In experiments on 2-3-, 7-8-, 10- and 16-day old rat pups, basic age differences have been observed in the effect of a precursor of catecholamine mediator, i.e. L-DOPA on the activity on the spinal and brain mechanisms of autogenic periodic motor excitation. Strong stimulation of the spinal motor rhythm was observed during the first week. At this period, supraspinal rhythm becomes even less evident. On further development, the effect is an opposite one. In 10- and 16-day rat pups, at the background of total inhibition of the spontaneous activity, administration of L-DOPA significantly increases the activity of supraspinal generator of the motor rhythm. The data obtained reveal ontogenetic changes in regulatory mechanisms of autogenic motor activity.  相似文献

Studies on muscle functional coupling mechanisms have detected excitatory and inhibitory reflex systems differing in their organization at the spinal level. The classic notions on inhibitory interactions between antagonists at the motoneuronal and premotoneuronal levels are supplemented with data on excitatory interactions. Mutual excitation of synergist muscles is also not the only possible type of interaction: inhibitory relationships are found in the system of synergists under certain physiological conditions. This organization of the motor centers of antagonists and synergists enhances the possibilities for fine coordination of their activity, inhibition and disinhibition mechanisms playing an important role in this system. Data on the inhibitory systems of the spinal cord, in particular, the inhibitory interactions between functionally coupled muscles, are reviewed.  相似文献

The method of transcutaneous electrical stimulation of the spinal cord (ESSC) has recently begun to be actively used for both experimental studies of human motor functions and the rehabilitation of motor function in patients with spinal cord pathology. The spinal cord is the most important center of the regulation of vital functions, and ESSC affects as spinal locomotor networks as the visceral system too, which should be taken into account for the development of an improved method of rehabilitation and its use in experiments on healthy volunteers. We present a review of studies on the possible mechanisms of ESSC effects on the peripheral and cerebral circulation, cardiovascular, respiratory, excretory, and digestive systems of mammals.  相似文献

The coordination and timing of muscle activities during rhythmic movements, like walking and swimming, are generated by intrinsic spinal motor circuits. Such locomotor networks are operational early in development and are found in all vertebrates. This review outlines and compares recent advances that have revealed the developmental and functional organization of these fundamental spinal motor networks in limbed and non-limbed animals. The comparison will highlight common principles and divergence in the organization of the spinal locomotor network structure in these different species as well as point to unresolved issues regarding the assembly and functioning of these networks.  相似文献

In experiments on 5-30-day rat puppies, under slight urethane narcosis, studies have been made on the late prolonged discharges in extensor muscles evoked by stimulation of contralateral tibial nerve before and after injection of DOPA. Within the first 16 days, the drug completely abolished or significantly reduced late discharges, increasing the spontaneous motor activity. Beginning from the 16th day, periods of potentiation of late discharges were observed. The data obtained are discussed in relation to the development of mechanisms of control of the activity of spinal generators which are responsible for late discharges and spontaneous motor activity.  相似文献

We have used calcium imaging to visualize the spatiotemporal organization of activity generated by in vitro spinal cord preparations of the developing chick embryo and the neonatal mouse. During each episode of spontaneous activity, we found that chick spinal neurons were activated rhythmically and synchronously throughout the transverse extent of the spinal cord. At the onset of a spontaneous episode, optical activity originated in the ventrolateral part of the cord. Back-labeling of spinal interneurons with calcium dyes suggested that this ventrolateral initiation was mediated by activation of a class of interneurons, located dorsomedial to the motor nucleus, that receive direct monosynaptic input from motoneurons. Studies of locomotor-like activity in the anterior lumbar segments of the neonatal mouse cord revealed the existence of a rostrocaudal wave in the oscillatory component of each cycle of rhythmic motoneuron activity. This finding raises the possibility that the activation of mammalian motoneurons during locomotion may share some of the same rostrocaudally organized mechanisms that evolved to control swimming in fishes.  相似文献

Seventy years ago George Romanes began to document the anatomical organization of the spinal motor system, uncovering a multilayered topographic plan that links the clustering and settling position of motor neurons to the spatial arrangement and biomechanical features of limb muscles. To this day, these findings have provided a structural foundation for analysis of the neural control of movement and serve as?a guide for studies to explore mechanisms that direct the wiring of spinal motor circuits. In this brief essay we outline the?core of Romanes's findings and place them in the context of recent studies that begin to provide insight?into molecular programs that assign motor pool position and to resolve how motor neuron position shapes circuit assembly. Romanes's findings reveal how and why neuronal positioning contributes to sensory-motor connectivity and may have relevance to circuit organization in other regions of the central nervous system.  相似文献

Central pattern generators are neuronal circuits that when activated can produce rhythmic motor patterns such as walking, breathing, flying, and swimming in the absence of sensory or descending inputs that carry specific timing information. General principles of the organization of these circuits and their control by higher brain centers have come from the study of smaller circuits found in invertebrates. Recent work on vertebrates highlights the importance of neuro-modulatory control pathways in enabling spinal cord and brain stem circuits to generate meaningful motor patterns. Because rhythmic motor patterns are easily quantified and studied, central pattern generators will provide important testing grounds for understanding the effects of numerous genetic mutations on behavior. Moreover, further understanding of the modulation of spinal cord circuitry used in rhythmic behaviors should facilitate the development of new treatments to enhance recovery after spinal cord damage.  相似文献

