Gordonia polyisoprenivorans CCT 7137 was isolated from groundwater contaminated with leachate in an old controlled landfill (São Paulo, Brazil), and cultured in GYM medium at different concentrations of sugarcane molasses (2%, 6%, and 10%). The strain growth was analyzed by monitoring the viable cell counts (c.f.u. mL?1) and optical density and EPS production was evaluated at the end of the exponential phase and 24 h after it. The analysis of the viable cell counts showed that the medium that most favored bacterial growth was not the one that favored EPS production. The control medium (GYM) was the one that most favored the strain growth, at the maximum specific growth rate of 0.232 h?1. Differences in bacterial growth when cultured at three different concentrations of molasses were not observed. Production of EPS, in all culture media used, began during the exponential phase and continued during the growth stationary phase. The highest total EPS production, after 24 h of stationary phase, was observed in 6% molasses medium (172.86 g L?1) and 10% (139.47 g L?1) and the specific total EPS production was higher in 10% molasses medium (39.03 × 10?11 g c.f.u.?1). After the exponential phase, in 2%, 6%, and 10% molasses media, a higher percentage of free exopolysaccharides (EPS) was observed, representing 88.4%, 62.4%, and 64.2% of the total, respectively. A different result was observed in pattern medium, which presented EPS made up of higher percentage of capsular EPS (66.4% of the total). This work is the first study on EPS production by G. polyisoprenivorans strain in GYM medium and in medium utilizing sugarcane molasses as the sole nutrient source and suggests its potential use for EPS production by G. polyisoprenivorans CCT 7137 aiming at application in biotechnological processes.  相似文献

Gordonia polyisoprenivorans CCT7137, exopolysaccharide-producing bacterium, was isolated from groundwater contaminated with leachate in a former municipal landfill site (São Paulo, Brazil). The strain was submitted to starvation in phosphate-buffered saline solution for 56 days so as to evaluate its behavior regarding cuturability and cell morphology. As a response to starvation, G. polyisoprenivorans CCT7137 presented reduction in viable cell count, cell size and cell shape alteration. The initial number of viable cells was 1.51 × 107 c.f.u. ml. After 7 days of starvation culturability dropped to 13.70% (2.07 × 106 c.f.u/ml) and, after 56 days, to 3.25% (4.93 × 105 c.f.u/ml). It was also observed that after 7 days of starvation the cell size presented an average reduction in the values of length, length/width ratio, volume and area of 50, 58, 40 and 42%, respectively. The length/width ratio showed a change of shape from rod to coccobacillus. Changes in cell dimensions and distribution of cell into classes were not significant after day 7 of starvation. The results obtained show that it is not necessary for the strain to starve for more than a week to obtain G. polyisoprenivorans CCT7137 size- reduced cells. The results also indicate the potential for its starved forms to be used in future tests in porous medium to study the production of exopolysaccharide in situ.  相似文献

Two culture modes, continuous and semi-continuous, of the decolorization fungus,Geotrichum candidum Dec 1, were compared to obtain a high treatment efficiency of molasses decolorization and a large productivity of peroxidase (DyP) to simultaneously decolorize dyes and molasses. The continuous culture ofG. candidum Dec 1 using a 5-l jar-fermentor showed high DyP activity at a low dilution ratio of 0.005h−1, and decolorization ratio of molasses of 80% was obtained concomitantly. Therefore, a semi-continuous culture was performed by repeated refill and draw. In this mode, approximately 1.5 liters of the culture broth was replaced per cycle when the decolorization ratio of molasses was near 80%. The molasses medium (1.0 liter per day) was treated and the peroxidase productivity in the drawn culture broth was 26.6 U/day, whereas the peroxidase productivity was 17.9 U/day in the continuous culture with a dilution rate of 0.005 h−1. Molasses “B” is a rich co-product of the sugarcane process. It is obtained from the second step of crystallization and is richer in fermentable sugars (50–65%) than the final molasses, with a lower non-sugar solid content (18–33%); this co-product also contains good vitamin and mineral levels. The use of molasses “B” for ethanol production could be a good option for the sugarcane industry when cane sugar prices diminish in the market. In a complex medium like molasses, osmotolerance is a desirable characteristic for ethanol producing strains. The aim of this work was to evaluate the use of molasses “B” for ethanol production using Saccharomyces cerevisiae ITV-01 (a wild-type yeast isolated from sugarcane molasses) using different initial sugar concentrations (70–291 g L−1), two inoculum sizes and the addition of nutrients such as yeast extract, urea, and ammonium sulphate to the culture medium. The results obtained showed that the strain was able to grow at 291 g L−1 total sugars in molasses “B” medium; the addition of nutrients to the culture medium did not produce a statistically significant difference. This yeast exhibits high osmotolerance in this medium, producing high ethanol yields (0.41 g g−1). The best conditions for ethanol production were 220 g L−1 initial total sugars in molasses “B” medium, pH 5.5, using an inoculum size of 6 × 106 cell mL−1; ethanol production was 85 g L−1, productivity 3.8 g L−1 h−1 with 90% preserved cell viability.  相似文献

