The stimulation of dicotyledonous leaf growth by light depends on increased H+ efflux, to acidify and loosen the cell walls, and is enhanced by K+ uptake. The role of K+ is generally considered to be osmotic for turgor maintenance. In coleoptiles, auxin‐induced cell elongation and wall acidification depend on K+ uptake through tetraethylammonium (TEA)‐sensitive channels (Claussen et al., Planta 201, 227–234, 1997), and auxin stimulates the expression of inward‐rectifying K+ channels ( Philippar et al. 1999) . The role of K+ in growing, leaf mesophyll cells has been investigated in the present study by measuring the consequences of blocking K+ uptake on several growth‐related processes, including solute accumulation, apoplast acidification, and membrane polarization. The results show that light‐stimulated growth and wall acidification of young tobacco leaves is dependent on K+ uptake. Light‐stimulated growth is enhanced three‐fold over dark levels with increasing external K+, and this effect is blocked by the K+ channel blockers, TEA, Ba++ and Cs+. Incubation in 10 mm TEA reduced light‐stimulated growth and K+ uptake by 85%, and completely inhibited light‐stimulated wall acidification and membrane polarization. Although K+ uptake is significantly reduced in the presence of TEA, solute accumulation is increased. We suggest that the primary role of K+ in light‐stimulated leaf growth is to provide electrical counterbalance to H+ efflux, rather than to contribute to solute accumulation and turgor maintenance.  相似文献

以2年生的流苏播种苗为材料,采用不同浓度(50、100、200、300 mmol·L^-1)NaCl溶液进行胁迫处理,研究盐胁迫对流苏的生长、Na^+和K^+分布格局、渗透调节物质的影响,以明确其耐盐阈值。结果表明:(1)随着NaCl胁迫浓度的增加,流苏幼苗生长量逐渐降低,盐害指数升高、存活率下降;幼苗耐盐阈值为98.693 mmol·L^-1(0.577%W/V)。(2)随着NaCl胁迫浓度的增加,流苏幼苗各器官中的Na^+含量持续增加,并在浓度为50 mmol·L^-1时表现为根>叶>茎,在其余各处理组表现为叶>根>茎;幼苗根、叶中的K^+含量表现为先增后减的变化趋势,茎中K^+含量总体表现为下降趋势,且器官中K^+含量表现为根>叶>茎;幼苗根部到茎部和茎部到叶部的离子选择性运输能力、各器官中的K^+/Na^+比值均呈下降趋势。(3)随着NaCl浓度的增加,流苏幼苗叶片可溶性糖、可溶性蛋白含量总体呈上升趋势,其脯氨酸含量呈先上升后下降的趋势。研究发现,流苏幼苗根系可通过对Na^+的吸收和累积来阻止其向地上部运输进而避免盐害发生;叶片和茎中通过提高对K^+的选择性吸收和累积,从而增大K^+/Na^+比值以减缓盐分对其生理代谢的伤害。  相似文献

