一株木霉菌挥发性物质的抑菌活性测定及成分分析   总被引:2,自引:0,他引:2  
目的:测定木霉菌XYT-12菌株的挥发性物质对植物病原真菌的抑制活性,并分析其挥发性物质的化学成分。方法:抑菌活性测定采用对扣法,化学成分分析采用气相色谱-质谱联用法(GC-MS)。结果:木霉菌XYT-12的挥发性物质对植物病原真菌有一定的抑制活性,以对番茄灰霉病菌的抑制作用最强,达到61.78%;木霉菌挥发性物质的主要成分为6-戊基-2H-吡喃-2-酮(6PP),在菌丝和PDB培养液挥发物中的相对含量分别为90.346%和88.729%。  相似文献

用水蒸气蒸馏法从曼陀罗中提取挥发油,并用气相色谱/质谱联用技术(GC-MS)对挥发油的化学成分进行分离鉴定,用气相色谱峰面积归一化法确定各组分的相对含量.结果从曼陀罗挥发油中鉴定出58种化合物,占总挥发油量的92.37%.其中主要成分为5,6-二氢-6-戊基-2H-吡喃-2-酮(44.29%)、二苯胺(12.50%)、四十四烷(10.41%)、二十烷(4.19%)、(E)-3-己烯-1-醇(2.38%)、3,7,11,15-四甲基-2-十六碳烯-1-醇(2.28%)等.  相似文献

【背景】由病原菌Sclerotium denigrans侵染引起的平贝母菌核病是其主要的鳞茎病害之一,给平贝母种植产业带来了巨大的损失。【目的】筛选出对平贝母菌核病具有拮抗效果的木霉菌株。【方法】以平贝母菌核病作为靶标菌,采用平板对峙试验、平板对扣法、圆盘滤膜法与发酵液抑菌试验筛选对平贝母菌核病具有拮抗效果的木霉菌株。采用顶空固相微萃取的方法检测拮抗效果较好的木霉菌挥发性成分;二硝基水杨酸(dinitrosalicylic acid, DNS)比色法测定木霉菌的β-1,3葡聚糖酶的活性;室内防效试验验证其对平贝母菌核病的防治效果。【结果】平板对峙试验发现木霉菌F1、F2和D6对平贝母菌核病菌的生长具有较强的抑制作用,其抑菌率分别为91.06%、87.00%和86.12%;平板对扣法发现木霉菌E17和A26对菌核病菌的抑制效果最为明显,抑菌率分别为74.96%和75.86%;圆盘滤膜法发现菌核病菌在F2、C6、D3、F4、A26、B30、D4和D6的琼脂培养基上均不生长,抑菌率达100%;发酵液抑菌试验表明木霉菌D3抑制效果最强,可完全抑制菌核病菌的生长,抑菌率为100%;对A26、D4、E8、E17和D3这5株木霉进行GC/MS挥发性产物分析,在E17发现了具有抗真菌活性的6-戊基-2H-吡喃-2-酮等活性物质;DNS比色法发现β-1,3葡聚糖酶活性最高的木霉菌为F1;室内防效试验测定发现D3能明显抑制平贝母鳞茎菌核病的病变,对平贝母菌核病具有潜在的生防活性。【结论】木霉菌D3在防治平贝母菌核病中是极具开发价值的菌种。  相似文献

木霉是一类具有重要生防价值的丝状真菌。文中首先对分离自浙江省绍兴市和广东省佛山市共12株棘孢木霉Trichoderma asperellum进行平板拮抗评价,然后采用顶空固相微萃取气质联用法(HS-SPME-GC-MS)检测拮抗性较好的两株菌的挥发性次级代谢产物。结果表明,棘孢木霉ZJSX5003和GDFS1009菌丝生长迅速,对尖孢镰孢菌Fusariumoxysporum抑制率分别达73%和74%。挥发性次级代谢产物主要是醇类和酮类,其中包含异丁醇、异戊醇、3-甲基-3-丁烯-1-醇、3-羟基-2-丁酮、2,3-丁二醇和6-正戊基-2H-吡喃-2-酮(6-PAP)。进一步通过体外抑菌试验,证实6-PAP具有较好的抑制尖孢镰孢菌的效果,为开发以木霉菌代谢产物如6-PAP为主要成分的生防制剂提供指导。  相似文献

从胶陀螺中分离得到麦角甾醇、丙二酸和葡萄糖3种单体化合物;6-戊基-4羟基-2-一,四氢化-2氢-吡喃(2H-Pyran-2-one,tetrahydro-4-hydroxy-6-pentyl)和6-戊基-2-,5,6-二氢化-2氢-吡喃(2H-Pyran-2-one,5,6-dihydro-6-pentyl)2种主要香气成分;Bulgarhodin(I)和Bulgarein(II)2种色素混合物。该菌中还含有碳原子数为13、14、15、16、17、20、21、36和44的一组饱和烃类化合物。  相似文献

