Allozyme investigation of the five Cimicifuga taxa in Korea was conducted to assess genetic and clonal diversity within populations and genetic divergence among populations and taxa. Levels of allozyme variation maintained in Korean Cimicifuga taxa were comparable to those for most herbaceous perennials. In general, samples excluding copies of the same multilocus genotype maintained higher levels of genetic diversity than the total samples within populations. Copies of homozygous genotypes at several loci resulting from clonal spread lead to decreased levels of genetic diversity within populations, indicating that clonal reproduction found in Cimicifuga affects population genetic structure. In general, more widely distributed species such as C. dahurica and C. japonica harbored higher levels of allozyme diversity than the other taxa examined. Although two varieties of C. heracleifolia are geographically and reproductively isolated, the genetic and clonal structure of var. bifida seems to resemble var. heracleifolia, indicating that the two varieties may have had a similar evolutionary history. However, the allozyme data strongly indicate that the two morphological types (Groups I and II) of C. simplex should be treated as separate species.  相似文献

Ligustrum ovalifolium is a semi-evergreen tree distributed in the western part of Japan and southern Korea. This species contains an insular endemic variety, L. ovalifolium var. pacificum; this variety occurs only on the Izu Islands located south of the Japanese mainland Honshu. We isolated 10 polymorphic microsatellite loci from the species, and characterized them for 21 individuals from a population of L. ovalifolium. The primers developed in this study yielded an average 12.2 alleles per locus and an average expected heterozygosity of 0.78. These markers will be powerful tools for studying the genetic differentiation within the species.  相似文献

Erythronium propullans is a narrow endemic restricted to two counties in southeastern Minnesota. It coexists at all sites with the widespread common white trout lily, E. albidum. Erythronium albidum has both sexual and asexual reproduction; it has been suggested that E. propullans reproduces strictly by vegetative means. Electrophoretic analysis of 15 enzyme systems encoded by a minimum of 37 genetic loci revealed considerable genetic variation within and among E. propullans populations. However, all measures of variability calculated from allozyme data demonstrate that E. propullans maintains lower levels of genetic variation than E. albidum. There is also evidence for limited gene flow among E. propullans populations, whereas all E. albidum populations are genetically similar. Clonal diversity is also lower in E. propullans. Only 21 clones were found in 100 individuals sampled whereas virtually every one of the 70 individuals of E. albidum sampled had a unique multilocus genotype. The clones of E. propullans cluster into 3 genetically dissimilar groups; variation within each cluster indicates that some sexual recombination does occur. We discuss the effects of clonality and endemism on genetic variation, reasons why the clonal habit might preserve genetic variation, and suggest that the lower variability in E. propullans is due primarily to the genetic bottleneck it experienced at the time of its origin. Isozyme data suggest that E. propullans is a derivative of E. albidum. Based on the degree of genetic similarity and information on the glacial history of the region, we suggest that E. propullans is a local derivative of E. albidum, having arisen no more than 9,000 years ago.  相似文献

Two of the four members of subsection Contortae of the genus Pinus occur in the southeastern United States: Pinus virginiana, which ranges throughout the southern and central Appalachian Mountains, and P. clausa, which is restricted to Florida and southern Alabama. We examined allozyme variation within P. virginiana and genetic relationships between this species and the two varieties of P. clausa (var. clausa and var. immuginata). P. virginiana maintains more genetic diversity at both the species (Hes = 0.139) and population (Hep = 0.128) levels than the other three species in the subsection, which may reflect the combination of its widespread distribution and the absence of cone serotiny. Genetic differentiation among populations in P. virginiana was relatively low (GST = 0.053), but significant contrasts in allozyme frequencies and genetic diversity were apparent between populations to the northwest vs. outheast of the Appalachian Mountains. These regional differences likely resulted initially from historical processes that occurred during the Pleistocene and early Holocene, and have been reinforced by modern selective pressures and barriers to gene flow. The mean genetic distance between populations of P. virginiana and P. clausa (D = 0.071) was greater than that between populations of the two varieties of P. clausa (D = 0.012), which suggests that the two varieties diverged at some point after the separation of the two species.  相似文献