Fusicoccin (FC) has long been known to promote K+ uptake in higher plant cells, including stomatal guard cells, yet the precise mechanism behind this enhancement remains uncertain. Membrane hyperpolarization, thought to arise from primary H+ pumping stimulated in FC, could help drive K+ uptake, but the extent to which FC stimulates influx and uptake frequently exceeds any reasonable estimates from Constant Field Theory based on changes in the free-running membrane potential (V m) alone; furthermore, unidirectional flux analyses have shown that in the toxin K+ (86Rb+) exchange plummets to 10% of the control (G.M. Clint and E.A.C. MacRobbie 1984, J. Exp. Bot.35 180–192). Thus, the activities of specific pathways for K+ movement across the membrane could be modified in FC. We have explored a role for K+ channels in mediating these fluxes in guard cells ofVicia faba L. The correspondence between FC-induced changes in chemical (86Rb+) flux and in electrical current under voltage clamp was followed, using the K+ channel blocker tetraethylammonium chloride (TEA) to probe tracer and charge movement through K+-selective channels. Parallel flux and electrical measurements were carried out when cells showed little evidence of primary pump activity, thus simplifying analyses. Under these conditions, outward-directed K+ channel current contributed appreciably to charge balance maintainingV m, and adding 10 mM TEA to block the current depolarized (positive-going)V m; TEA also reduced86Rb+ efflux by 68–80%. Following treatments with 10 M FC, both K+ channel current and86Rb+ efflux decayed, irreversbly and without apparent lag, to 10%–15% of the controls and with equivalent half-times (approx. 4 min). Fusicoccin also enhanced86Rb+ influx by 13.9-fold, but the influx proved largely insensitive to TEA. Overall, FC promotednet cation uptake in 0.1 mM K+ (Rb+), despite membrane potentials which were 30–60 mVpositive of the K+ equilibrium potential. These results tentatively link (chemical) cation efflux to charge movement through the K+ channels. They offer evidence of an energy-coupled mechanism for K+ uptake in guard cells. The electrophysiological properties of cultured human melanocytes were investigated using the whole-cell configuration of the patch-clamp technique. Depolarizations to membrane potentials more positive than -30 mV resulted in the rapid development (<1 ms to peak) of an inward current. The maximum peak current was observed at +10 mV and reached an average amplitude of about 270 pA. During the depolarizations, the current inactivated with a time constant of about 2 ms. The current was abolished by the addition of 0.3 μM tetrodotoxin, a blocker of voltage-gated Na+-channels, and disappeared when Na+ was omitted from the extracellular medium. In addition, the melanocytes contain at least two types of outward K+-current. The first type, observed in every cell, was highly sensitive (Ki 1 mM) to the K+-channel blocker TEA, required depolarizations beyond zero to be activated and did not inactivate. The second type was less regularly observed (10% of the cells). This current activated at more negative voltages (–20 mV), was resistant to TEA (20 mM) but was blocked by 2 mM 4-aminopyridine and inactivated rapidly during depolarizations. We conclude that human melanocytes are equipped with voltage-dependent Na+-channels, a delayed rectifying K+-current and a K+-current similar to the A-current in neurones.  相似文献

Summary Previous current/voltage (I/V) investigations of theChara K+ state have been extended by increasing the voltage range (up to +200 mV) through blocking the action potential with La3+. A region of negative slope was found in theI/V characteristics at positive PD's, similar to that already observed at PD's more negative than the resting level. These decreases in membrane currents at PD's more negative than –150 mV and at PD's close to 0 or positive are thought to arise from the K+ channel closure. Both the negative slope regions could be reversibly abolished by 0.1mm K+, 20mm Na+, more than 10mm Ca2+ or 5mm tetraethylammonium (TEA). The K+ channels are therefore blocked by TEA, closed by low [K+] o or high [Ca2+] o and are highly selective to K+ over Na+. With the K+ channels closed, the remainingI/V profile was approximately linear over the interval of 400 mV (suggesting a leakage current), but large rectifying currents were observed at PD's more positive than +50 mV. These currents showed a substantial decrease in high [Ca2+] o , sometimes displayed a slight shift to more positive PD's with increasing [K+] o and were unaffected by TEA or changes in [Na+] o . The slope of the linear part of theI/V profile was steeper in low [K+] o than in TEA or high [Na+] o (indicating participation of K+, but not Na+, in the leak current). Diethylstilbestrol (DES) was employed to inhibit the proton pump, but it was found that the leakage current and later the K+ channels were also strongly affected.  相似文献