目的:用GC-MS法对柽柳花乙醇提取物的化学成分进行分析。方法:采用微波提取法从柽柳花中提取化学物质,用GC-MS法对化学成分进行鉴定。结果:鉴定出了48种化合物,所鉴定化合物的含量占提取物的87.23%。结论:柽柳花乙醇提取物的主要化学成分为邻苯二甲酸二(2-甲氧基丙基)酯(24.94%),邻苯二甲酸二丁酯(15.65%),5-羟甲基-2-呋喃甲醛(8.63%),十六碳酸(6.65%),邻苯二甲酸-6-乙基-3-辛烷基丁基酯(3.67%),三十六烷(1.85%),3,6-二氧代-1-甲基-8-异丙基三环[6.2.2.0(2.7)]十二烷-4,6-二烯(1.78%),2-呋喃甲醛(1.46%),2,3-二氢-6-甲基-3,5-二羟基-4H-吡喃-4-酮(1.39%),二十七烷(1.30%),O-苯基甲基-L-丝氨酸(1.29%),2,3-二氢苯并呋喃(1.23%),二十一烷(1.21%),二十四烷(1.11%),1,1-二乙氧基己烷(1.03%),以上十五种化合物占总提取物的73.22%。  相似文献

为了探究新疆植物资源的药用活性,本研究通过菌丝生长速率测定法,以尖孢镰刀菌、立枯丝核菌和拟轮枝镰刀菌等3种棉花致病菌为靶标菌,对21种新疆药用植物提取物的抑菌活性进行筛选与测定,然后采用超高效液相色谱串联质谱(UPLC-MS/MS),以及相关数据库和文献对曼陀罗乙醇提取物质进行分析鉴定。结果表明,21种植物提取物中曼陀罗对3种棉花病原菌均具有较好的抑菌活性效果,并通过液质联用共鉴定曼陀罗乙醇提取物21个化学成分,主要包括10个生物碱类、4个脂质类、2个酚酸类、2个有机酸类和其他类化合物,其中生物碱类化合物为主要成分占总的47.62%,生物碱类物质因具有显著抑菌活性,可能是曼陀罗类物质的主要抑菌活性成分。  相似文献

以棘托竹荪(Dictyophora echinovolvata)菌托为研究对象,提纯菌托提取物有效抑菌天然产物,分析其抑菌活性组成。选择最佳提取溶剂及指示菌,采用色谱层析法分离纯化,GC-MS检测分析活性组分。结果表明:乙酸乙酯提取物对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、蜡状芽孢杆菌、标准摩根氏菌、肠炎沙门氏菌均有较好的抑制效果;以金色葡萄球菌为指示菌,乙酸乙酯提取物的抑菌作用对温度、紫外线均有较好的稳定性;对提取物进行分离,获得2个有抑菌活性组分DEVⅠ、DEVⅡ,采用GC-MS分析DEVⅠ、DEVⅡ中的化学组分,DEVⅠ中相对含量1%以上的成分有20个;DEVⅡ中相对含量在1%以上成分6个,其中具有抑菌活性的成分为2-呋喃甲酸和肉桂酸,相对含量分别为25.6%和54.2%,推测可能为棘托竹荪菌托抑菌物质的主要组成成分。  相似文献

人参(Panax ginseng)根系分泌物成分对人参致病菌的化感效应   总被引:10,自引:1,他引:10  
采用室内培养结合生物学测定的试验方法,研究了不同浓度人参(Panax ginseng)根系分泌物成分苯甲酸、邻苯二甲酸二异丁酯、十六酸和2,2-二(4-羟苯基)丙烷对人参立枯丝核菌(Rhizoctonia solani)、黑斑菌(Alternaria panax)、疫病菌(Phytophthora cactorum)、菌核菌(Sclerotinia schinseng)、锈腐菌(Cylindrocarpon destructans)和绿色木霉菌(Trichoderma viride)菌落生长及孢子萌发的化感效应.结果显示,不同浓度人参根系分泌物成分对人参致病菌及绿色木霉菌的化感效应存在显著差异.苯甲酸浓度与人参立枯丝核菌、菌核菌和锈腐菌菌落生长以及人参黑斑菌、锈腐菌孢子萌发呈负相关,与人参黑斑菌、绿色木霉菌菌落生长呈正相关;对人参疫病菌菌落生长的化感效应表现为低浓度和高浓度抑制,中浓度促进.邻苯二甲酸二异丁酯浓度与人参立枯丝核菌、黑斑菌、菌核菌和绿色木霉菌菌落生长以及人参黑斑菌孢子萌发呈负相关;对人参锈腐菌菌落生长和孢子萌发表现为低浓度和高浓度抑制,中浓度促进;对人参疫病菌菌落生长表现为低浓度和中浓度抑制,高浓度促进.2,2-二(4-羟苯基)丙烷浓度与人参立枯丝核菌、黑斑菌、疫病菌、绿色木霉菌菌落生长以及人参黑斑菌、锈腐菌孢子萌发呈负相关;对人参菌核菌、锈腐菌菌落生长表现中浓度促进,高浓度抑制.十六酸浓度与人参锈腐菌、疫病菌和绿色木霉菌菌落生长呈正相关,与人参锈腐菌孢子萌发呈负相关,对黑斑菌孢子萌发表现为中浓度抑制.4种根系分泌物的等量混合物浓度与人参致病菌及拮抗木霉菌菌落生长速率呈负相关.  相似文献

