哒嗪类化合物9403诱导小麦雄性不育的初步研究 总被引:10,自引:2,他引:10
9403是国内新合成的一种哒嗪类化合物,经初步研究证明,9403具有诱导小麦雄性不育的作用。试验以GENESIS化学杂交剂为对照,对9403诱导小麦雄性不育的作用进行了研究。结果表明;9403对试验的两个品种西农6419与BAU95在其花药发育的药隔期,用剂量0.75kg/hm^2进行一次叶面喷施,诱导雄性不育率可达到96%以上,人工饱和授粉证明9403对雌蕊的育性没有影响,平均人工饱和授粉结实率为88.6%,与GENESIS相比,9403的杀雄效果较不彻底且具有一定的药害。 相似文献

利用育性恢复基因(Rf3)的近等基因系1031-1、S-165和1031-1与S-165之间的正交与反交,创建了四个实验品系(1031-1、S-165、不育品系、反交品系);采用改进的蛋白质聚丙烯酰胺凝胶双向电泳技术,从发育遗传学的角度,对小麦T型细胞质雄性不育和可育株旗叶表达的相关蛋白产物进行差异分析。通过对旗叶蛋白的双向电泳分析,发现4个品系有相近的蛋白质双向电泳图谱。没有相应穗中差异蛋白质的出现。从蛋白质水平上证实了不育基因与恢复基因表达具有器官特异性特征。 相似文献

利用拟南芥中已克隆的雄性核不育基因MS2和水稻中假定雄性不育蛋白的保守区域,设计一对简并引物,并在太谷核不育小麦可育株及不育株花药中进行扩增,得到了一条134bp的片段。以该片段为基础,通过电子延伸得到一个长为1604bp的序列,该序列编码的氨基酸包含一段由200个氨基酸组成的雄性不育保守区。RT-PCR结果表明,该雄性不育同源序列只在小麦可育花药中表达,而在小麦败育花药、叶片和根中不表达,说明该雄性不育同源序列为花药发育特异基因。 相似文献

Cytoplasmic male sterility (CMS) is a maternally inherited trait that causes dysfunctions in pollen and anther development. CMS is caused by the interaction between nuclear and mitochondrial genomes. A product of a CMS-causing gene encoded by the mitochondrial genome affects mitochondrial function and the regulation of nuclear genes, leading to male sterility. In contrast, the RESTORER OF FERTILITY gene (Rf gene) in the nuclear genome suppresses the expression of the CMS-causing gene and restores male fertility. An alloplasmic CMS line is often bred as a result of nuclear substitution, which causes the removal of functional Rf genes and allows the expression of a CMS-causing gene in mitochondria. The CMS/Rf system is an excellent model for understanding the genetic interactions and cooperative functions of mitochondrial and nuclear genomes in plants, and is also an agronomically important trait for hybrid seed production. In this review article, pollen and anther phenotypes of CMS, CMS-associated mitochondrial genes, Rf genes, and the mechanism that causes pollen abortion and its agronomical application for rice are described. 相似文献

小麦光温敏雄性不育相关基因的DDRT-PCR分析及功能预测 总被引:10,自引:0,他引:10
以小麦光温敏雄性不育系BS210为试材,分别在北京顺义(可育环境)和安徽阜阳(不育环境)两种诱导育性表现的生态环境下种植,并于雌雄蕊分化期、药隔形成期、花粉母细胞形成期、四分体期和单核期5个时期提取小穗组织的RNA,利用DDRT-PCR方法分离不育系在光、温因子诱导下的差异表达mRNA,并通过反向Northern 进行验证.对获得的20条差异表达的EST进行了测序和BLAST分析,得到了4个候选基因的片段,对其进行5′Race 扩增、序列分析及功能预测.结果表明:它们分别与水稻的DNA修复重组蛋白基因rad50、小麦穗部表达的LRR重复序列型类受体激酶基因、玉米叶片坏死斑点L1s1基因的序列相似性分别为89%、89%和88%,另外还筛选到1个未知功能的新基因片段,它和rad50分别在可育和不育环境下表达的mRNA具有相同的5′端编码区,但3′端非编码区 相似文献

Endonuclease restriction fragment patterns of Pennisetum americanum L. mitochondrial DNAs (mtDNAs) from a cytoplasmic male-sterile (CMS-A1), fertile revertants and a normal fertile cytoplasm were variable, while chloroplast DNA from those lines lacked variation. Comparisons between mtDNAs of CMS-A1 (parental) and fertile revertant lines revealed the presence of a unique 4.7 kbp PstI fragment in the sterile line that was not detected in any of the revertant lines. A 9.7 kbp PstI fragment was found in all of the revertants, but not in the CMS-A1. Neither of those fragments was found in the normal cytoplasm mtDNA. Hybridization studies revealed two sets of multiple homologies: 1) the 4.7 kbp fragment had homology with a 10.9 kbp and a 13.6 kbp fragment; and 2) the 9.7 kbp fragment was homologous with the 13.6 kbp fragment. The presence of those two repeated mitochondrial sequences on the altered fragments suggests that they may be involved in the recombinational associated events with reversion from CMS to fertility in P. americanum.Florida Agricultural Experiment Station Journal Series No.7797. 相似文献

