We have studied the activities of Ca2+-stimulated ATPase in rat heart sarcolemma upon modulating the redox state of membrane thiol groups with dithiothreitol (DTT). The suitability of alamethicin to unmask the latent activity of this enzyme was also investigated. The Ca2+-stimulated ATPase in sarcolemma exhibited two activation sites — one with low affinity (Km = 0.70 ± 0.2 mM; Vmax = 10.0 ± 2.2 mol Pi/mg/h) and the other with high affinity (Km = 0.16 ± 0.7 mM; Vmax = 4.6 ± 0.8 mol Pi/mg/h) for Mg2+ATP. Alamethicin at a ratio of 1:1 with the sarcolemmal protein caused a 3-fold activation of Ca2+-stimulated ATPase without affecting its sensitivity to Ca2+ or Mg2+ATP. Treatment of sarcolemma with deoxycholate or sodium dodecyl sulfate resulted in a total loss of the enzyme activity; high concentrations of alamethicin also showed a detergent-like action on the sarcolemmal vesicles. DTT at 5–10 mM concentrations caused a 4–5 fold activation of Ca2+-stimulated ATPase in sarcolemma and this effect was observed to be dependent on the concentration of Mg2+ATP. DTT increased the affinity of the enzyme to Mg2+ATP at the high affinity site and enhanced the Vmax at the low affinity site in addition to increasing the sensitivity of Ca2+-stimulated ATPase to Ca2+. DTT protected the Ca2+-stimulated ATPase against deterioration by detergents and restored the enzyme activity after treatment with N-ethylmaleimide. The mechanism of action of DTT on Ca2+-stimulated ATPase may involve the reduction of essential thiols at the active site of the enzyme or its interaction with specific DTT-dependent inhibitor protein. No changes in the sensitivity of sarcolemmal Ca2+-stimulated ATPase to orthovanadate was evident in the absence or presence of DTT and alamethicin. The results suggest the use of both DTT and alamethicin for the determination of Ca2+-stimulated ATPase activity in sarcolemmal preparations.  相似文献

The protein-sensitized fluorenscence of Tb3+ has been used to characterize cation binding sites on synaptic vesicles. In the presence of 100 mM KC1, the vesicles display a single class of sites having affinities for Ca2+ and Mg2+ in the mM range. Of subcellular fractions synaptic vesicles have the greatest number of sites, which involve tyrosine residues. Morphine, which is known to deplete Ca from nerve terminals in vivo by a mechanism possibly involving low affinity sites on synaptic vesicles, does not compete in vitro for the sites monitored by Tb fluorescence. In addition, acute morphine treatment does not alter the binding of Tb to the synaptic vesicle fraction. It is suggested that the depletion of brain Ca induced by morphine is not mediated by a direct action of opiates on these membrane sites.  相似文献

Kinetic experimentation was used to characterize the Mg2+ and Mn2+ modulation of Ca2+ transport and ATPase activity in sarcoplasmic reticulum vesicles. In addition to its participation in the ATP·Mg complex as substrate for the ATPase, Mg2+ is an activator of phosphoenzyme progression to hydrolylic cleavage. It is shown that this activation is due to Mg2+ occupancy of an allosteric site easily accessible on the outer surface of the vesicles, rather than to participation in an antiport mechanism. The Mg2+ site is distinct from the Ca2+ binding sites which are involved in activation of enzyme phosphorylation by ATP, and Ca2+ translocation. The role of Mg2+ is quite specific, inasmuch as phosphoenzyme decay is much slower if the Mg2+ allosteric site is occupied by Ca2+. Conversely, competive occupancy of the Ca2+ sites by Mg2+ does not permit enzyme phosphorylation by ATP. Intermediate characteristics between Mg2+ and Ca2+ are displayed by Mn2+ which is well able to stimulate phosphoenzyme cleavage by occupancy of the Mg2+ allosteric site, and is also able (although at much slower rates) to activate enzyme phosphorylation, and undergo active transport by occupancy of the Ca2+ sites.  相似文献

