海水中DHA产生菌的筛选及一株高产菌的鉴定   总被引:2,自引:0,他引:2  
从海水中筛选产DHA的微生物,共采集海水样品280余份,用苏丹黑染色法得到160株产油脂菌株,在对60株脂肪粒较大的微生物用索氏抽提法提取油脂后,初筛得到油脂含量高于8%的菌株7株。对10株油脂产量较高的菌株进行复筛,编号7-3的菌株油脂含量达到15.9%,DHA在油脂中的含量达到45.2%,选用7-3作为目的菌株。对7-3进行形态特征、培养特征及生理生化特征鉴定,初步判定菌株7-3为酒香酵母属(Brettanomyces sp.)。  相似文献

【目的】确定新疆辐射污染土样中分离筛选获得的一株产多糖的耐辐射菌株W36-1的分类学地位和抗氧化特性。【方法】对该菌株进行的60Co辐照,结合菌落形态和菌丝特征显微观察、Biolog鉴定以及16S rRNA基因序列分析。采用苯酚硫酸法测定其多糖含量,采用Fenton法测定多糖的粗提物对超氧阴离子清除率;用DPPH法测定抗氧化性;采用邻苯三酚自氧化法测定对超氧阴离子的清除率。【结果】W36-1可耐5 kGy辐照,为一株欧文氏菌属(Erwinia)新种;其粗多糖含量为53.17%,该多糖对超氧阴离子清除率为75.63%,对DPPH清除率为62.43%,羟基自由基去除率为54.89%。【结论】菌株W36-1产的多糖具有抗氧化性。  相似文献

从牛的瘤胃中筛选获得一株能发酵生产琥珀酸的兼性厌氧菌。对其进行生理生化特性鉴定及16S rRNA基因分析。该菌株短杆状,无鞭毛,革兰氏染色阴性,V-P反应阴性,能发酵多种糖类产酸;其16S rRNA基因与琥珀酸放线杆菌的同源性高达99.8%,认为属于琥珀酸放线杆菌(Actinobacillus succinogenes),并将其命名为琥珀酸放线杆菌(Actinobacillus succinogenes)SW0580,保藏号CGMCC 1593。初步发酵试验表明该菌能发酵60g/L葡萄糖产生25.8g/L的丁二酸。  相似文献

[目的]探索獐子菌多糖(Sarcodon imbricatus (L. ex Fr.) Karst polysaccharide,SIK-1)的结构及对其调控小鼠免疫细胞和小鼠结肠癌细胞(CT26. WT)生长的影响。[方法]采用核磁共振谱(Nuclear Magnetic Resonance spectroscopy,NMR)检测SIK-1的结构,CCK-8法检测细胞增殖率、抑制率和吞噬率,Elisa试剂盒检测小鼠免疫B细胞分泌抗体Ig A、Ig G、Ig M的分泌率。[结果]SIK-1的1H NMR谱数据显示SIK-1含有α型吡喃糖和β型吡喃糖;13C NMR谱数据显示獐子菌多糖中含有6个糖残基,由3个α型糖残基和3个β型糖残基构成的重复结构单元。SIK-1质量浓度为5μg/m L和10μg/m L时能极显著地促进T细胞、B细胞和RAW264. 7细胞的增殖;且能抑制CT26. WT的生长,且呈一定的剂量关系; SIK-1质量浓度在15μg/m L时能显著促进B细胞分泌免疫因子Ig A、Ig G、Ig M; SIK-1质量浓度为5μg/m L时能显著提高巨噬细胞的吞噬能力。[结论]...  相似文献

精氨酸脱亚胺酶由于具有成为精氨酸营养缺陷型肿瘤如肝细胞瘤,黑素瘤治疗药物的潜力而被国内外多个科研机构进行研究。本文报道本研究室于无锡锡惠公园土样中筛选得到一株产精氨酸脱亚胺酶的菌株901,并对其进行了形态,生理生化特征分析及16S rRNA基因分析,该菌被鉴定为恶臭假单胞菌(Pseudomonas putida)。  相似文献

通过多点采样,多次反复筛选,从旱田土壤中筛选出1株具有高絮凝活性的细菌。细菌生长过程均可产生具有絮凝作用的胞外分泌物,经实验对内蒙天然碱碱泥具有高絮凝作用。通过对菌株的个体形态特征、菌落形态特征、运动性进行观察;同时对其接触酶、氧化酶等生理生化指标进行了测试,最终确定为粪产碱杆菌(Alcaligenes faecalis)。  相似文献

A simple and rapid method is described for the detection of amylase-producing micro-organisms on solid media using Remazol Brilliant Blue-starch as substrate. The blue starch is incorporated into nutrient agar and amylase production is detected by the disappearance of the colour around the colonies. The method which is non-destructive, allows for direct visualization and isolation of amylolytic micro-organisms from the environment without prior replication of colonies.  相似文献

