The effects of heat shock (HS); UV-B irradiation; and the consecutive action of these factors on the growth, development, and water supply of seven-day-old melon (Melo sativusSager., cv. Torpeda) seedlings were investigated. Depending on the HS severity, we observed growth stimulation (after treatment at 45°C for 1 h), growth retardation (after treatment at 45°C for 2 h or at 48°C for 1 h), or complete growth inhibition and cell death (after treatment at 45°C for 3 h or at 55°C for 1 h). UV-B irradiation (18.3 kJ/(m2h)), depending on its duration, stimulated (5–10 min), retarded (60 min), or resulted in complete growth inhibition and plant death (90 min). HS treatment (at 45°C for 1 h) prior to UV-B irradiation (for 1 h) favorably affected both the growth and water balance of seedlings. Apparently, the HS pretreatment increases the tolerance of seedlings to high doses of UV-B radiation. 相似文献

Synthesis of stress proteins after heat shock and different periods of UV-B radiation were investigated with marine diatom species from the North Sea Ditylum brightwellii, Lithodesmium variabile, Odontella sinensis, Thalassiosira rotula and the Antarctic diatom Odontella weissfloggii from the Weddell Sea. Algae were grown in an artifical sea-water medium under controlled laboratory conditions: light/dark regime of 12:12 h (7.2 W m?2), normal air (0.035 vol.% CO2) and 18° or 4 °C. All the tested diatom species can produce heat shock proteins (HSPS) of the 70 kDa family by in vivo labelling with [35S]-methionine. The same results were obtained for Odontella sinensis, Ditylum brightwellii and Odontella weissflogii by estimation of the in vitro translation products with poly-A-mRNA isolated from these organisms. However, Odontella weissflogii, a species relatively insensitive to UV-B irradiance, did not synthesize UV-induced HSPS, whereas the UV-sensitive diatom Odontella sinensis, as well as Lithodesmium variabile, produced all the observed HSPS after UV-B exposure. In addition, a protein of 43 kDa was found after UV-B irradiance of the temperate Odontella sinensis. The temperate marine diatom Thalassiosira rotula synthesized 70 kDa and 5 7 kDa proteins after a heat shock and a UV-B exposure of 2 h, but a 40 kDa protein could not be detected, whereas a 60 kDa protein was found after 2 h UV-B exposure. The results are discussed in view of a possible adaptation of O. weissflogii to an enhanced UV dose. 相似文献

An increase in ultraviolet(UV)B radiation on the earth's surface is a feature of current global climate changes.It hasbeen reported that alternative oxidase(AOX)may have a protective role against oxidative stress induced by environmentalstresses,such as UV-B.To better understand the characteristic tolerance of plants to UV-B radiation,the effects ofenhanced UV-B radiation on the activity and expression of AOX in red kidney bean(Phaseolus vulgaris)leaves wereinvestigated in the present study.The results show that the total respiration rate and AOX activity in red kidney bean leavesincreased significantly during treatment with enhanced UV-B.However,cytochrome oxidase(COX)activity did not changesignificantly.The H_2O_2 content was also markedly increased and reached a maximum of 4.45 mmol.L~(-1)·g~(-1)DW(dry weight)at 24 h of UV-B treatment,before dropping rapidly.Both alternative pathway content and alternative pathway activity wereincreased in the presence of exogenous H_2O_2.Immunoblotting analysis with anti-AOX monoclonal antibody revealed thatexpression of the AOX protein increased in red kidney bean leaves under enhanced UV-B radiation,reaching a peak at 72h.In addition,AOX expression in red kidney bean leaves was induced by exogenous H_2O_2.These data indicate that theincrease in AOX activity in red kidney bean leaves under enhanced UV-B radiation was mainly due to H_2O_2-induced AOXexpression. 相似文献

