Based on analysis of variation at ten allozyme loci in three age groups (25-35, 40-80, and more than 100 years of age) of plants and in seed embryos, demographic dynamics of the gene pools was studied in a small (60.5 ha) isolated relict population of chalk pine Pinus sylvestris var. cretacea Kalenicz. ex Kom. from the steppe zone of Ukraine. The observed grenotype proportions in these tree groups were shown to fit Hardy-Weinberg expectations, while in the embryos of their seeds, an excess of homozygotes was observed at five to nine loci. The mean observed heterozygosity in the sample of old (> 100 years of age) trees (H(O) = 0.225) was substantially lower than in trees of the two other age groups (H(O) = 0.307; 0.311), but significantly higher than in the corresponding embryo samples (H(O) = 0.183-0.207). No allele and genotype heterogeneity of the maternal trees and embryos of their seeds was found. However, heterogeneity was high when the progeny of trees of different ages, particularly in pairs with old trees, were compared.  相似文献

Samples of trees and seed embryos of five Pinus kochiana Klotzsch ex Koch populations in Crimea were comparatively studied at 12 allozymic loci for its genetic variability and mating system. It has been shown that the embryos of seeds reproduce the allelic diversity of maternal plants, but the level of their observed heterozygosity (Ho) is significantly lower, i.e., 0.286 vs. 0.189, respectively. Unlike maternal trees, embryos are characterized by a significant divergence between the observed distribution of genotypes and the theoretically expected one according to the Hardy–Weinberg law for the majority of analyzed loci. The frequency of cross pollination in populations varied from 0.384 to 0.673 according to single-locus (ts) estimates and from 0.639 to 0.841 according to multilocus (tm) estimates.  相似文献

Allozyme variation at ten polymorphic loci and mating system was studied in three small isolated relict populations (4.4 to 22 ha) and in three artificial stands of Pinus sylvestris from the Kremenets Hill Ridge and Maloe Poles’e. It was established that the mean heterozygosity of 130 to 140 year-old trees from natural populations (H O = 0.288; H E = 0.277) was substantially lower, compared to 30 to 40 year-old trees from artificial stands (H O = 0.358; H E = 0.330). The observed heterozygosity of seed embryos (H O = 0.169 and 0.180) was substantially lower than of the mature trees from populations and artificial stands, respectively. In the embryo samples, irrespectively of the forest stand origin, substantial hetedrozygote deficiency was observed (at six to eight loci), compared to the Hardy-Weinberg expectations. The proportion of cross pollination in the populations and artificial stands was low, t m = 0.588 to 0.721; and t m = 0.455 to 0.837, respectively.  相似文献

Reactive oxygen (ROS) and nitrogen (RNS) species play a signaling role in seed dormancy alleviation and germination. Their action may be described by the oxidative/nitrosative “window/door”. ROS accumulation in embryos could lead to oxidative modification of protein through carbonylation. Mature apple (Malus domestica Borkh.) seeds are dormant and do not germinate. Their dormancy may be overcome by 70–90 days long cold stratification. The aim of this work was to analyze the relationship between germinability of embryos isolated from cold (5 °C) or warm (25 °C) stratified apple seeds and ROS or nitric oxide (NO) production and accumulation of protein carbonyl groups. A biphasic pattern of variation in H2O2 concentration in the embryos during cold stratification was detected. H2O2 content increased markedly after 7 days of seeds imbibition at 5 °C. After an additional two months of cold stratification, the H2O2 concentration in embryos reached the maximum. NO production by the embryos was low during entire period of stratification, but increased significantly in germination sensu stricto (i.e. phase II of the germination process). The highest content of protein carbonyl groups was detected after 6 weeks of cold stratification treatment. Fluctuation of H2O2 and protein carbonylation seems to play a pivotal role in seed dormancy alleviation by cold stratification, while NO appears to be necessary for seed germination.  相似文献

