Inoculation of the potato cut surfaces with an incompatiblerace of Phytophthora infestans induced an accumulation of rishitin,but only a trace occurred when infected by a compatible race.When the tuber was cut, a large amount of steroid-glycoalkaloids(solanine) accumulated in the cut surface tissue, although onlya trace was found in intact tissue. Only a small amount of solanine,if any, was contained in the wound periderm tissue. Most ofthe solanine seemed to be distributed in tissue neighbouringthe newly formed meristematic tissue zone. Distribution of solanineas a function of distance from the cut surface was exponential.Infection of the cut surface by an incompatible race of Phytophthorainfestans reduced the accumulation of solanine. The higher theconcentration of zoospore used, the less the solanine content.It has been reported that the higher the concentration of zoosporesused for inoculation of the cut surface, the less the numberof renewed meristematic cells in the wound tissue. In experimentsusing fresh and aged tubers, a good correlation between thenumber of renewed meristematic cells and solanine content wasfound. The accumulation of solanine in the wound tissue andits reduction due to infection by an incompatible race may berelated to renewed meristematic cells formation and its reductioncaused by the infection. Cadmium is known to reduce photosynthesis and overall growth of plants. Plants adopt several mechanisms of Cd detoxification, such as accumulation of sulfur-rich compounds, like glutathione (GSH) and its precursor cysteine. The accumulation of GSH is regulated by the activity of ATP-sulfurylase, a rate-limiting enzyme in sulfur assimilation. The carbon of Cys is provided through photosynthesis. Thus, a plant with the higher photosynthetic potential and ATP-sulfurylase activity may have the higher contents of Cys and GSH and therefore may provide for a greater tolerance to Cd stress. Mustard (Brassica juncea L. Czern and Coss.) cvs. Varuna (high photosynthetic potential) and RH30 (low photosynthetic potential) were subjected to 0 and 200 mg Cd/kg soil, and the activity of ATP-sulfurylase, the contents of Cys and GSH, oxidative stress, and activities of antioxidant enzymes were studied. Under 200 mg Cd/kg soil, cv. Varuna showed an increased ATP-sulfurylase activity, the higher contents of Cys and GSH, and the net photosynthetic rate than cv. RH30. In contrast, the activity of superoxide dismutase, the contents of thiobarbituric acid-reactive substances, and H2O2, and electrolyte leakage were found to be greater in cv. RH30 showing an increased oxidative stress than cv. Varuna. However, the activities of ascorbate peroxidase and glutathione reductase were greater in cv. Varuna than cv. RH30. The results show that a greater ATP-sulfurylase activity, an enhanced production of Cys and GSH, and an efficient antioxidant enzyme system in the high photosynthetic mustard cv. Varuna helped to the reduce the oxidative stress maintaing high photosynthesis.  相似文献

In studies on the infection of potato cultivars with different field isolates of Phoma exigua var. foveata, cultivar × isolate interactions, although sufficiently large to influence the relative pathogenicities of isolates on different cultivars, were not substantial enough to affect cultivar ranking order. Cultivar rank was markedly influenced by pathogen isolate only when both field and culture collection isolates were compared. This suggests that the complications of cultivar × isolate interactions can be avoided in cultivar screening tests by the use of recent field isolates of high pathogenicity. It was found that cultivar was considerably more important than isolate in determining gangrene lesion size. The need to consider both lesion diameter and depth when estimating rot size following point inoculation of different cultivars was confirmed.  相似文献

The coding sequence of the wild-type, cys-sensitive, cysE gene from Escherichia coli, which encodes an enzyme of the cysteine biosynthetic pathway, namely serine acetyltransferase (SAT, EC 2.3.1. 30), was introduced into the genome of potato plants under the control of the cauliflower mosaic virus 35S promoter. In order to target the protein into the chloroplast, cysE was translationally fused to the 5'-signal sequence of rbcS from Arabidopsis thaliana. Transgenic plants showed a high accumulation of the cysE mRNA. The chloroplastic localisation of the E. coli SAT protein was demonstrated by determination of enzymatic activities in enriched organelle fractions. Crude leaf extracts of these plants exhibited up to 20-fold higher SAT activity than those prepared from wild-type plants. The transgenic potato plants expressing the E. coli gene showed not only increased levels of enzyme activity but also exhibited elevated levels of cysteine and glutathione in leaves. Both were up to twofold higher than in control plants. However, the thiol content in tubers of transgenic lines was unaffected. The alterations observed in leaf tissue had no effect on the expression of O-acetylserine(thiol)-lyase, the enzyme which converts O-acetylserine, the product of SAT, to cysteine. Only a minor effect on its enzymatic activity was observed. In conclusion, the results presented here demonstrate the importance of SAT in plant cysteine biosynthesis and show that production of cysteine and related sulfur-containing compounds can be enhanced by metabolic engineering.  相似文献

