实验在雄性SpragueDawley 大鼠上进行。实验动物被随机分为对照组和应激组, 应激组大鼠每天给予电击足底结合噪声的应激刺激, 每日2 次, 每次2 h 。应激组大鼠在接受连续15 d 的慢性应激刺激后, 其尾动脉收缩压与对照动物相比有显著升高。对照组为16-25 ±0-63kPa (n = 7) ; 应激组为19-55 ±1-45 kPa (n = 8, P< 0-05) 。用RTPCR 结合Southern 印迹核酸分子杂交技术观察到, 血管升压素(vasopressin, AVP)V1 受体mRNA 广泛存在于大鼠下丘脑、皮质、延髓等部位以及心脏、肝脏、肾脏等组织中。用定量PCR 方法观察到, 大鼠在接受慢性应激刺激之后, 其大脑顶叶皮质、下丘脑及延髓组织中AVPV1 受体mRNA 水平均显著低于正常大鼠( 顶叶皮质: P< 0-05 ; 下丘脑: P< 0-01 ; 延髓: P< 0-001) , 而心脏、肝脏及肾脏组织中的AVPV1 受体mRNA水平与正常大鼠相比均无明显差别( 心脏: P> 0-05 ; 肝脏: P> 0-05 ; 肾脏:P> 0-05) 。上述结果提示, 慢性应激刺激可引起大鼠不同部位脑组织AVPV1 受体合成水平下调, 可能导致  相似文献

实验在雄性SD大鼠中进行,用核酸斑点杂交技术观察下丘脑组织中血管升压素(AVP)基因转录水平变化,用异羟基洋地黄毒甙(GIG)标记的26个碱基长寡聚核苷酸作为检测探针。实验中观察到,用渗透压微泵向大鼠侧脑人连续注射微量血管紧张素Ⅰ(0.2nmol/h)2d后,可引起动物饮水量显著增加,下丘脑组织中AVP基因转录水平高,但无统计学显著意义。将实验动物日饮水量限制在与对照动物相同的每日饮水量之后,侧脑  相似文献

本实验观察了三种应激情况下,大鼠血浆,下丘脑,延髓,心肌,血管及肾上腺组织血管紧张素Ⅱ(AⅡ)含量,以及血浆皮质酮含量的。应激方式分为急性应激(冷水游泳,断肢创伤)与慢性应激(寒冷环境刺激)。结果表明:急性应激动物血中AⅡ剧烈增高,游泳组达对照值的900%,创伤组增至390%,慢性寒冷组增至134%;而组织AⅡ除肾上腺外,则以慢性寒冷组增加最明显,游泳组次之,创伤组无明显变化。血浆皮质酮各组均显著  相似文献

目的通过体外细胞实验研究,探讨血管紧张素受体1在血管紧张素Ⅱ诱导人星形胶质细胞活性氧产生和细胞老化中的作用。方法人星形胶质细胞随机分为三组:血管紧张素Ⅱ+Cand(坎地沙坦)组和血管紧张素Ⅱ+tempol组。血管紧张素Ⅱ组是用100nM血管紧张素Ⅱ刺激人星形胶质细胞3天,血管紧张素Ⅱ+Cand组和血管紧张素Ⅱ+tempol组先用血管紧张素受体1阻滞剂坎地沙坦(100nM)和氧自由基清除剂tempol(3mM)预处理,再用100nM血管紧张素II刺激人星形胶质细胞3天,利用β半乳糖苷酶染色评估细胞老化。不同剂量(0、1nM、10nM、100nM、1000nM和1000nM+坎地沙坦)的血管紧张素Ⅱ刺激人星形胶质细胞30min,DHE染色评估细胞内活性氧产生。结果血管紧张素Ⅱ引起人星形胶质细胞DHE染色表达增多和β半乳糖苷酶染色细胞增多。利用血管紧张素受体1阻滞剂坎地沙坦和氧自由基清除剂tempol预处理逆转了血管紧张素Ⅱ引起的星形胶质细胞老化。结论血紧张素Ⅱ是通过血管紧张素受体1和超氧阴离子产生引起星形胶质细胞的老化。  相似文献

