Protein kinase Cι (PKCι) is an atypical PKC isoform and participates in multiple aspects of the transformed phenotype in human cancer cells. We previously reported that frequent amplification and overexpression of PKCι were correlated with lymph node metastasis in primary esophageal squamous cell carcinomas (ESCC). In the present study, short interfering RNA-mediated silencing of PKCι revealed that this enzyme was required for cell migration, invasion, and resistance to anoikis. In vivo experiments showed that PKCι suppression decreased tumor growth in esophageal cancer xenografts and lung metastases in nude mice. At the molecular level, knockdown of PKCι in suspended ESCC cells caused a decrease in S-phase kinase-associated protein 2 (SKP2) that had been reported to promote resistance to anoikis via the PI3K/AKT pathway. AKT phosphorylation was abolished after PKCι suppression, but AKT activation could be refreshed by PKCι upregulation, suggesting that PKCι enhanced cell resistance to anoikis via the PKCι-SKP2-PI3K/AKT pathway. Addition of the proteasome inhibitor MG132 prevented the decrease of SKP2 in PKCι silenced cells, and polyubiquitin-SKP2 was elevated after PKCι depletion, showing that PKCι might regulate the expression of SKP2 through the ubiquitin-proteasome pathway in suspended cells. Furthermore, overexpression of SKP2 in PKCι-downregulated cells restored cell resistance to anoikis. Most importantly, PKCι expression significantly correlated with SKP2 in 133 ESCC tissues (P = 0.031). Taken together, our data show that PKCι promotes tumorigenicity and metastasis of human esophageal cancer and that SKP2 is a candidate downstream effector of PKCι signaling in ESCC.  相似文献

Cancer cells that survive fractionated irradiation can be radioresistant and cause tumor recurrence. However, the molecular mechanisms underlying the development of radioresistance in cancer cells remain elusive. The aim of this study was to investigate the role of WISP-1 in the development of radioresistance in esophageal carcinoma during fractionated irradiation. Radioresistant esophageal cancer cells were generated from normal esophageal cancer cells via fractionated irradiation, and expression levels of related proteins were determined by Western blot. Radiosensitivity of cells was established by clonogenic cell survival assays, and cell cycle distribution was evaluated by flow cytometry. Protein distributions were determined by immunofluorescence, and cell toxicity was evaluated by cell counting kit-8 assays. In vivo validations were performed in a xenograft transplantation mouse model. Our data indicate that WISP-1 plays an important role in the development of radioresistance in esophageal cancer cells during fractionated irradiation. The overexression of WISP-1 in esophageal cancer cells was associated with radioresistance. Depletion of extracellular WISP-1 by antibody neutralizing reversed radioresistance and directly induced mitotic catastrophe resulting in cell death. WISP-1 may be a candidate therapeutic target in the treatment of recurrent esophageal carcinoma after radiotherapy.  相似文献

目的为探讨P16、P27蛋白在食管鳞状上皮、增生上皮和癌变上皮中表达状况及其与鳞状细胞癌发生、进展和转移的相关性.方法采用SP免疫组织化学方法,检测72例(其中活检标本13例)食管癌组织中P16、P27蛋白的表达情况.结果 P16、P27在食管癌和增生性上皮均有阳性表达,但两者相比均无统计学意义(P>0.05),在正常上皮组均无阳性表达;P16、P27在高分化鳞癌组阳性表达率均显著高于低分化鳞癌组(P<0.05);P16、P27在伴有淋巴结转移的食管癌组与不伴有淋巴结转移组比较差异显著(P<0.05).P16在原发食管癌组阳性表达率和淋巴结转移癌中比较具有显著性差异(P<0.05).结论 P16、P27基因蛋白在食管癌组织中的表达与病理分化程度有关;P16的表达与转移癌的形成有关;P16、P27表达与患者性别、年龄、肿瘤发生部位、浸润深度无明显相关性.  相似文献