The population genetics and mating system of the understory tropical rain forest tree Psychotria faxlucens were studied at two plots in Los Tuxtlas, Veracruz, México, on the Gulf of México coast. This species is distylous and is pollinated by moths, mainly small hawkmoths. The seeds are dispersed by gravity and by frugivorous birds. Controlled pollinations indicate that the trees are self-incompatible and that intramorph pollinations have lower compatibilities than intermorph pollinations. The pollen: ovule ratio is high, suggesting obligate xenogamy. Using electrophoretic allozyme methods we found that eight out of 20 loci were polymorphic (P = 0.400), the observed heterozygosity (H) was 0.198, and the mean expected heterozygosity (H) was 0.495, both relatively high values compared with that reported for tropical trees. The genetic differentiation between the two plots is low, as shown by the heterogeneity in allele frequencies and the Fst (mean Fst seedlings = 0.031, mean Fst adults = 0.026), although for some loci the plot differentiation is statistically significant. The studied populations are near Hardy-Weinberg proportions, both for seedlings (mean F = 0.128) and adults (mean F = 0.075). From the fixation rate, an indirect estimate of the outcrossing rate at equilibrium gave a mean of t = 0.898 for plot 1 and 0.685 for plot 2. Direct single loci and multiloci outcrossing rate estimates were generally not statistically different from 1.0.  相似文献

We developed microsatellite markers for the fig-pollinating wasp Blastophaga nipponica Grandi using a dual-suppression-PCR technique. Twenty-one candidates of microsatellite loci were obtained, of which 15 yielded scorable patterns. The degree of polymorphism for the 15 loci was further characterized using summary statistics describing the genetic variation in 60 individuals from three natural populations in Japan. All 15 loci were polymorphic and yielded 2–27 alleles per locus. Overall observed heterozygosity (H O) and expected heterozygosity (H E) were 0.465 and 0.631, respectively. As expected, based on the inbreeding tendency of this species, the mean inbreeding coefficient (F IS) was high (= 0.255). These markers will contribute to studies on the population structure of this species.  相似文献

Polymorphism of young (14–16 years), middle-aged (70–80 years) and old (120–150 years) plants and their seed embryos have been studied using 10 and 20 allozyme loci from a population of Pinus Pallasiana D. Don from Mountain Crimea. It was determined that the old-aged trees had significantly lower level of expected heterozygosity of embryos in comparison with young trees. Supernumerary homozygotation of the embryos was caused by low level of cross-pollination in three studied samples of plants (t m = 0.537–0.637).  相似文献

The silk protein sericin has been identified as a potent antioxidant in mammalian cells. This study was conducted to examine the effects of sericin on preimplantation development and quality of bovine embryos cultured individually. When two-cell-stage embryos were cultured individually for 7 days in CR1aa medium supplemented with 0, 0.1, 0.5, or 1% sericin, rates of total blastocyst formation and development to expanded blastocysts from embryos cultured with 0.5% sericin were higher (P < 0.05) than those from embryos cultured with 0 or 1% sericin. When embryos were cultured individually for 7 days in the CR1aa medium supplemented with 0 or 0.5% sericin under two oxidative stress conditions (50 or 100 μm H2O2), the addition of sericin significantly improved the blastocyst formation rate of embryos exposed to 100 μm H2O2. However, the protective effect of sericin was not observed in development of embryos exposed to 50 μm H2O2. When embryos were exposed to 100 μm H2O2 during culture, the DNA fragmentation index of total blastocysts from embryos cultured with 0.5% sericin was lower than blastocysts derived from embryos cultured without sericin (4.4 vs. 6.8%; P < 0.01). In conclusion, the addition of 0.5% sericin to in vitro culture medium improved preimplantation development and quality of bovine embryos cultured individually by preventing oxidative stress.  相似文献

Ten polymorphic microsatellite markers were isolated from the dwarf bamboo species Sasa cernua and Sasa kurilensis. The applicability of these markers was confirmed by genotyping of open‐pollinated seeds and leaf samples from natural populations. Genotypes of seeds from each culm shared at least one allele from the maternal parent without contradiction. All 10 loci were polymorphic in S. cernua with 2–15 alleles (average HE = 0.532). Eight loci were polymorphic in S. kurilensis with 2–10 alleles (average HE = 0.532). These markers will be useful in detailing the extent of clonal and sexual reproduction in these species.  相似文献

The comparative analysis of genetic variation at 12 polymorphic isozyme loci, as well as the mating system, has been carried out in mature trees and their seed progenies in three small localities of Pinus brutia var. stankewiczii Sukacz. in the region between the town of Sudak and settlement of Novyi Svet in Crimea. We found that embryos maintain the same allelic diversity as parent trees; however, their observed heterozygosity, on average, is lower by 37.4%. The significant deviation of genotype distribution from the expected ratios caused by the deficiency of heterozygotes was observed at 8 out of 12 loci. The multilocus estimate of the out-crossing rate (t m ) in populations varied from 68.9 to 94.9% with the average of 80.7%.  相似文献