应用幼年小鼠经孕马血清促性腺激素(pregnant mares serum gonadotropin,PMSG)处理的动物模型,研究了卵泡从发育到闭锁动态变化过程中血管紧张素Ⅱ(AngⅡ)的作用。结果表明:(1)24日龄小鼠给予PMSG(10IU/只)后6d时,卵巢中出现大量闭锁卵泡,颗粒细胞DNA琼脂糖电泳显示了梯形条带;(2)随卵泡闭锁发生,卵巢AngⅡ含量增加;(3)AngⅡ显著拮抗FSH刺激颗粒细胞雌二醇生成的作用。我们认为,AngⅡ参与了对小鼠卵泡闭锁的调节。  相似文献

文献报道脑内存在血管紧张素Ⅱ系统。与此一致,本工作用氨基甲酸乙脂麻醉、箭毒制动、人工呼吸的大鼠观察到:(1)穹窿下器(SFO)、室旁核(NPV)或NPV的投射区:延髓头端腹外侧区(RVLM)、导水管周围灰质(PAG)、蓝斑(LC)内注入血管紧张素Ⅱ(AⅡ)均引起升压反应;(2)SFO升压反应可被双侧NPV或RVLM内预先注入[Sar1,Thr8]AⅡ(STAⅡ,AⅡ拮抗剂)明显衰减,NPV升压反应也可被RVLM内注入STAⅡ削弱;(3)双侧PAG用STAⅡ预处理后,AⅡ引起的NPV或SFO升压反应均明显减小;(4)NPV升压反应还可被双侧LC内预先注射STAⅡ衰减,但SFO升压反应不受影响。结合我们以往工作曾显示兴奋PAG或LC均可作用于RVLM引起升压反应,目前的结果表明:SFO内的AⅡ能神经元通过NPV内AⅡ能神经元,不仅可直接作用于RVLM引起升压反应,而且还可间接通过PAG作用于RVLM起升压作用,但LC不参与SFO升压反应。  相似文献

为检测血管紧张素Ⅱ(angiotensin Ⅱ,AⅡ)对小鼠胚胎干细胞(embryonic stem cells,ESCs)向心肌细胞方向分化的作用,采用10-4 mol/L维生素C诱导小鼠R1胚胎干细胞分化为心肌细胞. Western印记检测胚胎干细胞诱导分化的心肌细胞中表达血管紧张素Ⅱ1 型受体(angiotensin Ⅱ type 1 receptor,AT1R).诱导分化期间用1 μmol/L AⅡ刺激胚胎干细胞,计数搏动拟胚体的比例;诱导分化第14 d用real-time RT-PCR 和Western 印记检测心肌标志物的表达确定其作用. 结果显示,与对照组相比,1 μmol/L AⅡ处理组可显著增加搏动拟胚体的比例,上调心肌标志物mRNA的表达. 预先用1 μmol/L洛沙坦处理1 h后可显著阻碍这种上调作用. 本实验结果表明,AⅡ通过AT1R可促进小鼠R1胚胎干细胞向心肌细胞分化.  相似文献

在培养新生大鼠心肌细胞上,探讨一氧化氮(NO)在血管紧张素Ⅱ诱导的心肌细胞以大中的作用。结果显示,血管紧张素Ⅱ可使心肌细胞蛋白质含量显著增加,心肌细胞一氧化氮合酶(NOS)活性和培养液NO浓度明显降低。血管紧张素Ⅱ可明显降低心肌细胞eNOSmRNA水平。Saralsin和百日咳毒素(PTX)可抑制血管紧张素Ⅱ诱导的蛋白质含量增加、心肌细胞NOS活性减弱和培养液NO浓度降低。硝普钠提高心肌细胞培养  相似文献

Our previous studies have shown that central administration of angiotensin II (ANG II) causes vasopressin release in the near-term fetus in utero as evidence that the hypothalamic-neurohypophysial system has relatively matured before birth. However, it is still unknown whether the vasopressin controlling centers have been functionally developed in younger fetuses. This study determined fetal plasma vasopressin levels and hypothalamic vasopressin neuron activity in the chronically instrumented pre-term ovine fetuses. Introcerebroventricular (i.c.v.) administration of ANG II did not affect fetal plasma osmolality and sodium concentrations. However, fetal plasma vasopressin levels were significantly increased ( approximately 3-fold) in response to central injection of ANG II. Central ANG II also induced vasopressin-neuron activity marked with c-fos expression in the fetal hypothalamus at pre-term. In addition, the fetal organum vasculosum of the lamina terminalis and the subfornical organ were activated. The results suggest that hypothalamic-neurohypophysial system has been relatively intact and functional at 70% gestational age, and that central angiotensin is important in inducing fetal vasopressin release in utero.  相似文献