Clostridium bifermentans strain DPH-1 has already been found to dechlorinate perchloroethylene (PCE) tocis-dichloroethylene (cis-DCE)via trichloroethylene (TCE). In this study, our investigation on different culture conditions of this DPH-1 strain was extended to find a more efficient and cost effective growth medium composition for this DPH-1 strain in bioremediation practices. Temperature dependency of strain DPH-1 showed that the growth starting time and PCE degradation at 15°C was very slow compared to that of 30°C, but complete PCE degradation occurred in both cases. For the proper utilization of strain DPH-1 in more cost effective bioremediation practices, a simpler composition of an effective media was studied. One component of the culture medium, yeast extract, had been substituted by molasses, which served as a good source of electron donor. The DPH-1 strain in the medium containing molasses, in the presence of K2HPO4 and KH2PO4, showed identical bacterial multiplication (0.135 mg protein mL−1h−1) and PCE degradation rates (0.38 μM/h) to those of the yeast extract containing medium.  相似文献

Lactobacillus delbrueckii was grown on sugarcane molasses, sugarcane juice and sugar beet juice in batch fermentation at pH 6 and at 40°C. After 72 h, the lactic acid from 13% (w/v) sugarcane molasses (119 g total sugar l−1) and sugarcane juice (133 g total sugar l−1) was 107 g l−1 and 120 g l−1, respectively. With 10% (w/v) sugar beet juice (105 g total sugar l−1), 84 g lactic acid l−1 was produced. The optical purities of d-lactic acid from the feedstocks ranged from 97.2 to 98.3%.  相似文献

Previously, a Saccharomyces cerevisiae strain was engineered for xylose assimilation by the constitutive overexpression of the Orpinomyces xylose isomerase, the S. cerevisiae xylulokinase, and the Pichia stipitis SUT1 sugar transporter genes. The recombinant strain exhibited growth on xylose, under aerobic conditions, with a specific growth rate of 0.025 h−1, while ethanol production from xylose was achieved anaerobically. In the present study, the developed recombinant yeast was adapted for enhanced growth on xylose by serial transfer in xylose-containing minimal medium under aerobic conditions. After repeated batch cultivations, a strain was isolated which grew with a specific growth rate of 0.133 h−1. The adapted strain could ferment 20 g l−1 of xylose to ethanol with a yield of 0.37 g g−1 and production rate of 0.026 g l−1 h−1. Raising the fermentation temperature from 30°C to 35°C resulted in a substantial increase in the ethanol yield (0.43 g g−1) and production rate (0.07 g l−1 h−1) as well as a significant reduction in the xylitol yield. By the addition of a sugar complexing agent, such as sodium tetraborate, significant improvement in ethanol production and reduction in xylitol accumulation was achieved. Furthermore, ethanol production from xylose and a mixture of glucose and xylose was also demonstrated in complex medium containing yeast extract, peptone, and borate with a considerably high yield of 0.48 g g−1.  相似文献

