Summary Fourteen transgenic flax (Linum usitatissimum) lines, carrying a mutant Arabidopsis acetolactate synthase (ALS) gene selected for resistance to chlorsulfuron, were characterized for resistance to two sulfonylurea herbicides. Progeny of 10 of the 14 lines segregated in a ratio of 3 resistant to 1 susceptible, indicating a single insertion. Progeny of 1 line segregated in a 151 ratio, indicating two insertions of the ALS gene at independent loci. Progeny from 3 lines did not segregate in a Mendelian fashion and were likely the products of chimeric shoots. Resistance to chlorsulfuron was stably inherited in all lines. At the enzyme level, the transgenic lines were 2.5 to more than 60 times more resistant to chlorsulfuron than the parental lines. The transgenic lines were 25–260 times more resistant to chlorsulfuron than the parental lines in root growth experiments and demonstrated resistance when grown in soil treated with 20 g ha-1 chlorsulfuron. The lines demonstrated less resistance to metsulfuron methyl; in root growth experiments, the transgenic lines were only 1.6–4.8 times more resistant to metsulfuron methyl than the parental lines. Resistance was demonstrated in the field at half (2.25 g ha-1) and full (4.5 g ha-1) rates of metsulfuron methyl.  相似文献

Herbicide residues may affect seedlings during early stages of their development. We studied this possibility by the use of light and electron microscopy after incubation of germinating seeds ofPisum sativum L. andZea mays L. with different concentrations of chlorsulfuron and metsulfuron-methyl. By in vitro experiments, we have shown that both herbicides caused growth reduction of the very young roots, and severe ultrastructural alterations and injuries of the root caps of both species. Chlorsulfuron caused increase of electron-dense material in the vacuoles, cytoplasmic degeneration even in the inner secretory cell layers of the cap, and disruption of the amyloplast envelopes with release of the statolithic starch grains. In the initial cell complex of the root cap, the herbicides caused the formation of large concentric aggregates of the rough ER and wall disformations in the cells adjacent to this complex. Scanning electron microscopic observations revealed a decrease of the slime layer ensheathing the root cap and the subapical root surface.We conclude that even in early stages of seed germination, both herbicides seriously affect the gravity perception centre (consisting of the statocytes), and the secretory tissue of the root caps, thus probably disturbing the processes of gravitropism and the protective slime secretion of the roots.  相似文献

A study was made of the effect of the herbicides 2,4-D, amitrole, atrazine, chlorsulfuron, diclofop-methyl, diquat, glyphosate, paraquat and trifluralin on the nodulation of sub-clover (Trifolium subterraneum L. ‘Clare’), the growth ofR. trifolii TA1 in liquid nutrient medium and the ability of herbicide-treated inoculum to successfully nodulate sub-clover plants. As concentrations of amitrole, diclofop-methyl and glyphosate in the rooting environment increased from 0 to 20 mg ai L−1, nodulation decreased linearly. The other herbicides at these concentrations caused more severe decreases in nodulation. Growth ofR. trifolii TA1 in nutrient broth was significantly retarded by all concentrations of diquat, 2 mg ai L−1 of paraquat, 10 mg ai L−1 of glyphosate and 2 mg ai L−1 of chlorsulfuron. Other herbicides did not suppress rhizobial growth. Inoculation with TA1 that had been grown in the presence of amitrole, atrazine or glyphosate and then washed free of the herbicide decreased nodulation of sub-clover, indicating that these herbicides may physiologically influence the nodulating potential of certain strains of Rhizobium. Slight increases in root length and dry matter production were measured in roots and shoots of Pisum sativum seedlings, when germinated seeds were grown for eight days in a controlled environment chamber, on Nitch nutrient solution to which 10 mg L-1 soil humic acid (HA) had been added. A concentration of 100 mg L-1 of HA produced a small reduction in dry matter production of shoots. In the presence of herbicides, 10 mg L-1 cycluron or 100 mg L-1 prometone, root elongation and shoot growth was reduced by varying degrees with respect to the control. Nutrient solutions containing 100 mg L-1 cycluron, 10 or 100 mg L-1 alachlor, or as little as 0.1 mg L-1 of 2,4-D resulted in an almost complete suppression of the growth of pea seedlings. The addition of 10 or 100 mg L-1 of HA to nutrient solution containing the herbicide was found to be either ineffective in enhancing growth, or even further reduced growth, compared to samples grown in the presence of the herbicide alone. The results of this preliminary study suggest that the generally accepted view that humic substances exert a positive effect on plant growth may not occur when humic substances interact with herbicides.  相似文献