The influence of intracerebroventricular injections of the angiotensin II antagonist--saralasin on the cardiovascular reactions elicited by electrical stimulation of different structures of the hypothalamus in rabbit was studied. The saralasin in doses from 1 to 1.5 mg reduced arterial pressure by 9 +/- 0.2 mm Hg and decreased the amplitude of the hypertensive reactions elicited by electrical stimulation of the paraventricular, supraoptic, ventromedial, supramammillary nucleus of the hypothalamus, area hypothalamic anterior and lateral. It has been shown that most suppressing influence of the saralasin on amplitude of the hypertensive reactions elicited by electrical stimulation of the paraventricular (68%) and supraoptic (76%) nucleus of the hypothalamus which contain magnocellular neurosecretory neurons.  相似文献

Cysteamine (beta-mercaptoethylamine, CSH) has been reported to have various effects on the neuroendocrine system. Reports indicate CSH decreases pituitary oxytocin (OT) without affecting pituitary vasopressin (VP). However, preliminary studies from our laboratory strongly indicate that CSH has an effect on VP release. Experiments were conducted with dibenzyline-treated, urethane-anesthetized, male Sprague-Dawley (SD) rats. Rats were injected with 4 mU of standard VP and 4 mg/100 g of CSH. Administration of VP resulted in an increase in mean arterial pressure (MAP) of 23.5 +/- 3.2 mm Hg. Administration of CSH resulted in a consistent, immediate decrease in MAP of 13.0 +/- 2.0 mm Hg prior to an increase of 21.0 +/- 2.6 mm Hg. The effects due to VP and CSH were strikingly different; the CSH-induced MAP rise took longer to peak and to return to baseline. Both the VP- and CSH-induced MAP rise were markedly inhibited by a prior administration of a specific VP antagonist d(CH2)5[Tyr(Me)]AVP. In addition, the typical increase in MAP observed in SD rats following CSH administration was substantially reduced when the same dose was administered in homozygous diabetes insipidus (HODI) rats. The data presented here strongly suggest that CSH-induced MAP elevation is due to the release of VP from the pituitary gland.  相似文献

Whether or not 1-desamino-8-D-arginine-vasopressin (DDAVP) reduces blood pressure or affects the release of arginine vasopressin (AVP) and renin is controversial, although evidence suggests AVP and renin are important in maintaining blood pressure during hemorrhage. We therefore investigated the effect of DDAVP on endogenous release of AVP and renin and on blood pressure during hemorrhage in dogs. In the control group the hemorrhage was performed at a rate of 0.4 ml.kg-1.min-1 for 40 min from the femoral artery. The plasma AVP concentration and renin activity (PRA) increased progressively in response to the hemorrhage, from 7.5 +/- 0.5 to 40.3 +/- 7.3 pg.ml-1, and from 11.8 +/- 1.5 to 20.5 +/- 4.2 ng.ml-1.h-1, respectively, while blood pressure decreased slightly. In the DDAVP group, intravenous infusion of DDAVP (2.5 ng.kg-1.min-1 for 40 min) and hemorrhage were simultaneously performed. The plasma DDAVP concentration increased progressively to 218 +/- 21.0 pg.ml-1. There was no significant difference, however, between the control and DDAVP groups in the response of AVP, PRA and blood pressure. The results suggested that DDAVP may not affect the release of AVP and renin or blood pressure during hemorrhage.  相似文献

