An account is given of experiments on the effect of the humidityof an ambient air-stream on spore discharge in Sordaria fimicola.Change from saturated to dry (35 per cent. R. Rooted one-year shoots were grown for one season by sprayingtheir roots with nutrient solution. Iron supplied as Fe-EDTAat four concentrations resulted in plants which were respectively(a) severely chlorotic, (b) mildly chlorotic, (c) dark greenand healthy (controls), and (d) dark green but with slight reductionin growth. Severely deficient plants showed 40–70 per cent reductionsin growth as measured by fresh weight, shoot length, diameterincrease, leaf area, net assimilation and relative growth-rates.Dry weights were reduced 70–80 per cent and of the totaldry-weight increment a greater proportion remained in the leaves,which had a lower dry weight and higher water content per unitarea. However, because the initial old stem formed a greaterproportion of the total dry weight, the leaf area ratio remainedabout 11 per cent lower than in the controls. Severely deficientplants had, per unit of chlorophyll, a higher dry-weight increaseand net assimilation rate than the controls. Mild deficiency caused 10–20 per cent reductions in growthand net assimilation rate; the leaf area ratio was normal. Possible mechanisms of the effects of low iron supply are discussed,while the small growth reduction at the highest Fe-EDTA concentrationis attributed to chelate toxicity  相似文献

Simultaneous observations on extension growth and respirationrate (oxygen consumption) of 2-mm. sections excised from theextension zone of roots of pea (Pisum sativum) growing in distilledwater and 0·5 per cent. sucrose have yielded resultsclosely similar to those of Brown and Sutcliffe (1950). Respirationrate is not obviously correlated with growth rate either inwater or in sucrose, but it is strongly correlated with sectionlength. Respiration rate per unit section length (¬per unitfresh weight) shows a marked downward drift during extensionand is affected little by growth conditions. Tentative suggestionsare advanced to account for the small differences between driftsin o·5 per cent. sucrose and those in distilled water. Medium agitation produces an immediate and sustained stimulationof growth but no stimulation of oxygen uptake until the latergrowth stages. Thus respiration per unit section length is unaffectedby agitation at any stage. A typical growth response to ß-indolylacetic acid(IAA) was obtained, with a maximum stimulation (of about 35per cent.) at 1 part in 1011 and inhibitions increasing progressivelywith concentration beyond the threshold of about i part in 109.Both percentage stimulation and percentage inhibition of growthwere independent of the presence of sucrose. Respiratory responses to ß-indolylacetic acid werecomplex. In water no immediate response could be detected witheither a growth-stimulatory (10–11) or a growth-inhibitory(10–-8) concentration, while in 0·5 per cent. sucrosethe inhibitory concentration prevented the small immediate respiratoryrise due to the sucrose, probably by impeding sugar entry. Duringthe subsequent period of rapid growth (up to 36 hours) the smallrespiratory responses observed closely followed the small growthresponses to both concentrations of IAA, suggesting that theformer are the direct result of the differences in section lengthinduced by the auxin. When growth ceases (at 48 hours) sectionswhich have grown considerably in sucrose show respiratory ratesstill closely correlated with section length, whereas in waterboth concentrations of auxin induce marked depressions in respirationrate. It is concluded that ß-indolylacetic acid in bothgrowth-stimulatory and growth- inhibitory concentrations hasno direct effect on the activity of the respiratory enzyme systemof growing root cells. The small respiratory responses are bestexplained as resulting from differential changes in sectionsize and correlated changes in the enzyme complements of thegrowing cell.  相似文献

Further Observations on Light and Spore Discharge in Certain Pyrenomycetes   总被引:1,自引:0,他引:1  
A ‘spore-clock’ for studying the hourly rate ofspore discharge over a 24-hour period is described. A numberof the experiments reported in this paper have involved theuse of this apparatus. In Sordaria fimicola there is a distinct positive light-dischargereaction in a dark-conditioned culture, the rate of spore dischargeincreasing steeply to a peak 2–3 hours after brief stimulationby bright light. Although darkening a light-conditioned cultureleads to an immediate decrease in the rate of discharge, thereis no evidence of a delayed negative dark-discharge reaction. In S. verruculosa with a 12-hours light: 12-hours dark dailyreëgime, more spores are discharged in the dark than inthe light periods if the intensity of illumination is low. Withhigher light intensity there is no significant difference betweenthe number of spores discharged in light and dark periods. Asin S. fimicola there is a positive light-discharge reaction,the interval between stimulus and maximum response being muchlonger (8–12 hours). When a dark-conditioned culture istransferred to light for 48 hours and then returned to darknessfor a further 48 hours it is apparent that not only is therea positive light-discharge reaction but also a negative dark-dischargeresponse. The ‘plateau’ level of discharge is essentiallythe same in light and darkness. It is confirmed that in Hypoxylon fuscum light inhibits discharge.  相似文献

