Two hundred and fifty strains, all of them representatives of native Bradyrhizobium sp., isolated from soils cultivated with soybean have been characterized by their denitrification activity. In addition, the denitrification potential of those soils was also measured by evaluating the most-probable-number (MPN) of denitrifying bacteria and the denitrification enzyme assay (DEA). Of the 250 isolates tested, 73 were scored as probable denitrifiers by a preliminary screening method. Only 41 were considered denitrifiers because they produced gas bubbles in Durham tubes, cultures reached an absorbance of more than 0.1 and NO3− and NO2− were not present. Ten of these 41 were selected to confirm denitrification and to study denitrification genes. According to N2O production and cell protein concentration with NO3−, the isolates could be differentiated in three categories of denitrifiers. The presence of the napA, nirK, norC and nosZ genes was detected by production of a diagnostic PCR product using specific primers. RFLP from the 16S-23S rDNA spacer region (IGS) revealed that denitrifiers strains could be characterized as Bradyrhizobium japonicum and strains which were non-respiratory denitrifiers as B. elkanii.  相似文献

The performance of enriched sludge augmented with the B21 strain of Alcaligenes defragrans was compared with that of enriched sludge, as well as with pure Alcaligenes defragrans B21, in the context of a sulfur-oxidizing denitrification (SOD) process. In synthetic wastewater treatment containing 100–1,000 mg NO3-N/L, the single strain-seeded system exhibited superior performance, featuring higher efficiency and a shorter startup period, provided nitrate loading rate was less than 0.2 kg NO3-N/m3 per day. At nitrate loading rate of more than 0.5 kg NO3-N/m3 per day, the bioaugmented sludge system showed higher resistance to shock loading than two other systems. However, no advantage of the bioaugmented system over the enriched sludge system without B21 strain was observed in overall efficiency of denitrification. Both the bioaugmented sludge and enriched sludge systems obtained stable denitrification performance of more than 80% at nitrate loading rate of up to 2 kg NO3-N/m3 per day.  相似文献

Taking advantage of the good biocompatibility and high efficiency of nitrogen removal with microbes, nitrifying and denitrifying bacteria, are becoming increasingly more widely used for wastewater treatment and prevention of eutrophication. In this research, an aerobic nitrifying-denitrifying bacterium was successfully screened from activated sludge and identified as Pseudomonas sp. (CCTCC No M2010209) by the 16S rDNA sequence. The activity verification confirmed its nitrifying-denitrifying capability of removing ammonium, nitrate and nitrite nitrogen. The types of carbon sources and carbon-nitrogen ratio greatly influenced the removal efficiency of NH4 +-N and NO3 -N. When the initial concentrations of NH4 +-N and NO3 -N in synthetic wastewater were less than 70 and 50 mg/L, the nitrogen removal rates reached 94 and 90% in 9 h, respectively. Preliminary comparisons of nitrogen removal capacity between this isolate and other commercial preparations in the treatment of synthetic wastewater revealed its promising potential to be used in the actual wastewater treatment.  相似文献

Synechocystis sp. PCC 6701 has a brilliantly colored pigment, phycobiliprotein containing phycoerythrin. Culture medium was optimized by sequential designs in order to maximize phycobiliprotein production. The observed fresh weights after 6 days were 0.58 g/L in BG-11, 0.83 g/L in medium for Scenedesmus sp. and 0.03∼0.52 g/L in the other tested media. Medium for Scenedesmus sp. was selected to be optimized by fractional factorial design and central composite design since the medium maintained a more stable pH within a desirable range due to higher contents of phosphate. The fractional factorial design had seven factors with two levels: KNO3, NaNO3, NaH2PO4, Na2HPO4, Ca(NO3)2, FeEDTA, and MgSO4. From the result of fractional factorial design, nitrate and phosphate were identified as significant factors. A central composite design was then applied with four variables at five levels each: nitrate, phosphate, pH, and light intensity. Parameters such as fresh weight and phycobiliprotein contents were used to determine the optimum value of the four variables. The proposed optimum media contains 0.88 g/L of nitrate, 0.32 g/L of phosphate under 25 μE·m−2·s−1 of light intensity. The maximum phycobiliprotein contents have been increased over 400%, from 4.9 to 25.9 mg/L after optimization.  相似文献

