Trichomycete fungi are common endobionts of aquatic insect larvae, but little is known of their ecology. In this study, the seasonality of trichomycete colonization of larval black flies (Diptera: Simuliidae) was investigated in three streams in northwestern South Carolina. At least eight species of trichomycetes were found in two species of black flies, and 93.8% of 1819 larval black flies examined contained trichomycetes. Significant differences were found in the seasonal prevalence of Harpella melusinae, Simuliomyces microsporus, and Paramoebidium spp. at one of three sites. At this site, the lowest mean prevalence for H. melusinae occurred in winter (67%) versus the other seasons (96-100%), whereas mean prevalence was lowest in summer for Simuliomyces microsporus (1%) versus the other seasons (2-21%) and lowest in summer for P. spp. (9%) versus the other seasons (45-67%). Significant differences in levels of colonization among seasons were not detected. Conjugations of H. melusinae (representing early stages of sexual reproduction) occurred most frequently in the spring and winter (up to 14% of larvae). Sexual reproduction (represented by zygospores) of Legeriomycetaceae occurred most frequently in the spring and fall (up to 17% of larvae).  相似文献

Klastostachys reflexa, a new genus and species combination in the Harpellales, is established herein based on Stachylina reflexa, which was described in 1988. This gut fungus was found attached to the peritrophic matrix of small bloodworms, Cryptochironomus sp. (Chironomidae), in the Rocky Mountains of Colorado, USA. Klastostachys resembles Stachylina, a genus common in Chironomidae, but the unbranched thalli of Klastostachys disarticulate and disperse with the trichospores remaining attached to their generative cells. This manner of dissemination is unusual among Harpellales, being noted also for Carouxella spp., but members of that genus have zygospores attached at one pole to the zygosporophore (Type IV), whereas Klastostachys zygospores are medially attached to the zygosporophore at right angles (Type I).  相似文献

A total of 2063 mid- to late-instar larval black flies were collected from 64 stream sites in South Carolina and screened for the presence of the trichomycete fungus Harpella melusinae. Sixteen of 18 host species were colonized by H. melusinae on at least one occasion. Prevalence of H. melusinae in larvae of Simulium tuberosum cytospecies "A" was highest in acidic streams with low conductivity, whereas H. melusinae colonized larvae of Simulium verecundum most frequently in slower-moving streams. Ecological conditions, therefore, can serve as predictors of the prevalence of H. melusinae. Prevalence in host larvae was significantly lower in the Piedmont ecoregion than in the Mountain ecoregion. Prevalence did not differ in the host species S. verecundum across ecoregions, suggesting that different prevalences among host species might indicate some host preference. The prevalence of H. melusinae differed significantly between two univoltine host species (Simulium venustum and Prosimulium magnum) at the same site but not between two multivoltine host species (S. tuberosum cytospecies "FG" and S. tuberosum cytospecies "CDE"), suggesting that host life history could be important in determining fungal prevalence.  相似文献

A new genus of gut fungus in the Harpellales (Trichomycetes) with the type species Dacryodiomyces oklahomensis, found in Chironomidae larvae, is described from northeastern Oklahoma, USA. Baetimyces, belonging to the Legeriomycetaceae, is described here as a new genus. Its single species, B. ancorae, has been found growing in the hindgut of mayfly nymphs of the genus Baetis (Ephemeroptera, Baetidae) inhabiting a Pyrenean Mountains canal-stream from the northeastern region of the Iberian Peninsula. The new genus is characterized by having trichospores with two thick appendages of unequal length, and by zygospores perpendicularly and medially attached to the zygosporophore. The new genus may be related to Glotzia, Legeriomyes, and Zygopolaris, and similarities with these and other genera are compared and discussed. The new species often coexists in the same gut with Legeriomyces ramosus, which is reported for the first time in Spain.  相似文献

Studies of the interactions between two or more species of symbiotic microorganisms in a single host animal have lagged behind those involving interactions between hymenopteran or dipteran parasitoids. We screened the larvae of four species of black flies (Cnephia ornithophilia, Simulium furculatum, Simulium innoxium, and Simulium vandalicum) to determine if the colonization and prevalence of trichomycete fungi were influenced by ichthyosporeans, mermithid nematodes, microsporidia, or other species of trichomycetes. Differences in the layering of the peritrophic matrix of microsporidian-infected larvae, compared with that of uninfected larvae, were associated with greater levels of colonization by the midgut-inhabiting trichomycete Harpella melusinae. Colonization levels of different species of trichomycetes were significantly correlated with one another in only 2 of 11 cases. Aside from the consistent attachment of the trichomycete Simuliomyces microsporus to the ichthyosporean Paramoebidium chattoni, we found little evidence that prevalence and colonization levels of trichomycetes were influenced by other symbiotes, including other trichomycetes. This trend might reflect the physical separation of trichomycetes, which are gut specific, from other symbiotes. In addition, any combinations of symbiotes that were wholly incompatible would not have been observed in our field-collected material.  相似文献

