AIMS: Several enzymes from mycelial wastes of Penicillium chrysogenum fungal cultures from penicillin manufacture were studied as supplements for animal feeding. METHODS AND RESULTS: Proteins were precipitated with tannic acid, acetone, ethanol or polyethyleneglycol 6000. The levels of the enzyme activities found in the different precipitates were similar. The activities of invertase, beta-1,3-glucanase, lipase and tannase were higher and those of amylase, cellulase, pectinase, protease and xylanase were lower. Precipitation with tannic acid was selected as an easy and simple method. CONCLUSIONS, SIGNIFICANCE AND IMPACT OF THE STUDY: An enzymatic precipitate from these P. chrysogenum wastes, comparable to a commercial enzymatic additive used in animal feeding, was obtained.  相似文献

Eleven plant species were collected from the Blyde River Canyon area, Mpumalanga, South Africa. Methanolic crude extracts of the plant samples were concentrated to dryness, dissolved in sterile distilled water and bioassayed in the laboratory for their fungicidal potential against seven plant fungal pathogens, namely Botrytis cinerea, Fusarium oxysporum, Sclerotium rolfsii, Rhizoctonia solani, Verticillium dahliae, Botryosphaeria dothidea and Pythium ultimum, at equal concentrations. A modified agar dilution method was used to determine the inhibitory effect of the plant extracts on the mycelial radial growth of the plant pathogens. Antifungal activity was confirmed in all the plants, but not in all plant parts and also not against all the test organisms. The crude extract of Eucomis autumnalis performed best of all the plant extracts tested, as it showed significant antifungal activity against all seven of the plant pathogenic test organisms and compared favourably to the inhibition of the mycelial growth by a broad spectrum synthetic fungicide (carbendazim/ difenoconazole). The crude extract of Schrebera alata came out second best, in the sense that at least one plant part inhibited the mycelial radial growth of four of the seven test organisms by at least 50%.  相似文献

The oxygen transfer dynamics in a pilot plant external air-lift bioreactor (EALB) during the cultivation of mycelial biomass were characterized with respect to hydrodynamic parameters of gas holdup (), oxygen transfer coefficient (KLa) and superficial gas velocity (U g), and dissolved oxygen (DO). An increased flow rate of air supply was required to meet the increased oxygen demand with mycelial biomass growth. Consequently, an increase in air flow rate led to an increase in , KLa and the DO level. The enhancement of oxygen transfer rate in the cultivated broth system, however, was limited with highly increased viscosity of the mycelial broth. An increase in air flow rate from 1.25 to 2.00 v/v/m resulted in a low increment of oxygen transfer. The newly designed pilot plant EALB with two air spargers significantly improved processing reliability, aeration rate and KLa. The pilot plant EALB process, operated under a top pressure from 0 to 1.0 bars, also demonstrated a significant improvement of oxygenation efficiency by more than 20% in DO and KLa. The performance of the two sparger EALB process under top pressure demonstrated an efficient and economical aerobic system with fast mycelial growth and high biomass productivity in mycelial biomass production and wastewater treatment.  相似文献

The fermentation of Grifola frondosa was investigated in the shake flasks and a 5-L jar fermenter in batch and fed-batch modes. In the shake-flask experiments, the preferable mycelial growth and exopolysaccharide (EPS) production was observed at relatively low pH; maltose and glucose were preferred carbon sources for high mycelial production. The EPS was doubled after 13 d of cultivation when glucose was increased from 2% to 4%. Yeast extract (YE) (0.4%) in combination with corn steep powder (CSP) (0.6%) and YE (0.8%) in combination with CSP (1.2%) were preferred nitrogen sources for high mycelial production and EPS production, respectively. All plant oils tested significantly stimulate cell growth of G. frondosa but they failed to enhance EPS production. The EPS products usually consisted of two fractions of different molecular sizes varied by the plant oils used. The fed-batch fermentation by glucose feeding was performed when the glucose concentration in the medium was lower than 0.5% (5g/L), which greatly enhanced the accumulation of mycelial biomass and EPS; the mycelial biomass and EPS were 3.97g/L and 1.04g/L before glucose feeding, which reached 8.23g/L and 3.88g/L at 13 d of cultivation. In contrast, the mycelial biomass and EPS in the batch fermentation were 6.7g/L and 3.3g/L at 13 d of cultivation.  相似文献