The risks of gene flow between interfertile native and introduced plant populations are greatest when there is no spatial isolation of pollen clouds and phenological patterns overlap completely. Moreover, invasion probabilities are further increased if introduced populations are capable of producing seeds by selfing. Here we investigated the mating system and patterns of pollen-mediated gene flow among populations of native ash (Fraxinus excelsior) and mixed plantations of non-native ash (F. angustifolia and F. excelsior) as well as hybrid ash (F. excelsior × F. angustifolia) in Ireland. We analysed the flowering phenology of the mother trees and genotyped with six microsatellite loci in progeny arrays from 132 native and plantation trees (1493 seeds) and 444 potential parents. Paternity analyses suggested that plantation and native trees were pollinated by both native and introduced trees. No signs of significant selfing in the introduced trees were observed and no evidence of higher male reproductive success was found for introduced trees compared with native ones either. A small but significant genetic structure was found (φft=0.05) and did not correspond to an isolation-by-distance pattern. However, we observed a significant temporal genetic structure related to the different phenological groups, especially with early and late flowering native trees; each phenological group was pollinated with distinctive pollen sources. Implications of these results are discussed in relation to the conservation and invasiveness of ash and the spread of resistance genes against pathogens such as the fungus Chalara fraxinea that is destroying common ash forests in Europe.  相似文献

Changes in H2O2 and the main antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR), in endospermic and embryonic tissues were studied in developing and artificially dried Jatropha curcas seeds. Immature seeds were desiccation-tolerant at 80 days after flowering, as they were able to germinate fully after artificial drying on silica gel had reduced their water content to 10–12% of fresh weight. In both endospermic and embryonic tissues, H2O2 level and, consequently, lipid peroxide content, decreased during seed development as well as after artificial dehydration of developing seeds. All examined antioxidant enzymes except DHAR showed a decrease in total activity in mature stages as compared with early stages. Expression analysis of SOD genes revealed that the decrease in total SOD activities was related to the decrease in Cu/Zn-SOD expression, while the continuous activity of SOD during maturation was related to an increase in Mn-SOD expression. Artificial drying resulted in increased SOD and DHAR activity, irrespective of the developmental stage. Our results revealed weak participation of CAT and APX in H2O2 scavenging, as well as no significant alterations in GR activities either during maturation or after artificial drying. Changes in SOD and GR isoenzyme patterns occurred during maturation-related drying, but not after artificial drying. These results highlight the role of ascorbate-glutathione cycle enzymes (DHAR and GR) in H2O2 scavenging during maturation or after artificial drying of developing J. curcas seeds.  相似文献

