Three different allelic isozyme systems (two esterases, ESH and ESR, and a malic dehydrogenase, MDH) were analyzed in population samples of a species of ant, Pogonomyrmex barbatus, from Texas. Allelic frequencies were determined for several collection localities, and a number of significant differences were found. Principal component analysis was used to compare the patterns of variability of the allelic frequencies with environmental factors. Significant correlation was particularly evident with respect to weather and the pattern of variability in both esterases, and it is therefore suspected that natural selection is important in determining the allele frequency patterns. Observed and expected genotypic proportions were found in good agreement, generally, but in some localities homozygotes appeared in significantly greater numbers than expected. Heterotic selective maintenance was thus not indicated. Correlation found between patterns of variability in the enzyme systems themselves was consistent with the hypothesis that all three enzyme systems were affected by the environmental factors.Supported in part by USPHS Research Grants No. GM 15769, GM 11609, and GM 11546 and Texas A & I University Faculty Research Grant No. 449-N-68. Adult Drosophila melanogaster flies collected from populations broadly dispersed over ecological and geographic strata of North Carolina, and over a period of 4 years, were analyzed for alcohol dehydrogenase phenotypes by gel electrophoresis. Gene frequencies in spring-summer-fall field collections were remarkably stable over all strata. Two winter collections exhibited contrasting gene frequency changes. Fucus vesiculosus L. is one of the most widespread macrophytes in the northwestern Atlantic, ranging from North Carolina (USA) to Greenland (DK). We investigated genetic diversity, population differentiation, patterns of isolation by distance, and putative glacial refugial populations across seven locations from North Carolina (USA) to Cape Breton Island, Nova Scotia (Canada), with microsatellite analyses. Distinct northern versus southern (Delaware–North Carolina) populations were revealed by microsatellite data. Five of six microsatellite loci were fixed in populations in North Carolina, suggesting a recent founder event or a bottleneck, and the same homozygous genotype was found in herbarium materials collected on the North Carolina coast from more than 60 years ago. An additional set of individuals from the northern limit in Greenland was included in our analysis of mitochondrial intergenic spacer (mt IGS) haplotypes in the northwestern Atlantic. Remarkably, 184 of 188 F. vesiculosus specimens from North Carolina to Greenland shared the same haplotype. Recent colonization of the North American shore from Europe is hypothesized based upon the ubiquity of this common haplotype, which was earlier reported from Europe.  相似文献

Gene flow, in combination with selection and drift, determines levels of differentiation among local populations. In this study we estimate gene flow in a stream dwelling, flightless waterstrider, Aquarius remigis. Twenty-eight Aquarius remigis populations from Quebec, Ontario, New Brunswick, Iowa, North Carolina, and California were genetically characterized at 15 loci using starch gel electrophoresis. Sampling over two years was designed for a hierarchical analysis of population structure incorporating variation among sites within streams, streams within watersheds, watersheds within regions, and regions within North America. Hierarchical F statistics indicated that only sites within streams maintained enough gene flow to prevent differentiation through drift (Nm = 27.5). Above the level of sites within streams gene flow is highly restricted (Nm ≤ 0.5) and no correlation is found between genetic and geographic distances. This agrees well with direct estimates of gene flow based on mark and recapture data, yielding an Ne of approximately 170 individuals. Previous assignment of subspecific status to Californian A. remigis is not supported by genetic distances between those populations and other populations in North America. Previous suggestion of specific status for south-eastern A. remigis is supported by genetic distances between North Carolina populations and other populations in North America, and a high proportion of region specific alleles in the North Carolina populations. However, because of the high degree of morphological and genetic variability throughout the range of this species, the assignment of specific or subspecific status to parts of the range may be premature.  相似文献