We used data from 12 allozyme loci for two endemic Brassicaceae from Gran Canaria (the endangered narrow endemic Crambe tamadabensis and its more widespread congener C. pritzelii) to assess whether their genetic diversity patterns reflect their phylogenetic closeness and contrasting population sizes and distribution areas, and to derive conservation implications. Genetic diversity values are high for both species and slightly higher in C. tamadabensis, despite its narrow distribution in north‐western Gran Canaria. At odds with the generally high interpopulation diversity levels reported in Canarian endemics, values of GST in C. tamadabensis and C. pritzelii are rather low (0.067 and 0.126, respectively). We construe that the higher genetic structure detected in C. pritzelii is mainly a result of unbalanced allele frequencies and low population sizes at the edges of its distribution. The overall high allozyme variation detected in C. tamadabensis and C. pritzelii is nevertheless compatible with an incipient but consistent genetic differentiation between the two species, modulated by recurrent bottlenecks caused by grazing and drift. Our data suggest that conservation efforts aimed at maintaining the existing genetic connectivity in each species and ex situ conservation of seeds are the best strategies to conserve their genetic diversity.  相似文献

Genetic structure arises when limited gene flow between populations favours the development of distinct arrays of genetic characters within each population. Determining the spatial scale at which this differentiation occurs is critical to our understanding of population biology and microevolution of species. The genetic structure and spatial pattern of genetic variation in an endemic, clonal perennial, Wyethia reticulata E. Greene, was investigated using random amplified polymorphic DNA (RAPD) markers and allozyme alleles. Large stands (250–360 m2) were found to contain few genetic individuals. Despite the small population sizes and endemism of the species, W. reticulata was highly diverse genetically, with most of the variation (75–81%) distributed within populations. A population structure in full agreement with spatially defined populations was achieved only by combining RAPD and allozyme markers. Analysis using both types of markers appeared to provide estimates of genetic similarity between individuals that were most consistent with empirical data on plant distributions. We postulated that large, long-lived clones dominated genetic relationships within populations but also provided opportunities for gene flow between populations on a longer time scale. The two marker types yielded different estimates of between-individual similarity and revealed disparate patterns of population structure. This result will arise because allozymes and random DNA segments have dissimilar evolutionary dynamics with respect to mutation and selection.  相似文献

SUMMARY 1. Population genetic structure of Wormaldia tagananana, a caddis with a narrow geographic range and endemic to the Canary Islands, was investigated by studying allozyme variation at 11 putative loci in five of the eight extant populations on Tenerife and La Gomera. Genetic variability, population structure and gene flow were compared with those reported previously in more widespread Trichoptera, particularly Canarian populations of the non-endemic limnephilid Mesophylax aspersus, to examine the hypothesis that the Wormaldia , with its restricted range, would exhibit relatively little genetic variability and gene flow.
2. Despite it being a narrow-range island endemic, genetic variability in populations of W. tagananana is broadly similar to values reported for more widespread caddis.
3. Significant genetic population structure was observed in W. tagananana (overall F ST = 0.387), greater than that seen in M. aspersus and amongst the highest reported for lotic caddis to date. Several site- and island-specific alleles were reported, providing further evidence for the relative isolation of individual Wormaldia populations.
4. Significant deviations from Hardy–Weinberg equilibrium were found in four of five populations (overall F IS = 0.675). This could result from within-locality population substructuring, or offspring within a reach being the product of a limited number of matings.
5. This genetic evidence supports the hypothesis that the restricted range of W. tagananana is, at least in part, because of limited dispersal ability.  相似文献

Genetic variation and differentiation in six island populations of two species ( Cynopterus brachyotis Müller and Haplonycteris fischeri Lawrence) of Philippine fruit bats (Chiroptera, Pteropodidae) were analysed using allozyme electrophoresis. Cynopterus is eurytopic and widespread in southeast Asia; Haplonycteris is stenotopic and endemic to the Philippines. Genetic variability within populations is consistently higher in Cynopterus , but differentiation between populations is much more pronounced in Haplonycteris. Genetic variation is not significantly correlated with island size in either species, but a positive trend is present in both. Levels of gene flow are sufficiently low to allow differentiation by genetic drift alone in Haplonycteris ( Nm = 0.05), but not in Cynopterus ( Nm = 7.5). There is no significant association between genetic distance and distance between sampling sites; however, between-population differentiation is positively related to degree of geographic isolation during Pleistocene periods of low sea level and to vagility and consequent levels of gene flow among populations. Significant effects of population size on genetic differentiation were not found. Genetic distance matrices for the two species share a common structure that is similar to patterns of mammalian faunal similarity for the Philippines as a whole, suggesting similar effects of geographic and/or environmental factors.  相似文献