Several wild strains and mutants of Rhodotorula spp. were screened for growth, carotenoid production and the proportion of -carotene produced in sugarcane molasses. A better producer, Rhodotorula glutinis mutant 32, was optimized for carotenoid production with respect to total reducing sugar (TRS) concentration and pH. In shake flasks, when molasses was used as the sole nutrient medium with 40 g l−1 TRS, at pH 6, the carotenoid yield was 14 mg l−1 and -carotene accounted for 70% of the total carotenoids. In a 14-l stirred tank fermenter, a 20% increase in torulene content was observed in plain molasses medium. However, by addition of yeast extract, this effect was reversed and a 31% increase in -carotene content was observed. Dissolved oxygen (DO) stat fed-batch cultivation of mutant 32 in plain molasses medium yielded 71 and 185 mg l−1 total carotenoids in double- and triple-strength medium, respectively. When supplemented with yeast extract, the yields were 97 and 183 mg l−1 total carotenoid with a 30% increase in -carotene and a simultaneous 40% decrease in torulene proportion. Summary A new PVA-hydrogel matrix for yeast cell immobilization for xylitol bioproduction from sugarcane bagasse was studied. Five repeated-batch fermentation runs were carried out in medium based on sugarcane bagasse hemicellulosic hydrolysate with reuse of the entrapped biocatalyst. The system performance as well as the metabolic behaviour of cells entrapped into the matrix were evaluated. The biocatalyst remained stable and exhibited a similar fermentative profile in all the successive batches, demonstrating the viability of the system. At the end of the run, an average xylitol production was observed of 35.1 g l−1 and average xylitol yield and productivity of 0.58 g g−1 and 0.49 g l−1 h−1, respectively.  相似文献

Gordonia alkanivorans strain 1B is able to desulfurize dibenzothiophene (DBT) to 2-hydroxybiphenyl (2-HBP), the final product of the 4S pathway. However, both the cell growth and the rate of desulfurization can be largely affected by the nutrient composition of the growth medium due to cofactor requirements of many enzymes involved in the biochemical pathways. In this work, the effect of several metal ions on the growth and DBT desulfurization by G. alkanivorans was studied. From all the metal ions tested, only the absence of zinc significantly affected the cell growth and the desulfurization rate. By increasing the concentration of Zn from 1 to 10 mg L−1, 2-HBP productivity was improved by 26%. The absence of Zn2+, when sulfate was also used as the only sulfur source, did not cause any difference in the bacterial growth. Resting cells grown in the presence of Zn2+ exhibited a 2-HBP specific productivity of 2.29 μmol g−1 (DCW) h−1, 7.6-fold higher than the specific productivity obtained by resting cells grown in the absence of Zn2+ (0.30 μmol g−1 (DCW) h−1). These data suggests that zinc might have a key physiological role in the metabolism of DBT desulfurization.  相似文献

In this work, the production of 1,3-propanediol from glucose and molasses was studied in a two-step process using two recombinant microorganisms. The first step of the process is the conversion of glucose or other sugar into glycerol by the metabolic engineered Saccharomyces cerevisiae strain HC42 adapted to high (>200 g l−1) glucose concentrations. The second step, carried out in the same bioreactor, was performed by the engineered strain Clostridium acetobutylicum DG1 (pSPD5) that converts glycerol to 1,3-propanediol. This two-step strategy led to a flexible process, resulting in a 1,3-propanediol production and yield that depended on the initial sugar concentration. Below 56.2 g l−1 of sugar concentration, cultivation on molasses or glucose showed no significant differences. However, at higher molasses concentrations, glycerol initially produced by yeast could not be totally converted into 1,3-propanediol by C. acetobutylicum and a lower 1,3-propanediol overall yield was observed. In our hand, the best results were obtained with an initial glucose concentration of 103 g l−1, leading to a final 1,3-propanediol concentration of 25.5 g l−1, a productivity of 0.16 g l−1 h−1 and 1,3-propanediol yields of 0.56 g g−1 glycerol and 0.24 g g−1 sugar, which is the highest value reported for a two-step process. For an initial sugar concentration (from molasses) of 56.2 g l−1, 27.4 g l−1 of glycerol were produced, leading to 14.6 g l−1 of 1.3-propanediol and similar values of productivity, 0.15 g l−1 h−1, and overall yield, 0.26 g g−1 sugar.  相似文献