Coal-derived sodium humate was found to stimulate primary root growth of seedlings of cantaloupe (Cucumis melo L.) at 1000 mg L–1, lettuce (Lactuca sativa L.) and onion (Allium cepa L.) at 500 and 1000 mg L–1, as well as hypocotyl growth of cantaloupe at 1000 mg L–1. Growth enhancement was not due to release of nutrient elements by the product, and, in the case of lettuce and onion, was not due to increased availability or uptake of mineral elements. Growth stimulation of cantaloupe, however, was dependent on the provision of nutrient solution. Growth stimulation of onion roots under axenic conditions indicated an effect of the humate per se rather than a response mediated via microbial breakdown products.  相似文献

Removal of the apical 3 mm of the primary root of hydroponically-grown lettuce seedlings 3 or 5 days after sowing, prevented further elongation of the root and increased both the number and total length of lateral roots. The length of the lateral zone, i.e. the distance from the base of the parent root to the lateral nearest the tip, except on one occasion, remained the same as the control in both 3 and 5 day treatments, until the length of the decapitated root (which had ceased elongating) became limiting.Zeatin applied via the roots, at a concentration range from 3 × 10–10 M to 10–8 M reduced tap root extension growth at all concentrations. Lateral root emergence was enhanced by low zeatin concentrations and retarded by higher ones. In general, the lateral zone length was the same in cytokinin-treated plants as in untreated controls.  相似文献

When [14C]indol-3yl-acetic acid was applied to the apical bud of 5-day old dwarf pea seedlings which possessed unbranched primary roots, a small amount of 14C was transported into the root system at a velocity of 11–14 mm h-1. Most of the 14C which entered the primary root accumulated in the young lateral root primordia, including the smallest detectable (20–30 mm from the primary root tip). In older (8-d old) seedlings in which the primary root bore well-developed lateral roots, 14C also accumulated in the tertiary root primordia. Only very limited information exists on the plasticity in size and structure of fine root systems, and fine root morphology of mature trees as a function of environmental variation. Six northwest German old-growth beech forests (Fagus sylvatica L.) differing in precipitation (520 – 1030 mm year–1) and soil acidity/fertility (acidic infertile to basic fertile) were studied by soil coring for stand totals of fine root biomass (0–40 cm plus organic horizons), vertical and horizontal root distribution patterns, the fine root necromass/biomass ratio, and fine root morphology (root specific surface area, root tip frequency, and degree of mycorrhizal infection). Stand total of fine root biomass, and vertical and horizontal fine root distribution patterns were similar in beech stands on acidic infertile and basic fertile soils. In five of six stands, stand fine root biomass ranged between 320 and 470 g m–2; fine root density showed an exponential decrease with soil depth in all profiles irrespective of soil type. An exceptionally small stand fine root biomass (<150 g m–2) was found in the driest stand with 520 mm year–1 of rainfall. In all stands, fine root morphological parameters changed markedly from the topsoil to the lower profile; differences in fine root morphology among the six stands, however, were remarkably small. Two parameters, the necromass/biomass ratio and fine root tip density (tips per soil volume), however, were both much higher in acidic than basic soils. We conclude that variation in soil acidity and fertility only weakly influences fine root system size and morphology of F. sylvatica, but affects root system structure and, probably, fine root mortality. It is hypothesized that high root tip densities in acidic infertile soils compensate for low nutrient supply rates, and large necromasses are a consequence of adverse soil chemical conditions. Data from a literature survey support the view that rainfall is another major environmental factor that influences the stand fine root biomass of F. sylvatica.  相似文献

How plant seeds secure root penetration into soil to obtain good seedling establishment is one of the basic ecological problems. In this study, seminal root growth was investigated to clarify the cause of varietal difference of seedling establishment in direct seeding of rice in flooded paddy fields, with special reference to root tip rotation. In a field experiment, seedling establishment percentage had a weak correlation with seminal root elongation rate but was not correlated with apparent seedling weight in water, which has been reported to be the cause of floating seedlings resulting in poor seedling establishment. Root tip rotation was analyzed for indoor-grown seedlings using spectrum analysis: the maximum entropy method (MEM) was used. Maximum entropy method power spectrum analysis clarified that maximum MEM power density (practically corresponds to spiral angle) detected in the frequency range above 0.1 cycles mm-1 was highly and positively correlated to seedling establishment percentage in the field experiment. Maximum MEM power density in high correlation with seedling establishment was mostly found around frequencies of 0.2 cycles mm–1, which corresponded to 2.0–3.4 cycles of root tip rotation per day. From these results, root tip rotation (circumnutation) with a larger spiral angle was suggested to play an important role in the establishment of rice seedlings on flooded and very soft soil. A possible explanation for why a larger spiral angle was advantageous for seedling establishment is that if buoyancy and seedling weight are constant, a larger pushing force of the seminal root is available without causing floating of a seedling, due to the upward force being a reaction of the seminal root pushing force.  相似文献

