本文研究结果表明:紫外光照射1小时有利于兰花原球茎的增殖,照射0.5小时有利于原球茎的分化。较高剂量的紫外光照射可导致细胞核的变形、皱缩、线粒体液泡化。在一定照射剂量条件下,紫外光照射可提高细胞无丝分裂频率。  相似文献

本文研究了紫外、光对细胞膜γ—射线损伤的恢复作用。结果发现经γ—射线300Gy照射后再经紫外光照射5分钟的大白鼠红细胞悬浮液,不但不会加强细胞膜的损伤,反而对细胞膜有明显的恢复作用。经统计处理有显著差别(P<0.05). 实验还进一步证明:红细胞悬浮液经γ—射线或紫外光照射后, 在低温中释放的血红蛋白量与照射剂量成正比关系.可作为辐射(包括电离辐射或非电离辐射)对细胞膜损伤的一个方便易行的检查指标.  相似文献

利用离体生物测定法比较了13种植物源化合物对南方根结线虫(Meloidogyne incognita Chitwood)的毒力,并探讨了紫外光照射对这些化合物杀虫活性的影响。结果表明,对苯二酚、DL-薄荷醇、丁子香酚和苦豆碱对南方根结线虫的毒杀活性最强,邻苯二酚等6种化合物的毒杀活性次之,野靛碱、毒扁豆碱和间苯二酚的毒力较低。紫外光照射可明显降低苦豆碱和柠檬酸对南方根结线虫的毒杀活性,而其他11种化合物的毒杀活性则基本不受紫外光照射的影响。  相似文献

几种植物生长调节物质对大花蕙兰组培原球茎增殖的影响   总被引:11,自引:2,他引:9  
采用KC 培养基, 分别添加不同浓度的BA、KT、NAA 及KT+NAA 组合, 对大花蕙兰初代培养已分化出的原球茎进行了继代培养。结果发现:①BA 可加快原球茎的增殖速度, 但浓度超过0.5 mg/L 后增殖速度有所下降, 且原球茎变小呈暗绿色。②KT 的促进增殖效果好于BA, 但浓度超过1.0 mg/L 时, 增殖速度也有所下降, 原球茎变小。③NAA 促进原球茎增殖的效果明显好于BA、和KT, 但浓度大于1.0 mg/L 后增殖不再加快, 而且部分原球茎有变褐现象。④在KT 0.5~0.7 mg+NAA 0.7~1.0 mg/L 范围组合的KC 培养基上, 大花蕙兰原球茎增殖的速度和质量是比较理想的。考虑到较低的细胞分裂素和植物生长素有利于植物增殖过程中遗传性的稳定, 选择KT0.5 mg+NAA 0.7 mg/L 的KC 培养基作为大花蕙兰原球茎增殖培养基。  相似文献

本文探讨6-BA和2,4-D对铁皮石斛原球茎增殖和分化的影响。结果表明:铁皮石斛原球茎芽再生的较好培养基为MS,芽增长的较好培养基为MS + 6-BA 1 mg/L + 2,4-D 0.1 mg/L,原球茎再生芽生根和原球茎增殖的较好培养基也为MS。添加0.5 mg/L 2,4-D不利于铁皮石斛原球茎的保存,而不添加2,4–D或添加0.1 mg/L 2,4-D均有利于铁皮石斛原球茎保存,且有持续增长趋势。  相似文献

目的:探讨转基因MSC能否修复放射损伤的胸腺。方法:雌性BALB/C小鼠120只,随机分为4组:对照组30只,予以假照射;照射组30只,60Coγ射线胸腺单次照射,剂量为9Gy;照射+MSC组30只,9Gy照射后2小时输注转基因MSC;对照+MSC组30只,假照射后2小时输注转基因MSC。照射后1、7、14、21、28天检测各组小鼠血像、胸腺指数及胸腺病理。结果:照射+MSC组血像和胸腺指数在各个时间点均优于照射组,胸腺病理显示组织结构恢复明显加快。结论:转基因MSC能归巢至放射损伤胸腺并能促进胸腺的修复,为干细胞的临床应用提供了实验依据。  相似文献

