. Successful pregnancy requires that the female immune system tolerate the presence of a semi-allogeneic graft for 9 months. Oral contraceptive use can increase susceptibility to certain genital tract infections and sexually transmitted diseases in women. Moreover, treatment of mice and rats with female sex hormones is required to establish animal models of genital tract Chlamydia, Neisseria and Mycoplasma infection. This review describes what is currently known about the effects of the female sex hormones oestradiol and progesterone on innate and adaptive immune responses in order to provide a framework for understanding these sex differences. Data from both human and animal studies will be reviewed.  相似文献

Two independent trials were conducted to evaluate 1) the effect of cloprostenol (CLP; ICI 80996) on subsequent corpus luteum size and progesterone content and 2) the effect of CLP and CLP followed by HCG (1500 IU) at estrus on daily serum progesterone levels in Brahman cows.In Trial 1, cows were assigned as untreated controls (n=8) or to receive 500 μg CLP intramuscularly on day 8–12 postestrus (n=9). Corpora lutea were removed surgically and weighed on day 13 after the spontaneous or CLP induced estrus. CL progesterone (P4) was also monitored.In Trial 2, cows were assigned as untreated controls (n=15), to receive 500 μg CLP on day 9, 10 or 11 postestrus (n=10), or to receive CLP as above, plus 1500 IU HCG 12 hr after the CLP-induced estrus (n=10). Daily blood samples were collected from all cows from day 2 postestrus through the second estrus, thus encompassing the period of CL development and regression.The data generated in Trial 1 indicated that CLP depressed CL weight (2.7 vs 4.7 mg; P<.05) and CL P4 content (220.88 vs 367.43 μg; P<.05) as compared to untreated controls. Serum P4 during the time period corresponding with CL development was lower (P<.05) in CLP-treated cows in Trial 2. The most distinct reduction occurred from day 7 through 10. Postestrus treatment with 1500 IU HCG appeared to increase CL steroidogenic capacity, however, a significant (P<.05) difference was not detected between CLP + HCG and CLP or control groups.  相似文献

Twenty-four pregnant, light horse mares were used in a study to determine if an exogenous progestin or progesterone would alter serum concentrations of progesterone. On day 40 of gestation, mares were randomly assigned to one of three administration groups: 1) 250 mg of progesterone in oil every other day, 2) 22 mg of Altrenogest (Regumate, American Hoechst, Somerville, NJ 08876) orally every day, or 3) 10 ml of neobee oil (control) orally every day. The treatment period was from day 40 to 105. Pregnancy status was monitored on days 40, 60, 80, 100 and 105 and a single jugular blood sample was obtained daily from days 40 to 46, 69 to 75 and 99 to 105. Serum concentrations of progesterone were determined by radioimmunoassay. Concentrations of progesterone were similar (P>0.05) among groups at each sampling period. Overall concentrations of progesterone increased (P<0.001) from days 40 to 46. Injection of 250 mg of progesterone in oil failed (P>0.05) to maintain concentrations of progesterone in serum above baseline for 24 hr. Thus higher doses and/or more frequent injections would be needed in order to increase concentrations of progesterone above those seen in untreated controls. In summary, Altrenogest was found to be a nonstressful, convenient method of administering progestins to pregnant mares without altering their endogenous secretion of progesterone.  相似文献

Oestrogen-primed and withdrawn chicks were injected with oestradiol benzoate, progesterone, and/or the anti-oestrogens tamoxifen and 4-hydroxytamoxifen. Oestrogen receptors were studied in oviduct chromatin solubilized by mild digestion of purified nuclei with micrococcal nuclease. After a single injection of oestradiol benzoate, ultracentrifugation on sucrose gradients of chromatin extracts labelled with [3H]-oestradiol showed two peaks of oestradiol binding sites, sedimenting at 13--14 S and 7--8 S. After repeated injections of oestradiol benzoate, the 13--14 S peak increased more than the 7--8 S peak. After injection of anti-oestrogen alone or together with oestradiol benzoate, no [3H]oestradiol-binding or 4-hydroxy[3H]tamoxifen-binding peaks were detected in the chromatin. Injection of progesterone also produced an increase of the 13--14 S and 7--8 S chromatin oestradiol receptor. Progesterone receptor could only by detected in chromatin early after progesterone administration, and it sedimented in density gradients with the 12 S mononucleosome fraction. Tamoxifen injected together with progesterone gave higher levels of 13--14 S oestrogen binding sites than did progesterone alone. The presence of a 13--14 S peak of oestrogen binding sites in hormonal situations which promote a biological response in the chick oviduct, and the absence of this peak after administration of anti-oestrogens, suggest that this subfraction of chromatin contains elements involved in gene regulation.  相似文献

Staphylokinase is a highly fibrin-specific clot-dissolving agent that constitutes a promising drug for clinical development. It is of bacterial origin, and the majority of patients develop neutralizing Ab after its administration. Several antigenic regions, recognized by these Ab, have been identified, but the underlying immunogenic features of staphylokinase remain unknown. In this study, we show that staphylokinase is a T cell-dependent Ag, and that an immunological memory may be acquired, even without administration of staphylokinase. Thrombolysis with staphylokinase provokes the proliferation of staphylokinase-specific T lymphocytes, which remain elevated over 10 mo posttreatment. Interestingly, analysis of a large number of staphylokinase-specific T cell clones isolated from 10 unrelated donors revealed only six distinct immunogenic regions in the molecule. Moreover, five of the six regions are recognized by T lymphocytes from several individuals, indicating that these regions are not restricted to a single HLA-DR allele. Therefore, these new insights can guide the design of variants with a lower immunogenic profile in humans.  相似文献

Eighteen crossbred multiparous sows were allocated at random to one of two lactation lengths: 42 or 10 days. All sows were mated at the oestrus after weaning and from mating until day 26 post coitum they were bled every second day. Progesterone and oestradiol concentration in the plasma was determined by radioimmunoassay. Progesterone increased more rapidly between days 4 and 10 post coitum in early weaned sows and the oestradiol surge at mating was abnormally extended for the same group.  相似文献