WLR1, a biotype of Lolium rigidum Gaud. that had been treated with the sulfonylurea herbicide chlorsulfuron in 7 consecutive years, was found to be resistant to both the wheat-selective and the nonselective sulfonylurea and imidazolinone herbicides. Biotype SLR31, which became cross-resistant to chlorsulfuron following treatment with the aryloxyphenoxypropionate herbicide diclofop-methyl, was resistant to the wheat-selective, but not the nonselective, sulfonylurea and imidazolinone herbicides. The concentrations of herbicide required to reduce in vitro acetolactate synthase (ALs) activity 50% with respect to control assays minus herbicide for biotype WLR1 was greater than those for susceptible biotype VLR1 by a factor of >30, >30, 7,4, and 2 for the herbicides chlorsulfuron, sulfometuron-methyl, imazapyr, imazathapyr, and imazamethabenz, respectively. ALS activity from biotype SLR31 responded in a similar manner to that of the susceptible biotype VLR1. The resistant biotypes metabolized chlorsulfuron more rapidly than the susceptible biotype. Metabolism of 50% of [phenyl-U-14C]chlorsulfuron in the culms of two-leaf seedlings required 3.7 h in biotype SLR31, 5.1 h in biotype WLR1, and 7.1 h in biotype VLR1. In all biotypes the metabolism of chlorsulfuron in the culms was more rapid than that in the leaf lamina. Resistance to ALS inhibitors in L. rigidum may involve at least two mechanisms, increased metabolism of the herbicide and/or a herbicide-insensitive ALS.  相似文献

Summary A study was made of the effects of the herbicides 2,4-D (2,4-dichlorophenoxyacetic acid) and MCPA (4-chloro-2-methyl-phenoxyacetic acid) on ion uptake, leakage and growth of rice seedlings. Using isotopically-labelled solutions containing different concentrations of 2,4-D or MCPA, it was established that 10–4 M 2,4-D or MCPA effectively inhibited potassium ion uptake, while K-ion leakage from the roots occurred only at 10–3 M. The growth of the rice seedlings was markedly retarded even at low (10–6 M) concentrations, and the roots and shoots showed different tolerances to the herbicide. At 10–8 M herbicide, the effects were not injurious, but rather favourable. Reduction in root length by herbicides was not in accordance with dry-matter production.  相似文献

The effect of 10–6 and 10–4 M NiSO4 on cell growth, proliferation, and differentiation was studied over 48 h in seminal and lateral roots of five-day-old Triticum aestivum seedlings. 10–6 M NiSO4 did not significantly affect the root system, whereas 10–4 M NiSO4 inhibited its development. However, 10–6 M NiSO4 disturbed the contacts between the groups of closely related cells of the rhizodermis in the meristem. In the exodermis, an additional layer of cells was formed. At the nickel concentration of 10–4 M, cell divisions in the outer layers of the root cells and metaxylem ceased earlier than in other root tissues positioned both centripetally and acropetally. Differentiation of protophloem sieve elements was completed in the meristem but at a greater distance from the root tip. Cell elongation started at the same distance from the root tip as in control plants. The rate of elongation decreased, and acropetally it stopped. Therefore, the cells of the xylem and metaphloem started to differentiate, and primordia of lateral roots were initiated and formed closer to the root tip. At a lethal concentration (10–4 M), nickel induced necroses of elongating cells of the endodermis and pericycle. Nickel is supposed to enter the tissues of the central cylinder predominantly via the protoxylem and rapidly translocate along the xylem. The addition of 1-aminocyclopropane-1-carboxylic acid (ethylene precursor), or 2-chloroethylphosphonic acid (ethephon, an ethylene-releasing compound) decreased root dry weight and l-DOPA (l-3,4-dihydroxyphenylalanine) accumulation in hairy root cultures of Stizolobium hassjoo. The inhibition caused by ethephon-mediated ethylene release was alleviated by 0.5 mg CoCl2 l–1 as an inhibitor of ethylene biosynthesis. The action of ethylene was inhibited by 1.5 mg AgNO3 l–1. Ethylene thus lowers hairy root formation and l-DOPA production; CoCl2 decreases ethylene formation leading to a considerably improved root dry weight and l-DOPA production.  相似文献

