Muscarinic stimulation of cultured fibroblasts decreases initial rates of cAMP accumulation in response to hormones 50–70%. This inhibitory effect of muscarinic stimulation on cAMP accumulation in intact cells was desensitized 65–75% by a 60 min pretreatment with the muscarinic agonist carbachol (10 μM), with a t12 of 11 min. The carbachol pretreatment resulted in a diminished carbachol inhibition of adenylate cyclase in broken cell preparations. The phospholipid monooleylphosphatidate (MOPA) which also inhibited hormone-stimulated cAMP accumulation with a half maximal effect at 0.03 μM (as compared with 0.5 μM for carbachol), displayed many of the characteristics of muscarinic inhibition such as loss of activity with time of pretreatment. However, fibroblasts did not become desensitized to prolonged MOPA treatment; rather, it appeared that the MOPA was being inactivated. Also, the desensitization to carbachol did not prevent further inhibition by MOPA. The inhibitory effects of maximal doses of MOPA and carbachol in combination were no greater than the effect of carbachol alone, suggesting that they shared an intermediate in their inhibition of cAMP accumulation. These results are consistent with the hypothesis that muscarinic inhibition of adenylate cyclase is mediated by the formation of a phospholipid. However, the desensitization to the cholinergic stimulus does not appear to involve the intermediate, but rather a modification at the receptor level.  相似文献

In the isolated rabbit papillary muscle, experiments were carried out in order to elucidate whether or not cholinergic stimulation produces a differential antagonistic action on the positive inotropic effects mediated via β- and α-adrenoceptor stimulation. Carbachol (0.1–30 μM) alone scarcely affected the basal tension developed. The postive inotropic effects of phenylephrine (30 μM) in the presence of phentolamine and of isoprenaline, which were mediated via β-adrenoceptors, were markedly inhibited by carbachol. Carbachol (3 μM) shifted the dose-response curve for isoprenaline in a parallel manner, and that for phenylephrine with phentolamine to the right and downwards. Carbachol administered during induction of the positive inotropic effects via α-adrenoceptors by phenylephrine (30 μM) with pindolol or by methoxamine failed to inhibit these effects and increased further the tension developed. The dose-response curve for phenylephrine determined in the presence of pindolol was not affected by carbachol. The present results indicate that the cholinergic antagonism of the adrenergic action on the contractility of the mammalian ventricular myocardium is exerted specifically to the β-adrenoceptor-mediated action, but not to the α-adrenoceptor-mediated one.  相似文献

The inhibition of prostaglandin (PG) synthetase by nonsteroidal anti-inflammatory drugs (NSAID) is not well understood. Co-factors (glutathione and hydroquinone) are needed for maximum enzymatic activity in vitro, and we suggest that NSAID might inhibit PG synthetase partly by interfering with co-factor induced stimulation of the enzyme. This hypothesis was tested by:A) Examining the effect of glutathione, noradrenaline and hydroquinone on bull seminal vesicle (BSV) PG synthetase in vitro. The stimulatory effects were concentration-dependent.B) Three structurally distinct NSAID, indomethacin, aspirin and paracetamol, inhibited the stimulation by each co-factor in a concentration-related manner. Rats were fed ethanol (Lieber-DeCarli diet) for three weeks. Stimulation of cerebellar adenylate cyclase by calcium was measured in control (pair-fed), chronic-alcohol and alcohol-withdrawn animals. No differences in the sensitivity or maximal stimulation of this enzyme were observed among these groups. Ethanol in,vitro (1%) stimulated brain adenylate cyclase approximately 50% in the presence or absence of calcium. Chronic alcohol exposure in,vivo did not alter the sensitivity of adenylate cyclase to stimulation by alcohol in,vitro.  相似文献

An ATPase is demonstrated in plasma membrane fractions of goldfish gills. This enzyme is stimulated by Cl? and HCO3?, inhibited by SCN?.Biochemical characterization shows that HCO3? stimulation (Km = 2.5 mequiv./l) is specifically inhibited in a competitive fashion by SCN? (Ki = 0.25 mequiv./l). The residual Mg2+-dependent activity is weakly is weakly affected by SCN?.In the microsomal fraction chloride stimulation of the enzyme occurs in the presence of HCO3? (Kmfor chloride = 1 mequiv./l); no stimulation is observed in the absence of HCO3?. Thiocyanate exhibits a mixed type of inhibition (Ki = 0.06 mequiv./l) towards the Cl? stimulation of the enzyme.Bicarbonate-dependent ATPase from the mitochondrial fraction is stimulated by Cl?, but this enzyme has a relatively weak affinity for this substrate (Km = 14 mequiv./l).  相似文献

