Chronic cadmium toxicity tests were carried out withChironomus riparius Meigen using a static-with-replacement exposure procedure. Significant reductions in larval development, survival and production were detected at 0.15 mg Cd l–1 a value approximately 5000 times lower than the 48 h LC50 for fourth instar larvae. The first instar appears to be the most critical stage in terms of sensitivity, with the number of adults eventually emerging reflecting first instar mortality. Male emergence was slightly delayed at 0.15 mg Cd l–1. The results of this study illustrate the importance of chronic toxicity tests in identifying sensitive life cycle stages and explaining pollutant-induced stress.  相似文献

Laboratory studies were conducted to determine the potential molluscicidal activity of five Nigerian plant species used in alternative medicine practice as anthelmintics. Laboratory-bred adult Biomphalaria pfeifferi, the snail host of Schistosoma mansoni in Nigeria, and their viable 0- to 24-hour-old egg masses were separately exposed for 24 hours, in replicated bioassays, to 7.81–4 000 mg l?1 concentrations of crude ethanolic extracts of the leaves and roots of Annona senegalensis, Anogeissus leiocarpus, Crotalaria retusa, Dalbergia sissoo and Vernonia amygdalina. Fruits and stem bark of D. sissoo were also investigated. The 24-hour LC50 and LC90 values of extracts for target organisms were calculated using probit analysis. Susceptibility of target organisms to extracts varied with the plant species and morphological part. Egg masses and adult snails were most susceptible to D. sissoo fruit extract (LC90 values of 89.29 and 74.33 mg l?1, respectively) and least susceptible to V. amygdalina leaf extract (LC90 values of 11 864.09 and 6 241.61 mg l?1, respectively). Concentration-dependent behavioural responses and embryo deaths at the gastrula/exogastrula and/or prehatch snail stages of development were observed in exposed snails and egg masses, respectively. Tested extracts of A. senegalensis, A. leiocarpus, C. retusa and V. amygdalina did not show appreciable toxicities to B. pfeifferi egg masses or adults. For demonstrating significant molluscicidal activity (LC90 < 100 mg l?1) and toxicities to egg masses, we recommend further studies on the ethanolic extracts of D. sissoo fruits and roots.  相似文献