The responses of 14-day-old Arabidopsis thaliana (L.) Heynh. plants to UV-B irradiation (280–320 nm) and ABA treatment were investigated. Wild-type plants as well as ethylene-insensitive etr1-1 and ctr1-1 mutants were used. Theetr1-1 mutant considerably differed from the ctr1-1 one in the fresh weight production after UV-B treatment (29.5 kJ/m2). The irradiated etr1-1 plants fell well behind the nonirradiated ones during the first two days after stress, but by the 8th day, their weight attained 70% of control plant weight. In contrast, Ctr1-1 mutant weight comprised 70% of control level after two days of stress but, by the 8th day, it was only 56% of the weight of control plants. In wild-type and ctr1-1 plants, ABA, in the 8 × 10–6 to 2 × 10–4 M concentration range, increased the difference between the weights of nonirradiated and irradiated plants, but in etr1-1 plants, ABA decreased this difference. The etr1-1, ctr1-1, and wild-type plants were very similar in the dynamics of ethylene evolution after UV-B treatment (7.4 kJ/m2). In wild-type, etr1-1, and ctr1-1 plants, ABA, in a concentration-dependent manner, inhibited UV-B-induced ethylene evolution to the same extent. The results obtained show that ABA exerted an opposite effect on UV-B-dependent growth in the plants with active (wild type and ctr1-1) and blocked (etr1-1) ethylene signal pathway, whereas the inhibition of ethylene synthesis by ABA was not related to ethylene signal transmission. 相似文献

The results are generalized of many-year studies into the adaptive role of heat shock proteins in different animals, including the representatives of cold- and warm-blooded species that inhabit regions with different thermal conditions. Adaptive evolution of the response to hyperthermia can lead to different results depending on the species. The thermal threshold of induction of the heat shock proteins in desert thermophylic species is, as a rule, higher than in the moderate climate species. In addition, thermoresistant species are often characterized by a certain level of heat shock proteins in cells even at a physiologically normal temperature. Although adaptation to hyperthermia is achieved in most cases without changes in the number of heat shock genes, they can be amplified in some cases in termophylic species. The parallel synthesis of heat shock proteins and trehalose in response to heat shock did not allow the role of these compounds in the acquisition of thermotolerance by fungal cells to be established for a long time. This review analyses experimental data obtained with the use of mutant fungal strains and shows differences in the thermoprotective functions of trehalose and heat shock proteins in relation to cell membranes and macromolecules. Bean (Phaseolus vulgaris L. cv. Berbukskaya) seedlings were pre-treated with choline compounds, 19 mM 2-ethyltrimethylammonium chloride (Ch) or 1.6 mM 2-chloroethyltrimethylammonium chloride (CCh), during 24 h, then after 6 d the excised primary leaves were exposed to UV-B and high temperature stress. Chlorophyll (Chl) fluorescence, delayed light emission, accumulation of photosynthetic pigments, contents of thiobarbituric acid reactive substances, and activities of the active oxygen detoxifying enzymes (superoxide dismutase, ascorbate peroxidase, and glutathione reductase) were examined. Pre-treatment of plants with Ch or CCh enhanced the resistance of photosystem 2 (PS2) photochemistry to UV-B and heat injuries. The higher stress resistance can be explained by the increased activity of the detoxifying enzymes. The increased content of UV-B-absorbing pigments may also contribute to the enhanced resistance of choline-treated plants to UV-B radiation. 相似文献

Cucumis sativus L.) seedlings were irradiated or not irradiated with UV-B for several days in environment-controlled growth chambers. The
first leaves irradiated with UV-B were retarded in growth but simultaneously acquired a remarkably high tolerance to oxidative
stress, as induced by paraquat treatment, compared with the non-irradiated leaves. This enhanced tolerance was observed within
1d after the start of UV-B irradiation and was maintained during the 12 d period of UV-B treatment. The effects of UV-B on
several antioxidative enzymes were examined, and activities of superoxide dismutase, ascorbate peroxidase and guaiacol peroxidase,
but not of glutathione reductase, were found to be enhanced. However, activation of these enzymes occurred only from 6 d after
the start of irradiation. In contrast, accumulation of phenolic compounds was observed within 1d after the start of UV-B irradiation.
HPLC analysis of phenolic compounds showed the distinct enhancement of a substance, which may have antioxidative properties
in cucumber seedlings irradiated with UV-B. On the basis of these results, we conclude that not only antioxidative enzymes
but also other factors in cucumber seedlings irradiated with UV-B, such as phenolic compounds, may participate in the enhanced
tolerance to oxidative stress.