Ca2+ transport activity in synaptosomal membranes has been identified as having two major components: Ca2+-stimulated ATP hydrolysis and ATP-dependent CA2+ uptake. Both processes exhibit similar affinities for Ca2+ and operate maximally under identical buffer conditions. Subcellular fractionation studies revealed the Ca2+/Mg2+ ATPase and ATP-dependent CA2+ uptake activities to be highest in synaptic plasma membrane fractions 1 and 2, with lesser activity in synaptic vesicles and mitochondria. Progressive treatment with Triton X-100 activated, then decreased Ca2+/Mg2+ ATPase, Mg2+ ATPase and Ca2+ ATPase. ATP-dependent Ca2+ uptake was progressively decreased by similar treatment with Triton X-100. These studies illustrate that Ca2+ ATPase and ATP-dependent Ca2+ uptake may provide two important mechanisms for buffering of cytosolic Ca2+ at the nerve terminal. These systems may function to rapidly sequester cytosolic Ca2+ following a rise during depolarization and then extrude Ca2+ from the terminal against a concentration gradient. This regulation of cytosolic Ca2+, represented by two processes of the type seen in other plasma membranes, may play critical roles in calcium homeostasis in nerve cells.Footnote: Portions of this research were submitted by K. ATP and the divalent cations Mg2+ and Ca2+ regulated K+ stimulation of the Ca2+-transport ATPase of cardiac sarcoplasmic reticulum vesicles. Millimolar concentrations of total ATP increased the K+-stimulated ATPase activity of the Ca2+ pump by two mechanisms. First, ATP chelated free Mg2+ and, at low ionized Mg2+ concentrations, K+ was shown to be a potent activator of ATP hydrolysis. In the absence of K+ ionized Mg2+ activated the enzyme half-maximally at approximately 1 mM, whereas in the presence of K+ the concentration of ionized Mg2+ required for half-maximal activation was reduced at least 20-fold. Second MgATP apparently interacted directly with the enzyme at a low affinity nucleotide site to facilitate K+-stimulation. With a saturating concentration of ionized Mg2+, stimulation by K+ was 2-fold, but only when the MgATP concentration was greater than 2 mM. Hill plots showed that K+ increased the concentration of MgATP required for half-maximal enzymic activation approx. 3-fold.Activation of K+-stimulated ATPase activity by Ca2+ was maximal at anionized Ca2+ concentration of approx. 1 μM. At very high concentrations of either Ca2+ or Mg2+, basal Ca2+-dependent ATPase activity persisted, but the enzymic response to K+ was completely inhibited. The results provide further evidence that the Ca2+-transport ATPase of cardiac sarcoplasmic reticulum has distinct sites for monovalent cations, which in turn interact allosterically with other regulatory sites on the enzyme.  相似文献

Tb3+, a fluorescent trivalent cation with physicochemical properties similar to Ca2+, binds to peripheral nerve membrane vesicles prepared from the walking leg nerve bundle of the lobster (Homarus americanus). Saturable binding is measured for at least two classes of binding site. Bound Tb3+ can be displaced by other cations in the order: Ca2+ > Mg2+ = Zn2+ > NH4+. The binding of Tb3+ to the lower affinity site (KD(app) = 6.0 μM) is inhibitable by Na+, Mg2+ and Ca2+, whereas the higher affinity site (KD(app) = 2.2 μM) is only sensitive to Ca2+. Using this spectral probe the role of Ca2+ in peripheral nerve membrane function can be investigated.  相似文献

Acutolysin D, isolated from the venom of Agkistrodon acutus, possesses marked haemorrhagic and proteolytic activities. The molecular weight and the absorption coefficients (A 1% 280) of acutolyisn D have been determined to be 47,850 ± 8 amu and 9.3 by mass spectrometer and UV spectrum, respectively. The effects of metal ions on the conformation and activity of acutolysin D have been studied by following fluorescence, circular dichroism and biological activity measurements. Acutolysin D contains two Ca2+-binding sites and two Zn2+-binding sites determined by atomic absorption spectrophotometer. Zn2+ is essential for the enzyme activities of acutolysin D, however, the presence of 1 mM Zn2+ significantly decreases its caseinolytic activity and intrinsic fluorescence intensity at pH 9.0 due to Zn(OH)2 precipitate formation. Ca2+ is important for the structural integrity of acutolysin D, and the presence of 1 mM Ca2+ markedly enhances its caseinolytic activity. Interestingly, the caseinolytic activity which is inhibited partly by Cu2+, Co2+, Mn2+ or Tb3+ and inhibited completely by Cd2+, is enhanced by Mg2+. The fluorescence intensity of the protein decreases in the presence of Cu2+, Co2+, Cd2+ or Mn2+, but neither for Ca2+, Mg2+ nor for Tb3+. Zn2+, Ca2+, Mg2+, Cu2+, Mn2+, Co2+ and Tb3+ have slight effects on its secondary structure contents. In addition, Cd2+ causes a marked increase of antiparallel β-sheet content from 45.5% to 60.2%.  相似文献

