农杆菌介导寒地水稻遗传转化的研究   总被引:2,自引:0,他引:2  
通过农杆菌介导法,将拟南芥转录激活因子ICE1基因导入寒地品种垦鉴稻10号成熟胚愈伤组织中,将经潮霉素筛选得到的抗性愈伤进行分化,获得了一些再生抗性植株,部分植株经PCR检测为阳性,证明外源目的基因已整合到水稻的染色体上,从而建立了高效水稻遗传转化体系。实验表明:YEB和MS按适当比例混合作为悬浮培养基转化率较高;干燥除菌效果和转化率均优于羧苄青霉素、头孢霉素液体除菌;潮霉素采用低压-高压-低压方式筛选最好;AS和葡萄糖均能提高转化率。  相似文献

根癌农杆菌与来自水稻成熟种子盾片的愈伤组织共培养,将GUS基因导入水稻愈伤组织,并获得了转基因植株。通过比较影响根癌农杆菌转化频率的各种因素,表明激素配比为2,4-D1mg/L、TDZ0.5mg/L、NAA1mg/L时,可以大大促进籼稻愈伤组织的分化能力;酚类化合物的加入使农杆菌的转化频率提高8.9%-23.5%;共培养时农杆菌的稀释方式及适当调整潮霉素(hygB)的使用浓度影响到农杆菌的转化频率。  相似文献

以3个水稻品种的成熟胚诱导的良好胚性愈伤组织为受体,以LycB为目的基因,应用根癌农杆菌介导法对水稻进行遗传转化,同时以抗性愈伤率为依据,对影响转化的几个因素进行优化研究。结果表明:预培养4d、侵染5~10min、农杆菌菌液浓度OD600值0.7~1.0、共培养2d有利于提高转化率。经潮霉素筛选获得的抗性植株经PCR和PCR-Southern分析鉴定,初步证明外源基因LycB已整合到水稻的基因组中。  相似文献

Several maize inbreds were transformed with Agrobacterium tumefaciens EHA101 (pGIH). Transgenic maize plants were obtained. Frequency of transformation of maize inbred Suyu No. 1 can reach 8.1%. Results of PCR and Southern blot analysis proved that T-DNA was stably integrated into the genome of maize. Staining with X-gluc confirmed the expression of GUS gene in maize cells. The band amplified by inverse PCR showed that the copy number of transgene in three transformants was single. After long term of subculture, some hygromycin resistant calli lost their regeneration ability. Although Southern blot probed the integration of gusA gene in their genome, GUS activity cannot be detected in those calli. Southern blot analysis of HpaII digest DNA showed that transgenic gusA gene was highly methylated.  相似文献

金发草(Pogonatherum paniceum)是一种多年生岩生草本植物,在生态恢复和景观建设中起着重要的作用。利用根癌农杆菌介导转化金发草胚性愈伤组织,通过GUS(β-葡萄糖苷酸酶)瞬时表达率研究菌液浓度、浸染时间、乙酰丁香酮(AS)浓度、葡萄糖浓度、共培养时间等因素对金发草转化的影响,并利用确定的最佳条件将GUS基因转入金发草,获得稳定表达转化植株。结果表明:菌液浓度(OD600)为0.6,浸染时间为10min,添加20mg/L的乙酰丁香酮(AS)和10g/L的葡萄糖,共培养时间5d为最佳条件,GUS瞬时表达率最高。经过抗性筛选后最终获得阳性转基因植株频率为57%。再生植株经GUS染色和PCR检测证明,GUS基因已成功整合到金发草基因组中。此转化体系的建立为金发草的遗传改良及相关功能基因的研究奠定了基础。  相似文献

通过农杆菌介导法 ,将Bt毒蛋白基因的一种—cryIAb基因导入 2个云南水稻栽培品种(滇系 4号 ,合系 39号 )的愈伤组织中 ,经过潮霉素筛选后 ,获得了一批抗性苗 ,部分苗经PCR检测为阳性 ,GUS组织化学染色分析发现Bt基因已整合到水稻基因组 ,并由此建立了一套有效的水稻遗传转化体系。实验中发现培养器皿的透气性对水稻愈伤组织的形成、生长具有较大的影响 ,提高培养基的渗透势能够提高愈伤组织的分化率。选择合适的洗菌液渗透势能够大大降低洗菌对愈伤组织所造成的损伤 ,从而提高转化率。  相似文献