Bacterial biofilters used in marine recirculation aquaculture systems need improvements to enhance nitrogen removal efficiency. Relatively little is known about biofilter autochthonous population structure and function. The present study was aimed at isolating and characterizing an autochthonous denitrifying bacterium from a marine biofilter installed at a recirculation aquaculture system. Colonization of four different media in a marine fish farm was followed by isolation of various denitrifying strains and molecular classification of the most promising one, strain T2, as a novel member of the Pseudomonas fluorescens cluster. This strain exhibits high metabolic versatility regarding N and C source utilization and environmental conditions for growth. It removed nitrate through aerobic assimilatory metabolism at a specific rate of 116.2 mg NO3-N g dw−1 h−1. Dissimilatory NO3-N removal was observed under oxic conditions at a limited rate, where transient NO2-N formed represented 22% (0.17 mg L−1) of the maximum transient NO2-N observed under anoxic conditions. Dissimilatory NO3-N removal under anoxic conditions occurred at a specific rate of 53.5 mg NO3-N g dw−1 h−1. The isolated denitrifying strain was able to colonize different materials, such as granular activated carbon (GAC), Filtralite and Bioflow plastic rings, which allow the development of a prototype bioreactor for strain characterization under dynamic conditions and mimicking fish-farm operating conditions.  相似文献

A bacterial strain using pyridine as sole carbon, nitrogen and energy source was isolated from the activated sludge of a coking wastewater treatment plant. By means of morphologic observation, physiological characteristics study and 16S rRNA gene sequence analysis, the strain was identified as the species of Paracoccus. The strain could degrade 2,614 mg l−1 of pyridine completely within 49.5 h. Experiment designed to track the metabolic pathway showed that pyridine ring was cleaved between the C2 and N, then the mineralization of the carbonous intermediate products may comply with the early proposed pathway and the transformation of the nitrogen may proceed on a new pathway of simultaneous heterotrophic nitrification and aerobic denitrification. During the degradation, NH3-N occurred and increased along with the decrease of pyridine in the solution; but the total nitrogen decreased steadily and equaled to the quantity of NH3-N when pyridine was degraded completely. Adding glucose into the medium as the extra carbon source would expedite the biodegradation of pyridine and the transformation of the nitrogen. The fragments of nirS gene and nosZ gene were amplified which implied that the BW001 had the potential abilities to reduce NO2 to NO and/or N2O, and then to N2.  相似文献

Using vegetative propagules (ramuli) of the iota carrageenan producing red seaweed Solieria chordalis, a maximum growth rate of 6.8% d−1 was achieved when cultured at 20 °C and 100 μmol photon m−2 s−1 in seawater supplemented with 20 μM NO3-N or 10 μM NO3-N plus 10 μM NH4-N. Ramuli grew less well when nitrogen was supplied solely as NH 4 + . Maximum carrageenan content was observed at the same temperature, irradiance and salinity as growth, but at lower nitrogen concentrations. These findings are discussed in relation to results obtained from studies on other iota-carrageenan producing carrageenophytes.  相似文献

Both root architecture and plant N nutrition are altered by inoculation with the plant growth-promoting rhizobacteria (PGPR) Phyllobacterium strain STM196. It is known that NO3 and N metabolites can act as regulatory signals on root development and N transporters. In this study, we investigate the possible interrelated effects on root development and N transport. We show that the inhibition of Arabidopsis lateral root growth by high external NO3 is overridden by Phyllobacterium inoculation. However, the leaf NO3 pool remained unchanged in inoculated plants. By contrast, the Gln root pool was reduced in inoculated plants. Unexpectedly, NO3 influx and the expression levels of AtNRT1.1 and AtNRT2.1 genes coding for root NO3 transporters were also decreased after 8 days of Phyllobacterium inoculation. Although the mechanisms by which PGPR exert their positive effects remain unknown, our data show that they can optimize plant development independently from N supply, thus alleviating the regulatory mechanisms that operate in axenic conditions. In addition, we found that Phyllobacterium sp. elicited a very strong induction of AtNRT2.5 and AtNRT2.6, both genes preferentially expressed in the shoots whose functions are unknown.  相似文献

