The crystalline and microstructure of the regenerated cellulose fibers prepared from different solvents and technology processes were investigated by synchrotron wide-angle X-ray diffraction (WAXD) and small-angle X-ray scattering (SAXS). WAXD results indicated that the crystal orientation, crystallinity of Lyocell and IL-cell fibers were higher than those of Viscose and Newdal fibers. The size of micro-voids located in the cross-section of regenerated cellulose fibers was analyzed based on the results of SAXS. And the technology process had little effect on the radius of the micro-voids. The micro-voids in Viscose and Newdal fibers have longer length (L) and greater misorientation (BΦ) than that in Lyocell and IL-cell fibers. This reveals that the average void volumes of Viscose and Newdal fibers were larger. Furthermore, the regenerated cellulose fibers from dry-jet-wet-spinning process exhibited completely a higher E-modulus, tenacity than the fibers spun by wet-spinning method did.  相似文献

Untreated cellulose was directly and quickly dissolved in NaOH/thiourea/urea aqueous solution. The mechanism of dissolution was investigated by SEM, WXRD and (13)C NMR. The components of this solvent cannot dissolve cellulose on their own, and the interactions between NaOH and urea, as well as between NaOH and thiourea, play an important role in improving the dissolution of cellulose. Moreover, (13)C NMR spectra proved that NaOH, thiourea, and urea were bound to cellulose molecules, which brings cellulose molecules into aqueous solution to a certain extent and prevents cellulose macromolecules from associating. (13)C NMR spectra of the cellulose solution show that this novel mixture is a direct solvent. Optical microscopy and CP MAS (13)C NMR were used to study the process of dissolution. The results reveal that cellulose is dissolved completely and that cellulose I (cotton linter) first changes to amorphous cellulose chains in solution, and then to cellulose II during regeneration. Moreover, a new, more effective dissolution method is proposed, as confirmed by dynamic rheology measurements.  相似文献

Molecular morphologies and conformation transition of lentinan, a beta-(1-->3)-D-glucan from Lentinus edodes, were studied in aqueous NaOH solution by atomic force microscopy (AFM), viscometry, multiangle laser light scattering, and optical rotation measurements. The results revealed that lentinan exists as triple-helical chains and as single random-coil chains at NaOH concentration lower than 0.05M and higher than 0.08M, respectively. Moreover, the dramatic changes in weight-average molecular weight Mw, radius of gyration [s2](1/2), intrinsic viscosity [eta], as well as specific optical rotation at 589 nm [alpha]589 occurred in a narrow range of NaOH concentration between 0.05 and 0.08M NaOH, indicating that the helix-coil conformation transition of lentinan was carried out more easily than that of native schizophyllan and scleroglucan, and was irreversible. For the first time, we confirmed that the denatured lentinan molecule, which was dissolved in 0.15M NaOH to be disrupted into single coil chains, could be renatured as triple helical chain by dialyzing against abundant water in the regenerated cellulose tube at ambient temperature (15 degrees C). In view of the AFM image, lentinan in aqueous solution exhibited the linear, circular, and branched species of triple helix compared with native linear schizophyllan or scleroglucan.  相似文献

NaOH/urea aqueous solution has been used as a solvent for chitin for the first time. Effects of this solvent composition and temperature on the solubility and stability of chitin solution were studied with an optical microscope, from which 8 wt% NaOH/4 wt% urea concentrations were deduced as suitable and −20 °C as the appropriate temperature. The original and regenerated chitin were characterized by viscosity, elemental analysis, FI-IR and X-RD analysis, and the effect of solvent composition and temperature on chitin structure was investigated. It was inferred that 8 wt% NaOH/4 wt% urea solvent under low temperature adventitiously has little effect on chitin structure and the urea is of benefit to the stability of chitin solution. In addition, the rheological properties suggested that chitin aqueous solution in high concentration is a pseudoplastic fluid and that chitin aqueous solution in low concentrations is a Newtonian fluid. This chitin aqueous solution is sensitive to temperature and will transform it to a gel when temperature increases.  相似文献