There have been few investigations of the number of founding sources and amount of genetic variability that lead to a successful nonindigenous species invasion, although genetic diversity is believed to play a central role. In the present study, population genetic structure, diversity and divergence patterns were analysed for the zebra mussel Dreissena polymorpha [n=280 samples and 63 putative randomly amplified polymorphic DNA (RAPDs) gene loci] and the quagga mussel D. bugensis (n=136 and 52 loci) from 10 nonindigenous North American and six Eurasian sampling sites, representing their present‐day ranges. Results showed that exotic populations of zebra and quagga mussels had surprisingly high genetic variability, similar to those in the Eurasian populations, suggesting large numbers of founding individuals and consistent with the hypothesis of multiple colonizations. Patterns of genetic relationships indicate that the North American populations of D. polymorpha likely were founded by multiple source populations from north‐western and northcentral Europe, but not from southcentral or eastern Europe. Sampling areas within North America also were significantly divergent, having levels of gene flow and migration about twice those separating long‐established Eurasian populations. Samples of D. bugensis in Lakes Erie and Ontario were significantly different, with the former being more closely related to a native population from the Dnieper River, Ukraine. No evidence for a founder effect was discerned for either species.  相似文献

Morphological and genetic variation were studied in Plantago cordata Lam., an imperiled aquatic plant. The existence of distinct seasonal morphs was confirmed by numerical morphological analyses of specimens from 53 populations. Plants from two North Carolina populations possessed leaves that were significantly shorter, narrower, and fewer-veined than populations in other portions of the species range. Genetic variation within and among ten populations in seven states/provinces (Illinois, Missouri, New York, North Carolina, Ohio, Ontario, and Wisconsin) was examined electrophoretically. The species is an apparent allopolyploid with fixed heterozygosity observed at 67% of the loci surveyed. Electrophoretic variation was mostly partitioned among populations. The genetic identity among populations was high except for North Carolina populations which differed at 27% of the loci surveyed.  相似文献

Genetic characteristics of sei whales, Balaenoptera borealis, inhabiting the western North Pacific were analyzed at 17 microsatellite loci in a total of 89 whales obtained from the area between 37°N–45°N and 147°E–166°E in 2002 (N = 39) and 2003 (N = 50). All the loci analyzed were polymorphic over the samples, some of the loci had more than 10 alleles, indicating a high level of genetic variation within samples. No significant deviation from the expected Hardy–Weinberg genotypic proportion was observed at the 17 loci in the samples. No evidence of genetic heterogeneity in allele frequencies was observed between sexes within samples as well as between the two temporally different samples, indicating a single population of sei whales inhabiting the western North Pacific. We finally tested and demonstrated that the population appeared not to suffer from genetic bottleneck as a result of population decline from past commercial whaling.  相似文献

Summary Variation at polymorphic isozyme loci was analyzed in Nantucket pine tip moth (NPTM) populations from 5 geographic locations. At the North Carolina location, populations representing 3 generations at 3 local sites were also studied. Four of the loci investigated (LAP, MDH, -GPDH and AK), although variable, had few alleles per locus (3–5) and few differences among populations in allele frequencies. At each locus, all populations had the same allele at a high frequency.At the PGM locus, fifteen alleles were identified and allelic frequencies varied among populations. At least eight alleles were present within a population and, in most populations, two or more alleles had high frequencies that differed among populations. An excess of homozygotes over Hardy-Weinberg expectations was found for 7 out of the 10 populations studied, indicating the probable existence of some form of inbreeding structure or populational subdivision within sampled stands.Joint consideration of the results observed for PGM and the other four loci is counterindicative of neutrality at all loci and strongly indicative of genetic differentiation among locally disjunct populations.Published as Paper No. 6751 of the Journal Series of the North Carolina Agricultural Research Service  相似文献

