Tricladins A and B (1 and 2, resp.), new imidazolone-containing alkaloids, together with five known metabolites, bacillamides A, B (3 and 4, resp.), 20-hydroxyaflavinine (5), N-(2-phenylethyl)acetamide (6), and N(b)-acetyltryptamine (7), have been isolated from the crude extract of the ascomycete fungus Tricladium sp. The structures of 1 and 2 were elucidated primarily by NMR and MS methods. Compound 2 showed marginal cytotoxicity against MDA-MB-231 human breast cancer cells, whereas the known metabolite 4 displayed an inhibitory effect on HIV-1 replication in C8166 cells.  相似文献

A new compound (1), named diaporthelactone, together with two known compounds (2 and 3) were isolated from the culture of Diaporthe sp., a marine fungus growing in the submerged rotten leaves of Kandelia candel in the mangrove nature conservation areas of Fugong, Fujian Province of China. The new compound was elucidated to be 1,3-dihydro-4-methoxy-7-methyl-3-oxo-5-isobenzofuran-carboxyaldehyde (1), which showed cytotoxic activity against KB and Raji cell lines (IC50 6.25 and 5.51 microg mL(-1), respectively). Two known compounds, 7-methoxy-4,6-dimethyl-3H-isobenzofuran-1-one (2) and mycoepoxydiene (3), were also demonstrated to exhibit cytotoxic activities for the first time. All three compounds were assessed for antimicrobial activity.  相似文献

Extracts of cultures grown in liquid or on solid rice media of the fungal endophyte Ampelomyces sp. isolated from the medicinal plant Urospermum picroides exhibited considerable cytotoxic activity when tested in vitro against L5178Y cells. Chromatographic separation yielded 14 natural products that were unequivocally identified based on their 1H and 13C NMR as well as mass spectra and comparison with previously published data. Six compounds (2, 4, 5, 7, 9 and 11) were natural products. Both fungal extracts differed considerably in their secondary metabolites. The extract obtained from liquid cultures afforded a pyrone (2) and sulfated anthraquinones (7 and 9) along with the known compounds 1, 3, 6 and 8. When grown on solid rice medium the fungus yielded three compounds 4, 5 and 11 in addition to several known metabolites including 6, 8, 10, 12, 13 and 14. Compounds 4, 8 and 10 showed the strongest cytotoxic activity against L5178Y cells with EC50 values ranging from 0.2–7.3 μg/ml. Furthermore, 8 and 10 displayed antimicrobial activity against the Gram-positive pathogens, Staphylococcus aureus, S. epidermidis and Enterococcus faecalis at minimal inhibitory concentrations (MIC) of 12.5 μg/ml and 12.5–25 μg/ml, respectively. Interestingly, 6 and 8 were also identified as constituents of an extract derived from a healthy plant sample of the host plant U. picroides thereby indicating that the production of bioactive natural products by the endophyte proceeds also under in situ conditions within the host plant.  相似文献

Discovery of potential bioactive metabolites from sponge-associated bacteria have gained attraction in recent years. The current study explores the potential of sponge (Suberea mollis) associated bacteria against bacterial and fungal pathogens. Sponge samples were collected from Red sea in Obhur region, Jeddah, Saudi Arabia. Of 29 isolated bacteria belong to four different classes i.e. Firmicutes (62%), γ-Proteobacteria (21%), α-Proteobacteria (10%) and Actinobacteria (7%). Among them nineteen (65%) bacterial strains showed antagonistic activity against oomycetes and only 3 (10%) bacterial strains were active against human pathogenic bacteria tested. Most bioactive genera include Bacillus (55%), Pseudovibrio (13%) and Ruegeria (10%). Enzyme production (protease, lipase, amylase, cellualse) was identified in 12 (41%) bacterial strains where potential strains belonging to γ-Proteobacteria and Firmicutes groups. Production of antimicrobial metabolites and hydrolysates in these bacteria suggest their potential role in sponge against pathogens. Further bioactive metabolites from selected strain of Vibrio sp. EA348 were identified using LC-MS and GC–MS analyses. We identified many active metabolites including antibiotics such as Amifloxacin and fosfomycin. Plant growth hormones including Indoleacetic acid and Gibberellin A3 and volatile organic compound such as methyl jasmonate were also detected in this strain. Our results highlighted the importance of marine bacteria inhabiting sponges as potential source of antimicrobial compounds and plant growth hormones of pharmaceutical and agricultural significance.  相似文献

