Samples of wheat naturally infected in the field byFusarium culmorum (W.G.Sm.) Sacc. andFusarium graminearum Schwabe were analyzed for deoxynivalenol, 3-acetyldeoxynivalenol, and zearalenone. No zearalenone was detected at levels higher than 0.5mg/kg. Deoxynivalenol was present in 100% and 3-acetyldeoxynivalenol in 80% of the examined samples at levels of 0.21 to 30.4 mg/kg and 0.54 to 29.54 mg/kg, respectively. The mycotoxin levels in the chaff were 5 to 50 times higher than in the kernels. This is the first report on natural occurrence of 3-acetyldeoxynivalenol in wheat. This toxin, in addition to deoxynivalenol, was highly correlated with wheat head fusariosis. These findings suggest that more attention should be given to the occurrence of 3-acetyldeoxynivalenol in cereal grains during the growth as well as during storage.  相似文献

When wheat is stored under suboptimal conditions, a further mycotoxin increase of deoxynivalenol (DON), but especially of mycotoxins produced by storage fungi, e.g. ochratoxin A, is possible, lowering wheat quality and food safety. Different storage trials were conducted under suboptimal storage conditions.Fusarium survival during suboptimal storage was monitored by cultural technique and multiplex-PCR and set into relation to DON contents. Furthermore, XANES spectroscopy was applied on a selected storage trial in order to characterize sulfur speciation in low molecular weight (LMW) subunits of glutenin isolated from suboptimally stored wheat samples highly infected withFusarium and from wheat infected withAspergillus andPenicillium. Distinct changes in sulfur speciation were observed in grains infected with storage fungi, especially a significant increase of higher oxidation states (sulfoxide state, sulfonate state). A single isolates ofFusarium graminearum Schwabe KF 366 andFusarium culmorum (W.G.Sm.) Sacc. KF 365 were used to infect 10 genotypes (9 lines and one cultivar) of winter triticale, 1 rye cultivar and 1 wheat cultivar, and amounts of mycotoxins in kernels were analysed at the same stage of development. One genotype of triticale CHD 353/79 and rye “Chodan” were found to be most resistant towards both species infection with lowest amount of mycotoxins (deoxynivalenol) content in kernels and also the lowest yield reduction. The most susceptible line CZR 142 cumulated in kernels about ten times higher amount of mycotoxins (up 53 mg DON/kg and 16 mg 3AcDON/kg, and 5 mg zearalenone/kg). GenerallyF, culmorum formed higher level of mycotoxins in kernels of infected heads thanF. graminearum. In kernels of more susceptible genotypes except deoxynivalenol, 3 acetyldeoxynivalenol and zearalenone also were present.  相似文献

The susceptibility of six winter wheat cultivars toFusarium head blight has beenstudied. The lowest infected plants intensity, mean degree of head damage and fusariosis index exhibited cultivar Grana which also cumulated the lowest amount of trichothecenes (DON and derivatives ). AIMS: To assay sago starch from Papua New Guinea (PNG) for important mycotoxins and to test fungal isolates from sago for mycotoxin production in culture. METHODS AND RESULTS: Sago starch collected from Western and East Sepik Provinces was assayed for aflatoxins, ochratoxin A, cyclopiazonic acid, sterigmatocystin, citrinin and zearalenone and all 51 samples were negative. Frequently isolated species of Penicillium (13), Aspergillus (five) and Fusarium (one) were cultured on wheat grain, and tested for the production of ochratoxin A, cyclopiazonic acid, sterigmatocystin, citrinin, patulin and penicillic acid. All 12 isolates of P. citrinin and one of two A. flavipes isolates produced citrinin. A single isolate of A. versicolor produced sterigmatocystin. No other mycotoxins were detected in these cultures. CONCLUSIONS: No evidence was found of systemic mycotoxin contamination of sago starch. However, the isolation of several mycotoxigenic fungi shows the potential for citrinin and other mycotoxins to be produced in sago stored under special conditions. SIGNIFICANCE AND IMPACT OF THE STUDY: Sago starch is the staple carbohydrate in lowland PNG and the absence of mycotoxins in freshly prepared sago starch is a positive finding. However, the frequent isolation of citrinin-producing fungi indicates a potential health risk for sago consumers, and food safety is dependant on promoting good storage practices.  相似文献