In the investigations reported here, the length of zygospore incubation or “maturation” prior to the induction of meiosis was found to affect the inheritance pattern of chloroplast genes. The frequency of zygospores transmitting chloroplast alleles from both parents drops with increasing zygospore age following mating, while the frequencies of zygospores homoplasmic for maternal or paternal chloroplast alleles increase correspondingly. Since there is a negligible reduction in viability, zygospores which are initially biparental appear to become pure for the chloroplast genes from one or the other parent prior to the occurrence of cell division. These results are amplified in crosses of mt+ cells which have been irradiated with ultraviolet (uv) light or grown in the presence of the base analog, 5-fluorodeoxyuridine, which also perturbs maternal inheritance. Low doses of uv irradiation, applied to zygospores derived from crosses in which the maternal parent was also irradiated prior to mating, increase the biparental zygospore frequency while reducing the proportion of maternal zygospores. This indicates that at least some maternal zygospore clones are actually derived from zygospores which still contain both parental chloroplast genomes prior to the induction of germination. Thus, a subclass of zygospores must contain paternal chloroplast genomes which are either eliminated upon germination or are not expressed in the resulting zygospore clone. Tetrad analysis of biparental zygospores derived from uv-irradiated mt+ gametes demonstrates that the frequency of maternal chloroplast alleles in biparental zygospores decreases as they age. One result is an increase in the proportion of meiotic products homoplasmic for all paternal markers. The increased segregation of homoplasmic daughter cells during the meiotic divisions may result from a reduction in chloroplast ploidy by elimination of maternal genomes. Alternatively, it may reflect an altered ratio of maternal:paternal genomes due to continuous rounds of pairing and gene conversion between heterologous chloroplast DNAs leading to genetic drift within the DNA population of the organelle.  相似文献

Two pairs of stable diploid clones were obtained as aberrant forms among F1 progeny of an intragroup (intraspecific) cross between R-11-4 (mating type +) and M-16-4b (mating type -) of Group A of Closterium ehrenbergii Menegh. Each pair was derived from the two germination products of a single zygospore, and both clones were mating type minus. The cell size range of these four diploid minus clones was considerably above that of normal (haploid) Group A clones. Chromosome counts at the second meiotic metaphase indicated that these clones were diploid with approximately 200 chromosomes, which was double the number for normal Group A clones. Diploid minus clones conjugated normally with any haploid Group A plus clones, and yielded many triploid zygospores. Triploid zygospores germinated normally as did intragroup diploid zygospores. In metaphase I preparations, only bivalents were observed except on a few occasions where some uni- and multivalents were also detected. Viability of F1 progeny from triploid zygospores (55–74%) was somewhat lower than from diploid zygospores of Japanese Group A populations (65–90%), but higher than intergroup (interspecific) hybrid zygospores from Groups A, B and H (0–12%). In addition to lower viability, some F1 progeny from triploid zygospores exhibited slow vegetative growth. Almost all pairs of F1 clones from single triploid zygospores were of opposite mating type, similar to normal diploid zygospores of the intragroup cross. Morphological variability of F1 progeny of triploid zygospores was great. The apparently normal meiosis of triploid zygospores and the high viability of F1 progeny suggested that the genome of Group A contains several sets of chromosome complements with mechanisms by which bivalents are regularly formed in the first meiotic division.  相似文献

The presence of zygospores in the genus Orphella is newly described. We found zygospores in three species of the genus, O. catalaunica, O. coronata and O. helicospora, which are all the species of the genus known from the Iberian territory. Zygospores are associated with a heterothallic conjugating sexual process in O. coronata, whereas in O. catalaunica and O. helicospora, they form homothallically. In all instances, zygospores are consistently associated with an organized pattern of sterile cells, forming structures comparable to those present with asexual trichospores. We compare the ontogeny of Orphella zygospores with that found in the harpellid Genistellospora homothallica and discuss the possible close relationship of Orphella with Kickxellales (Zygomycetes). We report O. coronata in Spain for the first time, replacing all previous records of O. haysii. Results are supported with line drawings and photographs.  相似文献

There is a need for simple, inexpensive methods to maintain algal clones of constant genotype over long periods of time. Pandorina zygospores survive many environmental rigors which destroy the vegetative cells. The zygospores are the preferable from for storage of the alga and remain viable for at least 15 yr. storage procedures and germination techniques are described for zygospores of 2 species. These are compared with reports in the literature concerning other algal genera. General procedures for storage and maintenance of both vegetative cells and spores are proposed.  相似文献