The investigation of the Vietnamese marine sponge Spongia sp. led to the isolation of three new sesquiterpene phenols, langconols A–C (13), and one new sesquiterpene hydroxyquinone, langcoquinone C (4), together with two known meroterpenoids (5 and 6). Their structures were determined on the basis of spectroscopic analyses and comparisons with published data. Furthermore, the antibacterial assays of the isolates 16 suggested that 4 and 6 had significant antibacterial activities against Bacillus subtilis and Staphylococcus aureus, with MICs ranging from 6.25 to 25.0 µM, while 1 and 3 possessed significant antibacterial activities against B. subtilis with MICs of 12.5 and 25.0 µM, respectively. In contrast, cytotoxic assays of the isolated compounds 16, as well as compounds 715 previously isolated from this sponge, indicated that 1 and the previously reported anti-B. subtilis and anti-S. aureus sesquiterpene phenol 9 lacked cytotoxic activities against three human cancer cell lines (A549, lung cancer; MCF7, breast cancer; HeLa, cervix cancer) and a human normal cell line (WI-38 fibroblast).  相似文献

From the endophytic fungal strain Geotrichum sp. AL4, cultivated from the leaves of the neem tree (Azadirachta indica), four compounds, 1-4, were isolated from the AcOEt extract, including two new, chlorinated, epimeric 1,3-oxazinane derivatives. All compounds were assessed for their nematicidal activities against the nematodes Bursaphelenchus xylophilus and Panagrellus redivivus, and three out of the four isolates showed noticeable bioactivities.  相似文献

A new acetylenic fatty acid, 1 , has been isolated from the title sponge. The structure of the molecule was elucidated to contain an enyne and a thiophene by spectroscopic methods. Compound 1 showed a weak cytotoxic effect against NBT‐T2 rat bladder epithelial cells (IC50>20 μg/ml), and antimicrobial activity with minimal‐inhibitory concentrations (MIC) of 64 and 128 μg/ml against Staphylococcus aureus and Escherichia coli, respectively.  相似文献

Bioassay‐guided isolation of a fungal strain Nigrospora sp. MA75, an endophytic fungus obtained from the marine semi‐mangrove plant Pongamia pinnata, which was fermented on three different culture media, resulted in the isolation and identification of seven known compounds, 2, 3 , and 5 – 9 , from a medium containing 3.5% NaCl, while a new compound, 2,3‐didehydro‐19α‐hydroxy‐14‐epicochlioquinone B ( 10 ) was obtained from the medium containing 3.5% NaI. In addition, two new griseofulvin derivatives, 6‐O‐desmethyldechlorogriseofulvin ( 1 ) and 6′‐hydroxygriseofulvin ( 4 ), were isolated and identified from the rice solid medium. Dechlorogriseofulvin ( 2 ) and griseofulvin ( 3 ) were the major components in fermentation extracts of all these culture media, while compounds 1 and 4, 5 and 6 , and 10 were only present in the extract of respective culture medium. The structures of these compounds were elucidated by detailed spectroscopic analysis, and the absolute configuration of 1 was determined by CD measurement. Compounds 9 and 10 exhibited antibacterial activities toward five tested bacterial strains, while compounds 5, 6 , and 8 selectively inhibited MRSA, E. coli, and S. epidermidis, and compound 3 showed moderate activity against V. mali and S. solani. Moreover, compound 10 potently inhibited the growth of MCF‐7, SW1990, and SMMC7721 tumor cell lines with IC50 values of 4, 5, and 7 μg/ml, respectively.  相似文献

Two new phytotoxic γ‐lactones, pestalotines A and B ( 1 and 2 , resp.), along with 4‐oxo‐4H‐pyran‐3‐acetic acid ( 3 ) and 6‐hydroxyramulosin (=3,4,4a,5,6,7‐hexahydro‐6,8‐dihydroxy‐3‐methyl‐1H‐2‐benzopyran‐1‐one; 4 ), were isolateded from the culture of Pestalotiopsis sp. HC02, a fungus residing in the Chondracris rosee gut. Structures of the new metabolites were elucidated on the basis of their IR, NMR, and MS data. Pestalotines A and B ( 1 and 2 , resp.) significantly inhibited the radical growth of Echinochloa crusgalli with IC50 values of 1.85×10?4 and 2.50×10?4 M , respectively, comparable to that of 2‐(2,4‐dichlorophenoxy)acetic acid (0.94×10?4 M ) used as a positive control.  相似文献