目的 探讨血管生成拟态( vasculogenic mimicry,VM)与E-钙粘蛋白(E-cadherin,E-cad))在食管鳞癌(esophageal squamous cell carcinoma)组织中的表达及意义.方法 收集食管鳞状细胞癌术后标本100例和30例癌旁正常食管黏膜,应用免疫组化法和组织化学法检测食管鳞状细胞癌和正常食管黏膜组织中VM和E-cad的表达情况.结果 在食管鳞状细胞癌组织和正常食管黏膜组织中,VM和E-cad的阳性表达率分别为47.0%、48.0%和0%、70.0%,差异有统计学意义(P<0.05);VM及E-cad的表达与食管鳞癌的组织学分级、临床分期及淋巴结转移(P<0.05);VM与E-cad在食管鳞癌中的表达呈负相关(r=-0.865,P=0.000).多因素分析:PTNM分期、淋巴结转移、VM和E-cad的表达是影响食管鳞癌根治术后患者预后的独立因素(P<0.05);VM阳性组与阴性组的5年生存率分别为4.3%和64.2%,,差异有统计学意义(P=0.000);E-cad阳性组与阴性组5年生存率分别为60.4%和15.4%,差异有统计学意义(P=0.000).结论 具有VM结构的食管鳞状细胞癌的分化程度低,恶性度高,预后差,VM和E-cad表达的程度与食管鳞状细胞癌的进展和预后密切相关.  相似文献

目的:通过检测食管鳞癌标本中核因子E2相关因子1(NFE2L1)的表达情况,探究NFE2L1在食管鳞癌发生发展中的作用,为食管鳞癌的诊治以及预后评估提供新的思路。方法:收集我校附院2016-2017年经病理组织学检查诊断为食管鳞癌的手术标本40例及其对应的癌旁组织并从NCBI数据库下载GEO测序数据,应用定量PCR、免疫组化和生物信息分析等方法检测分析NFE2L1基因的表达情况。结果:在食管鳞癌组织中,NFE2L1表达阳性31例(77.5%),癌旁组织阳性表达17例(42.5%),两组比较差异显著有统计学意义(P0.001);进一步发现NFE2L1的阳性表达与肿瘤分化程度和淋巴转移相关(P0.05)。但在不同年龄、性别、浸润深度及不同部位之间差异无统计学意义。GEO数据分析结果显示NFE2L1在食管鳞癌组织显著高表达(P0.01),只是未达到显著差异表达的阈值标准(即变化倍数小于2倍)。结论:NFE2L1在食管鳞癌中高表达,表达的高低与食管鳞癌的发生进展密切相关。  相似文献

目的:探讨缺氧诱导因子-1α(HIF-1α)、生存蛋白(survivin)、细胞周期蛋白D1(cyclinD 1)在食管癌组织中的表达及其临床意义。方法:应用免疫组化技术检测50例食管癌组织和10例手术切除的远端正常食管组织中HIF-1α、Survivin、CyclinD 1蛋白的表达。结果:食管癌组织中HIF-1α、Survivin、CyclinD 1蛋白阳性表达率均与肿瘤浸润深度以及淋巴结转移相关(P〈0.05),Survivin阳性表达率与肿瘤分级相关(P〈0.05),HIF-1α与CyclinD 1的表达呈显著正相关(P〈0.05)。结论:检测HIF-1α、Survivin、CyclinD 1的蛋白的表达有助于判断食管癌的恶性程度以及推断其临床预后。  相似文献

Emodin, a tyrosine kinase inhibitor, is a natural anthraquinone derivative found in the roots and rhizomes of numerous plants. It reportedly exhibits an anticancer effect on lung cancer. Gefitinib (Iressa) is a selective epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor for human non-small cell lung cancer (NSCLC). However, the molecular mechanism of how emodin combined with gefitinib decreases NSCLC cell viability is unclear. The recombinase protein Rad51 is essential for homologous recombination repair, and Rad51 overexpression is resistant to DNA double-strand break-inducing cancer therapies. In this study, we found that emodin enhanced the cytotoxicity induced by gefitinib in two NSCLC cells lines, A549 and H1650. Emodin at low doses of 2-10 μM did not affect ERK1/2 activation, mRNA, and Rad51 protein levels; however, it enhanced a gefitinib-induced decrease in phospho-ERK1/2 and Rad51 protein levels by enhancing Rad51 protein instability. Expression of constitutively active MKK1/2 vectors (MKK1/2-CA) significantly rescued the reduced phospho-ERK1/2 and Rad51 protein levels as well as cell viability on gefitinib and emodin cotreatment. Blocking of ERK1/2 activation by U0126 (an MKK1/2 inhibitor) lowered Rad51 protein levels and cell viability in emodin-treated H1650 and A549 cells. Knockdown of Rad51 expression by transfection with si-Rad51 RNA enhanced emodin cytotoxicity. In contrast, Rad51 overexpression protected the cells from the cytotoxic effects induced by emodin and gefitinib. Consequently, emodin-gefitinib cotreatment may serve as the basis for a novel and better therapeutic modality in the management of advanced lung cancer.  相似文献