Following entrainment of nine selected species of basidiomyceteswith alternating 12 h light and 12 h dark periods, circadianrhythms with nocturnal peaks of spore discharge were found topersist in continuous light or dark for periods of 4 –9d. However, when light and dark periods were reversed followingentrainment, Piptoporus belulinus. The growth habit and certain developmental aspects of Platyceriumcoronarium (stag's horn fern) are described. Starting from theprimordial stage, the nest and pendulous leaves develop to maturityin 90 and 80 days respectively. The fertile lobe, which is partof the pendulous leaf, reaches its maximum size in 40 days.The morphogenesis of the nest leaf is more variable, and itmatures and deteriorates earlier than the pendulous leaf. Theacrostichoid sorus formation is completed in 3 weeks from inceptionand spore dispersal takes place when the fertile lobe is about100 days old. The area of the fertile lobe and number of sporesproduced were determined. On Knop's agar medium the gametophytesdevelop in 2 months and 85 per cent of them are unisexual (bothmale and female) and 15 per cent bisexual. Less than 1 per centof the gametophytes give rise to sporophytes. The juvenile leavesare simple, displaying open dichotomous venation; the firstnest and pendulous leaves are produced 24 months after the dateof spore germination. Five to 7 per cent of plants of Trifolium repens L. and T.pratenseL. and 100 per cent of plants of T. subterraneum L. were nodulatedby Rhizobium leguminosarum but none of T. hybridum L., T. glomeratumL. or T parvifirum Ehrh. The frequency of nodulation of T. pratenseby R. leguminosarum was much increased by breeding from susceptibleplants. Such plants were not nodulated by bacteria isolatedfrom any other cross-inoculation group, but remained fully susceptibleto R. trifolii. The nodules formed by R. leguminosarum are generallyassociated with lateral roots and are ineffective.  相似文献

Data are presented which show, when stomatal control is eliminated,that wheat leaves may lose 5–6 per cent. and Pelargoniumleaves 10–12 per cent. of their water without any reductionin the transpiration rate. Experiments in which Pelargonium and wheat leaves, with stomatalcontrol present, were submitted to cycles of changing watercontent also failed to establish any direct relation betweentranspiration rate and leaf water content. It is concluded that leaf water content over the range of 70–100per cent. of that present in the turgid state has no significanteffect in determining the rate of water loss from leaves. A repetition of Knight's experiment showed that stomata openedin still air and closed in moving air. This was not recordedby Knight, who used a porometer cup permanently attached tothe leaf. It is concluded that the higher transpiration raterecorded by Knight after a period of still air was due to widerstomatal aperture and not to the higher leaf water content assuggested by him.  相似文献

