粗毛栓菌粗酶液经聚丙烯酰胺凝胶电泳,在CBBG-250染色后,漆酶带处有一透明圈,经纯化漆酶和CBBG-250溶液直接作用以及菌体的培养结果证实,漆酶对CBBG-250具有一定的脱色降解作用,在漆酶活力达118u/ml时,CBBG-250溶液在595nm波长的光吸收60min内下降了32.4%。该粗毛栓菌产漆酶对工业染料废水也具有一定的降解脱色作用。  相似文献

本研究以PCR扩增的方法,从变色栓(菌Trametes versicolor)的基因组DNA中扩增出了一预期大小的DNA片断,并将其克隆到了pUCM-T载体上。经筛选、酶切、PCR鉴定,序列分析,证明该片断为变色栓菌漆酶基因的克隆。该基因的开放阅读框由1560个核苷酸组成,编码一个由519个氨基酸组成的多肽。与GeneBank中发表的Laccase基因(AY081188)序列比较发现,编码的氨基酸序列同源性为96%,而核苷酸序列的同源性为92%。  相似文献

根据铜结合区的保守氨基酸序列设计简并引物,扩增栓菌420(Trametessp.420)基因组,结合长距离反向PCR(LD-IPCR)技术,克隆得到新型漆酶同工酶B基因(lacB),包括结构基因(2255bp)及5′-和3′-非编码序列。lacB含12个内含子,其cDNA序列长1560bp,编码495aa成熟多肽和24aa信号肽。lacB与其它不同来源的真菌漆酶基因具有较高的同源性,而与植物、细菌、昆虫的漆酶基因同源性低于25%。将不含信号序列的lacBcDNA通过质粒pPIC9克隆到表达载体pPIC9K上,电击转化毕赤酵母GS115细胞,经BMM-ABTS平板筛选得到漆酶分泌阳性转化子。  相似文献

红栓菌(pycnoporus cinnabarius)在发酵培养3d后出现胞外漆酶活性峰。木素类似物对红栓菌胞外漆酶活性有诱导作用,阿魏酸、香兰素,愈创木酚和DL-β-苯丙氨酸诱导24h后,发酵液中漆酶活性分别是对照的2.8、4.3、3.5和1.7倍。发酵液经(NH_4)_2S0_4沉淀,Sephadex G-150、DEAE-Sephadex A-25、Sephadex G-25柱层析纯化后,冷冻干燥。高效液相色谱(HPLC)检测为一单峰,分子量为26000,含17种氨基酸,氨基酸总量占酶组成的64.27%。等离子光谱(ICP)分析表明漆酶含有铜。该酶反应的最适温度为30℃,与邻联苯甲胺反应最适pH为4.0,Km值为385μmol/L;与丁香醛连氮反应最适pH为5.8,Km值为833μmol/L。  相似文献

栓菌420(Trametes sp. 420)漆酶基因lacD以两种方式在巴斯德毕赤酵母(Pichia pastoris)进行异源表达,产生两种重组漆酶:rLacDx(具有天然N-末端)和rLacDe(N-末端带有8个额外的氨基酸残基)。摇瓶发酵18d,rLacDx和rLacDe的产量分别为1.21×105u/L、7.38×104u/L [以2,2′-连氮-3-乙苯-二噻唑-6磺酸(ABTS)为底物]。在高密度发酵条件下,rLacDx的产量增加到2.39×105u/L,同时其生产周期降至7.5 d。两种重组酶对愈创木酚底物的氧化特性相似,且在50℃和pH3~10的范围内均稳定。然而,rLacDx对底物ABTS的比活力(1761u/mg)高于rLacDe (1122u/mg),其表观Km值(427μmol/L)低于rLacDe (604μmol/L)。  相似文献

以白腐菌为出发菌株,利用紫外线(UV)进行诱变,筛选高产漆酶突变菌株。通过测定致死率绘制出发菌株的致死曲线,采用PDA-RBBR平板变色法进行初筛,ABTS检测酶活对突变株进行摇瓶复筛。结果表明:利用15 w紫外灯在照射距离为30 cm,照射时间为120 s,致死率为72.1%的条件下进行诱变处理,获得一株高产菌株,其酶活提高79.54%,经过5代传代培养,未见酶活下降,具有较好的遗传稳定性,进一步研究了初始pH值,接种量和培养基装液量等对诱变菌株产酶的影响,结果表明在最佳的培养条件pH值6.0,15%的装液量于28℃下,酶活达214.9 U/L。  相似文献

【目的】为提高漆酶产量,降低生产成本,以山核桃蒲壳作为基质,对粗毛栓菌D2固态发酵产漆酶的营养条件进行研究。【方法】对不同碳源、氮源、碳氮比、蒲壳含量对漆酶产量的影响进行分析。【结果】山核桃蒲壳是粗毛栓菌生长的良好载体,能够促进漆酶的合成。粗毛栓菌D2漆酶固态发酵培养基干物质组成为:山核桃蒲壳40%(质量比),玉米粉24%(质量比),菜籽饼粉36%(质量比)。发酵6 d时,漆酶活性为126.8 U/g干基。【结论】粗毛栓菌固态发酵山核桃蒲壳产漆酶具有效率高,生产成本低的优点,具有潜在的工业化应用前景。  相似文献

