The year 1970 saw the publication of Origin of Eukaryotic Cells by Lynn Margulis. This influential book brought the exciting and weighty problems of cellular evolution to the scientific mainstream, simultaneously breaking new ground and ‘re-discovering’ the decades-old ideas of German and Russian biologists. In this commemorative review, I discuss the 40 years that have elapsed since this landmark publication, with a focus on the ‘molecular era’: how DNA sequencing and comparative genomics have proven beyond all doubt the central tenets of the endosymbiont hypothesis for the origin of mitochondria and plastids, and, at the same time, revealed a genetic and genomic complexity in modern-day eukaryotes that could not have been imagined in decades past.  相似文献

Over the past 25 years, countless experiments have been conducted on the impact of increased atmospheric CO2 concentration on various plants and ecosystems. While this research was motivated to better understand and predict how rising CO2 will affect the structure and function of ecosystems in the future, it also shed light on some general, CO2-research independent, aspects in ecological research. Interestingly, it is these general aspects that continue to create confusion and lead to misinterpretation. Here, we focus on seven interrelated key issues including (1) the confusion between fluxes and pools, (2) the stoichiometric aspects of growth and biomass production, (3) resource allocation within organisms, (4) data scaling and the choice of a reference metric, (5) the consideration of time and timing (experimental duration, ontogenetic shifts), (6) confounding and second-order (indirect or feedback) effects, and (7) the key role of biodiversity. The principles deriving from addressing these issues relate strongly to each other. Their concurrent consideration requires experimenters and modellers to likewise maintain a broad, holistic perspective. In this synthesis, we attempt to show how appropriate consideration of these principles can greatly enhance the assessment of the validity, plausibility and generality of experimental and modelling results. We conclude that neglecting to adequately address these key issues in ecological research may lead to overestimations of measured responses and/or simplistic interpretations. Our examples mostly originate from research on plant responses to elevated atmospheric CO2, but are also applicable to other areas of ecological research. We provide a checklist for the planning of ecological field experiments and the interpretation of their results that may help in avoiding common pitfalls.  相似文献

Tissue Banking and Biomaterial Research Unit (TBBRU), the only tissue bank of Bangladesh, has been established to create an available supply of human tissue allografts for transplantation in Bangladesh. Since its establishment in 2003, TBBRU strictly follows the guidelines of tissue banking setup by the International Atomic Energy Agency, the European Association of Tissue Banks and the American Association of Tissue Banks. Though started serving from earlier, regular supply of tissue allografts from this bank were documented at the end of 2006. From January 2007 to December 2014, 3747 bones and 5772 amniotic sacs were collected from live tissue donors. During this period, 59,489 cc bone allografts and 23,472 pieces of amniotic membrane allografts were processed. In the same period, 58,483 cc bone allografts and 20,786 pieces membrane were supplied to different hospitals throughout the country on the basis of demand. The outcomes of the concerted efforts of tissue banking professionals and physicians were the restoration of health and hope of 3662 patients during the last 8 years.  相似文献

Alzheimer’s disease (AD) can be divided into sporadic AD (SAD) and familial AD (FAD). Most AD cases are sporadic and result from multiple etiologic factors, including environmental, genetic, and metabolic factors, whereas FAD is caused by mutations in the presenilins or amyloid-β (Aβ) precursor protein (APP) genes. A commonly used animal model for AD is the 3xTg-AD transgenic mouse model, which harbors mutated presenilin 1, APP, and tau genes and thus represents a model of FAD. There is an unmet need in the field to characterize animal models representing different AD mechanisms, so that potential drugs for SAD can be evaluated preclinically in these animal models. A mouse model generated by intracerebroventricular (icv) administration of streptozocin (STZ), the icv-STZ mouse, shows many aspects of SAD. In this study, we compared the non-cognitive and cognitive behaviors as well as biochemical and immunohistochemical alterations between the icv-STZ mouse and the 3xTg-AD mouse. We found that both mouse models showed increased exploratory activity as well as impaired learning and spatial memory. Both models also demonstrated neuroinflammation, altered synaptic proteins and insulin/IGF-1 (insulin-like growth factor-1) signaling, and increased hyperphosphorylated tau in the brain. The most prominent brain abnormality in the icv-STZ mouse was neuroinflammation, and in the 3xTg-AD mouse it was elevation of hyperphosphorylated tau. These observations demonstrate the behavioral and neuropathological similarities and differences between the icv-STZ mouse and the 3xTg-AD mouse models and will help guide future studies using these two mouse models for the development of AD drugs.  相似文献