Mitochondria prepared from young Arum spadix have enoughcytochrome c oxidase to account for the rate at which succinateis oxidized, but succinoxidase activity increases markedly asthe plants mature so that in old material cytochrome oxidaseactivity is only 10 per cent, of succinoxidase. 2. Disintegration of the mitochondria by vibration with ballotini,treatment with digitonin or incubation in the warm reveals anintra-mitochondrial cytochrome c oxidase probably active enoughto account for the fastest rates of succinate oxidation. 3. Succinic dehydrogenase activity is demonstrated and experimentswith p-chloromercuribenzoate indicate that it plays a part inthe oxidation of succinate. 4. Cyanide completely inhibits both external and internal cytochromeoxidase but even at the earliest stages it only reduces succinoxidaseby about 50 per cent. The Production and Distribution of Dry Matter in Maize after Flowering   总被引:3,自引:0,他引:3  
An experiment in which different groups of leaf laminae wereremoved, or ears shaded, shortly after silking showed that mostof the dry-matter increase after flowering was produced by upperleaves. The top five, the middle four, and the bottom six laminaeaccounted, respectively, for 26 per cent, 42 per cent, and 32per cent of the leaf area duration (D) of the laminae afterflowering; the estimated contributions of the three groups todry-matter production by the laminae after flowering were about40 per cent, 35–50 per cent, and 5–25 per cent,respectively. The sheaths provided about one-fifth of the totalleaf area and probably contributed about one-fifth, and laminaefour-fifths, of the total dry matter produced after flowering.The contribution from photosynthesis by the ear was negligible,presumably because its surface area was only 2 per cent of thatof the leaves. Leaf efficiency (dry matter produced per unitarea) decreased greatly from the top to the base of the shoot.When laminae were removed, the grain received a larger fractionof the dry matter accumulated after flowering, less dry matterremained in the stem, and the photosynthetic efficiency of theremaining leaves was apparently increased. When alternate laminae were removed at the time of silking (half-defoliation)D was decreased by 40 per cent, and the subsequent productionof dry matter decreased nearly proportionately, so that netassimilation rate (E) was not affected but grain dry weightwas decreased by only 32 per cent. At the final harvest, thegrain of half-defoliated plants constituted 80 per cent of thedry matter accumulated after flowering, compared with 65 percent for intact plants. Stem weight decreased from two weeksafter flowering in half-defoliated plants, but remained nearlyconstant in intact plants. When pollination was prevented andno grains formed, E during the first month after flowering wasunaffected; the dry matter that would have passed into the grainaccumulated in the stem and husks, not in the leaves. The decrease in stem weight caused by defoliation suggests thatpreviously stored dry matter was moved to the grain. That suchmovement is possible was shown by keeping prematurely harvestedshoots in the dark for two weeks with their cut ends in water;the dry weight of the grain increased and that of the stem,laminae, husks, and core decreased. Nevertheless, dry-matterproduction after flowering was more than sufficient for graingrowth, and previous photosynthesis probably contributed littleto the grain.  相似文献

Previous work on chlorophyll-deficiency mutations in pea andbarley has shown that a significant increase in mutations isinduced by storing seeds under various conditions which leadto losses of viability to about 50 per cent. The work here showsthat a detectable increase in mutation frequency is also associatedwith much smaller losses of viability. Pea seeds were storedat 35 °C and 16.5 per cent moisture content for 40 and 57d when viability fell from 99 to 93 and 82 per cent, respectively.At the same time mutation frequency (percentage of seeds containingrecessive point mutations) increased from 1.62 per cent in thecontrol treatment to about 3 to 4 per cent. Barley seeds at15.5 per cent moisture content were stored at 50 °C for42 and 54 h, and at 35 °C for 28 and 39 d. During theseageing treatments viability fell from 98 to 75, 26, 93 and 48per cent respectively and the mutation frequency increased fromzero to between about 0.3 to 0.9 per cent. In both species theinduction of mutation by ageing treatments was significant butthe differences between the various ageing treatments were not.It is concluded that there is probably no safe threshold lossof viability which completely avoids mutation, and these resultssupport the view that for genetic conservation seeds shouldbe stored under conditions which minimise loss of viability. Experiments were carried out on the effect of rate of air-flowand of humidity on the growth of Avena seedlings. The experimentalmethods used are described in some detail. The effect of air-flow on mesocoryl growth was considerablewhen unsaturated air was used, but with saturated air growthwas independent of air-flow. The effect must therefore be dueto variation in transpiration rate. A 10 per cent, variationin relative humidity, at a constant rate of flow, caused a 30per cent, variation in mesocotyl length. The coleoptiles showedless pronounced effects, but even in saturated air variabilitywas not eliminated. Under the conditions used a difference of 2·3 mm. inmesocotyl length (per 50 plants), i.e. 8 per cent., was significantat P = 0·05; for the coleoptiles a difference of 1 mm.(8 per cent.) was significant at this level.  相似文献

