During terminal development of antral follicles, granulosa cells progressively lose their proliferative activity. Romanov (ovulation rate = 3) and Ile-de-France (ovulation rate = 1) breeds of sheep were compared for fractions of proliferating granulosa cells, determined by in vitro continuous [3H] thymidine labelling. In both breeds, the fraction of proliferating cells decreased with increasing follicular size according to a sigmoid-shaped curve. After linearization, the slope of the regression line was higher (in absolute value) in Romanov, compared to Ile-de-France ewes (p = 0.02). In vivo FSH treatment led to a decrease in the slope of the regression line in Romanov ewes only (p = 0.03). These results suggest that during terminal follicular development (1) the rate of cell cycle exit is higher in granulosa cells of Romanov, compared to lie-de-France follicles, and (2) Romanov granulosa cells are more responsive to exogenous FSH in term of proliferation. These mechanisms may underlie differential dynamics of follicular development in poly- and mono-ovulating breeds of sheep.  相似文献

Expression of gonadotropin receptors and granulosa cell sensitivity to gonadotropin hormones by small (1-3 mm) and large (3.5-7 mm) follicles were compared in Romanov (ROM, ovulation rate = 3) and Ile-de-France (IF, ovulation rate = 1) ewes in the early and late follicular phase. In healthy follicles, LH receptor levels in granulosa cells increased with increasing follicular size (p < 0. 001) while FSH receptor levels decreased (p < 0.05). In granulosa cells of large follicles, LH receptor (LHR) mRNA levels were greater in the late than in the early follicular phase (p < 0.001, p < 0.05, for ROM and IF, respectively). In the early follicular phase, LHR levels in granulosa (p < 0.001) and theca cells (p < 0.05) of small follicles were greater in ROM than in IF ewes. FSH receptor mRNA levels in granulosa cells of small and large ROM follicles were greater than in the corresponding IF follicles (p < 0.05). Finally, a greater responsiveness (increase in cAMP secretion) to both FSH and hCG was observed by granulosa cells collected during the early follicular phase from ROM vs. IF ewes. Data provide evidence that the greater ovulation rate in the ROM as compared to the IF breed is associated with a greater gonadotropin responsiveness during the early follicular phase.  相似文献