The aim of this study was to develop a new inorganic-organic hybrid film. Nanohydroxyapaptite (nHAP) particles as the inorganic phase was mixed with cellulose in 7 wt.% NaOH/12 wt.% urea aqueous solution with cooling to prepare a blend solution, and then inorganic-organic hybrid films were fabricated by coagulating with Na2SO4 aqueous solution. The structure and properties of the hybrid films were characterized by high resolution transmitting electron microscopy (HRTEM), field emission scanning electron microscopy (FESEM), thermo-gravimetric analysis (TGA), Fourier transform infra-red (FT-IR) spectra, wide angle X-ray diffraction (WAXD) and tensile testing. The results revealed that the HAP nanoparticles with mean diameter of about 30 nm were uniformly dispersed and well immobilized in the hybrid film as a result of the role of the nano-and micropores in the cellulose substrate. A strong interaction existed between HAP and cellulose matrix, and their thermal stability and mechanical strength were improved as a result of good miscibility. Furthermore, the results of 293T cell viability assay indicated that the HAP/cellulose films had excellent biocompatibility and safety, showing potential applications in biomaterials.  相似文献

Glucose isomerase (D-xylose ketol-isomerase, EC 5.3.1.5.) catalyses the isomerization reaction of glucose and xylose. The small angle X-ray scattering (SAXS) data of glucose/xylose isomerase from Streptomyces rubiginosus were recorded for protein solution using synchrotron radiation. The experimental data were compared with theoretical scattering calculated on the basis of the known crystal structure (PDB code: 1OAD). The radius of gyration measured by SAXS (R(G)=3.30 nm) was almost identical and the maximum dimension in the distance distribution function was by about 2.5 % lower than the corresponding values calculated on the basis of the crystal structure.  相似文献

The small angle X-ray scattering (SAXS) data of xylanase XYNII (endo-1,4-beta-xylan xylanohydrolase EC 3.2.1.8) from Trichoderma longibrachiatum, an enzyme catalysing the reaction of accidental hydrolysis of beta-1,4-D-xylosidic linkages of xylan, were recorded for protein solution using synchrotron radiation. The experimental data were compared with those of theoretical scattering calculated on the basis of the known crystal structure. Microcomposite fibers of regenerated silk fibroin (RSF) and multiwalled carbon nanotubes (MWNTs) were successfully prepared by an electrospinning process from aqueous solutions. A quiescent blended solution and a three-dimensional Raman image of the composite fibers showed that functionalized MWNTs (F-MWNTs) were well dispersed in the solutions and the RSF fibers, respectively. Raman spectra and wide-angle X-ray diffraction (WAXD) patterns of RSF/F-MWNT electrospun fibers indicated that the composite fibers had higher β-sheet content and crystallinity than the pure RSF electrospun fibers, respectively. The mechanical properties of the RSF electrospun fibers were improved drastically by incorporating F-MWNTs. Compared with the pure RSF electrospun fibers, the composite fibers with 1.0 wt % F-MWNTs exhibited a 2.8-fold increase in breaking strength, a 4.4-fold increase in Young's modulus, and a 2.1-fold increase in breaking energy. Cytotoxicity test preliminarily demonstrated that the electrospun fiber mats have good biocompatibility for tissue engineering scaffolds.  相似文献

The weight-average molecular weight (Mw) and intrinsic viscosity ([eta]) of the alpha-(1-->3)-D-glucan (L-FV-II) from Lentinus edodes in 0.5 and 1.0 M NaOH aqueous solution containing urea, were studied by light scattering and viscometry. The Mw value of the glucan decreased with increase of the urea and NaOH concentration. A strong intermolecular hydrogen bonding confers water-insolubility on the glucan, but NaOH and especially urea, broke this hydrogen bonding leading to enhanced water-solubility. Use of 1.0 M urea-1.0 M NaOH as solvent broke not only intermolecular hydrogen bonds but also partial covalent bonds of the alpha-glucan in aqueous solution, resulting in a decrease of Mw and [eta]. The urea and NaOH concentrations, storage time with stirring, and mode of preparation of the polysaccharide in aqueous solution significantly affected the determination of Mw and [eta]. The dependences of specific rotation and fluorescence emission ratio of a probe on urea concentration showed that a change in the molecular conformation of the alpha-glucan in 0.5 M NaOH aqueous solution containing urea occurred in the range 0.4-0.6 M urea. The 0.5 M urea-0.5 M NaOH aqueous solution is a suitable solvent for the glucan, and the Mw and [eta] values obtained were 5.21 x 10(5) and 148 cm3 g(-1), respectively. Degradation of the glucan was obvious after storage for 15 months.  相似文献

