A new spectrophotometric procedure was developed for the quantitative determination of fucoidan and alginic acid and used for their analysis in extracts from biomass of 17 species of brown algae collected in coastal waters of the Kamchatka peninsula. In addition, neutral monosaccharides and mannitol were determined in the hydrolysis products of the alga biomass samples. The polysaccharide composition was shown to substantially depend on the algal species. The alginic acid content was maximal in the Alaria marginata blades; all the other representatives of the order Laminariales are also useful sources of the polysaccharides. At the same time, the fucoidan content is rather low in Laminariales. The highest content of fucoidan was found in Saundersella simplex, but Chordaria flagelliformis and Fucus evanescens are more practical fucoidan sources; the available supplies and the sugar composition make the latter alga the most suitable for the complex processing to prepare mannitol, fucoidan, and alginic acid.  相似文献

Sterile and reproductive tissues of five brown algae (Sargassum pallidum, Silvetia babingtonii, Fucus evanescens, Saccharina japonica, and Alaria ochotensis) from Russian Far East seas were compared for the content and monosaccharide composition of fucoidans as well as the content of laminarans and alginic acids. It was proved that reproduction has an apparent effect on fucoidan content and its monosaccharide composition. Fucoidan content in fertile tissues was shown to be 1.3–1.5 times as high as in sterile ones. Based on the present and previously reported data, we believe that fucoidan accumulation during the development of reproductive structure is a general trend for brown seaweeds. Reproduction also caused changes in the monosaccharide composition. As a result, fertile plants synthesized fucoidan with less heterogeneous monosaccharide composition in comparison with sterile ones. Structural changes of this polysaccharide are species-specific and perhaps depend on the type of the synthesized polysaccharide. The fertile plants of S. babingtonii yielded the highest fucoidan content [25% dry weight (dw)] among the tested species, whereas the maximum alginic acid content was found for the sterile tissues of S. japonica—up to 43% dw. There was no general trend in alginic acid content variation during sporulation. The alginic acid content was much higher in the sterile tissues of S. japonica in comparison with the fertile ones. On the contrary, the fertile tissues of A. ochotensis contained more alginic acid than the sterile ones.  相似文献

GC-MS of trimethylsilyl derivatives of the compounds present in the butanolic extract of biomass of brown seaweed Colpomenia peregrina from the Black Sea aided in identification of 24 components, including aliphatic hydroxy and keto and aromatic acids, glycerol, mannitol, floridoside, and monosaccharides. The polysaccharide composition of the biomass was also studied, with high sodium alginate and laminaran contents and a comparatively low level of fucoidan being revealed. The polysaccharides were isolated from the biomass by fractional extraction and purified by precipitation or ion exchange chromatography. The structures of alginic acid and laminaran were deduced from 13C NMR spectra and confirmed, in the case of laminaran, by methylation analysis. The sodium alginate was shown to contain more guluronic (G) than mannuronic acid (M) residues, the M/G ratio being 0.48. Laminaran was demonstrated to be a -glucan with 1 3 linkages in its backbone and 1 6 linkages in its branching points, which is characteristic of brown algae. Fucoidan turned out to be a complex heteropolysaccharide containing, in addition to fucose and sulfate, other neutral monosaccharides and uronic acids.  相似文献

The present study examines the polysaccharide composition of the brown alga Eisenia bicyclis. The major polysaccharide of E. bicyclis is shown to be sodium alginate (15.8% of the dry defatted alga weight). The yields of laminaran and fucoidan were 1.4% and 1.3% of the dry defatted alga weight, respectively. The alga contains laminaran (1,3;1,6-β-D-glucan) that has a unique structure, a high content of 1,6-linked D-glucose residues (the ratio of 1,3: 1,6 linkage is 1.5: 1), and an unusually high molecular weight (19–27 kDa). The fucoidan fractions isolated from E. bicyclis are sulfated and acetylated heteropolysaccharides that differ in their sulfate content and monosaccharide composition.  相似文献

We studied the possibility of stimulating physical activity in experimental animals using alginic acid, a polysaccharide isolated from a brown alga Laminaria cichorioides(family Laminaraceae). Seasonal changes in the content of alginic acid and fuccidan have been studied in four species of fucus algae from the Barents Sea: Fucus vesiculosus, F. distichus, F. serratus, and Ascophyllum nodosum. These polysaccharides are accumulated in the biomass of the algae during summer and autumn. To ensure complete processing of fucus algae it is recommended that the raw material be harvested within this period.  相似文献