The cystic fibrosis transmembrane conductance regulator (CFTR) protein is the only known apical glutathione (GSH) transporter in the lung. The purpose of these studies was to determine whether oral GSH or glutathione disulfide (GSSG) treatment could increase lung epithelial lining fluid (ELF) GSH levels and whether CFTR plays a role in this process. The pharmacokinetic profile of an oral bolus dose of GSH (300 mg/kg) was determined in mice. Plasma, ELF, bronchoalveolar lavage (BAL) cells, and lung tissue were analyzed for GSH content. There was a rapid elevation in the GSH levels that peaked at 30 min in the plasma and 60 min in the lung, ELF, and BAL cells after oral GSH dosing. Oral GSH treatment produced a selective increase in the reduced and active form of GSH in all lung compartments examined. Oral GSSG treatment (300 mg/kg) resulted in a smaller increase of GSH levels. To evaluate the role of CFTR in this process, Cftr knockout (KO) mice and gut-corrected Cftr KO-transgenic (Tg) mice were given an oral bolus dose of GSH (300 mg/kg) and compared with wild-type mice for changes in GSH levels in plasma, lung, ELF, and BAL cells. There was a twofold increase in plasma, a twofold increase in lung, a fivefold increase in ELF, and a threefold increase in BAL cell GSH levels at 60 min in wild-type mice; however, GSH levels only increased by 40% in the plasma, 60% in the lung, 50% in the ELF, and twofold in the BAL cells within the gut-corrected Cftr KO-Tg mice. No change in GSH levels was observed in the uncorrected Cftr KO mice. These studies suggest that CFTR plays an important role in GSH uptake from the diet and transport processes in the lung.  相似文献

To clarify the effects of storage temperature on potato components and acrylamide in chips, tubers from five cultivars were stored at various temperatures (2, 6, 8, 10, and 18 degrees C) for 18 weeks, and the contents of sugars, free amino acids in tubers, and acrylamide in chips after frying were analyzed. At temperatures lower than 8 degrees C, the contents of reducing sugars increased markedly in all cultivars, with similar increases in the acrylamide level and dark brown chip color. Free amino acids showed little change at the storage temperatures tested and varied within certain ranges characteristic of each cultivar. The contents of reducing sugars correlated well with the acrylamide level when the fructose/asparagine molar ratio in the tubers was <2. When the fructose/asparagine ratio was >2 by low-temperature storage, the asparagine content, rather than the reducing sugar content, was found to be the limiting factor for acrylamide formation.  相似文献

The nutrient contents of the two cultivare of Potato‐Irish Cobbler and Red Pontiac were quantitatively altered by Rhizoctonia bataticola during pathogenesis. The glucose content of both cultivare increased whereas sucrose, maltose and fructose contents decreased appreciably in quantity during the period of incubation. The total protein and lipid contents of both cultivare were also lowered quantitatively by the rot‐causing organism. The depletion of total carbohydrate, protein and lipid contents, was more in Irish Cobbler cultivar than in Red Pontiac cultivar.  相似文献

In order to explore a biotechnological method for improving potato tuber color and creating plants with increased anthocyanin contents, a potato UDP-glucose: flavonoid-3-O-glucosyltransferase (3GT) gene was inserted behind the GBSSI promoter of pBin19, and this construct was introduced into Solanum tuberosum L. cultivar Désirée plants by Agrobacterium-mediated transformation. Six independent transgenic lines overexpressing the 3GT gene were identified by PCR and Southern blot analysis from 18 kanamycin-resistant plants. Due to the expression of 3GT gene, the tuber color and the anthocyanin content were enhanced noticeably in the transgenic plants compared to the wild-type control plants. This result suggests that the 3GT gene can potentially be used to improve potato color and enhance levels of antioxidants in the diet.  相似文献

The incidence of potato virus Y (PVY) infection was assessed in samples of potato tubers, cv. Record, taken from Scottish seed stocks and English ware crops grown from some of these seed stocks. PVY was readily detected by ELISA of tuber sprouts. PVY-infected tubers were found in 10 seed stocks of 84 tested. The mean level of virus infection was 0.23%, 0.76% and 0.56% in Super Elite, Elite and AA stocks respectively. In 46 commercial ware crops grown from some of these seed stocks, a substantial proportion of the harvested tubers in all but one of the crops were infected with PVY, the mean percentage of infected tubers was 58.5%. Ware crops grown from seven seed stocks in which PVY had been detected (mean 6.2% infection in seed) contained a mean of 70% infected tubers, compared with 56% infection in crops grown from 39 stocks in which PVY was not detected in the seed tubers. The predominant PVY strain detected in the ware crops was the veinal necrosis strain (PVYvn).  相似文献