Vasopressin V(1b) receptor is specifically expressed in the pituitary and mediates adrenocorticotropin release, thereby regulating stress responses via its corticotropin releasing factor-like action. In the present study we examined catecholamine release in response to two types of stress in mice lacking the V(1b) receptor gene (V(1b)R(-/-) mice) vs. wild-type mice. There were no significant differences in the basal plasma levels of catecholamines between the two genotypes. In response to stress induced by forced swimming, norepinephrine (NE), but not epinephrine (E) or dopamine (DA), was increased in wild-type mice, whereas the increases in NE and DA were not observed in V(1b)R(-/-) mice. In wild-type mice, E, but not NE or DA, was increased in response to social isolation stress, whereas the increase in E was not observed in V(1b)R(-/-) mice. These results suggest that the V(1b) receptor regulates stress-induced catecholamine release. Because it has been suggested that arginine-vasopressin (AVP) is related to the development of depression, we also evaluated immobility time in the forced swimming test, and we found no significant change in V(1b)R(-/-) mice. Taken together, these findings suggest that, in addition to the previously elucidated effect on the hypothalamic-pituitary-adrenal axis, vasopressin activity via V(1b) receptors regulates stress-induced catecholamine release.  相似文献

Aging impairs shear-stress-dependent dilation of arteries via increased superoxide production, decreased SOD activity, and decreased activation of endothelial nitric oxide (NO) synthase (eNOS). In the present study, we investigated whether chronic increases in shear stress, elicited by increases in blood flow, would improve vascular endothelial function of aged rats. To this end, second-order mesenteric arteries of young (6 mo) and aged (24 mo) male Fischer-344 rats were selectively ligated for 3 wk to elevate blood flow in a first-order artery [high blood flow (HF)]. An in vitro study was then conducted on first-order arteries with HF and normal blood flow (NF) to assess shear stress (1, 10, and 20 dyn/cm(2))-induced release of NO into the perfusate. In HF arteries of both age groups, shear stress-induced NO production increased significantly. In 24-mo-old rats, the reduced shear stress-induced NO production in NF arteries was normalized by HF to a level similar to that in NF arteries of 6-mo-old rats. The increased NO production in HF arteries of 24-mo-old rats was associated with increased shear stress-induced dilation, expression of eNOS protein, and shear stress-induced eNOS phosphorylation. Wortmannin, a phosphatidylinositol 3-kinase inhibitor, reduced shear stress-induced eNOS phosphorylation and vasodilation. Superoxide production decreased significantly in HF compared with NF arteries in 24-mo-old rats. The decreased superoxide production was associated with significant increases in CuZn-SOD and extracellular SOD protein expressions and total SOD activity. These results suggest that stimulation with chronic HF restores shear-stress-induced activation of eNOS and antioxidant ability in aged arteries.  相似文献

The first inutero diagnosis of Sandhoff's disease was made in an at-risk fetus by the demonstration of deficient β-N-acetyl-hexosaminidase A and B activities in amniotic fluid components the day of amniocentesis. These enzymatic deficiencies were determined by enzyme assay and electrophoresis using 4-methylumbelliferyl-β-N-acetyl-glucosaminide as substrate. The concentrations of the neutral glycosphingolipids were quantified in amniotic fluid; the level of the glycosphingolipid substrate, globoside, was markedly increased in amniotic fluid from the at-risk fetus compared to that of fetal controls. In addition, ultrastructural examination demonstrated pathologic glycosphingolipid accumulation in uncultured amniotic cells. These enzymatic, chemical and ultrastructural procedures provided the rapid and accurate inutero diagnosis Sandhoff's disease within three days of amniocentesis. The inutero diagnosis was confirmed by the marked deficiencies of β-N-acetyl-hexosaminidase A and B in plasma and various tissues from the aborted fetus. These findings indicated that maternal hexosaminidases do not cross the fetal-placental barrier.  相似文献

Anterior pituitaries from normal rats were enzymatically dispersed and placed into monolayer cell culture in order to determine if and how angiotensin II (Ang II) mediates the in vitro release of ACTH and other pituitary hormones. Ang II stimulated ACTH secretion in a time dependent fashion. This release occurred at physiologic concentrations of Ang II and was linearly correlated with the log dose of Ang II. One hour pretreatment of the cells with cycloheximide, a inhibitor of protein synthesis, significantly decreased the cellular ACTH secretory response to Ang II. Ang 11 did not mediate the release of LH nor of ADH, a proposed stimulator of ACTH secretion.  相似文献
