采用免疫电镜技术对多头绒泡菌(Physarum polycephalum)是否含有类CyclinA蛋白以及该蛋白在有丝分裂周期各时相的定位进行了研究;并以抗CyclinA抗体封闭细胞内源类CyclinA蛋白的方法,探讨类CyclinA蛋白在多头绒泡菌细胞周期中的作用。免疫电镜结果表明,经抗CyclinA抗体标记的实验组细胞中的金颗粒密度明显高于对照组,说明多头绒泡菌细胞中含有类CyclinA蛋白。实验组样品中,细胞核的金颗粒密度很高,而细胞质的金颗粒密度与对照组的相仿,说明多头绒泡菌细胞中的类CyclinA蛋白是核蛋白。细胞核的金颗粒密度在S期最高,G2期的次之,早中期时明显降低,中期和中期以后与对照组的相近。这种金颗粒密度的变化反映了类CyclinA蛋白在细胞周期中的含量变化。以抗CyclinA抗体分别处理S期和G2期的多头绒泡菌细胞,处理后的细胞分别停滞在原来的时相,细胞核形态变得不规则,核内有空洞现象。处于有丝分裂前期的多头绒泡菌细胞经抗CyclinA抗体处理后,细胞核出现畸变。抗体处理结果说明类CyclinA蛋白是参与多头绒泡菌细胞周期多个转换过程调控的种重要蛋白,主要在S期/G2期和G2期/M期的转换以及走出有丝分裂期的进程中发挥作用。  相似文献

By screening a collection of one hundred combinations of thiazolidinone compounds, we identified one combination (M4) that synergistically inhibited the growth of H460 and H460/TaxR cells and tumor growth in H460/TaxR xenograft mice. A whole genome microarray assay showed that genes involved in negative regulation of microtubule polymerization or depolymerization, intracellular protein kinase cascade, positive regulation of histone acetylation, cell cycle arrest and apoptosis were upregulated. Further analysis proved that the four compounds act as either microtubule polymerization inhibitors or histone deacetylase inhibitors. They act synergistically targeting multiple proteins and leading to the regulation of cell cycle checkpoint proteins, including p53, p21, cdc25C and cdc2, the activation of caspases, JNK, p38 cascades and the inactivation of Akt. These events resulted in the G2/M cell cycle arrest and cell apoptosis. These data provide a new strategy for discovering anticancer drugs and drug combinations for drug-resistant cancers.  相似文献

The goal of the present work is to establish a correlation between the degree of histone post‐translational modifications and the effects caused by treatment of HT29 colon cancer cells with class I‐selective (MS‐275 and MC1855), class II‐selective (MC1568), and non‐selective (suberoylanilide hydroxamic acid (SAHA) histone deacetylase inhibitors (HDACi). This correlation could afford a mean to better understand the mechanism of action of new, more potent, and selective HDACi directly on the cells. To this end, LC coupled to MS was applied in studies of time and concentration‐dependent treatment with HDACi in HT29 cells. The results were correlated to their potency of histone deacetylase inhibition and to their effects on the cell cycle. The results indicate that the four tested inhibitors show a different pattern of time‐ and concentration‐dependent modification after treatment of HT29 cells. At the selected concentrations, they cause different histone hyperacetylation and different cell cycle effects. In particular, SAHA (non‐selective HDACi) affected hyperacetylation of all histones and caused massive cell death. MC1855 (class I‐selective HDACi, hydroxamate) proved to be more potent and less toxic (cell arrest in G2/M phase) than SAHA. MS‐275 (class I‐selective HDACi, benzamide) exhibited a higher degree of hyperacetylation of H4 and a lower degree of H2A, H2B, and H3 acetylation, causing a cell arrest in G0/G1 phase. On the contrary, MC1568 (class II‐selective HDACi) produced only a modest hyperacetylation of H4, was ineffective on the other histones, and showed no effect on cell cycle in HT29 cells.  相似文献

用免疫印迹技术检测到低等真核生物多头绒泡菌中含有两种与HeLa细胞SC35单克隆抗体反应的蛋白,其分子量分别为32.5kD和82.5kD,将其命名为PSCL32.5和PSCL82.5。用SDS—PAGE技术对PSCL32.5蛋白进行了纯化,用等电聚焦方法确定PSCL32.5蛋白的等电点为6.19。应用免疫印迹技术检测多头绒泡菌细胞周期不同时相的PSCL32.5含量,发现该蛋白的含量在细胞周期中是变化的,在S早期时含量最低,从S期到G2期含量逐步增高,G2期后期含量最高。  相似文献

