目的 检测极端环境分离出的真菌拮抗临床病原真菌的活性.方法 选用极端环境分离出的24株真菌菌株上清液及菌丝体的提取液,来进行临床病原真菌(孢子丝菌和犬小孢子菌)的拮抗试验,采用M38-A2产孢丝状真菌抗真菌药物敏感性试验方案和E-test纸片扩散法.结果 抑菌圈直径≥1.0 cm,具有较强拮抗孢子丝菌活性的极端环境真菌有8株;抑菌圈直径≥2.0 cm,具有极强拮抗犬小孢子菌的活性的极端环境真菌有1株.两种临床菌株的拮抗试验,90%的拮抗结果相一致.结论 70%极端环境真菌菌株上清液及菌丝体提取液具有拮抗临床病原真菌孢子丝菌和犬小孢子菌的活性.  相似文献

Wood-inhabiting fungi include many molds, wood-staining fungi, and decay fungi. Most of these fungal species can result in economic losses to wood users. Studies on molds, staining fungi, and decay fungi are necessary to be able to control their growth on wood and wood products. In this study, wood-inhabiting fungi were isolated from logs of 3 major Canadian hardwood species: sugar maple, white birch, and yellow birch. Two media were used for isolation. From these 3 wood species, a total of 1198 fungal cultures were obtained from summer- and winter-harvested logs in dry storage and under water sprinkling. The results showed that most fungal species were not host specific and affected all of the wood species tested. Frequently isolated molds were Alternaria alternata, Trichoderma species, and Mucor/Rhizopus (Zygomycota) species, frequently isolated staining fungi were Ophiostoma piceae and Ophiostoma piliferum, a frequently isolated bark saprophyte was Nectria cinnabarina, and frequently isolated decay fungi were taxa of the phylum Basidiomycota. More fungal species were isolated from summer-harvested logs than from winter-harvested logs. Fewer fungal cultures, especially decay fungi, were isolated from logs in early storage than from logs in late storage.  相似文献

【目的】明确湛江地区红树林滩涂海洋真菌的种类及其分布,为海洋真菌的开发利用研究奠定基础。【方法】运用稀释平板法从湛江市高桥及特呈岛红树林滩涂不同的潮位带(低、中、高)和不同树种(白骨壤、桐花树、木榄、红海榄)采集淤泥样品550份,采用真菌形态学和ITS序列分析技术进行多样性研究【。结果】分离获得海洋真菌274株,共鉴定出19属39种真菌,以曲霉属Aspergillus、青霉属Penicillium和木霉属Trichoderma真菌分离频率高,为湛江红树林滩涂优势真菌种类,尤其在中潮位带真菌种类最多。此外,分离获得真菌Talaromyces helicus,为中国新记录种。【结论】湛江红树林滩涂海洋真菌的种类十分丰富,具有潜在的开发和利用前景。  相似文献

The development of protocols for the conservation of fungi requires knowledge of the factors controlling their distribution, diversity, and community composition. Here we compare patterns of variation in fungal communities across New Zealand's Nothofagus forests, reportedly the most myco-diverse in New Zealand and hence potentially key to effective conservation of fungi in New Zealand. Diversity of leaf endophytic fungi, as assessed by culturing on agar plates, is assessed for three Nothofagus sp. growing in mixed stands from four sites. Host species was found to have a greater influence on fungal community assemblage than site. The leaf endophyte communities associated with Nothofagus solandri and Nothofagus fusca (both Nothofagus subgenus Fuscopora), were more similar to each other than either were to the community associated with Nothofagus menziesii (Nothofagus subgenus Lophozonia). The broad taxonomic groups isolated, identified on the basis of internal transcribed spacer (ITS) sequences, were similar to those found in similar studies from other parts of the world, and from an earlier study on the endophyte diversity in four podocarp species from New Zealand, but there were few matches at species level. Average levels of endophyte species diversity associated with single Nothofagus species and single podocarp species were similar, despite historical literature and collection data recording more than twice as many fungal species on average from the Nothofagus species. The significance of these findings to fungal conservation is discussed.  相似文献

Identified (n = 17) and unidentified (n = 1) fish-pathogenic fungal species from 10 genera of Oomycetes and soil fungi were isolated from 40 infected freshwater fish samples of the species Oreochromis niloticus niloticus (Nile tilapia) and Clarias gariepinus (African catfish). Samples were collected from various fish farms in the Nile Delta, Egypt. Nile tilapia were tested in aquaria for their susceptibility to the commonest Oomycetes species, Aphanomyces laevis and Achlya klebsiana, and also against the 2 most prevalent pathogenic soil fungi, Paecilomyces lilacinus and Phoma herbarum. Two techniques were used: water bath exposure and intramuscular (subcutaneous) injection. Water bath exposure to the 2 species of Oomycetes caused greater mortalities of O. niloticus niloticus than intramuscular injection, but the reverse was true of the soil fungal species. Regardless of the infection method, the 2 Oomycetes species were more potent pathogens than the soil fungal species. In both gills and mytomal muscles of fish infected by A. laevis and P. herbarum, we measured and compared with controls the oxidative stress parameters total peroxide (TP), lipid peroxidation (LPO) and nitric oxide (NO), as well as levels of the antioxidants vitamin E and glutathione (GSH), and superoxide dismutase (SOD) and catalase (CAT) activities. Infection by these 2 fungal species through either spore suspension or spore injection significantly increased oxidative damage in gills and induced marked decrease in most studied antioxidants. In addition, both routes showed similar effects and A. laevis depressed the antioxidants CAT, vitamin E and GSH more than P. herbarum.  相似文献