Batch culture kinetics of the red yeast, Xanthophyllomyces dendrorhous SKKU 0107, revealed reduction in biomass with glucose and lower intracellular carotenoid content with fructose. Figures were different when compared to sucrose, which is a disaccharide of glucose and fructose. In contrast, specific growth rate constant stayed between 0.094~0.098 h−1, irrespective of the carbon sources employed. Although the uptake rate of glucose was found to be 2.9-fold faster than that of fructose, sucrose was found to be a more suitable carbon source for the production of carotenoids by the studied strain. When sugar cane molasses was used, both the specific growth rate constant and the intracellular carotenoid content decreased by 27 and 17%, respectively. Compared with the batch culture using 28 g/L sugar cane molasses, fed-batch culture with the same strain resulted in a 1.45-fold higher cell yield together with a similar level of carotenoid content in X. dendrorhous SKKU 0107.  相似文献

Xanthomonas campestris pv phaseoli produced an extracellular endoinulinase (9.24 ± 0.03 U mL−1) in an optimized medium comprising of 3% sucrose and 2.5% tryptone. X. campestris pv. phaseoli was further subjected to ethylmethanesulfonate mutagenesis and the resulting mutant, X. campestris pv. phaseoli KM 24 demonstrated inulinase production of 22.09 ± 0.03 U mL−1 after 18 h, which was 2.4-fold higher than that of the wild type. Inulinase production by this mutant was scaled up using sucrose as a carbon source in a 5-L fermenter yielding maximum volumetric (21,865 U L−1 h−1) and specific (119,025 U g−1 h−1) productivities of inulinase after 18 h with an inulinase/invertase ratio of 2.6. A maximum FOS production of 11.9 g L−1 h−1 and specific productivity of 72 g g−1 h−1 FOS from inulin were observed in a fermenter, when the mutant was grown on medium containing 3% inulin and 2.5% tryptone. The detection of mono- and oligosaccharides in inulin hydrolysates by TLC analysis indicated the presence of an endoinulinase. This mutant has potential for large-scale production of inulinase and fructooligosaccharides.  相似文献

Because engineering of the 101.016-bp megaplasmid pKB1 of Gordonia westfalica Kb1 failed due to the absence of an effective transfer system, pKB1 was transferred by conjugation from G. westfalica Kb1 to a kanamycin-resistant mutant of Rhodococcus opacus PD630 at a frequency of about 6.2 × 10−8 events per recipient cell. Furthermore, pKB1 was transferred to G. polyisoprenivorans strains VH2 and Y2K and to Mycobacterium smegmatis by electroporation at frequencies of 5.5 × 103, 1.9 × 103, and 8.3 × 102 transformants per microgram plasmid DNA. The pKB1-encoded cadmium resistance gene cadA was used for selection in these experiments. Recombinant pKB1-containing G. polyisoprenivorans VH2 and M. smegmatis were then used to engineer pKB1. A kanamycin resistance cassette was inserted into the pKB1-encoded cadA gene, ligated to suicide plasmid pBBR1MCS-5, and the resulting plasmid was electroporated into plasmid-harboring strains. Homologous recombination between cadA on suicide plasmid and the respective sequence in pKB1 led to its integration into pKB1. Thus, two selection markers were accommodated in pKB1 to monitor plasmid transfer into Gordonia and related taxa for analysis of genes essential for rubber degradation and others. In this study, two transfer methods for large plasmids and strategies for engineering of pKB1 were successfully applied, thereby, extending the tool box for Gordonia.  相似文献

In the present paper, the degradation of phenanthrene, a model polycyclic aromatic hydrocarbon compound, by the Mycobacterium strain 6PY1 was optimized in a biphasic culture medium. The optimization and modeling were performed using the design of experiments methodology. The temperature, the silicone oil/mineral salts medium volume ratio, and the initial cell concentration, were used as the central composite design parameters. In all experiments, the phenanthrene was degraded to undetectable levels. Response surface methodology was successfully employed to derive an empirical model describing the rate and time of degradation and to deduce the optimal degradation conditions. As a result of the optimization processes, the optimal responses for the degradation rate, the volumetric degradation rate, and the 90% degradation time were estimated to be 0.172 mg h−1, 22 mg l−1 h−1, and 18 h, respectively.  相似文献