When soil was maintained at a mean water potential (Slatyer & Taylor, 1960) of the order of -80 J kg-1 at 25 cm depth throughout the growing period, much infection of potato tubers by Streptomyces scabies occurred, but when soil was irrigated to maintain it at potentials greater than - 13 J kg-1 at 10 cm depth infection was negligible. Until about 5 weeks after initiation, tubers were very susceptible to infection; irrigation during this period reduced scab considerably, but subsequent irrigation reduced it only slightly. At the low water potential, actinomycete populations on lenticels were high and bacterial populations low; these effects were reversed at the high water potential. Since also actinomycetes were more frequently isolated in the absence than in the presence of bacteria it was deduced that there was an interaction between these two groups. It is suggested that irrigation may decrease the population of S. scabies in tuber lenticels by increasing populations of bacteria antagonistic to it.  相似文献

The mobility of Cd in potato plants (Solanum tuberosum) was examined using both short‐term radioisotopic labelling with 109Cd and long‐term growth experiments in soil supplemented with Cd, with an emphasis on the pathways through which Cd is taken up by tubers. Split‐pot experiments showed that tubers and their associated stolons and stolon roots contribute only a minor fraction to the overall Cd absorption by the plant. Most of the Cd was absorbed by the basal roots. 109Cd absorbed from the soil was rapidly exported to other parts of the plant, especially the stem, with significant amounts appearing in the tubers within 30 h. Application of 109Cd to leaves showed that Cd can be rapidly distributed via the phloem to all tissues. The results suggest that unlike Ca, Cd has high mobility in plants in both xylem and phloem, and that stems may have an important role in transfer between these two pathways.  相似文献

Summary Effect ofN-acetyl-l-cysteine (NAC) administration on cysteine and glutathione (GSH) contents in rat liver and kidney was studied using intact and diethyl maleate (DEM)-treated rats and perfused rat liver. Cysteine contents increased rapidly, reaching peak at 10 min after intraperitoneal NAC administration. In liver mitochondria it increased slowly, reaching peak at 60 min. GSH content did not change significantly in these tissues. However, in liver and kidney depleted of GSH with DEM, NAC administration restored GSH contents in 60 and 120 min, respectively. Perfusion with 10 mM NAC resulted in 76% increase in liver cysteine content, but not in GSH content. Liver perfusion of DEM-injected rats with 10 mM NAC restored GSH content by 15%. Present findings indicate that NAC is an effective precursor of cysteine in the intact liver and kidney and in the perfused rat liver, and that NAC stimulated GSH synthesis in GSH-depleted tissues.  相似文献

Amyloplasts and starch containing plastids from green tissue – amylochloroplasts – from potato tubers ( Solanum tuberosum L., var. King Edward) were separated from other cell organelles by sedimentation in a discontinuous sucrose gradient. Their lipid composition was analysed with emphasis on galactolipids and phospholipids and the fatty acid compositions of these lipids. Irradiation of the tubers caused increased ratios of monogalactosyl diacylglycerol to digalactosyl diacylglycerol and of total galactolipids to total phospholipids in the plastid membranes. Furthermore, the degree of unsaturation of the fatty acids increased in all lipid classes analysed, this effect being most prominent in the galactolipids. The ultrastructural studies made on tuber tissue revealed that irradiation caused a change in starch grain size distribution concomitant with formation of membrane structures resembling grana within the envelope. In many cases prolamellar bodies and plastoglobuli were present.  相似文献

Soil fungal communities perform important ecological roles determining, at least in part, agricultural productivity. This study aimed at examining the fungal community dynamics in the potato rhizosphere across different development stages in two consecutive growing seasons (winter and summer). Microbial fingerprinting of rhizosphere soil samples collected at pre-planting, tuber initiation, flowering and at senescence was performed using ARISA in conjunction with Next Generation Sequencing (Illumina MiSeq). The epiphytic fungal communities on tubers at harvest were also investigated. Alpha-diversity was stable over time within and across the two seasons. In contrast, rhizospheric fungal community structure and composition were different between the two seasons and in the different plant growth stages within a given season, indicating the significance of the rhizosphere in shaping microbial communities. The phylum Ascomycota was dominant in the potato fungal rhizosphere, with Operational Taxonomic Units (OTUs) belonging to the genus Peyronellaea being the most abundant in all samples. Important fungal pathogens of potato, together with potential biological control agents and saprophytic species, were identified as indicator OTUs at different plant growth stages. These findings indicate that potato rhizosphere fungal communities are functionally diverse, which may contribute to soil health.  相似文献