High percentages of micro-calli and micro-derived embryos were produced from isolated asparagus microspores at late uninucleate stage on MS liquid medium supplemented with 1.0 mg l–1 2,4-D and 0.5 mg l–1 BA. Two types of calli, namely compact callus (CC) and loose callus (LC), were found. Plantlets were regenerated via organogenesis, when these calli were transferred onto MS solid medium supplemented with 1.0 mg l–1 BA and 0.2 mg l–1 IBA 6 weeks. Embryos were produced from liquid cultured microspores, or from solid cultured micro-calli. The frequencies of haploid plant production from organogenesis and embryogenesis were compared. Effects of plant growth regulators on callus production, plantlet regeneration, and haploid plant production were tested. Here, we report the establishment of an efficient, in vitro, shoot organogenesis, regeneration system for soybeans [Glycine max (L.) Merr.]. Mature soybean seeds were soaked for 24 h, the embryonic tips were collected and cultured on MSB5 medium supplemented with 3.5 mg l–1 N6-benzylaminopurine (BAP) for 24 h, and explants were transferred to MSB5 medium supplemented with 0.2 mg l–1 BAP and 0.2 mg l–1 indolebutyric acid. Use of embryonic tips yielded a higher regeneration frequency (87.7%) than regeneration systems using cotyledonary nodes (40.3%) and hypocotyl segments (56.4%) as starting materials. Regenerated embryonic tips were inoculated with Agrobacterium tumefaciens strain EHA105, which contains the binary vector pCAMBIA2301, and cultured for 20 h. Our results showed that the T-DNA transfer efficiency reached up to 78.2% and the transformation efficiency reached up to 15.8%. Population dynamics of Chaoborus flavicans larvae of various instars was studied from November 1986 to December 1987 in a eutrophic, fish-free pond, Japan. First and 2nd instar larvae were observed from late April to late October, indicating a reproductive period of about half a year. C. flavicans overwintered in the 4th instar larvae. In water column samples, total density of all instars was 680–23680 m-2, and pupal density 0–2600 m-2; larvae of the 1st, 2nd, and 3rd instars showed 5–6 density peaks in 1987, suggesting that 5–6 generations occur during a year (peaks of the 4th instar larvae were not clear, probably due to their longer development than those of younger instars). In sediment samples, no 1st and 2nd instar larvae were found, 3rd instar larvae were found occasionally but density of the 4th instar larvae was 280–18600 m-2, and pupal density varied between 0–502 m-2. Fouth instar larvae accumulated in sediment in the cold season and in the water column in the warm season; high temperature and low oxygen concentration were the most important factors limiting the distribution of larvae in the sediment in summer in the NIES pond. The dry weight of total C. flavicans larvae was 0.08–4.2 g m-2 in sediment samples and 24–599 μg l-1 (0.10–2.40 g m-2) in water column samples. Comparisons of maximum densities in the NIES pond in different years and in waters of different trophic status show that density is generally higher in eutrophic than in oligotrophic habitats. Nabis roseipennis Reuter nymphs demonstrated a preference for nuclear polyhedrosis virus (NPV) — infected over healthyAnticarsia gemmatalis Hübner larvae when offered a choice of larval prey in Petri dishes and on soybean. In Petri dishes, small (second-third instar) and large (fifth-sixth instar) nymphs attacked a significantly greater number of diseased than healthy larvae at all larval instars tested (first-fifth instars) and exposure periods (2, 5 and 24 h), except that at 2 h the number of 1st and 3rd instar larvae attacked by large nymphs did not differ significantly (P≤0.05). Nabis roseipennis caged with larvae on individual soybean plants in the greenhouse resulted in a generally low percentage of attack by small and large nymphs after 2 days, ranging from 5.6 to 36.7%. As in the Petri dishes, the nabids showed a significant preference for diseased larvae over healthy larvae attacked for all nabid and larval sizes on soybean, with the percentage of diseased larvae attacked ranging from 28.0 to 65.4% (P≤0.05). This preference for diseased larvae on soybean as well as in Petri dishes demonstrates that the preference was not due to the close proximity in which the host and prey were found in the Petri dishes. The preference for diseased larvae may be due to a reduction in a defensive response in late stages of disease. In this study, the mixture of mono- and di-rhamnolipids produced by Pseudomonas aeruginosa DS10-129 was characterized for its toxicity and modulatory effects on Cd availability to different bacteria. Gram-negative naturally bioluminescent Vibrio fischeri and recombinant bioluminescent Pseudomonas fluorescens, P. aeruginosa, Escherichia coli, and Gram-positive Bacillus subtilis were used as model organisms. Rhamnolipids reduced the bioluminescence of these bacteria in less than a second of exposure even in relatively low concentrations (30-min EC50 45–167 mg l−1). Toxicity of Cd to Gram-negative bacteria (30-min EC50 values 0.16 mg l−1 for E. coli, 0.96 mg l−1 for P. fluorescens, and 4.4 mg l−1 for V. fischeri) was remarkably (up to 10-fold) reduced in the presence of 50 mg l−1 rhamnolipids. Interestingly, the toxicity of Cd to Gram-positive B. subtilis (30-min EC50 value 0.49 mg l−1) was not affected by rhamnolipids. Rhamnolipids had an effect on desorption of Cd from soil: 40 mg l−1 rhamnolipids increased the water-extracted fraction of Cd twice compared with untreated control. However, this additionally desorbed fraction of Cd remained bound with rhamnolipids and was not available to bacteria. Hence, in carefully chosen concentrations (still effectively complexing heavy metals but not yet toxic to soil bacteria), rhamnolipids could be applied in remediation of polluted areas.  相似文献