Lipid and docosahexaenoic acid (DHA) accumulation into Schizochytrium G13/2S was studied under batch and continuous culture. Different glucose and glutamate concentrations were supplemented in a defined medium. During batch cultivation, lipid accumulation, 35% total fatty acids (TFA) occurred at the arithmetic growth phase but ceased when cell growth stopped. When continuous culture was performed under different glutamate concentrations, nitrogen-growth-limiting conditions induced the accumulation of 30–28% TFA in Schizochytrium. As the dilution rate decreased from 0.08 to 0.02 h−1, both cell dry weight and TFA content of the cell increased. Under a constant dilution rate of 0.04 h−1, carbon-limiting conditions decreased the TFA to 22%. Fatty acid profile was not affected by the different nutrient concentrations provided during continuous culture. Consequently, lipid accumulation can be induced through the carbon and nitrogen source concentration in the medium to maximise the TFA and subsequently DHA productivity by this microorganism.  相似文献

Osmotic stress restricts glycolytic flux, growth (rate and yield), d-lactate productivity, and d-lactate tolerance in Escherichia coli B strain SZ132 during batch fermentation in mineral salts medium with 10% (w/v) sugar. Addition of 1 mm betaine, a non-metabolized protective osmolyte, doubled cell yield, increased specific productivity of d-lactate and glycolytic flux by 50%, and tripled volumetric productivity (from 8.6 to 25.7 mmol l−1 h−1; 0.8 to 2.3 g l−1 h−1). Glycolytic flux and specific productivity in mineral salts medium with betaine exceeded that in Luria broth, substantially eliminating the need for complex nutrients during d-lactate production. In mineral salts medium supplemented with betaine, SZ132 produced approximately 1 mol d-lactate (90 g) per 100 g sugar (glucose or sucrose). The role played by a bacterial community composed ofPseudomonas putida, strain 21;Pseudomonas stutzeri, strain 18; andPseudomonas sp., strain 5, and by physical and chemical factors in the degradation of CN and SCN was studied. It was shown that the degradation of CN is determined both by the action of bacteria and by abiotic physical and chemical factors (pH, O2, temperature, the medium agitation rate, etc.). The contribution of chemical degradation was found to increase drastically at pH below 9.0; when air was blown through the medium (irrespective of the pH value); under active agitation of the medium; and when the medium surface interfacing air was increased. Even at elevated pH values (9.0-9.2), suboptimal for bacterial growth, the microbial degradation could account for at most 20–25 mg/1 of CN, regardless of its initial concentration. When CN and SCN were concurrently present in the medium, the former compound was the first to be degraded by microorganisms. The rate of bacterial degradation of SCN under continuous cultivation in a chain of reactors was found to depend on its concentration, the medium flow rate, agitation rate, and the pattern of carbon source supply and could exceed 1 g/(l day). CN and SCN are utilized by bacteria solely as nitrogen sources. The mechanism of CN and SCN degradation by the microbial community is discussed. Deceased.  相似文献

A new bacterial strain producing succinic acid was enriched from bovine rumen content. It is facultatively anaerobic, belongs to the family Pasteurellaceae and has similarity to the genus Mannheimia. In batch cultivations with D-glucose or sucrose the strain produced up to 5.8 g succinic acid l−1 with a productivity and a yield of up to 1.5 g l−1 h−1 and 0.6 g g−1, respectively. With crude glycerol up to 8.4 g l−1, 0.9 g l−1 h−1 and 1.2 g g−1 were obtained. The behavior of Streptomyces peucetius var. caesius N47 was studied in a glucose limited chemostat with a complex cultivation medium. The steady-state study yielded the characteristic constants μ max over 0.10 h−1, Y XS 0.536 g g−1, and mS 0.54 mg g−1 h−1. The product of secondary metabolism, ɛ-rhodomycinone, was produced with characteristics Y PX 12.99 mg g−1 and m P 1.20 mg g−1 h−1. Significant correlations were found for phosphate and glucose consumption with biomass and ɛ-rhodomycinone production. Metabolic flux analysis was conducted to estimate intracellular fluxes at different dilution rates. TCA, PPP, and shikimate pathway fluxes exhibited bigger values with production than with growth. Environmental perturbation experiments with temperature, airflow, and pH changes on a steady-state chemostat implied that an elevation of pH could be the most effective way to shift the cells from growing to producing, as the pH change induced the biggest transient increase to the calculated ɛ-rhodomycinone flux.  相似文献