External treatment of human erythrocytes with the diazonium salt of sulfanilic acid does not inhibit the Mg2+-dependent ATPase but does markedly inhibit the Ca2+-stimulated ATPase activity. Inhibition of the (Na+ + K+)-dependent activity is dependent upon the concentration of diazonium salt used. Treatment of membrane fragments does not irreversibly inhibit the (Na+ + K+)-dependent ATPase even though the diazonium salt binds covalently to membrane components. However, the Mg2+-dependent and Ca2+-stimulated ATPase activities are irreversibly inhibited. ATP and Mg-ATP will completely protect the (Na+ + K+)-dependent ATPase when present during treatment of membrane fragments with the diazonium salt, but only Mg-ATP will protect the Mg2+-dependent ATPase from inhibition. The Ca2+-stimulated ATPase activity is not protected.  相似文献

Neuronal ATPases comprise a wide variety of enzymes which are not uniformly distributed in different membrane preparations. Since purified vesicle fractions have Mg2+/Ca2+-ATPase, the purpose of the present study was to know whether such enzyme activities have a preferential concentration in a synaptic vesicle fraction in order to be used as markers for these organelles. Resorting to a procedure developed in this Institute, we fractionated the rat cerebral cortex by differential centrifugation following osmotic shock of a crude mitochondrial fraction and separated a purified synaptic vesicle fraction over discontinuous sucrose gradients. Mg2+/Ca2+-ATPase activities and ultrastructural studies of isolated fractions were carried out. It was observed that similar specific activities for Mg2+/Ca2+-ATPases were found in all fractions studied which contain synaptic vesicles and/or membranes. Although the present results confirm the presence of Mg2+ and Ca2+-ATPase activities in synaptic vesicles preparations, they do not favor the contention that Mg2+/Ca2+-ATPase is a good marker for synaptic vesicles.  相似文献

A fraction was isolated from human red cells capable of decreasing Ca2+-stimulated with a parallel increase of Mg2+-stimulated ATPase. This fraction seems to produce an increase of the apparent affinity for Ca2+. But, when the fraction concentration was increased, there was a decrement of Ca2+-stimulated ATPase up to the point of complete abolishment. At this point, Mg2+-stimulated ATPase reached the initial level of the Ca2+-stimulated ATPase.  相似文献

Heart sarcolemma has been shown to possess three catalytic sites (I, II and III) for methyl transferase activity (Panagia V, Ganguly PK and Dhalla NS. Biochim Biophys Acta 792: 245–253, 1984). In this study we examined the effect of phosphatidylethanolamine N-methylation on ATP-independent Ca2+ binding and ATPase activities in isolated rat heart sarcolemma. Both low affinity (1.25 mM Ca2+) and high affinity (50 µM Ca2+) Ca2+ binding activities were decreased following incubation of sarcolemmal membranes with AdoMet under optimal conditions for site II and III. Similarly, Ca2+ ATPase activities measured at 1.25 mM and 4 mM Ca2+ were depressed by phospholipid N-methylation. S-adenosyl homocysteine, a specific inhibitor of phospholipid N-methylation, prevented the depression of low affinity Ca2+ binding and Ca2+ ATPase activities, whereas the methylation-induced effect on the high affinity Ca2+ binding was not influenced by this agent. Pretreatment of sarcolemma with methyl acetimidate hydrochloride, an amino group blocking agent, also prevented the methylation-induced inhibition of both Ca2+ binding and Ca2+ ATPase. A further decrease in Ca2+ binding and Ca2+ ATPase activities together with a marked increase in the intramembranal level of PC was seen when membranes were methylated under the site III conditions in the presence of phosphatidyldimethylethanolamine as exogenous substrate. There was no effect of phospholipid methylation on sarcolemmal Na+-K+ ATPase and Mg2+ ATPase activities. These results indicate a role of phospholipid N-methylation in the regulation of sarcolemmal Ca2+ ATPase and low affinity ATP-independent Ca2+ binding.  相似文献