In continuation of our efforts to identify bioactive compounds from the Red Sea marine sponges, we have recently investigated the organic extract of the sponge Biemna ehrenbergi. This study resulted in the isolation of eight compounds including a new sterol, ehrenasterol (1), a new C24-acetylenic acid, biemnic acid (2), together with six known compounds including a hopanoid, three steroids and two nucleosides. The isolated compounds were identified as (22E)-ergosta-22-ene-8,14-epoxy-3,7-dione (1), (E)-tetracos-8-en-5-ynoic acid (2), (22E)-ergosta-5,8,22-trien-7-one-3β-ol (3), 32,35-anhydrobacteriohopanetetrol (4), (24R)-ergosta-6,22-diene-5,8-epidioxy-3-ol (5), melithasterol B (6), thymidine (7) and 2′-deoxyuridine (8). The structures of the isolated compounds were assigned by different spectral data including 1D and 2D NMR (COSY, HSQC, and HMBC) and high-resolution mass spectrometry. Compound 1 displayed inhibition zone of 20 mm at 100 μg/disc against Escherichia coli in the disc diffusion assay. Similarly, compounds 2 and 4 displayed inhibition zones of 20 and 18 mm respectively against Candida albicans at the same concentration. Compounds 13 displayed weak cytotoxic activity against human colon adenocarcinoma (HCT-116) cancer cell line.  相似文献

利用硅胶柱层析、制备型HPLC和重结晶等手段从普哥滨珊瑚分离的一株Talaromyces sp.真菌C21-1中筛选得到2个活性化合物,运用核磁共振、质谱和圆二色谱等技术鉴定这两个化合物分别为(R)-(-)-hydroxysydonic acid(1)和homodimeric WIN 64821(2),补充完善了化合物2的核磁共振信号归属,并对化合物进行抗菌活性和乙酰胆碱酯酶抑制活性的测定,发现化合物1对白色假丝酵母Canidia albicans和耐甲氧基青霉素的金黄色葡萄球菌Methicillin-resistant Staphylococcus aureus(MRSA)具有一定的抑制作用,其最低抑菌浓度(MIC)分别为0.075mmol/L和0.2mmol/L,对副溶血弧菌Vibrio parahemolyticus的抑制活性较弱,在0.2mmol/L浓度下的抑制率为17%;化合物2最大浓度0.2mmol/L条件下对这3种菌均没有明显的抑制效果。化合物2表现出剂量依赖的乙酰胆碱酯酶抑制活性,0.5mmol/L时抑制率达到35.1%。  相似文献

Two new sorbicillinoids, 1 and 2 , together with a novel benzofuranone derivative named phialofurone ( 3 ), were isolated from a deep‐sea sediment‐derived fungus, Phialocephala sp. Their structures were established on the basis of spectroscopic data. All compounds displayed cytotoxic effects against P388 (IC50 values of 11.5±1.4, 0.1±0.1, and 0.2±0.01 μM , resp.) and K562 (IC50 values of 22.9±0.8, 4.8±0.3 and 22.4±0.9 μM , resp.) cell lines.  相似文献

Fusarithioamide B (6), a new aminobenzamide derivative with unprecedented carbon skeleton and five known metabolites: stigmast-4-ene-3-one (1), stigmasta-4,6,8(14),22-tetraen-3-one (2), p-hydroxyacetophenone (3), tyrosol (4), and fusarithioamide A (5) were separated from Fusarium chlamydosporium EtOAc extract isolated from Anvillea garcinii (Burm.f.) DC. leaves (Asteraceae). The structure elucidation and complete assignment of the isolated metabolites were performed mainly by the aid of various NMR and MS data. Fusarithioamide B (6) has been assessed for antibacterial and antifungal activities towards various microbial strains by disc diffusion assay. It exhibited selective antifungal activity towards C. albicans (MIC 1.9?µg/ml and IZD 14.5?mm), comparing to clotrimazole (MIC 2.8?µg/ml and IZD 17.9?mm). Also, it possessed high antibacterial potential towards E. coli, B. cereus, and S. aureus compared to ciprofloxacin. Furthermore, 6 was tested for the in vitro cytotoxic effect against KB, HCT-116, BT-549, MCF-7, SKOV-3, and SK-MEL cell lines. It had selective and potent effect towards BT-549, MCF-7, SKOV-3, and HCT-116 cell lines with IC50s 0.09, 0.21, 1.23, and 0.59?μM, respectively compared to doxorubicin (IC50s 0.046, 0.05, 0.321, and 0.24?μM, respectively). Fusarithioamide B may provide a lead molecule for future developing of antitumor and antimicrobial agents.  相似文献