The impact of oxidative stress on the in vitro development of bovine embryos in synthetic oviduct fluid medium (mSOF) was assessed by using H2O2 as a stress inducer. In a preliminary experiment, a chemiluminescent method was used to measure the antioxidative capacity of the mSOF culture medium. Pyruvate was the mSOF component displaying the highest H2O2 degrading ability. Essential and nonessential amino acids also significantly reduced the H2O2 concentration, whereas lactate and glutamine were ineffective. The effect on further development of a short exposure of zygotes, 9–16‐cell stage embryos and blastocysts to 0 M; 107 M ; 106 M, and 105 M H2O2 in pyruvate‐free mSOF was evaluated. Developmental rates of the H2O2‐treated zygotes to the 5–8‐cell or blastocyst stages and survival of H2O2‐treated blastocysts were reduced in a dose‐dependent manner whereas the 9–16‐cell embryos were unaffected by those treatments. Blastocysts treated with H2O2 also tended to have lower numbers of bisbenzimide‐stained nuclei and showed increased nuclear fragmentation. Including pyruvate in the mSOF culture medium during a 105 M H2O2 pulse highly reduced the H2O2 concentration as measured by chemiluminescence and improved zygote and blastocyst development, but failed to prevent blastocyst nuclei degradation. These experiments suggest that bovine embryos show developmental change in sensitivity to exogenous H2O2, the 9–16‐cell embryos being more resistant than zygotes and blastocysts and that H2O2 and its toxic effects can be attenuated by including pyruvate in the medium. Hydrogen peroxide (H2O2) promotes seed germination of cereal plants and ascorbic acid which acts as antioxidant suppresses the germination of wheat seeds, but the role of H2O2 scavenging on germination during seed maturation has not been demonstrated. We investigated relationship of germination, ascorbate, H2O2 scavenging enzymes and sensitivity to ascorbic acid (AsA) maturing seeds of two typical wheat (Triticum aestivum L.) cultivars, cvs. Shirogane-Komugi and Norin61. Shirogane-Komugi had marked high germination ability than Norin61 during seed maturation. Although the H2O2 content had no difference in the two culti-vars, sensitivity to AsA of Norin61 seeds was higher than that of Shirogane-Komugi seeds during seed maturation. The sensitivity to AsA closely correlated with germination characteristic in the two cultivars. Especially, at 28 days after pollination (DAP), sensitivity to AsA in Norin61 seeds was remarkably high. At that stage, no significant differences were observed in endogenous AsA level, ascorbate peroxidase (APX, EC 1.11.1.11) and dehydroascorbate reductase (DHAR, EC 1.8.5.1) activities in the two cultivars. However, catalase (CAT, EC 1.11.1.6) activity and CAT mRNA in Norin61 were remarkably higher than in Shirogane-Komugi. Sensitivity to AsA at 35 and 42 DAPs kept high levels in Norin61, and endogenous AsA and CAT activity in the seeds were significantly higher than in Shirogane-Komugi. A comparative analysis of the gene pool state in natural populations and planted stands of Norway spruce and the degree of their infestation by the bark beetle in the Moscow region was conducted taking into account the dynamic state of communities (4 populations, 148 samples, 24 isoenzyme loci). The degree of infestation by the bark beetle of conditionally native communities is 0%; for planted stands, it is 90–100%; and for a short-term community, it is 15–20%. The comparison of “healthy” populations and those infested with bark beetle by average values of observed heterozygosity (H O) detected no significant differences. However, the test on allelic frequency heterogeneity demonstrated the difference of planted stands from conditionally native populations both by three loci (Fe-2, Idh-1, Mdh-3) and by the totality of 18 polymorphic isoenzyme loci; the short-term population differs from conditionally native population only by two loci. The value of the inbreeding coefficient by the Idh-1 locus is significantly higher in both populations infested with the bark beetle than in “healthy” populations. The results of conducted studies demonstrate the necessity of continuation of the study on the gene pool state in Norway spruce populations owing to the degree of their infestation by the bark beetle along with the study on the dynamic state of the communities; this can provide a key to solving the problem of the forest preservation from pests.  相似文献

  • The dormancy of seeds of upland cotton can be broken during dry after‐ripening, but the mechanism of its dormancy release remains unclear.
  • Freshly harvested cotton seeds were subjected to after‐ripening for 180 days. Cotton seeds from different days of after‐ripening (DAR) were sampled for dynamic physiological determination and germination tests. The intact seeds and isolated embryos were germinated to assess effects of the seed coat on embryo germination. Content of H2O2 and phytohormones and activities of antioxidant enzymes and glucose‐6‐phosphate dehydrogenase were measured during after‐ripening and germination.
  • Germination of intact seeds increased from 7% upon harvest to 96% at 30 DAR, while embryo germination improved from an initial rate of 82% to 100% after 14 DAR. Based on T50 (time when 50% of seeds germinate) and germination index, the intact seed and isolated embryo needed 30 and 21 DAR, respectively, to acquire relatively stable germination. The content of H2O2 increased during after‐ripening and continued to increase within the first few hours of imbibition, along with a decrease in abscisic acid (ABA) content. A noticeable increase was observed in gibberellic acid content during germination when ABA content decreased to a lower level. Coat removal treatment accelerated embryo absorption of water, which further improved the accumulation of H2O2 and changed peroxidase content during germination.
  • For cotton seed, the alleviation of coat‐imposed dormancy required 30 days of after‐ripening, accompanied by rapid dormancy release (within 21 DAR) in naked embryos. H2O2 acted as a core link between the response to environmental changes and induction of other physiological changes for breaking seed dormancy.