A survey of allozymic alleles and genetic diversity was made for 151 accessions of the American Barley Core Collection. A total of 25 alleles at ten loci were observed. Two loci were monomorphic. The average diversity index for individual loci ranged between 0.026 and 0.649. Most significant differences in allelic frequency and genetic diversity value were found between spring and winter barley. Spring barley showed a greatly higher average diversity than winter barley (t=2.124, P=0.071). The smallest differences in allelic frequencies and diversity values were observed between the two geographical regions, North and South America. Rare alleles were detected only in a few accessions. Seven rare alleles were associated with spring barley. The genetic similarities among the 151 accessions ranged from 0.20 to 1.00, which showed that a high level of genetic variability exists in this set of core accessions. Cluster analysis and principal coordinate analysis did not give clear-cut separation of different types of barley, but most of the winter barley accessions were closely associated. Genetic variation in North Amerindian populations: covariance with climate   总被引:1,自引:0,他引:1  
Allelic frequencies at seven polymorphic loci in 74 North Amerindian populations are examined relative to patterns of climatic variation. Canonical correlation analysis reveals strong and significant associations of heterozygosity at the ABO, Ss, Duffy, and P loci with climatic variability. Principal component analysis demonstrates that these loci tend to form correlated ensembles. Moreover, canonical correlation analysis of component scores provides support for an association between polymorphism at these loci and environmental variability. The results are concordant with two previous investigations which suggested a relationship between polymorphism for the ABO, Duffy, and Diego systems and climate. It is suggested that the examination of broad geographic patterns of genetic variation at multiple loci is a valuable, but underutilized, method of screening for the effects of long-term systematic pressures.  相似文献

Remarkable geographic and seasonal uniformities in gene and genotype frequencies were found for four enzyme loci. Principal component analysis revealed two patterns of allele frequency changes in three of the allozyme loci (Est-b, Est-c and Acph) and one pattern in the Mdh-d locus. These patterns accounted for 90% to 100% of the variability at these loci. Significant positive correlations were obtained between several of the components and the variability in the non-saponifiable and alkaloid fractions of the senita cactus host plant. The genetic variance within a single locality (Guaymas, Mexico) was as great as the variance between all localities sampled. This is interpreted as a local founder effect. The temporary nature of the rotting cactus breeding site, the delayed maturation and the differential maturation rate of females and males are three factors proposed to cause forced outbreeding and counter the local founder effect. A combination of selection resulting from a direct interaction of the genotype and the environment, in addition to migration, is proposed to maintain the uniform gene frequencies.  相似文献

Genetic variability was examined in nine sexual and three apogamous natural populations of the homosporous fern Pellaea andromedifolia by electrophoretic analysis of enzymes specified by eight loci. Genetic interpretations of heterozygous banding patterns were determined by segregational analysis of gametophytes. High levels of segregating heterozygosity characterized the sexually reproducing populations, and genotype frequencies at the five polymorphic loci were consistent with those expected under conditions of random intergametophytic mating. Multiple-banded patterns in the apogamously reproducing populations resembled those of heterozygous sexual individuals, but did not segregate. The results suggest that genetic variation in sexual homosporous vascular plants is produced by cross fertilization of genetically different gametes and may not result from pairing between homoeologous chromosomes carrying duplicated loci as previously thought.  相似文献

Adult Drosophila melanogaster from naturally occurring populations in the Eastern United States were examined by gel electrophoresis for their alcohol dehydrogenase (ADH) phenotype. The ADH enzymes were partially purified and characterized. Frequencies of the controlling alleles, Adh 4 and Adh 6, were discovered to vary in a clinal pattern. Adh 6 reaches a maximum frequency of about 0.90 in the South and minimum of about 0.50 in the North. Partially purified enzymes from the three Adh genotypes varied according to specific activity, substrate specificity, and heat stability. A differential influence of pH was indicated. Allele frequencies from seven polymorphic red cell antigen loci (ABO, Rh, MN, S, P, Duffy, and Diego) were examined in 144 Native American populations. Mean genetic distances (Nei's D) and the fixation index FST are approximately equal for the North and South American samples but are reduced in the Central American geographic area. The relationship between genetic distance and geographic distance differs markedly across geographic areas. The correlation between geographic distance and genetic distance for the North and Central American data is twice as large as that observed for the South American samples. This geographic difference is confirmed in spatial autocorrelation analyses; no geographic structure is apparent in the South American data but geographic structure is prominent in North and Central American samples. These results confirm earlier observations regarding differences between North and South American gene frequency patterns.  相似文献

Microsatellite loci were isolated from a Mahonia aquifolium cultivar. We describe the variability of 10 loci in invasive European and native North American M. aquifolium and their transspecies amplification in native Mahonia repens and Mahonia pinnata from North America and one species of the related genus Berberis (Berberis vulgaris), native to Europe. The markers should be useful to reveal the genetic origin of invasive Mahonia populations and differences in the genetic make up between invasive and native populations.  相似文献