Annual and seasonal rates of net nitrogen mineralization were determined for 19 sites in the spruce-fir forests of the Southern Appalachian Mountains. These sites included high and low elevation stands of red spruce (Picea rubens Sarg.) and Fraser fir (Abies fraseri (Pursh.) Poir.) on east and west exposures on Whitetop Mountain, Virginia; Mt. Mitchell, North Carolina; and Clingman's Dome in the Great Smoky Mountains National Park. Mineralization rates were determined using in situ soil incubations in PVC tubes with ion exchange resin bags placed in the bottom of the tubes to collect leachate. Throughfall was collected in resin bags placed in the top of the tubes. Average initial NH4-N + NO3-N ranged from 0.6 to 4.8 kg N/ha across all plots, and average mineralization rates ranged from 26 to 180 kg-N ha−1 yr−1. Throughfall ranged from 18 to 32 kg-N ha−1 yr−1 with NH4-N accounting for about two-thirds of the throughfall N across all sites. Throughfall and mineralization rates were not related to elevation or exposure. Summary Laboratory scale and pilot plant reactors were inoculated with an efficient denitrifier, Brachymonas denitrificans(CCUG 45880), in order to evaluate whether a bio-augmentation approach can be used to enhance biological nitrogen removal from tannery effluents. To determine the effectiveness of the introduced strain, denitrifying activity in the activated sludge was monitored by nitrate uptake rate (NUR) measurement of NO3-N. Fluorescent in situ hybridization (FISH) technique was used to monitor the growth of the augmented species. The laboratory scale nitrate removal efficiency with the introduced B. denitrificans (3.7±0.6 mg NO3-N gVSS −1 h −1) was higher than that of the activated sludge without the addition of the bacteria (3.5±0.7 mg NO3-N gVSS −1 h −1); the NUR in the pilot plant after and before the introduction of the strain was also of the magnitude of 12.0±1.4 and 10.6±1.4 mg NO3-N gVSS −1 day −1 , respectively. In situ hybridization results revealed that the introduced denitrifying bacteria significantly facilitated the development of a dense denitrifying bacterial population in the activated sludge, which enhanced in situ denitrification activity. FISH data indicated that once introduced, B. denitrificans remained abundant throughout the experimental period. The ability to seed a bioreactor with bacterial strain capable of removing target pollutants from tannery effluents in a mixed microbial community suggests that this approach could have commercial applications.  相似文献

The aim of this research was to test whether NH4 + and NO3 affect the growth, P demand, cell composition and N2 fixation of Cylindrospermopsis raciborskii under P limitation. Experiments were carried out in P-limited (200 μg l−1 PO4-P) chemostat cultures of C. raciborskii using an inflowing medium containing either 4,000 μg l−1 NH4-N, 4,000 μg l−1 NO3-N or no combined N. The results showed the cellular N:P and C:P ratios of C. raciborskii decreased towards the Redfield ratio with increasing dilution rate (D) due to the alleviation of P limitation. The cellular C:N and carotenoids:chlorophyll-a ratios also decreased with D, predominantly as a result of an increase in the chlorophyll-a and N content. The NH4 + and NO3 supply reduced the P maintenance cell quota of C. raciborskii. Consequently, the biomass yield of the N2-grown culture was significantly lower. The maximum specific growth rate of N2-grown culture was also the lowest observed. It is suggested that these differences in growth parameters were caused by the P and energy requirement for heterocyte formation, nitrogenase synthesis and N2 fixation. N2 fixation was partially inhibited by NO3 and completely inhibited by NH4 +. It was probably repressed through the high N content of cells at high dissolved N concentrations. These results indicate that C. raciborskii is able to grow faster and maintain a higher biomass under P limitation where a sufficient supply of NH4 + or NO3 is maintained. Information gained about the species-specific nutrient and pigment stoichiometry of C. raciborskii could help to access the degree of nutrient limitation in water bodies. Nannochloropsis sp. was grown with different levels of nitrate, phosphate, salinity and temperature with CO2 at 2,800 μl l−1. Increased levels of NaNO3 and KH2PO4 raised protein and polyunsaturated fatty acids (PUFAs) contents but decreased carbohydrate, total lipid and total fatty acids (TFA) contents. Nannochloropsis sp. grew well at salinities from 22 to 49 g l−1, and lowering salinity enhanced TFA and PUFAs contents. TFA contents increased with the increasing temperature but PUFAs contents decreased. The highest eicosapentaenoic acid (EPA, 20:5ω3) content based on the dry mass was above 3% under low N (150 μM NaNO3) or high N (3000 μM NaNO3) condition. Excessive nitrate, low salinity and temperature are thus favorable factors for improving EPA yields in Nannochloropsis sp.  相似文献