This study presents a comparative hierarchical analysis of variance applied to three classes of molecular markers within the blue marlin (Makaira nigricans). Results are reported from analyses of four polymorphic allozyme loci, four polymorphic anonymously chosen single-copy nuclear DNA (scnDNA) loci, and previously reported restriction fragment length polymorphisms (RFLPs) of mitochondrial DNA (mtDNA). Samples were collected within and among the Atlantic and Pacific Oceans over a period of several years. Although moderate levels of genetic variation were detected at both polymorphic allozyme (H = 0.30) and scnDNA loci (H = 0.37), mtDNA markers were much more diverse (h = 0.85). Allele frequencies were significantly different between Atlantic and Pacific Ocean samples at three of four allozyme loci and three of four scnDNA loci. Estimates of allozyme genetic differentiation (θO) ranged from 0.00 to 0.15, with a mean of 0.08. The θO values for scnDNA loci were similar to those of allozymes, ranging from 0.00 to 0.12 with a mean of 0.09. MtDNA RFLP divergence between oceans (θO = 0.39) was significantly greater than divergence detected at nuclear loci (95% nuclear confidence interval = 0.04–0.11). The fourfold smaller effective population size of mtDNA and male-mediated gene flow may account for the difference observed between nuclear and mitochondrial divergence estimates.  相似文献

Hydrogen peroxide (H2O2), a second messenger, plays a vital role in seed germination and plant growth, development as well as the acquisition of stress tolerance, while hydrogen sulfide (H2S) is considered as a new emerging cell signal molecule in higher plants. In the present study, soaking of H2O2 greatly improved germination percentage of Jatropha curcas seeds, stimulated the increase of l-cysteine desulfhydrase activity, which in turn induced accumulation of H2S. On the contrary, pretreatment of aminooxyacetic acid (AOA), inhibitor of H2S biosynthesis, eliminated H2O2 stimulated the increase of activity of l-cysteine desulfhydrase and accumulation of H2S as well as improvement of germination percentage. In addition, exogenously applied H2S also could improve germination percentage of seeds of J. curcas. These results suggested that pretreatment of H2O2 could improve germination percentage of J. curcas seeds and this improvement was mediated by H2S.  相似文献

Young healthy cotyledon and leaf explants of Rhinacanthus nasutus (L.) Kurz. were incubated on Murashige and Skoog (MS) medium supplemented with 1.0–5.0 mg/l 2, 4-dichlorophenoxyacetic acid (2,4-D) either alone or in combination with 0.3–1.5 mg/l indole-3-butyric acid (IBA). The optimum callus induction (100 %) was observed from cotyledon explants on MS medium supplemented with 4 mg/l 2, 4-D and 0.5 mg/l IBA. The friable, embryogenic callus when subcultured on half strength MS medium supplemented with IBA (3.0–5.0 mg/l) produced several somatic embryos at various stages of development (globular, heart, torpedo) after 45 days of culture. The highest frequency of callus embryogenesis was observed on ½MS medium supplemented with 4.0 mg/l IBA. Moreover, 47 % of incubated callus responded with a mean number of 16.3 somatic embryos per gram callus. For germination, somatic embryos at the torpedo stage were isolated and subcultured on ½MS medium supplemented with 0.5 mg/l each of 6-benzyladenine and indole-3-acetic acid. After 45 days of culture, plantlets developed with mean lengths of 3.8 cm. Somatic embryos at the torpedo stage were collected and suspended in a matrix of MS medium containing sodium alginate (3 % W/V), dropped into 100 mM calcium chloride (CaCl2·2H2O) solution for the production of synthetic seeds. Optimum growth ability of synthetic seed was obtained on MS medium supplemented with 0.2 mg/l gibberellic acid (GA3). Well developed healthy plantlets derived from somatic embryos and synthetic seeds were hardened and successfully transplanted to soil.  相似文献

The genetic diversity, subdivision, and differentiation of nine populations of Norway spruce (Picea abies (L.) Karst.) in Ukrainian Carpathians were studied using electrophoretic analysis of variability of enzyme systems in 346 trees aged from 80 to 150 years. Based on electrophoretic fractionation of enzymes extracted from seed endosperms in vertical slabs of 7.5% polyacrylamide gel, 20 loci of nine enzyme systems (ADH, ACP, DIA, GDH, GOT, MDH, LAP, FDH, SOD) were identified, and 71 allele variant were revealed. Each tree was heterozygous on average in 15.8% of its genes. The populations were characterized by low subdivision (F ST = 0.017) and differentiation (D N = 0.005). The main contribution to heterogeneity of population genetic structure was made by loci Dia-3, Lap-1, and Sod-3. Clustering and multivariate analysis revealed no observed trends in geographical or altitudinal position of the populations.  相似文献