Hoxa10 and Hoxd10 coordinately regulate lumbar motor neuron patterning   总被引:1,自引:0,他引:1  
The paralogous Hox genes Hoxa10 and Hoxd10 are expressed in overlapping domains in the developing lumbar spinal cord and surrounding mesoderm. Independent inactivation of these two genes alters the trajectory of spinal nerves and decreases the complement of motor neurons present in the lumbar spinal cord, whereas dual inactivation of these two genes has been shown to alter peripheral nerve growth and development in the mouse hindlimb. We have examined the organization and distribution of lumbar motor neurons in the spinal cords of Hoxa10/Hoxd10 double mutant animals. Double mutant animals have decreased numbers of lumbar motor neurons in both the medial and lateral motor columns. The anteroposterior position of the lumbar motor column is shifted caudally in double mutant animals, and the distribution of motor neurons is altered across individual spinal segments. Distinctions between classes of motor neurons based on positional specificity appear disrupted in double mutants. Double mutants also demonstrate abnormal spinal cord vasculature and altered kidney placement and size. Our observations suggest that Hoxa10 and Hoxd10 activity is required to specify the position of the lumbar motor column and to provide segmental specification and identity for the lumbar motor neurons.  相似文献

The organization of intracellular compartments and the transfer of components between them are central to the correct functioning of mammalian cells. Proteins and lipids are transferred between compartments by the formation, movement and subsequent specific fusion of transport intermediates. These vesicles and membrane clusters must be coupled to the cytoskeleton and to motor proteins that drive motility. Anterograde ER (endoplasmic reticulum)-to-Golgi transport, and the converse step of retrograde traffic from the Golgi to the ER, are now known to involve coupling of membranes to the microtubule cytoskeleton. Here we shall discuss our current understanding of the mechanisms that link membrane traffic in the early secretory pathway to the microtubule cytoskeleton in mammalian cells. Recent data have also provided molecular detail of functional co-ordination of motor proteins to specify directionality, as well as mechanisms for regulating motor activity by protein phosphorylation.  相似文献

Participation of the reticular formation and descending reticulospinal system in the motor control functions of the spinal cord is examined. The data indicate that the reticular formation may participate in the regulation of specific motor actions. This is shown by the results of experiments to analyze the properties of membranes of reticulospinal neurons and the principles of organization of cortico-reticular monosynaptic relays, and to the investigation of characteristics of responses of "ensembles" of reticular neurons in cats, and also by data obtained in a study of correlation of unit activity in the reticular formation with movements elaborated in rats. A review of the role of the microtubule motor dynein and its cofactor dynactin in the formation of a radial system of microtubules in the interphase cells and of mitotic spindle. Deciphering of the structure, functions, and regulation of activity of dynein and dynactin promoted the understanding of mechanisms of cell and tissue morphogenesis, since it turned out that these cells help the cell in finding its center and organize microtubule-determined anisotropy of intracellular space. The structure of dynein and dynactin molecules has been considered, as well as possible pathways of regulation of the dynein activity and the role of dynein in transport of cell components along the microtubules. Attention has also been paid to the functions of dynein and dynactin not related directly to transport: their involvement in the formation of an interphase radial system of microtubules. This system can be formed by self-organization of microtubules and dynein-containing organelles or via organization of microtubules by the centrosome, whose functioning requires dynein. In addition, dynein and dynactin are responsible for cell polarization during its movement, as well as for the position of nucleus, centrosomes, and mitotic spindle in the cell.  相似文献

To examine the involvement of supraspinal inputs in the maturation of motor activity patterns in the developing fetal lamb, we recorded spontaneous electromyographic activity from spinally innervated muscles at approximately 45, 65, and 95 days gestation (G45, G65, and G95; term = 147 days). At G45, fetal activity occurred in synchronized activity-inactivity cycles of approximately 2 min duration, with the activity phase lasting 22.2 ± 4.8 s and the inactivity phase lasting 95.4 ± 13.3 s (mean ± standard error of the mean, n = 5). At G65 and G95, the organization of activity was clearly different from that at G45 in that it was no longer cyclic, nor was the discharge of different muscles synchronized. By contrast, after spinal cord transection at G62, synchronised cyclic activity occurred in muscles innervated by segmental levels below the transection, both at G65 and G95. At G65 the duration of the activity phase of the cycle was 53.5 ± 6.0 s, while the inactivity phase lasted 171.6 ± 22.1 s; these durations did not alter between G65 and G95. Since spinal cord transection leads to the motor behavior of the G65 fetus reverting to the cyclic pattern characteristic of the G45 fetus, we conclude that supraspinal inputs begin to modulate the output of the spinal pattern generators at some stage between G45 and G65. The observation that spinally transected fetuses generate identical behavior at G65 and G95, both in terms of its cyclic character and the duration of cycles, suggests that spinal circuits undergo little autonomous development overthis period; that is, the altered behavior observed in the developing intact fetus reflects the influence of supra-spinal inputs on the motor circuits of the spinal cord. © 1997 John Wiley & Sons, Inc. Motor neurons that project axons to distinct limb-muscle targets are topographically organized such that central nervous system position reflects the location of the muscle in the limb. The central positioning of limb-projecting motor neurons arises during development through motor neuron migration followed by a period of coalescence into discrete groupings of motor neurons which project axons to an individual muscle. These so-called motor pools are a common feature of motor organization in higher vertebrates. Recent work has highlighted the critical role for armadillo family member catenin-dependent functions of the cadherin family of cell adhesion molecules in directing the organization of motor neurons. Cadherin function appears to be important for both the motor neuron migration and coalescence phases of the emergence of motor neuron topography.