在CO2浓度分别为当今CO2浓度(360 mL/L)和加富浓度(5 000 mL/L)条件下,研究了UV-B胁迫对亚心形扁藻(Platymonas subcordiformis (Wille) Hazen)的光合作用、膜脂过氧化和抗氧化酶活性的影响。实验结果表明:(1) UV-B单独作用下,亚心形扁藻的干重、光合速率、叶绿素a (Chl a)和类胡萝卜素(Car.)含量显著降低,CO2加富单独作用下,亚心形扁藻的干重和光合速率显著升高,叶绿素a和类胡萝卜素含量与对照相比没有显著变化,而UV-B与CO2共同作用则使亚心形扁藻的干重和光合速率与对照相比没有显著变化,叶绿素a和类胡萝卜素含量显著降低。(2) UV-B单独作用和CO2加富单独作用都使可溶性蛋白含量显著降低,UV-B与CO2共同作用下的可溶性蛋白含量比UV-B单独作用的要高。高CO2对藻的可溶性蛋白含量的变化在很大程度上归因于Rubisco蛋白的降低。(3)UV-B单独作用下,O2-. 产生速率、H2O2 含量和MDA含量显著升高,而CO2加富单独作用下,O2-. 产生速率、H2O2 含量和MDA含量显著降低,与UV-B单独作用相比,UV-B与CO2共同作用使O2-. 产生速率、H2O2 含量和MDA含量显著降低。说明CO2加富可以减少活性氧对亚心形扁藻的氧化胁迫,同时减少UV-B对亚心形扁藻的膜脂过氧化伤害。(4) UV-B单独作用下,SOD、POD、CAT、GR和GPX活性显著升高,高CO2 相似文献

以增强UV-B(10.08 kJ.m-2.d-1)辐射后的小麦根尖细胞为材料,采用间接免疫荧光标记技术,利用激光共聚焦扫描显微镜,观察分析小麦根尖分裂期细胞Ran蛋白在分裂周期的分布及形态变化。研究结果显示,正常细胞中,Ran蛋白在细胞分裂间期主要定位于核膜周边,在后期定位于赤道板上和纺锤体上,末期又回到子细胞核膜周边;增强UV-B辐射处理后,在细胞分裂间期和前期有点状荧光分布在核膜的周围;中期和后期点状荧光分布在细胞质中;在末期部分点状荧光又回到核膜的周围,部分仍分散在核内,且出现落后染色体、染色体桥、不均等分裂等染色体畸变类型和异常分裂现象。 相似文献

The marine algal species in the Baltic Sea are few due to the low sea water salinity. One of the few species that can be found is Fucus vesiculosus. Even this species is affected by the low salinity and becomes smaller in size in the Baltic. In present work the photosynthesis of F. vesiculosus in the northern Baltic (Bothnian Sea) was compared to the photosynthesis of F. vesiculosus in the Atlantic. Oxygen evolution was measured before and after exposure to 2.3 W of UV-B (280–320 nm) radiation for 5 h, as well as after 48 h recovery in low light. The plants were kept in their own sea water salinity as well as in a changed salinity, this to examine possible correlations between salinity and photosynthesis. The results show a significant higher initial maximal photosynthesis (P max) for Atlantic plants (10.3 nmol O2 g−1 FW s−1) compared to Baltic plants (4.0 nmol O2 g−1 FW s−1). The Baltic plants were found more sensitive to UV-B with a 40–50% decrease of P max as well as a lower degree of recovery (60–70% compared to 75–95% for the Atlantic plants). The higher salinity (35 psu) had a positive effect on the Baltic F. vesiculosus with increased P max as well as increased tolerance to UV-B. The lower salinity (5 psu) had a negative effect on the Atlantic plants with a decreased P max as well as a lower tolerance to UV-B. Pigment content was found higher in Atlantic F. vesiculosus. The pigment content decreased then the Atlantic plants were transferred to 5 psu. The concentration of Chl a as well as the total content of violaxanthin, diadinoxanthin and zeaxanthin in Baltic plants increased when transferred to 35 psu. The Atlantic F. vesiculosus can not survive the low salinity in the northern Baltic (died within 8 weeks). It is likely that a long time acclimation or adaptation to low salinity has taken place for F. vesiculosus in northern Baltic. The shoots of 16-day-old spring wheat plants (Triticum aestivumL., cv. Albidum 29) were subjected to heat shock (HS) at 40, 41, or 43°C for 10 min. The activity of the Hill reaction in chloroplasts isolated immediately after HS was 83, 61, and 30% of the initial value, respectively. The activity of the Hill reaction was also estimated after plant return to the initial growth conditions for one day. It was completely restored after heating at 40°C and achieved 82 and 30–33% of the initial level after heating at 41 and 43°C, respectively. Thereafter, the shoots were heated repeatedly at 42, 43, or 43.5°C for 10 min, and the activity of the Hill reaction was measured immediately or one day after this heating. Immediately after the second heating, the activity decreased again as compared to its value before heating. The percent of inhibition of the Hill reaction was similar in the control plants not subjected to preliminary HS and HS-treated plants independently of the temperature used. However, after one-day growth under normal conditions, the activity of the Hill reaction was restored almost completely in HS-treated plants but not more than by 10% in the control plants. The conclusion is that different mechanisms underlie the development of the tolerance to HS and recovery. Some plants were tested for the effect of HS (40°C) on their tolerance to photoinhibition. The degree of the Hill reaction inhibition after plant exposure to the light of 65–75 klx for 3 h was essentially similar in detached leaves from the HS-treated and unheated plants and comprised about 40% of the activity before light stress. After the leaves were returned to the low-light conditions for 3 h, the Hill reaction was restored and attained about 75% of that before photoinhibition in both HS-treated and untreated plants. The lack of the HS-induced stimulation of the Hill reaction recovery after photoinhibition is evidently related to the fact that heating and excess light damage different sites of photosystem II, which implies the different pathways for the recovery of its functional activity. 相似文献