Ovulation rate, in mixed-age groups of prolific and non-prolific ewe breed types, after administration of a range of doses of PMSG (0, 375, 750 and 1500 i.u.) during the follicular phase of the oestrous cycle, were compared in Ireland, Morocco and New Zealand. The ewes in Ireland and Morocco were from the Finnish Landrace and Galway, and D'Man and Timhadite breeds, respectively. In New Zealand Booroola Merino x Romney ewes which had been previously identified as heterozygous carriers (F+) of the Booroola high fecundity gene and purebred Romneys were used to represent the prolific and non-prolific genotypes respectively; in addition a group of Booroola Merino x Romney non-carriers (++) of the major gene were also included for comparison. Ovulation rate at the oestrus which preceded stimulation with PMSG was also measured in all animals. In all 3 locations the ewes of the prolific genotype had a greater ovulation rate after PMSG stimulation than did the non-prolific controls. However, this association between prolificacy and response to PMSG was removed when ovulation rate after PMSG was transformed by dividing by the ovulation rate observed before PMSG administration. Despite the differences in the genetic basis of their high prolificacy the pattern of response to PMSG over the range of dosages used was similar in Finnish Landrace, D'Man and Booroola Merino x Romney (F+) ewes and all breeds had means of about 10 ovulations in response to 1500 i.u. PMSG. Amongst the non-prolific breeds, the Timhadite was the most responsive to PMSG although it had the lowest natural ovulation rate.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The pattern of ovarian follicle development in maiden cyclic lambs was characterized using the definition of a follicle wave as the changes in the number of follicles among the days of the estrous cycle, as originally defined in cattle by Rajakoski in 1960. We also examined the steroid content relationships among follicles on Days 5 (Wave 1) and 14 (Waves 2 and 3) of the estrous cycle. In Experiment 1, the ovaries of 20 cyclic lambs (40 to 45 kg) were examined daily using transrectal ultrasonography for 1 or 2 estrous cycles (n = 31 cycles). The number of small (2 and 3 mm in diameter), medium (4 and 5 mm) and large (> or = 6 mm) follicles were aligned with the beginning and end of the average length estrous cycle and then compared among days. Identified follicles were defined as those that grew to > or = 4 mm and remained at > or = 3 mm for > or = 3 d. The number of identified follicles emerging (retrospectively identified at 2 or 3 mm) per ewe per day was also aligned with the average length estrous cycle. In Experiment 2, ewe lambs were ovariectomized on Day 5 (n = 6) or 14 (n = 5) of the estrous cycle, then follicle diameters and follicular fluid concentrations of estradiol and progesterone were compared among follicles. Data were analyzed by repeated measures ANOVA and compared among days using Fisher's LSD. In Experiment 1, either 2 (n = 10 cycles), 3 (n = 20 cycles) or 4 (n = 1 cycle) periods of emergence of identified follicles occurred during individual cycles, with estrous cycle lengths of 15.6 +/- 1.6, 16.1 +/- 1.1 and 17 d respectively. In animals with 2 or 3 periods of emergence of identified follicles, the total number of small, medium and large follicles differed (P < 0.05) among days of the estrous cycle showing a wave-like pattern. In Experiment 2, a single follicle collected on each of Days 5 and 14 of the cycle (6.2 +/- 0.2 and 3.9 +/- 0.2 mm in diameter) had a higher (P < 0.05) concentration of follicular fluid estradiol (36.2 +/- 4.4 and 50.9 +/- 21.6 ng/mL) than other follicles collected on the same day (next largest follicle: 4.3 +/- 0.3 and 3.5 +/- 0.4 mm; 4.3 +/- 0.9 and 18.2 +/- 6.7 ng/mL estradiol). The results showed that 1) there was a synchronous emergence of follicles associated with fluctuations in the number and size of follicles during the estrous cycle; 2) within a wave there was a hierarchy among follicles for diameter and steroid content; 3) ovarian follicle growth in ewe lambs occurred in 2 or 3 organized waves during the estrous cycle.  相似文献

In developing countries, cross‐breeding between local breeds and indigene or exotic breeds represents one of the main threats to the livestock diversity, leading to genetic dilution and loss of unique allelic combination underlying essential local adaptive traits. In this study, two Algerian sheep breeds, known to be highly admixed, were considered as a case study, to demonstrate how combination of different methodologies coupled with the use of specific softwares can be efficient to assess the spatial structuration of a hybrid zone, even in a case of extreme admixture. A fine sampling covering distribution areas of both breeds was implemented in order to study the admixture area and adjacent zones from a phenotypic (i.e., 19 quantitative traits were considered) and a genetic point of view (i.e., 21 microsatellites markers were used). Both approaches gave concordant patterns, highlighting areas with sheep most differentiated (or less admixed) for each breed. In detail, the region of Biskra appeared as the most preserved for the Ouled‐Djellal breed and the northwest of Laghouat was identified as the most preserved area for the Rembi breed. The approach proposed in the study offers a low‐cost solution to identify the most representative flocks of a breed, allowing the implementation of efficient conservation plans.  相似文献

The structure and the genetic diversity of the Churra, Lacha and Manchega sheep breeds have been analysed using hemotypes observed in eight loci. The three breeds are different in their hemotypes in terms both of quantity and quality. The proportions of unique hemotypes in Churra (60%), Lacha (61%) and Manchega (67%) revealed a high level of individual diversity within each breed. Racial genetic diversity follows the descending order of: Manchega-Lacha-Churra. The value of N:H (number of animals: total hemotypes) in the multi-racial population was 3.05.  相似文献