利用正交实验设计研究不同种类和浓度的氮源对铁皮石斛原球茎生长的影响,利用完全随机实验设计研究不同真菌诱导子对铁皮石斛原球茎生长和多糖积累的影响.结果表明硝态氮促进铁皮石斛原球茎鲜重和干重的增加( P <0.05);铵态氮促进铁皮石斛原球茎鲜重增加( P <0.05).无论鲜重还是干重,硝态氮和铵态氮的影响均不存在互作,最佳组合为:67-V培养液 100 mg/L (NH4)2SO4 800 mg/L KNO3 30 g/L蔗糖 200 g/L马铃薯提取汁.F检验的结果表明,14种真菌诱导子对铁皮石斛原球茎生物量的增加(鲜重与干重)均无显著性影响( P >0.05).但与对照铁皮石斛原球茎的多糖含量相比,真菌诱导子g6、g14、g5、g4处理可使其多糖含量分别提高14%、9%、6%、4%.本研究获得了有利于铁皮石斛原球茎生长的硝态氮和铵态氮的最佳搭配方案以及有利于铁皮石斛原球茎积累多糖的四种真菌诱导子,表明通过液体悬浮培养生产铁皮石斛原球茎及其多糖成分具有较好的开发应用前景.  相似文献

对大花蕙兰试管苗营养器官及原球茎的解剖学研究结果表明:根由复表皮、皮层和维管柱组成,根毛丰富,皮层发达,内皮层明显,初生木质部月多元型,中央具髓,根茎由表皮,基本组织和维管束构成,维管束散生,属周木型;叶为等面叶,在上下表皮处分布有成束的厚壁组织,叶肉无栅栏组织和海绵组织之分,细胞排列紧密,维管束鞘由机械组织构成。原球茎原生分生组织的原套仅一层细胞,在顶端分生组织后面的薄壁细胞中,存在胚性细胞,由胚性细胞经球状胚可发育成幼原球茎。  相似文献

Certain aspects of protocorm development in Vanda were examined ultrastructurally. The parenchymal cells of the protocorm accumulate substantial quantities of lipid, protein, and carbohydrate reserves which disappear gradually with the senescence of the parenchymatous region. The proteinaceous reserves appear initially as discrete bodies which become intimately associated with clusters of small tubules. The tubules eventually disperse throughout the cytoplasm and disappear following depletion of the protein bodies. The lipid reserves also appear as discrete bodies and are associated with an electron dense, laminated inclusion which appears to increase in size with the disappearance of the lipid bodies. While plastids in the meristematic cells differentiate a well-developed thylakoid system and contain little starch, those of the parenchymal cells contain large starch grains and numerous osmiophilic droplets and develop meager thylakoid systems. Membrane-bound crystalline structures of hexagonal and rhomboid cross section occur frequently in the cytoplasm of senescent parenchyma cells. Trichome initials, which differentiate from the epidermis, contain few conventional organelles and exhibit numerous membrane-bound structures containing many small crystalline inclusions. Numerous vesicles accumulate at the tips of the trichomes in spaces between the cell wall and the plasmalemma.  相似文献

以墨兰种子胚经培养诱导萌发形成的原球茎为材料,研究其生长的最适条件。结果表明:最佳基本培养基是MS培养基;NAA0.5mgL+BA1.0mgL-1有利于生长和分化;在培养基中加入1%蔗糖和0.1%活性炭最有利于原球茎生长;pH5.0偏酸环境适合于生长;每天给予15.5umolm-2S-116h的光照最佳。如将上述单因子组合在一起,其原球茎鲜重增长率比对照(MS培养基)高46%。在不同切割方式中,掰开法鲜重增长率比纵切法或横切法都高。  相似文献

用正交设计方法对铁皮石斛原球茎具有高效增殖影响的激素(BA,NAA,KT,马铃薯汁)以及配比进行研究,结果表明:以茎段为外植体在培养基Ms+BA2.0mg/L+NAA2.0mg/L+马铃薯汁10%+糖3%,具有很好的原球茎诱导作用,50d后诱导率达95.20%;原球茎在1/2MS+BA2.0mg/L+NAA1.0mg/L+KT0.5mg/L+糖3%的培养基上,以液体悬浮培养,原球茎增殖达49.032g/50d。  相似文献

The mat-forming cyanobacterium Phormidium murrayi West and West isolated from a meltwater pond on the McMurdo Ice Shelf was grown in unialgal batch cultures to evaluate the temperature dependence of ultraviolet radiation (UVR) effects on pigment composition, growth rate, and photosynthetic characteristics. Chlorophyll a concentrations per unit biomass were generally reduced in cells grown under UVR (low UV-A plus UV-B). In vivo absorbance spectra showed that the carotenoid/chlorophyll a ratio increased as a function of photosynthetically available radiation (PAR) and UVR exposure and varied inversely with temperature. Ultraviolet inhibition of growth (percentage reduction of μmax at each temperature) increased linearly with decreasing temperature, consistent with the hypothesis that net inhibition represents the balance between temperature-independent photochemical damage and temperature-dependent biosynthetic repair. There was no significant effect of UVR on photosynthesis over the first hour of exposure, but significant UV inhibition was observed after 5 days. Unlike growth, however, there was no apparent effect of temperature on the magnitude of UV inhibition of photosynthesis. These results imply that assays of UVR effects on photosynthesis are not an accurate guide to growth responses and that low ambient temperatures can have a major influence on the UV sensitivity of polar organisms. In a set of assays at 20° C (preacclimation under 300 μmol photons·m?2·s?1 and 20° C), growth was strongly depressed by UVR over the first day of exposure but then gradually increased over the subsequent 4 days, approaching the growth rates in the minus UVR control. This evidence of acquired tolerance indicates that the damaging effects of UVR will be most severe in environments where there is a mismatch between the timescale of change in exposure and the timescale of UV acclimation.  相似文献