We examined the adaptive importance of allozyme variation in wild barley (Hordeum spontaneum). The test involved a nested sampling design with four population groups, each representing a different environment, and a comparison of observed allozyme variation with that expected under the assumption that allozymes are not neutral. Measurements of plant fitness in indigenous and alien environments in reciprocal introductions of seeds and seedlings in the four environments provided a guideline for the expected pattern of allozyme variation. The results showed considerable variation in both the degree of regional and population subdivision and the pattern of the subdivision among loci. The observed pattern of variation was ambiguous. Although two alleles exhibited a pattern of distribution that cannot be explained by genetic drift as a function of geographic distance, we failed to detect either a significant relationship between genetic distance and environmental similarity or any favored epistatic allele combinations across the four environments. Our results suggest that interpretation of allozyme variation in wild barley as adaptive and directly related to local environment still needs justification. Although we could not reject the null hypothesis, a proposed methodology seeking a concordance between observed and "adaptive" (i.e., expected under hypothesis that allozymes are not neutral) allozyme variation may prove to be effective in resolving the neutralist-selectionist debate when applied to other species.  相似文献

Buffalograss, Buchloë dactyloides, is widely distributed throughout the Great Plains of North America, where it is an important species for rangeland forage and soil conservation. The species consists of two widespread polyploid races, with narrowly endemic diploid populations known from two regions: central Mexico and Gulf Coast Texas. We describe and compare the patterns of allozyme and RAPD variation in the two diploid races, using a set of 48 individuals from Texas and Mexico (four population samples of 12 individuals each). Twelve of 22 allozyme loci were polymorphic, exhibiting 35 alleles, while seven 10-mer RAPD primers revealed 98 polymorphic bands. Strong regional differences were detected in the extent of allozyme polymorphism: Mexican populations exhibited more internal gene diversity (He= 0.20, 0.19) than did the Texan populations (He= 0.08, 0.06), although the number of RAPD bands in Texas (n= 62) was only marginally smaller than in Mexico (n= 68). F-statistics for the allozyme data, averaged over loci, revealed strong regional differentiation (mean FRT=+ 0.30), as well as some differentiation among populations within regions (mean FPR=+ 0.09). In order to describe and compare the partitioning of genetic variation for multiple allozyme and RAPD loci, we performed an Analysis of Molecular Variance (AMOVA). AMOVA for both allozyme and RAPD data revealed similar qualitative patterns: large regional differences and smaller (but significant) population differences within regions. RAPDs revealed greater variation among regions (58.4% of total variance) than allozymes (45.2%), but less variation among individuals within populations (31.9% for RAPDs vs. 45.2% for allozymes); the proportion of genetic variance among populations within regions was similar (9.7% for RAPDs vs. 9.6% for allozymes). Despite this large-scale concordance of allozyme and RAPD variation patterns, multiple correlation Mantel techniques revealed that the correlations were low on an individual by individual basis. Our findings of strong regional differences among the diploid races will facilitate further study of polyploid evolution in buffalograss.  相似文献

Centaurea corymbosa Pourret (Asteraceae) is a narrow endemic species known only from six populations located in a 3-km2 area in the south of France. Earlier field experiments have suggested that pollen and seed dispersal were highly restricted within and among populations. Consistent with the field results, populations were highly differentiated for five allozyme loci and among-population variation fitted an isolation-by-distance model. In the present study, we investigated the genetic structure of C. corymbosa using six microsatellite loci. As with allozymes, microsatellites revealed no within-population structure and a large differentiation among populations. However, allozyme loci were less powerful than microsatellites in detecting the extent of gene flow assessed by assignment tests. The patterns of structuration greatly varied among loci for both types of marker; we suggest that differences in single-locus pattern could mainly be an effect of stochastic variation for allozymes and an effect of variation in mutation rate for microsatellites. In contrast to the multilocus results, the two most polymorphic microsatellite loci did not show any isolation-by-distance pattern. Our results suggest that highly variable loci might not always be the best suited markers to quantify levels of gene flow among populations.  相似文献