The number and growth rate of follicles within classes based on granulosa volume were determined for ovaries taken from groups of 4-5-year-old, fine-wool Merino ewes drawn at different times of the year from a single strain flock maintained at Armidale, N.S.W. The breeding season of the flock normally extends from February to October and the mean ovulation rate rises from about 0.5 in February to about 1.8-1.9 during April-May. Ewes sampled when they were anoestrous or had one (single-ovulatory) or two (twin-ovulatory) recent corpora lutea did not differ in respect to the mean total number of ovarian follicles, the mean number of follicles in individual classes, the time for follicles to complete their rapid growth stage, or the incidence of follicle atresia. However, the ovaries of twin-ovulatory ewes contained significantly more follicles in the two terminal classes within the rapid growth stage than did the ovaries of single-ovulatory or anoestrous ewes (2.2 v. 0.9 and 1.0). This difference was attributed to the differing numbers of follicles per day entering into the rapid growth stage (5.2, 4.5 and 3.7 respectively in twin-ovulatory, single-ovulatory and anoestrous ewes).  相似文献

Although genomic selection offers the prospect of improving the rate of genetic gain in meat, wool and dairy sheep breeding programs, the key constraint is likely to be the cost of genotyping. Potentially, this constraint can be overcome by genotyping selection candidates for a low density (low cost) panel of SNPs with sparse genotype coverage, imputing a much higher density of SNP genotypes using a densely genotyped reference population. These imputed genotypes would then be used with a prediction equation to produce genomic estimated breeding values. In the future, it may also be desirable to impute very dense marker genotypes or even whole genome re‐sequence data from moderate density SNP panels. Such a strategy could lead to an accurate prediction of genomic estimated breeding values across breeds, for example. We used genotypes from 48 640 (50K) SNPs genotyped in four sheep breeds to investigate both the accuracy of imputation of the 50K SNPs from low density SNP panels, as well as prospects for imputing very dense or whole genome re‐sequence data from the 50K SNPs (by leaving out a small number of the 50K SNPs at random). Accuracy of imputation was low if the sparse panel had less than 5000 (5K) markers. Across breeds, it was clear that the accuracy of imputing from sparse marker panels to 50K was higher if the genetic diversity within a breed was lower, such that relationships among animals in that breed were higher. The accuracy of imputation from sparse genotypes to 50K genotypes was higher when the imputation was performed within breed rather than when pooling all the data, despite the fact that the pooled reference set was much larger. For Border Leicesters, Poll Dorsets and White Suffolks, 5K sparse genotypes were sufficient to impute 50K with 80% accuracy. For Merinos, the accuracy of imputing 50K from 5K was lower at 71%, despite a large number of animals with full genotypes (2215) being used as a reference. For all breeds, the relationship of individuals to the reference explained up to 64% of the variation in accuracy of imputation, demonstrating that accuracy of imputation can be increased if sires and other ancestors of the individuals to be imputed are included in the reference population. The accuracy of imputation could also be increased if pedigree information was available and was used in tracking inheritance of large chromosome segments within families. In our study, we only considered methods of imputation based on population‐wide linkage disequilibrium (largely because the pedigree for some of the populations was incomplete). Finally, in the scenarios designed to mimic imputation of high density or whole genome re‐sequence data from the 50K panel, the accuracy of imputation was much higher (86–96%). This is promising, suggesting that in silico genome re‐sequencing is possible in sheep if a suitable pool of key ancestors is sequenced for each breed.  相似文献

MicroRNAs (miRNAs) regulate the development and growth cycle of hair follicles (HFs). The molecular mechanism by which miRNAs determine the development of HFs in the sheep foetus remains elusive. In this study, the expression profiles of miRNAs at 11 development periods (45, 55, 65, 75, 85, 95, 105, 115, 125, 135 and 145 d) in sheep foetus skin were analysed by high-throughput sequencing and bioinformatics analysis. A total of 72 conserved miRNAs, 44 novel miRNAs and 32 known miRNAs were significantly differentially expressed. qRT-PCR results for 18 miRNAs were consistent with the sequencing data. 85 d of foetal development was the starting point for secondary hair follicle (SF) development according to tissue morphology and cluster analysis. In SF development, the prolactin signalling pathway and platelet activation played important roles, and 10 miRNAs were potential candidate miRNAs in SF initiation.  相似文献

