Summary Calcium fluxes were measured simultaneously in brown trout fry maintained in an artificial soft water medium of [Ca] 20 mol·l-1 and pH 5.6, and exposed to each of six trace metals (Al, Cu, Fe, Ni, Pb, and Zn). The trace metal concentrations represented typical and maximum levels found in acid waters experiencing declining fishery status. In the absence of trace metals, evidence is presented which suggests that ca. 91% of Ca taken up from the external medium was by extraintestinal active transport. Calcium efflux was stimulated by both concentrations of Al, Cu, Fe, and Pb. Efflux was also stimulated by [Ni] 170 nmol·l-1 and [Zn] 3000 nmol·l-1. In some cases, response to increased efflux was stimulation of influx. Lack of stimulation of influx resulted in negative net Ca fluxes. Effects of sodium, potassium and calcium on salt-stressed barley   总被引:7,自引:0,他引:7  
We grew barley ( Hordeum vulgare L. CM 72) for a 28-day period and sequentially harvested plants every 3 or 4 days. Plants were salt-stressed with either NaCl or KCl (125 m M ) with or without supplemental Ca (10 or 0.4 m M final concentration, respectively). We determined tissue concentrations of Na, Ca, Mg, K. S, P, Fe, Mn, Cu and Zn for each harvest date by inductively coupled plasma spectrometry. Uptake (specific absorption rate) was calculated from the element content and growth rates. Salinity had significant effects on the uptake and concentrations of most elements. Mg and Mn concentrations declined with time. The concentrations of all other elements determined increased over time. Element uptake on a root dry weight basis declined with time. Three variables were significantly affected by salinity and correlated with growth; 1) the Ca concentration, 2) the total sum of the cation concentration (TC), and 3) the Mn concentration of the shoot. Salinity reduced Ca uptake and concentrations. Supplemental Ca increased Ca concentrations and was positively correlated with growth during salt stress. Salinity doubled TC, which was negatively correlated with relative growth rate (RGR). Relative growth rate declined at TC values above 150 m M . Salinity reduced the uptake and concentration of Mn. Manganese concentrations in the shoot were highly correlated with RGR. Relative growth rate declined at Mn concentrations below 50 nmol (g fresh weight)−1.  相似文献

We investigated the relationships between major nutrients (C, H, N, and P) and trace metals (Cu, Fe, and Mn) in seston samples from ten lake/lagoon systems in southern Brazil. The systems were characterized by a diverse set of limnological features, including surface areas from 10−1 to 102 km2, water color, a CDOM(440), from 1.4 to 12.9 m−1, and electrical conductivity from 50 to 100 000 μS cm−1. Seston concentrations also varied a great deal, 32-fold. The elemental (C: N, C: P, and N: P) and C: Chl-a ratios in the seston samples indicated, however, common features; i.e., most of the lakes were N-and/or P-limited, and the seston organic fraction was composed of nonvascular plants (e.g., phytoplankton). Our intersystem comparison revealed that the relative content of organic matter in seston and seston concentrations in lake water tended to correlate positively and negatively, respectively, with trace metal concentrations across the seston samples. Possible influences of elemental and C: Chl-a ratios on the association of metals with seston matrices, although theoretically important, were only partially evidenced here; positive correlations were found between C: N and also Org-H: N ratios with trace metal concentrations. We speculate that such results could be circumstantial, as the nature of the seston matrices appeared to be very similar among them. This hypothesis should thus be the theme of further research. In short, our findings suggest that C: N and Org-H: N ratios as well as the relative content of major nutrients in seston and seston concentrations can be importantly related to trace metal concentrations in seston samples. In discussing the results, however, we consider that metal-seston relationships depend on a variety of physical, chemical, and biological factors and/or variables other than those measured in this study, which could also contribute for defining and explaining variations in metal-seston concentrations in lake ecosystems.  相似文献