Spring wheat (Triticum aestivum L. cv. Svenno), oat (Avena sativa L. cv. Brighton) and glasshouse cucumber (Cucumis sativus L. cv. Bestseller F1) were cultured for a week after germination on complete nutrient solutions of three different dilutions (1, 25 and 50% of the full strength medium). K+(86Rb) and 45Ca were present during the whole culture period. Relative humidity (RH) was 50% except during the last day, when half the material was transferred to 90% RH. Efflux of labelled ions was then followed during eight hours on unlabelled solutions of the same composition as before, and at both 50% and 90% RH in the atmosphere. – Uptake of K+(86Rb) during growth tended to be saturated in the 25% medium. Contrariwise, the level of Ca2+ in the roots increased continuously with strength of the medium. At low concentrations cucumber roots were higher in Ca2+ than roots of oat or wheat, whereas all three species showed similar levels of Ca2+ in 50% medium. – At the lowest ionic strength, smooth efflux curves were obtained that could be resolved according to the three-compartment theory. At higher ionic strength, irregularities were observed, and more for Ca2+ than for K+; but for practical purposes compartment analysis with the same time constants could be applied as for the lowest concentration. – Discrimination between K+ and Rb+ differed between the roots, but not much between the shoots of different species. The roots of oat and wheat took up Rb+ preferentially over K+ in the 25% and 50% media; whereas K+ was preferred over Rb+ or little discrimination made in 1% medium and for cucumber. The shoots generally showed less discrimination than the roots. The main variability in discrimination between K+ and Rb+ thus appears to be localized in the tonoplasts of the roots cells. – Low RH around the shoots increased efflux of K+(86Rb) from the cytoplasm and vacuoles of the root cells as compared to the efflux at high RH. DNP (2,4-dinitrophenol) in the medium had the same effect as high RH around the shoots. The signal system that must exist between shoots and roots is discussed as a response to “drought” conditions. In relation to investigations of others, it is assumed that the effect of DNP may indicate that part of the chain between roots and shoots consists of metabolically influenced sites, whose output is influenced by the rate of water transport.  相似文献

Internal perfusion of tetraethylammonium ions (TEA) in squid axons produces a significant high frequency noise component. Although internal TEA suppresses the potassium conductance (G K) noise at relatively low frequencies, it induces high frequency noise which exceeds the intensity of the normal potassium and sodium noise. In addition, the induced noise is dependent on the presence of internal potassium ions (K+) suggesting that this source of noise arises from a modulation of the K+ conductance due to the blocking and unblocking of the K+ channel. The simplest model describing the TEA data is a two-step sequential pseudo-unimolecular reaction where TEA binds during an open conductance state. A unit channel conductance of 2 pS is estimated from the TEA data as well as noise induced by triethyldecylammonium (TEDA) ions. Thus, these data are consistent with the hypothesis that the channel is blocked whenever the quaternary ammonium ion binding site, located near or within the K+ channel, is occupied.  相似文献

The properties of one ATP-inhibited and one Ca2+-dependent K+ channel were investigated by the patch-clamp technique in the soma membrane of leech Retzius neurons in primary culture. Both channels rectify at negative potentials. The ATP-inhibited K+ channel with a mean conductance of 112 pS is reversibly blocked by ATP (K i = 100 m), TEA (K i =0.8 mm) and 10 mm Ba2+ and irreversibly blocked by 10 nm glibenclamide and 10 m tolbutamide. It is Ca2+ and voltage independent. Its open state probability (P o) decreases significantly when the pH at the cytoplasmic face of inside-out patches is altered from physiological to acid pH values. The Ca2+-dependent K+ channel with a mean conductance of 114 pS shows a bell-shaped Ca2+ dependence of P o with a maximum at pCa 7–8 at the cytoplasmic face of the membrane. The P o is voltage independent at the physiologically relevant V range. Ba2+ (10 mm) reduces the single channel amplitude by around 25% (ATP, TEA, glibenclamide, tolbutamide, and Ba2+ were applied to the cytoplasmic face of the membrane).We conclude that the ATP-dependent K+ channel may play a role in maintaining the membrane potential constant—independently from the energy state of the cell. The Ca2+-dependent K+ channel may play a role in generating the resting membrane potential of leech Retzius neurons as it shows maximum activity at the physiological intracellular Ca2+ concentration.This study was supported by the Deutsche Forschungsgemeinschaft (W.-R. This study focuses on the effects of K+ depolarization on neurite elongation of identified Helisoma neurons isolated into culture. Application of K+ to the external medium caused a dose-dependent suppression of neurite elongation. Lower concentrations of K+ were associated with a slowing in the rate of neurite elongation, whereas higher concentrations produced neurite retraction. Surprisingly, the effects of K+ depolarization were transient, and neurite elongation rates recovered towards control levels within 90 min even though the neurons remained in high-K+ solution. Identified neurons differed in the magnitude of their response to K+ depolarization; neurite elongation of buccal neuron B4 was inhibited at 5 mM K+, but elongation in B5 and B19 was not affected until concentrations of 25 mM. Electrophysiologically, K+ application evoked a brief period (5–10 s) of action potential activity that was followed by a steady-state membrane depolarization lasting 2 h or more. The changes in neurite elongation induced by K+ depolarization occurred in isolated growth cones severed from their neurites and were blocked by application of calcium antagonists. Intracellular free Ca2+ levels in growth cones of B4 and B19 increased and then decreased during the 90-min depolarization, corresponding to the changes in elongation. B4 and B19 showed differences in the magnitude, time course, and spatial distribution of the Ca2+ change during depolarization, reflecting their different sensitivities to depolarization.  相似文献