Endogenous cytokinin-like activity was detected in pea seedlings usingthe soybean callus bioassay. Higher levels of activity were found in two-day-oldseedling roots and in the root free zone of four-day-old seedlings compared tothe lateral root zone of four-day-old seedlings. By day six, the levels ofendogenous cytokinin-like activity was greatly reduced in both the lateral rootzone and root free zone. Decapitation of the root tip disrupted the spatialorganization of the root. Lateral roots were subsequently found along the entirelength of the root rather than in a discreet lateral root zone. Application of10–3 M dihydrozeatin to decapitated root tipsovercame the effect of root tip removal and restored the situation to what isnormally found in intact roots. There was little mobility of dihydrozeatin inthe root, with applied 3H-DHZ not moving from the root free zone,even 24 h after application.  相似文献

Low phosphorus availability regulates root hair growth in Arabidopsis by (1) increasing root hair length, (2) increasing root hair density, (3) decreasing the distance between the root tip and the point at which root hairs begin to emerge, and (4) increasing the number of epidermal cell files that bear hairs (trichoblasts). The coordinated regulation of these traits by phosphorus availability prompted us to speculate that they are synergistic, that is, that they have greater adaptive value in combination than they do in isolation. In this study, we explored this concept using a geometric model to evaluate the effect of varying root hair length (short, medium, and long), density (0, 24, 48, 72, 96, and 120 root hairs per mm of root length), tip to first root hair distance (0.5, 1, 2, and 4 mm), and number of trichoblast files (8 vs. 12) on phosphorus acquisition efficiency (PAE) in Arabidopsis. SimRoot, a dynamic three-dimensional geometric model of root growth and architecture, was used to simulate the growth of Arabidopsis roots with contrasting root hair parameters at three values of phosphorus diffusion coefficient (D e=1×10–7, 1×10–8, and 1×10–9 cm2 s–1) over time (20, 40, and 60 h). Depzone, a program that dynamically models nutrient diffusion to roots, was employed to estimate PAE and competition among root hairs. As D e decreased from 1×10–7 to 1×10–9 cm2 s–1, roots with longer root hairs and higher root hair densities had greater PAE than those with shorter and less dense root hairs. At D e=1×10–9 cm2 s–1, the PAE of root hairs at any given density was in the order of long hairs > medium length hairs > short hairs, and the maximum PAE occurred at density = 96 hairs mm–1 for both long and medium length hairs. This was due to greater competition among root hairs when they were short and dense. Competition over time decreased differences in PAE due to density, but the effect of length was maintained, as there was less competition among long hairs than short hairs. At high D e(1×10–7 cm2 s–1), competition among root hairs was greatest among long hairs and lowest among short hairs, and competition increased with increasing root hair densities. This led to a decrease in PAE as root hair length and density increased. PAE was also affected by the tip to first root hair distance. At low D e values, decreasing tip to first root hair distance increased PAE of long hairs more than that of short hairs, whereas at high D e values, decreasing tip to first root hair distance increased PAE of root hairs at low density but decreased PAE of long hairs at very high density. Our models confirmed the benefits of increasing root hair density by increasing the number of trichoblast files rather than decreasing the trichoblast length. The combined effects of all four root hair traits on phosphorus acquisition was 371% greater than their additive effects, demonstrating substantial morphological synergy. In conclusion, our data support the hypothesis that the responses of root hairs to low phosphorus availability are synergistic, which may account for their coordinated regulation.  相似文献

The herbicides 2,4-D, amitrole, atrazine, diclofop-methyl, diquat, paraquat and trifiluralin were applied at rates of 0, 2, 5 and 10 μg ai. g−1 to a sandy loam soil and allowed to degrade for 120 days. After this period, subterranean clover seedlings were transplanted into treated soil and the effect of herbicide residues on plant growth, number of nodules formed and nitrogenase activity was investigated. At all rates of atrazine and chlorsulfuron, and at all rates of amitrole in excess of 2 mg ai g−1 of soil, sufficient herbicide remained to be lethal to the seedlings. When amitrole was applied at the rate of 2 mg ai g−1 of soil, plant growth, nodulation and nitrogenase activity of plants were reduced. Residues of diquat reduced all plant parameters studied while, residues of 2,4-D reduced plant growth and nodule formation, but plant nitrogenase activity was unaffected. Residues of trifluralin had no effect on plant growth parameters but the number of nodules formed per plant was reduced. Residues of paraquat and diclofop-methyl had no effect on any of the plant parameters studied.  相似文献