The occurrence, species diversity and some aspects of taxonomical affinity and host selectivity of acaropathogenic fungi associated with phytophagous, saprotrophic and predacious mites in Poland and other European countries were investigated on wild and cultivated plants, in insect feeding sites under the bark and in decayed wood. From among 33 species of fungi affecting mites only five species of Entomophthorales were separated and the most numerous were Neozygites floridana mostly on Tetranychus urticae, N. abacaridis on a few eriophyid species, and Conidiobolus coronatus attacking gamasid mites most frequently of the genus Dendrolaelaps. The most frequent mite pathogens occurring in mite communities on plants and in wood infested by insects were of the genus Hirsutella. Until now 13 of their form-species have been recognized in these habitats, but only H. kirchneri, H. necatrix and H. thompsonii (including its variety synnematosa) can be treated as exclusive oligophagous pathogens of phytophagous mites, though their potential host range seems to embrace only selected eriophyid or tarsonemid mites. Taxonomical differentiation of fungal strains was based on close morphological observations and molecular analysis of ITS region sequences. Two new species of acaropathogenic fungi were described in these studies. Hirsutella danubiensis sp. nov. was found in the tetranychid T. urticae, whereas H. vandergeesti sp. nov. affected phytoseiid mites of the genera Amblyseius, Neoseiulus, Seiulus and Typhlodromus, and the tarsonemid Tarsonemus lacustris.  相似文献

Using the dilution plate method, 70 species and 31 genera were collected from 20 dust samples on glucose (28 genera and 64 species) and cellulose Czapek's agar (22 genera and 46 species) at 28° C. The most common fungi were Aspergillus niger, A. flavus, A. flavus var. columnaris, Phoma glomerata, Fusarium oxysporum, Penicillium chrysogenum and Mucor racemosus; and A. nidulans, Phoma humicola, Drechslera spicifera and Stachybotrys chartarum on the two media, respectively.Toxicity test showed that about 85% of the isolates tested were toxic to brine shrimp (Artemia salina). Thin layer chromatographic analysis revealed that 13 out of 23 toxic isolates produced known mycotoxins. Toxins identified were: aflatoxins B1 and B2, Kojic acid and trichodermine.  相似文献

Electrophoretic patterns of soluble proteins and enzymes during development from dry seed to first leaf were studied in wheat by means of disc electrophoresis. The patterns considered were those of alcohol dehydrogenase (ADH), peroxidase, and nonspecific esterases. Multiple forms (isoenzymes) were found for all these enzymes in dry seeds. The ADH pattern did not change during development. Esterases and peroxidase patterns changed both qualitatively and quantitatively. The occurrence of Fusarium spp. and associated mycotoxins in asparagus spears was evaluated in Poland in 2002 and 2003 and in Germany in 2002. Spears of two cultivars, Eposs and Gijnlim, were collected from two locations in Poland, Swidwowiec and Poznan, on sandy and sandy loam soil, respectively. Fusarium oxysporum and F. proliferatum were detected at an average incidence of 38.3% and 15.8% in the spear sections sampled, respectively. In stands of 11 (tested) cultivars of asparagus sampled in Germany on sandy soil, the same species dominated, however, they were less frequent than in Poland (26.6% and 5.6% of the spears infected with F. oxysporum and F. proliferatum, respectively). Chemical analyses revealed that fumonisin B1 (FB1) and moniliformin (MON) were present in some of the spears sampled in Poland. FB1 was not found and MON was not assessed in spears sampled in Germany in 2002, but F. proliferatum was able to form the toxin in vitro in the range from 101.4 up to 205.8 μg/kg maize kernel substrate. Asparagus samples in Poland contained FB1 at up to 5.6 μg/kg spear fresh weight. The highest MON concentration (1350 μg/kg) was detected in cultivar Eposs in Marcelin, Poland, in 2002. MON and FB1 were found in spears infected by both F. oxysporum and F. proliferatum, however, only the latter fungus was able to synthesize both toxins.  相似文献

Toxigenic Fusarium species are common pathogens of wheat and other cereals worldwide. In total, 449 wheat heads from six localities in Poland, heavily infected with Fusarium during 2009 season, were examined for Fusarium species identification. F. culmorum was the most common species (72.1% on average) with F. graminearum and F. avenaceum the next most commonly observed, but much less frequent (13.4 and 12.5% respectively). F. cerealis was found in 1.8% of all samples, and F. tricinctum was found only in one sample (0.2%). Subsequent quantification of the three major mycotoxins (deoxynivalenol, zearalenone and moniliformin) in grain and chaff fractions with respect to associated prevailing pathogen species uncovered the following patterns. Moniliformin (MON) was found in low amounts in all samples with F. avenaceum present. In contrast, deoxynivalenol (DON) and zearalenone (ZEA) were the contaminants of F. culmorum- and F. graminearum-infected heads. The highest concentration of DON was recorded in grain sample collected in Radzików (77 µg g?1). High temperatures in Central Poland during July and August accompanied with high rainfall in July were responsible for this high DON accumulation. Trichothecene, zearalenone, enniatin and beauvericin chemotypes were identified among 21 purified isolates using gene-specific PCR markers.  相似文献