Comparative study of phytochemical compositions of the most widespread brown algae species (one laminarian and four fucoid algae) from Barents Sea has been performed. A modified technique for mannitol determination in brown algae is proposed. It was revealed that fucus algae (fam. Fucaceae) contain 3% (of total dry weight) less mannitol than laminaria (Laminaria saccharina). The contents of alginic acid and laminaran in the Barents Sea fucoids are more than 10% less compared to laminaria. The alga L. saccharina contains almost two times more iodine than the species of fam. Fucaceae. The amounts of fucoidan and sum lipids in the Barents Sea fucoid algae is higher than in Laminaria saccharina (4–7% and 1–3%, respectively). In terms of contents of main biologically active compounds, fucus and laminarian algae from Barents Sea are inferior to none of the Far-Eastern species. The Barents Sea algae may become an important source of biologically active compounds.  相似文献

Crude fucoidan was extracted from the brown alga Undaria pinnatifida collected monthly from April to last July in Peter the Great Bay (Japan Sea, Russia). The amount of crude fucoidan rose markedly from April to June–July (from 3.2 to 16.0% dry weight) as the plant matures. An analysis of the monosaccharide composition of the fucoidan extracted showed that the alga synthesized polysaccharides with various structures which were dependent on the algae age. In juvenile plants collected in April–May, this was represented by sulfated manno-galactofucan containing up to 19–28 mol% of mannose and about 20 mol% of galactose, whereas in matured plants collected in June–July, the polysaccharide was represented by a sulfated galactofucan containing more than 38 mol% galactose. It is postulated that the production of sori causes a subsequent effect on fucoidan synthesis and leads to an enhanced of crude fucoidan content and an increased molar concentration of galactose. Crude fucoidan content in sporophylls increased 5 times, and galactose content in this polysaccharide rose s1.6 times with sori formation. The structural characteristics of the fucoidan extracted from sporophylls of Undaria collected in July were also studied. The fractionation of crude fucoidan on DEAE-Sephadex A-25 gave two fractions, F1 and F2 in equal quantities. F1 was characterized as manno-galactofucan sulfate and F2 was galactofucan sulfate. The molecular weights of both fractions were in a range of 30–80 kDa. Analysis of fucoidan structure using ESI-FTICR mass spectrometry showed the presence of mixed oligosaccharides consisting of fucose and galactose. Presumably, the polysaccharide molecules contain blocks built up of successively linked residues of fucose and galactose. These blocks are built from two to five or more residues of monosaccharides. According to IR-spectroscopy data, the main portion of sulfates is located at C2; in addition, sulfate esters are also present at C4 on the fucose and C3 and C6 of the galactose units.  相似文献

The capability to synthesize the extracellular polysaccharide (EPS) is widespread among eight mushroom species which accumulated 0.6–2.2 g/1 of EPS in submerged cultivation. Glucose, maltose, and mannitol were the most appropriate carbon sources for biomass and EPS production. Organic nitrogen sources appeared to be the most suitable nitrogen sources for biomass and EPS accumulation. The cultivation process in shake flasks was successfully reproduced in a laboratory fermentor with enhanced EPS production. The highest yield of EPS (3.8–4.0 g/1) was achieved in cultivation of Agaricus nevoi and Inonotus levis.  相似文献

A continuous micropropagation was established from protoplasts of thegreen alga Enteromorpha intestinalis. The effects of two differentcrude enzymes and the osmolarity at different concentrations of the enzymesolution on algal protoplast yields were tested. The optimal enzymecomposition for cell wall digestion and protoplast viability was 2%cellulase R 10 Onozuka and 2% Aplysie with 0.5 m mannitol. Largenumbers of Enteromorpha protoplasts were released (10.0 × 106protoplasts from 1 g fresh thalli) and settled on a rangeof substrata. Regeneration of the protoplasts followed the normal patternfor this species. Conditions for pure cultures and efficient systems offloating supports with nets were determined to optimise the product qualityof plantlets of Enteromorpha. A promising storage process has beendeveloped which involves including protoplasts in beads of alginic acid gel.Plants regenerated from protoplasts may also be used as seedstock tofacilitate propagation for macroalgal culture.  相似文献

The aims of this study were to characterise the composition of five seaweed species (Ascophyllum nodosum, Fucus serratus, Fucus vesiculosus, Laminaria hyperborea and Sargassum muticum), their extracts and commercial formulations, using thermogravimetry (TGA), energy dispersive X-ray microanalysis (EDX), Fourier-transform infrared spectroscopy (FTIR) and pyrolysis gas chromatography/mass spectrometry (Py-GC/MS). Analyses of the samples by TGA and EDX provided information on the proportions of algal cell wall, inorganic fractions and minerals. The main carbohydrate constituents of the five species and extracts were identified by their pyrolysis products, e.g. 1-(2-furanyl) ethanone, 5-methyl-2-furcarboxaldehyde, 2-hydroxy-3-methyl-2-cyclopenten-1-one, diannhydromannitol, 1,6-anhydromannopyranose and 1,6-anhydromannofuranose, using Py-GC/MS. The differences in relative intensities of the infrared bands of the five species were enhanced, especially after acid extraction compared with alkaline or neutral treatments, resulting in improved understanding of the compositional changes. In addition four commercial formulations and two acidic extracts of A. nodosum were evaluated for composition using the techniques. The dry matter, pH, electrical conductivity, ash, carbon and nitrogen content of the six preparations showed significant differences in composition. Variations in fatty acid, alginic acid, mannitol, laminarin and fucoidan content of the six formulations were reported. The results have shown that TGA, EDX, Py-GC/MS and FTIR are complementary techniques for rapid evaluation of seaweed materials and products.  相似文献

