Herbivory, mechanical injury or pathogen infestation to vegetative tissues can induce volatile organic compounds (VOCs) production, which can provide defensive functions to injured and uninjured plants. In our studies with ‘McNeal’ wheat, ‘Otana’ oat, and ‘Harrington’ barley, plants that were mechanically injured, attacked by either of two Oulema spp. (melanopus or cyanella) beetles, or infected by one of the three Fusarium spp. (graminearum, avenaceum, or culmorum), had significant VOC induction compared to undamaged plants. Mechanical injury to the main stem or one leaf caused the induction of one green leaf volatile (GLV) - (Z)-3-hexenol, and three terpenes (β-linalool, β-caryophyllene, and α-pinene) with all three grasses; wheat and barley also showed β-linalool oxide induction. The blend of induced VOCs after Fusarium spp. infestation or Oulema spp. herbivory was dominated by GLVs ((Z)-3-hexenal, (E)-2-hexenal, (E)-2-hexenol, (Z)-3-hexenyl acetate, and 1-hexenyl acetate) and β-linalool and β-caryophyllene; beetle herbivory also induced (E)-β-farnesene. Different ratios of individual VOCs were induced between the two Oulema spp. for each cereal grass and different ratios across the three cereals for each beetle species. Also, different ratios of individual VOCs were induced between the three Fusarium spp. for each cereal grass and different ratios across the three cereals for each fungal pathogen species. Our results are preliminary since we could not simultaneously measure VOC induction from controls with each of the ten different injury treatments for each of the three cereals. However, the comparison of mechanical injury, insect herbivory, and fungal infection has not been previously examined with VOC responses from three different plant species within the same family. Also, our work suggests large qualitative and quantitative overlap of VOC induction from plants of all three cereals having beetle herbivory injury when compared to infection injury from necrotrophic fungal pathogens.  相似文献

The cereal leaf beetle (CLB), Oulema melanopus (L.) (Coleoptera: Chrysomelidae), is an invasive pest in North America recently reported in the Canadian Prairies. We performed a series of laboratory assays to identify potential predators and a field study to quantify predation of CLB eggs. In no-choice Petri dish assays, ground beetles (Carabidae), rove beetles (Staphylinidae), and several common lady beetle species (Coccinellidae) were the most consistent predators of eggs and larvae. Nabis spp. (Hemiptera: Nabidae) and wolf spiders (Araneae: Lycosidae) consumed many larvae, but did not consume eggs. Hippodamia spp., Coccinella septempunctata (L.) (Coleoptera: Coccinellidae), and Pterostichus melanarius (Illiger) (Coleoptera: Carabidae) also fed on CLB eggs on potted plants when an alternative food source was available, Sitobion avenae (Fabricius) (Hemiptera: Aphididae). In our field study, we found an average of 24.5% of sentinel eggs disappeared over a 24?h period, likely due to predation. Our results suggest that generalist predators can play an important role in the biological control of CLB, and warrant further study.  相似文献

The cereal leaf beetle, Oulema melanopus (Coleoptera: Chrysomelidae), is a new invasive pest of cereals in western Canada. Host plant resistance is a potential option in the integrated management of this insect. Wheat (Triticum aestivum L.) genotypes are known to possess antixenosis against O. melanopus; however, mechanisms involving antibiosis are relatively less explored. We present results of an investigation exploring antibiotic resistance in wheat germplasm of Central Asian origin with putative resistance to O. melanopus. Our laboratory assessment of development and survivorship of O. melanopus on test genotypes indicated antibiotic properties in four of the six genotypes evaluated. Antibiosis effects were reflected through lower survivorship, extended developmental periods and low adult fitness. However, two genotypes were excellent hosts, and beetles had higher survivorship and fitness on them. The performance of O. melanopus on these two genotypes was comparable to that on the susceptible genotype CDC GO included as a check. The genotypes with antibiotic resistance identified by this research can now serve as sources of genetic resistance for breeding cultivars adapted to western Canadian conditions.  相似文献