The seasonal variation and distribution of microfungi in four soil types collected from two districts of Andhra Pradesh (India) were studied.Besides soil type and surface vegetation, it appears from the present study that soil moisture, organic matter, potassium, calcium, iron and phosphorus contents also may affect the fungal numbers favourably, while chlorides, total soluble salts, total nitrogen and manganese contents may have an adverse effect.Even alkaline soils harbour greater numbers of fungi, but small fluctuations in the pH seem to influence the fungal numbers in soils inversely.A total of 101 species representing 43 genera were isolated. These included 18 Phycomycetes, 5 Ascomycetes, 72 Fungi Imperfecti, 5 Mycelia Sterilia and a single Myxomycete. The order of occurrence of the chief genera of fungi isolated wasAspergillus, Penicillium, Fusarium, Pythium, Curvularia, Phoma, Cunninghamella, Rhizopus, Alternaria andTrichoderma.A large number of genera and species were found common to the forest, maize field, garden and uncultivated soils; and the fungal flora was also not very much different from those recorded from various parts of the world.  相似文献

Mites as Selective Fungal Carriers in Stored Grain Habitats   总被引:2,自引:0,他引:2  
Mites are well documented as vectors of micromycetes in stored products. Since their vectoring capacity is low due to their small size, they can be serious vectors only where there is selective transfer of a high load of specific fungal species. Therefore the aim of our work was to find out whether the transfer of fungi is selective. Four kinds of stored seeds (wheat, poppy, lettuce, mustard) infested by storage mites were subjected to mycological analysis. We compared the spectrum of micromycete species isolated from different species of mites (Acarus siro, Lepidoglyphus destructor, Tyrophagus putrescentiae, Caloglyphus rhizoglyphoides and Cheyletus malaccensis) and various kinds of stored seeds. Fungi were separately isolated from (a) the surface of mites, (b) the mites' digestive tract (= faeces), and (c) stored seeds and were then cultivated and determined. The fungal transport via mites is selective. This conclusion is supported by (i) lower numbers of isolated fungal species from mites than from seeds; (ii) lower Shannon–Weaver diversity index in the fungal communities isolated from mites than from seeds; (iii) significant effect of mites/seeds as environmental variables on fungal presence in a redundancy analysis (RDA); (iv) differences in composition of isolated fungi between mite species shown by RDA. The results of our work support the hypothesis that mite–fungal interactions are dependent on mite species. The fungi attractive to mites seem to be dispersed more than others. The selectivity of fungal transport via mites enhances their pest importance.  相似文献

The Federated States of Micronesia (FSM) is an island country in the western Pacific and is a known biodiversity hotspot. However, a relatively small number of fungi (236 species) have been reported till July 2021. Since fungi play major ecological roles in ecosystems, we investigated the fungal diversity of FSM from various sources over 2016 and 2017 and constructed a local fungal inventory, which also included the previously reported species. Fruiting bodies were collected from various host trees and fungal strains were isolated from marine and terrestrial environments. A total of 99 species, of which 78 were newly reported in the FSM, were identified at the species level using a combination of molecular and morphological approaches. Many fungal species were specific to the environment, host, or source. Upon construction of the fungal inventory, 314 species were confirmed to reside in the FSM. This inventory will serve as an important basis for monitoring fungal diversity and identifying novel biological resources in FSM.  相似文献

Fungal communities from gorgonians have been poorly documented and most studies of these communities have lacked defined sampling strategies. The objectives of this study were: (1) to estimate fungal diversity in Gorgonia ventalina; (2) to compare two sampling and tissue processing strategies: tissue fragments of different sizes vs. homogenized tissue. A total of seven genera and fourteen species of fungi were isolated on culture medium and identified by sequencing the nrITS. All but one species were new reports. In both treatments Aspergillus and Penicillium were the most common genera isolated. Most species isolated from fragments were not observed from homogenized tissue and vice versa. Reducing the size fragment increased significantly the number of species isolated per fragment. To better estimate fungal diversity in sea fans a strategy is proposed that combines sampling of small tissue fragments with homogenized tissue, since each technique yielded fungal species not detected by the other.  相似文献