Anthers of Morus indica L., with microspores at the uninucleate stage were cultured; and the influence of temperature and kinetin pretreatment on induction of androgenic calluses was examined. The effects of various pretreatments revealed that 24 h cold pretreatment increased the percentage of cultures inducing callus. First microspore division was observed after 16 to 20 days of culture. Th anthers split and developed embryogenic calluses on MB medium supplemented with NAA (0.5 mg l–1 and BA (1.0 mg l–1)) using 8% sucrose. Rhizogenesis was induced on medium supplemented with NAA and BA (each 0.5 mg l–1) with reduced myo-inositol (75 mg l–1). Cytological study of induced roots confirmed the haploid nature of calluses. Different type of embryos were initiated upon transfer of calluses to medium supplemented with NAA, BA (each 0.5 mg l–1), 2,4-d (1.0 mg l–1) and PVP (600 mg l–1). The concentrations of Cd, Pb, Cu and Zn inChironomus gr.thummi were determined for 4th instar larvae from the polluted Dyle River, tributary of the Scheldt River (Belgium). Comparison was made between larvae with deformed and normal menta. Deformed larvae showed higher overall metal concentrations than normal larvae. Especially Pb and Cu had higher concentrations in deformed larvae (16.22 mg kg–1 dry weight and 39.66 respectively) than in normal larvae (12.80 mg kg–1 dry weight and 35.70 respectively). No significant differences were found in the concentrations of Cd and Zn (mean [Cd] = 0.81 mg kg–1 dry weight and mean [Zn] = 313.12 mg kg–1 dry weight). There was no difference between the two larval groups as far as total length, dry weight and developmental stage of the imaginal discs are concerned.  相似文献

Successful application of microorganisms to heavy metal remediation depends on their resistance to toxic metals. This study contrasted the differences of tolerant mechanisms between Pb2+ and Cd2+ in Enterobacter sp. Microbial respiration and production of formic acid showed that Enterobacter sp. had a higher tolerant concentration of Pb (>1000 mg l−1) than Cd (about 200 mg l−1). Additionally, SEM confirmed that most of Pb and Cd nanoparticles (NPs) were adsorbed onto cell membrane. The Cd stress, even at low concentration (50 mg l−1), significantly enlarged the sizes of cells. The cellular size raised from 0.4 × 1.0 to 0.9 × 1.6 μm on average, inducing a platelet-like shape. In contrast, Pb cations did not stimulate such enlargement even up to 1000 mg l−1. Moreover, Cd NPs were adsorbed homogeneously by almost all the bacterial cells under TEM. However, only a few cells work as ‘hot spots’ on the sorption of Pb NPs. The heterogeneous sorption might result from a ‘self-sacrifice’ mechanism, i.e., some cells at a special life stage contributed mostly to Pb sorption. This mechanism, together with the lower mobility of Pb cations, caused higher microbial tolerance and removal efficiency towards Pb2+. This study sheds evident contrasts of bacterial resistance to the two most common heavy metals.  相似文献

We investigated the uptake, transport, storage and defence mechanisms in the freshwater crab, Potamonautes warreni, harbouring microbial gill infestations and exposed to increasing chronic (0.2, 0.5, 1.0 mg l–1) and acute (2.0 mg l–1) cadmium (Cd) concentrations under controlled laboratory conditions over a period of 21 days. Transmission electron microscopy and X-ray microanalysis revealed that the microbial gill fauna was eliminated on exposure to 0.2 mg Cd2+ l–1 and that Cd became increasingly adsorbed and incorporated into lamellar crystal deposits and permeated the cuticle of the gills of P. warreni. Degeneration of the apical membrane infoldings and vacuolation of epithelial cells occurred concurrently with pinocytosis, endocytosis and pronounced phagocytotic activity in the epithelia and haemal canal of the gills. Elevated Cd exposures (0.5 or 1.0 mg l–1) resulted in the swelling and dissociation of mitochondrial outer membranes together with an increase in transport of Cu, Cl and S by haemocytes in the haemal canal to epithelial tissues depleted in these elements. Cd also accumulated in tightly coiled concentric membrane whorls in the haemal canal, whereas the highest concentrations of Cd were found within aggregates of lysosome-like bodies in cuticulin-secreting cells of the gill stem. Chronic exposure to Cd induced increased fatigue and mild uncoordinated motor activity. In contrast, at an acute exposure of 2.0 mg l–1 over 48 h, P. warreni showed a time-specific rapid loss of motor function, although only mild cellular lesions occurred in the gill tissues. The significance of cellular changes in the gill epithelia and altered motor activity of P. warreni with increased waterborne Cd are discussed as potential biomarker responses in monitoring aquatic pollution.  相似文献