BACKGROUND AND AIMS: Dalbergia nigra, known as Brazilian rosewood, is an endangered tree species restricted to the Brazilian Atlantic Forest and has been intensively logged for five centuries due to its high-quality wood. The objective of the present study was to assess the genetic variation and structure in adults and saplings of the species from a large reserve of the Atlantic Forest, the Rio Doce State Park, and from two small surrounding fragments, one better preserved and another with a high degree of anthropogenic disturbance. METHODS: Analyses of genetic variation and structure were conducted by studying allozyme markers. Seven putative enzymatic loci were resolved, five of them being polymorphic. KEY RESULTS: The mean numbers of alleles per locus (A) were 1.93 and 1.73, while the percentages of polymorphic loci (P) were 93 and 73 % for adults and saplings, respectively. Saplings from the fragment with high anthropogenic disturbance exhibited the lowest values of A and P. The fragment that constitutes a conservation area exhibited genetic variation similar to the population from the large reserve. The observed (H(o)) and expected (H(e)) heterozygosities were not significantly different among the three populations. Only sapling populations showed F(ST) values (divergence among populations) significantly different from zero over all studied loci. The fragment with high anthropogenic disturbance exhibited considerable genetic divergence in relation to the above-cited populations. CONCLUSIONS: The evaluated populations displayed mean levels of genetic variation intermediate to those expected for narrow and widespread species. The results suggest that fragments with similar area and geographical distance from a large protected reserve can exhibit different levels of genetic variation, depending on the degree of anthropogenic disturbance. The considerable genetic variation in the protected fragment points to the importance of adequate conservation of small fragments for the preservation of genetic variation in D. nigra.  相似文献

Evolutionary theory predicts that species with small ranges and few individuals will exhibit low levels of genetic polymorphism. We investigated the population genetic structure of two locally endemic and two geographically widespread species of Astragalus. To facilitate direct comparisons among these congeners, three populations of each species were sampled in a consistent manner and scored for allozyme polymorphisms at 12 loci. Genetic polymorphism was lower in restricted A. linifolius and A. osterhouti than in widespread A. pectinatus. However, the restricted species do exhibit a moderate level of isozyme variation, comparable to that of widespread A. pattersoni and higher than has been detected in several other rare plant taxa. As measured by Wright's F statistics, there were no consistent differences between the restricted and widespread taxa with respect to the organization of genetic variation. FST values were low in all taxa, indicating little heterogeneity among populations.  相似文献

Few studies of genetic variation have been conducted on plants of the Pacific coastal desert and neighboring Andes of Peru, although the region has many endemic taxa. Enzyme electrophoresis was employed to examine allozyme diversities of four species of the family Malesher-biaceae, an endemic to the arid Andes and coastal desert. One population ofMalesherbia splendens and two ofM. tubulosa, both endemics of the department of Lima, one population ofM. weberbaueri var.weberbaueri, an endemic of the Andean department Huancavélica, and the two Lima populations ofM. scarlatiflora were studied. Fifteen loci were examined for all populations and an additional seven loci were resolved forM. weberbaueri andM. splendens. Malesherbia splendens, which is known from three populations, has a low mean number of alleles per locus (A), proportion of polymorphic loci (P), and expected heterozygosity (Hs) (A=1.214, P=0.214, Hs=0.057).Malesherbia weberbaueri (A=1.231, P=0.154, Hs=0.079) andM. scarlatiflora (A=1.364, P=0.273, Ht=0.083) both have average expected heterozygosities and relatively low mean numbers of alleles per locus and proportions of polymorphic loci. InM. tubulosa, all measures of genetic diversity are high in comparison with other endemics (A=1.818, P=0.364, Ht=0.206).Malesherbia tubulosa has high interpopulation differentiation, whereasM. scarlatiflora has low among-population diversity. The relatively low allozyme diversities, restricted habitats, narrow ranges ofM. splendens andM. weberbaueri, and the morphological similarities of all four species suggest that they evolved recently by founder events. Greater allozyme diversities inM. tubulosa could be attributable to its maintenance of larger populations in a greater variety of habitats.  相似文献