The sensitivity of the cereal leaf beetle, Oulema melanopus (Coleoptera: Chrysomelidae), to maize-expressed Bacillus thuringiensis (Bt) proteins was investigated in the present study. Neonate larvae of O. melanopus were caged on leaves of Cry3Bb1-expressing (MON88017) or Cry1Ab-expressing (MON810) Bt maize, the corresponding near-isolines, or two non-related, conventional maize varieties. Larval survival was reduced on Cry3Bb1-expressing, but not on Cry1Ab-expressing maize compared with conventional varieties. Differences among conventional varieties were also present. The amount of eaten leaf material, developmental time to prepupal stage, and prepupal weight did not differ between Bt maize varieties and their corresponding near-isolines. In an additional feeding study with newly emerged adults, survival and beetle weight did not differ when leaves of Cry3Bb1-expressing maize or the near-isoline were offered as food over 3 weeks. ELISA measurements revealed that larvae feeding on Bt maize contained rather high Cry3Bb1 or Cry1Ab levels, which were in the same order of magnitude as the leaves. In contrast, concentrations in feces were one order, and concentrations in prepupae and adults two orders of magnitude lower.  相似文献

In sequentially planted oat stands, the cereal leaf beetle (CLB), Oulema melanopus (L.) (Coleoptera: Chrysomelidae: Lemini), is found in greater numbers, and lays more eggs, on later planted (younger) oats (Avena sativa L.) (Poaceae). Plant characteristics that could explain this ovipositional preference were examined in a series of experiments. Cage and open field whole plant preference tests confirmed the attraction of ovipositing females to younger oats. A cage effect illustrated the role of plant architecture (plant height) in CLB host selection. Two multiple‐choice and one no‐choice excised leaf experiments determined that characteristics of individual leaves associated with leaf insertion level (leaf number from base to apex) and age influence ovipositional site selection. Leaves of higher insertion level have higher nitrogen content, but fewer eggs are laid on those leaves. Two experiments examining the interaction between total leaf nitrogen and leaf insertion level showed that only leaf insertion level affected oviposition choice. Published literature suggests variation in secondary plant compounds cannot explain O. melanopus ovipositional preference among leaves. Grass leaves of higher insertion level have more extensively developed cells associated with tissue toughness and hardness. The data and supporting literature suggest tissue toughness and hardness are deterring oviposition on oat leaves of higher insertion level. However, newly eclosed larvae are able to feed on leaves usually avoided as oviposition sites. The explanation for this result may be a lack of correlation between host suitability and ovipositional preference.  相似文献

Maize plants respond to feeding by arthropod herbivores by producing a number of secondary plant compounds, including volatile organic compounds (VOCs). These herbivore-induced VOCs are not only known to attract natural enemies of the herbivores, but they may also prime inducible defences in neighbouring plants, resulting in stronger and faster defence responses in these VOC-exposed plants. Among the compounds that cause this priming effect, green leaf volatiles (GLVs) have received particular attention, as they are ubiquitous and rapidly emitted upon damage. In this study, we investigated their effects under realistic conditions by applying specially devised dispensers to release four synthetic GLVs at physiologically relevant concentrations in a series of experiments in maize fields. We compared the VOC emission of GLV-exposed maize plants to non-exposed plants and monitored the attraction of herbivores and predators, as well as parasitism of the caterpillar Spodoptera frugiperda, the most common herbivore in the experimental maize fields. We found that maize plants that were exposed to GLVs emitted increased quantities of sesquiterpenes compared to non-exposed plants. In several replicates, herbivorous insects, such as adult Diabrotica beetles and S. frugiperda larvae, were observed more frequently in GLV-treated plots and caused more damage to GLV-exposed plants than to non-exposed plants. Parasitism of S. frugiperda was only weakly affected by GLVs and overall parasitism rates of S. frugiperda were similar in GLV-exposed and non-exposed plots. The effects on insect presence depended on the distance from the GLV-dispensers at which the plants were located. The results are discussed in the context of strategies to improve biological control by enhancing plant-mediated attraction of natural enemies.  相似文献