Summary The Ca2+ channel blockers felodipine and bepridil are known to affect selectively functions of calmodulin. We studied their effects on calmodulin binding and ATPase activities of calmodulin-containing and calmodulin-depleted rabbit heart sarcolemma. Both drugs as well as the specific anti-calmodulin drug calmidazolium at a concentration of 50 µM, inhibited the Ca2+-stimulated calmodulin binding to calmodulin-depleted sarcolemma. Within the concentration range of 3 to 100 µM all three drugs also progressively inhibited Ca2+ pumping ATPase in calmodulin containing sarcolemma, although the enzyme was assayed at saturating Ca2+ (100 µM). The inhibitory potency of calmidazolium and bepridil, but not that of felodipine, increased when the membrane protein concentration in the ATPase assay was lowered. At low membrane protein concentration 30 µM calmidazolium completely blocked calmodulin-dependent Ca2+ pumping ATPase, whereas the inhibition caused by 30 µM felodipine or bepridil remained partially. A similar inhibition pattern of the drugs was found in the calmodulin binding experiments. Within a concentration range of 3 to 30 µM, all three drugs had negligible effects on the basal Ca2+ pumping ATPase which was measured in calmodulin-depleted sarcolemma. In conclusion, the characteristics of the anti-calmodulin action of felodipine on the rabbit heart sarcolemmal Ca2+ pumping ATPase are not different from those of bepridil. The extent of the negative cooperativity with MgATP of the Ca2+-stimulated ATPase activity of sarcoplasmic reticulum has been studied with various membrane preparations and under various conditions. Preparations studied were fragmented sarcoplasmic reticulum vesicles, deoxycholate-solubilized and fractionated ATPase, triton extracted reticulum, vesicles reconstituted from either detergent, and limited trypsin digests of the reticulum. Conditions studied were suboptimal, optimal, and inhibitory Ca2+ concentrations; temperatures from 13 to 46 °C; 1 or 5 mm MgCl2; 0.1 m KCl, 0.1 m NaCl, or no added salt; and Triton or deoxycholate present in the assay. With preparations in which vesicles could accumulate Ca2+ ion, the ionophore A23187 was added to prevent inhibition by internal Ca2+ ions. Under all circumstances, the negative cooperativity of MgATP was present (Hill coefficient of 0.2 to 0.8), indicating the persistence of the properties of the enzyme molecule and its lipid environment giving rise to kinetic negative cooperativity. Attempts to measure the number of ATP sites by protection against N-ethylmaleimide inactivation and by binding of an analog suggested, but did not prove, that there was only one specific, active ATP binding site below 0.5 mm. These results are interpreted to be consistent with either of two mechanisms for ATP cooperativity of the Ca2+-stimulated ATPase activity of sarcoplasmic reticulum: (a) a single, high affinity ATP active site and a second, lower affinity “allosteric” activator site; or (b) a single ATP site which demonstrates two affinities through some kinetic mechanism such as a substrate-induced, slow transition.  相似文献