A plate method was developed to screen for xylose isomerase-producing microorganisms based on the use of 2,3,5-triphenyltetrazolium as an indicator of D-xylulose, the D-xylose isomerization product. A rapid and effective method for direct detection, selection and testing of microorganisms able to produce both cell-bound and extracellular true lipases is described. The method is based on formation of clearance zones on turbid solid media with emulsified olive oil around or under the colonies, cell fractions or culture supernatant of lipase-producing organisms. 【背景】利用微生物处理秸秆引起研究者的广泛关注。【目的】筛选生长速度快、木质纤维素降解酶活性强的真菌菌株,用于植物秸秆降解和高效利用。【方法】从自然界采集的样品中分离纯化真菌菌株,利用PDA-愈创木酚和PDA-羧甲基纤维素钠平板初筛,再经过液体发酵检测漆酶酶活、羧甲基纤维素酶酶活及菌丝生长速率复筛目的菌株,通过内转录间隔区(internal transcribed spacer,ITS)测序法对目的菌株进行鉴定,对目的菌株产漆酶和羧甲基纤维素酶活力进行测定及酶学性质研究。【结果】从样品中分离纯化到18株真菌,通过初筛筛选出9株产木质纤维素降解酶真菌菌株,再经过复筛,筛选出一株产漆酶、羧甲基纤维素酶活力高、菌丝生长快的菌株M1,经过分子生物学鉴定M1为糙皮侧耳(Pleurotus ostreatus),其漆酶酶活为(243.59±1.11)U/mL,羧甲基纤维素酶酶活为(36.03±0.63) U/mL。在5 d的培养期内,菌丝生长速率为(9.43±0.32) mm/d。对菌株M1的发酵粗酶液的酶学性质进行了检测分析,结果表明,所产的漆酶在pH5.0-6.5相对酶活为90%以上,在pH ...  相似文献

A plate assay based on the visible solubilization of small substrate particles and the formation of haloes on Petri dishes, containing a mixture of different dye-labelled polysaccharides as substrates, provides a specific, reliable and rapid simultaneous detection of corresponding polysaccharide-degrading microorganisms. It has potential for increasing the efficacy of screening of microorganisms, utilizing different polysaccharides, in large numbers of natural samples. Diversely colored insoluble forms of amylose, xylan and hydroxyethyl-cellulose (HE-cellulose) were prepared as chromogenic substrates by using the cross-linking reagent 1,4-butanediol diglycidyl ether and the dyes Brilliant Red 3B-A, Cibacron Blue 3GA and Reactive Orange 14. Using the method, the bacteria with amylase or xylanase or cellulase or a combination of these activities were screened from soil and sludge samples, selected and identified according to 16S rDNA sequencing.  相似文献

Glutathione transferases (GSTs; EC 2.5.1.18) form a group of multifunctional enzymes catalyzing the conjugation of a broad range of toxicologically important halogenated compounds to the tripeptide glutathione (GSH) with concomitant halogen ion release. In the present work, a rapid quantitative screening method for GSTs based on colorimetric measurement of halogen ions released from halogenated xenobiotics was developed. The assay is based on the color formation resulting from the reaction of Hg(SCN)2 with the released halogen ion of the substrate in the presence of Fe3+. The color intensity is proportional to the extent of the catalytic reaction, allowing a quantitative measurement of the GST catalytic activity. The assay can be performed using purified recombinant enzyme (the isoenzyme GmGSTU4-4 from Glycine max) or crude recombinant Escherichia coli cell lysates in 96-well microtiter plates. The suitability of the colorimetric assay for screening mutant GST variants derived from a directed evolution library was successfully evaluated. In addition, the assay was also used for screening GST synthetic inhibitors. It was concluded that the proposed colorimetric assay is selective and sensitive and allows the screening of large numbers of samples within a few minutes.  相似文献

A new method for monitoring reactions catalyzed by an immobilized enzyme, cross-linked penicillin acylase aggregates (PA CLEA), is suggested. Appropriate chromogenic substrates for spectrophotometric assay of catalytic activity of immobilized enzyme were chosen and their kinetic parameters determined. Active sites in PA CLEA preparations were titrated by the suggested method; it is shown that almost all active sites are retained during immobilization. This method is characterized as highly expressive, simple, and precise and may be used for control of PA immobilization efficiency as well as for study of operational, thermal, and pH stability of immobilized enzyme preparations.  相似文献

为了快速从土壤中定向筛选抗生素产生菌, 本研究利用宏基因组技术进行了土壤中抗生素产生菌快速筛选方法的探索。对6 种不同土质的土壤利用液氮冷冻法提取土壤基因组DNA 并用PVPP柱层析法进行纯化, 每克湿土可得到32.25?61.25 μg DNA, 片段大小为23 kb左右, 说明宏基因组DNA样品完整, 16S rDNA的 PCR结果证明了该基因组DNA纯度可用于后期的分子操作。利用Herbimycin产生菌掺入法进行了抗生素产生菌筛选方法学的验证。结果证明, 每克湿土掺入103个Herbimycin产生菌时, 利用Herbimycin合成基因簇的特定引物即可从所提取的土壤基因组DNA中扩增出目标条带, 并且可以利用传统菌种分离方法从土壤中重新分离出来被掺入到土壤的Herbimycin产生菌。这些结果证明本研究建立的定向抗生素产生菌筛选方法, 具有快捷、灵敏的特点, 大大减少传统筛选方法的工作强度和时间, 为微生物新药的研发建立一种全新的研究方法。  相似文献