Male genital morphology, allozyme allele frequencies and mtDNA sequence variation were surveyed in the butterfly species Lycaeides idas and L. melissa from across much of their range in North America. Despite clear differences in male genital morphology, wing colour patterns and habitat characteristics, genetic variation was not taxonomically or geographically structured and the species were not identifiable by either genetic data set. Genetic distances (Nei's D=0.002–0.078, calculated from allozyme data) between all populations of both species were within the range commonly observed for conspecific populations of other butterflies. The most frequent mtDNA haplotype was present in individuals of both species in populations from southern California to Wisconsin. We conclude that speciation has probably happened recently and the lack of genetic differentiation between the species is the product of either (1) recent or ongoing gene flow at neutral loci, and/or (2) an insufficiency of time for lineage sorting. The evolution of male genital morphology, wing colour patterns and ecological characteristics has proceeded more rapidly than allozyme or mtDNA evolution.  相似文献

Phosphoglucomutase (PGM; EC 2.7.5.1) isozyme variants were studied in a large number of inbred lines, crosses, and races of maize (Zea mays L.). Patterns of Mendelian inheritance demonstrated for PGM isozyme variants indicated that they are encoded by nuclear genes. Two unlinked loci, Pgm1 and Pgm2, located on the long arm of chromosome 1 and the short arm of chromosome 5, respectively, specify the observed electrophoretic variation on starch gels. No intra- or interlocus hybrid bands were found, suggesting that each isozyme band consists of a single polypeptide. PGM isozymes were present in all plant parts studied and the activity specified by both loci appears to reside in the cytoplasm. In studies of 520 racial collections of maize from Latin America, a single allele at each locus predominated in most collections. We characterized 10 polymorphic microsatellite loci from Nematostella vectensis, a burrowing anemone recently introduced to estuaries along the Pacific coast of North America and the southeast coast of England. Preliminary results indicate high variability and significant departures from Hardy–Weinberg equilibrium, the latter likely the result of population genetic structure and reproductive plasticity. Both results are consistent with earlier genetic analyses. These markers will be useful for resolving global patterns of introduction and for describing spatio‐temporal genetic structure at local and regional scales.  相似文献

Electrophoretic variation within and between North Atlantic minke whale samples(Balaenoptera acutorostrata) from West Greenland, Iceland, and Norway was investigated. In the West Greenland samples, 28 enzyme systems were examined, representing 36 loci, of which 6 were found to be polymorphic. In Icelandic and Norwegian samples, 22 enzyme systems were examined, representing 29 loci, of which 6 and 5 were found to be polymorphic, respectively. The average heterozygosity was 0.058 (SE=0.024) in samples from West Greenland, 0.074 (SE=0.028) in samples from Iceland, and 0.054 (SE=0.023) in samples from Norway. No significant deviations from the expected Hardy—Weinberg genotypic frequencies, within samples taken from the same area, were found. Significant differences in allele frequencies were observed, however, between samples from the three different areas. The average Nei's genetic distance was 0.014 and the averageF st value was 0.126. The genetic differences between the samples from the different areas indicate that those from West Greenland, Iceland, and Norway represented different breeding populations.  相似文献

The genetic variability of natural populations ofCapsella bursapastoris in North- and Middle-Europe has been estimated by means of enzyme assays. Zymograms of 81 populations have been developed. 17 loci could be identified, and 8 of them can be heterozygous. Genetic variability is greater between populations than within. No correlation between actual population sizes and genetic heterogeneity could be detected. Some electromorphs shift their frequencies proportionally to increasing adversity of climatic conditions, some appear to be constant over the whole area, and others are characterized by an apparently irregular variation pattern. Marginal populations comprise a significantly higher proportion of heterozygous plants than central ones. Apart from this clinal variation pattern, a mosaic pattern, strongly related to habitat conditions, was observed: genetic heterogeneity is greater in more intensively disturbed sites. The pattern of genetic variation in natural populations ofCapsella bursa-pastoris is rather highly influenced by the breeding system.  相似文献