The effects of fourteen metal ions (As3+, As5+, Cd2+, Co2+, Cr3+, Cr6+, Hg2+, Li+, Mg2+, Mn2+, Ni2+, Se4+, V5+, VO2+) on the proliferation and differentiation in mouse B16 melanoma cells cultivated in vitro were analyzed. Cell number assays, melanin, and protein measurements, a 3(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide reduction test (MTT survival test), and a clonal growth assay were performed. At 10−4 M, metal ions such as As3+, As5+, Cd2+, Cr6+, Se4+, V5+, VO2+, and, to a minor extent, Li+, Hg2+, and Co2+ significantly reduced the number of the B16 melanoma cells. For the same molar concentration, the order of the levels of cell toxicity of the metal compounds to B16 cells as measured by the MTT test was as follows: Hg2+>Cr6+=Cd2+>As3+, As5+>V5+, VO2+>Se4+=Ni2+=Co2+=Li+. An increased synthesis of melanin in B16 cells was noted after incubation with Co2+, Ni2+, Cd2+, and Li+, whereas Se4+ had, on the contrary, an inhibiting effect on melanogenesis.  相似文献

Dietary chitosans with different molecular weight Mw and the degree of deacetylation DDA (high molecular weight chitosan HCS with Mw 7.60 × 105 and DDA 85.5%, middle molecular weight chitosan MCS with Mw 3.27 × 104 and DDA 85.2%, chito-oligomer COS with Mw 0.99 × 103 and DDA 85.7% and water-soluble chitosan WSC with Mw 3.91 × 104 and DDA 52.6%) were used at the 1.05% level to feed mice for 90 days. Afterwards no pathological symptoms, clinical signs or deaths were observed. The body weight of mice in chitosan group and control group showed no significant difference. Although HCS, COS and WSC had no significant effect on the level of Fe, Zn and Cu in the tested mice’s liver, spleen, heart and kidney, MCS significantly increased the level of Fe, Zn and Cu in liver. Therefore dietary ingestion of chitosan did not depress the level of Fe, Zn and Cu in mice.  相似文献

Multiple mating is common in many species, but it is unclear whether multiple paternity enhances offspring genetic diversity or fitness. We conducted a survey on wild house mice (Mus musculus musculus), and we found that in 73 pregnant females, 29% of litters had multiple sires, which is remarkably similar to the 23–26% found in feral populations of Mus musculus domesticus in the USA and Australia, respectively. The question is: How has selection maintained multiple mating in these subspecies since the evolutionary divergence, ca. 2800–6000 years ago? We found no evidence that multiple paternity enhanced females’ litter size, contrary to the fertility assurance or genetic benefits hypotheses. Multiple paternity was associated with reduced mean and variance in offspring body mass, which suggests that females allocate fewer resources or that there is increased intrauterine conflict in multiple-versus single-sired litters. We found increased allelic diversity (though not heterozygosity) in multiple-sired litters, as predicted by the genetic diversity hypothesis. Finally, we found that the dams’ heterozygosity was correlated with the mean heterozygosity of their offspring in single-and multiple-sired litters, suggesting that outbred, heterozygous females were more likely to avoid inbreeding than inbred, homozygous females. Future studies are needed to examine how increased genetic diversity of litters and smaller mean (and variance) offspring body mass associated with multiple paternity affect offspring fitness.  相似文献

In this review article we give an overview of current knowledge with respect to redox-sensitive alterations in Na+ and Ca2+ handling in the heart. In particular, we focus on redox-activated protein kinases including cAMP-dependent protein kinase A (PKA), protein kinase C (PKC), and Ca/calmodulin-dependent protein kinase II (CaMKII), as well as on redox-regulated downstream targets such as Na+ and Ca2+ transporters and channels. We highlight the pathological and physiological relevance of reactive oxygen species and some of its sources (such as NADPH oxidases, NOXes) for excitation—contraction coupling (ECC). A short outlook with respect to the clinical relevance of redox-dependent Na+ and Ca2+ imbalance will be given.  相似文献