Abstract: Activation of the Ca2+/Mg2+ ATPase associated with highly purified Torpedo synaptic vesicles results in 45Ca2+ uptake. The accumulated 45Ca2+ is released by hypoosmotic buffer and by the Ca2+ ionophore A23187. Density-gradient centrifugation and permeation chromatography reveal that vesicular acetylcholine and the membrane-bound 45Ca2+ co-migrate, thus implying that 45Ca2+ is transported into cholinergic vesicles. ATP-dependent 45Ca2+ uptake follows saturation kinetics, with KmCa2+= 50 μM, KmATP= 5 μM, and Vmax= 3 ± 0.3 nmol Ca2+/mg protein/min. Treatment of the vesicles with mersalyl, dicyclohexyl-carbodiimide, and quercetin leads to inactivation of the Ca2+/Mg2+ ATPase and to comparable inhibition of 45Ca2+ transport. Ruthenium red and ouabain have no effect on either of these activities. Nigericin in the presence of external K+ is a potent inhibitor of 45Ca2+ translocation, whereas gramicidin activates transport. The proton translocator carbonylcyanide p-trifluoromethoxy-phenylhydrazone (FCCP) and FCCP + the ionophore valinomycin partially inhibit 45Ca2+ transport. By contrast, the above ionophores do not affect Ca2+/Mg2+ ATPase activity. Tentative mechanisms for ATP-dependent Ca2+ transport into cholinergic synaptic vesicles and the physiological significance of this process are discussed.  相似文献

The Ca2+/Mg2+ ATPase of the rat heart sarcolemmal particles was solublized with Triton X-100 after treating the membranes with trypsin and purified by high speed centrifugation, ammonium sulfate fractionation, hydrophobic chromatography and gel filtration. The purified enzyme was seen as a single protein band in nondenaturing polyacrylamide gel electrophoresis and its molecular weight by gel filtration was found to be about 240000. The enzyme utilized Ca-ATP or Mg-ATP as a substrate with high affinity sites (Km = 0.12 – 0.16 mM) and low affinity sites (Km = 1 mM). The enzyme also utilized CTP, GTP, ITP, UTP and ADP as substrates but at a lower rate in comparison to ATP. The enzyme was activated by Ca2+ (Ka = 0.4 mM) and Mg2+ (Ka = 0.2 mM) as well as by other cations in the order Ca2– > Mg2+ > Mn2+ > Sr2+ > Ba2+ > Ni2+ > Cu2+. The ATPase activity in the presence of Ca2+ was markedly inhibited by Mg2+, Mn2+, Ni2+ and Cu2+ whereas the monovalent cations such as Na+ and K+ were without effect. The enzyme did not exhibit Ca2+ stimulated Mg2+ dependent ATPase activity and was insensitive to calmodulin, ouabain, verapamil, D-600, oligomycin, azide and vanadate. Optimum pH for Ca2+ or Mg2+ ATPase activity was 8.5 – 9.0. In view of the possible ectoenzyme nature of the ATPase, its role in adenine nucleotide and Ca2+ metabolism in the myocardium is discussed.  相似文献

Ca2+-sensitive Mg2+-dependent ATP phosphohydrolase (EC 3.6.1.3, ATPase) was extracted from the plain synaptic vesicle fractions that were virtually devoid of contamination. The protein pattern of the ATPase preparation on SDS polyacrylamide gel electrophoresis closely resembled that of actomyosin from skeletal muscle. The finding suggests that the main components of the ATPase are actin- and myosin-like proteins of the brain (stenin and neurin). Microsome and synaptosomal plasmalemma fractions were extracted under the same conditions to examine the possibility that the ATPase extracted derived from contaminating particulates. An entirely different ATPase was extracted from microsomes, and no protein from plasma membranes. Although Ca2+-sensitive Mg2+-dependent ATPase was extracted from coated vesicle fraction, the electrophoretic pattern was dissimilar to that of the ATPase from plain synaptic vesicle fractions. It may be inferred that the whole complex of neurostenin is located in plain synaptic vesicles from the brain.  相似文献

