Two new C57BL/6H-2 mutants,B6.C-H- 2bm13 and B6.C-H- 2bm14 are described. They arose independently in C57BL/6 as spontaneous mutations of the gain and loss type. Complementation studies map the mutations in both bm13 and bm14 to theH-2D b gene. How ever, these two mutant strains are not identical, but occurred as independent mutations at the same locus, as shown by reciprocal graft rejection and by the inability of the (bm13 × bm114)F1 hybrid to accept C57BL/6 grafts. Serological studies by direct testing (cytotoxicity and hemagglutination) and by quantitative absorption demonstrated a decrease in the H-2Db private specificity H-2.2 in both bm13 and bm14 when compared to C57BL/6. This was confirmed by SDS-PAGE analysis using antisera detecting the H-2.2 specificity. Attempts to produce antibodies to either the gained or lost specificities of the two mutant strains failed.  相似文献

Three newH-2 b mutant strains, B6.C-H-2 bm9 , B6.C-H-2 bm10 and B6.C-H–2 bm11 , are described. The three mutant strains are of the gain and loss type as they reject skin grafts reciprocally with the parental C57BL/6Kh. The mutations, which arose independently, are all allelic at the same locus as 11 other mutant strains already described. By complementation and other studies the mutated gene has been shown to beH-2K b . The strains were typed directly and by absorption with antisera specific for H-2Kb and H-2Db private and public specificities and for Iab specificities. Each strain typed differently with these sera. The strain B6.C-H-2 bm9 was found to be serologically identical with C57BL/6. The strains B6.C-H-2 bm10 and B6.C-H-2 bm11 were found to have alterations in the private H-2Kb specificity, H-2.33, and in the public specificity, H-2.5, but to a different extent. B6.C-H- 2bm10 had a marked decrease in the amount of H-2.33 expressed on the splenic cell surface as compared to C57BL/6 and also has a marked decrease in the expression of H-2.5 on both spleen and red blood cells. In comparison, B6.C-H-2 bm11 has a decrease in the expression of H-2.33 but an increase in the expression of H-2.5 on both splenic and red blood cells. The other H-2b specificities appeared to be unaltered as compared with C57BL/6.  相似文献

C57BL/6 (H-2 b ) mice, and four mutants (B6.C-H-2 ba , B6-H-2 bg1 , B6-H-2 bg2 , B6-H-2 bh ) derived from this strain after separate mutations had occurred at the same locus within theH-2 complex, were analyzed to determine whether the mutations had led to anyH-2 (or Ia) difference which could be detected serologically. The strains were typed directly with antisera specific for H-2K and H-2D public and private specificities and for the Ia specificities; quantitative absorption studies were also performed for the relevant H-2Kb, H-2Dd and Iab specificities. In no case was any quantitative or qualitative difference detected serologically between any of the strains. In addition, by using a variety of techniques to produce and assay for antibody, we failed to produce any antisera between the parental strains and the four mutants. TheH-2 mutations therefore appear to give rise to a type of antigenic specificity which is recognized byT cells and which generateT, but notB cell responses; nor are they recognized by H-2 or Ia alloantisera. The location of the mutating locus within theH-2 complex was shown by the complementation method to be within theK orIA region and not in theIB region, since crosses of the mutant strains with B10.A(4R) or D2.GD failed to complement for a subsequent C57BL/6 skin graft.  相似文献

Lysis of ectromelia- or LCM virus-infected macrophage target cells by virus-specific cytotoxic T cells from mice immunized with the homologous virus occurred only where donors of T cells and target cells shared eitherH-2K orH-2D genes. With both viruses, use of T cell or target cell donors bearing mutations (B6.C-H-2ba, B6-H-2bh, B6-H-2bg1, and B6-H-2bg2), all of which apparently occurred in the same single genetic element in theH-2Kb region, abolished (H-2ba) or impaired (H-2bh,H-2bg1 andH-2bg2) lysis in T cell-target cell combinations that shared (apart from the mutations) all other genes in theK, I-A, orI-B regions of theH-2 complex. The data suggest that virus-induced antigenic patterns on infected B6.C-H- 2ba (mutant) cells are more different antigenically from those on C57BL/6 (wild type) cells than are those on infected cells from the other mutants -B6-H-2bh, B6-H-2bg1, and B6-H-2bg2. (B6.C-H-2ba× B6 -H-2bh)F1 mice behaved like B6-H-2bh, indicating no complementation, and confirming that theH-2K gene(s) involved in recognition of virus-infected cells by virus-specific T cells behave as a single element. These findings are discussed in relation to the nature of virus-induced antigenic patterns that are recognized by virus-specific cytotoxic T cells.  相似文献