The purpose of this study was to investigate the long-term bioaccumulation and elimination of Cd, Pb, Mn, Zn and Fe by Pinna nobilis tissues after their 90 day-transplantation period at Téboulba fishing harbor. During the transplantation period, the Cd, Pb, Mn, Zn and Fe concentrations in the different tissues of the mussels were measured before and after exposure period. Metal (Cd, Pb, Mn, Zn and Fe) accumulation in P. nobilis mussels varied depending on the analyzed tissue and the caging times. Notable differences in Cd, Pb, Mn, Zn and Fe accumulation patterns within the digestive gland, gills and muscle were found and may be due to the ability of each tissue to accumulate metals. During the depuration phase, the elimination of Cd, Pb, Mn, Zn and Fe depended on the target tissue and the metal speciation. Cd, Pb, Mn and Fe were eliminated rapidly from one organ and increased in other when compared to those of 90 day transplanted mussels. The increase of metal loads during the elimination phase is not clear and particularly what kind of processes is responsible for such response. However, it is reasonable to assume that metals increase is related to the existence of an accumulation/detoxification mechanism, which involves the transport of metals from an organ to another. The data obtained indicate that because of the significantly high quantities of Cd, Pb, Mn, Zn and Fe accumulated during the exposure phase, the transplanted mussels are suitable bioindicators for monitoring trace metals in marine ecosystem.  相似文献

Mice selected for aggression and coping (long attack latency (LAL), reactive coping strategy; short attack latency (SAL), pro-active coping strategy) are a useful model for studying the physiological background of animal personalities. These mice also show a differential stress responsiveness, especially in terms of hypothalamic-pituitary-adrenal axis reactivity, to various challenges. Since the stress response can increase the production of reactive oxygen species, we predicted that the basic oxidative status of the lines could differ. We found that LAL showed higher serum antioxidant capacity (OXY) than SAL, while no differences emerged for reactive oxygen metabolites (ROMs) or the balance between ROMs and OXY, reflecting oxidative stress. Moreover, the lines showed inverse relationships between ROMs or OXY and body mass corrected for age. The results indicate that variation in oxidative status is heritable and linked to personality. This suggests that different animal personalities may be accompanied by differences in oxidative status, which may predict differences in longevity.  相似文献

Reproductively mature male musk shrews and mice were bilaterally castrated. Epididymal sperm numbers and motility were assessed 0, 2, 4 and 6 weeks after surgery. Seminal vesicle weights and plasma concentrations of total androgens were also measured. In male musk shrews, 30% of the original epididymal sperm numbers were still present 2 weeks after castration and motile spermatozoa were present in 2 of 7 individuals. By 4 and 6 weeks after castration the numbers of spermatozoa remaining declined to about 10% and no sperm motility was noted. Seminal vesicle weights were maintained at about 30% of their original size even up to 6 weeks after castration. In male mice, epididymal sperm numbers, seminal vesicle weights, and androgen levels declined more dramatically after castration. Although androgen concentrations in gonadally intact male musk shrews were approximately 50% of the values in male mice, after castration the concentrations in musk shrews were approximately 2-fold higher than in mice at all times. The results suggest that post-castration retention of epididymal sperm and seminal vesicle weights in the male musk shrew as compared with male mice, is facilitated either by a relatively greater adrenal contribution to circulating androgen levels and/or greater target tissue sensitivity.  相似文献

镜像、影像等虚拟条件可能影响动物的觅食行为和活动时间分配。利用控制性围栏实验观察了同种个体实体、镜像及影像对ICR小鼠(Mus musculus)的觅食行为和活动时间分配的影响。发现实验鼠与实体接触时间显著高于与镜像、影像的接触时间;实体存在时,实验鼠显著增加了在低竞争风险区域贮藏食物量和巢外活动时间;影像存在时,实验鼠显著减少了巢外活动时间。结果提示ICR小鼠在实体、镜像、影像条件下表现出不同的觅食策略和活动时间分配,其觅食行为具有一定的可塑性。该结果可能为进一步研究ICR小鼠的行为及饲养管理提供一定参考。  相似文献

