Path coefficient analysis of the effects of stripe rust and cultivar mixtures on yield and yield components of winter wheat 总被引:2,自引:0,他引:2

Four club wheat cultivars and three two-component cultivar mixtures, planted at five frequencies, were grown in three environments in both the presence and absence of stripe rust. The effect of stripe rust on wheat yield was through the yield components, with weight of individual seed being the component most affected by rust. In some cases, yield component compensation was indicated by the presence of negative correlations among the yield components. Path analysis of the yield components revealed that components with the highest correlations to yield also had the largest direct effects on yield. Of the yield components, number of heads per unit area exerted the largest direct influence on yield. The direct effects of number of seeds per head and weight of individual seed were similar, although number of seeds per head was more important in the absence of rust than in its presence. The pure stands and mixtures differed considerably with respect to correlation coefficients, but were very similar for direct effects of yield components on yield. Breeding for resistance to the three rusts of wheat usually requires incorporation of genetically independent factors conferring resistance to each rust. Linked resistance genes in some alien translocation stocks permit concurrent transference of resistance for more than one rust. Alien derived resistances, however, are often reported to be associated with reduced yield and other undesirable characters. In our experience, backcross breeding when given a limited number of backcrosses (3–6) and with suitable selection procedures has resulted in lines giving yields higher or comparable to the recurrent wheat parent Kalyansona and resistance to one, two or all three rusts without any adverse effects. Some of the rust resistant derivatives also show resistance to Neovossia indica (Karnal bunt). The derivatives thus developed when used as parents in a breeding programme have produced several improved cultivars with high yields, superior grains and diversity for resistance to rust pathogens. One of the cultivars, named Vaishali (DL784-3), has been officially released for cultivation in the country. 相似文献

Yellow rust (causal agent: Puccinia striiformis f.sp. tritici) resistance in the UK wheat cultivar Guardian is developmentally regulated, resistance increasing as the plant matures. Yellow rust resistance was assessed under field conditions on plants after ear emergence to ensure maximum expression of resistance. Three quantitative trait loci (QTL) for yellow rust resistance were identified, being located on chromosomes 1B (QPst.jic-1B), 2D (QPst.jic-2D) and 4B (QPst.jic-4B). The largest resistance effect, QPst.jic-1B located to the same position on the long arm of chromosome 1B as the known durable source of yellow rust resistance, Yr29. Microscopic studies were carried out to determine what effect the resistance in Guardian had on the development of P. striiformis f.sp. tritici. While the adult plant resistance in Guardian did not prevent germinated urediniospores from establishing an effective infection site, the growth of hyphae within flag leaf tissue was significantly inhibited, slowing the development of microcolonies. 3,3-diaminabenzadine (DAB) and trypan blue staining indicated that this inhibition of hyphal growth was not associated with hydrogen peroxide accumulation or extensive plant cell death. 相似文献

Summary Wheat genotypes, including backcross derivatives of Thatcher carrying Lr10 and Lr23, substitution lines for Lr10 and Lr23 in Chinese Spring background and Chinese Spring and Thatcher were analysed against 21 pathotypes of leaf rust in seedling tests. Adult plant responses in all these stocks were observed in the field nurseries under exposure to the inoculum of the Indian virulent races of leaf rust. The seedling data demonstrated that both the substitution lines and the backcross derivatives for each gene carry identical pattern of infection for resistance. The high level of adult plant resistance in the substitution lines, in contrast to the backcross derivatives in Thatcher, has been postulated to be due to the combination of resistance contributed by Lr10 and adult plant Chinese Spring resistance or to Lr23 and Chinese Spring adult plant resistance. It has been suggested that genes Lr10 and Lr23 added to the Chinese Spring background provide sources for durable resistance, since Chinese Spring has continued to provide a moderate level of adult plant resistance to leaf rust for a very long time. 相似文献