The toxicity of deltamethrin, a synthetic pyrethroid insecticide, was determined under standardized conditions (ISO, 1982) in neonates and juveniles of Daphnia magna. Neonates (6 to 24 h old) were more sensitive than juveniles (48 to 72 h old). The 24- and 48-h EC50s (immobilization) in neonates were 0.113 and 0.031 µg l–1, respectively. The toxicity of deltamethrin was highly toxic. The 96-h EC50 was in the ppt (µg l–1) range. Toxicity tests with Daphnia may be used to detect toxic residues in water and sediment in areas treated with deltamethrin and other highly toxic pyrethroid pesticides.  相似文献

A white rot basidiomycete, Phanerochaete chrysosporium, was immobilized on loofa sponge (FBILS) discs. It removed ca. 37 and 71 mg Cd (II) g−1 from 50 and 200 mg l−1 aqueous solutions and up to 89% of 4-chloroanisole from a 10 mg l−1 aqueous solution. FBILS are physically strong and chemically recalcitrant, resisting temperature, mechanical agitation, and variations in pH without alteration to shape, structure or texture.  相似文献

Addition of 20 μM salicylic acid to Saussurea medusa cell cultures at day 6 resulted in jaceosidin and syringin productions up to 95 mg l−1 and 631 mg l−1 which were, respectively, about 2.5- and 2.7-fold higher than in the control. The biomass was increased from 8 to 12 g l−1. Expression of chalcone synthase gene (chs) increased sharply after 12 h treatment and was sustained up to 48 h; chalcone isomerase gene (chi) expression reached a peak at 24 h and decreased after 48 h; and phenylalanine ammonia-lyase activity increased by 7.5-fold (96 U mg−1 protein) higher than in the control after 24 h. These results indicate that salicylic acid enhances the production of jaceosidin and syringin which is accompanied by induction of the related phenylpropanoid biosynthetic enzymes.  相似文献

A protoplast-to-plant regeneration system has been established for sweet potato (Ipomoea batatas (L.) Lam.) and its wild relative, I. lacunosa L. Viable protoplasts, isolated from preplasmolyzed stems and petioles of in vitro-grown plants, were cultured on liquid MS (Murashige & Skoog 1962) medium that supported cell division and colony formation. Embryogenic calli of sweet potato were induced on agar-solidified MS medium supplemented with 3% (w/v) sucrose, 50 mg l-1 casamino acids, 0.2–0.5 mg l-1 2,4-d, 1.0 mg l-1 kinetin and 1.0 mg l-1 ABA. On average, 3 plants were regenerated from a single sweet potato callus subcultured on semi-solid MS medium containing 3% (w/v) sucrose, 800 mg l-1 glutamine, 2.0 mg l-1 BA or 1.0 mg l-1 kinetin and 1.0 mg l-1 GA3. Embryogenic calli of I. lacunosa L. were initiated on semi-solid MS medium containing 0.2–0.5 mg l-1 IAA and 1.0–2.0 mg l-1 BA. Summary A method for obtaining a high frequency of haploid asparagus embryos through anther culture was developed. Flowers collected from plants in the field in July, August and September 1990, for the genotype G203, were stored at 5°C for 24 h. Anthers were placed on Murashige and Skoog medium (MS) containing 500 mg l –1 casein hydrolysate, 800 mg l–1 glutamine, 2 mg l –1 NAA, 1 mg l –1 BA and 5 % sucrose at 32 °C in the dark for three to four weeks to induce calli. Calli were then grown at 25 °C with a 16 h photoperiod for three to four weeks. Developing embryos and calli were transferred to embryo maturation medium, MS containing 6% sucrose, 0.1 mg l –1 NAA, 0.1 mg l –1 kinetin and 0.65 mg l –1 ancymidol, for four weeks. More than 50% of the recovered mature embryos germinated on MS containing l mg l –1 GA3. Anthers with microspores at the late-uninucleate stage had the highest frequency of total and embryogenic calli formation, 40% and 15%, respectively. Each embryogenic callus usually produced 10–15 embryos. Aproximately 75 plants per 100 anthers cultured were recovered: 76% haploid, 22% diploid and 2% triploid. Effects of a juvenile hormone analog, pyriproxyfen, on various developmental stadia of the apterous form of the turnip aphid, Lipaphis erysimi (Kaltenbach), were bioassayed at three concentrations (50, 100, and 150 mg [AI]/l) in the laboratory. Supernumerary-molted nymphs with 1-3 extra molts were found when the first three nymphal instars were treated, and all these nymphs died prematurely. In contrast, all treated fourth instar nymphs molted normally to adults. The longevity of all pyriproxyfen-treated L. erysimi nymphs and adults and the fecundity of treated adults were reduced by 50%. Higher concentrations of pyriproxyfen (100 and 150 mg [AI]/l) caused sterility in adults from treated fourth instars; whereas the lowest concentration (50 mg [AI]/l) did not cause any adult sterility. A majority of adults from treated fourth instars and treated adults produced normal nymphs, and some adults produced dead nymphs with normal appendages or dead and deformed nymphs without any appendages. Normal nymphs produced by the adults from treated adults or treated at fourth instars developed more slowly from first instar to adult than the water-treated nymphs, and all adults in the new generation apparently reproduced normally. The potential role of pyriproxyfen in a vegetable aphid IPM program was discussed.  相似文献