Plant roots accumulate K+ over a range of external concentrations. Root cells have evolved at least two parallel plasma-membrane K+ transporters which operate at millimolar and micromolar external [K+]: high-affinity K+ uptake is energised by symport with H+, while low-affinity uptake is assumed to occur via ion channels. To determine the role of ion channels in low-affinity K+ uptake, a characterisation of the principal K+-selective ion channels in the plasma membrane of Arabidopsis thaliana (L.) Heynh. cv. Columbia roots was undertaken. Two classes of K+-selective channels were frequently observed: one inward (IRC) and one outward (ORC) rectifying with unitary conductances of 5 pS, 20 pS (IRCs) and 15 pS (ORC), measured in symmetrical 10 mM KCl. The dominant IRC (5 pS) and ORC (15 pS) were highly cation-selective (PCl PK < 0.025) but less selective amongst monovalent cations (PNaPK0.17–0.3). Both the IRC and the ORC were blocked by Ba2+, Cs+ and tetra-ethyl-ammonium, whereas 4-aminopyridine and quinidine selectively inhibited the ORC. The ORC open probability was steeply voltage-dependent and ORC activation potentials were close to the potassium equilibrium potential (EK+), enabling ORCs to conduct mainly outward, but occasionally inward, K+ current. By contrast, gating of the 5-pS IRC was weakly voltageependent and IRC gating was invariably restricted to membrane potentials more negative than EK+, ensuring K+ transport was always inwardly directed. Studies on channel activity were conducted for a large number of root cells grown at two levels of external [K+], one where K+ uptake is likely to be principally through channels (6 mM K+) and one where it must be energised (100 M K+). Shifting growth conditions from high to low K+ did not affect single-channel properties such as conductance and selectivity, nor the manifestation of the ORC and 20-pS IRC, but led to enhanced activity of the 5-pS IRC. Extrusion of H+ by intact barley (Hordeum vulgare L.) roots was automatically titrated. Simultaneously, uptake of K+ into the roots, transport of K+ through the roots, and (as a residual term) accumulation of K+ within the root tissue were determined. When no monovalent cation was present in the medium the steady rate of H+ release was close to zero. Addition of K+ stimulated H+ extrusion within less than 1 min. The stimulation of H+ release was apparently limited only by the movement of K+ through the apoplast of the roots. The steady rate of H+ extrusion depended on the availability of external K+ and saturated at a K+ concentration of about 100 mol· dm-3. Half-maximum rates of net K+ uptake and H+ extrusion were reached at a K+ concentration of about 10 mol·dm-3. With (slowly absorbable) sulfate as the only anion present, the stoichoimetry between H+ release and net K+ uptake was one. In conclusion, the uptake of K+ across the plasmalemma of the cells of the root cortex is electrically coupled to H+ extrusion.  相似文献

Summary Ultrastructural cytochemical localization of ouabain-sensitive, potassium dependent p-nitrophenyl phosphatase (K+-NPPase) of the Na+-/K+-ATPase complex and adenylate cyclase (cAMPase) activities, in washed inactivated human platelets, are described. The one-step lead-citrate method, under similar incubation conditions, was used to determine both activities. K+-NPPase appeared in both plasma membrane and the surface-connected canalicular system (SCCS) of the platelets. These data suggest a uniform distribution of the enzyme throughout membrane systems which are in contact with the external medium. cAMPase activity was strictly localized in tubules of the dense tubular system (DTS) when incubation medium contained prostaglandin E1, prostaglandin D2 or forskolin, at concentrations known to stimulate the enzyme in platelets that are intact. This fact and the inhibition of cytochemical reaction by thrombin confirm that the one-step lead-citrate method is a useful procedure in determining adenylate cyclase, abolishing the unfavorable conditions of previously reported methods.  相似文献