Phaedranassa schizantha (Amaryllidaceae) is an endangered species endemic to Ecuador and two varieties have been described: P. schizantha var. schizantha and P. schizantha var. ignea. We assessed population genetic structure and demographic patterns in 11 populations across the range of the species using 13 microsatellite loci. Our data show that genetic diversity was generally lower in the southern part of the range and was especially low in populations closest to cities. We found significant population differentiation (FST = 0.14, DEST = 0.34) and evidence of a genetic bottleneck. Genetic variation did not show isolation by distance. Instead, results suggest genetic barriers around two main cities. Bayesian analysis identified two genetic groups, neither of which represents either of the two varieties previously recognized. Coalescent analysis indicates a relatively recent colonization pattern between the two genetic groups (< 3000 generations). Conservation efforts need to be taken to facilitate genetic exchange between the groups, especially between locations that seem to be genetically isolated.  相似文献

Rhus michauxii, a rare plant species endemic to the southeastern United States, was previously known only from central North Carolina and one site in Georgia. An additional site, which is now believed to represent the largest known concentration of R. michauxii, was recently discovered at Ft. Pickett near Blackstone, Virginia. Morphological characteristics in several of the Ft. Pickett Rhus populations appear to be intermediate between R. michauxii and the widespread R. glabra, a closely related congener that co-occurs at Ft. Pickett. Although morphological evidence of hybridization between R. michauxii and R. glabra in North Carolina has been provided previously, genetic marker data are lacking. In the present study we examined levels of allozyme variation at 11 polymorphic loci within and among seven populations of R. michauxii, one population of R. glabra, and four putative hybrid populations at Ft. Pickett. Overall, R. michauxii had typical levels of within-population genetic variation when compared to other species with similar life-history characteristics. In contrast, the proportion of genetic variation among populations (G(ST)) was considerably lower than expected. Finally, R. michauxii and R. glabra appear to have a fixed allelic difference at the Idh2 locus. This enabled us to confirm hybridization in all four of the putative hybrid populations and one of the R. michauxii populations.  相似文献

Thirteen allozyme loci and 68 random amplified polymorphic DNA (RAPD) markers were analyzed to assess the genetic diversity and population structure of threatened Antirrhinum microphyllum (Scrophulariaceae), a narrow endemic of central Spain known from only four populations. According to allozyme data, species genetic diversity (p = 46.15%, A = 2.61, and H(e) = 0.218), as well as within-population genetic diversity (p = 44.23%, A = 2.10, and H(e) = 0.204), were high when compared to average estimates for other narrowly distributed plant species. Ninety-four percent of species genetic diversity corresponded to within-population genetic diversity. Nevertheless, significant differences were found among populations in allele frequencies of four of the six polymorphic loci, and three private alleles were detected. Inbreeding coefficients (F(IS)) suggest that populations are structured in genetic neighborhoods. The RAPDs also showed high levels of genetic diversity (p = 89.71% and H(e) = 0.188 at the species level, and p = 67.65% and H(e) = 0.171 at the population level). Nei's genetic distances estimated both from allozymes and RAPDs indicated low differentiation among populations. In spite of this, the low frequencies of certain alleles and the presence of private alleles indicate that efforts should be made to conserve all four remaining populations.  相似文献

The starfish Asterina pectinifera of the family Asterinidae is endemic Japanese species and commonly found in Japanese waters. In order to examine the degree of genetic variation and the maintenance mechanism of polymorphism within population, we studied the allozyme variation in five northern Japanese local populations of the starfish by electrophoresis. The species showed much higher genetic variability than many other shallow water echinoderms. Based on other allozyme studies and the ecological data, it was suggested that the high genetic variation of the starfish was closely related to the population size. Additionally, the relation between the degree of enzyme variation and the quaternary structure of enzymes was also examined, and the results suggested the close relation between the enzyme variability and functional constraints.  相似文献