The aim of this study was to determine whether an oral administration of melatonin, a known antioxidant, free radical scavenger and metal chelator, influences tissue accumulation and toxicity of cadmium (Cd) in mice exposed subchronically to the metal. The animals received drinking water containing 50 μg Cd/mL only or with additional 2, 4 or 6 μg/mL melatonin for 8 weeks. Melatonin co-treatment brought about a dose-dependent decrease in the renal, hepatic and intestinal Cd concentrations, and the renal and hepatic metallothionein levels followed a pattern similar to that of the Cd accumulation. Histopathological changes occurred only in the kidneys (glomerular swelling and focal tubular degeneration) in all mice from the Cd alone group. In mice co-treated with melatonin, only slight (2 μg/mL melatonin) or no damage (4 and 6 μg/mL melatonin) was seen. The Cd and melatonin treatments did not affect renal lipid peroxidation and iron concentration. These data indicate that orally administered melatonin together with Cd reduces tissue accumulation of this metal; in particular, the reduction of renal Cd accumulation by melatonin is probably responsible for the prevention of Cd-induced injury in this organ.  相似文献

In virtually every mammalian species examined, some males exhibit reflexive testosterone release upon encountering a novel female (or female-related stimulus). At the same time, not every individual male (or every published study) provides evidence for reflexive testosterone release. Four experiments using house mice (Mus musculus) examined the hypothesis that both the male's genotype and his degree of sexual arousal (as indexed by ultrasonic mating calls) are related to such variability. In Experiment 1, CF-1 males exhibited reflexive testosterone elevations 30 min after encountering female urine. CK males, on the other hand, did not exhibit testosterone elevations 20, 30, 50, 60, or 80 min after encountering female urine (Experiments 1 and 2) suggesting this strain incapable of reflexive release. In Experiment 3, we measured both mating calls and reflexive testosterone release in response to female urine in CF-1 and CK males. Most males of both strains called vigorously to female urine but not to water. But, only CF-1 males exhibited significant testosterone elevations to female urine. In Experiment 4, DBA/2J males called vigorously to females followed by testosterone elevations 30 min later. The first 3 experiments support the hypothesis that male genotype is an important variable underlying mammalian reflexive testosterone release. Statistically significant correlations between mating calls in the first minute after stimulus exposure and testosterone elevations 30 min later (Experiments 3 and 4) support the hypothesis that, in capable males, reflexive testosterone release is related to the male's initial sexual arousal.  相似文献

Lead is a heavy metal of considerable environmental and occupational concern and there is growing evidence that it is toxic to the human immune system. In this regard, this study examined the effect of lead (Pb) exposure to peritoneal macrophages (Mvarphis) of mice (Mus musculus) cultivated in DMEM medium supplemented with fetal bovine serum, in order to investigate cell damage related to cell death. Cells were exposed to two concentrations of inorganic lead [Pb(II)] for 4, 24 and 72h. Cell viability declined during the treatment, with responses including cell death, cellular damage and DNA damage. Cell death images were found in treated cells with an increase in Bax expression, but the inorganic lead failed to induce the loss of membrane asymmetry (Annexin V conjugates), suggesting that cell death was mainly due to necrosis induction. The effects of Pb(II) on the mechanisms of cell death is not completely understood, but the immunosuppression due to DNA damage and Mvarphis death is discussed here. We have previously shown the effect of inorganic lead in mitochondria and phagocytosis in Mvarphis, suggesting here a pathway for the effect of the metal on mechanisms of cell death, also discussing its effects on the immune system.  相似文献