The relationship between Rb+ influx and microsomal ATPase activity stimulated by K+ and Mg2++ K+ was investigated for roots of 7-day-old seedlings of oat (Avena sativa L., cv. Brighton). Different concentrations of K+ in the roots, K+root were produced by cultivating plants in complete nutrient solutions of different dilutions and dFifferent K+ concentrations at various temperatures. Experiments were performed in both light and darkness. The range of the influx/ATPase ratios was large with a factor of 5 or more between the highest and lowest values. In most cases, the highest ratios were obtained at low K+root and at high temperatures, and the lowest at high K+root and at low temperatures. At high temperatures (20 and 25°C) in the light, the influx/ATPase ratio was constant, independently of K+root, if K+ in the medium was kept constant but the bulk of the nutrient solution diluted. If K+ was varied and the other components of the medium kept constant, the normal relation of decreasing influx/ATPase ratio at increasing K+root was found; thus, Rb+ influx appears regulated by both the internal and external potassium conditions. Also in darkness, at 15°C and with K + in the medium varied, the influx/ATPase ratio was independent of K+root but in the corresponding light experiments, ratio and K+root had the normal, inverse relationship. The difference between light and dark conditions appears to indicate that growth rate is of importance for the relationship between energy input and transport. Our data lead to the concept of “flexible coupling” between transducers) of energy and ion carrier. Without excluding other possibilities, this may be one of the mechanisms for ecological adaptation to variations in the root medium.  相似文献

Root elongation by wheat seedlings (Triticum aestivum L. cv. Scout 66) was not inhibited by NaCl or KCl up to 130 mM in culture solutions or by high Na+ (2 mg g-1 FW) or K+ (4 mg g-1 FW) in the root tissue, provided that [Ca2+]>2 mM in the rooting medium. At [NaCl], [KCl], or [mannitol] >250 mOs, root elongation was progressively inhibited, irrespective of high [Ca2+]. In contrast, shoot elongation was sensitive to any diminution of water potential, and Ca2+ alleviated the toxicity only weakly. At solute concentrations <250 mOs, the following interactions were observed. Ca2+ alleviated Na+ and K+ toxicity to roots by at least three separate mechanisms. K+ was more toxic to roots than Na+, but Na+ was more toxic to shoots. Low levels of K+ relieved Na+ toxicity, but low levels of Na+ enhanced K+ toxicity. Tissue concentrations of Na+ were reduced by Ca2+ and K+ in the rooting medium, and tissue concentrations of K+ were enhanced by Ca2+ and Na+. Several hypotheses relating to salinity toxicity can be evaluated, at least for wheat seedlings. The osmoticant hypotheses (salinity intoxication occurs because of diminished water potential) is true for shoots at all salinity levels, but is true for roots only at high salinity. The Ca2+-displacement hypothesis (Na+ is toxic because it displaced Ca2+ from the cell surface) is correct, but often of minor importance. The K+-depletion hypothesis (Na+ is toxic because it causes a loss of K+ from plant tissues) is false. The Cl--toxicity hypothesis (the apparent toxicity of Na+ is induced by associated Cl-) is false. The results indicate that, apart from osmotic effects, high levels of Na+ in the rooting medium and in the tissues are not toxic unless Ca2+ is also deficient, a condition probably leading to inadequate compartmentation and excessive cytoplasmic accumulation. This study related growth to ion activities at plasma-membrane surfaces. Abstract