Summary In winter wheat (Triticum aestivum L.), the development of a methodology to estimate genetic divergence between parental lines, when combined with knowledge of parental performance, could be beneficial in the prediction of bulk progeny performance. The objective of this study was to relate F2 heterosis for grain yield and its components in 116 crosses to two independent estimates of genetic divergence among 28 parental genotypes of diverse origins. Genetic divergence between parents was estimated from (a) pedigree relationships (coefficients of kinship) determined without experimentation, and (b) quantitative traits measured in two years of field experimentation in Kansas and North Carolina, USA. These distances, designated (1 -r) and G, respectively, provided ample differentiation among the parents. The 116 F2 bulks were evaluated at four locations in Kansas and North Carolina in one year. Significant rank correlations of 0.46 (P = 0.01) and 0.44 (P = 0.01) were observed between G and grain yield and kernel number heterosis, respectively. Although (1 -r) was poorly associated with grain yield heterosis, G and midparent performance combined to account for 50% of the variation in F2 yields among crosses when (1 -r) was above the median value, whereas they accounted for only 9% of the variation among crosses when (1-r) was below the median. Leaf rust, caused by Puccinia triticina, is one of the most damaging diseases of wheat worldwide. Lr16 is a widely deployed leaf rust resistance gene effective at the seedling stage. Although virulence to Lr16 exists in the Canadian P. triticina population, Lr16 provides a level of partial resistance in the field. The primary objective of this study was to identify markers linked to Lr16 that are suitable for marker-assisted selection (MAS). Lr16 was tagged with microsatellite markers on the distal end of chromosome 2BS in three mapping populations. Seven microsatellite loci mapped within 10 cM of Lr16, with the map distances varying among populations. Xwmc764 was the closest microsatellite locus to Lr16, and mapped 1, 9, and 3 cM away in the RL4452/AC Domain, BW278/AC Foremost, and HY644/McKenzie mapping populations, respectively. Lr16 was the terminal locus mapped in all three populations. Xwmc764, Xgwm210, and Xwmc661 were the most suitable markers for selection of Lr16 because they had simple PCR profiles, numerous alleles, high polymorphism information content (PIC), and were tightly linked to Lr16. Twenty-eight spring wheat lines were evaluated for leaf rust reaction with the P. triticina virulence phenotypes MBDS, MBRJ, and MGBJ, and analyzed with five microsatellite markers tightly linked to Lr16. There was good agreement between leaf rust infection type (IT) data and the microsatellite allele data. Genetic diversity is an important safeguard against crop vulnerability to biotic and abiotic stresses. Coefficient-of-parentage (COP) values of 248 North American hard spring wheat (Triticum aestivum L. em. Thell) cultivars released from 1901 to 1991 were used to estimate the genetic similarity of cultivars. COP values were used: to (1) quantify germ plasm sources and their contributions to the North American hard spring wheat gene pool; (2) measure changes in genetic diversity through time; and (3) identify major groupings of related cultivars. Landraces and local cultivars that contributed to the formation of the gene pool were: spring wheat (64%), winter wheat (16%), T. turgidum var. durum L. (10%), and T. turgidum var. emmer L. (8%). Fife, Hard Red Calcutta, and Turkey Red accounted for 18%, 13%, and 8%, respectively, of the hard spring wheat origins. Era and Butte in the US, and Neepawa and HY 320 in Canada, were the most commonly used named parents of cultivars released from 1981 to 1991. Both Canada and US had the greatest level of similarity among new cultivar releases in the 1930s (Canada: r=0.39, US: r=0.34). Genetic similarity in the US declined to r=0.14 in the 1940s and remained relatively constant thereafter. Similarity among released Canadian cultivars remained relatively high until the 1970s when the introduction of new market classes resulted in a 50% reduction in genetic similarity to approximately the same level of similarity found in the US. Cluster analysis was used to group cultivars released after 1941 into 13 clusters of similar genotypes. Summary Inheritance of gliadin components in winter wheat has been studied by one-dimensional polyacrylamide gel electrophoresis. Single F2 grains from 36 intervarietal hybrid combinations have been analysed. The genetic analysis has revealed blocks, including 1–6 gliadin components, which are inherited as individual mendelian traits. About 80 variants of blocks have been detected. On the basis of the allelism test they are grouped into 6 series in accordance with the number of known gliadin-coding loci located on chromosomes of the homoeologous groups 1 and 6. Each series includes 8–18 blocks controlled by different alleles of one gliadin-coding locus. Blocks of components have been confirmed to be inherited codominantly in accordance to the gene dose in the triploid endosperm. The highest similarity between members of one series is observed in groups of blocks controlled by chromosomes ID and 6D. On the contrary, many blocks controlled by chromosomes 1A and 1B have no bands in common. The presented catalogue of blocks of components may be used to make up gliadin genetic formulae and to compare electrophoregrams obtained by different authors. Blocks of gliadin components are suitable genetic markers for use in revealing and studying heterogeneity of wheat varieties, in tracing their origin, in identifying recombinations, translocations and substitutions of the genetic material and in solving many other problems of the origin, evolution and selection of hexaploid wheat. 相似文献