The uranium-adsorbing abilities of seven plant wastes were investigated. High abilities to adsorb uranium from non-saline water containing 10 mg dm−3 of uranium were observed with a number of plant wastes tested. However, with seawater supplemented with 10 mg dm−3 of uranium, similar results were found only with chestnut residues. When the plant wastes were immobilized with formaldehyde, their ability to adsorb uranium was increased. Uranium and copper ions were more readily adsorbed by all plant wastes tested than other metal ions from a solution containing a mixture of seven different heavy metals. The selective adsorption of heavy metal ions differs with different species of plant wastes. The immobilization of peanut inner skin, orange peel and grapefruit peel increased the selectivity for uranium.  相似文献

AIMS: The aim of this work was to study the effect of high temperatures generated during composting process, on the phytopathogen fungus Fusarium oxysporum f.sp. melonis. This investigation was achieved by both in vivo (semipilot-scale composting of horticultural wastes) and in vitro (lab-scale thermal treatments) assays. METHODS AND RESULTS: Vegetable residues infected with F. oxysporum f.sp. melonis were included in compost piles. Studies were conducted in several compost windrows subjected to different treatments. Results showed an effective suppression of persistence and infective capacity, as this process caused complete fungal elimination after 2-3 days of composting. In order to confirm the effect of high temperature during this process, in vitro experiments were carried out. Temperature values of 45, 55 and 65 degrees C were tested. All three treatments caused the elimination of fungal persistence. Treatment at 65 degrees C was especially effective, whereas 45 degrees C eliminated fungal persistence only after 10 days. CONCLUSIONS: The composting process is an excellent alternative for the management of plant wastes after harvesting, as this procedure is able to suppress infective capacity of several harmful phytopathogens such as F. oxysporum f.sp. melonis. SIGNIFICANCE AND IMPACT OF THE STUDY: Fusarium oxysporum f.sp. melonis is a plant pathogen fungus specially important in the province of Almería (south-east Spain), where intensive greenhouse horticulture is very extended. High temperatures reached during composting of horticultural plant wastes ensure the elimination of phytopathogen microorganisms such as F. oxysporum f.sp. melonis from vegetable material, providing an adequate hygienic quality in composts obtained.  相似文献

Six ligno-cellulolytic fungi were tested regarding to examine their capability to grow on agricultural wastes and produce inocula for composting. Two residues were used: pepper plant wastes and almond shell residues. Results showed the latter as the most adequate substrate for growth of fungi tested. On the contrary, Trichoderma koningii, as well as HLC1 and HLC3, both fungi isolated from almond shell wastes, were able to persist in pepper plant wastes. Modifications of aeration and pH significantly influenced growth of Coriolus versicolor, HLC1 and Phanerochaete. flavido alba and P. flavido alba and Phlebia radiata, respectively, while P. flavido alba was the only microorganism whose growth was not significantly altered by temperature. In competitive assays, where fungi were growth together with other species, growth of both microorganisms isolated from almond shell residues, HLC1 and HLC3, were stimulated while T. koningii showed the better results in sterile conditions.  相似文献

Pleomorphic deterioration is a process where a fungal isolate loses the ability to produce conidia during repeated subculturing. We have previously isolated strains of the entomopathogenic fungus Metarhizium anisopliae that have irreversibly lost the ability to produce conidia and only produce mycelia when grown on agar. Gel electrophoresis was used to examine differences in intracellular protein patterns (urea-soluble proteins and urea-insoluble proteins (i.e., hydrophobins)) in conidiating and mycelial cultures of M. anisopliae. Two major proteins present in a conidiating culture and one from a mycelial culture were N-terminally sequenced but showed no homologies to known proteins. The presence of hydrophobins in conidiating and mycelial cultures was also examined, and it was shown that these proteins were abundant in conidiating cultures but not in mycelial cultures. We also used primers designed from regulatory genes involved in conidiation in Aspergillus nidulans. The amplified fragments were not homologous to A. nidulans genes.  相似文献