Single plants of white clover, grown in a controlled environmentand dependent for nitrogen on fixation in their root nodules,were defoliated once by removing approximately half their shoottissue. Their regrowth was compared with the growth of comparableundefoliated plants. Two similar experiments were carried out:in the first, plants were defoliated at 2.5 g, and in the secondat 1.2 g total plant d. wt. Defoliation reduced rate of N2 fixation by > 70 per cent,rate of photosynthesis by 83–96 per cent, and rate ofplant respiration by 30–40 per cent. Nodule weights initiallydeclined following defoliation as a result of loss of carbohydratesand other unidentified components. No immediate shedding ofnodules was observed but nodules on the most severely defoliatedplants exhibited accelerated senescence. The original rates of N2 fixation were re-attained after 5–6or 9 d regrowth, with increase in plant size at defoliation.In general, the rate of recovery of N2 fixation was relatedto the re-establishment and increase of the plant's photosyntheticcapacity. Throughout the growth of both defoliated and undefoliatedplants nodule respiration (metabolism) accounted for at least23 ± 2 per cent of gross photosynthesis. The unit ‘cost’of fixing N2 in root nodules, in terms of photosynthate, appearedto be unaffected by defoliation, except perhaps for plants veryrecently defoliated. Similarly, the percentage nitrogen contentsof shoot, root and nodules of defoliated plants became adaptedwithin a few days to those characteristic of undefoliated plants. A rough action-spectrum curve (in the range 400–600 mµ)for light-stimulated spore discharge in Sordaria fimicola iscompared with the absorption-spectrum curve of an ethyl-alcoholextract of the fungus. The two curves show some agreement. Amutant lacking black pigment (probably melanin) is shown torespond to light in its spore discharge as does the normal,but evidence is given that it is more sensitive to light. Itseems that the melanin in the perithecial wall of the normalstrain acts as a neutral filter reducing the effectiveness ofthe incident light.  相似文献

The nutrition of developing fruits of Ricinus communis was studiednear Perth, Western Australia, where the species grows as aweed on poor sandy soil. Fruits required 60 days to mature anddehydration of the capsule began 20 days before the seeds ripened.Mature seeds accumulated 49 per cent of the fruit dry matterand over 80 per cent of its P, Zn and Cu, 50–80 per centof its Mg, N, Fe and Mn, 41–46 per cent of its S and Caand 11–21 per cent of its K and Na. Losses of nutrientsfrom capsules during fruit ripening were: Zn, 73 per cent, P,42 per cent, Cu, 23 per cent and Mn, 8 per cent. Dry matter,N, K, S, Ca, Mg, Na and Fe were not withdrawn from capsules.Apparent retranslocation from capsules could have provided from6–28 per cent of the Zn, Mn, P and Cu in mature seeds.Seeds from plants on poor sandy soil were small but had adequatelevels of nutrients when compared with those from plants growingon a fertile loam. Concentrations of all nutrients except P were higher in youngcapsules than in young seeds, but levels of N, P, Mg, Fe, Znand Cu were higher in mature seeds than in mature capsules.The intake of most nutrients by fruits was out of phase withdry matter accumulation, especially in capsules, and the elementsappeared to accumulate in fruit parts independently of eachother. Glutamine accounted for over 85 per cent of the amino-Nin phloem sap destined for fruits. Potassium made up over 90per cent of the inorganic cations in phloem exudate. Of theminor elements in the exudate, Fe was present at highest concentrationand Cu at the lowest. The results showed that retranslocation from the capsule madea very small contribution to the nutrition of seeds. It is suggestedthat R. communis would require a sustained supply of soil nutrientsto ensure maximum seed yield, partly due to the restricted retranslocationof most nutrients from capsules. Twenty-eight-day-old plants of Silene coeli-rosa L. were maintainedin short days (SD) for 9 d (0–8) or exposed to 7 longdays (LD), or 7 SD with a 5 min exposure at 1700 h of each dayto far-red (FR), red (R) or 5 min FR/5 min R, or 7 dark-interrupted(di = 1700–1720 h) LD. Treatments were followed by twofurther SD. The mitotic index and G1 and G2 proportions weremeasured in the shoot apices of plants sampled at 2000 h ofeach day of each replicated treatment. Exposure to 7 LD (= 100per cent flowering) resulted in significant increases, relativeto the SD controls, in both the G2 proportion and the mitoticindex on d 0 to 3, 7 and 8. Five minute FR (= 0 per cent flowering)resulted in cell cycle responses similar to those in LD onlyfrom d 0 to 2. R and FR/R (both = 0 per cent flowering) didnot result in any increases in the G2 proportion in the apexapart from d 3 of FR/R. However 5 min FR/5 min R, and to a lesserextent 5 min R, did result in significant increases in the mitoticindex on d 0, 1, 7, and 8. diLD (= 8–10 per cent flowering)also prevented any significant increases in the G2 proportionon d 0 to 3, and 5 to 8 but the mitotic index was again higheron these days compared with control data. Thus the transitionto floral growth for 90 per cent of the plants is associatedwith changes in the cell cycle in the shoot apex measured asincreases in the G2 proportion at 2000 h of LD 0 to 3 and 7to 8. The sporulation process in Sordaria fimicola has been studiedunder controlled conditions, with particular reference to discharge.Discharge occurred with a circadian periodicity for the greaterpart of the sporulation life of the culture, with peaks at about18.00 hours in either light or darkness. The consistency ofthis phenomenon was confirmed by an analysis of previous work.Fluctuations in light intensity, temperature, relative humidity,carbon dioxide concentration, and vibration were eliminatedas far as possible, as causes of this periodicity. Minor fluctuationsin discharge rate with a period of less than 24 h were probablyrandom. The general nature of the periodicity was shown to be consistentwith some aspects of endogenous rhythms in other organisms.However, spore discharge in S. fimicola is very sensitive tolight, and it is readily entrained to discharge out of phasewith the natural period. The rate of development of two stages of spore formation (delimitationand maturation) were determined, and these were related to therate and periodicity of discharge. From the data it was impossibleto establish at which stage of sporulation the periodicity arose.  相似文献