目的探讨共济失调性毛细血管扩张症突变基因(Ataxia-telangiectasia mutated,ATM)mRNA在食管鳞状细胞癌组织中的表达及其临床意义。方法应用原位分子杂交方法检测52例食管正常黏膜、45例食管上皮内瘤变组织及63例食管癌组织ATM mRNA的表达。结果食管正常黏膜、食管上皮内瘤变及食管癌组织中ATM mRNA表达率分别为26.9%(14/52)、44.4%(20/45)及63.5%(40/63),食管癌组织中ATM mRNA表达率明显高于正常黏膜及上皮内瘤变(P≤0.05),ATM mRNA表达率与食管癌组织分级呈负相关(r=-0.312,P=0.013);食管正常黏膜和食管上皮内瘤变ATM mRNA表达率无明显差异(P=0.07),食管癌组织中ATM mRNA表达与患者年龄、性别、肿瘤浸润深度、淋巴结转移、临床分期及其它临床病理因素无关(P0.05)。结果 ATM mRNA在食管癌组织中异常表达,有望成为食管癌治疗的新靶点。  相似文献

目的:检测葡萄糖调节蛋白78(glucose—regulated proteins78,GRP78)在同一食管癌患者食管的鳞状细胞癌、不典型增生及正常鳞状上皮组织中的表达,研究GRP78与食管鳞状细胞癌发生发展的关系。方法:取90例食管鳞状细胞癌患者的病变组织,其病理切片中正常鳞状上皮组织、不典型增生组织和鳞状细胞癌组织均存在,用免疫组化的方法,检测GRP78在3种不同组织中表达的情况,分析G史P78与性别、年龄、浸润深度、分化程度、分期及淋巴结转移等参数之间的关系。结果:GRP78在食管正常鳞状上皮组织、不典型增生组织、鳞状细胞癌组织中的阳性表达率分别为7.8%、85.6%、47.8%,GRP78在正常食管组织、不典型增生组织、食管鳞癌组织3组中表达的差异有显著性意义(P〈0.01)。鳞状细胞癌组织中GRP78表达阴性者,年龄较阳性者大;GRP78表达与肿瘤分化程度、分期、淋巴结转移等有明显相关性。结论:GRP78在食管正常细胞向恶性细胞转化的过程中可能扮演了重要角色,检测GRP78的表达可能有助于对食管鳞状细胞癌的预防及早期诊断。  相似文献

Rad51 protein is essential for homologous recombination repair of DNA damage, and is over-expressed in chemo- or radioresistant carcinomas. The polycyclic hydrocarbon carcinogen benzo[a]pyrene (B[a]P) affects MAPKs transduction pathways. Gefitinib (IressaR, ZD1839) is a selective epidermal growth factor receptor tyrosine kinase inhibitor that blocks growth factor-mediated cell proliferation and ERK1/2 activation. We hypothesized that gefitinib enhances B[a]P-mediated cytotoxicity by decreasing ERK1/2 activation. Exposure of human lung cancer cells to gefitinib decreased B[a]P-elicited ERK1/2 activation and induced Rad51 protein expression. Gefitinib and B[a]P co-treatment decreased Rad51 protein stability by triggering degradation via a 26S proteasome-dependent pathway. Expression of constitutive active MKK1/2 vectors (MKK1/2-CA) rescues the decreased ERK1/2 activity, and restores Rad51 protein level and stability under gefitinib and B[a]P co-treatment. Gefitinib enhances B[a]P-induced growth inhibition, cytotoxicity and mutagenicity. Co-treatment with gefitinib and B[a]P can further inhibit cell growth significantly after depletion of endogenous Rad51 by siRad51 RNA transfection. Enhancement of ERK1/2 activation by MKK1-CA expression decrease B[a]P- and gefitinib-induced cytotoxicity, and B[a]P-induced mutagenicity. Rad51 protein protects lung cancer cells from synergistic cytotoxic and mutagenic effects induced by gefitinib and B[a]P. Suppression of Rad51 protein expression may be a novel lung cancer therapeutic modality to overcome drug resistance to gefitinib.  相似文献