内芽孢杆菌属细菌(Paenibacillus daejeoneusis)SS02,是一株能强烈抑制玉米纹枯菌(Rhizoctonia cerealis)、白色念珠菌(Candidal vaginitis)等病害真菌的拮抗菌株。SS02菌株培养液经硫酸铵分级盐析,SephadexG-75凝胶柱层析和DEAE-32纤维素柱层析分离纯化出一种抗菌蛋白,命名为SD22。蛋白电泳分析结果表明,此蛋白分子量为56000,等电点为6.4。此蛋白对热和紫外线稳定,对蛋白酶部分敏感。纯化后的SD22对玉米纹枯菌(Rhizoctonia cerealis)、油菜菌核病(Sclerotinia sclerotiorum)、苹果轮纹病菌(Physalospora piricala)、绿色木霉(Triclwdema viride)、绿色粘帚霉(Gliocladium viride)、弯孢霉菌(Curvula rialeafspot)、镰刀霉菌(Fusarium sp.)、赤霉菌(Fusarium headblight)、球孢白僵菌(Beauveria bassiana)、大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylo-coccus aureus)、枯草芽孢杆菌(Bacillus subtilis)、白色念珠菌(Candidal vaginitis)、冬瓜枯萎菌(Fusarium oxysporum Schl.emend,Snyder&Hause)等菌有很强的抑制作用。  相似文献

制备了欧美107杨树枝条和叶的乙醇粗提物及不同极性溶剂的萃取部分,并测定了它们对植物病原真菌的抑制活性。枝条和叶的乙醇粗提物对棉花枯萎病菌、小麦纹枯病菌、番茄早疫病菌、番茄枯萎病菌、黄瓜枯萎病菌、小麦赤霉以及玉米弯孢等7种植物病原真菌均具有一定的抑制作用。而枝条的乙醇粗提物对杨树溃疡病菌的菌丝生长有一定的促进作用。抗菌活性成分主要集中在乙酸乙酯萃取部分。  相似文献

Endophytic mycopopulation isolated from India’s Queen of herbs Tulsi (Ocimum sanctum) were explored and investigated for their diversity and antiphytopathogenic activity against widespread plant pathogens Botrytis cinerea, Sclerotinia sclerotiorum, Rhizoctonia solani and Fusarium oxysporum. 90 fungal isolates, representing 17 genera were recovered from 313 disease-free and surface sterilised plant segments (leaf and stem tissues) from three different geographic locations (Delhi, Hyderabad and Mukteshwar) during distinct sampling times in consequent years 2010 and 2011 in India. Fungal endophytes were subjected to molecular identification based on rDNA ITS sequence analysis. Plant pathogens such as F. verticillioides, B. maydis, C. coarctatum, R. bataticola, Hypoxylon sp., Diaporthe phaseolorum, Alternaria tenuissima and A. alternata have occurred as endophyte only during second sampling (second sampling in 2011) in the present study. Bi-plot generated by principal component analysis suggested tissue specificity of certain fungal endophytes. Dendrogram revealed species abundance as a function of mean temperature of the location at the time of sampling. Shannon diversity in the first collection is highest in Hyderabad leaf tissues (H'' = 1.907) whereas in second collection it was highest from leaf tissues of Delhi (H'' = 1.846). Mukteshwar (altitude: 7500 feet) reported least isolation rate in second collection. Nearly 23% of the total fungal isolates were considered as potent biocontrol agent. Hexane extract of M. phaseolina recovered from Hyderabad in first collection demonstrated highest activity against S. sclerotiorum with IC50 value of 0.38 mg/ml. Additionally, its components 2H-pyran-2-one, 5,6-dihydro-6-pentyl and palmitic acid, methyl ester as reported by GC-MS Chromatogram upon evaluation for their antiphytopathogenic activity exhibited IC50 value of 1.002 and 0.662 against respectively S. sclerotiorum indicating their significant role in antiphytopathogenic activity of hexane extract. The production of 2H-pyran-2-one, 5,6-dihydro-6-pentyl from M. phaseolina, an endophytic fungus is being reported for the first time.  相似文献