The gelatinization of waxy rice, regular rice, and potato starch suspensions (66% w/w moisture) was investigated by real-time small-angle X-ray scattering (SAXS) and wide-angle X-ray diffraction (WAXD) during heating and by fast ramp differential scanning calorimetry (DSC). The high-angle tail of the SAXS patterns suggested the transition from surface to mass fractal structures in the DSC gelatinization range. Amylose plays a major role in determining the dimensions of the self-similar structures that develop during this process as the characteristic power-law scattering behavior extends to lower scattering angles for regular than for waxy starches. Crystallinity of A-type starches is lost in the temperature region roughly corresponding to the DSC gelatinization range. At the end of the gelatinization endotherm, the B-type potato starch showed residual crystallinity (WAXD), while SAXS-patterns exhibited features of remaining lamellar stacks. Results indicate that the melting of amylopectin crystallites during gelatinization is accompanied by the (exothermic) formation of amorphous networks.  相似文献

The determination of the crystal orientation of regenerated cellulose fibers produced under different drawing regimes is presented. Orientation is determined by using wide-angle X-ray diffraction from a synchrotron source and by measuring the azimuthal width of equatorial reflections. The orientation parameter theta is then determined to compare fiber samples. By using a 500 nm beam size, clear differences between the crystal orientations of the skin and the core of the fibers are reported for a range of differently processed fibers for the first time. These results are shown to have implications for the mechanical properties of regenerated cellulose fibers. By applying tensile deformation to fiber bundles it is shown that the most misoriented samples undergo rapid decreases in the orientation parameter, which is an indication of crystal reorientation. However, the more highly oriented fibers undergo little reorientation. An average shear modulus for these fibers is determined by placing the data on a master curve and fitting with a model equation. By using another model for the fibers of low orientation and the shear modulus from the master curve analysis, it is shown that the deformation of less oriented fibers is dominated by shear between crystals, whereas the more oriented filaments are likely to undergo more significant chain deformation. By using a new model for fibers of low orientation, a parameter ksigma is introduced that gives the proportion of the fiber stress that is due to crystal shear. Systematic differences between this parameter for fibers of increasing initial orientation are reported. Moreover it is shown that the fibers of initially lower average orientation are governed by uniform strain, in agreement with the new model, whereas more highly oriented fibers deform under uniform stress. Furthermore, the model that we propose for misoriented domains and the use of a new factor dictating the proportion of shear stress may have general applications in materials engineering.  相似文献

We describe the capillary flow behavior of gels of beta-lactoglobulin (beta-lg) containing droplets of fibrils and the shear flow alignment of beta-lg fibers in dilute aqueous solutions. Polarized optical microscopy and laser scanning confocal microscopy are used to show that capillary shear flow does not affect the fibril droplet sizes in the beta-lg gels, the system behaving in this respect as a solution of compact colloidal particles under shear flow. Small-angle X-ray scattering (SAXS) on dilute aqueous solutions indicates that the fibers can be initially aligned under capillary shear, but this alignment is lost after 18 min of shear. Transmission electron microscopy experiments on the samples studied by SAXS suggest that the loss of orientation is due to a shear-induced breakup of the swollen fibril network. Dynamic and static light scattering on dilute beta-lg fibril aqueous solutions are used to show that before shear beta-lg fibrils behave as strongly interacting semiflexible polymers, while they behave as weakly interacting rods after 18 min of capillary shear.  相似文献