Wheat (Triticum aestivum L.) has been examined for allelopathic potential against annual ryegrass (Lolium rigidum). The bioassay technique, 'equal-compartment-agar-method', was employed to evaluate seedling allelopathy in a doubled-haploid (DH) population derived from cv Sunco (weakly allelopathic) and cv Tasman (strongly allelopathic). A significant difference in allelopathic activity was found among the DH lines, which inhibited the root length of ryegrass across a range from 23.7 to 88.3%. The phenotypic data showed that wheat allelopathic activity was distributed normally within this DH population and a substantial transgressive segregation for seedling allelopathic activity was also found. Analysis of restriction fragment length polymorphism (RFLP), amplified fragment length polymorphism (AFLP) and microsatellite (SSRs) markers identified two major QTLs on chromosome 2B associated with wheat allelopathy. The linkage analysis of genetic markers and the QTLs may improve genetic gains for the allelopathic activity through marker-assisted selection in wheat breeding. The development of wheat allelopathic cultivars could reduce the over-reliance of weed control on synthetic herbicides.Communicated by J. Dvorak 相似文献

Summary Genetic variability of endosperm esterase has been studied in 42 cultivars of Triticum aestivum L. 2n=6x=42. Different techniques, including sequential electrophoresis and electrofocusing, have been used with various substrates and esterase inhibitors. The electrophoretic patterns in each cultivar are described. Chromosomal location using the nullitetrasomic and ditelosomic lines of Chinese Spring was carried out in order to relate and/or locate the esterase genes to specific chromosomes. Most of the esterase isozymes located were in the long arm of the chromosomes of the homoeology group 3; but we have found six located in the short arms, five of them in the chromosome 3AS and one in the 3DS. This location increases the number of esterase genes described, because no esterase genes had been described so far in short arms of chromosomes of the homoeology group 3. The genetic control is discussed and, according to our results, between 12 and 15 loci, organized in five compound loci, control the endosperm esterases in wheat. Also one modifier gene modifying the mobility of two esterase bands and present in all the cultivars studied is postulated.This work was supported by a personal grant (L. Summary A study of spring bread wheat (Triticum aestivum) germ plasm developed at the International Maize and Wheat Improvement Center (CIMMYT) showed highly significant phenotypic variability for each component of partial resistance (namely, uredial appearance period, latency period, uredial number and uredial size) to Puccinia recondita f. sp. tritici. All of the wheat genotypes displayed longer uredial appearance and latency periods and decreased uredial number and uredial size when compared to the susceptible check cultivar Morocco. Positive correlations between uredial appearance period and latency period, and uredial number and uredial size, and negative correlations between uredial appearance and latency periods and uredial number and uredial size, inclusive, suggested that the components of partial resistance were either tightly linked or under pleiotropic genetic control. Compared to Morocco, all entries had slow disease progress in the field and variation occurred in the germ plasm for the area under the leaf rust progress curve. Disease progress was negatively correlated with uredial appearance and latency periods, whereas a positive correlation was observed with uredial number and uredial size. Certain genotypes displayed high levels of partial resistance resulting in low disease incidence in the field. 相似文献