The aim of this work was to determine the genera or species composition and the number of colony forming units of airborne bacteria and fungi, respectively, in two salt mines in Poland “Wieliczka” (Lesser Poland) and “Polkowice–Sieroszowice” (Lower Silesia). Both of them are working environments characterized by extreme conditions, and additionally “Wieliczka,” officially placed on the UNESCO World Heritage Sites’ list, plays a role of tourist attraction. There are also some curative chambers located in this mine. Air samples were taken once in December 2015, between 6:00 a.m. and 9:00 a.m. There were nine measurement points located about 200 m underground in “Wieliczka” and six measurement points located in the working shafts about 400 m underground in “Polkowice–Sieroszowice.” The total volume of each air sample was 150 L. Air samples, collected in individual measurement points of both salt mines, were inoculated on two microbiological media: potato dextrose agar and tryptic soy agar using the impact method. We identified 10 and 3 fungal genera in the “Wieliczka” Salt Mine and in “Polkowice–Sieroszowice,” respectively. The most common were fungi of the Penicillium genus. In both mines, the Gram-positive bacteria of genus Micrococcus were detected most frequently. Among identified microorganisms, there were neither pathogenic fungi nor bacteria. The most prevalent microorganisms detected in indoor air were Gram-positive cocci, which constituted up to 80% of airborne microflora. Our results showed that microorganisms recorded in the air samples are not a threat to workers, tourists or patients. Neither pathogens nor potentially pathogenic microorganisms, listed as BSL-2, BSL-3 or BSL-4, were detected. The microbes identified during our analysis commonly occur in such environments as the soil, water and air. Some of the detected bacteria are component of natural microflora of human skin and mucous membranes, and they can cause only opportunistic infections in individuals depending on their health condition.  相似文献

Mycotoxins produced by seven strains of Penicillium vulpinum (formerly Penicillium claviforme) isolated from different sources were studied. The strains were characterized by specific profiles of secondary metabolites and produced mycotoxins of different structural types. In addition to toxins already known for this fungal species (patulin, roquefortine, 3,12-dihydroroquefortine, oxalin, viridicatin, cyclopenin, and alpha-cyclopiazonic acid), the strains studied also produced indolyl-3-acetic acid, griseofulvin, meleagrin, and cyclopeptin.  相似文献

Thirty-nine isolates of fungi obtained from foodstuffs and soil samples from various parts of the world have been identified. The isolates were grown on a solid rice medium, and extracts were prepared with 50% aqueous methanol. The extracts were examined for toxicity in the following systems: (i) cytotoxicity to cultured normal human diploid skin fibroblasts (proliferating and nonproliferating) and mouse fibroblasts; (ii) skin toxicity after topical application on rats; and (iii) rat feeding tests in which rats were examined for death, overt pathological effects including congestion and hemorrhage of tissues, weight loss, food refusal, and uterine growth. Sixteen culture extracts were highly toxic as indicated by death, congestion and hemorrhage of tissues, and net weight loss. One half of the isolates were highly cytotoxic (50% lethal concentration, 0.01 to 5 micrograms/ml) as indicated by the ability to cause death and disintegration of 3T3 Swiss mouse fibroblasts and human diploid skin fibroblasts during 3 to 4 days in culture. The remainder were moderately cytotoxic (50% lethal concentration, 5 to 250 micrograms/ml). Four culture extracts were highly toxic by some clinical criteria but did not cause congestion and hemorrhage of tissues and were weakly cytotoxic (50% lethal concentration, 250 to 5,000 micrograms/ml). Six culture extracts exhibited moderate toxicity (weight loss only) and low cytotoxicity (50% lethal concentration, 3,000 to 50,000 micrograms/ml). Four culture extracts caused uterine enlargement as the major clinical sign, suggesting the presence of zearalenone. The toxigenicity of 392 strains ofAspergillus, representing 132 species and 19 varieties, was assessed on chicks and mice fed iso-caloric 50 % basal feed mixes of wheat and soybeans molded by these strains. On the basis of death numbers (3 or more of 6 on test), low weight gain of survivors, and low feed consumption over a 4-week period, 166 of the strains, representing 73 species and 9 varieties, were toxigenic. Ten species (A. alliaceus, A. avenaceus, A. clavato-flavus, A. janus, A. melleus, A. ochraceus, A. parasiticus, A. quercinus, A. sclerotiorum, andA. sulphureus) had strains that were markedly toxic to both animal types on one or both feeds; the other 63 species and 9 varieties had strains that were moderately to mildly toxic to one or both animal types on one or both feeds. The analysis of deoxynivalenol (DON) in naturally infected wheat samples, after having been separated into four fractions through laboratory sieves, showed very low levels of DON in the fraction of largest kernels >2.8 mm (0 up to 1 mg/kg). The highest concentration of DON was found in fractions 2.2 to 2.5 mm and <2.2mm with up to 14mg/kg and 15mg/kg DON, respectively. In two samples (fractions <2.2mm) nivalenol was detected in concentrations up to 1,4mg/kg.  相似文献