Uniaxially oriented films with high tensile strength were processed from ultrahigh-molecular-weight poly[(R)-3-hydroxybutyrate] (P(3HB)) by a method combining hot-drawing near the melting point of the polymer and two-step-drawing at room temperature. In a two-step-drawn and subsequently annealed film, P(3HB) molecular chains fall into two states: 2/1 helix (alpha-form) and planar zigzag (beta-form) conformations. The mechanism for generating the beta-form during two-step-drawing was investigated by time-resolved synchrotron wide- and small-angle X-ray scattering measurements (WAXD and SAXS), together with the measurement of stress-strain curves. It was found that the improvement of mechanical properties is due to not only the orientation of molecular chains but also the generation of the beta-form during the drawing. The crystal and molecular structures of the alpha-form remained unchanged until the yield point of the stress-strain curve. At the yield point, the long period obtained from SAXS doubled and a new reflection indicative of the beta-form was observed on the equatorial line in WAXD. The intensity of the reflection from the beta-form increased with an increase in the two-step-drawing ratio at room temperature. The SAXS pattern changed from a two-point reflection along the meridian to a cross pattern with streaking on the equatorial line, demonstrating the close alignment of shish-kebab structures. The reflection intensity, crystal orientation and crystal size of the alpha-form decreased during two-step-drawing. Based on these results, the beta-form is mainly introduced from the orientation of free molecular chains in the amorphous regions between alpha-form lamellar crystals, but the structural transformation of molecular chains also occurs from the alpha-form to the beta-form at the deformed lamellar crystals.  相似文献

The small angle X-ray scattering (SAXS) pattern of the homotetrameric asparaginase II from Escherichia coli was measured in solution in conditions resembling those in which its crystal form was obtained and compared with that calculated from the crystallographic model. The radius of gyration measured by SAXS is about 5% larger and the maximum dimension in the distance distribution function about 12% larger than the corresponding value calculated from the crystal structure. A comparison of the experimental and calculated distance distribution functions suggests that the overall quaternary structure in the crystal and in solution are similar but that the homotetramer is less compact in solution than in the crystal.  相似文献

Hydrogels with nanoscale structure were synthesized using amphiphilic poly(epsilon-caprolactone)-poly(ethylene oxide)-poly(epsilon-caprolactone) (PCL-b-PEO-b-PCL) triblock copolymers. Small-angle X-ray scattering (SAXS) studies show that the block copolymers form 30-40 nm structures in aqueous solution and that these patterns are retained, with some increase in length scale, following electron beam cross-linking. Lamellar nanostructures were observed by SAXS and atomic force microscopy (AFM), with SAXS indicating cylindrical structure as the block lengths become more different in length. It is demonstrated through Fourier transform infrared spectroscopy (FTIR), mass loss, and differential scanning calorimetry (DSC) that the PCL can be completely removed by hydrolysis in NaOH(aq) to form porous PEO hydrogels. These hydrogels retain active functional groups following PCL removal that serve as sites for further chemical modification.  相似文献

Solution blowing is an innovative process for spinning micro-/nano-fibers from polymer solutions using high-velocity gas flow as fiber forming driving force. Submicron-scale cellulose fibers were successfully solution blown by two improvement measures. First, cellulose solution was directly blown to fibers of 260-1900nm in diameter by raising the air temperature along the spinning line which was proved to accelerate the evaporation of solvent and fiber forming. Second, coaxial solution blowing technique was established with cellulose solution and polyethylene oxide (PEO) solution used as core and shell liquids, respectively. The core-shell structures of the fibers were examined by SEM and TEM. Cellulose fibers with diameter between 160nm and 960nm were further obtained after removing PEO shell. X-ray diffraction studies showed that the two kinds of submicron-scale cellulose fibers are mostly amorphous.  相似文献

Transparent and bendable regenerated cellulose films prepared from aqueous alkali (NaOH or LiOH)/urea (AU) solutions exhibit high oxygen barrier properties, which are superior to those of conventional cellophane, poly(vinylidene chloride), and poly(vinyl alcohol). Series of AU cellulose films are prepared from different cellulose sources (cotton linters, microcrystalline cellulose powder, and softwood bleached kraft pulp) for different dissolution and regeneration conditions. The oxygen permeabilities of these AU cellulose films vary widely from 0.003 to 0.03 mL μm m(-2) day(-1) kPa(-1) at 0% relative humidity depending on the conditions used to prepare the films. The lowest oxygen permeability is achieved for the AU film prepared from 6 wt % cellulose solution by regeneration with acetone at 0 °C. The oxygen permeabilities of the AU cellulose films are negatively correlated with their densities, and AU films prepared from solutions with high cellulose concentrations by regeneration in a solvent at low temperatures generally have low oxygen permeabilities. The AU cellulose films are, therefore, promising biobased packaging materials with high-oxygen barrier properties.  相似文献