Twenty-one of the world's prolific sheep breeds and strains were tested for the presence of the FecB mutation of BMPR1B and the FecX(I) mutation of BMP15. The breeds studied were Romanov (2 strains), Finn (2 strains), East Friesian, Teeswater, Blueface Leicester, Hu, Han, D'Man, Chios, Mountain Sheep (three breeds), German Whiteheaded Mutton, Lleyn, Loa, Galician, Barbados Blackbelly (pure and crossbred) and St. Croix. The FecB mutation was found in two breeds, Hu and Han from China, but not in any of the other breeds. The 12 Hu sheep sampled were all homozygous carriers of FecB (FecB(B)/FecB(B)) whereas the sample of 12 Han sheep included all three genotypes (FecB(B)/FecB(B), FecB(B)/FecB+, FecB+/FecB+) at frequencies of 0.33, 0.58 and 0.08, respectively. There was no evidence of FecX(I) in any of the breeds sampled.  相似文献

Large-scale evaluations of genetic diversity in domestic livestock populations are necessary so that region-specific conservation measures can be implemented. We performed the first such survey in European sheep by analysing 820 individuals from 29 geographically and phenotypically diverse breeds and a closely related wild species at 23 microsatellite loci. In contrast to most other domestic species, we found evidence of widespread heterozygote deficit within breeds, even after removing loci with potentially high frequency of null alleles. This is most likely due to subdivision among flocks (Wahlund effect) and use of a small number of rams for breeding. Levels of heterozygosity were slightly higher in southern than in northern breeds, consistent with declining diversity with distance from the Near Eastern centre of domestication. Our results highlight the importance of isolation in terms of both geography and management in augmenting genetic differentiation through genetic drift, with isolated northern European breeds showing the greatest divergence and hence being obvious targets for conservation. Finally, using a Bayesian cluster analysis, we uncovered evidence of admixture between breeds, which has important implications for breed management.  相似文献

Variations of associations between different behavioural patterns were analysed during post-embryonic development in the water stick insect,Ranatra linearis L. (Heteroptera: Nepidae). Behavioural patterns recorded were: quiescence, swimming, crawling, leg waving, body swaying, respiratory ascents breathing, respiratory descents, grooming and feeding activities (predatory attempts, foreleg movements, ingestion). A factorial analysis of correspondance stressed the contrast between the organization of larval behaviour and that of adult behaviour. No behavioural patterns were dropped from the repertoire during post-embryonic development ofRanatra linearis and no new patterns developed in adults outside the reproductive period. Larval behaviour was characterized by more frequent respiratory activities and grooming and adult behaviour was characterized by more frequent locomotory activities. At all larval stages grooming was associated with respiratory activities and crawling, but not with feeding activities.  相似文献

This study investigates the number and size of ovarian antral follicles in relation to plasma follicle stimulating hormones (FSH) and luteinizing hormone (LH) concentrations from birth to 26 weeks of age in ewe lambs of the Ouled Djellel breed, a non-seasonal breed of sheep. Plasma was collected from 10 ewe lambs at 14 sampling times (Week 0, i.e. <24h, Week 1 and every two weeks from Week 4 to Week 26, inclusive). At each of these stages, four ewe lambs were slaughtered, the ovaries recovered and weighed, and the number and size of the follicles determined from histological examination. The pattern for plasma FSH showed a peak at Week 10, a smaller peak at Week 18 and a very small peak at Week 24. The pattern for LH was similar until Week 24 when the largest peak occurred. Paired ovarian weight increased rapidly from birth to four weeks and then more slowly to 10 weeks, followed by a decline at 12 weeks and a gradual increase from 14 to 24 weeks of age. The number and total diameter of follicles > or =3 mm in diameter showed similar patterns of development--rising gradually from birth to Week 14, falling to Week 16 and then rising more rapidly to a peak at Week 24. Maximum follicle diameter declined from birth to Week 1, then rose rapidly to Week 4, followed by a more gradual rise to Week 14 and, thereafter, a more rapid increase to a peak of 7.23+/-0.16 mm at 24 weeks old. The number of follicles (<3 mm diameter) increased rapidly from birth to Week 10 and then declined to values similar to those at Weeks 1 and 4. First behavioural oestrus was observed at Week 24 and a corpus luteum was present on the ovary of one lamb at Week 24 and two lambs at Week 26. It was concluded that two or three peaks in plasma FSH and LH levels precede puberty and first ovulation in Ouled Djellel ewe lambs, and first ovulation occurred at 24-26 weeks of age. The increase in follicle number and size generally reflected the pattern of plasma FSH and LH levels.  相似文献