Hz1 (H-2 bm1 ) mice, an H-2 mutant strain derived from C57BL/6(H-2 b ), were either injected with vaccinia virus or had their spleen cells sensitized in vitro with syngeneic TNP-modified cells. The cytotoxic cells generated were tested for their activity against target cells that were either infected with vaccinia virus, TNP-modified, or both vaccinia infected and TNP-modified.Hz1 anti-TNP cytotoxic cells specifically lysed syngeneic target cells that were trinitrophenylated but not infected with vaccinia virus, while anti-vaccinia cells specifically lysed vaccinia infected target cells but not TNP-cells. Hz1 (H-2K bm1 D b ) anti-TNP effector cells killed B10.A(5R)-TNP (H-2K b D d ) targets, indicating that there is cross-reactivity between TNP-H-2Kb and TNP-H-2Kbm1. On the other hand, there is no cross-reactivity between vaccinia-H-2Kb and H-2Kbm1, since Hz1 anti-vaccinia effector cells did not kill vaccinia infected B10.A(5R) targets.Since Hz1 anti-TNP effector cells lysed B10.A(5R) target cells that were first infected with vaccinia virus and then derivatized with TNP, virus does not mask cross-reactive determinants shared by TNP-H-2Kb and H-2Kbm1. Additional experiments showed that Hz1 anti-TNP effector cells lysed TNP-modified and vaccinia infected B10.A(5R) target cells irrespective of the virus concentration used for infection or the time of addition of virus. Further, there are no detectable quantitative differences between C57BL/6 and Hz1 anti-TNP effector cells in their ability to kill TNP-5R targets.The cytotoxic effect of Hz1 anti-TNP effector cells on B10.A(5R)-TNP targets could not be blocked with TNP derivatized inhibitor cells that carry theH-2D d region allele. Mutational disparities derived from alleles of theH-2K andH-2D loci vary widely in their ability to induce neonatal tolerance. The more subtle mutations, such asK bm5 andK bm8, proved to be excellent tolerogens, but theK bm3 mutant (M505) turned out to be the poorest tolerogen yet studied of all H-2 alloantigens. By challenging tolerant animals with skin grafts from related mutants, it was found that expression of tolerance was highly specific. Although a minority of tolerant animals failed to discriminate between the Kb, Kbm5 and Kbm8 antigens, they never failed to discern Kb, Kbml and Kbm3 as distinctly different alloantigens.  相似文献

The line B6.M505 is congenic with C57BL/6JY and carries a mutant form of theH-2 b haplotype designatedH-2 bd . The mutant site 505 was located by the F1 tests in theK end of theH-2 gene complex. The M505 mice are histoincompatible with the B6.C(Hz1) line (haplotypeH-2 ba ) carrying another mutation in theK end ofH-2 b . Inability of M505 to complement Hz1 in tests with B6 skin grafting is considered as an evidence that the same gene was altered by both mutations. The gained H antigens of two mutants can cross-react in vivo as revealed by accelerated rejection of Hz1 skin grafts by B6 recipients presensitized with M505 spleen cells. The lost antigenic determinants are not identical as shown by accelerated rejection of B6 skin grafts by Hz1 hosts preimmunized with M505 spleen cells. Absorptions of the antiserum ASY-015, (d×a) anti-i, anti-H-2.33 with M505 spleen cells did not clear forH-2 i ,H-2 b andH-2 ba , and absorptions with Hz1 did not clear forH-2 i ,H-2 b , andH-2 bd . These results show that changes of histocompatibility determinants may be accompanied by loss of some haptenic determinants in the Hz1 and M505 mutations.  相似文献