Excised root cultures of Gloriosa superba reached 7.5 g dry wt l–1 and accumulated 240±40 g colchicine g–1 cell dry wt after 4 weeks growth. While all precursors (except trans-cinnamic acid) enhanced colchicine content of root cultures without adversely affecting root growth, treatment with p-coumaric acid + tyramine (each at 20 mg l–1) increased colchicine content to 1.9 mg g–1 cell dry wt.  相似文献

Oxygen is essential for all aerobic organisms. Higher plants need oxygen to sustain metabolism and growth. After experiencing anaerobic stress for a period of time, most plant tissues will be damaged. This study examined the physiological characteristics and anatomical structures in the root tips of rice seedlings (cultivars Xiushui09 and Chunyou84) in response to different rhizosphere oxygen environments. The results showed that moderate oxygen (MO: 2.5–3.5 mg L−1) increased the dry weight accumulation and formation of rice roots (including length, surface area, number of tips) in both genotypes. Compared to under normal conditions (NC), the magnitudes of increase in these four variables were 4.67, 66.40, 35.44 and 49.50% in Xiushui09 and 12.25, 15.59, 13.57 and 13.48% in Chunyou84, respectively. Low oxygen (LO: 0–1.0 mg L−1) stress decreased the root surface area but increased root volume and average diameter. LO and high oxygen (HO: >6.8 ± 0.21 mg L−1) stress each damaged the apical cells' ultrastructure, reduced the number of organelles, and increased electrical conductivity. Meanwhile, the root activity and respiration of rice seedlings decreased; the abscisic acid content increased compared to levels under NC. Under MO treatment, the cell membrane was not damaged, the root tip organelles were rich, the soluble protein content, root activity, respiration rate and gibberellic acid content increased compared to levels under NC; the magnitudes of these increases were 24.76, 36.00, 8.00 and 283.00% in Xiushui09 and 4.78, 40.00, 15.45 and 49.35% in Chunyou84, respectively. In conclusion, MO optimised rice root morphology and enhanced root physiological activity.  相似文献

Hairy roots of Rauvolfia micrantha were induced from hypocotyl explants of 2–3 weeks old aseptic seedlings using Agrobacterium rhizogenes ATCC 15834. Hairy roots grown in half-strength Murashige & Skoog (MS) medium with 0.2 mg indole 3-butyric acid l–1 and 0.1 mg -naphthaleneacetic acid l–1 produced more ajmaline (0.01 mg g–1 dry wt) and ajmalicine (0.006 mg g–1 dry wt) than roots grown in auxin-free medium. Ajmaline (0.003 mg g–1 dry wt) and ajmalicine (0.0007 mg g–1 dry wt) were also produced in normal root cultures. This is the first report of production of ajmaline and ajmalicine in hairy root cultures of Rauvolfia micrantha.  相似文献

Root lectins are believed to participate in the recognition between Rhizobium and its leguminous host plant. Among other factors, testing this hypothesis is difficult because of the very low amounts in which root lectins are produced. A double-antibody-sandwich enzyme-linked immunoassay, was used to determine nanogram quantities of pea lectin in root slime and salt extracts of root cell-wall material when pea seedlings were 4 and 7 d old. In addition, a critical NO 3 - concentration (20 mM) which inhibited nodulation was found, and the lectin present in root slime and salt extracts of root cell walls of 4- and 7-d-old peas supplied with 20 mM NO 3 - was comparatively determined. With the enzyme-linked immunoassay, lectin quantities ranging between 20 and 100 nanograms could be determined. The assay is not affected by monomeric mannose and glucose (pealectin haptens). The slime of the 4-d-old roots contained more lectin than the slime of the 7-d-old roots. Salt-extractable, cell-wall-associated lectin accumulated in the older roots. Nitrate affected slime and cell-wall production, and the extractability of cell-wall material in both age groups. The presence of NO 3 - increased lectin in the slime, most notably in the younger roots; the relative amount of lectin in the slime was almost doubled.