Garole, a prolific small sized sheep breed of India was introduced in the Mutton Project of the Institute during 1997 to explore the possibility of incorporating fecundity gene into Malpura (M) sheep of semi-arid region of Rajasthan. The data on reproduction and production traits of M and Garole × Malpura (GM) half-breds was used in the study to assess the improvement in evolved crossbred stock. The overall percent gain in prolificacy in GM was 52.38, which increased to 75.73 in third parity. The GM compared to M ewes also weaned more lambs per ewe lambed (1 versus 1.41). The lambing rate averaged 91.75 and 148.21% in M and GM ewes, respectively. The twin lambing percentage was only 4.71% in M flock while; it reached up to 51.10% in GM. Moreover, 5.72% of GM ewes produced triplets also. The genetic group non-significantly affect ewe efficiency (EE), but GM produced 3.3% more total lambs weight per ewe lambed at weaning and 6.15% more total lamb weight per ewe lambed at 6-month of age. The body weights at different ages of GM half-breds were lower (P < 0.01) compared to contemporary M lambs. The parity of ewes significantly affects body weight from birth weight to 12-month of age. The survivability of GM half-breds was almost at par with the local M sheep. Results revealed that the fecundity genes responsible for increasing prolificacy have been incorporated into the GM and it might prove a valuable germplasm towards evolving a new prolific strain of sheep. The crossing of Garole with M increased the ewe efficiency.  相似文献

The aim of this study was to analyze, through the use of microsatellite molecular markers, the genetic diversity and variability of nine Alpine sheep breeds reared in Italy (Bergamasca, Biellese, Schwarzbraunes Bergschaf, Tiroler Bergschaf, and Schnalserschaf), Germany (Brillenschaf and Weisses Bergschaf) and Slovenia (Bovška and Jezerzkǒ-Solčavska), and one Italian crossbreed population. Allelic richness was rather high in each breed highlighting a considerable genetic diversity. However, the study evidenced a significant departure from Hardy–Weinberg equilibrium in all analyzed breeds caused by a heterozygote deficiency. Such lack seems to be caused both to the presence of population substructure within breed and to a rather high level of inbreeding. The genetic differentiation among breed was rather low (FST = 0.057) but significant. The clustering analysis performed with STRUCTURE detected the presence of eight clusters; Schwarzbraunes Bergschaf and Tiroler Bergschaf were grouped together and Biellese and Bergamasca as well, evidencing a high similarity of their genetic make up. Reynolds’ genetic distance estimates confirmed the close relationship between these pairs of breeds. Moreover, Bovška, Jezerzkǒ-Solčavska and Brillenschaf resulted rather similar as expected according to their region of origin and to their common ancestors. Molecular coancestry confirmed as well such findings, suggesting that the studied breeds had origin from different ancestral populations. Concluding, microsatellite resulted a useful tool to investigate breed variability and to characterize Alpine sheep breeds. Obtained findings suggest the need to set up a conservation plan aiming to safeguard and increase the genetic variability of the studied breeds compromised by the high level of inbreeding. Microsatellites genotyping could help to monitor breed variability and to organize matings.  相似文献