Cyclic AMP phosphodiesterase can be extracted from sporangiophores of Phycomycesblakesleeanus. Activity is enhanced by 1–10 μM FAD and FMN but not by riboflavin. Moderate intensity of blue light also activates the enzyme, especially in the presence of 1mM GTP. The enzyme must be extracted and stored in the absence of blue light for this result. Forty times the intensity of red light has no effect. This finding is consistent with the very sudden transient drop in cyclic AMP level upon light stimulation in the intact sporangiophore.  相似文献

The agonist binding affinity of nicotinic acetylcholine receptor (nAChR) from Torpedocalifornica electroplax, as inferred from ability of agonist to inhibit specific curaremimetic neurotoxin binding to nAChR, is sensitive to the duration of exposure to agonist. The concentration of carbachol necessary to prevent one-half of toxin binding over a 30 min incubation with nAChR (K30) is 10 μM when toxin and carbachol are simultaneously added to membrane-bound nAChR, and 3 μM when nAChR are pretreated with carbachol for 30 min prior to the addition of toxin. These alterations in agonist affinity may be mimicked by modification of nAChR thiol groups. Affinity of nAChR for carbachol is decreased following treatment with dithiothreitol (DTT). Dithio-bis-nitrobenzoic acid treatment of DTT-reduced membranes yields K30 values of 5 μM for carbachol, while N-ethylmaleimide treatment of DTT-reduced nAChR produces nAChR with reduced affinity for carbachol, reflected in K30 values of about 400 μM. In the absence of Ca++, K30 values for carbachol binding to native and DTT-reduced nAChR are diminished 3–6 fold. These affinity alterations are not observed with d-tubocurarine (antagonist) binding to nAChR. Thus, Ca++ and the oxidation state of nAChR thiols appear to affect the affinity of nAChR for agonists (but not antagonists), and may therefore be related to agonist-mediated events in receptor activation and/or desensitization.  相似文献

From a study of the translation of synthetic polynucleotides in the E. coli and B. thuringiensis cell-free systems, it is shown that the stimulation of polypeptide synthesis by spermidine depends on the uracil content of messenger ribonucleic acid. An enzyme, ribonucleotide polymerase, isolated from the yeast phase of a fungus, Histoplasma capsulatum has been found to stimulate the incorporation of dTMP in the reaction catalysed by DNA polymerase from H. capsulatum and E. coli. The stimulation is dependent on the amount of ribonucleotide polymerase added. The data indicate that protein-protein interaction is responsible for the increase in DNA synthesis. It is suggested that ribonucleotide polymerase may be involved in supplying short RNA primers for DNA polymerase.  相似文献

The first precocious development of UDP glucuronyltransferase in the mammalian fetus in utero by a known compound of endogenous origin is described. Intraperitoneal injection of cortisol (8 mg) into maternal rats on days 14 and 15 of gestation stimulated fetal-liver transferase activity from near zero to 12 maternal levels by day 17; 0.3 mg dexamethasone, possessing a longer biological half-life, raised activity to full maternal level by day 16. In controls, injected with solvent only, fetal-liver transferase remained low on day 16. With both glucocorticoids, transferase stimulation was dose-dependent. Transferase activities were assayed in a range of digitonin concentrations from zero to above optimal for enzyme activation. Activities stimulated were towards o-aminophenol, p-nitrophenol, 1-naphthol and serotonin. Activities towards bilirubin, morphine and testosterone were not stimulated. The former group of activities are stimulated by glucocorticoids in culture and normally reach approximate adult levels just before birth; the latter group are not so stimulated on culture and normally reach adult levels after birth. Implications of these findings are discussed.  相似文献