目的:探讨血管生成拟态(vasculogenic mimicry,VM)与食管鳞癌临床病理特征的关系及其对患者预后的影响,并分析食管癌血管生成拟态的形成机制。方法:收集57例食管鳞癌石蜡包埋样本,进行过碘酸雪夫氏(PAS)及CD34免疫组织化学双重染色,结合HE染色,观察食管鳞癌血管生成拟态的发生情况。对患者临床病理和预后信息进行单因素分析,Kaplan-Meier生存比较和Cox风险模型分析。通过食管鳞癌细胞株Eca-109三维培养建立,观察RNAi沉默VE-cadherin对食管鳞癌Eca109血管生成拟态形成的影响。结果:食管鳞癌中VM表达的阳性率为54.3%,显著高于正常食管黏膜组织;VM在病理分型为低分化食管鳞癌的阳性表达率为78.9%,显著高于中高分化组(P0.05);III-Ⅳ期食管鳞癌患者VM阳性率显著高于Ⅰ-Ⅱ期食管鳞癌患者(P0.05);有淋巴结转移的食管鳞癌者VM阳性率明显高于无淋巴结转移者(P0.05)。单因素分析结果显示食管鳞癌VM的发生率与肿瘤的分化程度、TNM分期和淋巴转移显著相关。Kaplan-Meier生存分析显示有VM组食管鳞癌患者的生存期明显短于无VM组(P0.05);Cox分析显示VM是影响食管鳞癌患者预后的独立危险因素(RF=0.67)。三维培养结果显示Eca-109细胞在基质胶上形成典型的血管网状样结构,VE-cadherin-siRNA可有效抑制VE-cadherin在Eca109的表达,抑制体外培养的Eca109细胞VM的形成。结论:血管生成拟态是食管鳞癌一种独特的血液供应模式,与食管鳞癌的分化程度、TNM分期、淋巴转移密切相关,是食管鳞癌患者术后生存期的独立危险因素。  相似文献

目的:探讨原发性口腔鳞癌患者组织和血清中内皮抑素表达及与肿瘤分期、分级的关系。方法:采用免疫组化方法检测36例口腔鳞癌和12例正常口腔粘膜组织中内皮抑素表达情况。ELISA法检测36例口腔鳞癌患者术前血清内皮抑素水平,14例健康者血清做对照。结果:内皮抑素主要见于肿瘤组织细胞质。正常口腔粘膜中内皮抑素表达率为7.15%,口腔鳞癌组织中内皮抑素阳性率为76.44%,其中G1、G2、G3级阳性率分别为47.21%、79.17%、90.90%,病理分级间比较差异有统计学意义(P〈0.05)。口腔鳞癌患者血清中内皮抑素水平(49.62±1.72)ng/mL显著高于健康对照者(5.60±0.37)ng/mL(P〈0.05),TNM分期III、IV期肿瘤患者血清内皮抑素水平显著高于I和II期(P〈0.05)。结论:口腔鳞癌患者组织和血清中内皮抑素表达显著升高,并与肿瘤分期、分级相关,检测内皮抑素表达有助于判断口腔鳞癌恶性程度。  相似文献

Fatty acid synthase (FASN) overexpression has also been associated with a variety of human malignancies including tumor progression, aggressiveness, and metastasis. To investigate the role of FASN expression in esophageal cancer, we evaluated 60 cases of squamous cell carcinoma, 20 cases of adenocarcinoma, and 10 cases of normal esophageal tissues. We found that FASN was detected in 95?% human squamous cell carcinoma, and in 90?% human adenocarcinoma samples. However, all cases of normal esophageal epithelium did not express the protein of FASN. Further, to investigate the role of FASN in tumorigenesis and development, we analyze the growth and migration by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), colony formation and wound healing assay. We found that inhibition of FASN expression in TE13 cells by RNAi suppressed the growth of cells. Decreased FASN expression mitigated the migration of TE13 cells. These studies demonstrated the functional importance of FASN in esophageal tumorigenesis, and suggested that inhibiting FASN might be applied to treat esophageal cancer.  相似文献