The effect of Li+, Rb+ and tetraethylammoniumchloride (TEA) on the locomotor activity rhythm of Musca domestica was studied. Li+ as well as TEA lengthen the free run period t. This effect was more pronounced in animals with shorter period lengths. The effect of Rb? depends on the previous period length. In flies which had a short period, Rb+ lengthened, and in those with a long period Rb+ shortened t. Replacing the Li+ solution with water reverted the period lengthening partly. In the case of Rb? and TEA the rhythm became obscured after replacing the solutions with water. The involvement of K+ ions and transport ATPases in the expression of circadian rhythms is discussed.  相似文献

Summary K+ channels in cultured rat pancreatic islet cells have been studied using patch-clamp single-channel recording techniques in cell-attached and excised inside-out and outside-out membrane patches. Three different K+-selective channels have been found. Two inward rectifier K+ channels with slope conductances of about 4 and 17 pS recorded under quasi-physiological cation gradients (Na+ outside, K+ inside) and maximal conductances recorded in symmetrical K+-rich solutions of about 30 and 75 pS, respectively. A voltage- and calcium-activated K channel was recorded with a slope conductance of about 90 pS under the same conditions and a maximal conductance recorded in symmetrical K+-rich solutions of about 250 pS. Single-channel current recording in the cell-attached conformation revealed a continuous low level of activity in an apparently small number of both the inward rectifier K+ channels. But when membrane patches were excised from the intact cell a much larger number of inward rectifier K+ channels became transiently activated before showing an irreversible decline. In excised patches opening and closing of both the inward rectifier K+ channels were unaffected by voltage, internal Ca2+ or externally applied tetraethyl-ammonium (TEA) but the probability of opening of both inward rectifier K+ channels was reduced by internally applied 1–5mm adenosine-5-triphosphate (ATP). The large K+ channel was not operational in cell-attached membrane patches, but in excised patches it could be activated at negative membrane potentials by 10–7 to 10–6 m internal Ca2+ and blocked by 5–10mm external TEA.  相似文献

Action potentials in the afterdischarge of the ovulation hormone producing caudo–dorsal cells (CDC) of Lymnaea stagnalis are strikingly different from electrically evoked spikes in the silent resting and inhibited states of these cells. Spikes evoked in the silent states consist of one fast peak (80–100 mV; 10–15 ms). The overshoot is Na+ - and Ca2+ -dependent. Spikes are blocked in Na+ -free saline and by TTX. Repolarization is retarded by TEA. Co2+ increases the overshoot. Active state action potentials (60–80 mV) last up to 125 ms, due to activation of a slow component following the TTX-sensitive spike. The slow component is Na+ - and Ca2+ -dependent. In normal saline it is blocked by Co2+ and La3+. In Ca2+ -free saline the remaining part of the slow component is blocked by La3+ only. The slow component is voltage-dependent in a graded fashion. Activation is bound to the active state in which the CDC are depolarized by 20 mV. TEA and Ca2+ -free saline greatly increase spike duration in the active state. This suggests that, in addition to the classical TEA-sensitive channel, a Ca2+ -dependent K+ channel is involved in repolarization of active state action potentials. The underlying membrane properties and the functional significance are discussed in relation to the pacemaking mechanism of the CDC.  相似文献

The members of the RCK family of cloned voltage-dependent K+ channels are quite homologous in primary structure, but they are highly diverse in functional properties. RCK4 channels differ from RCK1 and RCK2 channels in inactivation and permeation properties, the sensitivity to external TEA, and to current modulation by external K+ ions. Here we show several other interesting differences: While RCK1 and RCK2 are blocked in a voltage and concentration dependent manner by internal Mg2+ ions, RCK4 is only weakly blocked at very high potentials. The single-channel current-voltage relations of RCK4 are rather linear while RCK2 exhibits an inwardly rectifying single-channel current in symmetrical K+ solutions. The deactivation of the channels, measured by tail current protocols, is faster in RCK4 by a factor of two compared with RCK2. In a search for the structural motif responsible for these differences, point mutants creating homology between RCK2 and RCK4 in the pore region were tested. The single-point mutant K533Y in the background of RCK4 conferred the properties of Mg2+ block, tail current kinetics, and inward ion permeation of RCK2 to RCK4. This mutant was previously shown to be responsible for the alterations in external TEA sensitivity and channel regulation by external K+ ions. Thus, this residue is expected to be located at the external side of the pore entrance.