Thiosulfate-cytochrome c-551 reductase derived from Chlorobiumthiosulfatophilum has been highly purified. The enzyme reduces cytochrome c-551 of C. thiosulfatophilum in the presence of thiosulfate while cytochrome c-555 of the organism is not reduced by the enzyme. Cytochrome c-555 reacts with the enzyme at an appreciable rate only in the presence of cytochrome c-551. However, the reduction rate of cytochrome c-551 by the enzyme is greatly enhanced on addition of a catalytic amount of cytochrome c-555. Therefore, cytochrome c-555 seems to function as an effector on thiosulfate-cytochrome c-551 reductase as well as it acts as the electron donor to the light-excited chlorobium chlorophylls.  相似文献

Phospho-N-acetylmuramyl-pentapeptide translocase, the initial membrane enzyme in the biosynthesis of peptidoglycan, requires a lipid microenvironment for function. n-Butanol was reversibly intercalated into membranes to perturb the hydrophobic interactions in this microenvironment in order to define further the role of lipid. In the concentration range for maximal stimulation of enzymic activity (0.12–0.18 M), n-butanol causes a 40% decrease in the fluorescence emission of the dansylated product, undecaprenyl diphosphate-(N?-dansyl)pentapeptide. Since no change in emission maximum occurs below 22°C in the presence of 0.12 M n-butanol, it is concluded that intercalation of this alkanol causes an increase in fluidity. Above 22°C this concentration of n-butanol causes both a decrease in the fluorescence emission and a red shift in the emission maximum. It is concluded that a polarity change as well as fluidity change occurs above 22°C. n-Butanol also causes a significant change in the phase transition experienced by the dansylated lipid product. Thus, it is possible with n-alkanols, e.g. n-butanol, to perturb lipid-translocase interactions resulting in an increase in fluidity in the microenvironment of the enzyme. This change in fluidity correlates with a stimulation of enzymic activity.  相似文献

The effect of prostaglandin E2 on vascular responses of the rabbit kidney to renal nerve stimulation and noradrenaline was examined invitro and insitu as a test of the hypthesis that prostaglandins of the E series may be involved in the regulation of adrenergic neuroeffector transmission. Intraarterial administration of prostaglandin E2 to the invitro kidney caused marked inhibition of vascular responses to nerve stimulation whereas the responses to noradrenaline were not significantly altered. In the insitu preparation, vascular responses to both nerve stimulation and noradrenaline were inhibited by prostaglandin E2 infusion, although its effect on responses to nerve stimulation was approximately twice that observed on responses to noradrenaline.It is concluded that prostaglandin E2 acts primarily at a prejunctional level of adrenergic neuroeffector transmission in the kidney, although a postjunctional effect has also been observed.  相似文献

In homogenates of Macacamulatta (Rhesus) or Cebusapella amygdaloid nuclear complex, adenylate cyclase activity was approximately doubled by either 10μM dopamine or 8mM NaF. In the presence of morphine, the stimulation by dopamine was reduced. A 90–100% inhibition of the dopamine stimulation was obtained with 20μM, and a 50% inhibition, with 5μM morphine. The effects of 10μM morphine on dopamine stimulation were reversed by 10μM naloxone. Morphine itself did not significantly affect the basal adenylate cyclase activity, but in the presence of 10μM morphine the stimulation by 8mM NaF was reduced approxiamtely 50%. The data suggest an action of morphine at a receptor site which is distinct from the dopamine receptor, but which inhibits the dopamine-stimulated adenylate cyclase. In addition, the cyclic GMP content of Cebus amygdala slices was reduced by 50–75% during incubation for 5–20 minutes with morphine. Maximum effects on cyclic GMP were obtained with 10μM, and half-maximum effects, with 0.1μM morphine. The effect of morphine on amygdala cyclic GMP was not reversed by naloxone. Thus, this action of morphine may not be receptor mediated, or may involve the interaction of morphine with receptors other than the opiate receptor.  相似文献