The mammalian homologue of the cdc2 gene of the fission yeast Schizosaccharomyces pombe encodes a p34cdc2 cyclin-dependent kinase that regulates the cell cycle of a wide variety of cell types. Resting murine T lymphocytes contained no detectable p34cdc2 protein, histone kinase activity, or specific mRNA for the cdc2 gene. Activation of the T cells by immobilized anti-CD3 resulted in the expression of specific mRNA late in the G1 phase of the cell cycle, and p34cdc2 protein was detectable at or near G1/S. At this point in the cell cycle, the protein was phosphorylated at tyrosine and displayed no H1 histone kinase activity. As the cells progressed through the cycle, the amount of specific mRNA and p34cdc2 increased, and H1 histone kinase activity was detectable when the cells were blocked at G2/M by nocodazole. The activation of T cells by phorbol dibutyrate induced the expression of IL-2R but failed to induce the synthesis of IL-2 or the expression of cdc2-specific mRNA. Under these conditions, the activated cells failed to enter the S phase of the cell cycle. Because the presence of IL-2 added exogenously during activation by phorbol dibutyrate resulted in the expression of cdc2-specific mRNA and progression through the cell cycle, either IL-2 or the interaction with IL-2R may be involved in the expression of cdc2 and regulation of the G1/S transition.  相似文献

CDKs play key roles in controlling cell cycle progression in all eukaryotes. In plants, multiple CDKs are present,among which the best characterized CDKs are PSTAIRE CDKs. In this study, we carried out Western blot,immunoelectron microscopy and antibody treatment with an anti-PSTAIRE monoclonal antibody to explore the subcellular localization and functions of PSTAIRE CDKs in Physarum polycephalum. The results of Western blot and immunoelectron microscopy showed that in P. polycephalum, a PSTAIRE CDK-like protein was 34 kD in molecular weight and located in both nuclei and cytoplasm. In nuclei, the protein was mainly associated with chromosomes and nucleoli. The expression of the PSTAIRE CDK-like protein in both the plasmodia and nuclei showed little fluctuation through the whole cell cycle. When treated with an anti-PSTAIRE monoclonal antibody at early S phase, the cells were arrested in S phase, and the mitotic onset of P. polycephalum was blocked for about 1 h when treated at early G2 phase.Our data indicated that the PSTAIRE CDK-like protein has a direct bearing on the mitosis.  相似文献

The histone deacetylase inhibitor and potential anti-cancer drug sodium butyrate is a general inducer of growth arrest, differentiation, and in certain cell types, apoptosis. In human CCRF-CEM, acute T lymphoblastic leukemia cells, butyrate, and other histone deacetylase inhibitors caused G2/M cell cycle arrest as well as apoptotic cell death. Forced G0/G1 arrest by tetracycline-regulated expression of transgenic p16/INK4A protected the cells from butyrate-induced cell death without affecting the extent of histone hyperacetylation, suggesting that the latter may be necessary, but not sufficient, for cell death induction. Nuclear apoptosis, but not G2/M arrest, was delayed but not prevented by the tripeptide broad-range caspase inhibitor benzyloxycarbonyl-Val-Ala-Asp.fluoromethylketone (zVAD) and, to a lesser extent, by the tetrapeptide 'effector caspase' inhibitors benzyloxycarbonyl-Asp-Glu-Val-Asp.fluoromethylketone (DEVD) and benzyloxycarbonyl-Val-Glu-Ile-Asp.fluoromethyl-ketone (VEID); however, the viral protein inhibitor of 'inducer caspases', crmA, had no effect. Bcl-2 overexpression partially protected stably transfected CCRF-CEM sublines from butyrate-induced apoptosis, but showed no effect on butyrate-induced growth inhibition, further distinguishing these two butyrate effects. c-myc, constitutively expressed in CCRF-CEM cells, was down-regulated by butyrate, but this was not causative for cell death. On the contrary, tetracycline-induced transgenic c-myc sensitized stably transfected CCRF-CEM derivatives to butyrate-induced cell death.  相似文献