Damaging threshold levels of root-knot nematode Meloidogyne incognita and root-rot fungus Fusarium solani on plant growth parameters, viz., plant length, fresh and dry weight of chilli were determined by conducting their pathogenicity trials in pot experiments. The results revealed a significant reduction in the plant growth parameters at and above the inoculum level of about 1000?J2 per plant of M. incognita and the highest reduction was recorded at 8000?J2 per plant. Significant reduction in plant growth parameters was recorded at 1.00?g mycelial mat of F. solani per plant, while the highest reduction was observed at 8.00?g mycelial mat per plant. The damaging threshold level was 1000?J2 per plant of M. incognita and 1.00?g mycelial mat of F. solani.  相似文献

Thirty‐nine plant species, representing 20 families from the subclasses Rosidae, Asteridae, Commelinidae and Liliidae, were collected from the Blyde River Canyon Nature Reserve, Mpumalanga, South Africa. Crude extracts were prepared and bioassayed, at equal concentrations, for their antifungal potential by determining the inhibitory effects on the mycelial growth of seven economically important plant pathogenic fungi. Statistically significant differences between plants and plant parts were observed as well as the resistance of different fungi to treatment with different plant extracts. The most significant mycelial growth inhibition was obtained with extracts from two species of the subclass Liliidae, namely Aristea ecklonii and Agapanthus inapertus. The crude extract of A. ecklonii performed best of all extracts as it totally inhibited the mycelial growth of all seven of the plant pathogenic test organisms and outperformed the inhibition by a broad spectrum synthetic fungicide (carbendazim/difenoconazole). Crude extracts of A. inapertus showed complete inhibition of four and strong inhibition of the remaining three plant pathogenic fungi. Although not as efficient as the previous two species, the extract of Anisopappus junodii, from the subclass Asteridae, also showed promising antifungal activity by completely inhibiting mycelial radial growth of two and strongly inhibiting that of the remaining five fungi.  相似文献

Summary Mycelial wastes of Rhizopus arrhizus, used in fermentation industries to produce lipases, were studied for their ability to absorb various heavy metal ions (Ni, Zn, Cd and Pb). Chelation of all these ions occurs by a chemical, equilibrated and saturatable mechanism, following the Langmuir adsorption model. Data transformation allowed us to calculate maximum uptake and dissociation constants of the sorption reaction. We also investigated the influence of pH on metal accumulation. Sorption capacity variations between different biosorbent types (Rhizopus, Mucor, Penicillium, and Aspergillus), could be related to their acidity. pH neutralisation during the sorption reaction considerably enhanced zinc chelation (up to 56 mg/g). Previous NaOH treatment of mycelial wastes also increased their capacity for metal sorption. We report R. arrhizus metal uptake curves versus pH, using a pH-stat system. Optimal adsorption was achieved at neutral pH for nickel and zinc, pH 5.0 for lead, and inhibition of chelation was observed when the pH decreased. These results illustrate the importance of pH during the adsorption process, indicating a competitive mechanism for chelation between heavy metal ions and protons at cell wall adsorption sites. Hemicellulose components from industrial viscose fibre production are characterized by a lower cellulose content than commercial xylan and the presence of a carboxylic acid fraction originating from the alkaline degradation of carbohydrates during the process. This substrate, after neutralization, can be used by Trichoderma reesei RUT C-30 for the production of cellulase-poor xylanases, useful for the pulp and paper industry. The yields of xylanase ranged up to almost 400 units/ml, with a ratio of carboxymethylcellulase/xylanase of less than 0.015. This crude xylanase enzyme mixture was shown to be superior to that obtained on beech-wood xylan when used for bleaching and, particularly, upgrading of hard-wood chemical pulp by selective removal of the xylan components. Biochemical studies indicate that the low cellulase production by T. reesei grown on these waste hemicelluloses is the result of a combination of at least three factors: (a) the comparatively low content of cellulose in these hemicellulosic wastes, (b) the inhibitory action of the carboxylic acid fraction present in the hemicellulosic wastes on growth and sporulation of T. reesei, and (c) the use of a mycelial inoculum that is unable to initiate the attack on the cellulose components within the carbon source. Beauvericin (BEA) is a proven and potent antibiotic compound useful for bio-control and a potential antifungal and anticancer agent for human. This study was to evaluate and optimize the nutrient medium for BEA production in mycelial liquid culture of a high BEA-producing fungus Fusarium redolens Dzf2 isolated from a medicinal plant. Among various organic and inorganic carbon and nitrogen sources, glucose and peptone were found the most favorable for the F. redolens Dzf2 mycelial growth and BEA production. Through a Plackett-Burman screening test on a basal medium, glucose, peptone, and medium pH were identified as the significant factors for mycelial growth and BEA production. These factors were optimized through central composite design of experiments and response surface methodology, as 49.0 g/L glucose, 13.0 g/L peptone and pH 6.6, yielding 198 mg/L BEA (versus 156 mg/L in the basal medium). The BEA yield was further increased to 234 mg/L by feeding 10 g/L glucose to the culture during exponential phase. The results show that F. redolens Dzf2 mycelial fermentation is a feasible and promising process for production of BEA.  相似文献