Protein kinase activities were extracted from anther tissue of wheat (Triticum aestivum L. cv. Zaragoza) and characterized with regard to the effects of polyamines, second messengers and plant growth hormones. The protein kinases were inhibited by polyamines and cyclic nucleotides, but were stimulated by the addition of auxins, gibberellic acid and kinetin. The dominant polyamine-sensitive kinase activity was partially purified and characterized. The optimal pH of the reaction was 7.5 to 8.0 and casein was the preferred exogenous substrate. Polyamines were inhibiting in the decreasing order of putrecine > spermidine > spermine. The results are discussed against the context of the anther culture technique. 相似文献

We constructed a genetic linkage map based on a cross between two Swiss winter wheat (Triticum aestivum L.) varieties, Arina and Forno. Two-hundred and forty F5 single-seed descent (SSD)-derived lines were analysed with 112 restriction fragment length polymorphism (RFLP) anonymous probes, 18 wheat cDNA clones coding for putative stress or defence-related proteins and 179 simple-sequence repeat (SSR) primer-pairs. The 309 markers revealed 396 segregating loci. Linkage analysis defined 27 linkage groups that could all be assigned to chromosomes or chromosome arms. The resulting genetic map comprises 380 loci and spans 3,086 cM with 1,131 cM for the A genome, 920 cM for the B genome and 1,036 cM for the D genome. Seventeen percent of the loci showed a significant (P < 0.05) deviation from a 1:1 ratio, most of them in favour of the Arina alleles. This map enabled the mapping of QTLs for resistance against several fungal diseases such as Stagonospora glume blotch, leaf rust and Fusarium head blight. It will also be very useful for wheat genetic mapping, as it combines RFLP and SSR markers that were previously located on separate maps. S. Paillard and T. Fertile, green plants were regenerated from immature inflorescence explants from each of four Canadian wheat cultivars. The cultivars were representative of four classes of Canadian wheat. Explants from immature inflorescences of three size ranges were cultured on two types of media: MSI/MSR, which contains 1650 mg l-1 NH4NO3and sucrose as a carbon source, and BII/BIR, which contains 250 mg l-1 NH4NO3and maltose as a carbon source. Regeneration from all cultivars was significantly better on BII/BIR media than on MSI/MSR media. On BII/BIR media, `AC Karma', `Plenty', and `Fielder' gave the highest number of shoots per 10 explants, where the explants were derived from immature inflorescences 5.1 to 10.0 mm in length. 'Columbus' did not regenerate on MSI/MSR medium, and regenerated poorly on BII/BIR medium. Differences were found between cultivars with regard to the number of regenerant plants produced with the best treatments: `Plenty' produced 16.1 shoots per 10 explants, `AC Karma' 12.4, `Fielder' 6.4, and `Columbus' 2.2. 相似文献