The association of enzyme activities in developing kernels with specific storage product accumulation at maturity was analyzed in different parts of Zea mays inbred OH43 kernels. Maize kernels were harvested at 20 and 55 days post-pollination and dissected into basal region, pericarp, embryo, lower endosperm, middle endosperm and upper endosperm. Mature (55 days pos(-pollination) kernel parts were analyzed for starch, total protein, zein and oil content. Immature (20 days post-pollination) kernel parts were assayed for activities of 15 enzymes of sugar and amino acid metabolism. Statistical analyses of the data suggested that glucokinase (EC 2.7.1.2) fructokinase (EC 2.7.1.4) and phosphofructokinase (EC 2.7.1.1 11) activities were primarily associated with oil accumulation, whereas ADP'-glueose pyrophosphorylasc (EC 2.7.7.27) and sucrose synthase (EC 2.4.1.13) activities were associated with starch accumulation. The results suggest that oil biosynthesis utilizes inveitase-mediated sucrose degradation in a pathway not requiring pyrophosphatc. whereas starch biosynthesis utilizes a sucrose synthase-mediated pathway of sucrose degradation in a pathway requiring pyrophosphatc. Additional groups of enzyme activities were associated with each oilier but not with any specific storage product and appeared to be associated with general metabolic activity.  相似文献

Aims Ecological systems, especially soils, have been recently recognized as an important source of atmospheric nitric oxide (NO). However, the study on the contribution of plants to atmospheric NO budget is significantly lagged. The specific objectives of this study are to reveal the phylogenetic variation in NO emission potential existing in various plant species and find out the possible leaf traits affecting NO emission potential.Methods We measured NO emission potential, leaf N and C content, C:N ratio, specific leaf area, net photosynthetic rate (P n) and estimated photosynthetic N use efficiency (PNUE) of 88 plant species. Further investigation of the relationships between NO emission potential and leaf traits were performed by simple linear regression analysis and pair-wise correlation coefficients analysis.Important findings Major results are as follows: (1) NO emission from plant species exhibited large variations, ranging from 0 to 41.7 nmol m ?2 h-1, and the species frequency distributions of NO emission potential could be fitted to a log-normal curve. (2) Among 88 species, NO emission potential was the highest in Podocarpus macrophyllus, but lowest in Zanthoxylum nitidum and Vernicia montana. (3) NO emission potential has strong correlation to leaf N content, P n and PNUE. The variations in NO emission potential among diverse plant species may be closely related to leaf N level and net photosynthetic ability.  相似文献

CYP3RNA, a double-stranded (ds)RNA designed to concomitantly target the two sterol 14α-demethylase genes FgCYP51A and FgCYP51B and the fungal virulence factor FgCYP51C, inhibits the growth of the ascomycete fungus Fusarium graminearum (Fg) in vitro and in planta. Here we compare two different methods (setups) of dsRNA delivery, viz. transgene expression (host-induced gene silencing, HIGS) and spray application (spray-induced gene silencing, SIGS), to assess the activity of CYP3RNA and novel dsRNA species designed to target one or two FgCYP51 genes. Using Arabidopsis and barley, we found that dsRNA designed to target two FgCYP51 genes inhibited fungal growth more efficiently than dsRNA targeting a single gene, although both dsRNA species reduced fungal infection. Either dsRNA delivery method reduced fungal growth stronger than anticipated from previous mutational knock-out (KO) strategies, where single gene KO had no significant effect on fungal viability. Consistent with the strong inhibitory effects of the dsRNAs on fungal development in both setups, we detected to a large extent dsRNA-mediated co-silencing of respective non-target FgCYP51 genes. Together, our data further support the valuation that dsRNA applications have an interesting potential for pesticide target validation and gene function studies, apart from their potential for crop protection.  相似文献