The conformation of polysaccharide PGG-Glucan, isolated from yeast cell walls, in aqueous solution was investigated by small angle x-ray scattering (SAXS) and multidetector gel permeation chromatography coupled with postcolumn delivery (GPC/PCD) techniques in comparison with scleroglucan. It was shown that both polysaccharides exhibit a rigid rod-like conformation in aqueous solution by SAXS experiments. The mass per unit length (M/L) and radius (R) of rod cross section of PGG-Glucan were measured to be 6300 daltons/nm and 1.89 nm, while those of scleroglucan are 2300 and 0.83, respectively. Utilizing a GPC/light scattering technique, the average aggregation number of PGG-Glucan is 9, while that of scleroglucan is around 3. From the comparison of the M/L and R of the respective rod cross sections as well as their aggregation number data, it is concluded that PGG-Glucan is composed of triple helices, which tend to aggregate as triplets in solution, whereas scleroglucan is composed of a single triple helix. The aggregation number distribution of PGG-Glucan was found to range from 1 to about 25 determined by GPC/PCD. From the observation of a Debye-Scherrer ring type of peak in the macroscopic scattering cross section of PGG-Glucan by SAXS, the existence of a small amount of ordered clusters of PGG-Glucan can be deduced. The "lattice parameter" of these ordered fasces-like clusters is consistent with the radius of the individual triple-helical rods forming a microfibrillar superstructure. These results indicate that higher aggregated forms of PGG-Glucan containing up to 8 triple helices behave as ordered fasces-like clusters. We conclude that PGG-Glucan is triple-helix aggregates formed by rigid rods stacking together side by side. We propose a molecular structural model for PGG-Glucan conformations.  相似文献

Attempts were made to enhance cellulose saccharification by cellulase using cellulose dissolution as a pretreatment step. Four cellulose dissolution agents, NaOH/Urea solution, N-methylmorpholine-N-oxide (NMMO), ionic liquid (1-butyl-3-methylimidazolium chloride; [BMIM]Cl) and 85% phosphoric acid were employed to dissolve cotton cellulose. In comparison with conventional cellulose pretreatment processes, the dissolution pretreatments were operated under a milder condition with temperature <130 °C and ambient pressure. The dissolved cellulose was easily regenerated in water. The regenerated celluloses exhibited a significant improvement (about 2.7- to 4.6-fold enhancement) on saccharification rate during 1st h reaction. After 72 h, the saccharification yield ranged from 87% to 96% for the regenerated celluloses while only around 23% could be achieved for the untreated cellulose. Even with high crystallinity, cellulose regenerated from phosphoric acid dissolution achieved the highest saccharification rates and yield probably due to its highest specific surface area and lowest degree of polymerization (DP).  相似文献

Cellulose films were prepared by dissolving lyocell fibers in LiCl/N,N-dimethylacetamide solvent and subsequently coagulating and drying them under ambient conditions. To introduce preferred orientation, the films were uniaxially drawn under air-dry and rewetted conditions, respectively. Preferred orientation was determined by birefringence measurements and by wide-angle X-ray scattering. Mechanical properties were characterized by means of tensile tests with films conditioned to standard temperatures and humidity. Drawing resulted in the substantial reorientation of cellulose, whereby the molecular chains in the amorphous regions exhibited clearly stronger reorientation than the crystalline fraction. The average degree of orientation was comparable to orientation achieved in spun cellulose fibers. Wet-drawing resulted in improved tensile strength and modulus of elasticity but reduced elongation at break. The mechanical properties of wet-drawn films are competitive with regard to cellophane and melt-blown cellulose films, particularly considering their high modulus of elasticity of up to 26 GPa, which is also comparable to values obtained for industrially produced cellulose fibers.  相似文献