Studies on the stress resistance of insect-pathogenic fungi are very important to better understand the survival of these organisms in the environment. In this study, we examined the cold activity (8 ± 1 °C for 7 days), UV-B tolerance (Quaite-weighted UV-B irradiance at 847.90 mW m−2 for 1, 2, 3, and 4 h), and wet-heat tolerance (45 °C for 1, 2, 3, and 4 h) of two isolates of Tolypocladiumcylindrosporum (ARSEF 3392 and 5558), one isolate of Tolypocladium geodes (ARSEF 3275), and two isolates of Tolypocladium inflatum (ARSEF 4772 and 4877) based on their germination, compared with Metarhizium robertsii (ARSEF 2575). After 3 h of UV-B exposure, T. cylindrosporum germinated at a greater rate than the other Tolypocladium species and had similar viability to that of the M. robertsii. Most Tolypocladium isolates, however, were less UV-B tolerant than M. robertsii. The T.cylindrosporum isolates were also the most thermotolerant, with similar tolerance to the M. robertsii. The isolates of T. inflatum and T. geodes, which had similar heat tolerance, were the least heat tolerant compared with the isolates of T. cylindrosporum and M. robertsii. After 4 h of heat exposure, the germination of T. inflatum and T. geodes isolates was not significantly different. For cold activity, both T.cylindrosporum isolates germinated to ca. 100% in only 3 days. Approximately 50% of the two T. inflatum isolates germinated, and less than 5% of T. geodes germinated after 3 days. All fungal isolates, however, completely germinated by the seventh day, except M.robertsii. The isolates of T. cylindrosporum, therefore, were the most heat and UV-B tolerant, and had the highest cold activity compared to the other species. 以成熟期不同的两个番茄品种同辉(早熟型)和霞光(晚熟型)为试材,模拟兰州地区12%和20%臭氧层减薄时增强的UV- B辐射(分别为T1=2.54 KJ·m-2·d-1和T2=4.25 KJ·m-2·d-1),研究了大田条件下增强UV-B辐射对其花粉生活力的影响以及雄蕊中4种内源激素(IAA、GAs、ZR和ABA)和多胺(Put、Spm和Spd)及脯氨酸的变化。结果表明:辐射抑制了同辉的花粉萌发和花粉管伸长,但只降低了霞光的花粉萌发率;两种辐射明显降低了两品种番茄雄蕊中的GAs含量,同时造成同辉雄蕊中Put和Spd含量明显增加,Spm和Put/Spd+Spm比值显著降低;霞光中3种多胺含量都显著减少,导致高辐射时Put/Spd+Spm比值上升;同辉番茄雄蕊的脯氨酸含量不受影响,但高辐射使霞光番茄雄蕊的脯氨酸含量降低。实验表明,两品种番茄花粉生活力的变化与增强UV-B辐射下雄蕊中GAs水平、Spm含量以及脯氨酸含量的减少有关。雄蕊中多胺和脯氨酸含量变化对UV- B辐射的响应说明霞光品种对UV-B辐射更敏感。 相似文献