The leaves and bark dichloromethane extracts of Ravensara anisata showed antifungal activity against the yeast Candida albicans and the phytopathogenic fungus Cladosporium cucumerinum in bioautographic TLC assays. Activity-guided fractionation afforded two new alpha-pyrones: 6R*-(4R*-acetoxy-2S*-hydroxy- 8-phenyloctyl)-5,6-dihydro-2-H-pyran-2-one and 6R*-(2S*-acetoxy-4R*- hydroxy-8-phenyloctyl)-5,6-dihydro-2-H-pyran-2-one. Their structures have been established by NMR spectroscopy, chemical methods and X-ray crystallographic analysis. The antifungal activity against C. albicans and C. cucumerinum was determined for both compounds.  相似文献

绿僵菌对小麦纹枯病菌的抑制作用研究   总被引:1,自引:0,他引:1  
在实验室条件下,研究了金龟子绿僵菌(Metarhizium anisopliae)对小麦纹枯病菌(Rhizoctonia cerealis)的拮抗作用及其机理。结果表明,金龟子绿僵菌与小麦纹枯病菌对峙培养以及在培养基中加入金龟子绿僵菌孢子悬浮液,对小麦纹枯病菌菌丝生长均有较好的抑制作用。测定了培养不同天数的金龟子绿僵菌Ma55发酵液对小麦纹枯病菌菌丝生长、菌核产生量及菌核萌发率的影响。结果表明,液体振荡培养25 d的金龟子绿僵菌Ma55发酵液对小麦纹枯病菌的菌丝生长、菌核产生量及菌核萌发具有显著的抑制作用,且Ma55发酵液中的抑菌活性物质具有较好的热稳定性。在光学显微镜下,未观察到Ma55对小麦纹枯病菌的重寄生现象,但发现金龟子绿僵菌与小麦纹枯病菌对峙培养处小麦纹枯病菌营养菌丝的细胞质变稀薄、菌丝部分消解或断裂。上述结果显示,金龟子绿僵菌对小麦纹枯病菌的拮抗机制主要是营养竞争、空间竞争及抗生作用等。  相似文献

All Basidiomycotina screened were sensitive to validamycin A, whereas most Ascomycotina and all Mucorales and Oomycetes were insensitive. Studies with Rhizoctonia cerealis and Fusarium culmorum showed that, in semi-solid culture, the antibiotic caused a decrease in colony radial growth rate and that this was associated with a decrease in mean hyphal extension rate and an increase in hyphal branching. However, the antibiotic did not alter the morphology of R. cerealis grown in liquid culture (shaken or stationary). Validamycin A caused a reduction in the number and viability of conidia produced by F. culmorum.  相似文献

Growth responses of some soil fungi to spatially heterogeneous nutrients   总被引:1,自引:0,他引:1  
Abstract The natural nutritional environments of most fungi are spatially non-uniform, yet the majority of studies of fungal growth take no account of this fact. An experimental system is described which permits the growth responses of eucarpic fungi to heterogeneously distributed nutrient resources to be studied. The system comprises tesselations of agar tiles of contrasting nutrient status separated by air gaps. Growth responses in such systems of Alternaria alternata, Mucor sp., Phoma foveata , Rhizoctonia solani and Trichoderma viride are described. Generally, the growth of the fungi reflected the nutrient status of the underlying substrate. There was evidence for growth in low-nutrient tiles being greater when high-nutrient tiles were included in the tessellation. Reproductive structures tended to be formed only in low nutrient tiles with Trichoderma and Rhizoctonia and only high nutrient tiles with Alternaria . Growth responses of Rhizoctonia were strongly asymmetric in nutritionally symmetric, but heterogeneous, tesselations. The consequences of the observations for fungal growth in heterogeneous environments such as soil is discussed.  相似文献

Ethyl acetate extracts of two strains of Trichoderma koningii were evaluated for antifungal activity against soilborne pathogenic fungi, Rhizoctonia solani, Sclerotium rolfsii, Macrophomina phaseolina and Fusarium oxysporum by poisoned food technique. Secondary metabolites, namely δ-decanolactone, 6-pentyl-α-pyranone and 6-(4-oxopentyl)-2H-pyran-2-one were isolated from T. koningii (T-8) extract while palmitic acid, 6-pentyl-α-pyranone and stigmasterol were isolated from T. koningii (T-11) extract. Secondary metabolites were identified by 1H NMR, 13C NMR and mass spectroscopic methods. These metabolites were evaluated for antifungal activity against the test pathogens 6-Pentyl-α-pyranone and 6-(4-oxopentyl)-2H-pyran-2-one exhibited excellent antifungal activity against S. rolfsii. The antifungal activity though slightly less was comparable to that of hexaconazole, a commercial fungicide.  相似文献