Lentinula edodes is considered an alternative recycling agent for agricultural wastes, and there have been several studies to understand the relationship between its growth and ligninolytic activity. We tested the effect of wood from viticulture pruning, extracted with solvents of differing polarity, on the biomass production and activity pattern of ligninolytic enzymes. The analysis was done by measuring the mycelial dry mass and enzyme activity of liquid growth medium during the culture of L. edodes, adding either single extracts or a combination of extracts. Polar extracts enhanced mycelial production, and the activity patterns of lignin peroxidase, manganese peroxidase, aryl alcohol oxidase, and laccase were comparable to their activities predicted by ligninolysis models proposed for other fungi. We conclude that the polar extracts could be useful for enhancing fungal biomass production and for modifying lignin degradation because the regulation of ligninolytic enzyme activity is differentially influenced by the polarity of the extract.  相似文献

Ergosterol, a membrane sterol found in fungi but not in plants, was used to estimate live mycelial biomass in ectomycorrhizae. Loblolly pine (Pinus taeda L.) seeds were sown in April 1993 and grown with standard nursery culture practices. Correlations between total seedling ergosterol and visual assessment of mycorrhizal colonization were high during July and August but low as ectomycorrhizal development continued into the growing season. Percentages of mycelial dry weight over lateral roots decreased from 9% in July to 2.5% in November because seedling lateral root dry weight accumulated faster than mycelial dry weight. Total ergosterol per seedling increased from July through February. As lateral root dry weight ceased to increase during winter months, ectomycorrhizal mycelia became the major carbohydrate sink of pine seedlings. No distinctive seasonal pattern of soil ergosterol content was observed. The impact of ectomycorrhizal fungi on plant carbohydrate source-sink dynamics can be quantitatively estimated with ergosterol analysis but not with conventional visual determination.  相似文献

The application of low-cost adsorbents obtained from plant wastes as a replacement for costly conventional methods of removing heavy metal ions from wastewater has been reviewed. It is well known that cellulosic waste materials can be obtained and employed as cheap adsorbents and their performance to remove heavy metal ions can be affected upon chemical treatment. In general, chemically modified plant wastes exhibit higher adsorption capacities than unmodified forms. Numerous chemicals have been used for modifications which include mineral and organic acids, bases, oxidizing agent, organic compounds, etc. In this review, an extensive list of plant wastes as adsorbents including rice husks, spent grain, sawdust, sugarcane bagasse, fruit wastes, weeds and others has been compiled. Some of the treated adsorbents show good adsorption capacities for Cd, Cu, Pb, Zn and Ni.  相似文献

Malaysia is blessed with abundant natural resources and bears a favorable climate for commercial cultivation of crops such as oil palm. In Malaysia the total plantation area of oil palm was 4,487,957 ha in 2008. It has been reported that in 2005 there was a total of 423 palm oil mills having production capacity of approximately 89 million tonnes of fresh fruit bunch (FFB) per year. Waste from the oil palm mill process include palm oil mill effluent (POME), generated mainly from oil extraction, washing and cleaning up processes. POME contains cellulosic material, fat, oil, and grease. Discharging untreated effluent into water streams may cause considerable environmental problems. The solid wastes generated are mainly decanter cake, empty fruit bunches, seed shells and fibre from the mesocarp. POME as well as the solid wastes may rapidly deteriorate the surrounding environment if not dealt with properly. Hence there is an urgent need for a sustainable waste management system to tackle these wastes. As these wastes are organic in origin, they are rich in plant nutrients. Composting of waste generated from palm oil mills can be good practice as it will be helpful in recycling useful plant nutrients. This review deals with various aspects of waste management practices in palm oil mills and the possibility of composting the wastes.  相似文献