【背景】海岛棉相对陆地棉更易感枯萎病,一旦发生很难根治,使得枯萎病逐渐成为威胁新疆海岛棉产业发展的主要病害,但其致病机理目前还不是十分明确。【目的】揭示棉花枯萎病菌的遗传变异和致病机理,同时获得带有绿色荧光蛋白(Green Fluorescent Protein,GFP)标记的棉花枯萎病菌转化子用于观察其侵染海岛棉的途径。【方法】采用农杆菌介导的遗传转化(Agrobacterium tumefaciens-Mediated Transformation,ATMT)方法,对棉花枯萎病菌7号生理小种st89进行了遗传转化并对转化条件进行优化。【结果】农杆菌介导的遗传转化法转化棉花枯萎病菌的最佳条件为:150 mg/L的潮霉素浓度能完全抑制棉花枯萎病菌的生长,浓度为200 mg/L的头孢噻肟钠能完全抑制农杆菌LBA4404生长,农杆菌起始浓度OD600为0.2,农杆菌预培养时间为8 h,棉花枯萎病菌分生孢子浓度为105个/mL,枯萎病菌孢子悬液和农杆菌LBA4404比例为1:1,乙酰丁香酮浓度为200μmol/mL,共培养时间为4 d,转化后培养温...  相似文献

【目的】建立分离自西藏林芝的能够产生一系列具有抗肿瘤活性的二酮哌嗪类化合物(epipolythiodioxopiperazine,ETP)的一株冬虫夏草定殖真菌——粘帚菌(Gliocladium sp.)6.102的遗传转化体系。【方法】利用农杆菌介导的方式建立了粘帚菌的遗传转化体系;用正交试验研究了影响转化效率的因素,包括共培养所用农杆菌与真菌孢子的比例、共培养的时间、pH值和乙酰丁香酮的浓度。【结果】成功建立了农杆菌介导的粘帚菌的遗传转化体系,得到了转化的最佳条件,其转化效率为50-100个转化子/106真菌孢子。通过农杆菌介导的方式分别将潮霉素B磷酸转移酶基因(hph)和绿色荧光蛋白基因(egfp)转入粘帚菌中,实现了表达并且可以稳定存在。【结论】首次建立了农杆菌介导的粘帚菌遗传转化体系,为研究ETP化合物的生物合成及其调控机制奠定了基础。  相似文献

利用农杆菌介导法将白细胞介素-2基因(il-2)导入番茄中,对影响其转化的因素进行了分析。结果表明:农杆菌菌种(EHA105和C58C1)、外植体类型(子叶和下胚轴)、带有不同筛选标记(Kanr、PPTr、Hygr)的载体质粒几个因素对芽诱导分化及转化均有影响。实验共接种转化2018个子叶和下胚轴外植体,获得了47株抗性再生株,对其进行il-2的PCR扩增检测,有44株呈阳性。PCR-Southern杂交证实PCR结果可靠,显示il-2基因已导入到番茄中。  相似文献

转基因育种是快速定向改良兰花育种目标性状的有效方法,但迄今未见有关墨兰转基因育种的研究报道。试验以‘企剑白墨’墨兰Cymbidium sinensis cv.‘Qijianbaimo’的根状茎为受体材料,研究了影响农杆菌介导墨兰遗传转化效率的因素,以建立有实用价值的墨兰遗传转化技术体系。结果表明,受体的预培养时间、乙酰丁香酮的添加方式及浓度、农杆菌工程菌液浓度(OD600)、侵染时间和共培养时间均对‘企剑白墨’根状茎的GUS瞬时表达率有显著影响。以预培养39 d的根状茎尖为材料,在添加200μmol/L乙酰丁香酮,OD600为0.9的工程菌液中侵染35 min后,转入添加200μmol/L乙酰丁香酮的共培养基中培养7 d时,‘企剑白墨’根状茎的GUS瞬时表达率最高,为11.67%。采用上述条件对‘企剑白墨’墨兰进行遗传转化,经PCR鉴定和GUS染色检测,从400株再生植株中获得了3株转基因植株,转化率为0.75%。研究表明通过农杆菌介导法对墨兰进行遗传改良是可行的。  相似文献

Cereal crops have been the primary targets for improvement by genetic transformation because of their worldwide importance for human consumption. For a long time, many of these important cereals were difficult to genetically engineer, mainly as a result of their inherent limitations associated with the resistance to Agrobacterium infection and their recalcitrance to in vitro regeneration. The delivery of foreign genes to rice plants via Agrobacterium tumefaciens has now become a routine technique. However, there are still serious handicaps with Agrobacterium -mediated transformation of other major cereals. In this paper, we review the pioneering efforts, existing problems and future prospects of Agrobacterium -mediated genetic transformation of major cereal crops, such as rice, maize, wheat, barley, sorghum and sugarcane.  相似文献