A.BY, B10.LPa, and B10.129(5M) mice were presensitized in vivo against B10.A(5R) cells and then restimulated in vitro by the same cells in the standard CML assay. The effector cells thus generated lysed not only B10.A(5R), but also C57BL/6 targets, indicating that, in addition to anti-H-2Dd response [measured on the B10.A(5R) targets], response to minor histocompatibility (H) antigens (measured on the C57BL/6 targets) also occurred. The latter response was directed against multiple minor H antigens in the case of the A.BY effectors, and against H-1 and H-3 antigens in the case of B10.129(5M) and B10.LPa effectors, respectively. The sensitization against minor H antigens occurred in the context of H-2Kb H-2Dd antigens, but by testing the response on C57BL/6 targets, only cells reacting with minor H antigens in the context of H-2Kb were assayed. The same effector cells were then tested against H-2b mutant strains, in which theH-2K b allele was replaced by a mutant one. All three effector types [A.BY, B10.LPa, and B10.129(5M)] behaved in a similar way: they all reacted with theH-2 bg1 mutant to the same degree as withH-2 b, they did not react at all or reacted only weakly with theH-2 bd andH-2 bh mutants, and they reacted moderately or strongly with theH-2 ba mutant. The degree of crossreactivity with the mutants reflects, with one exception, the degree of relatedness of these mutants toH-2 b, as established by other methods. The one exception is theH-2 ba mutant, which is the most unrelated toH-2 b, and yet it crossreacted strongly. Further testing, however, suggested that in this instance the crossreactivity was probably directed against H-2 antigens: the anti-H-2Dd effectors apparently crossreacted with the H-2Kba antigens. This finding is an example of cell-mediated crossreactivity between the products of two differentH-2 genes (H-2K andH-2D). It is also an example of anH-2 mutation generating an antigenic determinant known to be present in another strain.  相似文献

Structural studies of the H-2 gene products from a group of five closely related but independent C57BL/6 H-2 mutant mice were undertaken. Each of the mutants exhibits reciprocal graft rejection with the parent. The group is remarkable, however, because each member of this group can accept skin grafts from any other member. The results of biochemical analysis of the H-2 glycoproteins from two of these related mutants, bm5 and bm16, are presented in this report. Evidence is given that the H-2K molecules from these two mutants are identical to each other based on comparative tryptic peptide mapping profiles with the parent. From partial amino acid sequence analysis, K products of both mutants have at least one common difference from the parental type located at residue number 116. Definitive studies established that in both bm5 and bm16 a tryosine found in the parent molecule is substituted with a phenylalanine in the mutant. S (survivor) mutants were produced in mice for genetic analysis of host resistance to metastatic cancer. S-mutants S-27 and S-31 resist transplantation of lymphoma EL4 of parental C57BL/6J (B6) mice while they accept parental skin grafts. Mutant S-27 also resists formation of spontaneous metastases from intradermally growing EL4 tumor into lymp nodes; mutant S-31 is highly susceptible to EL4 metastases. Another mutant. H-2 bm26 (bm26), resists EL4 and rejects B6 skin grafts. Major histocompatibility complex (MHC) class I and class II gene expression was compared in these mutants and normal B6 mice. All three mutants tested, S-27, S-31, and bm26, expressed a low amount of K b mRNA in organspecific fashion. Mutant bm26 and S-31 expressed a low amount of Ab b mRNA and of Ab antigen on their spleen cells. Some oligonucleotide probes designed to hybridize to the second exon of the clss II MHC gene Ab b did not hybridize with DNA from all three mutants. These findings suggest extensive sequence alternations in the Ab b gene in mutants S-27, S-31, and bm26; they also suggest a major role of MHC in the control of host resistance to spontaneous metastases of the EL4 tumor. Biochemical analyses of the H-2K-gene products of the C57BL/6 mutant strains, bm3 and bm11, have been carried out in order to characterize the structural relationships among these antigens. From comparative tryptic peptide mapping of cyanogen bromide fragments from the mutant and parent Kb glycoproteins and from preliminary amino-acid sequence analyses, a number of discrete differences have been discerned. Two sites of difference at amino residues 77 and 89 in the Kbm3 glycoprotein are noted relative to Kb. An alteration at residue 77 similar or identical to that seen in Kbm3 is present in the K antigen of thebm11 mutant. Because our techniques sample only 75 to 80 percent of the extracellular portion of H-2Kb, other undetected changes are possible. However, our present findings are most consistent with the conclusion that only very limited differences exist between mutant and parent molecules. Further, taken together with CML (cell-mediated-lymphocytotoxicity) reaction patterns (Melief et al. 1980), the biochemical data support the hypothesis that the proposed structural alterations in theK b mutant glycoproteins are directly related to their observed immunological specificity.  相似文献