Summary Tissue cultures of maize carrying cms-T cytoplasm have been found to regenerate fertile, T-toxin resistant plants, with and without a selective treatment with T-toxin. Progeny of these plants were tested for mitochondrial sensitivity to T-toxin and the translation products synthesised by isolated mitochondria were analysed. The results confirm previous indications of a close correlation between susceptibility to T-toxin and the synthesis of a variant 13,000 Mr mitochondrial polypeptide. Interestingly, there appeared to be a critical level at about 33% maximum synthesis of the 13,000 Mr polypeptide above which male sterility and sensitivity to T-toxin are jointly expressed. The possibility that there is a causal link between synthesis of this additional mitochondrial polypeptide, pollen abortion and sensitivity to T-toxin is discussed.  相似文献

The degree of adaptation of herbivorous insects to their local flora is an important component of the evolutionary processes that lead to host plant specialization in insects. In this study we investigated geographic variations in the oviposition preference of the leaf beetle Oreina elongata Suffrian (Coleoptera: Chrysomelidae: Chrysolini) in relation to differences in host plant specialization, in the field. We focused on the mechanisms of host choice and asked whether potential differences among populations are due to variations in host plant ranking and/or host plant specificity. We performed a combination of simultaneous choice and sequential no‐choice experiments with two of the major host plants of the beetle [Cirsium spinosissimum (L.) and Adenostyles alliariae (Gouan) (Asteraceae)]. The results suggested that spatial variation in host plant specialization has resulted in differences between populations in some aspects of the oviposition choice of O. elongata, while other aspects seem unaffected. We found no variation in host plant ranking among populations, as estimated in simultaneous choice tests. In contrast, the sequential no‐choice test indicated that host plant specificity was lower in a population that never encountered the highest ranked plant in the field. This finding agreed with our expectations, and we discuss our results in relation to the commonly used hierarchical threshold model. The results suggested that the mechanism for the differences in specificity is the variation among populations in the general motivation to oviposit, rather than quantitative differences in relative preference for the two hosts. We stress that it is essential to establish which of the two mechanisms is most important, as it will affect the probability of evolutionary change in host plant ranking.  相似文献

Molecules transferred from males to females via seminal fluids are important to the study of insect reproduction because they affect female physiology, reproductive behavior, and longevity. These molecules (seminal fluid molecules or SFMs) interest applied entomologists because of their potential use in insect control. SFMs are also interesting because of their relatively rapid evolution and important role in post-mating sexual selection. We studied SFMs in Diaprepes abbreviatus, a major pest of numerous plant species of economic importance. Using radiolabeled-methionine (35S), we found that D. abbreviatus males synthesized proteins de novo in their reproductive tissues after mating. Males that were fed radiolabeled methionine transferred radioactivity to females beginning within the first 10 min of mating. Male-derived substances are absorbed from the female's reproductive tract into the hemolymph and circulated throughout the body, but are found primarily in the eggs and ovaries. As a result, SFMs may be a useful means of both horizontal (to mates) and vertical transfer (to offspring) of control agents between conspecifics.  相似文献

Complicated genital structures are widely known in many animals. It is assumed that an increasing morphological complexity of genital structures would also increase the difficulty of inserting and withdrawing the structures. We examined the insertion and withdrawal mechanisms of extremely elongated genitalia in the Japanese leaf beetle, Lema (Lema) coronata. Insertion and withdrawal processes are completed quickly. Investigation of genital morphology showed that there are no protractor or retractor muscles attached to the elongated part. Instead, the elongated part is tightly enveloped by a membrane. Because of the close fit between the elongated part and its surrounding membrane, eversion of the membrane allows for insertion of the elongated part, and retraction of the membrane induces withdrawal of the elongated part. This surrounding membrane makes it possible to store the median ejaculatory guide (MEG) in a small genital cavity, with the basal portion of the elongated part extending internally against the entrance. This unusual character state is observed only in members of the subgenus Lema, which also have the elongated part. Infection of the maize (Zea mays L.) with aflatoxigenic fungus Aspergillus flavus and consequent contamination with carcinogenic aflatoxin is a persistent and serious agricultural problem causing disease and significant crop losses worldwide. The rachis (cob) is an important structure of maize ear that delivers essential nutrients to the developing kernels and A. flavus spreads through the rachis to infect kernels within the ear. Therefore, rachis plays an important role in fungal proliferation and subsequent kernel contamination. We used proteomic approaches and investigated the rachis tissue from aflatoxin accumulation resistant (Mp313E and Mp420) and susceptible (B73 and SC212m) maize inbred lines. First, we compared rachis proteins from resistant and susceptible inbred lines, which revealed that the young resistant rachis contains higher levels of abiotic stress-related proteins and proteins from phenylpropanoid metabolism, whereas susceptible young rachis contains pathogenesis-related proteins, which are generally inducible upon biotic stress. Second, we identified A. flavus-responsive proteins in rachis of both resistant and susceptible genotypes after 10- and 35-day infection. Differential expression of many stress/defense proteins during rachis juvenility, maturation and after A. flavus challenge demonstrates that resistant rachis relies on constitutive defenses, while susceptible rachis is more dependent on inducible defenses.  相似文献