Barley ( Hordeum vulgare L. cv. CM 72) was grown for a 28-day period and stressed with treatments of 125 mol m−3 NaCl or KC1 with low Ca2+ (0.4 mol m−3 Ca2+) or high Ca2+ (10 mol m−3 Ca2+). Plants were harvested periodically so that relative growth rate (RGR), net assimilation rate (NAR) and leaf area ratio (LAR) could be calculated using the functional approach to plant growth analysis. Relative growth rate declined with time for all treatments, including controls. Salinity inhibited RGR relative to control values by day 10. High Ca2+ improved the growth of salt-stressed plants in both NaCl-salinity and KCl-salinity. KC1 proved more toxic than NaCl, especially for KCI-salinity plants with low Ca2+, which died by day 28. Net assimilation rate, but not LAR, was highly correlated with RGR for all treatments. This indicates that the photosynthetic-assimilatory machinery was limiting RGR and not the leaf area of the plant.  相似文献

Measurements were made of the influx of 45Ca into internodal cells of Chara corallina in solutions containing high concentrations of NaCl. Increasing salinity in the range 4–100mol m?3 NaCl resulted in a doubling of Ca2+ influx at the plasmalemma. A time-course of Ca2+ influx in 50 mol m?3 NaCl, 0.5mol m?3 CaCl2 showed that while influx at the plasmalemma increased only 1.5-fold, influx to the vacuole increased by up to 15-fold. This was interpreted as being due to inhibition of active Ca2+ efflux from the cell. The stimulation of Ca2+ influx by increasing salinity appeared to be principally a response to reduced turgor since similar stimulations were obtained when turgor was reduced by NaCl, Na2SO4 or mannitol. When cells were plasmolysed Ca2+ influx increased by 10–20-fold. The increased permeability was relatively specific for Ca2+ and was inhibitable by La3+. Survival of cells in high salt conditions was increased by 30 mmol m?3 La3+, which inhibited Ca2+ influx. Paradoxically, survival can also be extended by increasing external Ca2+ which leads to a higher influx. Therefore, it seems unlikely that the ameliorative effect of Ca2+ on the sensitivity of plants to high NaCl is mediated by Ca2+ entry across the plasmalemma. It seems more likely that the principal role of Ca2+ under these conditions is exerted externally through the control of membrane voltage and permeability.  相似文献

Twelve male 8-month-old lambs were used in a 6-month feeding experiment to determine the effects of dietary Mo, Mo + S and Zn supplements on the body retention and tissue accumulation of dietary Cu, Zn and Fe. The lambs were divided into four groups of three lambs each and each group was fed ad libitum one of four diets. A control diet was based on palm kernel cake (PKC) and grass hay. Three additional diets were the control supplemented with either Mo or Mo+S or Zn. At 3 months of the experiment, feces and urine were collected and sampled for 6 days. At the end of the experiment (6 months), blood was sampled and then the sheep were slaughtered. The liver and kidney were removed and sampled for chemical analysis. In comparison with the control, each dietary supplement decreased (P<0.05) the Cu concentration in the liver, but only the Mo+S supplement decreased it to a safe range of below 350 μg/g dry matter. This was accompanied by the body retention of dietary Cu of 24.6%, 6.7%, 2.5% and 6.5% for the control, Mo, Mo+S and Zn treatments, respectively. The blood plasma concentration of Cu was decreased (P<0.05) by the Zn supplement, but was not affected by other supplements (P>0.05). It was concluded that from the supplements tested, only Mo+S appeared to be effective in reducing the retention and liver accumulation of the dietary Cu to prevent chronic Cu toxicity in sheep fed PKC-based diets.  相似文献

Summary A study has been made of the influence of waterlogging on the distribution of trace metals between the various reservoirs in which they are held in a soil. Selective extractants have been used to remove metals held in 4 ways: soil solution and exchangeable; specifically adsorbed by inorganic sites; adsorbed or chelated by organic sites; adsorbed onto oxide surfaces.Waterlogging over a period of 16 weeks resulted in the release of both manganese and iron from the organic — and oxide — bound reservoirs to the soluble, exchangeable and inorganic reservoirs. Addition of both 1% dried grass (as an actively decomposing organic material) and 1% CaCO3 to the soil resulted in an acceleration of the metal redistribution.For manganese, selective extraction methods accounted for the distribution of all the metal in the reservoirs studied. In the case of iron however, there appeared to be some release from a reservoir not being extracted.  相似文献