Three new cytochalasins Z24, Z25, Z26 ( 1 – 3 , resp.) and one known compound, scoparasin B ( 4 ), were isolated from the fungus Eutypella sp. D‐1 isolated from the soil of high latitude of the Arctic. The structures of 1 – 3 were elucidated from spectroscopic data (NMR, MS). These compounds were evaluated for cytotoxic activities against several human tumor cell lines. Among them, compound 1 exhibited moderate cytotoxicity toward human breast cancer MCF‐7 cell line with IC50 of 9.33 μM .  相似文献

We have isolated and characterized two cDNAs from the marine sponge Geodia cydonium coding for a new member of a receptor tyrosine kinase of class II. The deduced amino acid sequence shows two characteristic domains: (i) the tyrosine kinase domain; and (ii) and immunoglobulin-like domain. The latter part shows high homology to the vertebrate C2 type immunoglobulin domain. This result demonstrates that immunoglobulin domains are not recent achievements of higher animals but exist also in those animals which have diverged from other organisms about 800 million years ago.  相似文献

Five new 9,11‐secosteroids 1, 2 , and 4 – 6 , and seven known analogs, 3 and 7 – 12 , with the same steroid skeleton, (5αH)3β,6α,11‐trihydroxy‐9,11‐secocholest‐7‐en‐9‐one, were isolated from the South China Sea gorgonian Subergorgia suberosa. Among them, 2 / 3 and 4 / 5 are C(24)‐epimeric mixtures, and 6 / 7 is an (E)/(Z) mixture of (C(24)?C(28)). Their structures and relative configurations were elucidated by using comprehensive spectroscopic methods including NOESY spectra. The absolute configuration of the steroidal nucleus was established by the modified Mosher method applied to 10 and on the basis of a common biogenesis for all of these compounds. All isolated compounds, 1 – 12 , and five synthetic acetylated derivatives, 12a – 12e , were evaluated for their cytotoxicities in vitro. Compounds 4 / 5, 11, 12 , and 12b – 12d showed cytotoxic activities against K562 cell line with the IC50 values ranging from 1.09 to 8.12 μM .  相似文献

Three new sesquiterpene acids, xylaric acids A-C (1-3, resp.), and a new tetralone (=3,4-dihydronaphthalen-1(2H)-one) derivative, 4, along with nine known compounds, xylaric acid D (5), hydroheptelidic acid (6), gliocladic acid (7), chlorine heptelidic acid (8), trichoderonic acid A (9), 16-(α-D-mannopyranosyloxy)isopimar-7-en-19-oic acid (10), 16-(α-D-glucopyranosyloxy)isopimar-7-en-19-oic acid (11), 5-carboxymellein (12), and naphthalen-1,8-diol 1-O-α-D-glucopyranoside (13) have been isolated from the solid culture of the ascomycete fungus Xylaria sp. associated with termite nest. The structures of these compounds were elucidated primarily by NMR experiments. The absolute configurations of compounds 1-3 and 5-9 were determined by combination of X-ray data and CD spectral analysis. The absolute configuration of 4 was assigned by Snatzke's method. Compounds 8 and 11 showed slight cytotoxicities against two cell lines A549 and SGC7901.  相似文献

A new bisabolane-type sesquiterpene, trichoderic acid (1), and a new acorane-type sesquiterpene, 2β-hydroxytrichoacorenol (2), along with three known compounds, cyclonerodiol (3), cyclonerodiol oxide (4), and sorbicillin (5), were isolated from the culture broth of Trichoderma sp. PR-35, an endophytic fungus isolated from Paeonia delavayi. Their structures were elucidated on the basis of their IR, MS, and 1D- and 2D-NMR analyses. The antibacterial and antifungal activities of 1-5 towards various types of bacteria and fungi were tested. Most of the compounds showed moderate or weak antimicrobial activities in an agar-diffusion assay.  相似文献