The objective of genome mapping is to achieve valuable insight into the connection between gene variants (genotype) and observed traits (phenotype). Part of that objective is to understand the selective forces that have operated on a population. Finding links between genotype–phenotype changes makes it possible to identify selective sweeps by patterns of genetic variation and linkage disequilibrium. Based on Illumina 50KSNP chip data, two approaches, XP‐EHH (cross‐population extend haplotype homozygosity) and FST (fixation index), were carried out in this research to identify selective sweeps in the genome of three Iranian local sheep breeds: Baluchi (= 86), Lori‐Bakhtiari (= 45) and Zel (= 45). Using both methods, 93 candidate genomic regions were identified as harboring putative selective sweeps. Bioinformatics analysis of the genomic regions showed that signatures of selection related to multiple candidate genes, such as HOXB9, HOXB13, ACAN, NPR2, TRIL, AOX1, CSF2, GHR, TNS2, SPAG8, HINT2, ALS2, AAAS, RARG, SYCP2, CAV1, PPP1R3D, PLA2G7, TTLL7 and C20orf10, that play a role in skeletal system and tail, sugar and energy metabolisms, growth, reproduction, immune and nervous system traits. Our findings indicated diverse genomic selection during the domestication of Iranian sheep breeds.  相似文献

The objective of the present experiment was to determine whether failure to become pregnant through out-of-season reproductive management regimes is attributable to a failure to stimulate normal corpora lutea (CL) production. Romney ewes and East Friesian Composite (EF) ewes were mated in summer, autumn, winter, and early and late spring after administration of intravaginal progesterone inserts (plus eCG in spring and summer) in a factorial (two breedsxfive mating periods) experimental design. Oestrus rate was determined from mating data, and the numbers of CL were determined by laparoscopy 9 days after ram introduction. Conception rate, pregnancy rate and litter size were determined by transabdominal ultrasonography 62 days after ram introduction. Proportions of ewes displaying oestrus did not differ significantly (75-100%) between seasons or breeds, except for the Romney ewes in summer (67%; P<0.05). Numbers of CL on Day 9 for EF ewes was lowest in autumn (1.5) and winter (1.5), and highest in late spring (2.1; P<0.05). In contrast, Romney ewes had the lowest CL count in summer (1.3), and the highest in autumn (1.8) and winter (1.6; P<0.05). Only in summer was there a difference between breeds (EF 1.8; Romney 1.3; P<0.01). The proportion of ewes that failed to conceive despite having one or more CL present was highest in late spring (EF 41%, Romney 43%), and lowest in autumn (EF 9%, Romney 4%) and winter (EF 14%, Romney 4%; P<0.05). Conception and pregnancy rates followed similar patterns with values for autumn (EF 91%, 91%; Romney 96%, 96%) and June (EF 86%, 82%; Romney 91%, 83%) being significantly (P<0.05) higher than in early (EF 50%, 40%; Romney 54%, 50%) and late spring (EF 44%, 36%; Romney 42%, 36%). Pregnancy rate in summer was higher for EF ewes (60%) than for Romney ewes (39%) but conception rates were not statistically different (EF 68%; Romney 60%). Numbers of foetuses identified at scanning was highest in autumn (1.5) and lowest in late spring (0.5 P<0.001): Litter size in pregnant EF ewes was highest in early spring and lowest in winter (1.8 versus 1.2; P<0.01), but for pregnant Romney ewes was highest in winter and lowest in early spring (1.9 versus 1.3; P<0.001). It was concluded that seasonal differences in the ability of ewes to conceive are not the consequence of failure to display oestrus or to ovulate, but probably are a result of failure of fertilisation or the establishment of pregnancy.  相似文献

Electrophoretic and hybridization studies indicated that a haemoglobin found in three rare Dutch breeds of sheep was the a chain variant HbD which had hitherto only been found in three individual native Yugoslav sheep. Two phenotypes, Hb DAB and Hb DB, were detected, and the D variant was always present in lower concentrations than the normal haemoglobins. The calculated gene frequencies for αD were 0.030, 0.082 and 0.029 in the Kempisch Heide, Veluws Heide and Mergelland breeds respectively.  相似文献