Many studies have shown that natural marine compounds can prevent biofouling by a broad spectrum of organisms without toxic effects, encouraging their use in antifouling (AF) coatings. Studies over the past 25 years of the natural product zosteric acid (ZA) are systematically organized in this review. ZA is a sulfated phenolic acid produced by the seagrass Zostera marina that has very promising AF potential against several micro- and macrofouling organisms. ZA was shown to have appropriate environmental fate parameters such as high water solubility, a low log P, low bioaccumulation, and no ecotoxicity, which demonstrated the potential of ZA as a safe AF agent. This review also highlights that ZA has been successfully incorporated into several types of coatings. The synthesis of analogs is also considered in this review, and it has allowed a better understanding of ZA structure–AF activity relationships and clarified the mechanism of action of ZA.  相似文献

Systematic genome-wide scans to date have shown that genes of major effect are not common causes of schizophrenia, but independent linkage studies looking for schizophrenia susceptibility genes are converging on a number of key chromosomal locations. The methanolic extract of pasuchaca (Geranium dielsiaum) (PsEx) was found to suppress blood glucose elevation after oral administration of sucrose, maltose, and starch, but not after oral administration of glucose, in the mouse. In vitro examination of the inhibitory effect of PsEx on maltase activity revealed that PsEx strongly inhibited mouse small intestine maltase activity. Taken together, these results suggest that the inhibitory effect of PsEx on α-glucosidase activity might contribute to delay in carbohydrate digestion and subsequent lowering of the blood glucose level, thereby leading to prevention and cure of diabetes.  相似文献

Propagation of the cycad Dioon edule aimed at sustainable management in the state of Veracruz, Mexico has been ongoing since 1990 under the supervision of staff at Francisco Javier Botanic Garden, with the principal objectives of addressing illegal trafficking and habitat destruction. Plant sales have been limited. Nevertheless these, along with the sale of other managed forest products, have given the cycad producers and other villagers enough incentive to conserve 80 hectares of cycad habitat and to discourage illegal collecting. This model was taken up by four similar nurseries in the buffer zones of two biosphere reserves in Chiapas for the propagation of four additional cycad species and two endangered Chamaedorea palms. A further biosphere reserve in Puebla hosts a similar nursery for the critically endangered D. caputoi. Here the producers are paid through the Reserve authority for cultivation and reintroduction of the cycad. All species were studied at the population level prior to and during nursery establishment. Cultivation knowledge has been passed on to the farmers as well as limited help in marketing. Seedling reintroduction experiments have been carried out but further demographic studies of D. edule and C. mirandae have given reason to re-think reintroduction strategies. There is great potential for these nurseries to act as shelter for rescued plants during civil engineering projects. The marketing problem is still an issue and has been approached by the involvement of conservation authorities in Chiapas to assist the producers with permit paperwork and to seek markets. This experience is an important example of botanic garden extension to rural communities in Mexico that covers several articles of the Convention on Biological Diversity.  相似文献

The bird-lemming hypothesis postulates that breeding success of tundra-nesting geese and waders in Siberia follows the cyclic pattern of lemming populations, as a result of predators switching from lemmings to birds when the lemming population crashes. We present 50 years of data on constant-effort catches of red knot Calidris canutus and curlew sandpiper C. ferruginea at an autumn migratory stopover site (Ottenby) at the Baltic Sea, supplemented with literature data on winter censuses of dark-bellied brent goose Branta b. bernicla and white-fronted goose Anser albifrons in northwestern Europe, and waders in Germany and Southern Africa. Number and proportion of juveniles in these bird populations (both our own and literature data) were compared with an index of predation pressure (calculated from the abundance of lemmings on the Taimyr peninsula), and climate indices for the North Eurasia and the North Atlantic regions. The index of predation pressure correlated significantly with the number of juveniles of red knot and curlew sandpiper, but not with number of adults. Also, this index correlated with the reproductive performance of geese and waders reported in the literature. Fourier analysis revealed a significant deviation from random noise with the maximum spectral density at the period length of 3 years for number of juvenile red knots and curlew sandpipers captured at Ottenby, abundance of lemmings, reproduction in arctic fox Alopex lagopus, and reproductive performance in geese on the Siberian tundra. Also, the date of passage at Ottenby for adult red knot and curlew sandpiper showed a spectral density peak at a period length of 3 years, the latter species also showing a peak at a period length of 5-6 years. Passage dates for adult red knot and curlew sandpiper were earlier in years of high predation pressure compared with years of low predation pressure. The fluctuations in reproductive success of the studied Siberian goose and wader species appear to be primarily influenced by biotic factors in the breeding area, rather than by abiotic factors, such as climate oscillations. Annual variations in migratory arctic bird populations may have far reaching effects in habitats along their migration routes and in their wintering areas. We suggest a link between lemming cyclicity in the Northern Hemisphere and predation pressure on Southern Hemisphere benthos, in which the signal is carried between continents by long distance migrating waders.  相似文献