Three species of hemipteran predators preyed differently upon 1st instarHeliothis punctiger Wallengren larvae.Cermatulus nasalis consumed more larvae thanOechalia schellenbergii which consumed more larvae thanTropiconabis nigrolineatus. All the species consumed significantly less 1st instar larvae on plants than what they consumed in Petri-dishes. Fifth instar predators showed significant differences in terms of prey consumption due to sex independent of searching conditions. Only 4th and 5th instars ofT. nigrolineatus attacked and captured 2nd instars ofH. punctiger larvae. The other 2 species however readily attacked and consumed 2nd instarH. punctiger larvae. Their prey consumption was similar in Petri-dishes and on plants. Only 5th instars ofT. nigrolineatus could subdue and capture 3rd instarH. punctiger larvae. Second instar pentatomids captured just one 3rd instar larva but older instars killed and ate more. Fourth instarH. punctiger larvae were immune to attacks by allT. nigrolineatus and younger pentatomids due to their defense ploys but 5th instar pentatomids could subdue and capture them. Microbial asymmetric oxidation of 2-butyl-1,3-propanediol was investigated for an efficient synthesis of S- and R-enantiomers of 2-hydroxymethylhexanoic acid (2-HMHA). From an intensive survey of the stocked bacterial strains, Acetobacter pasteurianus IAM 12073 and Pseudomonas putida IFO 3738 were found to show the highest S- and R-2-HMHA-producing activity, respectively. Under optimized conditions, A. pasteurianus (351 mg dry cell weight) and P. putida (642 mg dry cell weight) cells produced 12.0 g l−1 S-2-HMHA with 89% enantiomeric excess (e.e.) at 24 h of incubation and 5.1 g l−1 R-2-HMHA with 94% e.e. at 35 h of incubation from 2-butyl-1,3-propanediol.  相似文献

Field and laboratory studies were conducted to determine mollusk distributions in proximity to waste-water treatment plants (WTP's) in the upper Clinch River and to test the tolerance of two mollusk species to monochloramine and unionized ammonia, the major toxicants in domestic effluent. River reaches up to 3.7 km downstream of WTP's were devoid of freshwater mussels (Unionidae), and tolerance to effluents varied among snails, sphaeriid clams, and the asian clam Corbicula fluminea. Residential communities with septic systems had no measurable impact on mollusk assemblages downstream.Laboratory bioassays with glochidia of Villosa iris yielded the following results: 24 h EC50 and LC50 values of 0.042 mg l–1 and 0.084 mg l–1 monochloramine, respectively; and 24 h EC50 and LC50 of 0.237 mg l–1 and 0.284 mg l–1 unionized ammonia, respectively. Glochidia rank among the most sensitive invertebrates in their tolerance to these toxicants. The snail Pleurocera unciale unciale was moderately sensitive, with 96 h LC50 values of 0.252 mg l–1 monochloramine and 0.742 mg l–1 unionized ammonia. Monitoring of monochloramine and unionized ammonia concentrations 0.1 km below WTP outfalls indicated that monochloramine was the toxicant likely inhibiting mollusk recovery below these plants.The Unit is jointly supported by the U.S. Fish and Wildlife Service, Virginia Department of Game and Inland Fisheries, The Wildlife Management Institute and Virginia Polytechnic Institute and State University.  相似文献