通过检测愈伤组织中GUS基因的瞬间表达,研究农杆菌LBA4404/pCAMBIA1301介导多年生黑麦草的转化体系。通过对多年生黑麦草瞬间表达率的比较,确立了其遗传转化的最佳优化条件。研究发现,多年生黑麦草不同品种的转化率在25%~45%之间变化。多年生黑麦草遗传转化最佳优化条件是预培养10d的胚性愈伤组织、浓度为0.5~0.8OD的农杆菌菌液以及2d共培养时间。在共培养基中添加100μmol/L乙酰丁香酮能有效地提高植物瞬间表达率。两种侵染处理方法比较结果为滤纸滴加法比浸泡法更优。转化后对愈伤组织的干燥处理能抑制农杆菌过度繁殖,能改善愈伤状态,有利于提高转化率。  相似文献

Difficulties frequently encountered using direct DNA transfer methods for transformation of Javanica varieties of rice (Oryza sativa L.) have limited the application of biotechnology to these varieties. We now reportAgrobacterium-mediated transformation of Javanica cultivars Gulfmont and Jefferson that are, respectively, widely used or about to enter commercial cultivation in the southern USA. Vigorous, phenotypically normal, fertile plants expressing both the selectable marker and the gene of interest were obtained. Southern analysis showed that only one or two copies of the T-DNA insert were present. Sequence analysis of right border fragments of one line confirmed that insertion was into a coding region of rice nuclear DNA. This analysis also revealed the presence of relatively short regions of permuted T-DNA border sequences, similar to those found afterAgrobacterium-mediated transformation of dicots. Progeny analysis of lines bearing two copies showed co-segregation, indicating that they were located relatively closely on the same chromosome. The introduced genes were transmitted to the R1 and R2 generations in a Mendelian fashion, confirming the suitability of this approach for biotechnological improvement of elite rice cultivars.  相似文献

Although several reports on rice transformation have been published, producing transgenic plants ofIndica rice varieties is still problematic. We report an improved protocol for transformingIndica rice genotypes. An important agronomic MexicanIndica rice variety, Morelos A-92, was used. Calli derived from scutellum seeds were produced by using auxins and bombarded with 2 vectors, one harboring the reporteruidA gene and the other with thehptII gene conferring hygromycin resistance. The influence of the molar relation of these vectors (uidA-hptIII) in generating callus and plants expressing theuidA reporter gene was analyzed. Selection of bombarded calli was performed under 2 conditions: 50 mg/L of hygromycin with 3 subcultures and 80 mg/L of hygromycin with no subcultures. The best conditions were a 20∶1uidA-hptII molar vector relationship and selection at 80 mg/L of hygromycin, producing 14% of calli expressing GUS. The minimal callus size in regenerating plants was 3 mm. Transformed rice plants were generated with 4.6% efficiency, considering the initial number of bombarded calli. Heredity of theuidA gene behaved as a single locus in transformed rice plants. Homozygous plants were identified in the T1 generation by means of pollen staining.  相似文献

Experiments were conducted to produce transgenic barley plants following infection of immature embryos with Agrobacterium tumefaciens. Transformed callus was obtained using hygromycin resistance as a selectable marker and either green fluorescent protein (GFP) or -glucuronidase (GUS) as a reporter. Significantly reduced plant transformation frequencies were obtained with the GFP gene compared to GUS. However, GFP proved to be an excellent reporter of early transformation events and was used to compare four barley cultivars for efficiency in two phases of transformation: the generation of stably transformed barley callus and the regeneration of plantlets from transformed callus. Transformed callus was generated at a high frequency (47–76%) in all four cultivars. Regeneration of transformed plantlets was also achieved for all four cultivars although the frequency was much higher for Golden Promise than for the other three genotypes, reiterating that genotype is an important determinant in the regenerative ability of barley. This study has demonstrated for the first time that Agrobacterium-mediated transformation can be used to transform the Australian cultivars Sloop and Chebec.Communicated by W. Harwood  相似文献

为了培育出无筛选标记基因的转基因水稻,试验将loxp-hpt-loxp基因与成基因连锁在-起转化水稻方法,得到loxp-hpt—loxp—Bt转基因水稻植株,再与同质的带有ere基因的水稻杂交,以定向删除潮霉素抗性筛选标记。试验表明以水稻品种“皖粳97”为供试材料,将成熟胚来源的愈伤组织用根癌农杆菌EHA105/pCAMBIA1305.1感染后,筛选出抗性愈伤组织并获得再生植株。经PCR验证,得到20棵转基因水稻植株。  相似文献