This study was conducted to generate information regarding the diversity of fungi inhabiting creosote-treated wood in a storage yard for crosstie wastes in Gwangmyeong, Korea. Additionally, the resistance to polycyclic aromatic hydrocarbons (PAHs) of indigenous fungi that mainly occupy creosote-treated wood was evaluated. We isolated fungi from the surface and inner area of crosstie wastes and identified them using a combination of traditional methods and molecular techniques. Overall, 179 isolates including 47 different species were isolated from 240 sampling sites. The identified fungal species included 23 ascomycetes, 19 basidiomycetes, and 5 zygomycetes. Three species, Alternaria alternata, Irpex lacteus, and Rhizomucor variabilis, were the most frequently isolated ascomycetes, basidiomycetes, and zygomycetes, respectively. The results of this study showed that there was a large difference in the fungal diversity between the surface and the inner area. Additionally, zygomycetes and ascomycetes were found to have a greater tolerance to PAHs than basidiomycetes. However, two basidiomycetes, Heterobasidion annosum and Schizophyllum commune, showed very high resistance to PAHs, even in response to the highest concentration (1,000 ppm), which indicates that these species may play a role in the degradation of PAHs.  相似文献

Endophytic fungi represent an interesting group of microorganisms associated with the healthy tissues of terrestrial plants. They represent a large reservoir of genetic diversity. Fungal endophytes were isolated from the inner bark segments of ethnopharmaceutically important medicinal tree species, namely Terminalia arjuna, Crataeva magna, Azadirachta indica, Holarrhena antidysenterica, Terminalia chebula, and Butea monosperma (11 individual trees), growing in different regions of southern India. Forty-eight fungal species were recovered from 2200 bark segments. Mitosporic fungi represented a major group (61%), with ascomycetes (21%) and sterile mycelia (18%) the next major groups. Species of Fusarium, Pestalotiopsis, Myrothecium, Trichoderma, Verticillium, and Chaetomium were frequently isolated. Exclusive fungal taxa were recovered from five of the six plant species considered for the study of endophytic fungi. Rarefaction indices for species richness indicated the highest expected number of species for bark segments were isolated from T. arjuna and A. indica (20 species each) and from C. magna (18 species).  相似文献

A. chartarum was isolated from paper bag containing sugar and the other four from tomato sauce. A survey of the fungi isolated from the digestive tract of Panstrongylus megistus (insects vectors from Chagas' disease) used on xenodiagnosis was carried out. Two hundred and fourteen fungal strains were isolated from 180 nymphs. Aspergillus and Penicillium were the most predominant genera and some of their species were new records concerning insects. A great reduction in the fungal population was observed in the material that was positive for Trypanosoma cruzi.  相似文献

The unique mode of nutrition by carnivorous plants makes the facilitation of nutrient acquisition by mycorrhizal fungi seem unlikely. However, previously we have reported that the carnivorous plant Drosera rotundifolia can host at least eight species of fungal root endophyte. Although the function of these fungi remains unknown, their ubiquitous presence suggests that they may give plants a competitive advantage in both stressful and nutrient poor environments. The aim of this study was to examine the species of fungal endophyte colonising the roots of Pinguicula vulgaris and to compare them with the species isolated from D. rotundifolia found growing in the same location. Trichoderma spp. were isolated from every plant, whilst four other species were isolated once from single plants (Neonectria sp., Leohumicola spp., Cladosporium macrocarpum and Volutella ciliata). Although Trichoderma were isolated from both carnivorous plants, all of the other fungal endophytes were found colonising either P. vulgaris or D. rotundifolia. This observation may indicate a degree of host-specificity for these fungal species, despite the two host plants having very similar ecological strategies for living in a very nutrient poor and abiotically stressful environment.  相似文献

Endophytic fungi inhabit living plant tissues without causing disease symptoms. Although abundant, the extent of their contribution to fungal biodiversity remains unclear. Since endophytic fungi are poorly known, especially in the tropics, current estimates of fungal species are probably conservative. Here we tested strategies for sampling endophytic fungi in tropical plants. We compared the number of fungi isolated from 400 mm2 leaf pieces that were divided into increasingly small fragments. Leaf pieces were surface-sterilized, cut into fragments and plated on culture media. For a given area, cutting leaf pieces into smaller fragments significantly increased the number of fungal morphospecies recovered. There was a strong linear relationship between size of fragments and number of fungi isolated. By extrapolation, an estimated 16 +/- 3 fungi could be recovered from a 2 x 2 cm leaf piece, using infinitely small fragments. This represents a large part of the fungal diversity estimated to exist in leaf endophytes in a population. We conclude that reducing the size and increasing the number of leaf fragments will increase the number of fungal species isolated. This strategy will help to estimate real values of endophytic fungal diversity.  相似文献

Systemic mycoses in killer whales (Orcinus orca) are rare diseases, but have been reported. Two killer whales died by fungal infections at the Port of Nagoya Public Aquarium in Japan. In this study, the fungal flora of the pool environment at the aquarium was characterized. Alternaria spp., Aspergillus spp. (A. fumigatus, A. niger, A. versicolor), Fusarium spp. and Penicillium spp. were isolated from the air and the pool surroundings. The other isolates were identified as fungal species non-pathogenic for mammals. However, the species of fungi isolated from the environmental samples in this study were not the same as those isolated from the cases of disease in killer whales previously reported.  相似文献