The effects of chloramphenicol and kinetin on uptake and incorporationof 35S-methionine and some 14C-amino acids have been investigatedin leaf-disks of Nicotiana rustica in light and dark. Chloramphenicolin a concentration of 1 mg per ml inhibits the uptake of aminoacids from 30 to 60 per cent compared with the water control.The incorporation of amino acids into bulk protein is stronglyinhibited in light (40 to 70 per cent), but only to a smalldegree in dark (10 to 20 per cent), as revealed also by 14CO2-photosynthesisof the disks and following treatment with chloramphenicol indark. The stimulating effect of kinetin on uptake and incorporationof amino acids is dependent upon its concentration (10–5to 10–6 M ; but 10–4 M solution inhibits stronglyboth uptake and incorporation). The stimulation seems to influencemore incorporation than uptake processes. Possible interactionsof chloramphenicol and kinetin in the protein metabolism oftobacco leaves have been discussed. A study has been made, using C14 as a tracer, of the variationswhich occur according to the physiological condition of thecells in the distribution of carbon fixed, both in the lightand in the dark, by the diatom Navicula pelliculosa (Bréb.)Hilse during periods of 30 to 300 seconds. Fixation into thefollowing cell fractions was determined: (A) material solublein 80 per cent. ethanol but insoluble in benzene, (B) materialsoluble both in 80 per cent. ethanol and in benzene, and (C)material insoluble in 80 per cent. ethanol. Carbon fixed inphotosynthesis was incorporated, rapidly and in amounts representingup to 70 per cent. of the total fixation, into fractions B andC, as well as into fraction A. Considerable variation was foundin the proportions of carbon entering the three fractions inthe light; in actively growing cells the proportion enteringfraction C preponderated over that in B, corresponding to thesynthesis of protein, whereas in nitrogen-deficient cells fixationin B was the greater, corresponding to the synthesis of fat.These patterns changed only slowly, over periods of days, followingthe transfer of cells to altered conditions of nitrate supply.However, when ammonium nitrogen was supplied to nitrogen-deficientcells a marked change in distribution of carbon fixed occurredwithin 5 minutes, fixation in fraction B falling to a low valueand that in A rising correspondingly. In cells subjected toprolonged nitrogen-deficiency, fixation in fraction B fell toa relatively low value but the proportion which this fractionformed of the total dry matter in the cells rose as a resultof an increased rate of loss from the cells of constituentsother than lipides. The distribution of carbon fixed was alsodependent on light intensity. Fixation in fractions B and Crose relatively to that in A as light intensity was increasedup to 100 foot-candles but fell again at the highest intensityused, 2,000 foot-candles. These results are discussed with particularreference to the relationship between fat accumulation and photosynthesisin algae.  相似文献