A compound that stimulated the growth of an arbuscular mycorrhizal (AM) fungus was isolated from 75% methyl alcohol (MeOH) extracts of a brown alga, Laminaria japonica Areschoug, using high-pressure liquid chromatography (HPLC). This compound (Compound 1) was identified as mannitol by HPLC and nuclear magnetic resonance (NMR) spectroscopy. Compound 1 and purchased polysaccharides (alginic acid, fucoidan, carrageenan and mannan from marine algae) were tested for in vitro hyphal growth of an AM fungus, Gigaspora margarita Becker and Hall. Compound 1 (50–500 mg L−1) and carrageenan (1000 mg L−1) significantly stimulated the hyphal growth of germinating spores of Gi. margarita. The application of 100 mg L−1 of Compound 1 to trifoliate orange (Poncirus trifoliata Raf.) inoculated with Gi. margarita promoted root colonization and increased plant growth. These results suggest low concentrations of mannitol are among the reasons for enhanced hyphal growth and root colonization by the application of algal extracts. Other sugar alcohols (100–300 mg L−1 of xylitol, sorbitol and meso-erythritol) also increased the hyphal growth of Gi. margarita.  相似文献

The water-soluble polysaccharides of brown algae attract the increasing attention of researchers as an important class of polymeric materials of biotechnological interest. The sole source for production of these polysaccharides has been large brown seaweeds such as members of Laminariales and Fucales. A new source of water-soluble polysaccharides is suggested here: it is a filamentous brown alga Streblonema sp., which can be cultivated under controlled conditions in photobioreactors that allow obtaining algal biomass with reproducible content and quality of polysaccharides. The accumulation of water-soluble polysaccharides can be stimulated by macronutrient limitation. In response to nitrogen deficiency, Streblonema sp. accumulated water-soluble polysaccharides (WSPs) rich in laminaran. WSP accumulation started after 3–4 days following nitrate depletion and reached a plateau at around day 7. Polysaccharide accumulation was related to cellular nitrogen content. The critical internal N level that triggered the onset of polysaccharide accumulation was 2.3% dry weight (DW); at a cellular N concentration less than 1.4% DW, the polysaccharide synthesis stopped. Upon nitrate re-supply, mobilization of WSP occurred after 3 days. These results suggest that a two-stage cultivation process could be used to obtain large algal biomass with high water-soluble polysaccharide production: a first cultivation stage using nitrate-supplemented medium to accumulate algal biomass followed by a second cultivation stage in a nitrate-free medium for 3 to 7 days to enhance polysaccharide content in the alga.  相似文献

Extraction of Lessonia vadosa(Laminariales, Phaeophyta) collectedin three different localities near PuntaArenas in the south of Chile, with 3%aqueous sodium carbonate solutions gave asodium alginate yield in the range3.0–17.7% dry weight. Among the Mediterranean algae belonging to the family Cystoseiraceae analysed for diterpenic compounds, only Cystoseira elegans contains eleganolone, epoxy-eleganolone and elegandiol, as does the Atlantic alga Bifurcaria bifurcata. Surprisingly, for the seasonal Cystoseira elegans, these compounds are present only at the beginning of growth of the alga during Spring and they disappear in the Summer yielding important seasonal variation data for diterpenoids, and also for sterols, alginic acid,mannitol and cellulose.  相似文献

Fucoidans are matrix polysaccharides from marine brown algae, consisting of an α-l-fucose backbone substituted by sulfate-ester groups and masked with ramifications containing other monosaccharide residues. In spite of their interest as biologically active compounds in a number of homologous and heterologous systems, no convenient sources with fucanase activity are available yet for the degradation of the fucalean algae. We here report on the isolation, characterization, and culture conditions of a bacterial strain capable of degrading various brown algal fucoidans. This bacterium, a member of the family Flavobacteriaceae, was shown to secrete fucoidan endo-hydrolase activity. An extracellular enzyme preparation was used to degrade the fucoidan from the brown alga Pelvetia canaliculata. End products included a tetrasaccharide and a hexasaccharide made of the repetition of disaccharidic units consisting of α-1→3-l-fucopyranose-2-sulfate-α-1→4-l-fucopyranose-2,3-disulfate, with the 3-linked residues at the nonreducing end.  相似文献