In comparing the genetic organization and exploring the molecular basis of cytoplasmic male sterility (CMS) in wheat, mitochondrial DNAs (mtDNA) from Triticum aestivum, T. timopheevi, CMS alloplasmic wheat with T. aestivum nucleus and T. timopheevi mitochondria, and fertility-restored lines were compared by hybridization analysis with specific probes for three gene regions: CoxII, cob, and coxI. Minor differences between T. aestivum- and T. timopheevi-derived sources were found for gene regions for coxII and cob. For coxI, there are significant differences between T. timopheevi-derived mtDNAs and T. aestivum mtDNA extending beyond an 8 kb distance. All T. timopheevi-derived mtDNA sources have a chimeric gene region (orf256) with part of the upstream coxI gene region, including some coxI-coding region, preceding coxI. The part of orf256 that does not include any of coxI and the 3-flanking region of CMS coxI are not found in T. aestivum mtDNA. Neither orf256 nor the CMS 3-flanking region of coxI are found in T. timopheevi or T. aestivum chloroplastic or nuclear DNA. There do not appear to be DNA sequence differences for the three gene regions studied that are related to either CMS or fertility-restored states. 相似文献

利用具有粘果山羊草 Aegilops kotschyi 、易变山羊草 Ae.variabilis 、偏凸山羊草 Ae.ventricosa 和二角山羊草 Ae.bicornis 异源细胞质的粘、易、偏和二角型 1B/ 1R、非 1B/ 1R型小麦雄性不育系为母本 ,一些优良小麦品种 系 为父本进行杂交 ,就 4类异源细胞质 1B/ 1R、非 1B/ 1R型不育系的育性恢复性进行比较研究 ,结果表明 : 1 4类异质 1B/ 1R、非 1B/ 1R型杂种自交结实率均低于相应普质杂种 ,说明由于核质不协调 ,4类异源细胞质对不育系育性恢复表现出一定的负效应 ; 2 非 1B/ 1R型杂种自交结实率明显高于 1B/ 1R型杂种 ,并达极显著差异 ,说明恢复性除受异源细胞质影响外 ,还与核内染色体核型密切相关 ; 3 非 1B/ 1R型杂种恢复度均在 6 5 %以上 ,大于 80 %的组合占 6 0 %以上 ,且变异范围较小 ,有望进一步研究利用 ; 4 1B/ 1R型杂种恢复度不高且变异较大 ,与1B· 1B/ 1R杂合染色体在减数分裂过程中的异常行为密切相关 ;而非 1B/ 1R型杂种由于不含 1B· 1B/ 1R杂合染色体 ,减数分裂过程中染色体行为相对稳定 ,易恢复且恢复度高 ,很有实际利用前景 相似文献

In most flowering plant species, pollination and fertilization occur during the hot summer, so plants must have evolved a mechanism that ensures normal growth of their pollen tubes at high temperatures. Despite its importance to plant reproduction, little is known about the molecular basis of thermotolerance in pollen tubes. Here we report the identification and characterization of a novel Arabidopsis gene, THERMOSENSITIVE MALE STERILE 1 ( TMS1 ), which plays an important role in thermotolerance of pollen tubes. TMS1 encodes a Hsp40-homologous protein with a DnaJ domain and an a_ERdj5_C domain found in protein disulfide isomerases (PDI). Purified TMS1 expressed in Escherichia coli (BL21 DE3) had the reductive activity of PDI. TMS1 was expressed in pollen grains, pollen tubes and other vegetative tissues, including leaves, stems and roots. Heat shock treatment at 37°C increased its expression levels in growing pollen tubes as well as in vegetative tissues. A knockout mutation in TMS1 grown at 30°C had greatly retarded pollen tube growth in the transmitting tract, resulting in a significant reduction in male fertility. Our study suggests that TMS1 is required for thermotolerance of pollen tubes in Arabidopsis, possibly by functioning as a co-molecular chaperone. 相似文献

以粘、易、偏型非1BL/1RS不育系ms(kots)-90-110、ms(var)-90-110为母本,一些优良小麦品种(系)为父本,广泛进行测交和育性恢复性分析,结果表明:(1)粘、易、偏型非1BL/1RS不育系不同于相同细胞质背景下1BL/1RS易位型不育系,突出地表现为:不育性稳定,恢复源广泛,高恢复度恢复系在普通小麦中占较大比例,且F1育性变异幅度小,变异系数低,并可将二者视为选择优良恢复系的辅助指标(2)粘、易、偏型非1BL/1RS不育系安全克服了同质1BL/1RS不育系产生单倍体的缺点,不育系和测交F1无单倍体产生;(3)粘、易、偏型非1BL/1RS不育系持有的易恢复性特点,为常规育种的最新成果直接应用于杂种小麦组配强优势组合创造了极有利条件。 相似文献