The effectiveness of ultraviolet radiation for on-line sterilization of cheese whey was investigated. The effects of flow rate and residence time on the performance of three UV reactors having different gap sizes (18, 13, and 6 mm) were studied. Six flow rates and six residence times were tested with the three UV reactors. The cheese whey used in this study had a very high turbidity (4317 NTU), very poor transmittance in the UV radiation germicidal range ( approximately 0%), and high percentage of large solid particles ( approximately 20% > 100 microm). Although the cheese whey physical characteristics showed low probability of sterilization using UV radiation, the study showed that UV radiation can be used on-line to sterilize cheese whey if the proper reactor gap size and the appropriate residence time are used. There were combined effects of the flow rate and gap size. The cell removal efficiency increased with increases in residence time and decreases in the UV reactor gap size. Removal efficiency of 100% was not achieved in this study with the first UV reactor (18-mm gap size), whereas 100% removal efficiency was achieved with the second (13-mm gap size) and third (6-mm gap size) UV reactors at residence times of 2.0 and 0.5 h, respectively. The microbial decay rates achieved in this study were 4.94, 7.62, and 20.9 h(-)(1) using the first, second, and third UV reactor, respectively. Residence times of 3.3, 2.1, and 0.8 h would be required to completely destruct a microbial population of 5.95 x 10(6) cells/mL using the first, second, and third UV reactors, respectively. Although cheese whey sterilization using UV radiation seems to be a good alternative to pasteurization, increases in cheese whey temperature resulted in lamp fouling. If online sterilization is to be used, the fouling problem should be investigated and a maintenance scheme for the UV reactor should be developed.  相似文献

以授粉后不同发育时期的同色兜兰种子为材料,观察其形态特征和萌发过程,并探讨建立同色兜兰高效快繁体系的最佳条件。结果表明,种子发育中后期即授粉后210~240d为较适宜的采收期,授粉后210d的种子萌发率最高(达77.79%);1/4 MS和1/2MS为同色兜兰适宜的基本培养基,添加100mL/L椰乳或1g/L蛋白胨对种子萌发及原球茎生长和分化有明显的促进作用;添加1g/L活性炭对原球茎褐化有一定的抑制作用,但添加剂量不宜过大;添加香蕉汁和苹果汁对同色兜兰种子萌发和原球茎生长分化有抑制作用;暗处理对同色兜兰种子萌发无影响;分化后的原球茎在壮苗和生根培养基上培养120d即可得到4~5片叶、高3~5cm的同色兜兰健壮试管苗。  相似文献

研究了影响金线兰种子非共生萌发的因素,并应用正交试验研究基本培养基、6-BA、ZT、NAA四种因素对原球茎增殖的作用。结果表明:授粉类型对金线兰种子非共生萌发影响较大,异株异花、同株异花以及同株同花授粉所得的种子的萌发率分别为78.53%、69.62%、39.87%;蒴果成熟度以生长150d为宜,采收后萌发率可达78.59%;冷藏影响种子的活力,种子的萌发率随冷藏时间的延长而降低;使用次氯酸钠浸泡后的种子与对照相比,其萌发率无明显差异;NAA对原球茎增殖作用显著,适宜于原球茎增殖的培养基为1/2MS+ZT0.5mg/L+NAA1.0mg/L。  相似文献

在1/2MS基本培养基中,分别加入由野生春兰和华石斛根部分离到的内生真菌所制成的4种菌丝提取物,对象牙白种子萌发形成的原球茎进行诱导培养。结果表明:4种菌株诱导子中3种可不同程度地促进象牙白的生长和分化。对象牙白原球茎的诱导作用依次为HF26>CF6>CF8,与对照相比,HF26菌株显著提高了象牙白的鲜重(P<0.05),鲜重增长率分别为96.13%、86.86%和61.18%;干物质重分别增加了130.70%、96.50%和124.30%,均达到极显著水平(P<0.01)。此外,CF8菌株能促进根条数的增加;HF26菌株最有利于原球茎的增殖;CF6菌株在促进成苗和根原基的分化上效果显著。结论:在象牙白原球茎生长的不同阶段,能够促进其达到最佳生长状态的菌株是不同的。  相似文献