Hyperimmunization of B6.C-H-2bm?1 (H-2bm?1), a congenic mutant of C57Bl/6J (B6), with the C57Bl lymphoma EL4 resulted in the induction of antibodies with apparent EL4 specificity. EL4 reactivity was demonstrable in H-2bm?1 anti-EL4 sera by complement-mediated cytotoxicity, absorption, and enzyme-linked immunosorbent assay. By these same serological tests, H-2bm?1 anti-EL4 serum was found to be nonreactive with B6 normal lymphoid cells, embryonic fibroblasts, and two fibrosarcomas previously induced in B6 mice by methylcholanthrene. These data suggest that the serological response of H-2bm?1 to EL4 is directed against tumor-associated antigens on EL4. These findings indicate that congenic mutants which differ from the wild-type strain at MHC Class I subloci, but which do not evoke serological responses to MHC components, may provide convenient sources for preparing serological reagents directed against tumor-specific antigens.  相似文献

In the Simonsen spleen weight assay, theH-2K ba mutant does not respond against theH-2K bd mutant orH-2K bd /H-2K b hybrid, while the parentalH-2K b haplotype does respond. TheH-2K ba /H-2K b hybrid reacts strongly to bothH-2K bd andH-2K bd /H-2K b , indicating that the donor genotype could influence the reactivity against the same antigenic difference. The response of theH-2 ba mutant against a number of unrelated H-2 antigens does not differ from that of the parental haplotype. TheH-2K bd mutant reacts againstH-2K b andH-2K ba , and theH-2K b parent reacts against both theH-2K ba andH-2K bd mutants. The specific defect of reactivity in theH-2K ba mutant is effectively complemented by crossing with a number of unrelatedH-2 haplotypes. TheH-2 ka andH-2 fa mutants complement poorly compared to corresponding parental strains CBA and A.CA, while the B10.M (H-2 f ) strain does not complement at all (which is probably attributable to an undetectedH-2 mutation in the last strain). The data strongly suggest that the product of theH-2K locus-which is known to function as a transplantation antigen, lymphocyte activating determinant, and serologically defined antigen-also influences the immune response capacity against a mutant histocompatibility determinant.  相似文献

In an earlier paper, we presented evidence that two independent mutants of the bg series, B6-H-2 bm5 (bm5) and B6-H-2 bm16 (bm16) carry identical mutations such that tyrosine at residue number 116 of the H-2Kb molecule from the parent strain C57BL/6Kh is replaced by a phenylalanine in each of the two mutant molecules. In this paper, we demonstrate, using similar techniques, that the independent bg series mutants B6-H-2 bm6 (bm6), B6.C-H-2 bm7 (bm7), and B6.C-H-2 bm9 (bm9), which share biological properties with bm5 and bm16, can be grouped together because they share two identical mutations, one of which is common to bm5 and bm16, a Tyr to Phe interchange at residue number 116. In addition, a second mutation is at residue number 121, where a Cys in the H-2K molecule from 136 is substituted with an Arg in the mutant. Since all of the bg series mutants arose independently and share biological and biochemical characteristics, it is anticipated that study of these mutants could lead to some understanding of the high mutation rate in the Kb molecule.  相似文献

Cell-mediated lympholysis (CML) reactions were studied among four strains of C57BL/6 (B6) mice carrying mutant alleles (H-2 ba ,H-2 bd ,H-2 bg , andH-2 bh ) at thez1 locus in theK end ofH-2 b and the original B6 (H-2 b ) strain. Cross killing of target cells from lines that had not participated in the mixed lymphocyte reaction (MLR) was extensive, but usually less intense than that of target cells of stimulator cell genotype. The extent of CML crossreactivity could be limited by using cells from F1 hybrid mice as responders in MLR. In a comprehensive analysis of the cytotoxicity exerted by 20 MLR combinations with homozygous, and 10 MLR combinations with F1 hybrid responder cells, 19 different CML cytotoxicity patterns were identified, corresponding to at least 19 different CML target specificites. When the number of CML mismatches of each mutant with the originalH-2 b was calculated,H-2 ba was found to be most distinct fromH-2 b ,H-2 bs andH-2 bd were closest toH-2 b , andH-2 bh occupied an intermediate position. The validity of this sequence of relatedness is supported by published reports on skin graft survival times and on the interaction of T lymphocytes with virus-infected target cells using cells fromz1 locus mutants.  相似文献