Polyhydroxyalkanoates (PHAs), intracellular carbon and energy reserve compounds in many bacteria, have been used extensively in biodegradable plastics. PHA formation is influenced by nutrient limitations and growth conditions. To characterize the PHA accumulation in a new denitrifying phosphorus-removing bacterium Brachymonas sp. P12, batch experiments were conducted in which the electron acceptor (oxygen or nitrate) was varied and different concentrations of carbon (acetate), nitrogen (NH4Cl), and phosphorus (KH2PO4) were used. Polyhydroxybutyrate (PHB) was the dominant product during PHA formation when acetate was the sole carbon source. The PHB content of aerobically growing cells increased from 431 to 636 mg PHB g−1 biomass, but the PHB concentration of an anoxic culture decreased (−218 mg PHB g−1 biomass), when PHB was utilized simultaneously with acetate as an electron donor for anoxic denitrification. The specific PHB production rate of the carbon-limited batch, 158.2 mg PHB g−1 biomass h−1, was much greater than that of batches with normal or excess carbon. The effects of phosphorus and nitrogen concentrations on PHB accumulation were clearly less than the effect of carbon concentration. According to the correlation between the specific PHB production rate and the specific cell growth rate, PHB accumulation by Brachymonas sp. P12 is enhanced by nutrient limitation, is growth-associated, and provides additional energy for the biosynthesis of non-PHB cell constituents to increase the cell growth rate beyond the usual level.  相似文献

A short rod shaped, gram-negative bacterium strain Burkholderia sp. FDS-1 was isolated from the sludge of the wastewater treating system of an organophosphorus pesticides manufacturer. The isolate was capable of using fenitrothion as the sole carbon source for its growth. FDS-1 first hydrolyzed fenitrothion to 3-methyl-4-nitrophenol, which was further metabolized to nitrite and methylhydroquinone. The addition of other carbon source and omitting phosphorus source had little effect on the hydrolysis of fenitrothion. The gene encoding the organophosphorus hydrolytic enzyme was cloned and sequenced. The sequence was similar to mpd, a gene previously shown to encode a parathion-methyl-hydrolyzing enzyme in Plesiomonas sp. M6. The inoculation of strain FDS-1 (106 cells g−1) to soil treated with 100 mg fenitrothion emulsion kg−1 resulted in a higher degradation rate than in noninoculated soils regardless of the soil sterilized or nonsterilized. These results highlight the potential of this bacterium to be used in the cleanup of contaminated pesticide waste in the environment. A bacterial strain able to degrade dichloromethane (DCM) as the sole carbon source was isolated from a wastewater treatment plant receiving domestic and pharmaceutical effluent. 16S rDNA studies revealed the strain to be a Xanthobacter sp. (strain TM1). The new isolated strain when grown aerobically on DCM showed Luong type growth kinetics, with μmax of 0.094 h−1 and S m of 1,435 mg l−1. Strain TM1 was able to degrade other aromatic and aliphatic halogenated compounds, such as halobenzoates, 2-chloroethanol and dichloroethane. The gene for DCM dehalogenase, which is the key enzyme in DCM degradation, was amplified through PCR reactions. Strain TM1 contains type A DCM dehalogenase (dcmAa), while no product could be obtained for type B dehalogense (dcmAb). The sequence was compared against 12 dcmAa from other DCM degrading strains and 98% or 99% similarity was observed with all other previously isolated DCM dehalogenase genes. This is the first time a Xanthobacter sp. is reported to degrade DCM.  相似文献

The ability of an ecosystem to retain anthropogenic nitrogen (N) deposition is dependent upon plant and soil sinks for N, the strengths of which may be altered by chronic atmospheric N deposition. Sugar maple (Acer saccharum Marsh.), the dominant overstory tree in northern hardwood forests of the Lake States region, has a limited capacity to take up and assimilate NO3. However, it is uncertain whether long-term exposure to NO3 deposition might induce NO3 uptake by this ecologically important overstory tree. Here, we investigate whether 10 years of experimental NO3deposition (30 kg N ha−1 y−1) could induce NO3 uptake and assimilation in overstory sugar maple (approximately 90 years old), which would enable this species to function as a direct sink for atmospheric NO3 deposition. Kinetic parameters for NH4+ and NO3 uptake in fine roots, as well as leaf and root NO3 reductase activity, were measured under conditions of ambient and experimental NO3 deposition in four sugar maple-dominated stands spanning the geographic distribution of northern hardwood forests in the Upper Lake States. Chronic NO3 deposition did not alter the V max or K m for NO3 and NH4+ uptake nor did it influence NO3 reductase activity in leaves and fine roots. Moreover, the mean V max for NH4+ uptake (5.15 μmol 15N g−1 h−1) was eight times greater than the V max for NO3 uptake (0.63 μmol 15N g−1 h−1), indicating a much greater physiological capacity for NH4+ uptake in this species. Additionally, NO3 reductase activity was lower than most values for woody plants previously reported in the literature, further indicating a low physiological potential for NO3 assimilation in sugar maple. Our results demonstrate that chronic NO3 deposition has not induced the physiological capacity for NO3 uptake and assimilation by sugar maple, making this dominant species an unlikely direct sink for anthropogenic NO3 deposition.  相似文献