The plasma membrane was isolated from a calcareous red alga, Serraticardia maxima (Yendo) Silva (Corallinaceae), by aqueous two-phase partitioning. Its purity was examined with marker enzymes, Mg2+-dependent ATPase, inosine diphosphatase, cytochrome c oxidase and NADH-cytochrome c reductase, as well as the sensitivity of Mg2+-dependent ATPase to vanadate, azide and nitrate. The results showed that the isolated plasma membrane was purified enough to study its functions. Electron microscopic observations on thin tissue sections revealed that most vesicles of the isolated plasma membrane were stained by the plasma membrane specific stain, phosphotungstic acid-chromic acid. Mg2+- or Ca2+-dependent ATPases were associated with the plasma membrane. Ca2+-dependent ATPase was activated at physiological cytoplasmic concentrations of Ca2+ (0.1–10 μmol/L). However, calmodulin (0.5 μmol/L) did not affect its activity. The pH optimum was 8.0, in contrast to 7.0 for Mg2+-dependent ATPase. The isolated plasma membrane vesicles were mostly right side-out. To test for H+-translocation, right side-out vesicles were inverted; 27% of vesicles were inside-out after treatment with Triton X-100. The inside-out plasma membrane vesicles showed reduction of quinacrine fluorescence in the presence of 1 mmol/L ATP and 100 μmol/L Ca2+. The reduced fluorescence was recovered with the addition of 10 mmol/L NH4Cl, or 5 μmol/L nigericin plus 50 mmol/L KCl. UTP and CTP substituted for ATP, but ADP did not. Ca2+-dependent ATPase might pump H+ out in the physiological state. The acidification by this pump might be coupled with alkalinization at the calcifying sites, which induces calcification.  相似文献

Terbium (Tb3+) binding to skeletal muscle troponin C was studied by fluorescence spectroscopy and circular dichroism. Titrations indicate that Tb3+, like Ca2+, preferentially binds to the two high affinity Ca2+-Mg2+ sites (III and IV) inducing structural changes similar to those induced by Ca2+. Tb3+ readily displaces Ca2+ from these sites suggesting a K(Tb3+) ≥ 109 M?1 In 6 M urea, both Ca2+ and Tb3+ bind preferentially to a single site on troponin C. The effect of morphine on ATPase of synaptic plasma membranes (SPM) and synaptic vesicles isolated from the mouse brain was studied. The activity of synaptic vesicle Mg++-dependent ATPase from mice rendered morphine tolerant and dependent by pellet implantation was 40% higher than that from placebo implanted mice. However, the activities of Mg++-dependent ATPase and Na+, K+ activated ATPase of SPM of tolerant and nontolerant mice were not significantly different. The activity of synaptic vesicular Mg++-dependet ATPase was dependent on the concentration of Mg++ but not of Ca++; maximum activity was obtained with 2 mM MgCl2. On the other hand, Mg++-dependent ATPase activity of SPM was dependent on both Mg++ and Ca++, activity being maximum using 2 mM MgCl2 and 10?5 M CaCl2. It is suggested that this stimulation of ATPase activity may alter synaptic transmission and may thus be involved in some aspects of morphine tolerance and dependence.  相似文献

  • 2.2. Both Ca2+-dependent ATPase and Mg2+-dependent ATPase activities were decreased in the DOC-treated membranes; however, the depression of Mg2+-dependent ATPase activity was greater than that of Ca2+-dependent ATPase.
  • 3.3. The differential changes in Ca2+-dependent ATPase and Mg2+-dependent ATPase activities were apparent when incubations with DOC were carried out for different time intervals and at different temperatures.
  • 4.4. In DOC-treated preparations, the Km value for Ca2+-dependent ATPase was decreased whereas that for Mg2+-dependent ATPase was increased. The half maximal velocities of the Ca2+-dependent ATPase and Mg2+-dependent ATPase enzyme reactions in the treated preparations were obtained at a DOC: membrane protein ratio of 3.0 and 0.6, respectively.
  • 5.5. In the DOC-treated membranes exhibiting the half maximal velocities of enzyme reactions, the Ki value for Ca2+-dependent ATPase was drastically reduced but remained unchanged for Mg2+-dependent ATPase.
  • 6.6. The DOC treatment was associated with a loss of protein as well as phospholipids and resulted in changes in the ultrastructural integrity of the membrane.
  • 7.7. Varying degrees of decreases in the activities of sarcolemmal adenylate cyclase. (Na-K+)-ATPase. 5'-nucleotidase and calcium binding were seen upon DOC treatment.
  • 8.8. The extent of reduction in Ca2+-dependent ATPase and Mg2+-dependent ATPase activities were also different when the membrane was treated with a non-ionic detergent, Lubrol PX.
  • 9.9. These data suggest that Ca2+-dependent ATPase in heart sarcolemma is more resistant than Mg2+-dependent ATPase to detergent treatments and further indicate some differences in the properties of these enzymes.