Summary A study of pollen development in wheat was made using transmission electron microscopy (TEM). Microspores contain undifferentiated plastids and mitochondria that are dividing. Vacuolation occurs, probably due to the coalescence of small vacuoles budded off the endoplasmic reticulum (ER). As the pollen grain is formed and matures, the ER becomes distended with deposits of granular storage material. Mitochondria proliferate and become filled with cristae. Similarly, plastids divide and accumulate starch. The exine wall is deposited at a rapid rate throughout development, and the precursors appear to be synthesized in the tapetum. Tapetal cells become binucleate during the meiosis stage, and Ubisch bodies form on the plasma membrane surface that faces the locule. Tapetal plastids become surrounded by an electron-translucent halo. Rough ER is associated with the halo around the plastids and with the plasma membrane. We hypothesize that the sporopollenin precursors for both the Ubisch bodies and exine pollen wall are synthesized in the tapetal plastids and are transported to the tapetal cell surface via the ER. The microspore plastids appear to be involved in activities other than precursor synthesis: plastid proliferation in young microspores, and starch synthesis later in development. Plants treated with the chemical hybridizing agent RH0007 show a pattern of development similar to that shown by untreated control plants through the meiosis stage. In the young microspore stage the exine wall is deposited irregularly and is thinner than that of control plants. In many cases the microspores are seen to have wavy contours. With the onset of vacuolation, microspores become plasmolyzed and abort. The tapetal cells in RH0007-treated locules divide normally through the meiosis stage. Less sporopollenin is deposited in the Ubisch bodies, and the pattern is less regular than that of the control. In many cases, the tapetal cells expand into the locule. At the base of one of the locules treated with a dosage of RH0007 that causes 95% male sterility, several microspores survived and developed into pollen grains that were sterile. The conditions at the base of the locule may have reduced the osmotic stress on the microspores, allowing them to survive. Preliminary work showed that the extractable quantity of carotenoids in RHOOO7-treated anthers was slightly greater than in controls. We concluded that RH0007 appears to interfere with the polymerization of carotenoid precursors into the exine wall and Ubisch bodies, rather than interfering with the synthesis of the precursors. 相似文献

Twenty-two cultivars and lines of winter and spring wheat (Triticum aestivum L.) were studied, most for the first time, for their anther culture response. The response was genotype dependent. Plants grown in the field gave higher callus induction frequency than those grown in the greenhouse and the controlled environment chamber. Donor plants grown in a season of low drought stress as compared to a season of severe drought stress resulted in a higher frequency of callus induction. Spherical microcalli were observed in two wheat genotypes in some of only those anthers that were placed with only one loculus in contact with the medium. Wheat lines that were more responsive to anther culture were identified. 相似文献

MYB转录因子是植物最大的转录因子家族之一,广泛参与植物各种生理生化过程。该研究通过对小麦基因组测序数据库进行同源搜索,利用电子克隆技术从紫色籽粒小麦品种‘高原115’中分离得到了一个新的MYB基因TaMYB3-4 D。结果表明,TaMYB3-4D仅含有一个内含子,其编码蛋白含有2个连续的MYB结构域,为典型的R2R3-MYB蛋白。TaMYB3-4D系统发生关系上与调控花青素合成的MYB基因亲缘关系较近。TaMYB3-4 D与bHLH基因ZmR瞬时表达能够诱导白色胚芽鞘中花青素的合成。此外,TaMYB3-4 D基因仅在‘高原115’含花青素的种皮和胚芽鞘中表达,在根、茎、叶中均未表达。研究表明,TaMYB3-4 D基因是一个具有调控花青素合成代谢功能的R2R3-MYB基因,很有可能参与小麦花青素的生物合成。 相似文献

采用开顶箱(OTC)法和遮光网技术,设置100 n L/L臭氧熏气与3个辐射减弱梯度结合,模拟臭氧浓度升高和太阳辐射减弱的复合大气背景。用BIOLOG生态测试板,采用孔平均颜色变化率法(AWCD)测定冬小麦根际土壤微生物利用不同碳源的能力,计算微生物群落多样性指数,对不同碳源的利用率进行了主成分分析。两年试验结果显示,臭氧熏气与太阳辐射减弱复合作用,降低了土壤微生物对碳源的利用速度和利用总量;除了聚合物以外其它碳源利用率显著降低;对土壤微生物多样性没有直接的影响;对碳源降解的抑制效应大于增强的O3与减弱的太阳辐射两因素各自的单独作用。太阳辐射减弱20%,一定程度上增加了对聚合物类的分解。O3熏气条件下太阳辐射减弱,糖类、胺类代谢变异度较高,受环境影响较大。 相似文献