Infection by L13, a temperature-sensitive mutant of gene 42 of phage T4, the structural gene for dCMP hydroxymethylase, previously was shown not to form T4 DNA at nonpermissive temperatures. Yet the enzyme activity was found in extracts. Since inactivation of the enzyme was not reversible, we have examined acid-soluble extracts of cells infected at nonpermissive temperature by tsL13 for 5-hydroxymethyldCMP in order to determine whether the enzyme functioned in vivo. A double mutant of tsL13 and amB24 (5-hydroxymethyldCMP kinase) did not form the nucleotide at nonpermissive temperature, but the control, amB24, formed large quantities. From these results and previous temperature-shift studies it is suggested that the enzyme is normally activated to function in vivo between 5 and 8 minutes after infection.  相似文献

A new type of enzyme electrode has been developed for invivo electrochemical measurements which allows discrimination between ascorbic acid and catecholamines and their metabolites. The electrode employs the enzyme ascorbic acid oxidase held between the voltammetric electrode and solution by a dialysis membrane or immobilized on the outer surface of serous membrane from rat small intestine. The electrode gives linear calibration curves for all catecholamines and metabolites independent of any ascorbic acid concentrations significant in physiological measurements. The electrode has been tested in brain slice measurements and shown to respond to releases of catecholamines initiated by potassium ion stimulation.  相似文献

High-dose carbachol (10?3 M) has previously been shown to cause NaCl absorption in short-circuited rabbit ileum. The mechanism of this effect may be norepinephrine release induced by carbachol activation of presynaptic nicotinic receptors on adrenergic neurons. Norepinephrine then interacts with postsynaptic α-adrenergic receptors on intestinal mucosal cells to stimulate neutral NaCl absorption and inhibit electrogenic bicarbonate secretion. The present paper examines the in vitro intestinal ion transport effects of DMPP an agent which is more specific than carbachol on nicotinic cholinergic receptors. DMPP (10?5 M) caused a transient increase followed by prolonged depression of the short-circuit current, increased NaCl absorption and increased tissue conductance. This effect was antagonized by hexamethonium and phentolamine. It is concluded that nicotinic cholinergic agents stimulate norepinephrine release from adrenergic nerves and effect intestinal ion transport just as norepinephrine does.  相似文献

The pyrimidine analogs, 5-fluoroorotate and 5-azacytidine, have been shown to stimulate the basal level as well as the cortisone, tryptophan, and casein hydrolysate-induced levels of the rat liver enzyme, tyrosine aminotransferase. This stimulation was most marked in the case of dietary and hormonal induction when the analog was given 4–6 hr prior to the administration of the inducer. When tryptophan induced tyrosine aminotransferase, maximal stimulation by the analog occurred if it were given 2 hr prior to the administration of the amino acid. The optimal stimulatory dose of 5-azacytidine was 5 mg/kg body weight whereas 5-fluoroorotate gave its highest stimulation at a dose of 60 mg/kg. Of several orotic acid analogs tested, only the chloro-analog had an effect similar to the fluoro-congener.Utilizing quantitative immunochemical precipitation and pulse labeling in vivo, it was demonstrated that the administration of 5-fluoroorotate or 5-azacytidine at doses of 60 and 5 mg per kg, respectively, while causing a stimulation in the basal level of tyrosine aminotransferase, did not result in any change in the rate of enzyme synthesis. Furthermore, after cortisone induction of the enzyme, the delayed administration of these analogs caused either a further stimulation in the level of the enzyme or the maintenance of a high level while the enzyme activity decayed in animals not given the analogs. The rates of synthesis either showed no change or a decrease while the amount of enzyme was increasing. Prelabeling of the enzyme in vivo after induction with cortisone and followed by the administration of 5-fluoroorotate resulted in a marked decrease in the t12 of the decay rate of the enzyme measured either by loss of radioactivity or by loss of enzyme activity. These studies suggest that these analogs act in some manner to prevent enzyme turnover by an inhibition of enzyme degradation.  相似文献

5-Azacytidine, when added to growing E.coli K12, causes a decrease in DNA methylation assayed invitro. This decrease is greater when E.coli DNA is used as substrate than when calf thymus DNA is used. The decrease in activity is not due to the inhibition of protein synthesis caused by this drug, since neither chloramphenicol nor rifampin causes a decrease in enzyme activity. The effect is specific for the DNA(cytosine-5)methylase; the methylation of adenine is not affected. The concentration of drug that inhibits the DNA methylase by 50% is the same concentration that inhibits cell growth by 50%.  相似文献