The heather beetle Lochmaea suturalis which is native to northwest Europe has been released as a biocontrol agent for heather in New Zealand. We have isolated and optimized eight microsatellite loci from New Zealand beetles. These loci provide markers with high polymorphism ranging from four to 20 alleles per locus. Observed heterozygosity averaged 0.631 per locus. These results suggest the markers are useful for population studies that will contribute to assessment of L. suturalis as a biocontrol agent.  相似文献

Ascorbic acid is a well‐known antioxidant and cellular reductant with an intimate and complex role in the response of plants to ozone. It is clear from a number of studies that sensitivity to ozone is correlated with total ascorbic acid levels, and that a first line of defence against the reactive oxygen species generated in the apoplastic space by ozone is ascorbic acid. For activity, ascorbic acid must be in the fully reduced state. Therefore, both the rate of ascorbic acid synthesis and recycling via dehydroascorbate and monodehydroascorbate reductases are critical in the maintenance of a high ascorbic acid redox state. Active transport of ascorbic acid across the plasma membrane is necessary to achieve reduction of oxidized ascorbic acid by cytoplasm‐localized reductases. It has been known for some time that the chlorotic lesions produced by exposure to ozone are not unlike lesions produced by the hypersensitive response to avirulent pathogen attack. Surprisingly, activation of a defence gene‐signalling network by both ozone and pathogens is influenced by the level of ascorbic acid. Indeed, in addition to acting simply as an antioxidant in the apoplastic space, ascorbic acid appears to be involved in a complex phytohormone‐mediated signalling network that ties together ozone and pathogen responses and influences the onset of senescence.  相似文献

Activity of ribulose 1,5-bisphosphate (RuBP) carboxylase in leaf extracts of the constitutive Crassulacean acid metabolism (CAM) plant Kalanchoe pinnata (Lam.) Pers. decreased with increasing leaf age, whereas the activity of phosphoenolpyruvate (PEP) carboxylase increased. Changes in enzyme activities were associated with changes in the amount of enzyme proteins as determined by immunochemical analysis, sucrose density gradient centrifugation, and SDS gel electrophoresis of leaf extracts. Young developing leaves of plants which received high amounts of NO 3 - during growth contained about 30% of the total soluble protein in the form of RuBP carboxylase; this value declined to about 17% in mature leaves. The level of PEP carboxylase in young leaves of plants at high NO 3 - was an estimated 1% of the total soluble protein and increased to approximately 10% in mature leaves, which showed maximum capacity for dark CO2 fixation. The growth of plants at low levels of NO 3 - decreased the content of soluble protein per unit leaf area as well as the extractable activity and the percentage contribution of both RUBP carboxylase and PEP carboxylase to total soluble leaf protein. There was no definite change in the ratio of RuBP carboxylase to PEP carboxylase activity with a varying supply of NO 3 - during growth. It has been suggested (e.g., Planta 144, 143–151, 1978) that a rhythmic pattern of synthesis and degradation of PEP carboxylase protein is involved in the regulation of -carboxylation during a day/night cycle in CAM. No such changes in the quantity of PEP carboxylase protein were observed in the leaves of Kalanchoe pinnata (Lam.)