TheH-2 restriction pattern of cytolytic T lymphocytes (Tc) and T lymphocytes which mediate a delayed-type hypersensitivity response (Td) directed against infectious Sendai virus was investigated usingH-2 mutant mice. Peritoneal (PM) and bone marrow-derived (BMM) macrophages and lung fibroblasts (LF) from inbred, intra-H-2 recombinant, H-2 mutant, and hybrid mice were infected with murine cytomegalovirus (MCMV) under centrifugal enhancement. At the concentration of virus employed, peritoneal macrophages from strains carrying Kd, Kb, Dd, KS and/or Ds, K4 and/or D4 alleles could be infected to a level of 80%–100%, as assessed by viral antigen expression or loss of Fc receptors. Cells lacking these haplotypes and carrying Kk, Kf, Dk, Df, or Db were resistant, yielding levels of infection below 20% . The background (non-H-2) and class II genotype and the S allele did not influence the proportions of cells infected. Furthermore, sensitivity was dominant in the F, progeny of H-2 b x H-2 k and H-2d x H-2 k crosses, and was not compromised by thebm1, bm3, bm10, or bm14 mutations in the al or2 regions of Kb orD b. The proportions of cells able to release infectious virus were low, but paralleled the frequencies of viral antigen expression. The class I genotype also determined susceptibility to MCMV infection in BMM and LF, although up to 35% of H-2 k BMM and 46% of H-2 k LF could be infected. The findings are consistent with an association between K and D antigens and a cellular receptor for MCMV on all three cell types.  相似文献

To study the interactions between T cells and class I MHC products, we developed in vitro a T-cell line reactive to H-2Kb stimulating cells and derived T-cell clones from it. Although the T-cell line could proliferate in the absence of exogeneous T-cell growth factors when stimulated with H-2Kb spleen cells, each of the derived T-cell clones required both H-2Kb stimulating cells and an external source of T-cell growth factor for its propagation. Each of the T-cell clones was also cytolysic for H-2Kb target cells. Such T-cell clones allowed the comparison of the antigenic requirements for proliferation and cytolysis. By using H-2K b mutant mice, we found that while the original anti-H-2Kb T-cell line reacted with each of the six mutants tested, the individual T-cell clones could be distinguished in terms of their reactivity pattern. Similar fine specificity patterns were found when H-2K b mutant cells were used as stimulating or target cells for any given T-cell clone. Each of the three monoclonal H-2Kb-specific antibodies reacting with different epitopes of the H-2Kb molecule totally inhibited H-2Kb-induced proliferation and lysis by the T-cell clones. Further blocking studies involved use of Fab antibody fragments and definition of their reactivity on cells from the H-2K b mutants. In preparation for functional analyses, a study of the binding of H-2Kb-specific monoclonal antibodies (mAb) to formaldehyde (FOR)-fixed H-2b spleen or tumor cells revealed that three of nine mAb tested had lost reactivity with the FOR-fixed cells, whereas the reactivity of the other mAb generally did not diminish. Comparison of the reactivity of these mAb on untreated H-2K bm mutant cells and on FOR-treated H-2Kb cells suggests that for three mAb the total loss of reactivity on the latter could be a consequence of the alteration by FOR of lysine 89, which is substituted by alanine in mutant bm3. H-2KPb-specific alloreactive polyclonal or monoclonal CTL, all of which had retained reactivity with bm3 target cells, had also retained reactivity with FOR-fixed H-2b cells as indicated by cold target inhibition studies. The H-2Kb-specific CTL were probably reactive with conformational determinants of H-2Kb, which are dependent on the integrity of both the 1 and the 2 domains of the H-2Kb molecule. Immune response (Ir) genes mapping in theI region of the mouseH-2 complex appear to regulate specifically the presentation of a number of antigens by macrophages to proliferating T cells. We have investigated the possibility that similarIr genes mapping in theH-2K andH-2D regions specifically regulate the presentation of target antigens to cytotoxic effector T cells. We report that the susceptibility of targets expressing specific non-H-2 H alloantigens to lysis by H-2-compatible, H-antigen-specific cytotoxic effector T cells is controlled by polymorphicH-2K/D genes. This control of susceptibility to lysis is accomplished through what we have defined operationally as antigen-specific regulation of non-H-2 H antigen immunogenicity. High immunogenicity of the H-4.2 alloantigen is determined by a gene mapping in theH-2K region ofH-2 b . However, high immunogenicity of H-7.1 is determined by a gene mapping in theH-2D region ofH-2 b . High immunogenicity of the H-3.1 alloantigen is determined by genes mapping in both theH-2K andH-2D regions ofH-2 b . Therefore, genes mapping in theH-2K andH-2D regions serve a function in presenting antigen to cytotoxic effector T cells. This function is analogous to that played byI-regionIr genes expressed in macrophages which present antigen to proliferating T cells. We present arguments for classification of theseH-2K/D genes as a second system ofIr genes and discuss the implications of twoH-2-linkedIr-gene systems, their possible functions, and their evolution.  相似文献