The recent data on the pathways of carotenoid biosynthesis, which resulted in revision of the earlier concepts, are analyzed. Trisporoid diversity and their role in the sexual process of mucoraceous fungi, resulting in formation of sexual cells (zygospores) are discussed. Zygospore formation in different strains of the Closterium peracerosum-strigosum-littorale complex was examined in this unicellular isogamous charophycean alga to shed light on gametic mating strains in this taxon, which is believed to share a close phylogenetic relationship with land plants. Zygospores typically form as a result of conjugation between mating-type plus (mt+) and mating-type minus (mt) cells during sexual reproduction in the heterothallic strain, similar to Chlamydomonas. However, within clonal cells, zygospores are formed within homothallic strains, and the majority of these zygospores originate as a result of conjugation of two recently divided sister gametangial cells derived from one vegetative cell. In this study, we analyzed conjugation of homothallic cells in the presence of phylogenetically closely related heterothallic cells to characterize the reproductive function of homothallic sister gametangial cells. The relative ratio of non-sister zygospores to sister zygospores increased in the presence of heterothallic mt+ cells, compared with that in the homothallic strain alone and in a coculture with mt cells. Heterothallic cells were surface labeled with calcofluor white, permitting fusions with homothallic cells to be identified and confirming the formation of hybrid zygospores between the homothallic cells and heterothallic mt+ cells. These results show that at least some of the homothallic gametangial cells possess heterothallic mt-like characters. This finding supports speculation that division of one vegetative cell into two sister gametangial cells is a segregative process capable of producing complementary mating types.  相似文献

The first two non-Mendelian gene mutations to be identified in Chlamydomonas moewusii are described. These putative chloroplast gene mutations include one for resistance to streptomycin (sr-nM1) and one for resistance to erythromycin (er-nM1). In one- and two-factor reciprocal crosses, usually over 90% of the germinating zygospores transmitted these mutations and their wild-type alternatives from both parents (biparental zygospores); the remaining zygospores transmitted exclusively the non-Mendelian markers of the mating-type "plus" parent. Among the biparental zygospores, a strong bias in the transmission of non-Mendelian alleles from the mating-type "plus" parent was indicated by an excess of meiotic and postmeiotic mitotic progeny that were homoplasmic for non-Mendelian alleles from this parent compared to those that were homoplasmic for the non-Mendelian alleles from the mating-type "minus" parent. At best, weak linkage was detected between the sr-nM1 and er-nM1 loci. Non-Mendelian, chloroplast gene markers in Chlamydomonas eugametos and Chlamydomonas reinhardtii showed a predominantly uniparental mode of transmission from the mating-type "plus" parent in crosses performed under the same conditions used for the C. moewusii crosses.  相似文献

The zygospore of Chlamydomonas is a diploid resting stage that provides protection from environmental extremes. The remarkable abiotic stress resistance of the zygospore can be explained, in part, by the presence of a massive wall that includes a sporopollenin‐containing surface layer ( Van Winkle‐Swift and Rickoll 1997 ). A Chlamydomonas monoica Strehlow zygospore‐specific mutant strain (D19) was obtained previously by screening for loss of chloroform resistance in zygospore populations derived from self‐mating of post‐mutagenesis clones. Exposure of D19 zygospores to solar UV radiation or germicidal radiation also resulted in a pronounced decrease in survival of D19 zygospores relative to wildtype zygospore survival. Similarly, resistance to NaCl‐induced osmotic shock was reduced in D19 zygospores, especially when exposed to very high (e.g., 20% w/v) salt concentrations. Mature zygospores of C. monoica exhibit a UV‐induced blue surface autofluorescence that may indicate the presence of phenolic wall components. The intensity of zygospore autofluorescence was significantly reduced in D19 zygospores. As revealed by TEM, the surface layer of mature homozygous D19 zygospores was disrupted, suggesting a defect in wall assembly. Zygospore‐specific chloroform sensitivity, UV sensitivity, and reduced autofluorescence cosegregated in tetrads derived from D19 heterozygotes (i.e., if a progeny clone from a cross involving D19 and a normal strain was found to be chloroform sensitive, it was always also UV sensitive and showed reduced autofluorescence), indicating that all three characteristics were the consequence of the same Mendelian mutation.  相似文献

Treatment of zygospores of Choanephora cucurbitarum with KMnO4, NaClO or H2O2 effectively activated the spores and induced their germination. The optimum concentration of KMnO4 for activation of zygospores was 0.25 to 0.5%. Zygospores were not able to germinate in darkness even after activation by KMnO4. When zygospores from 40 to 50-day-old cultures were treated with 0.5% KMnO4 solution for 60 min before incubation on water agar at 24% under light, about 50% germinated in 10 days. KMnO4 treatment killed more than 99% of residual mycelial fragments, sporangiospores and sporangiola in the zygospore suspension. During germination disappearance of oil droplets in zygospores occurred prior to the cracking on zygospore wall. Both sporangial germination and mycelial germination were found. Moreover, sporangiole germination was observed for the first time.  相似文献