In this study, using a bioassay-guided isolation and purification procedure, we obtained 3-chloro-2,5-dihydroxybenzyl alcohol from a marine-derived Ampelomyces species that effectively inhibited larval settlement of the tubeworm Hydroides elegans and of cyprids of the barnacle Balanus amphitrite. The inhibitive effect on larval settlement was nontoxic and the EC50 of 3-chloro-2,5-dihydroxybenzyl alcohol ranged from 3.19 μg ml−1 to 3.81 μg ml−1 while the LC50 was 266.68 μg ml−1 for B. amphitrite cyprids; EC50 ranged from 0.67 μg ml−1 to 0.78 μg ml−1, and LC50 was 2.64 μg ml−1 for competent larvae of H. elegans, indicating that inhibitive effect of this compound was nontoxic. At a concentration of 50 μg per disc, this compound showed strong inhibitive effects on the growth of 13 out of 15 marine bacterial species tested in disc diffusion bioassay. Overall, the high inhibitory activities against bacteria and larval settlement as well as the non- or low-toxic nature of this compound to the barnacle and polychaete larvae suggest this compound could be a potent antifoulant and/or antibiotic.  相似文献

A Pseudomonas sp. strain, which can utilize quinoline as its sole carbon, nitrogen and energy source, was isolated from activated sludge in a coking wastewater treatment plant. Quinoline can be degraded via the 8-hydroxycoumarin pathway. We quantified the first two organic intermediates of the biodegradation, 2-hydroxyquinoline and 2,8-dihydroxyquinoline. We tracked the transformation of the nitrogen in quinoline in two media containing different C/N ratios. At least 40.4% of the nitrogen was finally transformed into ammonium when quinoline was the sole C and N source. But addition of an external carbon source like glucose promoted the transformation of N from NH3 into NO3 , NO2 , and then to N2. The product analysis and gene characteristics indicated that the isolate accomplished heterotrophic nitrification and aerobic denitrification simultaneously. The study also demonstrated that quinoline and its metabolic products can be eliminated if the C/N ratio is properly controlled in the treatment of quinoline-containing wastewater.  相似文献

A new basidiomycete, Trametes sp. 420, produced laccase at 6,810 U l−1 (268 mg, 25.4 U mg−1 protein for guaiacol) in glucose medium and 7,870 U l−1 (310 mg) in cellobiose medium with induction by 0.5 mM Cu2+ and 6 mM o-toluidine. Laccase isozyme E (LacE) was the sole laccase in the fermentation products. It was stable at pH 5–9 and below 70°C over 30 min. The K m values of LacE for four substrates (guaiacol ABTS, 2,6-dimethoxyphenol and syringaldazine) varied from 5 to 245 μM. The activity of LacE was strongly inhibited by NaN3 but not by EDTA or dimethylsulfoxide. LacE at 0.5 U l−1 could decolorize industrial dyes. The open reading frame of the lacE gene was 2,130 bp and was interrupted by 10 introns. It displayed a high homology to laccases from other fungi. Nannochloropsis sp. was grown to the exponential phase and transferred to the high CO2 (2,800 μl l−1) and irradiance (100 μmol photons m−2 s−1) condition with different levels of nitrate and phosphate for 72 h, then the photosynthetic activity and inorganic carbon acquisition of the alga were measured. The apparent photosynthetic efficiency (α) of Nannochloropsis sp. decreased with increasing NO3 concentration from 150 to 3,000 μM, and the high nitrate-grown cells showed the lowest levels of light-saturated photosynthetic rate (P m), while the low nitrate-grown cells showed the highest levels of dark respiration rate (R d). The maximal light-saturated photosynthetic rate and the minimal dark respiration rate were seen under the middle nitrate condition. When the nitrate concentration ranged from 150 to 3,000 μM, the affinity for inorganic carbons of Nannochloropsis sp. increased sharply with the increasing NO3 concentration to 300 μM and then decreased significantly. The middle phosphate-grown cells exhibited the highest light-saturated photosynthetic rate and apparent photosynthetic efficiency, however, the affinity for inorganic carbons of Nannochloropsis sp. was the maximum under the low phosphate condition. It was shown that the appropriate nitrogen and phosphorus levels were of vital importance to the photosynthesis of cells.  相似文献