漆酶是一种绿色高效的多酚氧化酶,在降解双酚A方面具有巨大潜力。为降低发酵产漆酶的成本及考察漆酶在双酚A降解中的能力,本研究以麸皮和柚皮为主要基质,优化了栓菌固态发酵产漆酶条件,对优化后获得的漆酶在双酚A降解中的应用进行了研究。结果表明,在培养基组分为:麸皮和柚皮粉比例为6:4(W/W)、固液比1:2.5(W/V)、铜离子2%(W/W)、蔗糖3%(W/W)、硝酸钾2%(W/W)、稻壳20%(W/W)的条件下,栓菌发酵产漆酶的酶活最高,发酵11d后,酶活可达到38.4U/gds。当双酚A初始浓度为10μg/mL时,在55℃条件下酶解140min后,双酚A基本降解完全。  相似文献

In a medium containing 40 g ethanol l–1, laccase production by Trametes versicolor was 2.6 unit per ml of the supernatant, which was over 20 times higher than that without ethanol. Laccase activity with ethanol was quite comparable to that with the well-known inducers such as veratryl alcohol, xylidine and guaiacol. With other white-rot fungi, Coriolus hirsutus and Grifola frondosa, ethanol had a similar stimulatory effect on laccase production.  相似文献

毛栓菌原生质体制备和再生及单核菌株产漆酶特性   总被引:1,自引:0,他引:1  
毛栓菌Trametes hirsuta能有效地降解木质素,在生物燃料、制浆和饲料工业等方面具有很高应用价值。为了获得遗传性能稳定的T. hirsuta单核菌株,研究了其菌丝生长培养基的类型、菌丝生长时间(菌龄)、酶解时间、原生质体纯化离心速度和原生质体再生培养基类型对T. hirsuta YJ-9-1原生质体制备与再生的影响;采用DAPI染色和锁状联合缺失的观察,从再生株中筛选单核菌株并考察其产酶特性。结果表明:采用YGM菌丝生长培养基、88h菌龄、1h酶解时间、4,000r/min原生质体纯化离心速度以及YGMS再生培养基,最终可获得密度大约为5.0×106个/mL的原生质体悬浮液和9.1%的再生率;从200株再生菌株中筛选出了3株单核菌株,其中一株单核菌株D-2-1的漆酶产量比原菌T. hirsuta YJ-9-1明显提高,在第12天其漆酶酶活为771.67U/L,是原菌的1.51倍。  相似文献

Diverse food wastes, apple, orange and potato, were screened for laccase production, under solid-state fermentation conditions, by the white-rot fungus Trametes hirsuta. Potato peelings gave the highest activity, reaching about 5000 U l–1 within 8 days. These values are higher than those reported to date.  相似文献

La3+ and Ce3+, either singly or a mixture, promoted crocin production of Crocus sativus callus but Nd3+ had little effect and all metal ions were toxic above 100 microM. La3+ (60 microM) promoted growth of callus significantly but increased crocin only slightly. Ce3+ (40 microM) significantly promoted crocin production but had little effect on cell growth. La3+ (60 microM) and Ce3+ (20 microM) together gave the highest dry weight biomass (20.4 g l(-1)), crocin content (4.4 mg g(-1)) and crocin production (90 mg l(-1)) which were, respectively, 1.7-fold, 4.2-fold and 7.1-fold of those without additions. Nd3+ (40 microM) only slightly promoted cell growth and crocin production.  相似文献

In the current studies, we used Lineweaver-Burke analysis to examine the role of 1-hydroxybenzotriazole (HBT) in the oxidation of various compounds by laccase from Trametes versicolor. At low concentrations, HBT was a competitive inhibitor of the oxidation, but at high concentrations, it was a noncompetitive inhibitor. Analysis of the oxidation of ferrocytochrome c by the laccase-HBT couple showed that increasing the concentration of ferrocytochrome c did not affect the V(max) but reduced the apparent K(m). In addition, in the manganese peroxidase-Mn(II) reaction, which is a typical oxidation system by mediator, the apparent K(m) and V(max) increased as the concentration of the substrate 2,6-dimethoxyphenol was increased. These results indicate that HBT is involved in the binding of laccase and substrates that laccase cannot oxidize alone.  相似文献

Production of the indole alkaloids, ajmalicine or catharanthine, in cell suspension cultures of Catharanthus roseus was enhanced by cerium (CeO2 and CeCl3), yttrium (Y2O3) and neodymium (NdCl3). The yield of ajmalicine in these treated-cultures reached 51 mg l–1 (CeO2), 40 mg l–1 (CeCl3), 41 mg l–1 (Y2O3) and 49 mg l–1 (NdCl3) while catharanthine production reached to 36 mg l–1 (CeO2) and 31 mg l–1 (CeCl3). A major portion of increased alkaloids was released into medium in these treatments. But Sm2O3, SmCl3, La2O3, LaCl3, complex of chromium (III)-titanium (IV) and NaSeO4 treatments had little effect on alkaloid production of C. roseus cell cultures.  相似文献

Agrochemicals, industrial compounds and their transformation products have been assayed for their ability to enhance laccase production in liquid cultures of Trametes versicolor, when added at 0.5 mM. After 3 days of treatment, enzymatic activity in the culture medium was increased 14-fold by 4-n-nonylphenol and 24-fold by aniline. Laccase activity was enhanced 10-fold by oxidised derivatives of the herbicide diquat, 17-fold by N,N-dimethyl-N-(5-chloro,4-hydroxyphenyl)urea and 22-fold by 9-fluorenone.  相似文献

Grape seeds were used by Trametes hirsuta as a substrate for laccase production giving 23 kU l–1, which was 10-fold the value attained in the cultures with no lignocellulosic waste addition. The dyes, Indigo Carmine and Bromophenol Blue, were easily decolourised (100% in 24 h) by the extracellular liquid obtained in such cultures, whereas Methyl Orange (65% in 24 h) and Phenol Red (36% in 24 h) were more resistant to degradation. This shows the specificity of laccase towards different dye structures.  相似文献
